Immature cotyledons of open-pollinated seeds from five walnut (Juglans regia L.) cultivars were excised from fruits at 6–11 weeks after full pistillate bloom and grown on a sequence of media to induce somatic embryogenesis. Globular, heart, cotyledonary and complete somatic embryos were obtained. Embryogenic cultures were maintained for more than a year by repetitive embryogenesis in which the roots, cotyledons and hypocotyls of somatic embryos formed additional adventive somatic embryos. Mature somatic embryos required a cold treatment of 8–10 weeks at 2–4°C to overcome apical dormancy. Selected plantlets derived from these somatic embryos were grown to young plants in soil. In addition, somatic embryogenesis was induced in J. hindsii (Jeps.), Jeps., and in Pterocarya sp., another member of the Juglandaceae.  相似文献

The carbohydrate, protein, lipid, total RNA and DNA at four stages in the development of the eggs of Tetraclita squamosa rufotincta Pilsbry, Balanus perforatus Brug., and Pollicipes cornucopia Darwin have been determined. The initial caloric content varies with species, that of the large egg of Tetraclita being extremely high; on a relative volume-specific basis, however, differences, although still present, are much less variable. A comparison has been made with other species. Those eggs in which lipid predominates over protein in Stage I lose more lipid during development. The relatively large quantities of reserves in Tetraclita may serve as metabolic substrate during the development of the planktonic larvae under nutrient-poor conditions.  相似文献

研究了盐氮处理条件下盐地碱蓬种子成熟过程中的离子积累以及种子萌发特性,以理解盐地碱蓬在种子发育及萌发过程中对高盐低氮生境的适应性。结果表明,种子成熟过程中,不同浓度盐氮处理下(0.5和5 mmol/L NO3--N;1和500 mmol/L NaCl),与果皮和果枝相比, 胚中Na+、K+、Cl- 和NO3-离子含量几乎没有变化。所有盐氮处理下Na+ 和Cl-都是果皮和果枝中高于胚中,尤其是在高盐处理下。高盐处理下,K+ 和NO3-含量呈现相反的趋势。高氮时无论高盐还是低盐,果皮中NO3-离子含量高于胚中,而果枝中NO3-离子含量低于胚中。而低氮时果皮及果枝中NO3-离子含量均显著低于胚中。与高氮环境下收获的种子相比,低氮环境下收获的种子萌发率,萌发指数,活力指数都要明显高。上述结果说明,盐地碱蓬种子成熟过程中存在完善的离子调控机制,保护胚免受Na+ 和Cl-等有害离子的伤害并且促进K+ 和NO3-等营养离子的积累。低NO3--N下收获的种子对外界的NO3-含量比较敏感,施以较高浓度的NO3-能够促进种子萌发,提高萌发指数和活力指数,可能与盐地碱蓬长期适应高盐低氮生境有关。  相似文献

During 2002–2004, three laboratories in Canada and France collaborated to improve initiation of somatic embryogenesis (SE) in jack pine (Pinus banksiana Lamb.), eastern white pine (P. strobus L.), maritime pine (P. pinaster Ait.), and Scots pine (P.␣sylvestris L.), giving particular attention to the effects of (1) N-(2-chloro-4-pyridyl)-N′-phenylurea (CPPU) versus various concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) and benzyladenine (BA), (2) differences in basal nutrient media, i.e., macro- and microelements, and (3) gelling agent concentration. The work was carried out separately at␣each laboratory, but the details of media compositions were shared and tested on their respective species. Results indicate that the developmental stage of the zygotic embryo (ZE) and genotype effects had a large influence on SE initiation, and that genetic effects were consistent over time. Different species responded differently to PGR types and concentration, basal nutrient media, trace elements, and their combinations. Currently, our best initiation rates based on a selected group of genotypes, optimal development stage of ZE, and medium are 3.9% for jack pine, 54.6% for eastern white pine, 76.2% for maritime pine, and 19.7% for Scots pine.  相似文献

