Influence of moderate chilling stress on vascular bundle sheath cell (BSC) and especially mesophyll cell (MC) chloroplasts of mature maize leaves was studied by electron microscopy and stereology. Plants of two inbred lines of maize, differing in their photosynthetic activity, and their F1 hybrids were cultivated during autumn in heated or unheated glasshouse. Generally, chilling temperatures resulted mainly in the decrease in stereological volume density (VD) of both granal and intergranal thylakoids of MC chloroplasts, while the ratio of granal to all thylakoids (granality) was less affected. The VD of peripheral reticulum and plastoglobuli usually increased after cold treatment of plants. The volume of MC chloroplasts usually increased under chilling stress, the shape of the chloroplasts changed only slightly. The ultra-structure of chloroplasts differed between individual genotypes; chilling-stressed hybrid plants showed positive heterosis particularly in the granal thylakoids' VD of MC chloroplasts.  相似文献

The influence of chilling (8 °C, 5 d) at two photon flux densities [PFD, L = 200 and H = 500 μmol(photon) m−2 s−1] on the gas exchange and chlorophyll fluorescence was investigated in chilling-tolerant and chilling-sensitive maize hybrids (Zea mays L., K383×K130, K185×K217) and one cultivar of field bean (Vicia faba L. minor, cv. Nadwiślański). The net photosynthetic rate (P N) for the both studied plant species was inhibited at 8 °C. P N of both maize hybrids additionally decreased during chilling. Changes in the quantum efficiency of PS2 electron transport (ΦPS2) as a response to chilling and PFD were similar to P N. Measurements of ΦPS2CO2 ratio showed that in field bean seedlings strong alternative photochemical sinks of energy did not appear during chilling. However, the high increment in ΦPS2CO2 for maize hybrids can indicate reactions associated with chill damage generation. At 8 °C the non-photochemical quenching (NPQ) increased in all plants with chilling duration and PFD. The appearance of protective (qI,p) and damage (qI,d) components of qI and a decrease in qE (energy dependent quenching) took place. NPQ components of field bean and maize hybrids differed from each other. The amount of protective NPQ (qE + qI,p) components as part of total NPQ was higher in field bean than in maize hybrids at both PFD. On 5th day of chilling, the sum of qE and qI,p was 26.7 % of NPQ in tolerant maize hybrids and 17.6 % of NPQ in the sensitive one (averages for both PFD). The increased PFD inhibited the ability of all plants to perform protective dissipation of absorbed energy. The understanding of the genotypic variation of NPQ components in maize may have implications for the future selection of plants with a high chilling tolerance.  相似文献

The differences between two maize (Zea mays L.) inbred lines and their F1 hybrids in their response to chilling periods of various duration (1, 2, 3 or 4 weeks) and subsequent return to optimum temperatures were analysed by the measurement of the photosystem (PS) 1 and 2 activity, the photosynthetic pigments' content and the activity of antioxidant enzymes. The PS2 activity and the chlorophyll content decreased in plants subjected to 3 or 4 weeks of chilling, but not in those subjected to 1 or 2 weeks of chilling. This decrease was more pronounced in inbreds compared to their hybrids. The activity of superoxide dismutase did not much change with the increasing length of chilling period in the inbreds but decreased in the hybrids, the glutathione reductase activity increased in both types of genotypes but more in the inbred lines, while for ascorbate peroxidase and catalase the changes in parents-hybrids relationship did not show any specific trend. The PS1 activity and the carotenoids' content was not much affected.  相似文献

