Purpose

In the USA, several studies have been conducted to analyze the energy consumption and atmospheric emissions of Warm-mix Asphalt (WMA) pavements. However, the direct and indirect environmental, economic, and social impacts, termed as Triple-Bottom-Line (TBL), were not addressed sufficiently. Hence, the aim of this study is to develop TBL-oriented sustainability assessment model to evaluate the environmental and socio-economic impacts of pavements constructed with different types of WMA mixtures and compare them to a conventional Hot-mix Asphalt (HMA). The types of WMA technologies investigated in this research include Asphamin® WMA, Evotherm? WMA, and Sasobit® WMA.

Methods

To achieve this goal, supply and use tables published by the U.S. Bureau of Economic Analysis were merged with 16 macro-level sustainability metrics. A hybrid TBL-LCA model was built to evaluate the life-cycle sustainability performance of using WMA technologies in construction of asphalt pavements. The impacts on the sustainability were calculated in terms of socio-economic (import, income, gross operating surplus, government tax, work-related injuries, and employment) and environmental (water withdrawal, energy use, carbon footprint, hazardous waste generation, toxic releases into air, and land use). A stochastic compromise programming model was then developed for finding the optimal allocation of different pavement types for the U.S. highways.

Results and discussion

WMAs did not perform better in terms of environmental impacts compared to HMA. Asphamin® WMA was found to have the highest environmental and socio-economic impacts compared to other pavement types. Material extractions and processing phase had the highest contribution to all environmental impact indicators that shows the importance of cleaner production strategies for pavement materials. Based on stochastic compromised programming results, in a balanced weighting situation, Sasobit® WMA had the highest percentage of allocation (61 %); while only socio-economic aspects matter, Asphamin® WMA had the largest share (57 %) among the asphalt pavements. The optimization results also supported the significance of an increased WMA use in the U.S. highways.

Conclusions

This research complemented previous LCA studies by evaluating pavements not only from environmental emissions and energy consumption standpoint, but also from socio-economic perspectives. Multi-objective optimization results also provided important insights for decision makers when finding the optimum allocation of pavement alternatives based on different environmental and socio-economic priorities. Consequently, this study aimed to increase awareness of the inherent benefits of economic input–output analysis and multi-criteria decision making through application to emerging sustainable pavement practices.  相似文献

As the use of recycled materials and industrial by-products in asphalt mixtures is increasing, we investigated if recycled additives modify the genotoxicity of fumes emitted from asphalt. Fumes were generated in the laboratory at paving temperature from stone-mastic asphalt (SMA) and from SMA modified with waste plastic (90% polyethylene, 10% polypropylene) and tall oil pitch (SMA-WPT). In addition, fumes from SMA, SMA-WPT, asphalt concrete (AC), and AC modified with waste plastic and tall oil pitch (AC-WPT) were collected at paving sites. The genotoxicity of the fumes was studied by analysis of DNA damage (measured in the comet assay) and micronucleus formation in human bronchial epithelial BEAS 2B cells in vitro and by counting mutations in Salmonella typhimurium strains TA98 and YG1024. DNA damage was also assessed in buccal leukocytes from road pavers before and after working with SMA, SMA-WPT, AC, and AC-WPT. The chemical composition of the emissions was analysed by gas chromatography/mass spectrometry. The SMA-WPT fume generated in the laboratory induced a clear increase in DNA damage in BEAS 2B cells without metabolic activation. The laboratory-generated SMA fume increased the frequency of micronucleated BEAS 2B cells without metabolic activation. None of the asphalt fumes collected at the paving sites produced DNA damage with or without metabolic activation. Fumes from SMA and SMA-WPT from the paving sites increased micronucleus frequency without metabolic activation. None of the asphalt fumes studied showed mutagenic activity in Salmonella. No statistically significant differences in DNA damage in buccal leukocytes were detected between the pre- and post-shift samples collected from the road pavers. However, a positive correlation was found between DNA damage and the urinary metabolites of polycyclic aromatic hydrocarbons (PAHs) after work shift, which suggested an association between occupational exposures during road paving and genotoxic effects. The chemical aspects associated with the incorporation of petroleum hydrocarbons‐ and metals‐affected soil has been studied extensively with respect to pavement properties, leaching behavior, sensitivities to moisture damage, and function group analysis. These studies provide information that can be used to evaluate the stability of these constituents in soils that have been incorporated as an ingredient in asphalt. These studies also indicate that cold‐mix asphalt incorporating contaminated soil will be highly stable and perform adequately as an end product. Maximum chemical performance is achieved when the asphalt is comprised of high contents of pyridinic, phenolic, and ketone groups, which can be achieved by selectively choosing the source material. If the situation requires special stability or redundancy, small amounts of shale oil and lime can be used as additives. Situations and conditions that favor the presence of inorganic sulfur, monovalent salts, and high‐strength solutions in the asphalt should be avoided because these conditions decrease the chemical stability of the asphalt cement by disrupting the functional group‐aggregate bonds and by increasing the overall permeability. However, these conditions are not typically expected in the anticipated uses of asphalt cement to stabilize contaminants in soil using Environmentally Processed Asphalt? (EPA?)* or Asphaltic Metals Stabilization? (AMS?)** remedial technologies.  相似文献

