Lymph node cells from the posterior cervical and mesenteric lymph nodes of immune rats passively protected syngeneic recipient rats against Angiostrongylus cantonensis better than cells from the spleen, thymic and inguinal lymph nodes either as reduced worms burdens and/or stunted growth. No antibody was detected in the sera of recipient rats after transfer of the cells and before infection which suggested that the protection was cell- rather than antibody-mediated.  相似文献

Splenectonuzed rats infected with Angiostrongylus cantonensis had more and longer worms and lower anti-A. cantonensis antibody titres compared to intact and sham operated rats. Splenectomy, however, had no effect on the capacity of rats to mount a cell-mediated immune response, as assessed by the in vitro uptake of tritiated thymidine by peripheral blood lymphocytes following stimulation by PHA and adult worm antigen. Adoptive protection and antibody production against A. cantonensis could be transferred with immune spleen cells. This protection was strongly dose and time dependent. Protection was adoptively conferred with 3 × 109 spleen cells transferred 7 days before infection.  相似文献

A protective immunity against the cestode Hymenolepis nana was transferred with serum taken from actively immunized mice. All of 17 pooled sera examined, which were taken from mice immunized for 3 or more weeks, were strongly effective. Intraperitoneal injection of a total of 3·0 ml serum made the recipient mice (4–5 weeks old) almost completely immune. In almost all the mice given immune serum no cysticercoids were found on day 4. In mice receiving immune serum, oncospheres hatched, invaded the intestinal villi and differentiated to stage II or III larvae, but failed to develop to fully developed cysticercoids. The degree of protection conferred by serum transfer was similar to, but slightly weaker than that stimulated by active immunization. The major effect of immune serum was damaging hatched oncospheres in both the intestinal lumen and the villi within 1 day post infection.  相似文献

Significant protection against infection with 10 or 30 metacercariae of Fasciola hepatica was conferred on naive rats by the passive transfer of serum derived from rats which had been exposed to primary and challenge infections with 5 or 10 and 30 or 20 metacercariae respectively. Immune serum did not have a pronounced effect on the mortality of metacercariae in vitro. However, its presence was associated with the formation of a precipitate on the tegument of each metacercaria and in the culture medium. The precipitate contained rat antibody and other components, presumably parasite antigens, which elicited the formation of antibody when the precipitate was injected into rats. Viability of metacercariae cultured in immune and normal sera as well as freshly excysted specimens was tested in rats by intraperitoneal infection. Metacercariae cultured in immune serum did not develop. By comparison with the viability of freshly excysted metacercariae, that of some metacercariae cultured in normal serum was impaired; this was attributed to inadequacies in the culture technique. A relationship between precipitate formation in vitro and impaired viability of metacercariae in vivo has yet to be established.  相似文献

Infections of M. corti in rats were compared with those in mice. The recoveries of parasites and their distribution were examined for 60 days after infection. During this period continuous and extensive multiplication occurred in mice. In rats there was an initial multiplication of tetrathyridia in the first 10 days followed by a decline in numbers. The relative distribution of tetrathyridia between the peritoneal cavity and liver was similar in both hosts.  相似文献

The serum of Helix pomatia agglutinates enzyme-treated erythrocytes and also possesses opsonizing properties. The agglutinating as well as the opsonizing activity could be inhibited by N-acetylglucosamine, indicating an identicalness of these serum components. As this observation supports the hypothesis that agglutinins may function as opsonins, purified agglutinins from the albumin gland of Helix pomatia, from the sponge Axinella polypoides, and Con A were utilized to sensitize foreign cells prior to their injection into the hemocoel of H. pomatia. Helix agglutinin revealed a strong opsonic effect on the elimination of the nonself particles from the circulation of the snail. It is assumed that serum opsonins of H. pomatia may couple certain nonself materials to the surface of cells in different clearance organs and that hemocytes possess membrane-associated agglutinins which mediate their attachment to trapped foreign particles.  相似文献

Dobson C. & Owen M. E. 1978. Effect of host sex on passive immunity in mice infected with Nematospiroides dubius. International Journal for Parasitology8: 359–364. Female C3H but not Quackenbush (Q) mice harboured fewer Nematospiroides dubius than male C3H and Q mice. Both strains lost worms 21 days after infection. C3H and Q mice became progressively immune to infection following 4 sequential doses of N. dubius larvae and showed a sex resistance to infection. Hypothymic nu/nu CBA Balb/c mice did not show these effects on N. dubius infection. The reciprocal transfer of male and female immune mesenteric lymph node cells (IMLNC) to syngeneic male and female recipients showed that the female environment enhanced the protective qualities of both male and female IMLNC but the male environment suppressed these effects. Gonadectomized male and female recipients of male and female IMLNC had levels of infection similar to the entire female recipients. Serum from immune female donor mice protected both male and female recipients better than immune serum from male donors, but female mice in each treatment group were better protected than male mice. Immune serum transferred greater levels of protection then IMLNC to recipient mice against N. dubius infections. These data are consistent with the conclusion that the male environment suppresses lymphocyte activity and the production of protective antibodies and additionally may depress the effectiveness of sensitized lymphocytes and antibodies in ejecting N. dubius. On the other hand the female environment does not appear to adversely affect the mobilization of the protective immune response and may enhance immune effector mechanisms in protecting mice against N. dubius infections.  相似文献

