ABSTRACT Hermodactylus tuberosus Mill. (Iridaceae) is a species with a very short style and a tricarpellar unilocular ovary. For ornamental purposes it is usually reproduced by means of tubers, although both vegetative and seed reproduction occur in nature. However, its capsules do not contain many seeds in comparison to the high number of ovules inside the ovary. To understand this conflicting process a study on H. tuberosus pollen-pistil interaction was carried out. Pollen viability was evaluated by different techniques. Germination rate was tested in vitro and in vivo after stigma, intrastylar and intraovarian pollinations. A high percentage of the pollen grains were found to be viable resulting in a high percentage of in vitro germination and a low rate of germination on the stigma. Indeed, many pollen tubes developed after intrastylar and intraovarian pollinations. Several anomalies characterized pollen germination and pollen tube growth in vitro and in vivo. Observations from field trials indicated that capsules were only produced from open or hand, cross- pollinated flowers, confirming that the plant is self- incompatible and not apomictic. In addition, all the mature capsules contained only a few mature seeds suggesting the existence of pre- and postzygotic seed set inhibition. 相似文献

ABSTRACT The morphology, cytology and viability of Hermodactylus tuberosus L. (Iridaceae) pollen were examined from the first mitosis until maturation and after anther opening. During maturation, the pollen coat becomes modified, and the vegetative cell cytoplasm accumulates several types of reserve substances. In the vegetative cell cytoplasm, starch is quickly utilised whereas lipid inclusions of different dimensions, shape and composition occur during pollen maturation. Pollen from opened anthers have a thin pollen coat; the cytoplasm has mostly lipid reserves, and many small vesicles and vacuoles. It is similar in size or larger than pollen located inside the anther, and its viability does not decrease until one day after anther dehiscence. Large osmiophilic bodies, different from those of the vegetative cell cytoplasm, are present in the generative cell cytoplasm starting from the first stage of pollen development. The poorly developed pollen coat in pollen from opened anthers suggests that it plays a minor role in attracting insects for pollination. The size and structural and ultrastructural features of mature pollen indicate that it does not undergo dehydration and possesses sufficient vigour for immediate germination. 相似文献

Pollen of Crocus biflorus Miller subsp. biflorus from natural habitats of Tusculum (Frascati, near Rome, Italy) has been studied in order to compare its structure and physiology to pollen of other Crocus species belonging to the Crocus sativus group. Mature pollen grains are rounded, 60 μm in diameter, in-aperturate (but with surface incisions where exine is lacking). DAPI staining reveals a spindle-shaped generative nucleus which is intensely fluorescent, and vegetative nucleus which is less fluorescent, and is elongated with numerous lobes. At anthesis the pollen is bicellular, but about 2% of tricellular grains occur among the pollen grains released from the anthers as well as on both naturally or handpollinated stigmas. Pollen germination is low in vitro, but higher in vivo. The pollen tubes are of normal shape. An electron-dense surface coat is sometimes visible on the exine, which in many cases, is detached from the exine. The vegetative cytoplasm is very rich in glycolipid bodies surrounded by endoplasmic reticulum. The generative cell has a lobed cell wall and is surrounded by the vegetative nucleus. 相似文献

