The data considered in the paper indicate that a tumor clone resulting from cell transformation, in order to develop into an overt neoplasm, should overcome a microenvironmental constraint. This destroys intercellular contacts and cell interactions with extracellular matrix required for induction and maintenance of epithelium differentiation. The possible reasons for this lie in mutations of genes that control cell adhesion molecules and integrins, as well as proteases secreted by a tumor. These events lead to partial loss of differentiation antigens by a cell or to their incorrect localization in a cell. Simultaneously, the expression of embryo-specific genes is unblocked, leading to overexpression of embryonic antigens and their abnormal secretion into blood, which results in appearance of oncofetal serum markers. Discussed from this point of view are alpha-fetoprotein, the carcinoembryonic antigen, and the prostate-specific antigen, which are used as tumor markers.  相似文献

Rajasekariah G. R., Rickard M. D. and Mitchell G. F. 1980. Immunization of mice against infection with Taenia taeniaeformis using various antigens prepared from eggs, oncospheres, developing larvae and strobilocerci. International Journal for Parasitology10: 315–324. Antigens were collected during in vitro incubation of oncospheres, 3-week-old larvae and strobilocerci of T. taeniaeformis. Supernatants of these in vitro products centrifuged at 500 g contained antigens which stimulated a significant degree of protective immunity when injected into mice. However, centrifugation of the strobilocercus preparation at 4500 g yielded supernatants which failed to induce immunity. Suspensions of eggs and oncospheres disrupted by sonication stimulated a high level of immunity as also did 4500 g supernatants of the sonicated preparations. Centrifugation of sonicated oncospheres at 100,000 g yielded a supernatant which stimulated significantly less immunity than the 4500 g supernatant, although the protective capacity was not totally abolished. The pellet from 100,000 g centrifugation of sonicated oncospheres induced almost absolute immunity. These results are consistent with the suggestion that the ‘functional’ antigens in the preparations tested may initially be membrane-associated or particulate in nature and that sonication causes partial solubilization. Supernatants prepared from homogenised strobilocerci and centrifuged at 4500 g also stimulated protective immunity and presumably contain soluble antigens. No evidence is available to suggest whether or not the strobilocercus antigens which stimulated protective immunity are identical to those found in oncospheral preparations. Immunity was stimulated by subcutaneous, intraperitoneal and intramuscular injections of antigen and both Freund's complete adjuvant and Bordetella pertusiss vaccine were effective as adjuvants. Using sonicated oncospheres, a high level of immunity was stimulated without the use of adjuvant.  相似文献

In spite of the advent of several newer modalities in molecular and immunological methods of diagnosis, the diagnosis of smear negative tuberculosis is a major problem even today. Use of antibody detection ELISA for the purpose has been evaluated by many authors using a variety of antigens derived from mycobacteria. These antigens are reported to have many advantages and disadvantages. Here we review commonly used antigens used for detection of antimycobacterial antibodies for diagnosis of tuberculosis. Each antigen is reviewed for its application in diagnosis, sensitivity, specificity, cross-reactivity and clinical application.  相似文献

A group of antigens of 24-kD Mr from rat testes were characterised biochemically. These antigens were part of a larger molecule of approximately 200 kD. On treatment with disulfide bond reducing agent, the 200-kD molecule was reduced to subunits. Immunoreactivity was confined to a doublet of approximately 24 kD and a single band of approximately 50 kD Mr after the reduction. Glycoprotein in nature, this antigen shared immunoreactive epitopes with a 40-kD antigen on human spermatozoa. Antiserum raised in rabbits against the 24-kD antigen from rat testes reacted with antigens on the acrosome of human spermatozoa. Agglutination of sperm could be induced by the antiserum. The carbohydrate residue could be removed by mannosidase digestion. Chemical deglycosylation studies showed a slight decrease in molecular weight. Immunoreactivity was however not completely lost after chemical deglycosylation. Isoelectric focusing of the antigen identified nine isoelectric species. Two relatively minor species showed immunoreactivity. Acrosome-reacted spermatozoa showed loss of antigens from acrosome.  相似文献

