To aid in the development of artificial diets for mass rearing parasitioids, we investigated the anatomical changes in the digestive tract during feeding behaviour of larval Trichogramma australicum (Hymenoptera : Trichogrammatidae). Larvae begin to feed immediately upon eclosion and feed continuously for 4 h until replete. Feeding is characterised by rhythmic muscle contractions (ca 1 per s) of the pharynx. Contractions of the pharyngeal dilator muscles lift the roof of the lobe-shaped pharynx away from the floor of the chamber, opening the mouth and pumping food into the pharyngeal cavity. Another muscle contraction occurs about 0.5 s later, forcing the bolus of food through the oesophagus and into the midgut. The junction of fore- and midgut is marked by a cardiac valve. The midgut occupies most of the body cavity and is lined with highly vacuolated, flattened cells and a dispersed layer of muscle cells. In the centre of the midgut, food has the appearance of host egg contents. Food near the midgut epithelial cells has a finer, more homogeneous appearance. This change in the physical properties of the gut contents is indicative of the digestion process. In the prepupa, where digestion is complete, the entire gut contents have this appearance. After eclosion, the vitelline membrane remains attached to the posterior end of the larva. We believe this attachment to be adaptive in two ways: (1) to anchor the larva against the movements of its anterior portion, thereby increasing the efficiency of foraging within the egg; and (2) to prevent a free-floating membrane from clogging the mouthparts during ingestion.  相似文献

The process of oogenesis was studied in the bisexual species, Pratylenchus penetrans, and the monosexual species, P. zeae. The nucleus of the oocyte of P. penetrans underwent two divisions after sperm penetration. The chromosome number of P. penetrans observed at metaphase of the first maturation division was 2n = 10 and the reduced chromosome number observed at anaphase of the first maturation division was n = 5. Two polar bodies were found within the egg, indicating that this species reproduces by amphimixis. The nucleus o f the oocyte of P. zeae underwent one mitotic division and the chromosome number was 2n = 26. The presence of only 1 polar body indicates that this species reproduces by mitotic parthenogenesis. The development of the embryo was similar in P. penetrans and P. zeae. Unsegmented eggs were usually deposited by females. Following the 9-celled stage, the number of cells increased rapidly until a blastula was formed. Cell differentiation immediately followed, as evidenced by the formation of darker and larger inner endodermal cells and smaller ectodermal cells. Six to 7 days after egg deposition, the first stage larva was coiled three to f o u r times within the egg shell. During the first molt, the styler apex was formed first, then the larva moved frequently and vigorously and the styler was repeatedly thrust into the egg shell. Finally, the shell was broken and the second stage larva emerged. It took 10 days from the unsegmented egg to hatching at 23 C.  相似文献

Laboratory experiments were conducted to study the characteristics of parasitism of diamondback moth,Plutella xylostella (L.), a worldwide pest of crucifers, by a larval parasite,Diadegma semiclausum Hellén. Results showed that the greater the host larval density, the lesser was the percentage of larvae parasitized. The area of discovery decreased with increasing parasite density, but k-value remained practically unchanged. Parasitism reduced food consumption in parasitised larvae. No difference was observed in duration of larval instars between the parasitized and healthy larvae. As the frequency of superparasitism increased, the proportion of production of female progeny also increased. Presence of parasite larva within the host larva deterred superparasitism greater than the presence of parasite egg. Presence of larva of eitherD. semiclausum or another larval parasite,Cotesia plutellae Kurdjumov, within host larva deterred multiparasitism by either species greater than the presence of egg of either parasite species.Diadegma semiclausum was much more aggressive thanC. plutellae in parasitisingP. xylostella larvae.  相似文献

The external features of a shortened, apluteal development (lacking a pluteus larva) are described. Some features are unusual for echinoids. The large egg is distinctively marked by dark and pale coloured yolk. The sperm entry point is marked by a dark yolk spot and the first cleavage plane in most embryos is through the meridian on which the sperm entry point lies. Dark yolk in the animal hemisphere segregates largely to one blastomere in the two-cell embryo and pale yolk segregates to the other as a result of yolk movements during the first cell cycle. Progeny of the pale-yolk blastomere form adult oral structures and progeny of the dark-yolk blastomere form adult aboral structures. There is no feeding planktonic pluteus larva. The gastrula develops into a demersal vestibula larva with bilateral symmetry. The plane of symmetry is coincident with the Carpenter axis that defines a plane of symmetry through the madreporite in adult echinoderms. The coincidence shows that the anterior ambulacrum is vegetal with respect to egg polarity and the interradius originating at the madreporite is animal. The bilateral symmetry of the vestibula offers insight into the origin of radial symmetry in echinoderms and the body plan of an echinoderm ancestor.  相似文献

