A total of 286 genotypes were collected from 39 countries of the world and were evaluated to determine the phenotypic diversity
for 17 quantitative traits. Higher degree of coefficient of variation were recorded for grain yield−5 (52.46%), biomass−5 (45.73%), fresh pod width-10 (47.24%), dry pod weight−10 (40.33%), plant height−1 (35.25%), harvest index (32.70%) and number of branches−5. Cluster-II clearly reflected that late genotypes were having lightest pods weight, shortest pod width, and pod length; low
grain yield, biomass and harvest index. Width, and pod length; low grain yield, biomass and harvest index. While genotypes
in Cluster-III were in contrast to Cluster-II having heaviest pods weight, longest pods width and length, highest grain yield,
biomass and harvest index. Higher PC−1 values have been determined for days to flower initiation which consequently were contributing weighed positive to days to
pods picking, days to flower completion, days to dry pod appearance, days to plant harvesting while negatively contributed
to yield producing traits, indicating that late flowering pea germplasm emphasizes more on the vegetative growth and was low
yielding. However, higher PC−2 values have been obtained for number of branches−5, grain yield and biomass while lower values for days to flowering, days to pods picking, days to flower completion, days

A 2–8-fold increase in the activity of glutamate dehydrogenase (GDH), accompanied by an alteration of the GDH isoenzyme pattern, was observed in detached pea shoots floated on tap water (preincubated shoots). Sugars supressed the process, whereas NH
+

and various metabolites as well as inhibitors of energy metabolism and protein synthesis were ineffective. The subcellular distribution pattern revealed evidence that the GDH isoenzymes are exclusively located in the mitochondrial matrix. The alterations in GDH activity occurring in preincubated shoots are restricted to the mitochondria.An experimental device suitable for studying the GDH function in isolated intact mitochondria has been established. Allium sativum germplasm collected from different parts of India was subjected to critical analysis of 14 easily noticeable morphological traits. Variability encountered in quantitative as well as qualitative, bulb and plant traits was documented. -Mannosidase and ß-N-acetylglucosaminidase were purifiedfrom extracts of cotyledons of germinating Pisum sativum L.A 13-fold purification of a-mannosidase free from ß-N-acetylglucosaminidaseactivity was achieved by precipitation in ammonium sulphate,column chromatography on DEAE-cellulose, and treatment with2 M pyridine. ß-N-Acetylglucosaminidase was purified200-fold by the use of (NH4)2SO4, and chromatography on ConcanavalinA1-Sepharose and Sephacryl-200. This preparation showed no measurablecontamination by -mannosidase activity. Both glycosidases appearto be glycoproteins and demonstrate optimal activity at pH valuesof 4.04.5. Both glycosidases appear to have very similarmolecular weights, with -mannosidase being slightly larger thanß-N-acetylglucosaminidase. Summary The possibility of obtaining instant pure breeding lines by matromorph seed development in Pisum sativum L. has been investigated. Two types of maternal parents, namely, homozygous for the recessive marker genes and heterozygous for the dominant marker genes were pollinated with Lathyrus odoratus and the P174 variety of Pisum sativum L. carrying dominant markers. For both pollinators, induction of matromorphy by prickle pollination, irradiated pollen and IAA treatment was examined. Promising matromorphs were identified in the M1 generation which were studied in the M2 generation for assessing their genetic status with respect to homozygosis. The success of pod set varied from zero to 28% with a varying number of matromorphic seeds following different treatments. The possible mechanisms for matromorphic origin have been discussed. The evidence presented herein favours induction of matromorphy in peas for the production of homozygous stocks. In addition, the recovery of double recessive seed markers of the maternal parents along with plant markers from the paternals has prospective implications in plant breeding as an alternative tool to recurrent back crossing. 相似文献

Pisum sativum L. is one of the plant species where infraspecific genome size variation, up to 1.29-fold between cultivars, has been reported. The present investigation deals with a Feulgen cytophotometric analysis of this phenomenon in 25 wild accessions, landraces, and cultivars of widely different geographic origin. Differences between accessions were maximally 1.054-fold in single experiments but proved to be nonreproducible upon repeated measurements. Seedlings of the same accession often differed significantly, up to 1.056-fold, but values from root and shoot tips in one individual were not significantly correlated, indicating the absence of true genome size variation between plants. Upon calibration against Allium cepa a 1C value of 4.42 pg is estimated for Pisum sativum. Altogether the data suggest that, contrary to the divergence in the literature data and recent reports on DNA content variation, the pea has a stable genome size. 相似文献