Asphalt fumes are complex mixtures of aerosols and vapors containing various organic compounds, including polycyclic aromatic hydrocarbons (PAHs). Previously, we have demonstrated that inhalation exposure of rats to asphalt fumes resulted in dose-dependent induction of CYP1A1 with concomitant down-regulation of CYP2B1 and increased phase II enzyme quinone reductase activity in the rat lung. In the present study, the potential genotoxic effects of asphalt fume exposure due to altered lung microsomal enzymes were studied. Rats were exposed to air or asphalt fume generated under road paving conditions at various concentrations and sacrificed the next day. Alveolar macrophages (AM) were obtained by bronchoalveolar lavage and examined for DNA damage using the comet assay. To evaluate the systemic genotoxic effect of asphalt fume, micronuclei formation in bone marrow polychromatic erythrocytes (PCEs) was monitored. Lung S9 from various exposure groups was isolated from tissue homogenates and characterized for metabolic activity in activating 2-aminoanthracene (2-AA) and benzo[a]pyrene (BaP) mutagenicity using the Ames test with Salmonella typhimurium YG1024 and YG1029. This study showed that the paving asphalt fumes significantly induced DNA damage in AM, as revealed by DNA migration in the comet assay, in a dose-dependent manner, whereas the micronuclei formation in bone marrow PCEs was not detected even at a very high exposure level (1733 mg h/m3). The conversion of 2-AA to mutagens in the Ames test required lung S9-mediated metabolic activation in a dose-dependent manner. In comparison to the controls, lung S9 from rats exposed to asphalt fume at a total exposure level of 479+/-33 mg h/m3 did not significantly enhance 2-AA mutagenicity with either S. typhimurium YG1024 or YG1029. At a higher total asphalt fume exposure level (1150+/-63 mg h/m3), S9 significantly increased the mutagenicity of 2-AA as compared to the control. However, S9 from asphalt fume-exposed rats did not significantly activate the mutagenicity of BaP in the Ames test. These results show that asphalt fume exposure, which significantly altered both phases I and II metabolic enzymes in lung microsomes, is genotoxic to AM and enhances the metabolic activation of certain mutagens through altered S9 content.  相似文献

Genotoxic effects of bitumen fumes in Big Blue transgenic rat lung   总被引:1,自引:0,他引:1  
Road paving workers are exposed to bitumen fumes (CAS No. 8052-42-4), a complex mixture of volatile compounds and particles containing carcinogenic and non-carcinogenic polycyclic aromatic hydrocarbons. However, epidemiological and experimental animal studies failed to draw unambiguous conclusions concerning their toxicity. In order to gain better insights on their genotoxic potential, we used an experimental design able to generate bitumen fumes at road paving temperature (temperature: 170 degrees C, total particulate matter: 100mg/m3) and perform a nose-only exposure of Big Blue transgenic rodents 6h/day for five consecutive days. The mutagenic properties of bitumen fumes were determined by analyzing the mutation frequency and spectrum of the neutral reporter gene cII inserted into the rodent genome. We previously observed in mouse lung, that bitumen fumes did not induce an increase of cII mutants, a modification of the mutation spectrum, nor the formation of DNA adducts. Since DNA adducts were found in the lungs of rats exposed to asphalt fumes in similar conditions, we decided to carry out an analogous experiment with Big Blue rats. A DNA adduct was detected 3 and 30 days after the end of treatment suggesting that these genetic alterations were quite steady. Thirty days after exposure, the cII mutant frequency was similar in control and exposed rats. In addition, a slight but not significant modification of the mutation spectrum associated with an increase of G:C to T:A and A:T to C:G transversions was noticeable in the treated animals. Then, these data failed to demonstrate a pulmonary mutagenic potential for bitumen fumes generated at road paving temperature in our experimental conditions despite the presence of a DNA adduct. These results may provide information concerning the pulmonary mechanism of action of this aerosol and may contribute to the occupational health hazard assessment.  相似文献

Purpose

With building construction and demolition waste accounting for 50 % of land fill space, the diversion of reusable materials is essential for Perth”s environment. The reuse and recovery of embodied energy-intensive construction materials during civil engineering works programs can offer significant energy savings and assist in the mitigation of the carbon footprint.

