The coleoptile curvature bioassay of agar diffusates from successive cambial-region sections of Pinus silvestris L. stem tissue reveals the existence of a wave-like pattern, possibly of auxin translocation. Spectrofluorometric determinations of indolyl-3-acetic acid (IAA) in these diffusates gave only a poor correlation. Probably another phytohormone, also an auxin, plays a more important role in the pattern.
The amplification in the amplitude of the pattern, brought about by apically applied IAA, cannot be induced by the synthetic auxin, 2,4-dichlorophenoxyacetic acid.  相似文献

Summary A new gel-type support poly-N-vinylcaprolactam for microbial cell immobilization is presented. The method allows one to obtain beads of biocatalyst in a single step. The properties of beads obtained using different types of gel stabilizers were compared; the best stabilizer was found to be tannin. The method developed was used for entrapment of viable bacterial cells and fungal spores. A method is described for the routine determination of absolute amounts of free indolyl-3-acetic acid (IAA) in plant material. The time required for a determination is in the order of 2 hr. Several samples can be handled at the same time. Depending on the contents of IAA, the amount of fresh plant material varies from 0·05 to 5 g. Losses are corrected for by applying 14C-labeled IAA.  相似文献

We investigated the effect of γ-irradiation (4–50 Gy) of barley seeds (Hordeum vulgare L., cv. Nur) on the content of endogenous phytohormones–stimulators of plant growth and development: indol-3-acetic acid (IAA), indolyl-3-butyric acid (IBA), zeatin and abscisic acid (ABA). The ratio (IAA + IBA + zeatin)/ABA from the third to the seventh day of germination has been measured. It was shown that the changes in the content of phytohormones as a function of the radiation dose were nonlinear. In the dose range of 4–20 Gy, phytohormones balance was changed due to increased content of growth stimulators and decreased ABA content. Using a dose of 50 Gy led primarily to a decrease in the content of growth stimulators and an increase in ABA content, and the ratio (IAA + IBA + zeatin)/ABA shifted toward ABA content.  相似文献

The pH-driven accumulation of [3H]indolyl-3-acetic acid (IAA) has been found to occur in membrane vesicles of lupin (Lupinus albus L.) hypocotyls. Most of this association of auxin with membranes is very sensitive to osmotic shock, high concentrations of permeable weak acids, incubation at 20° C for 20 min and to some ionophores. Long incubation times also depress the ability to accumulate radioactive IAA but this ability can be partially restored by a treatment that presumably reconstitutes the pH gradient across the membranes. Two specific inhibitors of auxin transport, N-1-naphtylphthalamic acid and 2,3,5-triiodobenzoic acid, stimulate net IAA uptake with an optimum at about 10-6 M (pH 5.0). At least two auxin carriers appear to be present in the lupin membrane vesicles. An uptake carrier seems to be saturated at 10-7 M IAA in the presence of N-1-naphtylphthalamic acid, but higher IAA concentrations are needed to saturate an efflux carrier. A new ELISA method is proposed for differential quantitative determination of free (indolyl-3-acetic acid; IAA) and bound (indolyl-3-acetyl-L-aspartate) forms of natural auxins. There is similarity in results obtained by this and some traditionally used methods. The standard error of determination of the active form of IAA by our method is 1.5-2.0 times less than that using the traditional method. The method of quantitative differential determination of the main natural auxins does not require preliminary sample preparation, and this shortens assay time. The developed method has been used for practical determination of different forms of endogenous IAA in wheat and dandelion ovaries subjected to minimal treatment. This method can be used to investigate changes in the ratio of various hormonal forms of auxins that differ in their physiological activity in reproductive organs of angiosperms at various stages of reproduction.  相似文献

Cytochalasin B (CB) inhibits the elongation growth of maize roots, and that of wheat coleoptile segments incubated in indolyl-3-acetic acid, by over 30% after a lag period of about 60 min. This long lag is not due to poor tissue penetration by the inhibitor, but seems to reflect a property of the process inhibited by CB. The only visible ultrastructural change accompanying growth inhibition is the accumulation of secretory vesicles in the vicinity of dictyosomes, which occurs between 90 and 300 min. However, a massive accumulation of vesicles is seen after 120 min in root cap cells which possess very active dictyosomes. The results indicate that CB does not inhibit elongation growth by interfering with cytoplasmic streaming. S ummary : Micro-organisms isolated from rhizospheres and rhizoplanes of wheat plants, and from root-free soil, produced growth regulating substances with the properties of gibberellins and indolyl-3-acetic acid (IAA). Substances inhibiting extensions of pea plant internodes and lettuce hypocotyls were also produced, especially by bacteria from the root region of seedlings 6 days old. Bacteria producing growth promoting substances were most abundant on roots of older plants. Seedlings grown aseptically with added gibberellic acid (GA3) and IAA, or grown with a soil inoculum, developed similarly and differed in their morphology from those grown aseptically without additives.  相似文献