Pistil structure and pollen tube growth were investigated inLeptospermum myrsinoides and L. continentale (Myrtaceae). BothL. myrsinoides and L. continentale pistils consist of an ovarywith five locules, a style and a five-lobed dry, papillate stigma.A centrally located stigmatic cleft is present but extends onlyto the base of the stigma. Pollen germinates and grows intercellularlythrough the stigma into the central transmitting tissue of thestyle. Pollen tubes do not grow down the stigmatic cleft. Atthe base of the style the transmitting tissue separates intofive, each tract leading through the placenta to one of thefive locules. The pollen tubes continue to grow intercellularlythrough these five tracts entering the locules between the lobesof the placenta. Pollen tubes are smooth-walled and straightwhilst in the transmitting tissue of the style but produce shortlateral branches at regular intervals when in the locules. Branchingcontinues until pollen tubes enter ovules. It is suggested thatthe observed branching in the locules is a result of pollentubes following a chemotropic or thigmotropic pathway to theovules. The effect of sucrose, H2BO3, KNO3, Ca(NO2)2.4H2O and MgSO4.7H2O on pollen germination of Phytolacca dodecandra L. (endod)in a liquid medium was investigated. Sucrose and H3BO3 werecritical to pollen germination. A concentration of 10% sucroseand 161.8 µm H2BO3 gave over 70% germination. The germinationof pollen was not enhanced by Ca(NO3)2.4H2O, KNO3 and MgSO4.7H2O.Endod pollen was dehydrated over CaCl2 and stored in gelatincapsules in cryogenic vials at 175 °C, 1±1°C and 24±2 °C. The pollen moisture content atcollection was approx. 7.8% (f. wt basis) and dehydration overCaCl2 reduced it to about 1.4%. Pollen stored at 1±1°C and 175 °C maintained viability for over 6months. Pollen stored at room temperature lost viability within4 weeks of storage. Pollination with cryopreserved pollen resultedin normal fruit set. Abstract Mature pollen of C. thomasii Ten. (Iridaceae) has been studied from a morphological and physiological point of view and compared to that of C. sativus L., C. thomasii pollen is roundish, with a 83 μm diameter and a 14% of anomalous grains. Percentage of the positive alcoholdehydrogenase (ADH +) pollen grains is about 90% whereas the in vitro germinated rate reaches 55%. Exine is homogeneously thickened (2.5 μm) with randomly distributed thinner zones and 5 μm thick intine. Cytoplasm of vegetative and generative cells is very rich of small and large smooth vesicles and vesicles coated by ribosomes. The generative cell shows a thin ondulated pectocellulosic wall. Its nucleus is intensely fluorescent after treatment with the 4,6-diamidino-2-phenylindole (DAPI) fluorochrome. Although many ultrastructural aspects of the C. thomasii pollen are common in C. sativus. L., C. thomasii pollen is smaller, more regularly structured and germinates in vivo and in vitro in higher percentage than that of C. sativus. 相似文献

Abstract Fruit and seed set were obtained in the triploid Crocus sativus (saffron) either by hand crossing with pollen of the diploid C. cartwrightianus or by free pollination. The morphology of the capsules and seeds was similar in both Crocus species. The embryo is comparatively small, whereas the endosperm is formed of numerous layers of cells with hemicellulose-thickened walls and a cytoplasm containing few lipid and protein reserves. Seed germination gave rise to a white prophyll from which a green leaf emerged. At the base of this leaf, a small corm without tunics developed, which, after leaf desiccation, became dormant. These corms sown separately in pots were again capable of growth and emitted a single long green leaf. Vegetative development of new corms was similar in the diploid and hybrid saffron plants. In sites of past saffron cultivation the beds were found to contain degenerated corms and corms at different development stages. The absence of fruit and seed set in emasculated and self- and out-pollinated plants suggests that saffron does not form apomictic embryos. 相似文献

以2年生桔梗植株为材料,采用液体培养法研究了培养基种类、PEG、蔗糖、pH以及培养温度、培养时间对桔梗花粉离体萌发生长的影响,结果表明:(1)浓度为100~150 g.L-1的PEG可显著促进桔梗花粉萌发和花粉管的生长;200~250 g.L-1PEG显著促进花粉萌发,但对花粉管生长的作用不显著。(2)100 g.L-1的蔗糖有利于花粉萌发和花粉管生长,高浓度蔗糖(200 g.L-1)有明显抑制作用;(3)桔梗花粉离体萌发和花粉管生长的适宜培养基为ME3+BK+10%蔗糖+150 g.L-1PEG(pH5.8);(4)25~40℃条件下桔梗花粉均可较好萌发,以30℃培养1.5 h为最佳培养条件。 相似文献

