The effect of copper and zinc ions on sulphur oxidation by Acidithiobacillus thiooxidans, strain SFR01, isolated from anaerobic sewage sludge was assessed, resulting in tolerance levels up to 20 and 200 mmol l–1 for copper and zinc, respectively. The tolerance levels obtained were higher than the concentration of copper and zinc usually found in the collected sewage sludge. The tolerance levels obtained indicate no constraints for sludge bioleaching of those metals due to their toxicities to the indigenous A. thiooxidans.  相似文献

Tests with rainbow trout in mixtures of ammonium chloride with copper sulphate and of phenol with zinc sulphate have shown that the threshold of toxic concentration for a 50% mortality occurs in solutions for which a value of I is obtained by summing the concentrations of the individual poisons expressed as fractions of their individual threshold concentrations. With ammonia-copper mixtures this method of predicting the threshold concentration becomes progressively less adequate as lower percentage mortalities are considered.  相似文献

In experiments with rapeseed (Brassica napus L., cv. Westar) plants, it was confirmed that copper was considerably more toxic than zinc. The toxic effects of 50 and 150 μM CuSO4 were comparable to those of 1000 and 2500 μM ZnSO4. The analysis of the effects of these concentrations of copper and zinc on photosynthetic pigment contents and on the rate of lipid peroxidation did not reveal any reasons for different toxicities of these heavy metals (HM). Among biological effects studied, significant differences were found in the organ distribution of these metals in plants grown on both the standard medium and the medium with high concentrations of copper or zinc. Copper retained in the roots in relatively small amounts and was poorly transported over the aboveground part of the plants. It stayed mainly in the lower leaves, and its distribution changed only a little during the recovery of plants following the HM treatment. In contrast, zinc proved to be highly mobile, it was concentrated in the upper leaves and actively transported when the plants were transferred to a medium with the optimal HM concentrations. High copper concentrations slowed strongly zinc uptake by the roots but practically did not change its movement over the plant. In contrast, high zinc concentrations facilitated copper uptake by the roots but reduced its transfer to the aboveground organs. The data presented here allow us to hypothesize that biological peculiarities of organ and cellular distribution of copper and zinc in plants and interaction of these HM play an important role in the toxic effects of high concentrations of these HM and the mechanisms of adaptation to them at industrial environmental pollution used by rapeseed plants.  相似文献

Effects of mixtures of chloride salts of cadmium, copper and zinc on survival, whole body residues, and histopathology of mummichog, Fundulus heteroclitus (L.), were investigated in synthetic sea water at 20‰ salinity and 20°C. Mixtures of Cu2+ and Zn2+ as indicated by 96 h bioassay studies produced more deaths than expected on the basis of toxicities of individual components. Concentrations of Cd2+ not ordinarily lethal exerted a negative effect on survival of fish intoxicated by salts of copper, zinc, or both.
Atomic absorption determinations of Cd, Cu, and Zn residues in mummichog which survived 96 h exposures to each of these toxicants provided useful indices of total body burdens for these metals. Residues from survivors held in mixtures, especially Cd2+ and Zn2+ mixtures, did not conform to patterns observed for single elements. Whole body aggregates of Cd, Cu, and Zn from dead mummichogs were of limited worth owing to possible accumulation of these metals from the medium after death.
Renal and lateral line canal lesions were noted in all fish subjected to copper concentrations of 1 mg/1 and higher. Renal lesions observed in fish immersed in mixtures of Cu2+ and Cd2+ assumed a damage pattern characteristic of Cd2+; with mixtures of Cu2+ and Zn2+, lesion were typical of Cu2+-induced damage. Lesions induced in lateral line epithelium by Cu2+ were not affected by either Cd2+ or Zn2+. Epithelia lining the oral cavity were necrotized by the caustic action of high levels of Zn2+ (60 mg/1) and of Cu2+ (8 mg/1).  相似文献

In the present study, the serum and hair levels of zinc, selenium, and copper were determined in children with iron-deficiency anemia (IDA). A total of 52 anemic children aged 1–4 yr constituted the study group. Fortysix healthy children acted as controls. The copper and zinc levels were measured with an atomic absorption spectrophometer. Serum and hair selenium was determined by a spectroflourometric method. The serum zinc and selenium concentrations in the IDA group were found to be significantly lower and serum copper significantly higher than those in the controls (p<0.05). Lower iron, zinc, and selenium concentrations (p<0.001) but not copper were found in hair (p>0.05).  相似文献