Tolerance limits, at which 50% of larvae could survive high temperature and low salinity for 24 h, were determined for the yolk-sac larvae of Clyde and North Sea herring (Clupea harengus L.), cod (Gadus morhua L.) and flounder (Platichthys flesus L.) during early development and starvation. Clyde and North Sea herring, cod and flounder from hatching to the end of the yolk-sac stage, could withstand 21–23.5 °C, 20.5–23 °C, 15.5–18 °C and 21.5–24°C, respectively. The temperature tolerance was reduced by about 3.5–4 °C for Clyde herring and cod, 4–4.5 °C for North Sea herring and 8–8.5 °C for flounder when the larvae reached the point-of-no-return (PNR, when 50% of larvae, although still alive, are no longer strong enough to feed). The lowest salinity tolerance between hatching and the end of yolk-sac stage was 1–1.5‰ for Clyde and North Sea herring, 2–3‰ for cod and 0–1‰ for flounder. In no instance was there a loss of tolerance to low salinity during starvation. In fact, tolerance improved somewhat until the larvae became moribund. At hatching Clyde and North Sea herring larvae were negatively buoyant with a sinking rate of 0.35–0.4cm · s−1 which steadily decreased until the larvae became moribund. Cod and flounder larvae, however, were positively buoyant at hatching but became progressively less buoyant and, by the end of the yolk-sac stage they were negatively buoyant with a sinking rate of 0.06–0.07 cm · s−1. This sinking rate then decreased slightly until the PNR stage. The low salinity tolerance of all three species varied in a similar fashion to buoyancy.  相似文献

Cloned mRNAs identify three programs of gene expression in cotton (Gossypium hirsutum L.) embryos that are associated with the maturation (reserve accumulation) stage, the postabscission stage, which is marked by expression of Late-embryogenesis-abundant (Lea) mRNAs, and germination (broadly defined as including all events through early postgerminative growth). In order to test if the regulation of these programs is the same in other dicotyledonous species, their expression was studied in normal and cultured maturation-stage, postabscission-stage, and mature embryo-stage embryos or seed of oilseed rape (Brassica napus L.), soybean (Glycine max [L.] Merr.), and tobacco (Nicotiana tabacum L.) using cotton and other cDNA probes. During postabscission, Lea mRNAs accumulated in all test species and were induced in earlier maturation-stage embryos by excision and culture on basal medium. Abscisic acid often enhanced this induction in the test species. Germinationspecific mRNAs were induced in cultured maturationstage and postabscission-stage embryos of all test species. These results indicate that the regulation of embryonic and germination programs is similar in all dicotyledons tested. Properties of thylakoids isolated from leaves of three salt tolerant species, Avicennia germinans L., Avicennia marina var resinifera, and Beta vulgaris L., were not affected by the salinity in which the plants were grown. With increase in the growth salinity from 50 to 500 millimolar NaCl, there were no major effects on the per chlorophyll concentrations of lipids or proteins, or on the rates of uncoupled electron transport per chlorophyll mediated by either the whole chain or the partial reactions of photosystems I and II. Responses of the partial and whole chain reactions to variation in the sorbitol and NaCl concentrations in the assay media were independent of the salinity experienced during leaf growth and not substantially different from those of a salt-sensitive species, Cucurbita sativus L. Uncoupled rates of electron flow from water to p-benzoquinone mediated by photosystem II were insensitive to the NaCl concentration unless thylakoids were rendered Cl deficient by treatment with uncoupler under alkaline conditions. Loss of 65% to 85% of the photosystem II activity in these Cl-deficient thylakoids was restored by addition of 10 to 20 millimolar Cl.  相似文献

The influence of increased and decreased salinity on early larvae of the mussel Mytilus edulisL. and the starfish Asterias rubensfrom the White Sea is studied. Fertilized egg cells, embryos, larvae, and yearlings of the two species were compared. Salinity tolerance changes with the stage of development of mussels and starfish. Morphological changes occurring in eggs and embryos exposed to extremely high and low salinities are reported.  相似文献

