Two strains of Penicillium digitatum and one strain of Penicillium italicum were exposed to various levels of sorbic acid and potassium sorbate, and the MICs were determined. Selected strains of the molds were then repeatedly exposed to subinhibitory levels of the compounds to determine whether increased tolerance might develop. The MIC of sorbic acid (pH 4.75) to P. digitatum was between 0.02 and 0.025%. The MIC of sorbate (pH 5.5) to two strains of P. digitatum and P. italicum was found to be between 0.06 and 0.08%. Increasing levels of sorbate resulted in increasing growth suppression of the molds. Populations of P. digitatum were tested for increased tolerance to sorbic acid, and none was found. Individual molds that started from the same parent colony were examined for increased tolerance to potassium sorbate. Two P. digitatum strains developed no observable increased tolerance, but P. italicum developed a slight increase in tolerance to sorbate. When spores of P. italicum and P. digitatum were exposed to higher levels of sorbate for prolonged times, the fungicidal or fungistatic activity of the inhibitor was dependent upon pH, length of exposure time, level of sorbate, and the mold strain.  相似文献

The present study investigated the antifungal activity of octanal against Penicillium italicum and P. digitatum. Results showed that octanal exhibited strong antifungal activity against the test pathogens in a dose-dependent manner. Scanning electron microscopy observation revealed that octanal obviously altered the morphology of P. italicum and P. digitatum hyphae by causing the loss of cytoplasm and distortion of mycelia. A rapid increase in the membrane permeability of P. italicum and P. digitatum was observed after treated with octanal at minimum inhibitory concentration or minimum fungicidal concentration, evidenced by the release of cell constituents, the extracellular conductivity and the extracellular potential of hydrogen. In addition, octanal apparently induced a decrease in total lipid contents of P. italicum and P. digitatum cells. These results suggested that the antifungal activity of octanal against P. italicum and P. digitatum can be attributed to the disruption of the cell membrane integrity and the leakage of cell components.  相似文献

Panomycocin, a novel exo-beta 1,3 glucanase, was tested as an antifungal agent against green and blue mold diseases, the most important causes of post harvest decay in citrus fruits. All tested isolates of Penicillium digitatum and Penicillium italicum were susceptible to panomycocin in vitro. Effective panomycocin concentrations for 50% growth inhibition (MIC-2) for P. digitatum and P. italicum were 2 and 1 μg ml−1, respectively. Complete (MIC-0) growth inhibition of all isolates observed at a panomycocin concentration of 16 μg ml−1. Treatment of spores with panomycocin at values lower than the MIC-0 led to slower germ tube elongation and mycelium growth. In tests on fruit, panomycocin at concentrations equal to in vitro MIC-0 value protected lemon fruit from decay.  相似文献

Aims: This study aimed at modelling the effect of ethanol vapours, in the range 0·7–7·5 kPa, on the inactivation of dry‐harvested conidia of Penicillium chrysogenum, Penicillium digitatum and Penicillium italicum. Methods and Results: Survival curves were modelled by a Weibull model: log (N/N0) = ?1/2·303 (t/α)β. The shape parameter β was different from one in all cases, indicating that the classical first‐order kinetics approach is the exception rather than the rule. Survival curves exhibited upward concavity (β < 1) with the notable exception of P. chrysogenum at ethanol vapour pressures 0·7 and 1·5 kPa. The scale parameter α (h) varied greatly depending on the ethanol vapour pressure and on the species. Conclusions: For safety reasons, it is recommended not to exceed an ethanol vapour pressure of 3·3 kPa. At 2·8 kPa, more than 4 log10 reductions in viable conidia were achieved for all the species after 24‐h exposure. Significance and Impact of the Study: Ethanol has GRAS status in the USA and represents an interesting alternative to fungicides. The effectiveness of ethanol vapours to inactivate dry‐harvested conidia of some Penicillium was demonstrated in this study.  相似文献

