In a light-dark (LD) regimen, sperm, first apyrene and then eupyrene, start moving out of the fused testes of the Mediterranean flour moth, Anagasta kuehniella, toward the beginning of the scotophase. At 27° ± 2°C, the sperm mass remains in the proximal part of the vasa deferentia for 10 to 12 hr and then passes rapidly into the seminal vesicles, remains in these organs for about 5 hr, and is then transported to the ductus ejaculatoris duplex where it becomes available for ejaculation. The phases of sperm movement appear to be closely related to sperm development, and the reproductive activity of the moths. In isolated abdomens there is a significant reduction in the amount of sperm released from the testes, but normal periodicity of sperm release and movement continues in either LD or continuous dark (DD) regimens, and rapid phase shifting occurs when a LD regimen is reversed. All stages of sperm movement are disrupted in continuous light (LL), but normal periodicity is usually resumed when isolated abdomens of the LL moths are placed in LD or DD regimens. Normal periodicity also occurs in moths paralyzed with tetrodotoxin or procaine. Removal of any one of the four abdominal ganglia from LL moths does not prevent increased sperm release when the moths are placed in LD, though with each ganglion there is some disruption of the normal pattern of movement down the vasa deferentia. It is thought that the testes and vasa deferentia down to at least the seminal vesicles represent a semiautonomous complex in which periodicity is maintained by endogenous circadian activity in cells of the testes (and possibly the vasa deferentia) or more probably in a peripheral control center.  相似文献

Abdominal injection of 1 μg aqueous 20-hydroxyecdysone into Anagasta kuehniella, anytime prior to the initiation of sperm release from the testes, prevents the impending release of eupyrene sperm bundles. Apyrene sperm release is not prevented and there is complete recovery of eupyrene release by the following cycle 24 hr later. If 20-hydroxyecdysone is administered on consecutive days, no eupyrene bundles are released and although apryene sperm release continues, it diminishes with time. The effect of 20-hydroxyecdysone in preventing eupyrene release is dose dependent. Administration of decreasing 20-hydroxyecdysone dosages results in increasing numbers of eupyrene bundles released. When a single injection of 20-hydroxyecdysone is administered to isolated abdomens, recovery time of eupyrene sperm release is slower than in whole moths and total recovery is not seen even by 5 days after administration. Apyrene sperm release is also affected to a greater extent than in whole moths, and in some cases, no apyrene release was detected at all. Treatment with 20-hydroxyecdysone prevents cupyrene bundles from passing through the testicular basilar membrane into the vasa efferentia, thus causing a build up of bundles near the basilar membrane but no disintegration of these eupyrene sperm bundles.  相似文献

Sperm production and movement from the fused testes into the male reproductive tract of the common cutworm Spodoptera litura were studied in insects maintained in a 12 h:12 h light dark (LD) regime. Two types of sperm bundles, eupyrene (nucleated) and apyrene (anucleate) were present in the adult testes. Eupyrene bundles constituted about 25% of the total. Descent of spermatozoa from the testes into the upper vas deferens (UVD) first occurred about 24-30 h before adult eclosion. On entering the reproductive tract, eupyrene spermatozoa remained in bundles while apyrene bundles became dissociated before they reached the UVD. Downward movement of both eupyrene and apyrene spermatozoa within the male tract occurred in a daily rhythm. Sperm descent from the testes into the UVD occurred during the early scotophase, followed by their further descent into the seminal vesicle (SV) during the photophase. Spermatozoa remained in the SV for only a short duration, whence sperm quickly passed through the lower vas deferens into the duplex, which acted as the main sperm storage organ until mating was initiated. During mating 80% of sperm left the duplex, but mating did not influence the number of sperm bundles that subsequently descended into the duplex or the rate of their descent. There was no evidence of sperm reflux. Rearing in constant light (LL) and in constant dark (DD) reduced the number of eupyrene sperm present in the testes of adults that emerged in LL and DD compared to controls (LD), although there was no significant effect on the number of apyrene sperm in the testes. The rhythmic pattern of sperm descent was suppressed in both LL and DD regimes, and the number of sperm in the duplex was adversely affected, with a marked impact in LL reared insects. Male longevity, mating behaviour, oviposition and fertility were found to be more severely affected in LL than in DD.  相似文献