The possible role of C2H4 metabolism in mediating the responses of plants to C2H4 is re-examined. It is demonstrated that (i) the effects of inhibitors upon C2H4 action do not correspond with their effects on metabolism, (ii) elicitors of C2H4 effects do not have appropriate effects on C2H4 metabolism, (iii) inhibitors of C2H4 metabolism do not affect the response of plants to C2H4. It is concluded that metabolism of C2H4 is not linked to the mode of action of the growth regulator.Abbreviations DTC

The physiological and biochemical consequences of treating Le (tall) and le (dwarf) pea seedlings with varying quantities of the gibberellins [3H]GA20 and GA1 have been investigated. Although the percentage uptake of these compounds from the site of application on the 3 stipules was low and most of the applied GA remained unmetabolised in situ, the quantitative relationship between GA translocation and GA dosage was found to be linear for GA1 but saturating for GA20. The movement of the GAs and their subsequently produced metabolites was mainly acropetal. They accumulated in greatest quantity in the apical extremities of the shoot. Overall, the extent to which GA20 was metabolished in le seedlings was considerably less than in Le pea seedlings. Although all le tissues contained significantly less [3H]GA1 than their Le counterparts, phenotypic effects of the le mutation were apparent only on internode and tendril development. Increased tissue growth, consequent upon GA treatment, was also apparent only in the internodes and tendrils of le plants. For internodes, GA1 content determined the mid-logarithmic-phase growth rate and, consequently, final length. For tendrils, GA20 rather than GA1 may be the primary stimulatory agent.Abbreviations GA

The evolution of the total amount of DNA in epicotyls and of the amount of DNA per cell nucleus in epicotyl cortex cells during germination was followed in two closely related pea varieties, Pisum sativum cv. Finale and Pisum sativum cv. Rondo. Under etiolating conditions, growth of the cv. Rondo occurs only by cell elongation which is preceded by endomitotic DNA synthesis, while in the cv. Finale growth is the result of cell elongation accompanied by endomitotic DNA synthesis and cell division. The maximum C-level attained in both cultivars under etiolating conditions is 8 C (C=haploid amount of DNA in a gamete cell). Both the maximum C-level reached and the percentage of cells reaching this C-level seem to be under strict genetic control. In both cultivars, light inhibits the endomitotic DNA replication.Neither gibberellic acid (GA3), nor AMO 1618 alter the maximum C-level or the percentage distribution of the C-classes. Both growth regulators are effective, although in an opposite way, only in tissues where cell division occurs or where endomitotic DNA synthesis is blocked, as in light-grown pea epicotyls. 相似文献