In order to control postharvest rot of satsuma mandarin (mandarin) fruits, a new strain of Bacillus amyloliquefaciens JBC36 (JBC36) was isolated from rhizosphere of a mandarin orchard and tested for its suppression of decay due to green and blue molds caused by Penicillium digitatum and Penicillium italicum, respectively, and its mode of action was investigated. In addition, carnauba wax-based and paraffin oil-based coating formulations were developed to increase the control efficacy of the antagonist. The strain JBC36 at 108 CFU mL?1 inhibited incidence of green and blue molds on wounded mandarin fruits with control efficacies of 88 and 80.2%, respectively. Mycelial growth and spore germination of P. digitatum and P. italicum were strongly inhibited in the presence of JBC36 or antagonistic metabolites. In order to determine antifungal activity, three kinds of antibiotics were isolated by reverse-phase high performance liquid chromatography (RP-HPLC) and identified as lipopeptide families, iturin A, fengycin and surfactin by RP-HPLC and thin-layer chromatography (TLC) analysis. Fengycin was further identified as C16 fengycin A by liquid chromatography/electrospray ionisation-mass spectrometry (LC/ESI-MS) and mass spectrometry/mass spectrometry (MS/MS) analysis. Volatile organic compounds from the antagonist also reduced the mycelial growth of P. digitatum and P. italicum. Carnauba wax-based and paraffin oil-based coating formulations containing 108 CFU mL?1 of JBC36 efficiently decreased the incidence of green mold with control efficacy of 91 and 80.9%, respectively. Overall, the antagonistic rhizobacterium JBC36 is a promising biocontrol agent for use in preventing postharvest spoilage of mandarin fruits by green and blue molds.  相似文献

Fungal antifungal proteins (AFPs) have attracted attention as novel biofungicides. Their exploitation requires safe and cost-effective producing biofactories. Previously, Penicillium chrysogenum and Penicillium digitatum produced recombinant AFPs with the use of a P. chrysogenum-based expression system that consisted of the paf gene promoter, signal peptide (SP)-pro sequence and terminator. Here, the regulatory elements of the afpA gene encoding the highly produced PeAfpA from Penicillium expansum were developed as an expression system for AFP production through the FungalBraid platform. The afpA cassette was tested to produce PeAfpA and P. digitatum PdAfpB in P. chrysogenum and P. digitatum, and its efficiency was compared to that of the paf cassette. Recombinant PeAfpA production was only achieved using the afpA cassette, being P. chrysogenum a more efficient biofactory than P. digitatum. Conversely, P. chrysogenum only produced PdAfpB under the control of the paf cassette. In P. digitatum, both expression systems allowed PdAfpB production, with the paf cassette resulting in higher protein yields. Interestingly, these results did not correlate with the performance of both promoters in a luciferase reporter system. In conclusion, AFP production is a complex outcome that depends on the regulatory sequences driving afp expression, the fungal biofactory and the AFP sequence.  相似文献

Cultural conditions under which Penicillium digitatum and P. italicum develop were an important factor in determining the extent of their cellulolytic activity. At pH 5.5–7.5 of the culture medium, Cx-cellulase activity was correlated with mycelial dry weight. However, at pH 4.5 and more so at pH 3.5, activity was markedly reduced while fungal growth was not affected. Cx-cellulases of both species were not induced by the presence of carboxymethyl cellulose as a carbon source and were defined as constituent enzymes. Cellulase activity of the two Penicillia on different carbon sources was detected prior to the initiation of sporulation. A sporeless mutant of P. digitatum exhibited cellulolytic activity similar to that of the normal strain, suggesting no role for sporulation in the Cx-cellulase synthesis. Cx-cellulase activity in Valencia oranges started during the early stages of pathogenesis, before the appearance of disease symptoms. Correlation between the cellulase activity and the severity of the disease symptoms was apparent during the first three days after inoculation. At the end of the incubation period both fungi almost reached their maximum enzymatic activity, whereas the disease index continued to rise gradually until total fruit rot was achieved. A possible role of Cx-cellulases of the two Penicillia in the early stages of pathogenesis was suggested.  相似文献

Water extracts of rind, essential oil and juice from oranges, also citrus pectin and citric acid promoted the formation of lesions when spores of Penicillium digitatum were placed in wounds 1·0 mm deep in flavedo of oranges; fructose, glucose and sucrose had little effect. Rind extracts were less effective in wounds 0·5 mm deep but orange juice and pectin still increased infection. None of the substances allowed the parasite to infect fruit through unwounded surfaces. Germination of spores in water increased as spore concentration decreased but was poor even at low concentrations. Almost all spores germinated in aqueous extracts of flavedo, albedo or whole rind, or in wounds on the surface of fruit. Fructose, glucose, sucrose and xylose were less effective but still caused over two-thirds of spores to germinate but only in the presence of phosphate buffer. Without buffer, germination was little different from that in water. Arabinose and galactose stimulated germination to a lesser extent but with the same phosphate effect. Carboxymethylcellulose and pectin did not affect germination. A variety of substances containing nitrogen increased germination but to different degrees, decreasing in the order, casamino acids, yeast extract, ammonium salts, nitrate. Thiamin and to a lesser extent biotin were also effective. Volatile substances from rind infected with P. digitatum stimulated spore germination and growth of germ tubes. The significance of these results is discussed in relation to infection.  相似文献