The serum concentrations of copper, zinc, iron, and cobalt and copper/zinc ratio were investigated in horses infected with equine herpesvirus-1 (EHV-1). Nine horses were naturally infected with the virus and nine healthy horses served as controls. The concentrations of copper, zinc, iron, and cobalt were determined spectrophotometrically in the blood serum of all horses. The results were (expressed in micrograms per deciliters) copper 2.80 ± 0.34 vs 1.12 ± 0.44, zinc 3.05 ± 0.18 vs 0.83 ± 0.06, iron 2.76 ± 0.17 vs 3.71 ± 0.69, cobalt 0.19 ± 0.37 vs 0.22 ± 0.45, and copper/zinc ratio 0.72 ± 0.38 vs 1.41 ± 0.36 for control vs infected group, respectively. In conclusion, copper and zinc concentrations of the infected group were lower than the control group (p < 0.001), whereas iron concentration and the copper/zinc ratio of the infected group were higher than the control group (p < 0.05 and p < 0.001). The cobalt concentration was not found to be statistically different between two groups. It might be emphasized that copper/zinc ratio was significantly affected by the EHV-1 infection, so it could be taken into consideration during the course of infection. Essential elements, mainly selenium and zinc, were involved in protection against oxidative stress in cells. Oxidation could lead to the formation of free radicals that have been implicated in the pathogenesis of many diseases, including leukemia. Leukemia is a neoplastic disease that is susceptible to antioxidant enzyme and essential elements alterations. This study was undertaken to examine the levels of essential elements, antioxidant enzymes activities, and their relationships with different types of leukemia. Serum selenium, zinc, and copper concentrations, red blood cell glutathione peroxidase (GPx) activities, plasma Cu−Zn superoxide dismutase (Cu−Zn SOD) activities and lipid peroxidation (LPO) levels were determined in 49 patients with different types of leukemia before initial treatment. Serum selenium and zinc concentrations were lower in leukemia patients than those of controls (p<0.01). Serum copper concentration was higher in leukemia patients than that of controls (p<0.01). The activities GPx and Cu−Zn SOD were significantly increased in leukemia patients, especially with acute leukemia (AL), acute lymphoid leukemia (ALL), and acute nonlymphoid leukemia (ANLL) (p<0.05), whereas no difference was found between those of chronic myelogeneous leukemia and the controls. The levels of LPO were normal as controls. Serum selenium concentration was not correlated with GPx, and serum levels of zinc and copper were not related to Cu−Zn SOD. Serum zinc levels had a negative correlation with the absolute peripheral blast cells, whereas serum copper had a positive correlation with the absolute peripheral blast cells. Increased GPx and Cu−Zn SOD activities and normal levels of LPO, which were a protective responses, were an indicator of mild oxidative stress; it mights indicate that the essentials elements alterations in leukemia patients were mostly dependent on tumor activity. Changes of their levels demonstrated that there are low selenium, zinc, and high copper status in leukemia patients. The decrease of plasma zinc and increase of the Cu/Zn ratio could be the index that showed an unfavorable prognosis of acute leukemia.  相似文献

Meals of 12 diets were prepared from conventional foods with precautions against contamination by metallic elements because of epidemiologic associations between ischemic heart disease and the metabolism of magnesium, calcium, copper, and zinc. Magnesium, calcium, copper, and zinc were measured by atomic absorption spectrometry with satisfactory accuracy and precision. The mean daily amount of copper in the diets was less than the apparent adult requirement. Mean amounts of magnesium and zinc were close to apparent requirements; however, adults consuming amounts less than one standard deviation below these means may be depleting body stores and be at risk of pathology. Magnesium and copper were highly correlated (r=0.849,P=0.0001) in meals. This and other significant correlations probably will prevent the relationships of these elements to ischemic heart disease from being elucidated by epidemiology. Metabolic experiments will be necessary to differentiate among several hypotheses.  相似文献

The toxic milk (tx) mouse is a rodent model for Wilson disease, an inherited disorder of copper overload. Here we assessed the effect of copper accumulation in the tx mouse on zinc and iron metabolism. Copper, zinc and iron concentrations were determined in the liver, kidney, spleen and brain of control and copper-loaded animals by atomic absorption spectroscopy. Copper concentration increased dramatically in the liver, and was also significantly higher in the spleen, kidney and brain of control tx mice in the first few months of life compared with normal DL mice. Hepatic zinc was increased with age in the tx mouse, but zinc concentrations in the other organs were normal. Liver and kidney iron concentrations were significantly lower at birth in tx mice, but increased quickly to be comparable with control mice by 2 months of age. Iron concentration in the spleen was significantly higher in tx mice, but was lower in 5 day old tx pups. Copper-loading studies showed that normal DL mice ingesting 300 mg/l copper in their diet for 3 months maintained normal liver, kidney and brain copper, zinc and iron levels. Copper-loading of tx mice did not increase the already high liver copper concentrations, but spleen and brain copper concentrations were increased. Despite a significant elevation of copper in the brain of the copper-loaded tx mice no behavioural changes were observed. The livers of copper-loaded tx mice had a lower zinc concentration than control tx mice, whilst the kidney had double the concentration of iron suggesting that there was increased erythrocyte hemolysis in the copper-loaded mutants.  相似文献