Methods

A streamlined life cycle assessment, with limited focus, was carried out to determine the carbon footprint and embodied energy associated with a 100-m section of road base. A life cycle inventory of inputs (energy and materials) for all processes that occurred during the development of a 100-m road section was developed. Information regarding the energy and materials used for road construction work was obtained from the Perth-based firm, Cossill and Webley, Consulting Engineers. These inputs were inserted into Simapro LCA software to calculate the associated greenhouse gas emissions and embodied energy required for the construction and maintenance of a 100-m road section using. Two approaches were employed; a traditional approach that predominantly employed virgin materials, and a recycling approach.

Results and discussion

The GHG emissions and embodied energy associated with the construction of a 100-m road section using virgin materials are 180 tonnes of CO2-e and 10.7 terajoules (TJ), respectively. The substitution of crushed rock with recycled brick road base does not appear to reduce the carbon footprint in the pre-construction stage (i.e. from mining to material construction, plus transportation of materials to the construction site). However, this replacement could potentially offer environmental benefits by reducing quarrying activities, which would not only conserve native bushland but also reduce the loss of biodiversity along with reducing the space and cost requirements associated with landfill. In terms of carbon footprint, it appears that GHG emissions are reduced significantly when using recycled asphalt, as opposed to other materials. About 22 to 30 % of greenhouse gas (GHG) emissions can be avoided by replacing 50 to 100 % of virgin asphalt with Reclaimed Asphalt Pavement (RAP) during the maintenance period.

Conclusions

The use of recycled building and road construction materials such as asphalt, concrete, and limestone can potentially reduce the embodied energy and greenhouse gas emissions associated with road construction. The recycling approach that uses 100 % reused crushed rock base and recycled concrete rubble, and 15 % RAP during the maintenance period could reduce the total carbon footprint by approximately 6 %. This large carbon saving in pavement construction is made possible by increasing the percentage of RAP in the wearing course.  相似文献

Recirculating aquaculture systems are highly intensive culture systems that actively filter and reuse water, thus minimizing water requirements and creating relatively small volumes of concentrated waste (compared to flow-through aquaculture systems). Vermicomposting, which uses earthworms to stabilize and transform organic wastes into valuable end-products, has been proposed as an alternative treatment technology for high-moisture-content organic wastes from agricultural, industrial and municipal sources. This study was conducted to determine if the effluent solids from a large recirculating aquaculture facility (Blue Ridge Aquaculture, Martinsville, Virginia) were suitable for vermicomposting using the earthworm Eisenia fetida. In two separate experiments, worms were fed mixtures of solids removed from aquaculture effluent (sludge) and shredded. Mixtures containing 0%, 5%, 10%, 15%, 20%, 25%, and 50% aquaculture sludge (dry weight basis) were fed to the worms over a 12-week period and their growth (biomass) was measured. Worm mortality, which occurred only in the first experiment, was not influenced by feedstock sludge concentration. In both experiments worm growth rates tended to increase with increasing sludge concentration, with the highest growth rate occurring with feedstocks containing 50% aquaculture sludge. Effluent solids from recirculating aquaculture systems mixed with shredded cardboard appear to be suitable feedstocks for vermicomposting.  相似文献

Organisms of the genera Pseudomonas, Chromobacterium, and Bacillus capable of degrading asphalt were isolated by enrichment cultures. The asphalt degradation by these organisms varied from 3 to 25% after incubation for 1 week. The effects of temperature, pH, and atmosphere of incubation on asphalt degradation were investigated and were shown to vary with different organisms on the same substrate.  相似文献