In laboratory trials, fumigation with acetaldehyde vapour at 0–75 to 20 % concentration (v/v) for 0–5 to 120 min at 21 oC killed six species of post-harvest citrus pathogens grown on agar media. The fungicidal effect of acetaldehyde vapour at this temperature was a function of concentration and exposure. Penicillium digitatum and P. italicum were not as susceptible to inhibition of spore germination as Alternaria citri, A. tenuis, Colletotrichum gloeosporioides and Glomerella cingulata. A concentration of 10 % acetaldehyde vapour at 21 oC for 10 min was fungitoxic to all the pathogens tested.  相似文献

A new antagonist/pathogen combination is reported, with Bacillus pumilus showing strong antagonistic activity against Penicillium digitatum. B. pumilus (1.6 × 1010 to 1.6 × 1012*cfu ml-1) gave significant control of P. digitatum infection in Valencia orange, which was as effective as imazalil (500 μg ml-1) and was significantly better than benomyl treatment (500 μg ml-1). When lower concentrations of B. pumilus (1.9 × 107 to 1.9 × 109 cfu ml-1) were tested on Washington Navel orange and Lisbon lemon fruit, the antagonist caused significant control of P. digitatum infection at two inoculum levels (6.5 × 104 and 6.5 × 105 spores ml-5). Concentrations of both the pathogen and the antagonist affected the biocontrol effect.  相似文献

Trichoderma viride spores applied in water to apparently uninjured skin of oranges do not cause lesions. Adding orange juice, rind extract, citric acid or orange essential oil did not influence infection. Oranges became infected only when the stem-end cuts or wounds deeper than 6 mm into oil vesicles were inoculated. Sound oranges in contact with decayed oranges did not become infected. Diphenyl-impregnated wrappers reduced infection. A mixed inoculum of T. viride and Penicillium digitatum caused as fast rotting as P. digitatum, which caused faster rotting than T. viride alone. Lesions infected with P. digitatum could become infected by T. viride but those caused by T. viride did not become infected by P. digitatum. T. viride was antagonistic to P. digitatum in vivo and in vitro, possibly because it produces a heat-labile diffusible substance toxic to P. digitatum.  相似文献

【目的】采后柑橘极易受指状青霉(Penicillium digitatum)侵染而发生严重的绿霉病腐烂,生物防治因具有安全、有效、环保等特点近年来备受关注。论文旨在研究荧光假单胞菌(Pseudomonas fluorescens)ZX对采后柑橘绿霉病的防治效果,揭示P.fluorescensZX对P.digitatum可能存在的作用机制。【方法】以"北碚447"锦橙果实为试材,先分别接种20μL拮抗菌培养液、滤液(培养液离心后,上清经0.22μm滤膜过滤)、菌悬液(培养液离心后,菌体用无菌水反复洗涤并用无菌水重悬)和热杀死液(培养液高温高压灭菌),2 h后接种20μL P. digitatum孢子悬浮液(1×10~4spores/m L),所有果实于20oC、90%相对湿度环境下恒温恒湿培养8 d后,测定果实的发病率和病斑直径;制备柑橘皮培养基,进行平板抑菌试验,探索P. fluorescens ZX对P. digitatum孢子发芽情况的影响;采用两板对扣法和生物熏蒸法研究P.fluorescensZX挥发性次级代谢产物的抑菌作用;利用插入式细胞培养皿等分析P.fluorescensZX和P.digitatum之间竞争的营养物质;同时,测定P.fluorescensZX的生长曲线,利用结晶紫染色法评估P. fluorescens ZX的生物膜形成能力。【结果】P. fluorescens ZX不同处理液之间对采后锦橙绿霉病的作用效果差异显著,菌悬液抑菌效果最好,经菌悬液处理的果实,发病率和病斑直径分别仅为40.83%和1.78 cm;不论是在柑橘皮固体培养基上对峙培养还是在液体培养基中混合培养,菌悬液和原液的作用效果较好,固体平板上,相对抑制率达到了35%–45%,液体培养基中,P. digitatum孢子12 h后的发芽率不超过27%;P. fluorescens ZX产生的挥发性物质具有抑菌作用,经P. fluorescensZX熏蒸处理的锦橙果实,发病率和病斑直径都显著降低;营养竞争试验结果表明,P. fluorescens ZX能更快速有效地消耗柑橘皮培养基中的营养,并和P. digitatum竞争葡萄糖、果糖、蔗糖、天冬氨酸、苏氨酸、丝氨酸、亮氨酸、精氨酸和脯氨酸等营养物质;同时,P. fluorescens ZX生命力强,培养4 h后即进入对数生长期,约24 h后形成成熟的生物膜。【结论】P. fluorescens ZX可能通过抑制P. digitatum孢子发芽、营养与空间竞争、形成生物膜、产生抑菌物质等方式抑制P.digitatum的生长繁殖,有效防治采后锦橙绿霉病。  相似文献

