石楠拟盘多毛孢毒素产生条件初探   总被引:1,自引:0,他引:1  
石楠拟盘多毛孢是引起草莓根腐病的优势病原菌之一, 本实验在明确该病菌毒素是主要致病物质的基础上, 利用叶圆片法对该菌的产毒条件进行了初步探讨, 结果表明, 除光照影响不明显外, pH值、温度、振荡及培养时间等对此菌产生毒素均有显著影响, 其最适产毒条件为:培养基初始pH值为自然pH值(约为6.2)、温度为25°C、黑暗、静置培养5 d~7 d。此外, 实验还发现该菌产生的粗毒素对玉米、黑麦和绿豆种子萌发、根或芽的延伸生长都有明显的抑制作用。  相似文献

【目的】探讨利用tri5-PCR鉴定产毒镰孢菌的可行性以及tri5阳性菌株的产毒特性和产毒条件。评估鸡舍空气和固体基质中镰孢菌分离株中产单端孢霉烯族毒素潜力的发生率。【方法】利用编码单端孢霉烯族毒素合成酶的起始基因(tri5基因)对139株镰孢菌分离株进行PCR检测,并对tri5阳性菌株进行产毒培养,通过免疫亲和柱-高效液相色谱方法来检测培养产物中的T-2毒素和HT-2毒素含量。【结果】共筛选到42株tri5阳性菌株,其中分离自鸡舍空气中的10株tri5阳性菌株经产毒培养后7株菌株产生T-2毒素(1.36-5 ng/mL)或HT-2毒素(6.1-17.1 ng/mL)。在5℃-20℃间隔24 h变温、光照与黑暗间隔24 h交替、前期振荡后期静止的培养条件下培养9 d镰孢菌的产毒量最高;镰孢菌的产毒量与温度和时间显著相关,而与菌丝体干重无显著相关性。【结论】与传统鉴定产毒镰孢菌的方法比较,tri5-PCR更适用于快速、准确地检测大量鸡舍环境样本中的产毒镰孢菌。本研究为养殖环境的产毒镰孢菌的危害预警和控制提供技术支撑和理论依据。  相似文献

本对嗜线虫致病杆菌(Xenorhadbus nematophilus)产生抗生素的发酵培养基和发酵条件进行了研究,同时对该菌代谢过程pH值、还原糖、总糖、氨基氮与抗生素产量的关系进行了分析,通过筛选该菌对碳源和氮源的要求,用正交试验初步确定了该菌产素的最佳发酵培养基和条件为:玉米粉1%,大豆粉3%,蔗糖1%,蛋白胨1.5%,KH2PO40.02%,MgSO40.2%,活化剂T0.1%;发酵培养基的起始pH值在6.0-8.0,种龄16h,接种量4%,500mL摇瓶装量15-150mL的条件下培养72h可获得较高的抗生素产量;产素量与菌代谢过程中pH、还原糖、总糖和氨基氮的变化有一定关系,通过培养基和培养条件的研究使该菌的产抗生素能力提高了56.3%。  相似文献

【目的】从青海大骨节病区小麦麦穗中分离的内生真菌中筛选产T-2毒素的菌株,并研究影响其合成该毒素的条件。【方法】采用种子胚芽抑制试验和抑菌试验从分离所得的菌株中筛选产毒菌株;利用薄层层析和高效液相检测待测菌株产物,复筛出产T-2毒素的菌株。通过显微形态学观察及ITS序列分析对筛选出的菌株5-5m-1进行鉴定。应用单因素筛选方案研究固体培养时间、温度以及液体培养转速、初始p H等对其产T-2毒素的影响,并采用正交试验进一步优化。【结果】菌株5-5m-1的显微形态与梨孢镰孢菌(Fusarium poae)相似;ITS序列分析显示,该菌株与F.poae的相似度也较高。其产T-2毒素的最佳条件为:玉米固体培养基、日温25°C/夜温15°C、光暗交替。【结论】5-5m-1菌株为梨孢镰孢菌,培养条件对其产T-2毒素能力有很大影响。实验结果将为进一步研究T-2毒素产生的机制和防止真菌毒素污染提供参考。  相似文献

针对赤霉菌生长与产次级代谢产物赤霉素的关系进行摇瓶静息培养研究。在不同菌龄及不同底物浓度的条件下,菌丝产素能力有较大不同。实验数据表明:在菌龄96h,葡萄糖浓度为5gfL时,菌丝静息培养72h后,培养液中赤霉素含量最高,达1100μg/mg;同时单位时间单位菌丝产率也最大,为7.1μg/mg·h;在该菌龄条件下,当葡萄糖浓度为1dL时,赤霉素转化效率最高,为0.8484。该研究有助于进一步优化发酵工艺,降低赤霉素生产成本。  相似文献

真菌毒素是产毒真菌产生的次生代谢产物,以木霉、曲霉、毛霉、青霉和根霉为主的丝状真菌是农产品中常见的产毒真菌。阐明农产品中产毒致病微生物的基因组序列信息是揭示真菌特殊遗传性状的基础。截至2013年12月,共有子囊菌门中204个种和担子菌门中62个种的基因组序列已经测序或公布,它们的基因组大小大部分在30-40 Mb。整理了部分已完成基因组测序的具有产毒能力或致病力的真菌基因组信息,并对真菌测序方案、主要产毒真菌及其比较基因组学的研究进展进行了综述。  相似文献