The extracellular peroxidase from cultures of Inonotus radiatus and of peanut (Arachis hypogeaL.) cells as well as the mycelial peroxidase from Trametes versicolor were used for studies of immobilizing this protein either by its free amino or its carboxyl groups. The immobilization process was carried out either on keratin proteins derived from feathers or on polyamide coated over silica gel. Coupling was established either through the free amino or carboxyl groups. In general the indolyl-3-acetic acid oxidase activity of fungal peroxidases exceeds that of peanut peroxidase. When the peroxidase of the three sources was immobilized on the matrices by the free amino groups, little if any effect on the IAA oxidase activity could be measured. However, immobilization through the carboxyl groups resulted in a drastic reduction of indole-3-acetic acid oxidase activity. Since identical amounts of peroxidase were linked in all cases, the loss of indolyl-3-acetic acid oxidase activity implies that the carboxyl group is essential for the active site.  相似文献

An in vitro bioassay system for floral bud formation has been established using Nicotiana tabacum L. cv. MC, explants excised from floral stalks cultured on modified Murashige-Skoog medium containing excess auxin and antiauxin. Three auxins. indolyl-3-acetic acid (IAA), 4-chloroindolyl-3-acetic acid and 5,6-dichloroindolyl-3-acetic acid, were tested for floral bud-forming activity; IAA was most efficient. Three antiauxins. 5,7-dichlorindolyl-3-isobutyric acid (5,7-Cl2-HBA), p -chlorophenoxyiso butyric acid and 2,3,5-triiodobenzoic acid, were tested for the ability to reverse the inhibition of floral bud formation caused by excess IAA. Only 5,7-Cl2-HBA was very effective. Leaf exudates from short day- and long day-treated tobacco plants were added to the bioassay system which contained 1 µ M 6-benzylaminopurine, 5 µ M IAA and 5 µ M 5, 7-Cl2-HBA, Interestingly, only the leaf exudate from short day-treated plants stimulated floral bud formation. Moreover, the buds produced grew into large. well-developed ones with shapes similar to those of the natural floral buds of intact tobacco plants.  相似文献

Summary Abscisic acid (ABA) moves basipetally and laterally in maize (Zea mays L.) root segments placed horizontally; its transport properties are thus similar to those of the growth-inhibiting substances produced by the root cap. We studied the effects of auxin (indolyl-3 acetic acid) on formation of the primary polarity axis in zygotes of the brown algae Fucus vesiculosus. Within the first 2.5 h after fertilization, the zygotes release this phytohormone in the environment. The treatment of developing zygotes with the inhibitor of indolyl-3-acetic acid transport from the cell triiodobenzoic acid at 5 mg/l arrests the auxin secretion and leads to its accumulation in the cells. This causes a significant delay in zygote polarization. The treatment of zygotes with the exogenous indolyl-3-acetic acid at 1 mg/l stimulates cell polarization and formation of a rhizoid process. When auxin was added to the medium with triiodobenzoic acid, the inhibitory effect of the latter was fully relieved. It has been proposed that the content of indolyl-3-acetic acid in the environment is a key factor in the induction of polarity of the F. vesiculosus zygotes.  相似文献

Elongation, indolyl-3-acetic acid (IAA) and abscisic acid (ABA) levels, – gas chromatography-mass spectrometry quantification –, in the elongating zone were analysed for maize ( Zea mays L., Cv. LG11) roots immersed in buffer solution with or without zeatin (Z). The effect of Z depends on the initial extension rate of roots. The slower growing roots are more strongly inhibited by Z (10−7−10−5 M ) and they show a greater increase in IAA and ABA content. When compared to the rapidly growing roots, the larger reactivity of the 'slow'ones cannot be attributed to a higher Z uptake as shown when using [14C]-Z. It is suggested that Z could regulate root elongation by acting on the IAA and/or ABA level. The comparative action of these two hormones is discussed.  相似文献

Naphthalene-1-acetic acid (NAA) binding by membrane fractions derived from maize has been re-evaluated. Using a computer curve-fitting procedure only one major type of NAA binding, in terms of binding affinity, could be identified. Auxins, antiauxins and structurally related compounds have been tested for their competitive effect on NAA binding and the inhibitor constants for a number of these have been determined. Extracts from various plant species have been examined for their NAA binding ability, but all showed much less binding than maize leaf or coleoptile preparations. Endogenous contents of indolyl-3-acetic acid (IAA) and abscisic acid (ABA) were quantified in excised roots of Catasetum fimbriatum (Orchidaceae) cultured in vitro on solidified Vacin and Went medium with 1, 2, 4, 6, 8 and 10 % sucrose, as well as 2 % sucrose plus mannitol. Maximum root growth was observed in media with 4 % sucrose and 2 % sucrose plus 2.2 % mannitol, suggesting that a moderate water or osmotic stress promotes orchid root growth. Contents of both ABA and IAA increased in parallel to increasing sucrose concentration and a correlation between root elongation and the ABA/IAA ratio was observed. Incubating isolated C. fimbriatum roots with radiolabeled tryptophan, we showed an accumulation of IAA and its conjugates.  相似文献