Burrowing, iocomotory and other movements of the echiuran Ochetostoma caudex have been examined and discussed. A continuous body cavity enables the worm to undergo peristaltic waves to pump water through the burrow without causing locomotion. The animal is capable of both forward and backward locomotion in its burrow. During forward locomotion, retrograde peristaltic waves are utilized which advance the animal in a step-wise fashion. Pressure changes within the coelom during burrowing, locomotion and during irrigation movements have been measured with the use of electronic recording techniques and the results interpreted in relation to direct visual observation. The structural and functional specializations for burrowing are discussed and compared with the activities of Priapulus caudatus, Sipunculus nudus and Bonellia viridis.  相似文献

Fertilization in the clear egg (1 mm in diameter) of the ctenophore Beroe ovata and, in particular, the positioning and movements of pronuclei, and their relationship to the larval oral-aboral axis have been observed. Fertilization can take place anywhere on the egg surface. The sperm pronucleus remains at its entry site and becomes surrounded by a specialized zone (30–50 μm in diameter) beneath the surface referred to as the sperm pronuclear zone or SPZ and devoid of large cortical granules. Polyspermy has been observed to be frequent; each pronucleus is surrounded by its own SPZ. Only the egg pronucleus migrates with a continuous velocity (averaging 18 μm/min) and moves beneath the surface directly toward the immobile sperm pronucleus. In polyspermic eggs, the egg pronucleus can probe several SPZ, each containing a single sperm nucleus, before it finally enters one SPZ and fuses with the chosen sperm pronucleus. These migrations of the egg pronucleus occur over several millimeters and take hours, but the mechanism underlying the motion or how the egg pronucleus decides which SPZ to enter is not yet known. Under our experimental conditions the mitotic apparatus and the first cleavage plane which defines the oral-aboral axis of the larva (see Reverberi (1971). “Experimental Embryology of Marine and Fresh-Water Invertebrates.” North-Holland, Amsterdam. for review) pass through the point of sperm entry. During fertilization and cleavage, movements of a cortical autofluorescent material are clearly seen. This material is segregated into micromeres as cleavage progresses.  相似文献

The coconut hispine beetle, Brontispa longissima (Gestro), supposedly originated from Papua New Guinea and Indonesia but has recently invaded Southeast and East Asian countries where it has been causing serious damage to Cocos nucifera L. This insect also occurs on the Southwest Islands off Kyushu Island in Japan. To evaluate the potential northward range expansion of this insect in Japan, we investigated its cold tolerance at 0, 5, and 10 °C (egg, larva, pupa, and adult), 13 °C (adult), and 15 °C (egg and hatched larva). At 15 °C, few eggs hatched, and the larvae that hatched died within a few days of hatching. At 13 °C, Ltime95 was estimated to be 23 days for adults, with the most cold-tolerant developmental stage at 10 °C. At all developmental stages, Ltime95 of B. longissima was estimated to be 19 days at 10 °C, 8 days at 5 °C, and 5 days at 0 °C, suggesting the cold tolerance of this beetle is very low. Considering average daily temperatures, it is unlikely that B. longissima can establish itself north of Amami-Oshima Island, located in the far south off the main island of Japan.  相似文献

The first detailed morphological study of larvae, pupae and adults of a species of the hooded beetles (Coleoptera: Corylophidae) – Sericoderus lateralis – is presented. Histological sectioning, scanning and transmission electron microscopy, laser confocal microscopy and 3D-computer reconstruction were used. For the first time we report that according to the morphometric data of S. lateralis, at least some corylophid beetles have three larval stages. A phylogenetic position of Corylophidae within a cucujoid-cleroid clade is confirmed, and also the placement of Sericoderini within a corlyophid subgroup, which does not include Periptycinae and Foadiini. The larvae of Sericoderus are mainly characterized by plesiomorphic features compared to those of other corylophid tribes, notably Peltinodini and Rypobiini. Morphological and developmental consequences of miniaturization are discussed. Corylophid beetles display much less specific and far-reaching morphological consequences of miniaturization compared to Ptiliidae. We report the presence of unique modifications in the neural system not shared with any other insects, such as a distinctly asymmetric supraoesophageal ganglion in first instar larva, and a total displacement of the brain to the thorax in the adult stage. A highly unusual feature of the digestive tract is the sclerotised, V-shaped ventral wall of the pharynx. Developmental and size dependent changes in the relative volume of different organs are addressed. All organ systems change allometrically in the development of S. lateralis. Allometric trends in the volume of organs confirm that the factors limiting miniaturization are the size of the neural system, associated with the number and size of neurons (most critical for first instar larva), the mass of the skeleton, the egg size, and consequently the volume of the reproductive system (for free-living insects).  相似文献

