Abstract

The effect of the growth retardants on the structure of Pea seedlings coltured in nutritive solution. – The addition of CCC (2-Chloro-ethyltrimethyl ammonium chloride) and AMO 1618 (4-Hydroxyl-5-isopropyl-2 methyl-phenyl-trimethylammonium chloride. 1-piperidine carboxylate) to Pea seedlings (Pisum sativum L. var. Gloria di Quimper) promotes the usual modifications induced by growth retardants on higher plants. CCC appears less effective than AMO 1618; CCC inhibits growth only at 102-M. concentration, on the contrary 5×10-5M. AMO 1618 inhibits strongly the growth of the seedlings both in the light and in darkness. CCC and AMO 1618 operate similarly as far as the inhibition of expansion growth, the increase of the stem diameter, and the decrease of the apical dominance are concerned. 10-2M. CCC stimulates both the growth of roots and the secondary roots formation, on the contrary 2,5×10-4M. AMO 1618 inhibits strongly the growth of the roots. AMO 1618 affects more strongly than CCC the expantion growth of the leaves. Leaves of the AMO 1618 treated plants are greener than the control plants. Plants treated with CCC and AMO 1618 are smaller because these chemicals inhibit the expantion growth of the cells. The increase of the stem diameter induced by CCC and AMO 1618 is due to the stimulation of the mitotic activity of the cambium. AMO and CCC induce a decrease of the size of the vessels and the sieve tubes. In the sieve tubes of the treated plants and slime plugs appear near to the sieve plates many slime bodies. AMO and CCC did not affect the mitotic activity of the apical meristems; in fact the plants grown in the presence of the growth retardants, show a normal primary body. AMO and CCC delay the lignification process. Chloroplasts of this Pisum sativum variety show prolamellary bodies associated to a good lamellar system. Starch granules are always present. Starch was never found in the chloroplasts of the treated plants. The general picture of the effects induced by growth retardants in Pea seedlings show so many modifications that it is very difficult to believe, like some Authors suggest, that all the effects produced by growth retardants are due to the inhibition of gibberellin biosynthesis.  相似文献

Abstract

Action of cyanide on growth and greening of seedlings of Picea abies and Larix decidua.—The effects of cyanide on growth and greening of dark germinated seedlings of Picea abies and Larix decidua are studied. While growth of Picea seedlings is scarcely inhibited, the effect of the inhibitor on Larix seedlings is very drastic. Greening of Picea seedlings is never inhibited while, at stronger concentrations, cyanide weakly inhibits greening of Larix seedlings. When seedlings are treated with cyanide and salicylhydrossamic acid, an inhibitor of mithocondrial respiration cyanide non-sensible, growth is inhibited more strongly than when cyanide only is used and greening too is inhibited. Cyanide is shown to be able to reach megagametophyte and embryo and to be incorporated steadily.  相似文献

Physiological dormancy of scarified seeds of Townsville stylo (Stylosanthes humilis H.B.K.) was released by seleno-L-methionine (SeM), but not by L-methionine. This regulating effect was impaired by inhibitors of ethylene biosynthesis and action; in the first case SeM action was restored by 2-chloroethylphosphonic acid (CEPA) and 1-aminocyclopropane-1-carboxylic acid (ACC). The Se-aminoacid proved to be toxic in a time-dependent manner to seedling growth, inhibiting primarily the hypocotyl expansion. This toxicity is suggested to trigger ethylene biosynthesis, which would promote germination of dormant seeds.  相似文献

Epinodosin, an ent-kaurane diterpenoid isolated from Isodon japonica var. galaucocalyx, had a biphasic, dose-dependent effect on root growth and a strong inhibitory effect on root hair development in Lactuca sativa L. seedlings. Lower levels of epinodosin (25–100 μM) significantly promoted root growth, but higher concentrations (150–200 μM), by contrast, had inhibitory effects. In addition, all of the tested concentrations (20–80 μM) inhibited root hair development of lettuce seedlings in a dose-dependent manner. Further investigations on the underlying mechanism revealed that the promotion effect of epinodosin (25–100 μM) resulted from increasing the cell length in the mature region and enhancing the mitotic activity of meristematic cells in lettuce seedling root tips. On the other hand, epinodosin at higher concentrations inhibited root growth by strongly affecting both the cell length in the mature region and the division of meristematic cells. Comet assay analysis demonstrated that the decrease of mitotic activity of root meristematic cells was due to DNA damage induced by higher levels of epinodosin.  相似文献