We have determined the plasma concentrations of copper, zinc, copper/zinc ratio, and carbonic anhydrase activity in horses infected with Babesia equi. The study was conducted in 14 horses with the disease and 10 healthy animals that served as controls. The infection was confirmed by the clinical manifestations of the disease and by Giemsa staining of thin blood smears showing the parasites inside red blood cells. The horses with piroplasmosis had lower plasma levels of zinc, elevated copper, and increased activity of carbonic anhydrase. Consequently, the copper/zinc ratio was also higher than in the healthy controls.  相似文献

Puberty associated with intense physical activity results in oxidation stress. Zinc supplementation may benefit antioxidant capacity although it may also affect iron and copper status. This study evaluated the effect of zinc supplementation on antioxidant, zinc and copper status of physically active male football players (13 years ± 0.4 years), divided in two groups and studied during 12 weeks: Zn‐supplemented (Zn‐SUP, 22 mg Zn d?1 as zinc gluconate, n = 21) and placebo (PLA, n = 26). At baseline, there was no significant difference in biochemical indices between the two groups. After treatment, plasma zinc and erythrocyte iron increased in both groups (p < 0.001); urinary zinc increased (p < 0.001) only in Zn‐SUP, and erythrocyte zinc decreased (p = 0.002) only in PLA. Plasma iron and copper decreased (p = 0.01 and p = 0.015, respectively) only in Zn‐SUP. Plasma ferric‐reducing ability and plasma conjugated dienes increased, and erythrocyte osmotic fragility decrease in both groups, although the latter two were significantly lower in Zn‐SUP compared to PLA (p < 0.01). In conclusion, our study indicates that the use of 22 mg d?1 of supplemental zinc during 12 week in adolescent athletes did not affect growth, improved markers of antioxidant status but reduced plasma iron and copper. Therefore, it appears that the use of zinc supplementation by healthy adolescent athletes benefits their antioxidant capacity but impairs copper and iron nutritional status. The acute toxicity of copper, zinc and manganese and copper-zinc and copper-manganese mixtures were determined for juvenile longfin dace, Agosia chrysogaster in hard water bioassays (mean=218 mg 1−1 CaCO3). Copper-zinc was the most lethal toxicant (96-h L.c.50= 0.21 mg 1−1 copper and 0.28 mg 1−1 zinc) and exhibited a more than additive toxicity which was in contrast to the additive toxicity of copper-manganese mixtures (96-h L.c.50= 0.45 mg 1−1 copper and 64.0 mg 1−1 manganese). The toxicity of copper (96-h L.c.50= 0.86 mg 1−1) and zinc (96-h L.c.50= 0.79 mg 1−1) to the fish was similar but both were considerably more lethal than manganese (96-h L.c.50= 130 mg 1−1).  相似文献

The study describes the concentrations of zinc, magnesium, copper, iron, and biochemical markers of seminal vesicle and prostate in seminal plasma of different categories of infertile men. The zinc level in oligasthenospermic subjects was significantly higher than in azoospermic, asthenospermic, and oligospermic subjects. The γ-glutamyl transpeptidase activity in both oligoasthenospermic and azoospermic subjects were significantly lower than in asthenospermic and oligospermic individuals. There was no significant difference in iron, copper, and magnesium levels and acid phosphatase activity among the different infertile categories. A positive correlation between zinc and acid phosphatase (r=0.84, r=0.73; p<0.05), and between copper and fructose (r=0.81, r=0.72; p<0.05) was observed in oligoasthenospermic and azoospermic men, respectively. On the basis of our results, it may be postulated that there is a direct effect of the trace elements zinc and copper on acid phosphatase and fructose, respectively, in oligoasthenospermic and azoospermic subjects.  相似文献