Background

Schistosome eggs must traverse tissues of the intestine or bladder to escape the human host and further the life cycle. Escape from host tissues is facilitated by secretion of immuno-reactive molecules by eggs and the formation of an intense strong granulomatous response by the host which acts to exclude the egg into gut or bladder lumens. Schistosome eggs hatch on contact with freshwater, but the mechanisms of activation and hatching are poorly understood. In view of the lack of knowledge of the behaviour of egg hatching in schistosomes, we undertook a detailed dynamic and correlative study of the hatching biology of Schistosoma japonicum.

Methodology/Principal Findings

Hatching eggs of S. japonicum were studied using correlative light and electron microscopy (EM). The hatching behaviour was recorded by video microscopy. EM preparative methods incorporating high pressure freezing and cryo-substitution were used to investigate ultrastructural features of the miracidium and extra-embryonic envelopes in pre-activated and activated eggs, and immediately after eggshell rupture. Lectin cytochemistry was performed on egg tissues to investigate subcellular location of specific carbohydrate groups.

Conclusions/Significance

The hatching of S. japonicum eggs is a striking phenomenon, whereby the larva is liberated explosively while still encapsulated within its sub-shell envelopes. The major alterations that occur in the egg during activation are scission of the outer envelope-eggshell boundary, autolysis of the cellular inner envelope, and likely hydration of abundant complex and simple polysaccharides in the lacunal space between the miracidial larva and surrounding envelopes. These observations on hatching provide insight into the dynamic activity of the eggs and the biology of schistosomes within the host.  相似文献

Amino acid levels were determined in the honeydew of pea aphids infected with A. smithi, and compared to those of the holidic diet of the host in order to study the effect of parasitism on amino acid assimilation. Honeydew of both normal and infected aphids contained large amounts of arginine, histidine and homoserine but little of the other 20 amino acids and amides present in the diet. Aphids infected with an egg stage of the parasite assimilated amino acids at reduced rates compared to controls, possibly indicating that infection inhibited protein synthesis in the host aphid. Larval growth of the parasite caused renewed amino acid utilization in the host, the effect being more pronounced for homoserine than for arginine and histidine. The results suggest that the growing A. smithi larva depletes the host of specific nutrients according to its own requirements, thus causing regulatory responses in host feeding and food assimilation.  相似文献

Embryonic invasion into the tissue of genetically different organisms has been known only in mother-embryo interactions of viviparous organisms. Hence, embryonic invasions have been thought to occur only within the same or closely related species. For endoparasitic Hymenoptera, which are oviposited in their host egg but complete their development in the later stages, entry into the host embryo is essential. To date, the entry of these parasitoids is known to be accomplished by either egg deposition directly into the embryo or by the newly hatched larva boring into the embryo. However, Copidosoma floridanum is a polyembryonic parasitoid whose development is characterized by a prolonged embryonic stage, and which lacks a larval form during its host embryogenesis. We have analyzed the behavior and fate of C. floridanum embryos co-cultured with their host embryo in vitro. Here, we show that the morula-stage embryo of C. floridanum actively invades the host embryo. Histological analyses have demonstrated that C. floridanum embryonic invasion is associated with adherent junction to host cells rather than causing an obvious wound on the host cells. These findings provide a novel case of embryonic invasion into a phylogenetically distant host embryo, ensuring cellular compatibility with host tissues.  相似文献