Experimental investigations were carried out to evaluate the effect of continuous and delayed exposure of power frequency electromagnetic fields at 5, 50 and 100 μT on germinating Vicia faba seedlings as a model system. These studies included physical parameters (length and girth of primary roots, number as well as length of lateral roots and imbibition), major biochemical constituents (total sugar, protein, and fat) and activities of important housekeeping enzymes (amylases, proteases, and lipase) at 2, 4, and 8 days of growth. Also, mitotic index and rate of DNA synthesis were studied at day 8 of growth. There was no significant change in physical parameters and major biochemical constituents between control and experimental groups. Also, the comparison between the control and experimental group of seeds showed that α-amylase activity significantly decreased at 5, 50 and 100 μT on day 2 and 4 of growth. β-amylase and protease (37C & 50C) showed a significant decrease in activity on day 2 and 4 of growth at 100 μT, whereas activity of lipase significantly decreased only on day 2 of growth at 100 μT. At day 8 of growth, all enzyme activities reverted back to the same as control. Also, there was a significant increase in mitotic index as well as 3H-thymidine uptake at 100 μT delayed exposure on day 8. The present study suggests that exposure to power frequency electromagnetic fields up to 100 μT on germinating seedlings does not cause any permanent damage since the initial alteration under the magnetic fields in some important housekeeping enzymes involved in the onset of seed germination were returened to control values on day 8 of growth. Also, the growth of the germinated seedlings was found to be enhanced by the application of power frequency magnetic fields (100 μT) as evidenced by mitotic index and 3H-thymidine uptake.  相似文献

Abstract

Significance of integuments in caryopses of « Triticum durum » Desf. treated with X-rays. Experiments were carried out on Triticum durum « Cappelli », to test the influence of seed coats on the damage by X-rays. The damage was evaluated in terms of germination, seedling growth and chromosomal aberrations. Seeds in which the embryo was covered (with pericarp and spermoderm), uncovered (without pericarp) and bared (without pericarp and spermoderm), were irradiated at the total exposure of 1000, 2000 and 4000 R of X-rays. The growth of seedlings coming from covered and uncovered embryos, X-rayed at 1000 R, results better than that of bared embryos. The chromosomal aberrations analysis carried out in root meristem cells at anaphase, during the first mitotic cycle, show the highest damage in seedlings coming from uncovered and bared embryos and the type of aberrations was almost chromatidic (B'). We therefore forward the hypothesis that the removal of the integuments (pericarp and spermoderm) could possibly accelerate the process of germination, and could also produce in the embryo a greater sensitivity to X-rays.  相似文献

Abscisic acid, a potent growth inhibitor inhibits hypocotyl growth ofRaphanus sativus seedlings. Phenolic compounds,viz., trans-cinnamic acid, chlorogenic acid, ferulic acid, salicylic acid, tannic acid and quercetin when applied with ABA, antagonize ABA action and restore normal seedling growth. Gibberellic acid promotes hypocotyl growth and on combined application with ABA, the ratio of their concentrations determines the course of the resultant growth. This interaction can be modulated by phenolic compounds. Phenolic compounds in low concentrations when present together with GA and ABA, favour GA-induced growth by antagonizing the inhibitory influence of ABA. The inhibitory action of abscisic acid on a wide range of growth processes is so far known to be reversed only by growth promoting hormones,viz., IAA, GA and cytokinins. Antagonistic action of phenolic compounds towards ABA, and increasing the action of GA when present together with GA and ABA, establishes a dual role to this class of compounds; balancing the effect of both growth promoting and growth inhibiting hormones. The ethyl acetate extract of the Bacillus sp. EJ-121 culture broth exhibited growth inhibitory activity on a lettuce (Lactuca sativa L.) seedlings assay. Bacillus sp. EJ-121 was identified as Bacillus cereus by the morphological characteristic and nucleotide sequence of the 16S rDNA. The bioassay-guided fractionation of the ethyl acetate extract led to the isolation of two compounds. Their structures were deduced by spectroscopic methods and determined as sodium vanillate (1) and 2-aminobenzoic acid (2). Both compounds 1 and 2 inhibited more than 90% of root length at 50 ppm (0.26 and 0.36 mM, respectively) while they had a limited effect on shoot growth at the same concentration level. Roots and shoots of lettuce seedlings showed severe deterioration at 100 ppm. In order to study the fundamental structure–activity relationship, several structurally related benzoic acid derivatives were also assayed. The existence of a polar carboxyl moiety seemed to be responsible for the stronger activity.  相似文献