The ultrastructure and dimensions of chloroplasts in leaf mesophyll cells were quantitatively examined in three parental inbred lines of maize (Zea mays L.) and their four hybrids subjected to two types of four-week low-temperature (LT) treatment: the abrupt onset of chilling temperatures (“severe chilling”, SC) and the gradual, more moderate one (“moderate chilling”, MC). The relationship between the response of individual genotypes to one or the other type of chilling was analyzed as well as the possibility to predict the behaviour of chloroplasts in hybrids from that of their parents. Although selected parameters of chloroplast ultrastructure (e.g. volume densities of granal and intergranal thylakoids, plastoglobuli, and peripheral reticulum) and dimensions changed due to the exposure of maize plants to LT, no general pattern of such changes was found for this species due to the observed intraspecific variability. The response of some genotype to SC could not be predicted from its behaviour under MC (and vice versa) and no clear rules could be applied for the inheritance of chloroplast response to chilling in the general sense. Thus, great caution should be always taken when interpreting the results of studies aimed at the dissection of chloroplast ultrastructure as affected by LT, particularly in case such studies are made with one genotype or under one type of chilling only.  相似文献

Young plants of maize inbred lines CE777, CE704, and CE810 and their F1 hybrids displaying a positive heterotic effect in various photosynthetic characteristics were exposed to low temperature during their early growth developmental stage. The photochemical activity of isolated mesophyll chloroplasts and the contents of photosynthetic pigments in leaves of stressed and non-stressed plants were compared with the aim to find out the possible changes in the relationship between parents and hybrids, and to determine the genetic basis of heterosis in F1 generation. Strong decrease in the content of chlorophylls was observed for all genotypes examined when plants were subjected to low growth temperature. Similar change was recorded for Hill reaction activity (HRA) of inbred lines but not of their F1 hybrids, and no significant response at all was found for photosystem 1 (PS1) activity or the total carotenoids content. The intraspecific variation due to differences between genotypes was found for most of photosynthetic characteristics examined. This variation was caused by the additive and dominance genetic effects. Positive dominance was the main cause of positive heterosis in HRA and in the contents of photosynthetic pigments and was much more pronounced in the stressed plants compared to the non-stressed ones. The maternal additive effects participated in the inheritance of contents of photosynthetic pigments in plants exposed to low temperature, too. A betA gene encoding choline dehydrogenase from Escherichia coli was transformed into cotton (Gossypium hirsutum L.) via Agrobacterium-mediated transformation. Transgenic cotton plants exhibited improved tolerance to chilling due to accumulation of glycinebetaine (GB). The results of our experiment showed that GB contents of leaves of transgenic lines 1, 3, 4, and 5, both before and after chilling stress, were significantly higher than those of wild-type (WT) plants. At 15°C, transgenic lines 1, 3, 4, and 5 exhibited higher germination capacity as determined by the germination speed and final germination percentage and, displayed less inhibition in seedling shoot growth rate than WT plants. Under chilling stress, transgenic lines 4 and 5 maintained higher relative water content, upper carbon dioxide (CO2) fixation capacity and PSII electron transfer rate, better osmotic adjustment (OA), a lower percentage of ion leakage, and less lipid membrane peroxidation when compared with WT plants. Chilling resistance of the transgenic lines was demonstrated to be positively correlated with GB content under chilling stress. The high levels of GB in transgenic cotton plants might not only protect the integrity of cell membrane from chilling damage, but also be involved in OA which alleviated chilling induced water stress. Moreover, under chilling-stressed conditions, transgenic cotton plants enhanced stomatal conductance, PSII electron transport rate, and further leaf photosynthesis through accumulating high levels of GB.  相似文献

Many C4 plants, including maize, perform poorly under chilling conditions. This phenomenon has been linked in part to decreased Rubisco abundance at lower temperatures. An exception to this is chilling‐tolerant Miscanthus, which is able to maintain Rubisco protein content under such conditions. The goal of this study was to investigate whether increasing Rubisco content in maize could improve performance during or following chilling stress. Here, we demonstrate that transgenic lines overexpressing Rubisco large and small subunits and the Rubisco assembly factor RAF1 (RAF1‐LSSS), which have increased Rubisco content and growth under control conditions, maintain increased Rubisco content and growth during chilling stress. RAF1‐LSSS plants exhibited 12% higher CO2 assimilation relative to nontransgenic controls under control growth conditions, and a 17% differential after 2 weeks of chilling stress, although assimilation rates of all genotypes were ~50% lower in chilling conditions. Chlorophyll fluorescence measurements showed RAF1‐LSSS and WT plants had similar rates of photochemical quenching during chilling, suggesting Rubisco may not be the primary limiting factor that leads to poor performance in maize under chilling conditions. In contrast, RAF1‐LSSS had improved photochemical quenching before and after chilling stress, suggesting that increased Rubisco may help plants recover faster from chilling conditions. Relatively increased leaf area, dry weight and plant height observed before chilling in RAF1‐LSSS were also maintained during chilling. Together, these results demonstrate that an increase in Rubisco content allows maize plants to better cope with chilling stress and also improves their subsequent recovery, yet additional modifications are required to engineer chilling tolerance in maize.  相似文献