The egg-larval parasitoid Chelonus inanitus induces in its host Spodoptera littoralis two major developmental effects, namely a precocious onset of metamorphosis followed by a developmental arrest in the prepupal stage. Along with each egg, the wasp injects polydnavirus and venom into the host egg. The polydnavirus has been shown to play a major role in inducing the developmental arrest while the parasitoid larva is instrumental in inducing the precocious onset of metamorphosis. Here we report that experimental dilution of haemolymph of polydnavirus-containing larvae can partially prevent the developmental arrest while injection of native, but not of heat-treated, haemolymph or plasma from polydnavirus-containing larvae into nonparasitized larvae could induce developmental arrest in 14-15% of the larvae. This illustrates that heat-labile factors present in haemolymph play a role in causing developmental arrest. Injection of parasitoid medium increased the proportion of larvae entering metamorphosis precociously while injection of antibodies against a parasitoid-released protein had the opposite effect; this indicates that this protein and possibly other parasitoid-released substances are involved in inducing the precocious onset of metamorphosis. Analysis of the plasma proteome of nonparasitized, parasitized and polydnavirus-containing larvae revealed that the developmental effects are associated with only minor differences: eleven low abundant viral or virus-induced proteins and five parasitoid-released proteins were seen at specific stages of the host.  相似文献

In a very dry environment (0% r.h.) the case plays an important role in the physiology of the Tinea pellionella larva. Absence of the case leads to a reduction of oxygen consumption and a great loss of body water. At 0% r.h. the rate of water loss from a larva without its case, is twice that from a larva remaining within its case.In a very humid environment (r.h. higher than 95%) the case absorbs a very large quantity of water and becomes very heavy, but it does not interfere with larval physiology. With or without their cases the weight of the larvae stays stable, and their oxygen consumption does not change.When the humidity changes abruptly, the case acts as a buffer and assists in the regulation of the water balance of the larva. If the humidity decreases the case slows the rate of body water loss; if the humidity increases, it very quickly builds up a reserve of atmospheric water around the larva's body.  相似文献

Eggs of the European skipper, Thymelicus lineola, containing pharate first-instar larvae were brought into the laboratory at the beginning of each month from September to May and maintained at 20°C, 14L:10D. No emergence was observed within 40 days of collection before December. From that time on, while total emergence was similar, the duration of the emergence period decreased over the season from > 120 days in December to < 14 days in May. There was however no marked change in the seasonal supercooling point values of diapausing larvae.The rate of emergence from eggs collected in March was influenced by photoperiod, being slower under short-day conditions. However for any given photoperiod tested, the rate of emergence increased with an increase in temperature. The possible adaptive significance of a two-phase diapause, where diapause termination is photoperiodically controlled in species that overwinter as an egg or as a pharate first-instar larva within the egg chorion in exposed habitats, is discussed.  相似文献

Insect embryo development is a complex process which requires nuclear and cellular division, cell shape alteration, and cell movement. This process needs to be orchestrated in a specific spatial and temporal fashion. Different insect species, despite similarities, present distinct morphogenetic pathways. We used the dipteran R. americana as a comparative model for embryo morphogenesis studies, following embryo development with different histochemical and immunohistochemical procedures. Despite the phylogenetic proximity with D. melanogaster, R. americana presents a peculiar morphogenesis. We show that at the initial phases of development, from egg fertilization to blastoderm formation, R. americana is similar to Drosophila. The first cleavages are nuclear and cellularization only begins after nuclei spread throughout the egg’s cortex. However after this stage a series of cell movements establishes a short compact germ band anlage, which gastrulates in a pattern quite different from Drosophila. After gastrulation the germ band elongates anterior–posteriorly and segmentation occurs simultaneously along the embryo. Embryo development from egg fertilization to larva hatching takes about 12 days. Our results show that R. americana presents a different morphogenetic pathway which does not fit in the current short, intermediate or long germ band classification.  相似文献

Smales L. R. The life history of Labiostrongylus eugenii, a nematode parasite of the Kangaroo Island Wallaby (Macropus eugenii): development and hatching of the egg and the free living stages. International Journal for Parasitology7: 449–456. Labiostrongylus eugenii (Trichonematidae) occurs in the stomach of the Kangaroo Island Wallaby. Egg morphology is similar to that of other strongyloids. When incubated at 25°C embryogenesis is completed in about 30 h. An incomplete moult occurs within the egg, and larvae hatch at a sheathed second-stage 4312–8312 h later. Development occurred at all temperatures between 2° and 37°C with an optimum about 25°C and an upper limit near 37°C. The hatching process is very rapid, taking about 2 min. It is signalled by increased larval activity followed by a change in shell permeability. The larva hatches at that pole of the shell which has become plastic.The sheathed second-stage larva measures 659.50 ± 22.54 μm by 27.98 ± 1.22 μm. Its internal structures are concealed by a mass of opaque granules which were demonstrated as neutral lipid by oil red O staining. A second incomplete moult at 3–4 days results in a doubly sheathed infective larva from which the lipid gradually disappears. The mouth never appears patent and the larvae neither feed nor grow but rather decrease in size with age. Optimal temperatures for larvae range between 15°–25°C with 37°C about the upper limit. The significance of this developmental pattern is discussed.  相似文献

