Four experiments were done to characterize the interactions of copper, iron, and ascorbic acid with manganese in rats. All experiments were factorially arranged Dietary Mn concentrations were less than 1 micrograms/g (Mn0) and 50 micrograms/g (Mn+). Dietary Cu was less than 1 mg/g (Cu0) and 5 micrograms/g (Cu+); dietary Fe was 10 micrograms/g (Fe10) and 140 micrograms/g (Fe140). Ascorbic acid (Asc) was not added to the diet or added at a concentration of 10 g/kg diet. Experiment 1 had two variables, Mn and Cu; in Experiment 2, the variables were Mn and Asc. In Experiment 3, the variables were Mn, Cu, and Asc; in Experiment 4, they were Mn, Cu, and Fe. Definite interactions between Mn and Cu were observed, but they tended to be less pronounced than interactions between Mn and Fe. Cu depressed absorption of 54Mn and accelerated its turnover. In addition, adequate Cu (Cu+), compared with Cu0, depressed liver, plasma, and whole blood Mn of rats. Absorption of 67Cu was higher in animals fed Mn0 diets than in those fed Mn+. Ascorbic acid depressed Mn superoxide dismutase activity and increased Cu superoxide dismutase activity in the heart. The addition of ascorbic acid to the diet did not affect Mn concentration in the liver or blood. Absorption of 54Mn was depressed in rats fed Fe140 compared with those fed Fe10. Interactions among Fe, Cu, and Mn resulted in a tendency for Mn superoxide dismutase activity to be lower in rats fed Fe140 than in rats fed Fe10. Within the physiologic range of dietary concentrations, Mn and Cu have opposite effects on many factors that tend to balance one another. The effects of ascorbic acid on Mn metabolism are much less pronounced than effects of dietary Cu, which in turn affects Mn metabolism less than does Fe.  相似文献

Autopsied liver tissue samples collected from 42 males and 31 females were analyzed for copper, manganese and zinc using atomic absorption spectrometry (AAS). With the exception of two liver samples for which the copper levels were determined to be 74.8 and 104.0 g/g (dry weight), hepatic copper concentrations were found to range from 1.7 to 32.4 g/g with a mean concentration of 14.2 g/g and standard deviation of 7.0 g/g. Manganese concentrations (with the exception of one sample having 12.9 g/g) ranged from 0.22 to 4.6 g/g with a mean of 2.26 ± 1.00 g/g. Hepatic zinc levels averaged 118.3 ± 44.4 g/g and ranged from 38.5 to 231.3 g/g. There were no apparent trends for the levels of any metals versus age nor were there any differences in average hepatic metal concentrations for males and females. © Rapid Science 1998.  相似文献

The absorption of Cu2+, Zn2+, and Mn2+ by leaf tissue of 4-month old sugarcane plants (Saccharum officinarum L., var. H53-263) has been investigated. After the “apparent free space” fraction was desorbed, the absorption of Cu2+, Mn2+, and Zn2+ yielded a curve typical of many ion uptake processes when measured as a function of the external concentration. However, only 1 absorption mechanism was evident for each cation. The pH optimum for Cu2+ and Zn2+ uptake was 5.0 to 6.0, whereas that for Mn2+ absorption was 4.5 to 6.0. Absorption was competitively inhibited by H+, and this inhibition was reversible when 0.5 mm Ca2+ was present. Cu2+ and Zn2+ were absorbed through the same carrier sites, as concluded from their mutually competitive activities. Mn2+ was absorbed through a second, independent mechanism. Uptake of each cation was strongly inhibited by uncouplers of oxidative phosphorylation, by Amytal and Nembutal2, by 5 × 10−2m succinate, and by ADP and Pi. Absorption of Cu2+, Zn2+, and Mn2+ was concluded to be coupled to oxidative phosphorylation, and specifically to energy-conservation Site I.  相似文献

Four sheep, each prepared with a rumen fistula and reetrant cannula in the proximal duodenum, were used to study the effects of ruminal administration of nitrilotriacetic acid on solubilities of zinc, copper, manganese, and iron in rumen and duodenal digesta. The sheep received a pelleted diet and were dosed with 0, 300, 600 and 1200 microgram of nitrilotriacetic acid per gram of diet via the rumen fistula. Higher concentration of soluble zinc, manganese, and iron but not copper, were found in the rumen of the sheep when they were dosed with nitrilotriacetic acid. The concentrations increased with increasing dose of the acid. However, only the solubilty of iron was increased in the duodenal digesta. Concentrations of soluble zinc and manganese in the rumen increased, whereas copper decreased, during the first 2 h after feeding. The pattern was reversed thereafter. Changes in the concentrations of soluble during 6 h afther feeding were comparatively small. It is concluded that the solubilty of iron in the stomach of sheep is increased by ruminal administration of nitrilotriacetic acid.  相似文献