【目的】从3种蓝莓根际土壤中分离细菌,探究蓝莓根际土壤细菌多样性,并筛选具有产酸、促生长、抑菌性能的菌株,为蓝莓专用微生物肥料的研究提供优质菌株资源和理论基础。【方法】选用5种培养基分离3种蓝莓根际土壤细菌,并进行16S rRNA基因测序和系统发育分析。筛选产酸、产吲哚-3-乙酸(indole-3-acetic acid,IAA)和铁载体、固氮、溶磷和抑制灰葡萄孢生长的菌株,挑选最适菌株制备菌剂进行蓝莓苗盆栽实验验证促生能力,并检测菌剂对蓝莓元素吸收和根际土壤肥力的影响。【结果】从3种蓝莓根际土壤分离得到124株细菌,挑选70株代表性菌株进行16S rRNA基因测序,分布于3个门21个属,其中芽孢杆菌属(Bacillus)、假单胞菌属(Pseudomonas)、链霉菌属(Streptomyces)和红球菌属(Rhodococcus)为优势分离菌群。代表性菌株中,21.4%的菌株能产酸,21.4%的菌株产吲哚-3-乙酸,47.1%的菌株具有固氮潜力,65.7%的菌株具有解磷能力,14.3%的菌株能产铁载体。少量菌株同时具有产酸、产IAA、固氮、解磷和抑菌等能力。选取具有产酸和多种促生特征的菌株绿针假单胞菌CSM-70和双鱼假单胞菌CSM-129进行盆栽蓝莓苗处理,发现2株菌均能显著促进蓝莓苗的生长发育并调控根际土壤pH,其中菌株CSM-70处理还显著促进了蓝莓叶片氮、磷元素的吸收,提升了土壤速效钾、碱解氮的含量。【结论】蓝莓根际细菌多样性高且蕴藏着丰富的促生长菌株,绿针假单胞菌CSM-70和双鱼假单胞菌CSM-129能够促进蓝莓苗生长、调控根际土壤pH和肥力,并促进植株养分吸收,具有蓝莓专用微生物菌剂研制与应用的潜力。  相似文献

【目的】探讨耐重金属细菌对铅(lead, Pb)和镉(cadmium, Cd)胁迫下菊苣(Cichorium intybus L.)幼苗Pb、Cd抗性和黄酮生物合成的调控作用。【方法】在不同浓度Pb和Cd [(200+20) mg/kg、(400+40) mg/kg、(800+80) mg/kg]处理下,接种菌株JB19并测定菊苣幼苗生长指标、Pb和Cd含量、抗氧化酶活性、总黄酮含量和黄酮生物合成相关基因表达量。【结果】菌株JB19可显著提高不同浓度Pb和Cd处理下菊苣的生物量和叶绿素含量;减少氧化损伤,地上部和根部Pb、Cd含量均降低,其中在(Pb200+Cd20) mg/kg处理下H2O2和丙二醛(malondialdehyde, MDA)含量分别比对照降低了25.7%和26.1%,地上部Pb、Cd含量降低的幅度最大,分别降低了53.2%和54.1%;加强了菊苣幼苗的抗氧化酶防御系统,黄酮生物合成相关基因均显著上调,其中在(Pb400+Cd40) mg/kg处理下黄酮类化合物的含量增加了105.2%,查尔酮异构酶基因上调最显著,达458....  相似文献

Rice (Oryza sativa L.) seedlings inhibited the growth of hypocotyls and roots of cress (Lepidium sativum L.) seedlings when both seedlings were grown together. Two growth inhibiting substances were found in the culture solution in which rice seedlings were hydroponically grown for 14 d. One growth inhibitor was further purified. This suggests that the rice seedlings may produce growth inhibiting substances, acting as allelochemicals to other plants, and release them from their roots into the environment.  相似文献

Abstract

Action of some respiratory inhibitors on growth and greening of seedlings of Picea and Larix.—The effects of sodium azide, arseniate, jodoacetate, fluoride and 2,4-dinitrophenol on growth and greening of dark germinated seedlings of Picea abies and Larix decidua were studied.

All the inhibitors showed a negative influence on growth (determined as fresh weight) of Picea seedlings and even more remarkably on Larix seedlings.

