The oxygen uptake rate of aggregated mycelia is decreased to an extent which, in the case of a typical spherical aggregate, could be estimated depending on its diameter, mycelial density, oxygen diffusivity, and so forth. Equations were presented in this paper to evaluate the oxygen uptake rate of an mold pellet. A favorable agreement was found between the calculation and the experiment.  相似文献

The amino acid pool of yeast cells, Saccharomyces cerevisiae, incubated with galactose remains at a constant level for 100 minutes. This is 30 minutes beyond the time at which the oxidative phase of the induced-enzyme formation begins. Washed yeast cells, the pools of which have been depleted 60 per cent by incubation with glucose, do not replenish their pools as do washed cells incubated without a substrate. These facts indicate that the induced enzymes are formed at least partially from pool-replenishing amino acids. The time of onset of pool depletion is the time at which the aerobic fermentation phase of induced-enzyme formation begins for cells incubated with galactose. With 0.1 per cent galactose the respiratory phase begins at 100 minutes but no aerobic fermentation nor pool depletion occurs. The rates of respiration and aerobic fermentation are constant for four glucose concentrations from 0.1 to 1.0 per cent. The amount of aerobicfermentation is proportional to the initial concentration of glucose. Amino acid pool depletion occurs for all concentrations but depletion ceases and is followed by pool replenishment after aerobic fermentation is complete. Ultraviolet radiations, which delay the appearance of the respiratory phase of induced-enzyme formation, completely eliminate both the appearance of aerobic fermentation and pool depletion. The results indicate an intimate association between aerobic fermentation and amino acid pool depletion.  相似文献

为了提高酵母菌麦角固醇的产量,采用摇瓶培养法,对筛选出的一株酵母菌YN2产麦角固醇发酵条件进行了研究。结果表明,酵母菌YN2产麦角固醇适宜的培养基配方为:酵母粉1%,牛肉膏2.5%,葡萄糖8%,K2HPO4 0.3%,MgSO4 0.15%,该菌株产麦角固醇最适培养条件为:培养温度28℃,起始pH6.5,发酵时间72h。在优化的实验条件下,麦角固醇含量可达2.2%,100ml发酵液中麦角固醇产量达25.30mg。  相似文献

该研究采用耐高温型酵母,在不同葡萄糖浓度(5%~30%wt)下进行了乙醇间歇发酵的动力学研究,确定了适合该酵母的最佳底物浓度范围为16%~20%(wt)。同时选取合适的动力学模型,通过实验数据的非线性性拟合,得出了不同底物浓度下对应的动力学参数值,并分析了各动力学参数值随底物浓度增加而变化的趋势。结果显示,该酵母的最大比生长速率μmax随着葡萄糖浓度的增加而有所降低,且呈线性关系:μmax=0.3161-4.1820×104s(100g/L相似文献

固定化细胞和固定化酶一样,作为固液两相的非均相催化反应系统,其反应速度无疑受到内外扩散的影响。世界各国的化学工程学者在固定化酶反应动力学方面研究得较多,固定化细胞反应动力学研究虽有些报道,但主要集中在以海藻酸钙为载体的固定化系统,且载体形状为球形颗粒。对于其他材料的不同形状颗粒载体的固定化细胞动力学研究报道较少。陶瓷作为固定化细胞载体是我们研究的一种固定化细胞的新型载体,在机械强度,孔径大小,细胞与之结合牢固度及其稳定活性,再生性能方面有其特有的优越性[10]。为了给在生产实践中使用这种新型载体提供理论依据,本文采用球型陶瓷和拉西环陶瓷为载体固定化酵母,对它们的发酵动力学进行了比较研究。  相似文献

In cells of Saccharomyces cerevisiae growing aerobically for 24 hr, acetyl-coenzyme A synthetase [acetate: CoA ligase (AMP), EC 6.2.1.1] was localized principally in the microsomal fraction. On density gradients, the enzyme in such cells behaved as a low-density particle, readily separable from the soluble proteins. After 48 hr of incubation, the cells showed a bimodal distribution of enzyme, with most of the activity now sedimenting with the mitochondrial fraction and only a smaller amount with the microsomal fraction. By using density gradients, two forms of synthetase were obtained from these cells: one band denser and the other band less dense than the intact mitochondria. In all preparations containing synthetase activity, appreciable levels of phospholipids were also detected.  相似文献

In the polyol fermentation by Candida zeylanoides KY6166, which occurred preferentially by keeping the pH of medium at acidic side (below 4.0), phosphate ion played a precise role in the conversion of erythritol fermentation to d-mannitol fermentation. Enzymatic studies on the conversion mechanism provided the following evidences.

The enzymes involved in pentosephosphate cycle were considerably depressed in polyol production phase in which intracellular pH ranged from 5.5 to 5.7. Particularly transaldolase responsible for the synthesis of erythrose 4-phosphate and fructose 6-phosphate from glyceraldehyde 3-phosphate plus d-sedoheptulose 7-phosphate was significantly depressed at pH 5.5. Besides, transketolase which participated directly in the formation of erythrose 4-phosphate from fructose 6-phosphate was significantly inhibited by phosphate ion. Glucose 6-phosphate dehydrogenase was slightly inhibited by phosphate ion.

This paper presents a modified random network model to illustrate how groups can form in the absence of evolutionary forces, assuming groups are collections of entities at any level of organization. This model is inspired by the Zero Force Evolutionary Law, which states that there is always a tendency for diversity and complexity to increase in any evolutionary system containing variation and heredity. That is, in the absence of evolutionary forces, the expectation is a continual increase in diversity and complexity at any level of biological hierarchy. I show that, when modeled, this expectation of increasing variation results not only in the formation of groups, but also in a higher probability of group formation than is found in a model that is purely random.  相似文献

Production of polyols such as erythritol, d-mannitol and d-arabitol by citric acid-producing yeasts occurred only when the medium-pH was controlled at acidic pH, as described in the previous papers.