The reactivity of antibodies with dimeric and monomelic peptide antigens was compared by ELISA. A panel of highly purified synthetic peptides of HIV-1 representing defined regions, 598–609 and 524 533 (fusion domain) of gp41 and 306–320 of gpl20, were used as antigens in the ELISA. These peptides were selected and synthesized taking into account the level of sequence conservation of various strains and hydrophilicity. The analysis included sera from 52 HIV-1 infected individuals and 53 HIV-1 negative controls. Both peptides from gp41 were found to be particularly immunoreactive with sera from HIV-1 infected individuals. The frequency of reactivity to the selected peptide from gp120 (V3 loop) in infected individuals was 82%. An interesting observation was that the dimeric peptide antigens had a detection rate more than 4-fold higher than the monomeric antigens. We found that lower levels of antibodies could be detected with dimeric antigens. The peptides reacted with few sera other than HIV-1 positive sera. These results implicate the potential dimeric peptide antigens to be utilized in the serodiagnosis of HIV-1 infection.  相似文献

Abstract An in vitro assay involving the adherence of Bordetella pertussis to Vero cells was used to quantify the inhibitory effects of monoclonal antibodies (McAb) on adherence. McAbs to agglutinogens (fimbriae), filamentous haemagglutinin (FHA), lymphocytosis promoting factor (LPF) and to an X-mode specific outer membrane protein were tested. X-mode cells adhered to the Vero cells to a greater extent than C-mode cells. McAb to agglutinogen 2 inhibited the adherence of B. pertussis bearing homologous agglutinogen. McAbs to LPF, FHA, agglutinogen 3 and X-mode specific outer membrane protein also partially inhibited the adherence of B. pertussis to Vero cells.  相似文献

Gram negative bacteria have lipopolysaccharides (LPS) that are critical for their survival. LPS molecules are composed of antigenic exopolysaccharide chains (O antigens). We are interested in discovering the enzymes involved in the biosynthesis of O antigens in Pseudomonas aeruginosa. The common polysaccharide antigen contains α-linked d-rhamnose residues. We have now synthesized GDP-d-rhamnose by a convenient synthesis in aqueous solution, and have shown that it can be used without extensive purification as the donor substrate for d-rhamnosyltransferase (WbpZ) from the P. aeruginosa strain PAO1. The availability of this nucleotide sugar preparation allows for characterization of d-rhamnosyltransferases.  相似文献

Abstract In a study of 718 women referred for specialist investigation for recurrent urinary tract infections, 250 (34.8%, P <0.01) were non-secretors. The proportion of non-secretors among the women with renal scars (42.6%) was higher than that found for women with no evidence of renal scars (36.6%). Among 29 patients in whom symptoms began in childhood or adolescence, 51.7% were non-secretors. The proportion of non-secretors among individuals with renal scars in this study (42.6%) and that reported in the accompanying paper for Swedish children (40%) suggests that non-secretion might influence the pathogenic sequelae of these infections. Possible host-parasite interactions underlaying the increased proportion of non-secretors among women with recurrent urinary tract infections and those leading to development of renal scars are discussed.  相似文献

Summary. Two monoclonal antibodies (mAbs), CC17 and IL-A67, which are specific for the bovine equivalent of the CD5 antigen, Bo5, were each found to react with the cells of some animals but not others. The cattle tested were all positive for one or both of the mAbs, but the level of expression on cells expressing both determinants was slightly lower than that on cells expressing either of the determinants on their own. Both mAbs precipitated an antigen of 67kD. However, sequential immunoprecipitation experiments with cells that reacted with both mAbs demonstrated that the determinants are present on two different sets of molecules. These findings suggested that the mAbs recognize two co-dominantly expressed allelic forms of Bo5. This was confirmed in family studies, with groups of full- and half-sibling offspring of sires and dams of defined phenotypes. These experiments also showed that the gene encoding the Bo5 antigen is not linked to the major histo-compatibility complex (MHC). The frequencies of the two alleles, which have been designated Bo5.1 and Bo5.2, in the cattle populations tested were 100% and 0%, respectively, in Bos taurus, and 10% and 90%, respectively, in Bos indicus.  相似文献