Enzymes are industrially applied under increasingly diverse environmental conditions that are dictated by the efforts to optimize overall process efficiency. Engineering the operational stability of biocatalysts to enhance their half-lives under the desired process conditions is a widely applied strategy to reduce costs. Here, we present a simple method to enhance enzyme stability in the presence of monophasic aqueous/organic solvent mixtures based on the concept of strengthening the enzyme's surface hydrogen-bond network by exchanging surface-located amino acid residues for arginine. Suitable residues are identified from sequence comparisons with homologous enzymes from thermophilic organisms and combined using a shuffling approach to obtain an enzyme variant with increased stability in monophasic aqueous/organic solvent mixtures. With this approach, we increase the stability of the broad-spectrum amino acid racemase of Pseudomonas putida DSM 3263 eightfold in mixtures with 40% methanol and sixfold in mixtures with 30% acetonitrile.  相似文献

The separation of tryptophan enantiomers was carried out with medium-pressure liquid chromatography using BSA (bovine serum albumin)-bonded silica as a chiral stationary phase. The influence of various experimental factors such as pH and ionic strength of mobile phase, separation temperature, and the presence of organic additives on the resolution was studied. In order to expand this system to preparative scale, the loadability of sample and the stability of stationary phase for repeated use were also examined. The separation of tryptophan enantiomers was successful with this system. The data indicated that a higher separation factor (α) was obtained at a higher pH and lower temperature and ionic strength in mobile phase. Addition of organic additives (acetonitrile and 2-propanol) in mobile phase contributed to reduce the retention time of L-tryptophan. About 30% of the separation factor was reduced after 80 days of repeated use.  相似文献

Formation of noncovalent complexes between alpha-chymotrypsin (CT) and a polyelectrolyte, polybrene (PB), has been shown to produce two major effects on enzymatic reactions in binary mixtures of polar organic cosolvents with water. (i) At moderate concentrations of organic cosolvents (10% to 30% v/v), enzymatic activity of CT is higher than in aqueous solutions, and this activation effect is more significant for CT in complex with PB (5- to 7-fold) than for free enzyme (1.5- to 2.5-fold). (ii) The range of cosolvent concentrations that the enzyme tolerates without complete loss of catalytic activity is much broader. For enhancement of enzyme stability in the complex with the polycation, the number of negatively charged groups in the protein has been artificially increased by using chemical modification with pyromellitic and succinic anhydrides. Additional activation effect at moderate concentrations of ethanol and enhanced resistance of the enzyme toward inactivation at high concentrations of the organic solvent have been observed for the modified preparations of CT in the complex with PB as compared with an analogous complex of the native enzyme. Structural changes behind alterations in enzyme activity in water-ethanol mixtures have been studied by the method of circular dichroism (CD). Protein conformation of all CT preparations has not changed significantly up to 30% v/v of ethanol where activation effects in enzymatic catalysis were most pronounced. At higher concentrations of ethanol, structural changes in the protein have been observed for different forms of CT that were well correlated with a decrease in enzymatic activity. The batch culture degradation of NASA wastewater containing mixtures of citric acid, methylhydrazine, and their reaction product was studied. The organic contaminants present in the NASA wastewater were degraded by Achromobacter sp., Rhodococcus B30 and Rhodococcus J10. While the Achromobacter sp. showed a preference for the degradation of the citric acid, the Rhodococcus species were most effective in reducing the methylhydrazine and the reaction product. Removals of more than 50% were observed for citric acid, methylhydrazine and the reaction product when the NASA wastewater was inoculated with the microbes in batch cultures. Simulation and chemical characterization of citric acid and hydrazine mixtures show that the interaction is partly of a chemical nature and leads to the formation of a conjugated UV/Visible absorbing compound. An 'azo' carbonyl derivative of the citric acid, consistent with the spectral data obtained from the investigation, has been proposed as the possible product.  相似文献

Diethyl l-aspartate was polymerized by a bacterial protease from Bacillus subtilis (BS) in organic solvent at a temperature between 30 and 50 degrees C to yield alpha-linked poly(ethyl l-aspartate) having an M(w) of up to 3700 and a maximum polymer yield of 85%. The best polymerization conditions were the 40 degrees C polymerization of diethyl l-aspartate using 30% protease BS containing 4.5 vol % water in acetonitrile for 2 days. Poly(ethyl l-aspartate) was readily depolymerized by the enzyme into the oligomeric and monomeric l-aspartate in aqueous acetonitrile. Poly(sodium aspartate) prepared by the saponification of poly(ethyl l-aspartate) was readily biodegradable by activated sludge obtained from the municipal sewage treatment plant. Also, poly(sodium aspartate) was depolymerized by the hydrolase enzyme into the monomeric aspartate. These results may indicate the sustainable chemical recycling and biorecycling of this polymer.  相似文献