A series of metal carboxylates containing pyridine N-oxide are prepared via one pot synthesis and solid phase synthesis. The structural variations from metal to metal are observed. In the case of reactions of manganese(II) acetate with pyridine N-oxide in the presence of aromatic carboxylic acids, polymeric complexes with bridging aromatic carboxylate as well as bridging pyridine N-oxide are observed. Whereas, the reaction of copper(II) acetate with pyridine N-oxide in the presence of an aromatic carboxylic acid led to mononuclear or binuclear paddle wheel carboxylate complexes with monodentate pyridine N-oxide. Co-crystal of two neutral complexes having composition [Cu2(OBz)4(MeOH)2][Cu2(OBz)4(pyO)2] (where OBz = benzoate, pyO = pyridine N-oxide) each neutral parts have paddle wheel structure. Solid phase reaction of zinc chloride with sodium benzoate prepared in situ and pyridine N-oxide leads to a tetra-nuclear zinc complex.  相似文献

Impaired cellular homeostasis of metals, particularly of Cu, Fe and Mn may trigger neurodegeneration through various mechanisms, notably induction of oxidative stress, promotion of α-synuclein aggregation and fibril formation, activation of microglial cells leading to inflammation and impaired production of metalloproteins. In this article we review available studies concerning Fe, Cu and Mn in Parkinson's disease and Wilson's disease. In Parkinson's disease local dysregulation of iron metabolism in the substantia nigra (SN) seems to be related to neurodegeneration with an increase in SN iron concentration, accompanied by decreased SN Cu and ceruloplasmin concentrations and increased free Cu concentrations and decreased ferroxidase activity in the cerebrospinal fluid. Available data in Wilson's disease suggest that substantial increases in CNS Cu concentrations persist for a long time during chelating treatment and that local accumulation of Fe in certain brain nuclei may occur during the course of the disease. Consequences for chelating treatment strategies are discussed.  相似文献

This article describes a study in which four trace elements (Se, Mn, Cu, and Fe) were analyzed in the blood serum of the patients with colorectal cancer from the Moravian region of the Czech Republic. Atomic absorption spectrometry with graphite furnace atomization was used for analysis of selenium and manganese and with flame atomization for analysis of copper and iron. The observed serum concentrations in adenocarcinoma colorectal patients of selenium were significantly lower (41.8 ± 11.6 μg/L) and those of manganese (16.3 ± 4.5 μg/L) and iron (2.89 ± 1.23 mg/L) were significantly higher as compared to the age-matched control group. Copper serum content (0.95 ± 0.28 mg/L) did not significantly differ as compared to healthy population.  相似文献

Acute experiments were conducted on 82 cats; a study was made of the influence of copper, manganese and cobalt on the chemical sensitivity of interoceptors. Administration of copper chloride caused inversion of the reflex reactions of the blood pressure and respiration to acetylcholine, cobalt chloride weakened these reactions, and manganese chloride intensified them. It is supposed that changes in the interoceptive reflexes observed depended chiefly on the direct action of microelements on the functional condition of the interoceptors.  相似文献

Abstract Absorption of copper and manganese by sunflower roots from solution cultures of varying composition was followed by measuring the concentrations of the metals appearing in whole roots, root cell sap and xylem exudate. Total copper in the fibrous roots was linearly related to the concentration of copper in the external solution but the concentration of copper released to the xylem exudate was buffered somewhat against the changes made externally. No such buffering was observed for managenese. A copper-sensitive electrode, responsive only to free cupric ions was used in conjunction with total copper analysis by atomic absorption spectrophotometry to show that little of the copper (usually < 1%) existed as a free ion in any phase of the system. Copper in the xylem exudate may be strongly complexed. An electron paramagnetic resonance spectrum of the xylem exudate indicated that manganese probably was a free divalent ion. Calculation of the electrochemical potential gradient for free cupric ions showed that no special metabolically-linked mechanism need be postulated to account for absorption of copper (or manganese) other than that necessary to maintain the transmembrane potential.  相似文献