Summary Data on metal-induced elemental changes in eukaryotic unicellular organisms, such as protozoa, are rare. This study focused on two species of ciliate protozoa,Aspidisca cicada andVorticella convallaria, both common and abundant in activated sludge. Elemental changes in cells exposed to aluminium, copper, and zinc were determined by scanning electron microscopy-electron probe X-ray microanalysis. X-ray emission spectra obtained from cells 2 h after resuspension in Chalkley's medium (control) showed clear, characteristic peaks for magnesium, silicon, phosphorus, sulphur, potassium, and copper. These elements were also routinely detected in all cells resuspended in metal solution. Spectra obtained from cells treated with aluminium or zinc showed additional distinct peaks for aluminium and zinc. In copper-treated cells enhanced copper peaks were seen. Mean aluminium levels were low inA. cicada control cells (0.14mg/g of dry weight) but higher in cells treated with 0.50 ppm (0.69 mg/g of dry weight) and 2.00 ppm aluminium (2.07 mg/g of dry weight).A. cicada was ten times more sensitive to copper than to the other metals, and thanV. convallaria. There was a significantly higher concentration of copper inA. cicada cells treated with 0.20 ppm copper (2.94 mg/g of dry weight) than in control cells (2.16 mg/g of dry weight). InA. cicada cells treated with 0.50 and 2.00 ppm zinc significantly higher levels of zinc (2.41 and 2.07 mg/g of dry weight, respectively) were observed than in the control (0.41 mg/g of dry weight). After treatment with 0.50 ppm and 2.00 ppm aluminium, aluminium was significantly higher in the treatedV. convallaria cells (1.58 and 0.67 mg/g of dry weight, respectively) than in control cells (0.14 mg/g of dry weight). After treatment with 2.00 ppm copper there was significantly more copper in treatedV. convallaria cells (3.63 mg/g of dry weight) than in controls (2.08 mg/g of dry weight). InV. convallaria cells treated with 0.50 ppm and 2.00 ppm zinc there was an increase in the amount of zinc in cells (1.30 and 2.68 mg/g of dry weight, respectively) compared with control cells (0.34 mg/g of dry weight). Data on other changes in intracellular elements in both species, after 2 h exposure to aluminium, copper, and zinc medium are given. In both species, there was uptake and/or accumulation of aluminium, copper, and zinc when the external concentration of the metal was increased. Intracellular elemental levels were altered by sublethal and toxic external concentrations of the metals studied.A. cicada was ten times more sensitive to copper than to aluminium or zinc, and thanV. convallaria to the metals studied.  相似文献

Viral hepatic diseases, especially those induced by the hepatitis B virus, can progress into more serious pathological outcomes and eventually to hepatocellular carcinoma. A growing body of evidence indicates that many trace elements play important roles in a number of carcinogenic processes that proceed through various mechanisms. To examine the status of trace elements during the development of hepatic carcinoma, we determined the selenium, iron, copper, and zinc levels and copper-to-zinc ratios in the serum of patients at different stages of viral hepatic disease. We observed significant changes in the selenium, iron copper, and zinc levels in the serum of patients having hepatocellular carcinoma, relative to those of healthy controls (p<0.05). The mean serum copper level in patients with hepatocellular carcinoma was significantly higher than that of the control group. In contrast, the mean selenium, iron, and zinc levels in patients having hepatocellular carcinoma were significantly lower than those of the control group. In addition, the mean zinc level in the serum of patients with hepatic cirrhosis was significantly lower than that of the control group (p<0.05). Moreover, we found markedly elevated Cu: Zn ratios (p<0.05) in patients having hepatic cirrhosis or hepatocellular carcinoma. Our findings imply that the levels of some trace elements, such as selenium, iron, copper, and zinc, and Cu:Zn ratios, might serve as biomarkers for the increased severity of viral hepatic damage.  相似文献

This study investigates the comparative strategies of accumulation under standardised laboratory conditions of the essential metals zinc and copper, and the non-essential metal cadmium by three crustaceans of different taxa; vizPalaemon elegans Rathke (Malacostraca: Eucarida: Decapoda),Echinogammarus pirloti (Sexton & Spooner) (Malacostraca: Peracarida: Amphipoda) and the barnacleElminius modestus Darwin (Cirripedia: Thoracica).The decapodP. elegans regulates body zinc concentrations to a constant level (ca. 79 µg Zn g–1) over a wide range of dissolved metal availabilities until regulation breaks down at high Zn availabilities and net accumulation begins. The amphipodE. pirloti accumulates zinc at all dissolved zinc concentrations but at a low net rate such that the accumulation strategy approaches that of regulation. The barnacleE. modestus accumulates zinc to high body concentrations with no significant excretion of accumulated zinc. In the case of copper,P. elegans similarly regulates body copper concentrations to a constant level (ca. 129 µg Cu g–1) over a range of dissolved copper availabilities until regulation breaks down at high copper concentrations. Both the amphipodE. pirloti and the barnacleE. modestus on the other hand accumulate copper at all dissolved copper exposures with no evidence of regulation. All three crustaceans accumulate the non-essential metal cadmium at all dissolved cadmium concentrations without regulation.Heavy metal accumulation strategies therefore vary between crustacean taxa and between metals. Uptake rates for zinc and cadmium have been estimated for the three crustaceans and can be interpreted in terms of cuticle permeability and way of life of each crustacean. Examination of these uptake rates provides an insight into possible reasons behind the adoption of particular metal accumulation strategies.  相似文献