A comparison is made of the vertical distribution, life cycles, and ecology of two littoral species of the sexually dimorphic isopod, Dynamene, on rocky shores of northwest France. Adult D. bidentata (Adams) prefer middle and lower eulittoral crevices and empty Balanus perforatus Brug. tests, and juveniles a wide variety of algae in the same zones. Males are capable of killing B. perforatus by wedging themselves tightly in the operculum so preventing feeding and respiration. Adult Dynamene magnitorata Holdich prefer empty Balanus crenatus Brug. tests, channels within sponges such as Halichondria and Hymeniacidon, and spaces within and between ascidian colonies, e.g., Dendrodoa and Didemnum, in the lower eulittoral and upper sublittoral. Juveniles are found in the same zones on a restricted number of algal species, in particular Chondrus crispus Stack. There is little overlap in the habitat preferences of the two species. Availability of food and tolerance of desiccation do not appear to be the primary factors responsible for the maintenance of zonation patterns; temperature would appear to be the main factor involved.The stages of the life cycle of the two species are the same but the sex-ratio in Dynamene magnitorata is much lower, and this is possibly due to the fact that the male may only spend one breeding season in the reproductive habitat as opposed to the two spent by male D. bidentata.Populations of D. bidentata from different latitudes show a tendency to increase the length of their reproductive and growth phases at more southerly locations so that ovigerous females and juveniles occur throughout the year. Winter air temperatures are a possible cause of these differences in timing.Stereoscan electron microscope observations are included to clarify the identification of the three species of Dynamene found on the Atlantic coasts of Europe and northwest Africa.  相似文献

Somatic embryos were obtained from immature cotyledons of Lupinus angustifolius, L. albus and L. mutabilis but not from L. luteus. Different kinds of basal media and plant growth regulators in primary and secondary culture were tested. The best induction media were based on B5 and were supplemented with 5 mg I-1 2,4-D alone or with 0.25 mg I-1 kinetin. Mature stage somatic embryos were obtained on media containing ABA (0.1–0.5 mg I-1) and a high NH4/NO3 ratio. Embryo germination and plantlet development occurred on MS media supplemented with glutamine or GA3.  相似文献

The objective of the present study was to assess the role of salinity-induced expression of pyrrolline 5-carboxylate synthetase (P5CS), P5CS activity, and proline accumulation on salinity tolerance in Brassica genotypes. A pot culture experiment was conducted with four Brassica genotypes viz. CS 52, CS 54, Varuna, (B. juncea) and T 9 (B. campestris) under control and two salinity levels, i.e., 1.65, 4.50 and 6.76?dS?m?1. Proline contents increased with increasing levels of salinity, and the highest content were recorded at post-flowering stage in CS 52 and CS 54. Activity of P5CS recorded at flowering stage was highest at higher level of salinity, with CS 52 and CS 54 recording highest activity. Gene expression of P5CS, which regulates the synthesis of proline, was higher in CS 52 and CS 54 under salt stress than Varuna and T 9. Comparison of partial nucleotide as well as amino acid sequence showed conserved domains, and inter and intra generic relatedness of these genes. The study suggests that salinity-induced expression of P5CS, pyrrolline-phosphate synthetase activity and proline accumulation may serve as one of the mechanism of salinity stress tolerance in Brassica genotypes.  相似文献

Although laboratory-reared species of the genus Peromyscus—including deer mice—are used as model animals in a wide range of research, routine manipulation of Peromyscus embryogenesis and reproduction has been lagging. The objective of the present study was to optimize conditions for oocyte and/or embryo retrieval and for in vitro culturing. On average, 6.4 oocytes per mouse were recovered when two doses of 15 IU of pregnant mare serum gonadotropin (PMSG) were given 24 h apart, followed by 15 IU of hCG 48 h later. Following this hormone priming, females mated overnight with a fertile male yielded an average of 9.1 two-cell stage embryos. Although two-cell stage embryos developed to 8-cell stage in Potassium Simplex Optimized Medium (KSOM; Millipore-Chemicon, Billerica, MA, USA) in vitro, but not further, embryos recovered at the 8- to 16-cell stages developed into fully expanded blastocysts when cultured in M16 media in vitro. These blastocysts had full potential to develop into late stage fetuses and possibly into live pups. As a result of the present work, all stages of Peromyscus preimplantation development are now obtainable in numbers sufficient for molecular or other analyses. These advances provide the opportunity for routine studies involving embryo transfer (e.g., chimeras, transgenics), and preservation of genetic lines by cryopreservation.  相似文献