The effect of dexamethason on Babesia hylomysci and B. microti was investigated in LACA mice. The drug enhanced both infections by depressing the immune mechanisms of the host when treatment was initiated before parasite inoculation, but had no effects on established and subpatent infections. The degree of parasitemia in the treated mice seemed to depend on the tropism of either parasite toward mature erythrocytes or reticulocytes. B. hylomysci, which favors mature erythrocytes, produced fulminating infections in treated mice. B. microti, which prefers reticulocytes, produced similar parasitemia patterns in treated and untreated mice, but only the treated mice succumbed to the infection. The drug, which suppressed cellular proliferation in the spleens of infected animals, together with its direct lympholytic effects, drastically changed the architecture of the organ.  相似文献

Comparisons were made between glucose, sucrose, and dimethyl sulfoxide (DMSO) as cryoprotective agents for the hemoprotozoan parasite, Babesia rodhaini, using infectivity for mice as the criterion of survival. Concentrations of the cryoprotectants tested were from 0.1 to 0.5 M for the sugars, and 1.5 to 2.5 M for DMSO. Glucose and sucrose were comparable as cryoprotectants, although glucose reduced infectivity of the parasites slightly more than did sucrose at above-freezing temperatures. When sucrose and DMSO were compared for cryoprotection during cooling to ?196 °C at nominal rates of 5, 100, and 500 °C/min, parasite survival varied with the type and concentration of cryoprotectant, but was higher in blood containing DMSO at all three cooling rates. The percentages of parasites that survived cooling at 100 °C/min and frozen storage in the presence of DMSO ranged from 20 to 36%.  相似文献

The pathology of severe Babesia argentina infections in splenectomized calves was studied. The calves were infected by intravenous inoculation of 109–1010B. argentina and given 0.1 mg/kg betamethasone to enhance the parasitemia. Hematological changes observed during detailed studies of the course of infection in eight calves, three of which subsequently died, included thrombocytopenia, leucopenia, and reduced fibrinogen levels. The prothrombin time and partial thromboplastin time were prolonged in all three calves tested, and pathological levels of fibrinogen degradation products were detected in both of two calves tested. Massive pulmonary edema was a constant finding at autopsy of 24 fatal cases. Histopathological examination revealed widespread fibrin thrombi in capillaries and larger vessels of lung, in capillaries of renal glomeruli, and in hepatic sinusoids. The findings established the occurrence of disseminated intravascular coagulation in the acute infections studied.  相似文献

Dalgliesh R. J. and Stewart N. P. 1982. Some effects of time, temperature and feeding on infection rates with Babesia bovis and Babesia bigemina in Boophilus microplus larvae. International Journal for Parasitology12: 323–326. Percentages of larval ticks in which Babesia bovis and B. bigemina parasites could be detected (infection rates) were determined after the larvae had been exposed to temperatures between 9°C and 27°C for periods of 1–35 days and then either fed on calves or heated at 37°C to stimulate babesial development. Infection rates with both species increased during 2–4 weeks after the larvae hatched, regardless of the temperature of exposure. Infection rates with B. bovis were higher after exposure of larvae to 14°C than to 27°C. This effect was less pronounced with B. bigemina. Infection rates were higher in fed larvae than in unfed, ‘heat stimulated’ larvae. The findings indicate that infected larval ticks become more efficient vectors of Babesia during the first 2–4 weeks after hatching and that repeated sampling of a tick population is necessary to determine valid infection rates.  相似文献

Babesia rodhaini parasites in murine blood containing 1.5 m DMSO were frozen at two rates, as judged by the duration of the “freezing plateau”, then cooled to ?196 °C and rewarmed at two rates to detect interactions between the duration of the plateau and rates of subsequent cooling and rewarming. Infectivity tests showed that fast and slow freezing (plateau times of about 1 sec and 30 sec, respectively) had similar effects on parasite survival when cooling was at 130 °C/min and warming was at 800 °C/min. However, when either the cooling rate was increased to 3500 °C/min or the warming rate was decreased to 2.3 °C/min, fast freezing decreased parasite survival more than did slow freezing. It is suggested that fast freezing accentuated the damaging effects of fast cooling and slow warming by increasing intracellular ice formation.  相似文献

