Comparative observations of Bipolaris sorokiniana and Curvularia geniculata conidia germination as influenced by culture age and temperature showed some distinct differences, but generally established the ability of these organisms to function under similar conditions. Total germination of B. sorokiniana conidia was favored by increasing culture age from 20 to 60 days and temperature to 25°C; total conidia germination of C. geniculata was favored by increasing temperature to 25°C, but increasing culture age decreased germination. These reactions seem associated with conidia age. Maximum proportional intra-population germination of conidia of each organism also varied with culture age and temperature. At temperatures of 5°C and 15°C, amplitude of maximum proportional germination of both organisms increased as culture age was increased from 20 to 40 days and then decreased among 60-day-old cultures. At 25°C and above, amplitude of maximum proportional germination of conidia of both organisms decreased from each older culture. Progressively increasing temperature at a given culture age increased the amplitude of maximum proportional germination up to 25°C for conidia of B. sorokiniana, but generally decreased it for conidia of C. geniculata (except 20-day-old cultures). Frequency (specific 2 h interval) of maximum proportional intrapopulation germination of B. sorokiniana shifted from 6 h to 2 h in response to increasing temperature and culture age; conidia from youngest cultures of C. geniculata shifted to intervals of 4 h and 2 h in response to increasing temperature to 25°C, but among conidia from 60-day-old cultures, frequency shifted to 6 h intervals at all temperatures. Above 25°C, maximum proportional germination of C. geniculata conidia from cultures of all ages occurred at 6 h. It was concluded that the germination response of B. sorokiniana and C. geniculata conidia to temperature and culture age (and, subsequently, conidia age) are enough similar that these organisms could function in a potential ‘disease complex’ on Poa pratensis and Agrostis palustris.  相似文献

In order to assess fluid domains in the genome of Dasypyrum villosum, Feulgen/DNA cytophotometric determinations and molecular and cytological DNA-DNA hybridization experiments were carried out in resting embryos and developing seedlings from yellow and brown caryopses belonging to different populations. The cytophotometric data showed that the basic amount of nuclear DNA is, on average, 12% higher in 2-day-old seedlings from yellow caryopses as compared to those from brown caryopses. It increases in each individual during seed germination, to a higher extent in seedlings from yellow caryopses than in those from brown caryopses. DNA content also differs up to 13% between plants within a caryopsis-colour group and up to 40% between populations. Dot-blot hybridization of a 396-bp D. villosum-specific DNA repeat to genomic DNA extracted from embryos in dry seeds, or from seedlings belonging to single progenies of plants from different populations, confirmed the cytophotometric results. The redundancy in the genome of sequences hybridizing to the 396-bp element differs significantly both between populations and between plant progenies within a population. During seed germination these sequences are the more amplified the less they are redundant in the genome of resting embryos, and amplification occurs to a significantly-greater extent in seedlings from yellow caryopses than in those from brown caryopses. 3H-labelled 396-bp sequences hybridize at or near the telomeres of most chromsome pairs though only to the shorter of the two subtelocentric pairs. The hybridization level is higher in seedlings from yellow caryopses that in those from brown caryopses, and a linear correlation exists between the number of silver grains counted over the labelled regions of each chromosome pair in the two groups of seedlings. Possible control mechanisms of the observed changes in the nuclear genome, and the role of these changes in developmental pregulation and environmental adaptation, are discussed.  相似文献