Many species of Western Australian Cyperaceae (sedges) are vital components of the indigenous flora but commonly display low seed set, poor seed quality and intractable seed dormancy. We report the effects of incubation temperature and in vitro growth media on whole seed germination compared with extracted zygotic embryo growth in Tetraria capillaris, T. octandra, Lepidosperma drummondii and L. tenue. No germination was observed from intact whole seeds of all test species regardless of the treatment evaluated. In contrast, excised zygotic embryos of all study species exhibited significant increases in growth when cultured at 15°C compared to embryos incubated at 25°C; however, optimal media for embryo growth were genera specific. Extracted embryos of T. capillaris and T. octandra exhibited maximum percentage growth (30 and 40%, respectively) at 15°C on ½ MS medium with no plant growth regulators required. In the case of L. drummondii and L. tenue 1 μM thidiazuron was a necessary addition to the ½ MS medium resulting in 40 and 77% growth of embryos (at 15°C), respectively. Incubation of extracted embryos at 25°C (regardless of medium treatment) resulted in <10% embryo growth for T. octandra and L. tenue, while the remaining two species (L. drummondii, T. capillaris) showed no embryo growth at 25°C on any medium treatment.  相似文献

Heat shock of living tissue induces the synthesis of a unique group of proteins, the heat shock proteins. In plants, the major group of heat shock proteins has a molecular mass of 15 to 25 kilodaltons. Accumulation of these proteins to stainable levels has been reported in only a few species. To examine accumulation of the low molecular weight heat shock proteins in a broader range of species, two-dimensional electrophoresis was used to resolve total protein from the following species: soybean (Glycine max L. Merr., var Wayne), pea (Pisum sativum L., var Early Alaska), sunflower (Helianthus annuus L.), wheat (Triticum aestivum L.), rice (Oryza sativa L., cv IR-36), maize (Zea mays L.), pearl millet (Pennisetum americanum L. Leeke, line 23DB), and Panicum miliaceum L. When identified by both silver staining and incorporation of radiolabel, a diverse array of low molecular weight heat shock proteins was synthesized in each of these species. These proteins accumulated to significant levels after three hours of heat shock but exhibited considerable heterogeneity in isoelectric point, molecular weight, stainability, and radiolabel incorporation. Although most appeared to be synthesized only during heat shock, some were detectable at low levels in control tissue. Compared to the monocots, a higher proportion of low molecular weight heat shock proteins was detectable in control tissues from dicots.  相似文献

Six plant species metabolized 18O-labeled 12-oxo-cis,cis-10,15-phytodienoic acid (12-oxo-PDA) to short chain cyclic fatty acids. The plant species were corn (Zea mays L.), eggplant (Solanum melongena L.), flax (Linum usitatissimum L.), oat (Avena sativa L.), sunflower (Helianthus annuus L.), and wheat (Triticum aestivum L.). Among the products was jasmonic acid, a natural plant constituent with growth-regulating properties. The pathway is the same as the one recently reported by us for jasmonic acid synthesis in Vicia faba L. pericarp. First, the ring double bond of 12-oxo-PDA is saturated; then β-oxidation enzymes remove six carbons from the carboxyl side chain of the ring. Substrate specificity studies indicated that neither the stereochemistry of the side chain at carbon 13 of 12-oxo-PDA nor the presence of the double bond at carbon 15 was crucial for either enzyme step. The presence of enzymes which convert 12-oxo-PDA to jasmonic acid in several plant species indicates that this may be a general metabolic pathway in plants.  相似文献