This work evaluates the (cor-)relations between selected biochemical responses to toxic Cd and the degree of Cd sensitivity in a set of pea genotypes. Ten genotypes were analysed that differ in their growth response to Cd when expressed as root or shoot tolerance indices (TIs). Concentrations of non-protein thiols (NPTs) and malondialdehyde (MDA), activity of chitinase, peroxidase (POX), and catalase significantly increased in all pea genotypes treated with Cd. Cd-sensitivity of genotypes was correlated with relative increases in MDA concentration as well as activities of chitinase and POX, suggesting similar Cd stress effects. Activities of ascorbate peroxidase (APX) decreased, but concentrations of glutathione (GSH) increased in the less Cd-sensitive genotypes. Differences in root and leaf contents of Cd revealed no correlation with TI, metabolic parameters, and enzyme activities in Cd-treated plants, respectively, except that shoot Cd concentration positively correlated with shoot chitinase activity. Toxic Cd levels inhibited uptake of nutrient elements such as P, K, S, Ca, Zn, Mn, and B by plants in an organ- and genotype-specific manner. Cd-sensitivity was significantly correlated with decreased root Zn concentrations. The results show both similarities, as well as distinct features, in Cd toxicity expression in genotypes of one species, suggesting that independent and multi-factorial reactions modulate Cd sensitivity on the low-tolerance level of plants. As a prelude to domestication efforts, variation of agronomic traits was determined among accessions of the polytypic, xerophytic cucurbit, coyote gourd [Cucurbita digitata subsp. digitata (DIG), palmata (PAL), cylindrata (CYL), and cordata (COR)] and among and within their progeny. Oil content in 60 accession seed lots (x = 27.8%, CV 21.4%) was more variable than protein content (x = 33.1%, CV = 13.5%). Punicic acid (c,t,c-9,ll,13-octadecatrienoic acid) levels in seed oils were also variable (x = 12.0%, CV = 20.6%) among accession seed lots. Substantial differences among and within subspecies were noted in 40 progeny lines for fruit/plant (x = 55.2, CV = 47.5%), seed wt/plant (x = 0.89 kg, CV = 51.1%), seed wt/fruit (x = 17.4 g, CV = 39.8%), seed no./fruit (x = 356, CV= 30.8%), 100-seed wt (x = 4.8 g, CV = 21.6%) and fruit diameter (x = 77 mm, CV 4.8%). Correlations among parameters suggested selection for fruit/plant to be the most effective primary strategy for seed yield improvement, but among high fruit-yielders, selection for seed wt/fruit was also important. The two subspecies PAL and CYL exhibited superior seed wts/plant. CYL types matured high fruit-loads, but their fruits were smaller and contained a smaller number of lighter seed than PAL or PAL hybrids. In contrast, PAL and PAL hybrids displayed lower fruit-yields/ plant but their fruits were larger and contained higher seed wts/fruit than their CYL counterparts. Root wt/plant was also variable (x = 1.55 kg, CV = 63.2%). Roots of DIG were larger and less branched than those of other subspecies. Proximate and liquid Chromatographic analyses of selected accession seed lots and controlled crosses failed to reveal advantages for the inclusion of specific types in a breeding program for the development of high oil or high/low punicic acid lines. 相似文献

Summary Soluble proteins of pea seed were investigated by quantitative immunological methods. Vicilin, legumin, pea seed lectin (PEA), 26 albumins and a globulin (B1) were detected and observed during seed development, germination and under different extraction and fractionation procedures. Vicilin and legumin were found to be immunologically distinctly different. Legumin was found to be comprised of two similar proteins, Legumin species I and II. Vicilin, but no legumin, was detected in the embryonic axis.Three albumins, B1 and PEA were found to be synthesized after the onset of legumin synthesis.Among the pea lines investigated, one line exhibited distinct differences with respect to the albumins and PEA.Some observations indicate that PEA might interact with other seed proteins of pea. 相似文献

Summary A half diallel analysis involving nine cultivars showed that additive as well as non-additive gene effects were important for the inheritance of seed yield per plant, 100-seed weights, protein content and potassium per cent. For remaining traits non-additive genetic components were important. Overdominance was observed for all traits except for 100-seed weight, which expressed partial dominance. Parents PMR-T10, EC21857, EC109182, T163 and EC109189 were good general combiners for seed yield, seed weight and quality traits. In general there was a good relationship between per se performance and the gca effects of the parents for all traits. Cross combinations such as LMR8 x EC109182,LMR8 x PMR-T10,LMR8 x EC21857,PMRT10 x EC21857 and P23 x EC21857 were found promising. The seed yield was positively correlated with other quality traits. Protein had a positive correlation with methionine and phosphorus. All the values of correlation co-efficients were non-significant except for yield with potassium, 100-seed weight and protein with methionine, indicating that yield and quality attributes can be improved simultaneously by simple selection procedures. 相似文献

Some properties of ornithine carbamoyltransferase from chloroplasts isolated from leaves of Pisum sativum L. (cv Marzia) were compared with those of the enzyme partially purified (316-fold) from shoots of seedlings after 3 weeks of cultivation.
Both preparations showed a pH optimum at pH 8.3 and had the same affinity to ornithine (Km = 1.2 millimolar) as well as to carbamoyl phosphate (Km = 0.2 millimolar). The approximate molecular weight determined by gel sieving was 77,600.
A desalted ammonium sulfate precipitate from 14-day seedlings (inclusive roots and senescing cotyledons) was applied on a column of anion exchanger. The elution pattern showed one peak of ornithine carbamoyl-transferase activity. This elution pattern was the same as observed for the enzyme from chloroplasts.
The results suggest the presence of one form of ornithine carbamoyl-transferase in pea seedlings.