目的:探讨被动吸烟对雌性Wistar大鼠卵巢结构和功能的影响。方法:雌性Wistar鼠32只,随机分为对照组和被动吸烟组,对照组在自然环境下饲养,吸烟组在自制的吸烟箱内被动吸烟3个月,吸烟三个月后,对两组分别进行秤重,然后处死。用测量仪器测量母鼠卵巢的大小、重量,并做光、电镜观察,免疫组织化学检测ER、PR、FSHR、LHR在卵巢中的表达,所得数据进行统计学处理。结果:吸烟三个月大鼠卵巢充血性囊性退变,皮质萎缩,卵泡颗粒细胞排列紊乱;生长卵泡的颗粒细胞松散变性,排列不整齐;卵泡数减少,成熟的卵母细胞数目明显减少;髓质血管收缩,减少;间质疏松。②电镜对照组可见多个卵泡细胞,细胞内有空白颗粒及高密度颗粒。吸烟组可见多个颗粒细胞层,内有增粗的内质网、核糖体;核周隙局部增宽,线粒体模糊;线粒体水样变、空泡样变,线粒体嵴断裂、模糊;内质网脱颗粒样变。与没有吸烟的正常大鼠B组比较,吸烟大鼠的ER、PR、FSHR、LHR减少(P<0.05)。结论:被动吸烟组雌性卵巢充血性囊性退变,皮质萎缩,卵泡颗粒细胞排列紊乱。  相似文献

Babesia gibsoni causes canine babesiosis. Here, we describe the identification and characterization of a novel gene, BgP22, containing an open reading frame of 621 bp and encoding a 22-kDa protein from B. gibsoni, as a serodiagnostic candidate. The recombinant BgP22 (rBgP22) was expressed and used as an antigen to produce anti-rBgP22 sera in mice. Using these anti-rBgP22 sera, a native 22-kDa protein was recognized by Western blot analysis and observed in the membrane of the parasites by immunofluorescent antibody tests (IFAT). The enzyme-linked immunosorbent assay (ELISA) using the rBgP22 detected specific antibodies to this protein in the sera of dogs experimentally and naturally infected with B. gibsoni in chronic stage. Furthermore, it did not show a cross reaction with the closely related apicomplexan parasites, indicating that the rBgP22 could be used as a diagnostic antigen for a detection of the chronic carrier stages of B. gibsoni infection.  相似文献

An attenuated live vaccine containing Babesia bovis and B. bigemina cultured in vitro with a serum-free medium was assessed for its clinical protection conferred of naïve cattle, under natural tick-challenge in a high endemicity zone to Babesia spp. Three groups of six animals were treated as follows: group I (GI) received a vaccine derived from parasites cultured with a free-serum medium; group II (GII) were immunized with the standard vaccine, with parasites cultured in a medium supplemented with 40% (v/v) bovine serum; and a control group (GIII) inoculated with non-infected bovine erythrocytes. Inocula were administered by IM route. Experimental animals were kept during 23 days after vaccination in a cattle farm free of ticks and Babesia spp. Thereafter, cattle were moved to a high endemicity farm for natural exposure to Babesia spp. transmitted by Rhipicephalus microplus ticks. Protection against clinical babesiosis was observed in bovines belonging to GI (100%) and GII (83.33%), while the control animals (GIII) were not protected, and showed severe clinical signs, closely related to babesiosis, were observed for at least three consecutive days during the challenge. These were fever, anemia, which were measured simultaneously, and circulating parasites were detected by optic light microscopy. All cattle showed B. bovis and B. bigemina in stained blood films during the challenge; B. bovis antibody titers were higher than those to B. bigemina in GI and GII, and lower titers were determined in GIII. The protective capacity of the vaccine derived from B. bovis and B. bigemina cultured in vitro in a serum-free medium was demonstrated.  相似文献