We studied changes in the chlorophyll (Chl) fluorescence components in chilling-stressed sweet potato (Ipomoea batatas L. Lam) cv. Tainung 57 (TN57, chilling-tolerant) and cv. Tainung 66 (TN66, chilling-susceptible). Plants under 12-h photoperiod and 400 μmol m−2 s−1 irradiance at 24/20 °C (day/night) were treated by a 5-d chilling period at 7/7 °C. Compared to TN66, TN57 exhibited a significantly greater basic Chl fluorescence (F0), maximum fluorescence (Fm), maximum fluorescence yield during actinic irradiation (Fm′ ), and the quantum efficiency of electron transport through photosystem 2, PS2 (ΦPS2). Chilling stress resulted in decrease in the potential efficiency of PS2 (Fv/Fm), ΦPS2, non-photochemical fluorescence quenching (NPQ), non-photochemical quenching (qN), and the occurrence of chilling injury in TN66. Chilling increased the likelihood of photoinhibition, characterized by a decline in the Chl fluorescence of both cultivars, and photoinhibition during low temperature stress generally occurred more rapidly in TN66.  相似文献

The effect of the natural compound S-methylmethionine (SMM) on the functioning of the photosynthetic apparatus, the efficiency of photosynthesis and the synthesis of stress-induced phenoloids and anthocyanins involved in defence was investigated in young maize plants exposed to moderate and severe chilling stress. Damage to PSII was observed as a reduction in the value of variable fluorescence (Fv/Fm) which could be detected even after few hours of mild chilling stress. At temperatures below 10°C, the reduction in Fv/Fm was more pronounced. Changes in the value of net photosynthesis exhibited a similar tendency. SMM has a moderating effect on this reduction and its protective effect was more pronounced under long-lasting chilling stress and at the lowest temperatures. Monitoring of fluorescence intensities and ratios correlated with the levels of stress defence compounds. The fluorescence intensities were found to increase over the course of chilling stress in response to SMM, with the highest values being recorded in plants exposed to the longest period of stress. A similar tendency was observed for the quantity of anthocyanins. The results confirm the complex role of SMM, which is manifested both in preserving the ability of the photosynthetic apparatus to function and in stimulating the synthesis of metabolites involved in stress defence.  相似文献

Abstract The leaves of olive are long lived and likely to experience both chilling and high temperature stress during their life. Changes in photosynthetic CO2 assimilation resulting from chilling and high temperature stress, in both dim and high light, are investigated. The quantum yield (φ) of photosynthesis at limiting light levels was reduced following chilling (at 5°C for 12 h), in dim light by approximately 10%, and in high light by 75%; the difference being attributed to photoinhibition. Similar reductions were observed in the light-saturated rate of CO2 uptake (Amax). Decrease in Amax correlated with a halving of the leaf internal CO2 concentration (ci), suggesting an increased limitation by stomata following photoinhibition. Leaves were apparently more susceptible to photoinhibitory damage if the whole plant, rather than the leaf alone, was chilled. On return to 26 °C, I he photosynthetic capacity recovered to pre-stress levels within a few hours if leaves had been chilled in high light for 8 h or less, but did not fully recover from longer periods of chilling when loss of chlorophyll occurred. Leaves which were recovering from chilling in high light showed far more damage on being chilled a second time in high light. Three hours in high light at 38 °C reduced φ by 80%, but φ recovered within 4h of return to 26 °C. Although leaves of Olive are apparently less susceptible to photoinhibitory damage during chilling stress than the short-lived leaves of chilling-sensitive annual? crops, the results nevertheless show that photoinhibition during temperature stress is potentially a major factor influencing the photosynthetic productivity of Olive in the field.  相似文献