Females of the cotton leafworm (Spodoptera littoralis) reared in long day conditions (LD 16:8 h) and mated to males kept throughout the whole period of development in continuous light (LL) oviposit very small numbers of mostly sterile eggs. It was found that in control males reared from the first larval instar in long day conditions there was a large accumulation of euperene sperm bundles in their testes on day 1 after imaginal moult. On day 10 of adult life the number of the sperm bundles was very small. In males kept from the first instar in continuous light there was also high number of sperm bundles on day 1 after imaginal moult but it did not decrease significantly on day 10 as was observed in controls. Transfer of different developmental stages of S. littoralis from long day conditions to continuous light resulted in a big difference in the density of eupyrene sperm bundles in their testes. In control insects reared through the whole of their development in long day conditions there was a significant decrease in the density of eupyrene sperm bundles on day 10 of adult life. By contrast, in males in continuous light, regardless of their developmental stage when transferred, there was either no change in density of sperm bundles in day 10 adults or there was a significant increase in comparison with day 1 adults. The highest density of eupyrene sperm bundles was observed in day 10 adults when they were transferred to continuous light shortly before moulting to the last instar (as day 4 larvae in the last stadium or day 1 pupae). Generally, the density of eupyrene sperm bundles on day 10 of adult development was about 2–2.5 times higher in males in continuous light then those under long day conditions. The results presented here indicate that the last larval instar and the pupa are the stages most sensitive to constant light treatment, which greatly reduces the amount of eupyrene sperm bundles released from the testes.  相似文献

Like other Lepidoptera, the silkworm (Bombyx mori) has both nucleated eupyrene and anucleated apyrene sperm that are derived from the same spermatocysts. The former type is responsible for egg fertilization, while the function of the latter is still uncertain. Many hypotheses have been presented concerning the role of the apyrene sperm in mating and fertilization, but none is supported by a convincing experimental approach. The aim of the present study was to enhance the production of apyrene sperm in vitro by using different concentrations of fetal bovine serum (FBS), namely 20%, 30% and 40%, in the culture medium used for cultivating the naked spermatocysts isolated from the silkworm testes at 0 hr, 120 hr, and 192 to approximately 360 hr after the fourth molt. Cultivation of 0-hr spermatocysts was not successful. The development of spermatocysts into eupyrene and apyrene sperm bundles was slightly slower in vitro than in vivo. The overall growth percentage of both eupyrene and apyrene bundles was satisfactory when the spermatocysts were cultivated in TC-100 culture medium containing 30% FBS.  相似文献

Sperm deliver the male complement of DNA to the ovum, and thus play a key role in sexual reproduction. Accordingly, spermatogenesis has outstanding significance in fields as disparate as infertility treatments and pest-control, making it a broadly interesting and important focus for molecular genetics research in a wide range of species. Here we investigate spermatogenesis in the model lepidopteran insect Bombyx mori (silkworm moth), with particular focus on the gene PMFBP1 (polyamine modulated factor 1 binding protein 1). In humans and mouse, PMFBP1 is essential for spermatogenesis, and mutations of this gene are associated with acephalic spermatozoa, which cause infertility. We identified a B. mori gene labeled as “PMFBP1” in GenBank’s RefSeq database and sought to assess its role in spermatogenesis. Like in mammals, the silkworm version of this gene (BmPMFBP1) is specifically expressed in testes. We subsequently generated BmPMFBP1 mutants using a transgenic CRISPR/Cas9 system. Mutant males were sterile while the fertility of mutant females was comparable to wildtype females. In B. mori, spermatogenesis yields two types of sperm, the nucleated fertile eupyrene sperm, and anucleated unfertile apyrene sperm. Mutant males produced abnormal eupyrene sperm bundles but normal apyrene sperm bundles. For eupyrene sperm, nuclei were mislocated and disordered inside the bundles. We also found the BmPMFBP1 deficiency blocked the release of eupyrene sperm bundles from testes to ejaculatory seminalis. We found no obvious abnormalities in the production of apyrene sperm in mutant males, and double-matings with apyrene-deficient sex-lethal mutants rescued the ΔBmPMFBP1 infertility phenotype. These results indicate BmPMFBP1 functions only in eupyrene spermatogenesis, and highlight that distinct genes underlie the development of the two sperm morphs commonly found in Lepidoptera. Bioinformatic analyses suggest PMFBP1 may have evolved independently in lepidoptera and mammals, and that despite the shared name, are likely not homologous genes.  相似文献