Abstract

The geomagnetic field effect hypothesis and the variability among replications in germination and growth tests in Triticum. – During previous eperiments on germination and growth of Triticum, we have often found many cases of heteroscedasticity and of statistically significant differences among contemporary replications (i.e. germinators). In such experiments all the caryopses were oriented in the same way inside the germinators, whilst the germinators were randomly set inside the thermostats: therefore the seeds of different germinators might result differently oriented. Some researchers report that seeds differently oriented according to the lines of force of the geomagnetic field, may respond with different germination, growth rates and growth directions of shoots and roots (magnetotropism). The present investigation was designed to ascertain if these effects might be responsible of the variability among the contemporary replications. The different caryopses orientation within laboratory geomagnetic field, does not give evidence of any effect on germination, growth rate and direction. The caryopses show no germination sensibility neither related to different age, light and temperature conditions, neither to different cultivars, neither to varying orientation from the geomagnetic North every 15° and to varying time of ? preorientation ? in dry conditions. Differences between the variability of randomly and North oriented subsamples are not observed. No statistically significant differences are observed among shoots and roots lenghts of seedlings from differently oriented caryopses neither in different environmental conditions, nor as function of the orientation (24 angles of 15°), nor after different preorientations, nor after repeating the test on 608 subsamples (12.000 seedlings). Therefore the variability among contemporary replications is not imputable to the geomagnetic field. The variability and the distributions structure analysis suggest that both the heteroscedasticity and the significant differences among the replications may be due to an interaction among the caryopses inside the germinators through the substrate. This is probably related to the cultivar characteristics. We emphasize that an accurate evaluation of the variability is particularly necessary in germination and growth tests.  相似文献

Kummerow, Jochen. (U. Chile, Santiago.) Endogenous fluctuations of germination capacity in Dactylis glomerata. Amer. Jour. Bot. 50(9): 915–920. Illus. 1963.—Dactylis glomerata caryopses, ‘New Zealand’ strain, furnished by the Ministerio de Agricultura of Chile, were enclosed in test tubes, carefully sealed with a Bunsen flame, and stored under constant conditions. Their germination capacity was periodically determined. In 1958/1959, a 2-peak annual germination curve was obtained. The germination increase was related with a diminution of the water content of the caryopses. The same material was seeded in 8 different zones of the Chilean Central Valley between 28°34′ and 40°35′ S. lat. The same germination procedures were practiced with the caryopses harvested in these 8 parcels. In the 1960/1962 period, 1-peak germination curves were found. We did not find the same relation between water content and germination as found in 1958/ 1959. There exists a certain relation between the velocity of germination and caryopsis origin. Abstract

Germination and spontaneous chromosome mutations in Triticum durum Desf. during the first two years of storage of the seed. — Spontaneous chromosome aberrations were recently observed in the meristem of radicles of seedlings raised from seeds (caryopses) of Triticum durum cv. Cappelli erop 1966. Germination and frequency of chromosomal aberrations, starting from the ripening of the caryopses to the end of the second year of their storage at room temperature were investigated. Cytological analysis and germination tests were accomplished at monthly intervals. It was shown that: 1) soon after ripening, the germination process was very slow, but chromosomal damage was high; 2) at the end of the dormancy period the percentage of aberrant anaphases and of chromatid breaks fell to a «minimum». This low chromosomal damage lasted for almost the whole of the two years of storage; 3) the nuclear damage was esclusively of chromatide type. This result and fact that the cytological damage is decreasing with time, are interpreted as evidence that the mutagenic agent(s) responsible is (are) more active during the germination period than during the quiescence of the embryo.  相似文献

Abstract

Five populations belonging to three subspecies of Dianthus rupicola Biv. (D. rupicola subsp. rupicola, D. rupicola subsp. aeolicus, and D. rupicola subsp. lopadusanus) and growing in different geographical areas of Sicily were tested for seed germination at various temperatures. All populations showed high germination rates with an optimum temperature between 15°C and 25°C. Efficient seed germination might contribute significantly to the preservation of these subspecies which are currently exposed to several environmental threats.  相似文献

Abstract

The effects of temperature and salinity (NaCl) on germination of Hordeum maritimum With, (halophyte) and H. murinum L. (glycophyte) seeds were investigated. Dehulled caryopses were used for monthly germination trials, starting from November (120 days of after‐ripening in darkness at 20±1°C). Trials were continued for one year. Differences in germination response between the two species were observed, confirming that H. Maritimum is better adapted to high salinity levels and to variations in external temperature than H. murinum. H. maritimum showed a germination control mechanism related to after‐ripening time and based on seed dormancy break/resumption. At higher temperature (30°C), thermodormancy was also recorded. No germination strategies were observed in H. murinum that is relatively insensitive to the combined effects of temperature and salinity. Thus, in virtually all treatments, H. murinum exhibited a higher germination rate compared with H. maritimum, as early as 72 h after imbibition, suggesting that dormancy, both in the presence or absence of salt, is totally abolished by early fall or at the latest in winter.  相似文献