To determine the effects of phosphorus nutrition on chilling tolerance of photosynthetic apparatus, tomato (Lycopersicon esculentum Mill. cv. Kenfengxin 2002) plants were raised under different P contents and subjected to 7 d of chilling at 9/7 °C. After chilling (2 h or 7 d) plant growth, P content in tissue, gas exchange and chlorophyll fluorescence were measured. Decreasing P concentration [P] in the nutrient solution markedly reduced plant growth and the chilled plants exhibiting higher optimum [P] than the unchilled plants. Decreasing [P] significantly decreased light saturated net photosynthetic rate (PNsat), maximum carboxylation velocity of Rubisco (Vcmax), maximum potential rate of electron transport contributed to Rubisco regeneration (Jmax), quantum efficiency of photosystem (PS) 2 (ΠPS2) and O2 sensitivity of PNsat (PSO2) and this trend was especially apparent in chilled plants.  相似文献

Over-expression of chloroplastic glycerol-3-phosphate acyltransferase gene (LeGPAT) increased unsaturated fatty acid contents in phosphatidylglycerol (PG) of thylakoid membrane in tomato. The effect of this increase on the xanthophyll cycle and chloroplast antioxidant enzymes was examined by comparing wild type (WT) tomato with the transgenic (TG) lines at chilling temperature (4 °C) under low irradiance (100 μmol m−2 s−1). Net photosynthetic rate and the maximal photochemical efficiency of photosystem (PS) 2 (Fv/Fm) in TG plants decreased more slowly during chilling stress and Fv/Fm recovered faster than that in WT plants under optimal conditions. The oxidizable P700 in both WT and TG plants decreased during chilling stress under low irradiance, but recovered faster in TG plants than in the WT ones. During chilling stress, non-photochemical quenching (NPQ) and the de-epoxidized ratio of xanthophyll cycle in WT plants were lower than those of TG tomatoes. The higher activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in TG plants resulted in the reduction of O2 −· and H2O2 contents during chilling stress. Hence the increase in content of unsaturated fatty acids in PG by the over-expression of LeGPAT could alleviate photoinhibition of PS2 and PS1 by improving the de-epoxidized ratio of xanthophyll cycle and activities of SOD and APX in chloroplast.  相似文献

The function of chloroplast ferredoxin quinone reductase (FQR)-dependent flow was examined by comparing a wild type tobacco and a tobacco transformant (ΔndhB) in which the ndhB gene had been disrupted with their antimycin A (AA)-fed leaves upon exposure to chilling temperature (4 °C) under low irradiance (100 μmol m−2 s−1 photon flux density). During the chilling stress, the maximum photochemical efficiency of photosystem (PS) 2 (Fv/Fm) decreased markedly in both the controls and AA-fed leaves, and P700+ was also lower in AA-fed leaves than in the controls, implying that FQR-dependent cyclic electron flow around PS1 functioned to protect the photosynthetic apparatus from chilling stress under low irradiance. Under such stress, non-photochemical quenching (NPQ), particularly the fast relaxing NPQ component (qf) and the de-epoxidized ratio of the xanthophyll cycle pigments, (A+Z)/(V+A+Z), formed the difference between AA-fed leaves and controls. The lower NPQ in AA-fed leaves might be related to an inefficient proton gradient across thylakoid membranes (ΔpH) because of inhibiting an FQR-dependent cyclic electron flow around PS1 at chilling temperature under low irradiance.  相似文献