Bombyx spermatogonia are bipotential, producing nucleate eupyrene sperm and anucleate apyrene sperm. An in vitro cultivation of spermatocysts of Bombyx mori from spermatocytes to matured sperm was established. The present experiment made clear that: (i) spermatocysts must be isolated; (ii) constant shaking at 45 r.p.m. was necessary; and (iii) the addition of Bombyx hemolymph (BH) was indispensable for successful cultivation. In the absence of BH, spermatogenesis proceeded normally for 2 or 3 days and, thereafter, spermatocytes and sperm bundles began to degenerate. The best results for normal eupyrene spermatogenesis were obtained when culture medium containing BH of the corresponding stage was used in every exchange of the medium at 72 h intervals. None or only a small number of apyrene sperm bundles was produced by this culture system when spermatocysts from larval testes were used, although eupyrene spermatogenesis proceeded normally to form matured, or squeezed, sperm bundles.  相似文献

Lepidopteran spermatogenesis is dichotomous, producing eupyrene (nucleated) and apyrene (anucleated) spermatozoa. The eupyrene precedes the apyrene spermatogenesis. The timing of the switchover from eupyrene to apyrene spermatogenesis was determined by cultivating testes of accurately aged codling moth larvae in a medium containing mammalian serum but neither hemolymph nor insect hormones. In cultures, eupyrene spermatogenesis occurred in testes dissected from either 4th or 5th instar larvae, probably due to macromolecular factor-like activity of the serum of the medium. But apyrene spermatogenesis occurred only in testes explanted during or after the fourth day of the 5th instar larva. It is concluded that: (1) An apyrene spermatogenesis inducing factor (ASIF) becomes active on the fourth day of the 5th instar larva in addition to the already existing macromolecular factor. (2) Primary spermatocytes can develop into either eupyrene or apyrene spermatozoa. (3) The apyrene spermatogenesis commitment and pupal commitment of other tissues coincide about the fourth day of the 5th instar larva.  相似文献

This paper describes the physiological mechanism of action of chlorfluazuron on testicular development and spermatogenesis when sublethal doses (LD10: 1.00 ng/larva or LD30: 3.75 ng/larva) are applied topically to the cuticle of newly moulted fifth instars of the common cutworm Spodoptera litura (F.) (Lepidoptera, Noctuidae). These doses disrupt the growth and development of testes by decreasing the volume and weight of testes and thickness of testes sheath as compared with that of the controls. Sublethal doses of chlorfluazuron also significantly reduce the protein content of the testis, but do not affect the carbohydrate and lipid contents in newly emerged treated males when measured in μg/mg of testis as compared with that of the controls. Additionally, such doses disrupt spermatogenesis by reducing the number and size of eupyrene and apyrene sperm bundles in the testis. Very few or no eupyrene sperm bundles are observed in vas deferens of pre‐ and newly moulted adults compared with controls. This result shows that the transfer of sperm bundles from testes to vas deferens is delayed in treated males. The effects of chlorfluazuron on testicular development and spermatogenesis is thought to be one of the factors responsible for the reduction in fecundity, fertility and hatchability caused by sublethal doses of chlorfluazuron.  相似文献

The effects on sex pheromone-releasing, or calling behaviour, of diel photoperiods of varying daylength, of light cycle phase shifts, and of continuous illumination were investigated in Trogoderma glabrum females. On light régimes with 8 to 20 hr daylengths, calling maxima tended to centre close to photophase midpoints. Although influencing the time of day at which calling occurred, daylength had little effect on the amount of activity or the length of the calling period. When 16 : 8 LD light cycles were advanced or delayed by 4 hr, the time of day at which calling peaks were observed shifted within 2 to 4 cycles so that a constant phase relationship with photoperiod was maintained. Daily calling peaks were evident in groups of females exposed to between 1 and 5 days of continuous illumination, but mean calling time occurred earlier in the day as light exposures were lengthened. It was concluded that a circadian rhythm of calling behaviour exists in T. glabrum females. and that the rhythm can be entrained to 24 hr periodicity by photoperiod.  相似文献

Pharbitis nil, strain Violet, subjected to various photoperiods(24-hr cycle at 24?C) bloomed about 10 hr after light-off whenthe light period was 10 hr or longer, and about 20 hr afterlight-on when the light period was shorter. The higher the temperature(20–30?C) during the dark period, the later the time offlower-opening, with the temperature during the last half ofthe dark period having a stronger effect than that during thefirst half. In continuous dark or light, flower buds of Pharbitis openedabout every 24 hr at all temperatures tested between 20 and28?C, which suggests the participation of a circadian rhythmin determining the time of flower-opening. A light pulse given6–12 or 28–36 hr after the onset of the dark periodgreatly advanced the phase of this rhythm (8–10 hr). The morphology of the male genital organs and sperm ultrastructure were studied by light and transmission electron microscopy in adult males of two species of the genus Micropterix (Micropterigidae; Zeugloptera; Lepidoptera). The results are compared with findings from other primitive Lepidoptera and Trichoptera, and evaluated from a phylogenetic viewpoint. Both Micropterix species examined agree with regard to essential characters of the male genital organs. The paired testes are separate and show no internal compartmentalization. The male genital organs contain only mature nucleate (eupyrene) spermatozoa. Anucleate (apyrene) spermatozoa characteristically found in all other Lepidoptera were not observed. The eupyrene spermatozoa are filiform, measure 100 μm in length, and contain an elongated nucleus, 2 mitochondrial derivatives without paracrystalline materials, and a 9 + 2 axoneme without accessory tubules; the nucleus extends for almost the entire length of the spermatozoa. The absence of apyrene spermatozoa in Micropterix is in contrast to their presence in another species of the same family.  相似文献