Glutamate dehydrogenase [l-glutamate:NAD+ oxidoreductase (deaminating) EC 1.4.1.2]has been purified 487-fold from pea stem mitochondria. The enzyme has a specific activity in the presence of 1 mm CaCl2 of 54 Enzyme Commission (EC) units. Calcium, manganese, and zinc ions activate the reductive amination reaction. The [Ca2+]0.5 for activation by calcium is 9 μm. The extent of activation by calcium changed during purification and storage. The oxidative deamination was slightly inhibited by calcium. The pH optimum for the reductive amination reaction was 8.0 and for the oxidative deamination was 9.2. At pH 8.0 and in the presence of 1 mm CaCl2 with the ionic strength held constant the enzyme showed normal kinetics for the reductive amination reaction. Under identical conditions except for the absence of CaCl2 the oxidative deamination reaction showed normal kinetics for glutamate. There was substrate activation at high NAD+ concentrations and these concentrations were avoided in the kinetic analysis. A steady-state kinetic analysis showed that a simple mechanism could not be in effect and a partially random mechanism is proposed. 相似文献

Essential oils obtained by hydrodistillation from different organs (flowers, leaves, stems and roots) of Tunisian coriander (Coriandrum sativum L.) was analyzed. GC and GC-MS analysis enabled us to identify 64 compounds and revealed great qualitative and quantitative differences between the analyzed parts. In all organs, the main compound was (E)-2-dodecenal, followed by (E)-2-tridecenal, gamma-cadinene, (Z)-myroxide, neryl acetate and eugenol. Multivariate analysis (PCA) revealed a high similarity in the essential oils composition between upper leaves and flowers in one hand and basal leaves, roots and stems on the other hand. Pyruvate dehydrogenase complex is associated with intact chloroplasts and mitochondria of 9-day-old Pisum sativum L. seedlings. The ratio of the mitochondrial complex to the chloroplast complex activities is about 3 to 1. Maximal rates observed for chloroplast pyruvate dehydrogenase complex activity ranged from 6 to 9 micromoles of NADH produced per milligram of chlorophyll per hour. Osmotic rupture of pea chloroplasts released 88% of the complex activity, indicating that chloroplast pyruvate dehydrogenase complex is a stromal complex. The pH optimum for chloroplast pyruvate dehydrogenase complex was between 7.8 and 8.2, whereas the mitochondrial pyruvate dehydrogenase complex had a pH optimum between 7.3 and 7.7. Chloroplast pyruvate dehydrogenase complex activity was specific for pyruvate, dependent upon coenzyme A and NAD and partially dependent upon Mg2+ and thiamine pyrophosphate. 相似文献

The organelles of soybean (Glycine max (L.) Merr.) protoplasts were separated using a recently developed procedure which allows rapid (3-h) recovery of a fraction enriched for coated vesicles (CVs). As determined by marker-enzyme enrichment and ultrastructural analysis of isolated membrane fractions, endoplasmic reticulum, Golgi membranes, glucan-synthase-II (EC 2.4.1.34)-containing membranes (putative plasma membrane), mitochondria, and CVs were enriched in separate fractions in a sucrose density gradient. Glucan synthase I (EC 2.4.1.12) had the highest specific activity in the Golgi-enriched and CV-enriched fractions and was found to comigrate with CVs upon rate-zonal centrifugation of a CV-enriched fraction. For further elucidation of the role of these latter organelles in cell-wall regeneration, freshly isolated protoplasts were pulsed with [3H]glucose for 20 min, and the disappearance of label from the organelles was followed for the ensuing 1 h. Although a CV-enriched fraction contained glucan synthase I, it contained very small amounts of labelled polysaccharide during the period of study. Pulse-chase experiments with [3H]glucose helped to confirm the role of the Golgi apparatus in secretion of matrix polysaccharides by protoplasts.Abbreviations CV(s)