The relationships between electron transport and photosynthetic carbon metabolism were measured in maize (Zea mays L.) leaves following exposure to suboptimal temperatures. The quantum efficiency for electron transport in unchilled leaves was similar to that previously observed in C3 plants, although maize has two types of chloroplasts, mesophyll and bundle sheath, with PSII being largely absent from the latter. The index of noncyclic electron transport was proportional to the CO2 assimilation rate. Chilled leaves showed decreased rates of CO2 assimilation relative to unchilled leaves, but the integral relationships between the quantum efficiency for electron transport or the index of noncyclic electron transport and CO2 fixation were unchanged and there was no photoinhibition. The maximum catalytic activities of the Benson-Calvin cycle enzymes, fructose-1,6-bisphosphatase and ribulose-1,5-bisphosphate carboxylase, were decreased following chilling, but activation was unaffected. Measurements of thiol-regulated enzymes, particularly NADP-malate dehydrogenase, indicated that chilling induced changes in the stromal redox state so that reducing equivalents were more plentiful. We conclude that chilling produces a decrease in photosynthetic capacity without changing the internal operational, regulatory or stoichiometric relationships between photosynthetic electron transport and carbon assimilation. The enzymes of carbon assimilation are particularly sensitive to chilling, but enhanced activation may compensate for decreases in maximal catalytic activity.  相似文献

Transgenic cotton plants from several independently-transformed lines expressing a chimeric gene encoding a chloroplast-targeted Mn superoxide dismutase (SOD) from tobacco exhibit a three-fold increase in the total leaf SOD activity, strong Mn SOD activity associated with isolated chloroplasts, and a 30% and 20% increase in ascorbate peroxidase and glutathione reductase activities, respectively. The Mn SOD plants did exhibit a slightly enhanced protection against light-mediated, paraquat-induced cellular damage but only at 0.3 µM paraquat. In addition, photosynthetic rates at 10°C and 15°C were similar to those of controls, and the immediate recovery of photosynthesis after a 35-min exposure to 5°C and full sun was only slightly better than that for wild-type plants. The recovery for longer exposure times was comparable for both genotypes as was the deactivation of the H2O2-sensitive, Calvin-cycle enzyme, stromal fructose 1,6-bisphosphatase (FBPase). Compared to the controls, Mn SOD plant leaves in full sun prior to chilling stress had a lower activation of FBPase, a higher ratio of oxidized to reduced forms of ascorbate, and a higher total glutathione content. After 35 min at 5°C in full sunlight, total glutathione had risen in control leaves to 88% of the Mn SOD plant values, and oxidized to reduced ascorbate ratios were higher for both genotypes. However, an 80% increase in the ratio of oxidized to reduced glutathione occurred for Mn SOD plant leaves with no change for controls. This increased demand on the ascorbate-glutathione cycle is circumstantial evidence that high Mn SOD activity in the chloroplast leads to increased H2O2 pools that could, in some manner, affect photosynthetic recovery after a stress period. We postulate that the pool sizes of reduced ascorbate and glutathione may restrict the ability of the ascorbate-glutathione cycle to compensate for the increased activity of SOD in cotton over-producing mitochondrial Mn SOD in chloroplasts during short-term chilling/high light stress.  相似文献

The response of the photosynthetic apparatus to low temperature periods differed among three hybrids of maize (Zea mays L.) grown in a phytotron. Light-saturated photosynthetic rates, leaf chlorophyll content, and mesophyll cell photosynthetic unit density all declined with increasing duration of low temperature. No single metabolic or physiological parameter appeared to control the response of the three hybrids to low temperature stress. Among all temperature treatments, net photosynthetic rate on a leaf area basis was more closely correlated with leaf chlorophyll content than with any other measured parameter. Final shoot dry weight was most highly correlated with stomatal conductance to CO2.  相似文献