Dormant, intact Avena fatua L. (wild oat) seeds germinate poorly at 20 °C. Removing the hulls slightly increased germination. Treatment with smoke solutions increased the germination of both intact seeds and caryopses. Exogenous GA3, alone or in the presence of smoke solution, increased the germination of caryopses, while ACC shows a tendency to increase germination of caryopses only when applied in combination with smoke solution. Results suggest that GA3 and ethylene, but not smoke solutions, are involved in the regulation of α-amylase activity during germination. However, the participation of smoke solutions in the control of ACC oxidase activity cannot be excluded.  相似文献

  • Seed germination of different populations of Aegilops geniculata Roth collected along a latitudinal gradient was studied under different drought stress in order to find populations suitable for improving drought tolerance in wheat. Different accessions of Aegilops neglecta Req. ex Bertol., Triticum aestivum L. and T. durum Desf. were used as comparison.
  • Under full hydration, germination was high in all populations, but increasing drought stress led to reduced and delayed germination. Significant differences in final germination and mean time to germinate were detected among populations. Wheat, durum wheat and the southern population of Ae. geniculata were not significantly affected by drought stress, germinating similarly under all treatments. However, seed germination of the northern populations of Ae. geniculata was significantly reduced under high water stress treatment. Differences between populations of the same species could not be explained by annual rainfall across populations’ distributions, but by rainfall during seed development and maturation.
  • Differences in the germination responses to drought found here highlight the importance of source populations as criteria for genotype selection for pre‐breeders.

The untwisting movement of the hygroscopic awn of wild oats (Avena fatua L.) causes the caryopses to rotate in anticlockwise direction and to dislodge from the spikelet. While untwisting, the awn generates more torque than is required to separate the caryopsis. The periodic shattering of caryopses caused by awn movement may be biologically significant in that the chances of having favorable moisture conditions for caryopses germination in the field at the time of dispersal are increased. Abstract

The autecology of the Sardinian endemics Aquilegia barbaricina Arrigoni et Nardi and A. nugorensis Arrigoni et Nardi were investigated. Peaks of anthesis and seed dispersal were recorded for five populations occurring in two distinct habitats, one riparian and one rupicolous. Germination tests were carried out on seed lots belonging to each population by sowing seeds at 10, 15, 20, 25 and 25/15°C. In addition, seeds were incubated for 2 months at either 25°C (summer), 5°C (winter) or 25°C for 2 months plus 2 months at 5°C (summer followed by winter–SW), and then moved to the germination temperatures. Embryo measurements were taken during pre-treatments and germination. Experimental seed burials were carried out for two populations of each species. Both species dispersed in summer. The population of A. nugorensis occurring on rocky outcrops differed in phenology from both the other A. nugorensis population from riparian vegetation and from A. barbaricina. Both species showed morphophysiological seed dormancy, with <50% germination under laboratory conditions. All riparian populations germinated only after the SW pre-treatment, while the rupicolous population germinated at 25°C, without any pre-treatment. Low germination percentages were observed in the experimental seed burials, suggesting the ability for both species to form a persistent soil seed bank.  相似文献