目的:探讨乙型肝炎病毒(HBV)DNA载量与其血清标志物的相关性。方法:运用荧光定量聚合酶链反应(FQ-PCR)、酶联免疫 吸附实验(ELISA)分别检测503 例患者HBV-DNA 载量和HBV 血清标志物。根据HBV 血清标志物结果分为大三阳组、小三阳 组、少见模式组、抗体阳性及全阴组,比较各组间HBV-DNA的阳性率及定量值。结果:在大三阳组、小三阳组、少见模式组、抗体 阳性及全阴组HBV-DNA 的阳性率分别为90%、65.1%、65.2%、2.0%,HBV-DNA 的定量结果(logHBV-DNA) 别为6.32± 1.96、 2.01± 1.68、3.48± 2.52 (抗体阳性及全阴组阳性例数过低,不纳入统计)。大三阳组HBV-DNA的阳性率显著高于小三阳组(P<0. 05),大三阳组、小三阳组HBV-DNA 的阳性率与少见模式组比较,差异均无统计学意义(P>0.05),但大三阳组、小三阳组、少见模式 组HBV-DNA 的阳性率均显著高于抗体阳性及全阴组(P<0.01)。HBsAg、HBeAg 阳性组HBV-DNA 的阳性率分别显著高于HBsAg 、HBeAg 阴性组(P<0.01)。小三阳组、少见模式组HBV-DNA 载量均显著低于大三阳组(P<0.01),少见模式组HBV-DNA 载量 显著高于小三阳组(P<0.05)。结论:HBV-DNA的阳性率与HBeAg、HBsAg相关;HBV-DNA载量与HBV 血清标志物模式相关。  相似文献

Epizootiological surveys of Leucocytozoon caulleryi infection in chickens in Japan, Taiwan, Philippines, Singapore, Malaysia and Thailand were undertaken by means of the immunodiffusion test. The rate of infection of L. caulleryi confirmed by the examination of parasites in the peripheral blood of chickens coincided with that of positive antibody response in the immunodiffusion test. Antibodies against L. caulleryi were found in chickens in all the countries surveyed in the present investigation. The prevalence of L. caulleryi infection in chickens was confirmed by the immunodiffusion test. Several chickens in each country showed the presence of serum antigens of L. caulleryi at the times of serum sample collection. These results seemed to indicate that the immunodiffusion test is a method efficient enough to be applicable to the epizootiological surveys and diagnosis of L. caulleryi infection in chickens in the field. As a result, the antibodies or soluble antigens in the sera of chickens infected with L. caulleryi present, respectively, in each country may have the same immunological characters.  相似文献

Recent approaches toward the immunotherapy of neoplastic disease involve the introduction of expression-competent genes for interleukin-2 (IL-2) into autologous malignant cells. Treatment of tumor-bearing experimental animals with the IL-2-secreting cells successfully induces partial and at times complete remissions. In most instances, however, although delayed, progressive tumor growth continues. Here, certain of the characteristic of B16 melanomas (H-2b) persisting in C57BL/6 mice (H-2b) treated with an IL-2-secreting, melanoma-antigen-positive cellular immunogen (RLBA-IL-2 cells) are described. Unlike the melanoma cells first injected, B16 cells recovered from mice treated with RLBA-IL-2 cells were deficient in the experssion of MHC class I, but not class II determinants. Deficient MHC class I expression correlated with the cells' resistance to cytotoxic T lymphocytes (CTL) from the spleens of mice immunized with RLBA-IL-2 cells. Melanomas persisting in mice treated with non-IL-2-secreting, melanoma-antigen-positive cell constructs (RLBA-ZipNeo cells) were also deficient in the expression of MHC class I determinants, and the melanoma cells were resistant to CTL from mice immunized with RLBA-ZipNeo cells. Thus, the expression of melanoma-associated antigens rather than IL-2-secretion correlated with deficient MHC class I expression by the persistent melanomas. This point was substantiated by the expression of MHC class I antigens by melanomas persisting in mice treated with IL-2-secreting, melanoma-antigen-negative LM cells (LM-IL-2); it was equivalent to that of melanomas in untreated mice. The involvement of MHC class I antigens in the immune resistance of persistent melanoma cells from mice treated with the melanoma-autigen-positive immunogens was indicated by the effect of interferon (IFN) orN-methyl-N-nitro-N-nitrosoguanidine (MNNG) on the susceptibility of the cells to anti-melanoma CTL. Treatment of the resistant melanomas with IFN or MNNG stimulated MHC class I antigen expression and restored the cells' sensitivity to CTL from mice immunized with IL-2-secreting or nonsecreting, melanoma-antigen-positive cellular immunogens. Prior treatment of the treated cells with antibodies to MHC class I determinants inhibited the cells' susceptibility to CTL from mice immunized with RLBA-IL-2 cells.  相似文献