There are two sperm morphs of silkworm, the nucleated spermatozoa (eupyrene) and anucleated spermatozoa (apyrene). Eupyrene sperm cannot complete fertilization successfully without the apyrene sperm. Here a modified rapid and efficient method for sperm identification was developed, after 10 s of fixation in paraformaldehyde and 30 s of 4′6-diamidino-2-phenylindole (DAPI) or propidium Iodide (PI) staining, the sperm bundles can be detected easily using a fluorescence microscope. Sperm maturation process of silkworm from the fifth instar larvae to the adult was described with the above method, the precise time of earliest elongate apyrene bundles was detected on day 2 of pre-pupation, with a ratio of 5% in total sperm bundles, after which the percentage of apyrene sperm bundles increased rapidly and attained a relatively stable ratio of 75% at the end of pupation and nearly 80% after eclosion. Delayed mating leads to apyrene sperm accumulation and damaged fertilization. Previous study showed that ecdysone can increase the frequency of apyrene sperm bundles in vitro. Here 20-hydroxyecdysone (20E) was injected into hemolymph of the 2-d-old fifth instar larvae, the worms entered into mounting period after three days injection, but no apyrene sperm bundles were induced unless day 2 of pre-pupation. Interestingly, maturation of eupyrene sperm bundles were accelerated, and the ratio of eupyrene sperm bundles increased and exhibited a dose-dependent effect after 20E injection, which indicated that the development of eupyrene sperm can be accelerated by ecdysone before pupation of silkworm in vivo. Recent work has shown that a high fluoride (F) intake in rodents leads to histopathologic changes in the germinal epithelium of testes and to zinc deficiency in the testis and several other organs. The purpose of the present study was to determine whether an elevation of testicular zinc concentration during fluoride exposure could protect the testes of bank vole from damage. The elevation of testicular zinc was achieved by exposing the bank voles to a long photoperiod (16 hr light/8 hr dark). The zinc concentration in the testes of bank voles kept under the long photoperiod was 38% higher than that in animals exposed to a moderate photoperiod (12 hr light/12 hr dark). Fluoride exposure (200 μg F/ml drinking water) during 4 months decreased additionally p > 0.05 zinc concentration in the testes of bank voles kept under the moderate photoperiod. The same animals also exhibited histopathologic changes in the germinal epithelium. By contrast, these disturbances were not observed in animals maintained in the long photoperiod. This experiment suggests that an increase in testicular zinc due to a long photoperiod prevents seminiferous tubules from a damage induced by fluoride in bank voles. The protective effects of zinc (or a long photoperiod) did not appear to be related to a decrease in testicular fluoride accumulation or lipid peroxidation.  相似文献

In Spodoptera littoralis, testicular sperm release occurs in a daily rhythm, which is controlled by endogenous circadian oscillator located in the male reproductive system. Although this rhythm is essential for male fertility, factors that initiate and maintain daily sperm release are not understood. In this study, we investigated a modulatory role for ecdysteroids in the sperm release rhythm and identified the source of ecdysteroids in adult males. We found that the onset of sperm release occurs two days pre-eclosion and coincides with a significant decrease in haemolymph ecdysteroids levels. 20-HE injection into the pupae prior to the first sperm release delayed its initiation and disrupted the developing rhythm in a dose dependent manner. 20-HE injection into adults depressed the number of sperm bundles leaving the testes. A day before the initial sperm release, ecdysteroid levels in the haemolymph and testes begin to oscillate in a circadian fashion. Ecdysteroid rhythms continue throughout imaginal life and correlate with the rhythm of sperm release. In each cycle, testicular sperm release coincides with a trough in testicular ecdysteroid concentration. Rhythmic changes in ecdysteroid levels are regulated by an endogenous circadian oscillator that continues to function in decapitated males. The generation of a complete cycle of ecdysteroid release by testes cultured in vitro indicates that this oscillator is located in the gonads. The haemolymph ecdysteroid levels are significantly lower and arrhythmic in males with removed testes, indicating that the testes are an important ecdysteroid source that may contribute to oscillations in haemolymph ecdysteroid levels.  相似文献