Greening was significantly inferior to that of control plants only for groups of Picea seedlings treated with sodium azide, at all the concentrations used, and for groups treated with fluoride and arseniate at the highest concentrations.

All the groups of Larix seedlings, instead, treated with inhibitors, with the exception of the ones treated with sodium fluoride at the lowest concentrations, presented a chlorophyll content inferior to that of control groups.

From the above results the different influence of inhibitors on Picea and on Larix seedlings might be due to the different metabolic pathways through which the pigment synthesis in the two species under study takes place.  相似文献

The effects of temperature and photoperiod on the ability of wheat (Triticum aestivum L.) seedlings to show induced responses (increased accumulation of hydroxamic acids, Hx) upon infestation by the bird cherry-oat aphid, Rhopalosiphum padi L. were evaluated under laboratory conditions. Induction of Hx was significantly higher at lower temperatures. No such clear trend was found for the photoperiod effect. The significant effect of environmental conditions on growth rate of seedlings and the significant negative correlation between growth rate prior to infestation and induction of Hx suggested that environmental effects on induced responses were at least partially mediated through their effect on plant growth rate. After statistically uncoupling the effect of environmental conditions from the effect of plant growth rate, the effect of temperature on induction of Hx was no longer significant. Therefore, the temperature effect was mediated by plant growth rate.  相似文献

Summary The positive geotropic responses of the primary roots of Zea mays and Pisum sativum seedlings depend upon at least one growth inhibiting factor which arises in the root cap and which moves basipetally through the apex into the extending zone. The root apex (as distinct from the cap) and the regions more basal to the extending zone are not sources of growth regulators directly involved in the geotropic response. A difference in the concentration or effectiveness of the inhibitory factor(s) arising in the cap must be established between the upper and lower halves of a horizontal root. Positive geotropic curvature in a horizontal root is attributable, at least in part, to a downward lateral transport of inhibitor(s) from the upper to the lower half of the organ.  相似文献

芽胞杆菌是目前植物病害生物防治研究最多的一类微生物,其在自然界中分布广泛,开发潜力大。【目的】为了探究从杏树根际土壤分离的芽胞杆菌BJ-6的分类地位及其防病促生作用。【方法】本研究测定了芽胞杆菌BJ-6的形态和生理生化特征,通过PCR扩增了该菌的16S rRNA、gyrA和gyrB基因并进行了序列测定,通过多基因聚类分析确定其分类地位,平板对峙法测定抗菌谱,盆栽幼苗实验验证其对甜瓜细菌性果斑病的防治效果和对甜瓜的促生作用。【结果】结合形态特征、生理生化特性及多基因序列分析建立的系统进化树,确定菌株BJ-6为解淀粉芽胞杆菌(B. amyloliquefaciens),抑菌实验发现该菌株对15种植物病原菌均有不同程度的抑菌活性,盆栽实验结果发现该菌株发酵液对甜瓜细菌性果斑病有很好的防治效果,并对甜瓜苗有很好的促生作用。【结论】BJ-6属于解淀粉芽胞杆菌,抑菌谱广,且具有防病促生作用,具有进一步开发为生防制剂的前景。  相似文献

The growth of wheat seedlings (Triticum sativum) is inhibited by abscisic acid (ABA). The inhibition increases with the concentration of ABA (from 10-6M to 5 × 10-5M) and is stronger in the case of coleoptiles and first leaves than in roots. In contrast, naphthaleneacetic acid (ANA), at 10-5M, exerts its greatest inhibitory effect on the roots. The inhibitory effect of ABA on coleoptiles can be partially overcome by kinetin and to a much smaller degree by gibberellic acid. Neither of these two compounds, at 10-5M, had any effect on the ABA-induced inhibition of root growth. The RNA and DNA contents per plant organ are considerably reduced after treatment of the seedlings with ABA, particularly in the coleoptiles and the first leaves. The incorporation of uracil-2-14C and uridine T (G) into RNA of treated seedlings is reduced in the case of coleoptiles and first leaves, but considerably enhanced in roots. The mechanism of the action of ABA is discussed in the light of these results.  相似文献

Abstract

INHYBITION OF NODULATION IN METEOR PEA, BY NICTOPHASIC TREATMENT WITH FAR-RED RADIATION. — A remarkable inhybiting effect of far-red light (given to the aerial part of the plants as a flash befor and during the nictophase) on Pea root nodulation, has been achivied through some preliminary experiments. The relations between irradiated aerial parts and the activities of the underground organs in general, are being studied in the picture of photo-chromoperiodism.