The fungus Penicillium digitatum, the causal agent of green mould rot, is the most destructive post‐harvest pathogen of citrus fruit in Mediterranean regions. In order to identify P. digitatum genes up‐regulated during the infection of oranges that may constitute putative virulence factors, we followed a polymerase chain reaction (PCR)‐based suppression subtractive hybridization and cDNA macroarray hybridization approach. The origin of expressed sequence tags (ESTs) was determined by comparison against the available genome sequences of both organisms. Genes coding for fungal proteases and plant cell wall‐degrading enzymes represent the largest categories in the subtracted cDNA library. Northern blot analysis of a selection of P. digitatum genes, including those coding for proteases, cell wall‐related enzymes, redox homoeostasis and detoxification processes, confirmed their up‐regulation at varying time points during the infection process. Agrobacterium tumefaciens‐mediated transformation was used to generate knockout mutants for two genes encoding a pectin lyase (Pnl1) and a naphthalene dioxygenase (Ndo1). Two independent P. digitatum Δndo1 mutants were as virulent as the wild‐type. However, the two Δpnl1 mutants analysed were less virulent than the parental strain or an ectopic transformant. Together, these results provide a significant advance in our understanding of the putative determinants of the virulence mechanisms of P. digitatum.  相似文献

Summary Copper adsorption by Rhizopus arrhizus, Cladosporium resinae and Penicillium italicum was studied using a copper-selective electrode. Copper adsorption by C. resinae and P. italicum obeyed the Freundlich and Langmuir isotherms for single-layer adsorption whereas R. arrhizus followed the BET isotherm for multi-layer adsorption. Temperature had little effect on adsorption over the range 4–25°C. Mineral acids were effective for desorption of copper from preloaded biomass, the efficiency of desorption increasing with decreasing pH. Other cations were also capable of copper desorption with zinc showing the greatest efficiency and sodium the lowest.  相似文献

Different liquid formulations of anise, coriander and black cumin essential oils were used for preparing some biocides. The prepared formulations were tested for their antimicrobial activity against some post-harvest pathogenic microorganisms. The tested microorganisms were Fusarium oxysporum (Dray rot of potato), Alternaria alternata (Black rot of tomato), Penicillium italicum, P. digitatum, (Blue and green rot of orange, respectively), Botryitus cinerea (Gray rot of strawberry) and Erwinia carotovora (Soft rot of potato).The results revealed that the different formulations showed a complete inhibition effect on the growth of most of the tested microorganisms. Also, the antimicrobial activity of the formulated essential oils did not affect the formulation process compared with the original oil.  相似文献

Abstract

On the presence of « hyphycellariola » (swollen celles) and conidia in three lichenized fungi cultivated « in vitro ». — The Authors refer to the presence of « cellariola » (and « hyphycellariola ») found in pure cultures « in vitro » of the mycobiont of Xanthoria parietina and describe the morphology of conidia in the fungus isolated from Sarcogyne similis and Acarospora fuscata.

As far as the « cellariola » and « hyphycellariola » are concerned, it is the first note relative to the mycobionts of Lichens.

The Authors report the data of the utilized medium and the morphological observations.