Summary Subpopulations of normal bronchial epithelial cells were identified using a series of murine monoclonal antibodies. These antibodies were used to stain intact bronchial epithelial cells in culture by indirect immunofluorescence. LAM 2 reacted with 80%, LAM 6 with 75%, LAM 7 with 60%, and LAM 8 with 5% of these cells. Sections of human bronchial epithelium were also stained with these antibodies by immunoperoxidase methods. LAM 2 was found to bind with 80%, LAM 6 with 65%, LAM 7 with 50%, and LAM 8 with less than 1% of bronchial epithelial cells. LAM 2 stained both columnar epithelial cells and basal cells; LAM 6 stained mainly basal cells and only a small proportion of columnar cells; LAM 7 showed specificity for basal cells; LAM 8 distinctly stained single cells in the basal cell layer. These antibodies were previously shown to react with the surface membrane of human lung carcinomas, ranging from the broad reactivity of LAM 2 with small cell and non-small cell lung cancers to the highly restricted reactivity of LAM 8 with small cell carcinomas of the lung. Thus, membrane antigens have been identified in bronchial epithelial cells by monoclonal antibodies which exhibit a similar range of cellular reactivity in vitro as in vivo. Inasmuch as these antibodies recognize subsets of cells which could not be easily distinguished by morphologic characteristics, these reagents may be useful in classifying bronchial epithelial cells.  相似文献

Helminth parasites of teleost fish have evolved strategies to evade and manipulate the immune responses of their hosts. Responsiveness of fish host immunity to helminth antigens may therefore vary depending on the degree of host-parasite counter-adaptation. Generalist parasites, infective for a number of host species, might be unable to adapt optimally to the immune system of a certain host species, while specialist parasites might display high levels of adaptation to a particular host species. The degree of adaptations may further differ between sympatric and allopatric host-parasite combinations. Here, we test these hypotheses by in vitro exposure of head kidney leukocytes from three-spined sticklebacks (Gasterosteus aculeatus) to antigens from parasites with a broad fish host range (Diplostomum pseudospathaceum, Triaenophorus nodulosus), a specific fish parasite of cyprinids (Ligula intestinalis) and parasites highly specific only to a single fish species as second intermediate host (Schistocephalus pungitii, which does not infect G. aculeatus, and Schistocephalus solidus, infecting G. aculeatus). In vitro responses of stickleback leukocytes to S. solidus antigens from six European populations, with S. solidus prevalence from <1% to 66% were tested in a fully crossed experimental design. Leukocyte cultures were analysed by means of flow cytometry and a chemiluminescence assay to quantify respiratory burst activity. We detected decreasing magnitudes of in vitro responses to antigens from generalist to specialist parasites and among specialists, from parasites that do not infect G. aculeatus to a G. aculeatus-infecting species. Generalist parasites seem to maintain their ability to infect different host species at the costs of relatively higher immunogenicity compared to specialist parasites. In a comparison of sympatric and allopatric combinations of stickleback leukocytes and antigens from S. solidus, magnitudes of in vitro responses were dependent on the prevalence of the parasite in the population of origin, rather than on sympatry. Antigens from Norwegian (prevalence 30–50%) and Spanish (40–66%) S. solidus induced generally higher in vitro responses compared to S. solidus from two German (<1%) populations. Likewise, leukocytes from stickleback populations with a high S. solidus prevalence showed higher in vitro responses to S. solidus antigens compared to populations with low S. solidus prevalence. This suggests a rather low degree of local adaptation in S. solidus populations, which might be due to high gene flow among populations because of their extremely mobile final hosts, fish-eating birds.  相似文献

Abstract The location of lipoteichoic acid and 3 cell wall-associated protein antigens in fresh isolates of Streptococcus mutans (serotype c ) and in variants derived from these parental strains by repeated subculture in vitro has been examined by electrophoretic and dot-immunobinding techniques. Following subculture there was a marked shift in the distribution of all four antigens from the cell wall to the extracellular culture supernatants. It is therefore apparent that that the decreased hydrophobicity and the impaired ability of the subcultured variants to colonise in vivo, as observed by others, cannot be correlated with changes in a single surface molecule.  相似文献