Two types of sperm, nucleate eupyrene and anucleate apyrene, occur in the silkworm as in other lepidopteran species. Hormones and other substances have been assumed to play important roles in sperm dimorphism. We established an in vitro cultivation system for silkworm spermatocytes, and found that apyrene sperm are not produced when spermatocytes from larval testes are cultivated, though eupyrene spermatocytes develop normally into mature sperm. Based on the fact that ecdysteroid titers increase rapidly and peak 1 day after spinning, and that the amount of glycogen reaches its peak 1 day before the spinning stage, we studied the effects of adding glucose and/or 20-hydroxyecdysone to the culture medium. The experiments disclosed a significant additive effect of both substances on apyrene sperm production.  相似文献

Female moths of Bombyx mori were artificially inseminated with cryopreserved semen. The fertility of inseminated females varied from 0% to 76.9% depending on the strain. Addition of fresh semen from triploid males, which are infertile but whose semen includes intact apyrene sperm, greatly improved fecundity of cryopreserved semen from normal males. Frozen apyrene sperm from the triploid donors also improved the fecundity of females, inseminated with cryopreserved normal semen, but less than fresh semen from triploid males. Fertilization success in B. mori requires the presence of both, intact eupyrene and apyrene sperm. Our results show that eupyrene sperm tolerate the cryopreservation process better than apyrene sperm. Hence, we recommend to add apyrene sperm from the triploid donors as helper sperm routinely to cryopreserved semen in artificial insemination. This may advance the application of cryopreservation as a routine technique to maintain silkworm resources. The technique may also be applicable to other moth and butterfly species which, like B. mori, possess eupyrene and apyrene sperm.  相似文献

Studies were made of the effects of blue, green, red and far-red (FR) light on the circadian rhythm of leaf movement of Coleus blumei × C. frederici, a short day plant. Under continuous illumination with blue light, there was a significant lengthening of the period of the rhythm to about 24.0 hr, as compared to 22.5 hr in continuous darkness. Under continuous red light, the period length was significantly shortened to 20.5 hr. Under continuous green or FR, the period length was not significantly different from the dark control. It was observed that under continuous FR illumination, the leaves tended to oscillate in a more downward position. Eight-hr red light signals were effective in advancing the phase of the rhythm as compared to a control under continuous green light. Blue light signals were effective in delaying the phase of the rhythm. FR light signals were ineffective in producing either delay or advance phase shifts. Far-red light did not reverse the effects of either red or blue light signals. On the basis of these results it is suggested, that pigments which absorb blue or red light, rather than phytochrome, mediate the effect of light on the circadian rhythm of leaf movement.  相似文献

Sublethal concentrations of the bisacylhydrazine moulting hormone agonists, RH-5849, and tebufenozide (RH-5992) were fed to sixth (final) instar larvae of Spodoptera litura. Both RH-5849 and tebufenozide adversely affected the mating success of S. litura when the surviving treated males were crossed with normal females. The ecdysone agonists decreased the longevity of treated males and of untreated females when crossed with treated males. The number of eggs laid by untreated females mated to treated males was decreased, and the fertility (percentage of hatching success) of the resulting eggs was reduced. These effects on male reproductive success were at least in part explained by a reduction in the number of sperm transferred during mating. The adverse effects of tebufenozide on male reproductive function were qualitatively the same as those of RH-5849, but tebufenozide was active at lower concentrations. To understand the reason for these adverse effects on male reproduction, we investigated the effects of the insecticides on male reproductive physiology. Male reproductive tract development and testicular volume of resulting adult moths were adversely affected by sublethal larval exposure to the ecdysone agonists. Dose-dependent reductions occurred in the production of eupyrene and apyrene spermatozoa in the adult testes, and in the number of spermatozoa released from the testes into the male reproductive tract. The descent into the male tract of both eupyrene and apyrene sperm was found to start at the normal stage of development in both normal and treated insects, but the daily rhythm of sperm descent was subsequently disturbed in the insecticide-treated moths. This affected the numbers of sperm in the upper vas deferens (UVD), seminal vesicle (SV), and duplex (duplex). Injections of RH-5849 given to pharate adult or newly emerged adult S. litura also caused drastic reduction in the number of sperm in the upper regions of the male tract, when measured 24 h after injection. The possible importance of pest population reduction through the sublethal anti-reproductive effects of insecticides is discussed.  相似文献