Green mold, caused by Penicillium digitatum, is the most destructive post-harvest disease in citrus. Secondary metabolites produced by fungal phytopathogens have been associated with toxicity to their respective host through the interaction with a wide range of cell targets. Natural products have also been described as important molecules for biocontrol and competition in their respective environment. For P. digitatum, the production of indole alkaloids, tryptoquialanines A and B, have been reported. However, their biological role remains unknown. Mass Spectrometry Imaging (MSI) technique was applied here for the first time to monitor the secondary metabolites produced on the orange surface during infection in order to gain insights about the P. digitatum-citrus interaction mechanisms. Through the combination of MSI and molecular networking it was possible to report, for the first time, the production of tryptoquivalines and fumiquinazolines by P. digitatum and also the accumulation of tryptoquialanines on the fruit surface from 4 to 7 d post inoculation. P. digitatum was also evaluated concerning the ability to sinthesize indole alkaloids in vivo in the different citrus hosts. The biological role of tryptoquialanines was investigated and tryptoquialanine A was submitted to insecticidal bioassays that revealed its high toxicity against Aedes Aegypti, suggesting an important insecticidal action during orange decay.  相似文献

Summary

The Author observing the great disagreement of the terminology usually adopted for the apomictic phenomena, examines the whole series of the phenomena noted both in regards of the formation of the gametophyte and of the embryo. The forming of the gametophyte is fixed on the base of nature of its original cell, normally a haploid spore or »haplospore« (phenomenon of the »haplospory« following the regular meiosis), rarely a diploid spore or »diplospore« (phenomenon of the »diplospory« following the irregular meiosis, as for instance in the case of restitution nucleus, etc.). The original cell can be at least represented by the same cell that usually is the spore mother cell, whether for the outward causes, or for inward causes is lacking the usual reductional process. For the failure of the meiotic process, such a cell cannot be called »spore«.

A cell of the somatic tissue of the sporophyte can even become the original cell of the gametophyte always in case of the absence of the meiosis. These two last cases of apospory are respectively called: »somatic apospory« and »gonial apospory« (= germinal apospory) (Chiarugi 1926).

The formation of the embryo is fixed on the base of the nature of the fecundation. The principal phenomena of the fecundation are represented by the following cases: »amphigamy« (or eugamy, or gamy), »semigamy, »pseudogamy«, »apogamy« »parthenogenesis«. The A. uses the term »apogamy« both for the diploid eggs and for the haploid eggs in which the stimulation of the division is produced by the pollen-tube or by male nuclei, in absence of the gamy.

The term »parthenogenesis« is restricted to the division of the haploid or diploid eggs in which the stimulation is not caused by pollen-tube nor even for the great reason, by the male nuclei existing in it.  相似文献

Green and blue mold postharvest diseases are the most vital negative components influencing the local market of citrus fruits. Citrus fruits were collected, and fungi were isolated. Among the fungal isolates identified, Penicillium digitatum and Penicillium italicum recorded the highest occurrence of 39.5 and 25.6%, respectively. In this work, we extracted chitosan from Aspergillum niger ATCC 9642. Fourier transform infrared spectroscopy was utilized to confirm the functional groups of the obtained compound, which exhibited the main characteristic bands of O–H stretching at 3302 cm-1, and C–O–C band at 1125 cm-1. A. niger ATCC 9642 chitosan had the degree of deacetylation of 88.5%, a molecular weight of 1.8 × 105 Da, and viscosity of 7.3 centipoises; these values were comparable to those for standard shrimp chitosan. Ultraviolet-visible light spectra revealed the presence of A. niger ATCC 9642 chitosan-AgNPs composite. Using antifungal and spore germination assays, it was found that this composite exhibited effective antifungal action against P. digitatum and P. italicum compared with a chitosan standard. In a comet assay, the percentage of tail DNA was considered as a parameter that indicated DNA damage. The comet parameter increased significantly (P < 0.05) with A. niger ATCC 9642 chitosan–AgNPs composite, and the increase was dose-dependent. The increase in the DNA damage positively correlated with the inhibition performance of the A. niger ATCC 9642 chitosan–AgNPs composite.  相似文献

Microorganisms are increasingly exploited as a source of new biological control agents. Genus Penicillium is a source of novel bioactive molecules which can be used as antifungal agents. The objective of this study was to evaluate the antifungal potential of Penicillium strains. Culture filtrates of two Penicillium species were tested for their antifungal potential by well diffusion assays. Filtrate of Penicillium isolates showed high antifungal effects on mycelial growth of Fusarium oxysporum, Fusarium solani, Macrophomina phaseolina, Aspergillus japonicus var aculeatus and Cladosporium cladosporioides. But Penicillium italicum inhibit the fungal growth from 45 to 68% as compared to Penicillium simplissimum (25–68%). However in case of A. japonicus var aculeatus, Penicillium spp. extracts were equally effective and reduce the colony growth up to 68%. However, P. simplissimum extract was least effective in case of M. phaseolina, where it decreased the colony growth only 25%.  相似文献