A cytoplasmic dynein ATPase has been identified in three species of unfertilized sea urchin eggs, Strongylocentrotus droebachiensis, S. purpuratus, and Arbacia punctulata. The enzyme was partially purified by sucrose gradient density centrifugation, and its polypeptide chain weight and composition were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The protein has enzymatic characteristics in common with flagellar dynein. It is activated nearly equally by Mg2+ and Ca2+, has no activity in the presence of K+ and EDTA, shows a specificity for ATP over other nucleoside triphosphates, and is inhibited by vanadate anion. On SDS-PAGE, the enzyme shows two major bands at 320,000 and 385,000 daltons, comigrating with certain ciliary and flagellar dynein polypeptides. The enzyme, given the name “egg dynein,” constitutes 2 to 4% of the total cell protein in the unfertilized egg and maintains this high value from fertilization through the late blastula stage. It appears to be equally distributed throughout the embryo at the 16-cell stage. Possible functions of egg dynein are discussed and models for dynein-microtubule mediated movements within the cytoplasm are presented.  相似文献

Drosophila is an extremely useful model organism for understanding how innate immune mechanisms defend against microbes and parasitoids. Large foreign objects trigger a potent cellular immune response in Drosophila larva. In the case of endoparasitoid wasp eggs, this response includes hemocyte proliferation, lamellocyte differentiation and eventual encapsulation of the egg. The encapsulation reaction involves the attachment and spreading of hemocytes around the egg, which requires cytoskeletal rearrangements, changes in adhesion properties and cell shape, as well as melanization of the capsule. Guanine nucleotide metabolism has an essential role in the regulation of pathways necessary for this encapsulation response. Here, we show that the Drosophila inosine 5''-monophosphate dehydrogenase (IMPDH), encoded by raspberry (ras), is centrally important for a proper cellular immune response against eggs from the parasitoid wasp Leptopilina boulardi. Notably, hemocyte attachment to the egg and subsequent melanization of the capsule are deficient in hypomorphic ras mutant larvae, which results in a compromised cellular immune response and increased survival of the parasitoid.  相似文献

Micromalthus debilis 33 ), has one of the most bizarre life cycles of any metazoan. Reproduction is typically by thelytokous, viviparous, larviform females, but there is also a rare arrhenotokous phase. The active first‐instar (triungulin) larva develops into a legless, feeding (cerambycoid) larva. This form either pupates, leading to a diploid adult female, or develops into any of three subsequent types of reproductive paedogenetic forms: (1) a thelytokous female that produces triungulins via viviparity; (2) an arrhenotokous female that produces a single egg that develops into the short‐legged (curculionoid) larva, eventually devouring its mother and becoming a haploid adult male; or (3) an amphitokous female that can follow either of the two above reproductive pathways. We speculate that Micromalthus is dependent on maternally transmitted bacteria for the ability to digest rotting wood, and that these bacteria are senescent in males, causing males to be obligately cannibalistic. Obligate male cannibalism, in turn, would have dramatically increased the cost of males, and have created a strong selective advantage for cyclic thelytoky and the other features of the Micromalthus life cycle that minimize the role of the male.  相似文献

Infections by schistosomes result in granulomatous lesions around parasite eggs entrapped within the host tissues. The host and parasite determinants of the Schistosoma mansoni egg-induced granulomatous response are areas of active investigation. Some studies in mice implicate Tumor Necrosis Factor (TNF) produced in response to the infection whereas others fail to find a role for it. In addition, in the mouse model, the S. mansoni secreted egg antigen omega-1 is found to induce granulomas but the underlying mechanism remains unknown. We have recently developed the zebrafish larva as a model to study macrophage recruitment and granuloma formation in response to Schistosoma mansoni eggs. Here we use this model to investigate the mechanisms by which TNF and omega-1 shape the early granulomatous response. We find that TNF, specifically signaling through TNF receptor 1, is not required for macrophage recruitment to the egg and granuloma initiation but does mediate granuloma enlargement. In contrast, omega-1 mediates initial macrophage recruitment, with this chemotactic activity being dependent on its RNase activity. Our findings further the understanding of the role of these host- and parasite-derived factors and show that they impact distinct facets of the granulomatous response to the schistosome egg.  相似文献