Despite the well-known importance of an adequate colostral immunoglobulin (Ig) transfer to calf health and survival, failed transfer of passive immunity (FTPI) remains a widespread problem in dairy farming. The aim of this study was to investigate the management factors associated with FTPI in newborn calves, evaluating particularly the combined effect of delivery time, amount and quality of the first colostrum meal. The study was conducted from March to August 2014 on 21 Italian dairy farms. Farmers were asked as first to answer a farm-level questionnaire on calf management. Blood sampling was then performed on overall 244 calves (1 to 5 days of age) born from Holstein cows, and a sample of the first colostrum meal of each calf was collected. Individual information on calves and the respective colostrum management were recorded. Serum and colostrum Ig concentrations were assessed by electrophoresis. A mixed effects multivariable logistic regression model was used to investigate the association of the variables obtained from both the management questionnaire and the individual calf data with FTPI (calf serum Ig concentration <10.0 g/l). A cumulative colostrum management score (CMS) that considered delivery time, amount and quality of the first colostrum meal was generated for 236 calves, with higher values indicating better colostrum management. Overall, 41.0% of the calves were found having FTPI, and within-farm percentage of FTPI was over 20.0% in 71.4% of the farms. The risk of having FTPI was higher both for Holstein purebred calves compared with Holstein-beef crossbreds and for females compared with males. Moreover, it increased by 13% with every hour of delay of the first colostrum meal provision since birth, whereas it decreased by 59% and 3%, respectively, with every additional liter of colostrum given and every additional gram of Ig per liter contained in the colostrum fed. Calf serum Ig concentration varied significantly according to the CMS, increasing by 1.53 g/l with every additional CMS point. In order to completely avoid FTPI, calves should receive at least 2.5 l of high-quality colostrum (Ig concentration >87.6 g/l) within 1.0 h from birth. Considerable improvements are still needed about colostrum management for newborn calves in dairy farms. The results of this study will help in developing farm-specific programs for reducing the occurrence of FTPI.  相似文献

Passive transfer of natural resistance to Schistosoma mansoni (PR-1 strain) has been successfully accomplished in the snail intermediate host, Biomphalaria glabrata (PR albino, M-line strain). Injection of serum (cell-free hemolymph) from a naturally schistosome-resistant strain of B. glabrata (10-R2) into PR albino snails induced a complete protection from a primary infection with the parasite in 29 of 48 snails (60.4%). In comparison, inoculation of homologous PR albino serum or heterologous proteins (fetal calf serum) had no effect. Moreover, this protection could be induced 24 hr prior to, or 24 hr after, exposure to the parasite, although heating of 10-R2 serum to 70 C for 30 min destroyed its protective ability. When in vitro transformed sporocysts were preincubated in 10-R2 or PR albino serum and then were injected into susceptible snails, a high level of infection (88.5 and 83.3%, respectively) was produced in both groups. Thus, the 10-R2 serum factor does not appear to be mediating specific parasite recognition by host hemocytes. Alternatively, our results suggest that 10-R2 serum possesses a heat-labile factor which specifically activate B. glabrata hemocytes to encapsulate and destroy sporocysts whereas PR albino serum lacks this factor.  相似文献

Previous work had shown that a sequence homologous to the gene encoding class II hydrophobin cerato-ulmin from the fungus Ophiostoma novo-ulmi, the causal agent of Dutch Elm Disease (DED), was present in a strain of the unrelated species Geosmithia species 5 (Ascomycota: Hypocreales) isolated from Ulmus minor affected by DED. As both fungi occupy the same habitat, even if different ecological niches, the occurrence of horizontal gene transfer was proposed. In the present work we have analysed for the presence of the cerato-ulmin gene 70 Geosmithia strains representing 29 species, isolated from different host plants and geographic locations. The gene was found in 52.1 % of the strains derived from elm trees, while none of those isolated from nonelms possessed it. The expression of the gene in Geosmithia was also assessed by real time PCR in different growth conditions (liquid culture, solid culture, elm sawdust, dual culture with O. novo-ulmi), and was found to be extremely low in all conditions tested. On the basis of these results we propose that the cerato-ulmin gene is not functional in Geosmithia, but can be considered instead a marker of more extensive transfers of genetic material as shown in other fungi.  相似文献

Artemisone was evaluated, in in vitro and in vivo, for control of bovine babesiosis caused by Babesia bigemina and Babesiabovis parasites. In vitro, artemisone reduced parasitemia in a dose-dependent manner: the inhibitory effects increased gradually, reaching a maximum inhibition of 99.6% and 86.4% for B. bigemina and B. bovis, respectively 72 h after initiation of treatment with initial parasitemia of 0.5%. In calves infected with either B. bigemina or B. bovis artemisone treatment was well tolerated and prevented development of acute babesiosis in all animals except for one B. bovis-infected calf. The treatment did not eliminate all blood parasites, and recovered animals carried a persistent low-level infection. Treatment with artemisone may be useful as an alternative drug for preventing the pathology that results from babesiosis, without interfering with acquired immune protection following recovery from an acute babesiosis infection or vaccination.  相似文献

The indirect fluorescent antibody test was used for the serological comparison of the parasites called Babesia argentina and Anaplasma marginale in Australia and Bolivia, South America. Cross-testing performed with antigens and antisera prepared in the two continents proved serological identity for both parasites. Because a similar study by Goldman & Rosenberg (1974) showed serological identity between B. bovis and B. argentina, by the law of priority the small Babesia of Australia and South America should be called B. bovis.