对番茄褐斑病菌(Helminthosporiurn carposaprum)产毒条件和毒素对不同番茄品种的毒性进行了研究,结果表明,不同的pH值、温度、光照条件、培养天数所制备的毒素毒力差异显著,病菌的最佳产毒条件是室温25℃,光照12 h、pH值为6、振荡培养15 d;不同植物对病菌毒素的敏感性不同。  相似文献

【目的】优化蜡样芽胞杆菌R75E菌株产胶原酶的条件,并通过蛋白分离纯化技术获得高纯度胶原酶。【方法】利用单因素及正交试验优化蜡样芽胞杆菌R75E产胶原酶的发酵条件及发酵培养基,将发酵液离心除菌后得到粗酶液,对其依次通过硫酸铵分级沉淀、Butyl FF疏水层析及SuperdexTM 200凝胶过滤层析等方法对目标胶原酶进行分离纯化,利用SDS-PAGE电泳检测其纯度。【结果】优化后发酵条件为培养温度41°C、接种量6%、培养时间36 h,优化后发酵培养基为葡萄糖10 g/L、蛋白胨5 g/L、起始p H 7.0,粗酶液酶活力较优化前提高了2.9倍;将该粗酶液经过一系列纯化后得到纯度超过90%的胶原酶产物,其纯化倍数和回收率分别为18.4和1.1%。【结论】获得蜡样芽胞杆菌R75E的最佳产酶条件,并对胶原酶分离纯化的方法进行了探索,为微生物胶原酶的开发应用奠定基础。  相似文献

The fungus Alternaria alternata is a common spot‐producing plant pathogen. During the past decade, tobacco brown spot disease caused by this fungus has became prevalent in China and lead to significant losses. To better understand the molecular pathogenesis of this fungus, the aapk1 gene encoding a cAMP‐dependent protein kinase catalytic subunit was cloned, sequenced and characterized. The aapk1 deletion mutants were identified from hygromycin‐resistant transformants by PCR strategy and confirmed by Southern blot analysis and RT‐PCR. The aapk1 deletion mutant exhibited reduced vegetative growth and was less toxic than the wild‐type strain sd1. Deletion of aapk1 also delayed disease development on detached tobacco leaves. Thus, we propose that the cAMP signalling pathway is involved in mycelia growth and pathogenic phenotype of Alternaria alternata.  相似文献

Alternaria citri, the cause of Alternaria black rot, and Alternaria alternata rough lemon pathotype, the cause of Alternaria brown spot, are morphologically indistinguishable pathogens of citrus: one causes rot by macerating tissues and the other causes necrotic spots by producing a host-selective toxin. To evaluate the role of endopolygalacturonase (endoPG) in pathogenicity of these two Alternaria spp. pathogens, their genes for endoPG were mutated by gene targeting. The endoPGs produced by these fungi have similar biochemical properties, and the genes are highly similar (99.6% nucleotide identity). The phenotypes of the mutants, however, are completely different. An endoPG mutant of A. citri was significantly reduced in its ability to cause black rot symptoms on citrus as well as in the maceration of potato tissue and could not colonize citrus peel segments. In contrast, an endoPG mutant of A. alternata was unchanged in pathogenicity. The results indicate that a cell wall-degrading enzyme can play different roles in the pathogenicity of fungal pathogens. The role of a cell wall-degrading enzyme depends upon the type of disease but not the taxonomy of the fungus.  相似文献

从云南高原地区的竹叶病理组织上分离到1株可产生红棕色素的丝状真菌ZL07-8。通过对分生孢子梗、分生孢子、喙等的形态学研究及对ITS和5.8S rRNA序列的分子系统学分析,发现此菌为链格孢属小孢子种群成员。然而该菌具有区别于已知种的特征,如分生孢子梗常单生,少数成簇,无分枝;分生孢子大多单生,较少链生,孢身较大;寄主为华西箭竹,故立为竹链格孢新种。干制培养物ZL07-8D保藏于山东理工大学生命科学学院实验室。  相似文献

美人蕉是重要的园林绿化植物和生物资源。为了明确美人蕉黑斑病的病原菌种类,并为病害防治提供理论指导,调查了昆明市美人蕉黑斑病的发生情况,采用组织分离法对病原物进行了分离和纯化,并进行了致病性测定。通过形态学和rDNA ITS序列分析,将昆明美人蕉黑斑病的病原菌鉴定为链格孢Alternaria alternata。  相似文献