We present a detailed analysis of the variety, quantity and distribution of the amino acids detected in organic residues after acid hydrolysis. Such organic residues are produced in the laboratory after the vacuum ultraviolet (VUV) irradiation of several astrophysically relevant ice mixtures containing H(2)O, CO, CO(2), CH(3)OH, CH(4) and NH(3) at low temperature (10-80 K), and subsequent warm-up to room temperature. We explore five experimental parameters: the irradiation time, the temperature, the ice mixture composition, the photon dose per molecule and the substrate for the ice deposition. The amino acids were detected and identified by ex-situ liquid chromatography analysis of the organic residues formed after warming the photolysed ices up to room temperature. This study shows that in all experiments amino acids are formed. Their total quantities and distribution depend slightly on the experimental parameters explored in the present work, the important requirement to form such molecules being that the starting ice mixtures must contain the four elements C, H, O and N. We also discuss the effects of the chemical treatment needed to detect and identify the amino acids in the organic residues. Finally, these results are compared with meteoritic amino acid data from the carbonaceous chondrite Murchison, and the formation processes of such compounds under astrophysical conditions are discussed.  相似文献

In the present study, bovine milk xanthine oxidase activity in various aqueous-organic mixtures and the effects of pH, temperature, and lyophilization on the enzyme activity have been investigated. The enzyme was incubated with xan-thine as the substrate in Sorenson’s phosphate buffer (pH 7.0) containing 0.1 mM EDTA, and the activity was determined spectrophotometrically in the absence and presence of different fractions of nine water-miscible organic solvents at 27–50°C and at different pH values ranging from 6 to 9. The organic solvents reduced the enzyme activity to different extents. In spite of these inhibitory effects, the enzyme showed relatively good stability in the aqueous-organic mixtures compared with the aqueous medium. Expression of the bacterio-opsin gene in Escherichia coli has been described in the accompanying papers. We now describe rapid and efficient methods for the purification of the E. coli-expressed bacterio-opsin. Bacterio-opsin can be extracted from E. coli membranes in a denatured form by using an organic solvent containing chloroform, methanol, water, and triethylamine. The bacterio-opsin, enriched to 30-50% in the extract, can be further purified to 90% by ion-exchange chromatography on DEAE-Trisacryl or hydroxylapatite chromatography in organic solvents or by preparative sodium dodecyl sulfate gel electrophoresis. In appropriate aqueous phospholipid/detergent mixtures, up to 80% of purified protein refolds and binds retinal covalently to regenerate the bacteriorhodopsin chromophore. When reconstituted into phospholipid vesicles, bacteriorhodopsin from E. coli shows the expected proton pumping activity in response to illumination.  相似文献

Studies were performed to elucidate the optimal reaction conditions (pH, temperature, ammonia concentration and biocatalyst loading) for bioconversion of trans-cinnamic acid (t-CA) to L-phenylalanine (L-Phe) by L-phenylalanine ammonia lyase (PAL) containing Rhodotorula glutinis cells. All treatments with permeabilizing agents stimulated L-Phe production and also enhanced instability of the catalyst, except Triton X-100 which gave a superior (56%) increase in conversion as compared to the control and a significant stabilization of PAL enzyme. Inclusion of several activity modifiers and stabilizer additives in reaction mixtures were shown to enhance the yield of L-Phe and maintained PAL stability over several successive incubations during the bioconversion process. Maximum stabilization of PAL and enhancement of L-Phe production was achieved with addition of 20% polyhydric alcohol (glycerol). The production of L-Phe continued to the fifth cycle and the total yield increased 2.3 times compared to the yield produced by the control (without glycerol addition) during the repeated batch process. Reducing agents such as 2-mercaptoethanol and thioglycolic acid were added to the bioconversion mixture in order to reduce the effects of oxygen on PAL catalyst life. Production of L-Phe by addition of 400 mgL(-1) of thioglycolic acid was maximized over the control by 55%. When both 20% glycerol and 400 mgL(-1) thioglycolic acid were simultaneously present in the reaction mixture, reuseability and stability of biocatalyst (PAL) were extended to eight consecutive cycles and conversion rate and overall productivity of L-Phe were higher than that of the control. These results may lead to improvements in the production of the essential amino acid L-Phe.  相似文献