Four superoxide dismutase (SOD) (E.C. 1.15.1.1) isozymes were present in whole tissue homogenates of Musca domestica when examined by polyacrylamide gel electrophoresis. One of the isozymes contained manganese, and the other three contained copper and zinc. All were observed in each of the body tagma (head, abdomen, and thorax) and at each developmental stage (egg to adult). The copper- and zinc-containing isozymes purified from newly emerged, adult M. domestica had a relative molecular weight of 34,800 as determined by gel filtration chromatography but consisted of two equal-size subunits of 16,000 as measured by sodium dodecylsulfate polyacrylamide gel electrophoresis. An isoelectric point between 4.8 and 5.1 was measured. Approximately 2 mol each of copper and zinc were present per dimer. The three copper, zinc isozymes were identified as charge variants. The amino acid composition of the enzyme was similar to that of copper, zinc-containing superoxide dismutases from other sources. Purified housefly copper, zinc superoxide dismutase was neither deactivated nor able to protect lactic dehydrogenase against deactivation in the presence of light and rose bengal, a known generator of singlet oxygen. The role of SOD in the phototoxic reaction involving rose bengal is discussed.  相似文献

Plasma essential trace elements, selenium, copper, zinc, and iron concentrations and the levels of immunoregulatory cytokines, interleukin-1β (IL-1β), interleukin-2 receptor (IL-2r), IL-6, IL-8, and tumor necrosis factor-α (TNF-α) were evaluated in patients with cutaneous leishmaniasis (CL) to investigate a possible role of these cytokines on selenium, zinc, copper, and iron homeostasis in CL patients. Plasma albumin levels were measured as an index of nutritional status. Plasma selenium, zinc, and iron concentrations, and IL-2r levels were significantly lower, and copper concentrations and IL-1β, IL-8, IL-6 and TNF-α levels were significantly higher in patients with CL than those of healthy controls. There was no significant difference in plasma albumin levels between two groups. There were positive important correlations between plasma selenium and IL-2r, copper and IL-6, and copper and IL-1β, and negative correlations between selenium and IL-8, iron and TNF-α, and zinc and IL-1β contents in patients with CL. Our results showed that plasma trace element contents change in patients with CL. These changes may not be a result of a specific deficiency from dietary inadequacies or imbalances, but, probably, a result of a part of the defense strategies of an organism that is regulated by immunoregulatory cytokines.  相似文献

A mixed culture of a chlorobenzoate-(3-CBA)-degradingPseudomonas aeruginosa, strain 3mT, and a phenol/cresols-degradingPseudomonas sp., strain CP4, simultaneously and efficiently degraded mixtures of 3-CBA and phenol/cresols. However, strains 3mT and CP4 usedortho- andmeta-ring cleavage pathways, respectively. Degradation of 3-CBA was complete when the 3-CBA was equal in amount to or less than that of phenol. CP4/3mT inoculum ratios (w/w) of 1:1 or 1:2 gave the most effective degradation of both the substrates in the mixture. The mixed culture degraded equimolar mixtures of 3-CBA/phenol up to 10mm. The estimation of the number of variables operating in cases of toxic action toward living organisms is discussed. The toxicity of glucose, sodium chloride, and copper nitrate solutions to Polycelis nigra has been investigated and also that of copper nitrate solutions whose osmotic pressure was adjusted to a constant level by the addition of glucose or of sodium chloride. It is shown that hypertonic solutions of copper nitrate are abnormally toxic but that the osmotic variable is not the factor responsible for this abnormally high toxicity. The lack of data which might elucidate such problems is indicated. During the course of this work it was observed that mixed solutions of copper nitrate + sodium chloride and copper nitrate + glucose exhibit toxicities greater than those expected from consideration of the separate toxicities of the components of the mixtures.  相似文献