Relations with the action of gibberellic acid (GA) are preliminary attempted. The inhibiting action of GA is far less striking in comparison with that of far-red radiation. Kinetin favores, at some extent, the nodulation process.  相似文献

Centaurea ragusina L. (Asteraceae) is an endemic Croatian plant species, which developed xeromorphic characteristics as a consequence of its natural environment – vertical limestone cliffs above the Adriatic sea. Cytogenetic status of C. ragusina long-term culture (94th subculture) and C. ragusina seedlings was analysed and compared after 4 weeks of growth. Cytogenetic stability was investigated in root meristem cells of C. ragusina cultured plants originated from Pen đa (cliffs near Dubrovnik) and seedlings originated from three different localities in south Adriatic (Penđa, Pasjača – cliffs near Dubrovnik and island of Komiža) using mitotic index and mitotic and chromosomal abnormalities as parameters. Mitotic indices of cultured plants and ‘Penđa’ seedlings were similar and showed significant increase compared to mitotic indices of ‘Komiža’ and ‘Pasjača’ seedlings. Although the highest number of mitotic abnormalities was recorded in root meristem cells of cultured plants, it was only a bit higher than in root tips of ‘Pasjača’ and ‘Penđa’ seedlings, while that of ‘Komiža’ was two times lower compared to cultured plants. Pattern of analysed mitotic abnormalities was very similar in root tips of cultured plants and ‘Pasjača’ and ‘Penđa’ seedlings, with exception of ‘Komiža’ seedlings. Presented results suggest that long-term cultivation of C. ragusina has almost no effect on culture ageing considering similar distribution of scored mitotic abnormalities as in ‘Penđa’ and ‘Pasjača’ seedlings.  相似文献

Amylase inhibitor producing actinobacteria were isolated and characterized from terrestrial environment and there is no much report found from marine environment, hence in the present study, 17 strains isolated from the rhizosphere sediments of mangroves were tested for their amylase inhibition ability. Seawater requirement test for the growth of actinobacteria found that the strains SSR-3, SSR-12 and SSR-16 requires at least 50% and SSR-6 requires at least 25% seawater for their growth. The inhibition activity of both prokaryotic and eukaryotic amylase was tested by using Bacillus subtilis and Aspergillus niger. The maximum amylase activity (40mm) produced by the A. niger was taken as positive control, when the test actinobacteria strains grown in the medium they inhibited amylase activity and was evidenced by the reduction in inhibition zone (14–37 mm) similarly the amylase produced by the Bacillus subtilis was also recorded maximum (35 mm) amylase activity and was taken as positive control, and the test atinobacterial strains reduced enzyme action(12–33 mm) it varied levals. This indicates that the actinobacteria strains were controlled amylase enzyme activity in both the cases. The strain SSR-10 was highly effective and SSR-8 was less effective in inhibiting eukaryotic amylase produced by A. niger. The strain SSR-2 was effective and SSR-6 showed very less effect in inhibiting the prokaryotic amylase produced by the B subtilis.  相似文献

Background: As the climate warms, plant interactions between shrubs and conifer seedlings may affect migration of boreal trees into alpine areas; however these interactions have not been widely tested across conifer life stages.

Aims: Determine the role of shading by Betula glandulosa shrubs on early Picea mariana recruitment in the forest-tundra ecotone of the Mealy Mountains, Labrador (Canada).

Methods: Four shrub treatments were established with varying degrees of shade (unaltered, trimmed, cleared, shaded). Emergence and growth of P. mariana in treatment plots were compared to control plots without shrubs.

Results: Seedling emergence was facilitated; seedlings beneath shrubs emerged at greater levels (16% ± 5.1%) than in the control (0.5% ± 0.3%); however, evidence of facilitation post-emergence was weak, likely due to stronger interactions with seedbed species at this stage. Vertical growth of young seedlings was greatest in control and shaded plots, but needle production significantly decreased with increased shade suggesting that seedlings may compete with shrubs for light early in life.

Conclusions: Although facilitation has been previously implicated as an important mechanism in stressful environments, our study indicates that the early life stages of some conifers at the tree line are not strongly facilitated by shrubs as has been previously suggested for adults.  相似文献