Antioxidant enzyme activities were determined at the first,third and fifth leaf stages of four inbred lines of maize (Zeamays L.) exhibiting differential sensitivity to chilling. Plantswere exposed to a photoperiod of 16:8 L:D for one of three treatments:(a) control (25C), (b) control treatment plus an exposure toa short-term chilling shock (11C 1 d prior to harvesting),and (c) long-term (11 C constant) chilling exposure. Catalase(CAT; EC 1.11.1.6 [EC] ), ascorbate peroxidase (ASPX; EC 1.11.1.11 [EC] ),superoxide dismutase (SOD; EC 1.15.1.1 [EC] ), glutathione reductase(GR; EC 1.6.4.2 [EC] ), and mono-dehydroascorbate reductase (MDHAR;EC 1.6.5.4 [EC] ) activities were assessed. Reducing and non-reducingsugars and starch concentrations were determined as generalmetabolic indicators of stress. Reduced activities of CAT, ASPX,and MDHAR may contribute to limiting chilling tolerance at theearly stages of development in maize. Changes in levels of sugarand starch indicated a more rapid disruption of carbohydrateutilization in comparison to photosynthetic rates in the chilling-sensitiveline under short-term chilling shocks and suggested a greaterdegree of acclimation in the tolerant lines over longer periodsof chilling. The aim of the present study was to assess the tolerance of Hevea brasiliensis to chilling temperatures since rubber production has been extended to sub-optimal environments. PB260 clone was used to analyze the responses of leaves chilled at 10°C during 96 h, as well as their recovery at 28°C. Some key parameters were used to evaluate photosynthetic apparatus functioning, membrane damage (electrolyte leakage) and oxidative stress. A short-term response versus a long-term one have been recorded, the time point of 24 h, when stomata closure was effective, being the border between the two responses. P n decreased dramatically at 1 h, and Fv/Fm was slightly affected. NPQ reached its maximal level between 4 and 7 h. Lipid peroxidation and membrane lysis were observed between 48 and 96 h. Activities of antioxidant enzymes increased, along with the induction of antioxidant gene expression. Finally, the plants were capable to recover (net photosynthetic rate, photochemical efficiency, antioxidant enzymes activities) when placed back to 28°C showing that PB260 can withstand long-term chilling.  相似文献

Experiments were carried out to investigate the changes in CO2 assimilation, photon allocation, and photosynthetic electron flux in leaves of cucumber (Cucumis sativus L.) plants after chilling stress. Chilling significantly decreased CO2 assimilation, the energy flux via linear electron transport (J PS2) and non-constitutive thermal dissipation (J NPQ) but increased fluorescence and constitutive thermal dissipation (J f,D) in chilling-sensitive genotype Jinyan No. 4. In contrast, chilling had little effects on J NPQ and J f,D although CO2 assimilation and J PS2 were inhibited in chilling-tolerant genotype Jinchun No. 3. In parallel with the reduction in J PS2, electron flux to oxygenation and carboxylation by ribulose-1,5-bisphosphate carboxylase/oxygenase all significantly decreased while electron flux to O2 significantly increased, especially in chilling-sensitive genotype. Thermal and fluorescence dissipation were the main energy dissipation pathways whilst water-water cycle was an important electron sink when photosynthetic carbon reduction was suppressed after chilling. Chilling sensitivity of the photosynthetic apparatus was related to the operation of different photoprotection mechanisms.  相似文献

Chilling‐induced photosynthetic impairment was examined in leaves of maize (Zea mays L.) seedlings of two cultivars, one adapted to western Europe and one adapted to Mexican highlands. Three experiments were performed in a controlled environment. The effects of chilling night temperatures, of chilling at high light intensity and of variable chilling day temperatures on photosynthetic parameters, were evaluated. Chilling in the dark period resulted in stomatal limitation of net photosynthesis. Chilling at moderate to high light intensities caused chilling‐dependent photoinhibition of CO2 uptake. Photobleached maize leaves did not resume normal photosynthetic function. Maize cv. Batan 8686 from the highlands of Mexico was less susceptible to photosynthetic damage than maize cv. Bastion adapted for cultivation in W. Europe, when exposed to chilling night temperatures, or to mild chilling photoinhibitory conditions.  相似文献