  • Even though exhaustively studied, dormancy alleviation in diaspores of Urochloa humidicola (Rendle) Morrone & Zuloaga remains a mystery. To clarify this, we asked the following question: could dormancy alleviation in diaspores of this species be associated with ageing, GA/ABA balance and remaining structures of the panicoid spikelet?
  • We answer this question using diaspores of U. humidicola cv. BRS Tupi as a biological model, a cultivar with a wide edaphoclimatic range in Neotropical areas and whose diaspores possess ‘deep dormancy’ when dispersed. We analysed both germination and early plant development using a split‐plot model.
  • Our findings demonstrate that dormancy alleviation in diaspores of U. humidicola is a synergic phenomenon driven by crosstalk between age, GA/ABA balance and remaining structures of the panicoid spikelet covering caryopses, since this interaction acts on the dynamics of germination and early plant development.
  • We demonstrate that: (i) spreading germination time is a maternal survival mechanism of this species, which has repercussions for occupational aggressiveness of the species; (ii) remaining structures of the panicoid spikelet covering caryopses are the main modulator of embryo development. These structures control the after‐ripening process, which is modulated by some molecular factor. We also highlight that it is necessary to review concepts about dormancy of dispersal units in this grass species.

Abstract

Limonium mansanetianum is catalogued as critically threatened (CR) species and it is included in Valencian Catalogue of Threatened Plant Species. Limonium mansanetianum is a gypsicolous species, which only lives in a restricted area to south-centre of Valencia province (Spain). The species is a low-branched woody shrub with summer flowering. The influence of incubation temperature (10°, 15°, 20° and 25°/20?°C) and salinity (0%–3.0% NaCl) on seed germination of L. mansanetianum was studied. Best seed germination was obtained in distilled water controls. Seed germination decreased with an increase in salinity and few seeds germinated at 2.5% and 3.0% NaCl. Optimal temperature regime for germination was 15?°C where germination in 0.5% and 1.0% NaCl was not affected. Recovery and hypersaline conditions experiments showed that L. mansanetianum seeds displayed a greater tolerance to high salinity and temperature stress before germination.  相似文献

Abstract

The germination of spring and winter wheat lines of exaploid Triticum « Denti de Cani ». — The dormancy in the seeds of two lines of Triticum « Denti de Cani » (which is spontaneous in Sardinia), one with solid stem (CP line), a spring line, the other with hollow stem (CV line), an winter line, has been studied. Germination was carried out in the dark, in Petri dishes at the constant temperatures of 5°, 10°, 20°, 23°, 26°, 30° and 35°C, using full ripe seeds, and seeds in different stages of after-ripening up to one year of age. The increase in % germination, for increasing temperatures above 5°C, is clearly conditioned by the progress of after-ripening in the seeds. In fact it was seen that, in general for the two lines, percentages over 50% of seeds germinated at 3 days were reached: at 10° and 20° after 15 days from the full ripening; at 23°C after 30 days; at 26°C after 50 days; at 30°C after about 100 days and at 35°C only after about 4–5 months from the harvest. During the experiment at 5°C it was observed that, during the first year of life of seeds and especially in the CP line, this temperature produces a clear slowing down in germinations after first year from the ripening, only the CV seeds — not the CP which remain very much inhibited — reach germination values over 50% at 3 days. It has also been demonstrated that the CV are more sensitive than the CP, in the first initial period of after-ripening (15 and 30 days), to the non-inhibiting activity of low temperatures (5° and 10°C) and that, between these, the 10°C temperature promotes the germination more clearly than the 5°C temperature. The results obtained have shown that the dormancy wears off in the spring CP-line much more slowly than in the winter CV-line. The CP-seeds remain in a relative dormancy condition for a long time, which causes a significative delay in germination, up to 100 days from the full ripening stage.  相似文献

Background: Fire is an important ecological factor in the Cerrado (Brazilian savanna). However, comparative studies on the effect of high temperatures experienced during fires on seed germination of native and invasive grass species are few.

Aims: To assess germination responses to simulated fire temperatures by seeds of invasive and native Cerrado grasses.

Methods: Heat-shock treatments (50 °C, 70 °C, 90 °C, 110 °C, 130 °C or 150 °C) were applied to seeds of 10 species of native and invasive grasses. For each temperature, the seeds were heated in a dry-air flow for 2 or 5 min. This combination of temperatures and exposure times simulated the soil conditions during typical Cerrado fires.

Results: Temperature treatment was significantly related to germination, and the effect varied according to species. Heat shock did not increase germination in either the native or the invasive species. Exposure time was important for only two species, and four species showed a significant increase in mean germination time.