Enterobacter cloacae was found to be associated with the pollen of several Mediterranean pines. The bacterium was detected only in mature pollen of Pinus halepensis, P. brutia, and P. pinea. E. cloacae is considered to be an obligatory endophyte based on its occurrence in disinfected male cones and the successful inoculation of seedlings of the above 3 species with E. cloacae AS1 isolated from pollen of P. halepensis used as a model strain. Strain AS1 was able to produce indolyl-3-acetic acid (IAA) from L-tryptophan in culture, and this was probably the source of the increased IAA content in the germination medium of pollen. In addition, strain AS1 promoted adventitious root formation in mung bean (Vigna radiata) cuttings. However, it was not possible to obtain bacterium-free pollen to elucidate its role in pollen germination.  相似文献

Differential growth of the nodal regions of graviresponding Tradescantia fluminensis (Wandering Jew) was analysed with special respect to the extension-restricting epidermal cells of the opposite growing and growth-inhibited organ flanks. Gravicurvature of horizontally gravistimulated isolated nodes depends on auxin (indolyl-3-acetic acid, IAA) and shows a node-specific profile in which the third node below the tip showed the greatest response. Exogenously supplied gibberellic acid induced no gravitropic growth. Vertically oriented isolated nodes supplied with exogenous IAA showed, on an electron microscopical level, conspicuous membrane invaginations with adjacent wall depositions restricted to the outer tangential epidermal cell walls. Their number was more than doubled by exogenously supplied Ca2+, which inhibited IAA-induced growth. No such changes could be detected in water-incubated segments or inner tissues of IAA-supplied segments. Gravistimulated differential growth of nodes of intact shoots and of nodal segments was characterized by changes similar to the ones induced by exogenous IAA, with greatly increased numbers of wall depositions within the epidermal cells of the growth-inhibited upper organ flank. Similar to the gravistimulated wall depositions, an asymmetric distribution pattern of Ca2+ was detected in the epidermal cell walls employing x-ray energy spectrum analysis (EDX). The results indicate that growth of nodes of Tradescantia fluminensis is regulated via IAA-induced secretion and subsequent infiltration of wall components enabling wall extension. The data support the hypothesis that temporary differential growth during gravicurvature of Tradescantia fluminensis is mediated by the antagonistic effect of Ca(2+)-ions on the infiltration of IAA-induced wall-loosening components into the outer, extension-restricting epidermal walls thereby inhibiting growth.  相似文献

Epidermal strips from well-watered faba-bean plants were subjected to a range of abscisic acid (ABA) and indolyl-3-acetic acid (IAA) concentrations (10-5 to 1 mM) in the presence or absence of CO2 in light or dark. ABA had inhibitory effect on abaxial stomatal apertures in all the concentrations studied and retained them closed even after addition of KCl (SO and 100 mM) to the incubation medium. It also influenced stomatal responses to CO2. In the presence of CO2 apertures were greater than in its absence in light as well as in darkness. This relationship remained unchanged also after addition of KCl. The action of ABA inhibited accumulation of potassium in the guard cells. IAA stimulated stomatal opening and its effect was quite opposite to ABA; in the presence of CO2 the apertures were smaller than in its absence. IAA, however, was able to inhibit the closing effect of darkness, CO2, and ABA, and stimulated potassium accumulation in the guard cells. Simultaneous action of ABA+IAA manifested effects of both substances.  相似文献

The ability of the actinomycetes and coryneform bacteria isolated from the root tissues of winter rye to produce auxin in a liquid culture was studied. The isolates of coryneform bacteria produced indolyl-3-acetic acid (IAA) into the medium in the amount of 9.0–95.0 μg/ml and the isolates of actinomycetes in the amount of 39.5–83.0 μg/ml. The maximal IAA accumulation in culture liquid of actinomycetes coincided, in general, with the beginning of the stationary growth phase. The dependences of IAA synthesis by actino-mycetes on the composition and pH of nutrient medium, tryptophan concentration, and aeration conditions were determined. Biological activity of the bacterial IAA was assessed. Treatment of winter rye seeds with coryneform auxin-producing bacteria increased the germination capacity and enhanced an intensive seedling growth in vitro.  相似文献

Callus derived from hypocotyls of periwinkle, Catharanthus roseus, responded to culture on nutrient media supplementedwith IAA, BA, and zeatin with shoot formation at low frequencies. However, shoot regenerating callus could be very successfully propagated and subcultured. Alkaloid profiles of callus derived from the original explants (hypocotyls) as well as callus derived from regenerated shoots were almost identical. Subcultures of old callus (initiated in 1978) failed completely to grow shoots.