In order to elucidate the conversion mechanism of citric acid fermentation to polyol fermentation, the effect of pH on the activities of enzymes involved in polyol synthesis and tricarboxylic acid cycle was studied. Shifting down of the medium-pH from 5.5 to 3.5 led immediately to the change of intracellular pH, from 6.5~6.7 to 5.5~5.7. Such the change affected remarkably on the activities of intracellular enzymes. Citrate synthase was significantly depressed at pH 5.7, but isocitrate lyase and phosphoenolpyruvate carboxykinase were reversely stimulated at this pH.

In some yeast strains incapable of polyol production, the change of medium-pH reflected directly on intracellular pH, whereby almost all enzymes were inhibited.

From these results, the conversion of citric acid production to polyol production was explained by the change in the enzyme activities caused by lowering of intracellular pH.  相似文献

Wall Relaxation and the Driving Forces for Cell Expansive Growth   总被引:15,自引:4,他引:11  
When water uptake by growing cells is prevented, the turgor pressure and the tensile stress in the cell wall are reduced by continued wall loosening. This process, termed in vivo stress relaxation, provides a new way to study the dynamics of wall loosening and to measure the wall yield threshold and the physiological wall extensibility. Stress relaxation experiments indicate that wall stress supplies the mechanical driving force for wall yielding. Cell expansion also requires water absorption. The driving force for water uptake during growth is created by wall relaxation, which lowers the water potential of the expanding cells. New techniques for measuring this driving force show that it is smaller than believed previously; in elongating stems it is only 0.3 to 0.5 bar. This means that the hydraulic resistance of the water transport pathway is small and that rate of cell expansion is controlled primarily by wall loosening and yielding.  相似文献

长江流域生态系统格局复杂,多种社会经济、政策和自然因素对土地利用变化的影响使得生态环境发生变化。分析了2000年至2015年长江流域生态系统格局和演变特征,及主要驱动力对生态系统变化的贡献。15年间,共有约6.4万km~2的生态系统类型发生变化,城镇增长67.5%,农田缩减7.5%,森林增加2.1%,剧烈的生态系统变化集中于下游,以及中上游的大城市,城镇聚集区以及退耕还林区。生态系统景观破碎化程度和景观多样性提高。上、中、下游生态系统格局、构成差异较大,15年间,上游和下游森林显著增加,下游城镇显著扩张、农田和湿地显著缩减,上游湿地增加最为显著。城镇化是生态系统格局演变的首要驱动力,对生态系统变化的贡献率达48.0%,长江下游城镇化的贡献率高达64.5%。生态保护与恢复工程是第二驱动力,对生态系统变化的贡献率为32.8%,在上游高达47.8%。水资源开发和农业开发贡献率分别为8.5%和9.9%,此外,气候变化促使高原湖泊面积增大。为保护长江流域生态系统的可持续发展,需划定生态保护红线,合理规划城市化进程中的土地利用,保护优质耕地,禁止重要湿地的开发。  相似文献

在小型悬浮床反应器研究絮凝颗粒酵母酒精连续发酵工艺的基础上,建立了单釜反应器有效容积0.5m3、二级串联的中试装置。以淀粉糖化液为底物,考察了悬浮床反应器由小试到中试的放大效应,提出了该反应器工业放大时对危险截面表观气速应加以限制的原则,以保证絮凝酵母颗粒不被剪切破坏。进而在平均稀释率为0.10h-1的条件下.中试装置稳定运转一个月,获得终点发酵液中酒精浓度80~85g/L,残糖低于5.0g/L,设备平均生产酒精8.4g/L·h的结果。  相似文献

建立筛选利用木糖为碳源产乙醇酵母模型,获得一株适合利用木质纤维素为原料产乙醇的酵母菌株。样品经麦芽汁培养基培养后,以木糖为唯一碳源的筛选培养基初筛,再以重铬酸钾显色法复筛。通过生理生化和26D1/D2区对筛选得到的菌株进行分析和鉴定,该菌初步鉴定为Pichia caribbica。经过筛选得到的菌株Y2-3以木糖(40g/L)为唯一碳源发酵时:生物量为23.5g/L,木糖利用率为94.7 %,乙醇终产量为4.57 g/L;以混合糖(葡萄糖40 g/L,木糖20 g/L)发酵时:生物量为28.6 g/L,木糖利用率为94.2 %,葡萄糖利用率为95.6%,乙醇终产量为20.6 g/L。Pichia caribbica是可以转化木糖及木糖-葡萄糖混合糖为乙醇的酵母菌株,为利用木质纤维素发酵乙醇的进一步研究奠定了基础。  相似文献

This landscape-scale study combines analysis of multitemporal satellite imagery spanning 30 years and information from field studies extending over 25 years to assess the extent and causes of land use and land cover change in the Loitokitok area, southeast Kajiado District, Kenya. Rain fed and irrigated agriculture, livestock herding, and wildlife and tourism have all experienced rapid change in their structure, extent, and interactions over the past 30 years in response to a variety of economic, cultural, political, institutional, and demographic processes. Land use patterns and processes are explored through a complementary application of interpretation of satellite imagery and case study analysis that explicitly addresses the local–national spatial scale over a time frame appropriate to the identification of fundamental causal processes. The results illustrate that this combination provides an effective basis for describing and explaining patterns of land use and land cover change and their root causes.  相似文献