Conclusions: Species showed different tolerances to high temperatures. It was not possible to differentiate the native and invasive grasses only by their tolerance to high temperatures, suggesting that fire alone may not be an efficient management tool to control the invasive species studied here.  相似文献

Avena fatua L. florets (caryopses enclosed by lemma and palea) were partially dormant at 10–20 °C and did not germinate at temperatures outside this range. After-ripening florets at 25 °C for 12 weeks completely removed dormancy. Caryopses (florets without lemma and palea) were able to germinate totally at 20 °C. Karrikinolide (KAR1) and gibberellic acid (GA3) applied at 10–25 °C partially or markedly induced germination of dormant florets and caryopses, respectively. Both florets and caryopses were more sensitive to KAR1 than to GA3. To obtain similar effects, 1,000 to 10,000 times lower concentrations of KAR1 than GA3 were required. After-ripening with time gradually increased sensitivity of caryopses to these regulators. Likewise, after-ripened, non-dormant caryopses were sensitive to KAR1 and GA3. Inhibitors of gibberellin biosynthesis, ancymidol, paclobutrazol and flurprimidol inhibited the effect of KAR1. This inhibition was reversed by GA3. Caryopses pre-incubated in water with ancymidol or paclobutrazol in the presence or absence of KAR1 germinated completely but with different rates after transfer to GA3. KAR1 probably requires gibberellin biosynthesis to stimulate germination of dormant Avena fatua L. caryopses. Both KAR1 and GA3 increased α-amylase, β-amylase and dehydrogenases activities during imbibition before visible germination occurred.  相似文献

Abstract

Swertia chirayita, a critically endangered medicinal herb, is being over-harvested in the wild. Understanding seed germination is a pre-requisite to ensure species conservation. The germination of seeds collected from six microhabitats was studied at 20°C, 25°C, and 30°C, both under a 14/10 h light/dark photoperiod and in continuous darkness. Two-way ANOVA indicated that microhabitat and temperature significantly affect seed germination, germination rate, germination recovery (GR), and GR rate. Overall, the seeds collected from under canopy showed a significantly (p < 0.05) higher germination than those from open habitats, at 20°C, 25°C, and 30°C (14/10 h light/dark photoperiod). Germination was negligible in continuous darkness but after transfer to a 14/10 h light/dark photoperiod, the seeds from under canopy significantly recovered at 20°C and at 25°C (p < 0.05), and showed the highest germination percentage compared to seeds collected from tree base, stump base, shrubberies, and grassy slope. Similarly, at 30°C, seeds from under canopy recorded the highest GR percentage. In general, seed germination, mean germination rate, seed GR, and GR rate were significantly greater (p < 0.05) at 25°C. Among the microhabitats tested, variation in GR rate was significant (p < 0.05). Seeds were confirmed to be positively photoblastic.  相似文献

Abstract

Effects of temperature and sea water on germination behaviour of Althenia filiformis Petit seeds. - Germination capacity and energy of Althenia filiformis Petit seeds have been investigated, 90 and 180 days after ripening, to carry out a preliminary study on ecology of this species.

This species, halophite and hydrophyte, is spreaded along the coast shores of middle-west mediterranean sea and atlantic shores of Morocco, Spain, Portugal and France.

Seeds were soaked in the dark, at 10°, 20°, 30°C, in solutions at different salt concentration: sea water; sea water diluted in deionized water at ratios (v/v) 1: 2, 1: 4, 1: 8; sea water plus 26 gr/l NaCl; deionized water, as control.

The experimental results show that germination is reduced and delayed when seeds are soaked in progressively concentrated salt solutions; in sea water plus 26 gr/l NaCl seed germination is inhibited.

Seeds pretreated by soaking at 3°C for 10 days in sea water diluted (1:1) by deionized water did not show, when soaked in salt solution at weak and middle concentration, any delay in germination in comparison with unpretreated seeds. On the contrary, pretreated and unpretreated seeds sown in sea water at 30°C had shown, 180 days after ripening, a significant depression in germination values as compared with seeds sown at 20°C.  相似文献