Chronic administration of chlorpromazine produced alterations in trace metal concentrations in caudate nucleus, frontal cortex, and cerebellar hemisphere of guinea pigs. Manganese concentration following chronic chlorpromazine rose significantly in the caudate nucleus and cerebellar hemisphere whereas iron concentration rose most significantly in the caudate nucleus. Copper content was decreased in all regions examined. The possible significance of increased manganese and iron in the caudate nucleus is discussed in relationship to the clinical problems of chlorpromazine-induced dyskinesias.  相似文献

Copper/zinc (Cu/ZnSOD) and manganese (MnSOD) superoxide dismutases which catalyze the dismutation of toxic superoxide anion, O inf2 sup– , to O2 and H2O2, play a major role in protecting cells from toxicity of oxidative stress. However, cells overexpressing either form of the enzyme show signs of toxicity, suggesting that too much SOD may he injurious to the cell. To elucidate the possible mechanism of this cytotoxicity, the effect of SOD on DNA and RNA strand scission was studied. High purity preparations of Cu/ZnSOD and MnSOD were tested in an in vitro assay in which DNA cleavage was measured by conversion of phage X174 supercoiled double-stranded DNA to open circular and linear forms. Both types of SOD were able to induce DNA strand scission generating single- and double-strand breaks in a process that required oxygen and the presence of fully active enzyme. The DNA strand scission could be prevented by specific anti-SOD antibodies added directly or used for immunodepletion of SOD. Requirement for oxygen and the effect of Fe(II) and Fe(III) ions suggest that cleavage of DNA may be in part mediated by hydroxyl radicals formed in Fenton-type reactions where enzyme-bound transition metals serve as a catalyst by first being reduced by superoxide and then oxidized by H2O2. Another mechanism was probably operative in this system, since in the presence of magnesium DNA cleavage by SOD was oxygen independent and not affected by sodium cyanide. It is postulated that SOD, by having a similar structure to the active center of zinc-containing nucleases, is capable of exhibiting non-specific nuclease activity causing hydrolysis of the phosphodiester bonds of DNA and RNA. Both types of SOD were shown to effectively cleave RNA. These findings may help explain the origin of pathology of certain hereditary diseases genetically linked to Cu/ZnSOD gene.  相似文献

A number of essential trace elements play a major role in various metabolic pathways. Selenium (Se), manganese (Mn), copper (Cu), zinc (Zn), and iron (Fe) are essential trace elements that have been studied in many diseases, including autoimmune, neurological, and psychiatric disorders. However, the findings of previous research on the status of trace elements in patients with schizophrenia have been controversial. We studied these elements in patients with a DSM-IV diagnosis of schizophrenia and compared them with sex- and age-matched healthy controls. Plasma Cu concentrations were significantly higher (p<0.01) and Mn and Fe concentrations were lower (p<0.05 and p<0.05, respectively) in schizophrenic patients than in controls. Se and Zn concentrations and protein levels did not differ between patients and healthy controls. These observations suggest that alterations in essential trace elements Mn, Cu, and Fe may play a role in the pathogenesis of schizophrenia. However, findings from trace element levels in schizophrenia show a variety of results that are difficult to interpret.  相似文献

The role of trace elements in the pathogenesis of liver cirrhosis and its complications is still not clearly understood. Serum concentrations of zinc, copper, manganese and magnesium were determined in 105 patients with alcoholic liver cirrhosis and 50 healthy subjects by means of plasma sequential spectrophotometer. Serum concentrations of zinc were significantly lower (median 0.82 vs. 11.22 micromol/L, p < 0.001) in patients with liver cirrhosis in comparison to controls. Serum concentrations of copper were significantly higher in patients with liver cirrhosis (median 21.56 vs. 13.09 micromol/L, p < 0.001) as well as manganese (2.50 vs. 0.02 micromol/L, p < 0.001). The concentration of magnesium was not significantly different between patients with liver cirrhosis and controls (0.94 vs. 0.88 mmol/L, p = 0.132). There were no differences in the concentrations of zinc, copper, manganese and magnesium between male and female patients with liver cirrhosis. Only manganese concentration was significantly different between Child-Pugh groups (p = 0.036). Zinc concentration was significantly lower in patients with hepatic encephalopathy in comparison to cirrhotic patients without encephalopathy (0.54 vs. 0.96 micromol/L, p = 0.002). The correction of trace elements concentrations might have a beneficial effect on complications and maybe progression of liver cirrhosis. It would be recommendable to provide analysis of trace elements as a routine.  相似文献