The correlation between water content and viability of maizepollen grains was studied on the basis of the germination abilityof pollen from a single cross hybrid. There was found to beclose correlation between viability of the grains and theirtolerance to desiccation. Most of the pollen grains in the hybridexamined survived a reduction by almost 50 per cent of the originalwater content without loss of normal function. With water lossgreater than this, less vigorous pollen grains died or losttheir ability to form pollen tubes. Consequently, when pollinationwas carried out using pollen with a water content reduced bymore than 50 per cent, only the most tolerant pollen grainssurvived to take part in the competition which precedes fertilization.Dry pollen grains required a longer period to establish adhesionto the stigma surface and to initiate pollen tubes than pollengrains with higher water content, but otherwise their behaviourwas normal. If more than 80 per cent of the original water contentwas lost, disturbances occurred in the physiology of the grainssurviving the treatment. This was exhibited as death or a reductionin rate of pollen tube growth. Drying of pollen by an amount which does not irreversibly damagethe more tolerant grains could possibly be used as a means ofpollen selection. Previous studies have demonstrated that pollen of sorghum [Sorghumbicolor (L.) Moench] loses capacity to both germinate in vitroand to set seed in vivo soon after being shed. The current studyevaluates the capacity for dehydrated pollen to effect in vitrogermination, reduce tetrazolium chloride, and set seed on cytoplasmicmale sterile plants. Morphological changes during pollen germinationwere examined by scanning electron microscopy (SEM). Close to70% of the pollen germinated in 5 min, or less, when collectedat 80% relative humidity (RH) and stored in sealed glass vials.Pollen tubes elongated autotropically with atmospheric humidityapparently being a controlling factor in the process. Pollendehydrated at 50% RH and 25°C for 15-30 min neither germinatedin vitro, reduced tetrazolium chloride, nor set seed on malesterile plants. Rehydrating the pollen did not restore the capacityfor germination. SEM micrographs demonstrated that elongatingpollen tubes encircled the pollen grain and were contiguousto the surface. In Croatia and the surrounding area, the genus Iris is represented with about 20 Alpine-Dinaric, Mediterranean and Pannonian taxa from the subgenera Iris and Limniris. We researched pollen morphology of all taxa by using scanning electron microscopy. All pollen grains are sulcate with few palynological features, which could have taxonomic importance on different classification levels: shape of dry pollen grains; shape, outline and approximate size of hydrated pollen grains, approximate size of the sulcus, and ornamentation of the sulcus membrane and of the exine. At least four pollen types were recognised and taxonomically delimited to the series level: two characterise the subgenus Iris, section Iris (series Elatae and Pumilae), and two the subgenus Limniris, section Limniris (series Laevigatae, Sibiricae and Spuriae). Taxonomic implications of pollen morphology of their critical groups and taxa have been evaluated. Possible phylogenetic implications of pollen morphology of the genus Iris on the Southern European area were suggested: the subgenus Iris is more advanced than the subgenus Limniris; I. graminea seems to be the most primitive member of the subgenus Limniris on the Southern European territory; and pollen morphology of the population of the subspecies I. sibirica subsp. erirrhiza from the Mountain Bjelolasica could represent a link between the subgenera Limniris and Iris in the territory of Southern Europe. The possible pathway of evolution of the genus Iris on the territory of Southern Europe was suggested: from the subgenus Limniris, through the “linked taxon” I. sibirica subsp. erirrhiza from the Bjelolasica Mountain, to the subgenus Iris, series Pumilae, and finally to the series Elatae. 相似文献

We studied nectar characteristics during the long flowering period (late June to end of November) in two populations of Linaria vulgaris (L.) Mill. spontaneously growing in the Botanical Gardens of Siena University (Tuscany, central Italy). The two populations were close to each other but they differed in blooming period. Plants of population 1 sprouted in May and flowered from the end of June to the end of September. Population 2 sprouted at the end of August and flowered from September to the end of November. Differences in nectar production and composition were found between and within populations. Flowers of population 1 produced a very small amount of nectar (not collectable) that remained on the nectary surface. The quantity of nectar increased in late September, when each flower produced 2–3 μl of nectar that flowed into the spur. Total sugar concentration was 175.8 mg/ml in young flowers. Flowers of population 2 produced 5–8 μl of nectar with a total sugar concentration of 200.9 mg/ml in the young stage. In bagged senescent flowers nectar volume decreased in both populations and nectar sugar concentration decreased down to 11.6 mg/ml in population 2 and increased up to 289.6 mg/ml in population 1. For both populations, the decrease in nectar volume in bagged flowers may have been due to water loss by evaporation. In population 2, the decrease in sugar concentration may have been due to nectar reabsorption that was never observed in population 1. Inflorescence and floral ontogeny of the perennial, herbaceous crop Crocus sativus L. were studied using epi-illumination light microscopy. After production of leaves with helical arrangement a determinate inflorescence forms which becomes completely transformed into a single terminal flower. In some cases, bifurcation of the inflorescence meristem yields two or three floral meristems. The order of floral organs initiation is outer tepals – stamens – inner tepals – carpels. Stamens and outer tepals are produced from the lateral bifurcation of three common stamen-tepal primordia. Within each whorl, organs start developing unidirectionally from the adaxial side, except for the stamens which begin to grow from the abaxial side. Specialized features during organ development include interprimordial growth between tepals forming a perianth tube, fusion at the base of stamen filaments, and formation of an inferior ovary with unfused styles. 相似文献