Purified spinach (Spinacea oleracea L.) and barley (Hordeum vulgare L.) ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activase supported 50 to 100% activation of substrate-bound Rubisco from spinach, barley, wheat (Triticum aestivum L.), soybean (Glycine max L.), pea (Pisum sativum L.), Arabidopsis thaliana, maize (Zea mays L.), and Chlamydomonas reinhardtii but supported only 10 to 35% activation of Rubisco from three Solanaceae species, tobacco (Nicotiana tabacum L.), petunia (Petunia hybrida L.), and tomato (Lycopersicon esculentum L.). Conversely, purified tobacco and petunia Rubisco activase catalyzed 75 to 100% activation of substrate-bound Rubisco from the three Solanacee species but only 10 to 25% activation of substrate-bound Rubisco from the other species. Thus, the interaction between substrate-bound Rubisco and Rubisco activase is species dependent. The species dependence observed is consistent with phylogenetic relationships previously derived from plant morphological characteristics and from nucleotide and amino acid sequence comparisons of the two Rubisco subunits. Species dependence in the Rubisco-Rubisco activase interaction and the absence of major anomalies in the deduced amino acid sequence of tobacco Rubisco activase compared to sequences in non-Solanaceae species suggest that Rubisco and Rubisco activase may have coevolved such that amino acid changes that have arisen by evolutionary divergence in one of these enzymes through spontaneous mutation or selection pressure have led to compensatory changes in the other enzyme.  相似文献

Sand dune ecosystems are one of the areas most affected by the introduction of invasive species which represents a threat for biodiversity conservation. Their invasion patterns and spread may depend on their salinity tolerance, besides other factors. To test this hypothesis, we investigated the effects of salt stress on seed germination and on the activity of antioxidant enzymes (catalase, CAT; ascorbate peroxidase, APX; peroxidase, POX; and glutathione reductase, GR) in two legume species, an invasive, Acacia longifolia (Andrews.) Willd., and a native, Ulex europaeus (L.), very common in the sand dunes of the coast of Portugal. Salt stress was induced by adding NaCl at different concentrations, 0, 50, 100 and 200 mM, for 15 days. Results showed that the highest germination percentages were obtained in distilled water (control) and that, with increasing salt concentration, seed germination was delayed and decreased in both species. Inhibition of germination was higher in the native species, only 3% of seeds germinated at 100 mM and no seeds germinated at 200 mM NaCl. In the invasive species, the reduction was higher at 200 mM NaCl (16%). Considering the coefficient of germination velocity, a decrease in both species with increasing NaCl concentration was observed. The CAT and GR activities decreased in A. longifolia with increasing salinity. In turn, APX activity significantly increased as NaCl concentration increased while the POX activities declined at the highest NaCl concentration. On the other hand, at 50 mM NaCl lower activity of CAT and APX and higher GR and POX were found in U. europaeus. In both species, protein content increased as NaCl concentration increased. In addition, it seems that APX activities play an essential role in the scavenging reactive oxygen species (ROS). These results suggest that the seeds of the invasive legume A. longifolia are more tolerant to salinity than the native legume U. europaeus, and seem better equipped to handle the physiological stress of high salinity, which may contribute to its invasive ability in sand dunes.  相似文献

Embryo rescue technique is widely utilized to save developing embryo from abortion in interspecific hybridization of plants. In Brassica species, success of an embryo rescue depends on the age of embryo, developmental stage of embryo, and media composition. In this study, ten crosses were conducted between diploid oilseed species B. rapa × tetraploid B. napus and B. rapa × tetraploid B. juncea along with their reciprocal crosses. The immature embryos from parents and crosses were cultured at 14 d after pollination in MS and ½ MS medium without any growth regulators for direct embryogenesis. Embryos were found at different stages of development during isolation, from globular to cotyledonary, indicating that rate of embryo development depends on genotype. Embryos isolated at the cotyledonary stage showed the highest regeneration percentage (av. 83.29%) and required the lowest time to regenerate (5.8 d) compared with another type of embryo, torpedo stage, cultured. Embryo rescued in ½ MS media accounted for 10–51%, 4.0–61%, 6.0–21%, and 5.0–43% greater shoot length, root length, number of leaves, and days to flowering, respectively, in different cross combinations compared with full-strength MS media. The results indicated that the rate of regeneration from embryo is higher in ½ MS media compared with full-strength MS media. Percent regeneration from the cultured embryos differed significantly in different cross combinations. Days required for regeneration from embryos, length of siliqua, and the number of ovules per siliqua accounted for high heterosis and broad sense heritability. The findings of this experiment would be useful in developing cost-effective embryo rescue protocol and desired interspecific hybrids through embryo rescue in Brassica species.