通过大量筛选从银杏茎中分离到一株内生真菌(编号为No.1028),其代谢产物对重要作物真菌病害病原菌具有明显的抑制作用。结果表明:离体条件下内生真菌发酵液对番茄早疫病菌、黄瓜枯萎病菌、菜豆炭疽病菌、葡萄炭疽病菌、水稻纹枯病菌、黄瓜立枯病菌均有较强的抑制作用,对上述不同病原菌的抑制率分别为:66.7%、48.3%、64.6%、36.5%、57.1%和23%。同时也研究了不同碳源、氮源、无机离子对内生真菌No.1028生物学特性和生长的影响。结果发现玉米粉、黄豆饼粉比较适合其生长和抑菌代谢产物的合成,Na+对抑菌代谢产物的合成有较明显的促进作用。  相似文献

Alternaria infectoria E . G . Simmons was isolated from naturally infected eggs of the fig wax scale insect , Ceroplastes rusci L . (Homoptera: Coccidae) , that were showing different degrees of shriveling . This fungus proved to be pathogenic to eggs , nymphs and adults of this pest . The fungus had significant effects on both the mortality and hatchability of the eggs ( P = 0 . 0001) . Three days after exposure to the fungus , nearly one - third (31 . 5%) of the fungus - treated eggs became discolored and 17% showed moderate shrinking but no mortality . Fifteen days post - application , 91% of the fungus - treated eggs were diseased (74 . 8% of them were dead) due to A. infectoria, while no disease developed on the untreated eggs . Hatching decreased by 39 . 6% over a 15 - day period in the fungus - treated eggs compared to the control . Additionally , crawlers hatching from fungus - treated eggs became infected . The fungus seemed to induce crawlers to enter the settling stage . Seventy - two per cent of the fungus - treated crawlers versus 9% of the untreated controls entered the settling stage 6 days after exposure to the fungal inoculum . The highest level of nymphal mortality attributed to A. infectoria occurred when 30% (wet w:v blended mycelium in water containing 0 . 5% w:v Metamucil) inoculum was applied , and a high relative humidity was maintained for the following 48 h . This is the first record of A. infectoria on the fig wax scale and for the genus Alternaria as an entomopatho genic fungus .  相似文献

Atopy to the fungus Alternaria is strongly associated with respiratory disease. The prevalences of asthma and of allergy to Alternaria are high amongst children living in rural towns of south-eastern Australia. In such towns, airborne allergenic spores have been proposed to arise from nearby crops, but this has not been tested and crops are unlikely to be the only sources of Alternaria . We sought to identify sources and factors that influence concentrations of spores of Alternaria detected in rural towns. Over two years, we sampled spores in two towns (Wagga Wagga and Moree, New South Wales, Australia), in nearby wheat and cotton crops during harvesting and control periods, in a cotton gin and a grain shed. Alternaria was present in both towns throughout the study, and above the crops, at the gin and grain shed. Daily and annual concentrations were amongst the highest recorded worldwide and peaks persisted for six months in Wagga Wagga and ten months in Moree. Crop maturation affected the spore load in the air more than the actual days of harvest. Regression analysis showed that the overall spore concentrations above towns correlated with those above crops. Variables of rainfall and maximum temperature correlated with concentrations in both towns, and additionally wind direction in Wagga Wagga. In conclusion, crops and produce handling released spores into the air that reached nearby rural towns, with peaks in spore concentrations following warm temperatures and recent rainfall.  相似文献

A novel host-selective phytotoxin was isolated from Alternaria solani fungus. Correlation between blighting of tomato plants and resistance of the pollen to the substance was found to be -0.75. A fungus capable of using vegetable tissues for multiplication in soil was isolated and identified as Aspergillus flavus based on morphological characteristics and sequence similarity of ITS and 28S. When grown in liquid medium prepared from the same vegetable tissues used in soil amendment, the isolate of A. flavus produced a substance capable of preventing disease development of black leaf spot of mustard cabbage caused by Alternaria brassicicola and inhibiting the germination of A. brassicicola conidia. The inhibitory substance was fungistatic, and was very stable under high temperature and high or low pH value. It was soluble in ethanol or methanol, moderately soluble in water, and insoluble in acetone, ethyl acetate or ether. The inhibitor is not a protein and has no charges on its molecule. This is the first discovery of the production of a fungistatic substance by this deleterious fungus. Results from this study suggest the possession of a strong competitive saprophytic ability by A. flavus, which in turn may explain the widespread occurrence of this fungus in soils. Production of a fungistatic substance when A. flavus was grown in medium prepared from vegetable tissues suggests the importance of antibiotic production in its competitive saprophytic colonization of organic matters in soils.  相似文献

An isolate of Alternaria crassa (Saac.) Rands was obtained from a naturally occurring leaf-spotting disease of Jimsonweed (Datura stramonium L.) plants. The fungus was induced to sporulate and used to reproduce the disease on seedlings in greenhouse and field inoculation tests. The fungus grew best at 25–35°C, and germinated best at 25–30°C. Infectivity on Jimsonweed was reduced at temperatures below 20°C and above 30°C. In greenhouse host range studies, conidia of the fungus were highly pathogenic to Jimsonweed, moderately pathogenic to ‘Marion’ and ‘Beefsteak’ tomatoes, and non-pathogenic to several other crop and weed species representing seven botanical families. These findings indicate that A. crassa has potential as a biological herbicide for controlling Jimsonweed.  相似文献
