The characteristics of acyl CoA:cholesterol acyltransferase (ACAT; EC 2.3.1.26) in microsomes prepared from human term placenta were studied and the rate of incorporation of [1-14C] oleoyl CoA into cholesteryl esters was measured. The apparent Km of the enzyme for [1-14C] oleoyl CoA was 38 ± 9 μm and the V for the reaction was 15 ± 6 pmol × mg? protein × min?1. The Hill coefficient for the reaction was 1.2, indicative of some degree of positive cooperativity. Cholesterol, added to the incubation mixture, did not influence ACAT activity, indicating that endogenous microsomal cholesterol served as an effective substrate for the placental ACAT enzyme. However, [1,2-3H]cholesterol in the presence of oleoyl CoA was incorporated into cholesteryl esters by placental microsomes. When progesterone was present in the incubation mixture at a concentration of 20 μm, ACAT activity was inhibited 50%. Pregnenolone, 5α-dihydroprogesterone, 17α-hydroxyprogesterone, deoxycorticosterone, dehydroisoandrosterone, androstenedione, testosterone, and estradiol-17β also inhibited ACAT activity, whereas corticosterone, cortisol, and estriol had little effect. These results are supportive of the view that ACAT activity in human placenta may be regulated by endogenously synthesized steroid hormones.  相似文献

The analysis of fecal ovarian steroids provides a powerful noninvasive method to obtain insights into ovulatory cycles, gestation length, and the timing of sexual interactions relative to the periovulatory period in wild primates. Techniques developed to collect and assay feces from free-ranging muriqui monkeys (Brachyteles arachnoides) for estradiol and progesterone yield the first explicit reproductive data on this species, and provide the first opportunity to evaluate the timing of observed copulations with muriqui ovarian cycles. Hormonal profiles from seven females indicate average cycle lengths of 21.0 ± 5.4 days (n=20). Females conceived after 3–6 ovulatory cycles. Gestation length averaged 216.4 ± 1.5 days for the five females for which conception cycles were sampled. Discrete copulation periods spanned an average of 2.1 ± 1.2 days (n=29), with intervals between these concentrated periods of copulations averaging 15.6 ± 6.7 days (n=20). There were no significant differences among females in cycle lengths, copulation period lengths, or copulation interval lengths. Ejaculation was visible following 71.8 ± 26.7% of copulations during the females' preovulatory periods. All females copulated outside the periovulatory period. The proportion of copulation days outside the periovulatory period was slightly greater (p=0.08) for primiparous females (64.8 ± 28.3%) than for multiparous females (28.7 ± 19.7%). Am. J. Temperature effects on antifreeze metabolism were investigated in two populations (northern and southern) of the golden rod gallfly, Eurosta solidaginis. Sorbitol production was temperature dependent and was triggered by short-term exposure to < +10°C. The maximal rate sorbitol synthesis occurred at 0°C. For both populations, sorbitol was rapidly catabolized during warm acclimation at +20°C. During the first 12 h of warm acclimation, sorbitol levels decreased by 36% (19.7 ± 0.6) to 12.6 ± 1.2 μg/mg) and by 83% (to 3.3 ± 1.7 μg/mg) after 48 h in the northern population. The southern population decreased sorbitol levels 64% (11.8 ± 0.69 to 4.2 ± 0.62) after 48 h. The southern population resynthesized more sorbitol than did the northern population upon re-exposure to 0°C. Glycerol levels increased linearly during the experimental period independent of temperature.  相似文献

Changes in the brain’s neuroactive steroid levels, behavior in the open field, and the anxious-phobic status of male and female rats in the course of development have been studied. An increase in the motor and exploratory activity and emotionality in rats of both sexes in the pubertal period and a decrease in their values in mature and old animals have been detected. Anxiety has no sexual dimorphism in adult animals; it is significantly higher in males than in females in the prepubertal and pubertal periods of development and is higher in old females than in males of the same age. An increase in the level of corticosterone in some brain structures in maturing and old rats has been found; the testosterone concentration increases in one-month-old and adult animals but decreases in old individuals, while the estradiol concentration in all studied brain structures of male and female rats was low in all periods of postnatal life. Correlation analysis has shown modulation by steroid hormones of the changes in behavioral responses during development.  相似文献

This study was designed to investigate the role of dopamine in the control of aldosterone secretion in three frequently used laboratory animals. Five New Zealand rabbits, five mongrel dogs and five rhesus monkeys received metoclopramide (MCP) (200 μg/kg iv) and blood samples were collected at 0,5,15,30 and 45 minutes after drug administration. MCP had no effect on plasma aldosterone concentrations at any sampling time in the rabbits or dogs. However, MCP produced a rapid and marked increase in plasma aldosterone from 6.5±0.6 ng/dl to 18.1±2.8 ng/dl at 5 min. and a maximum level of 40.5±4.4 ng/dl at 10 min. after drug administration in the monkeys. MCP had no significant effect on plasma cortisol or plasma renin activity levels in the three species. Prolactin rose in the monkeys from 8.6±1.2 ng/ml to a maximum of 123.5±8.5 ng/ml at 15 min. after MCP. Administration of MCP resulted in a rise in plasma 18-hydroxycorticosterone in the monkeys from 12.5±1.4 ng/dl to a maximum concentration of 50.0±5.1 ng/dl 15 min. after drug administration. Plasma corticosterone, 11-deoxycorticosterone, and 18-hydroxydeoxycorticosterone were not altered by MCP. Although unlikely, it is possible that ketamine may have accounted for some of the changes in plasma aldosterone and 18-hydroxycorticosterone observed after metoclopramide in the monkeys. The findings suggest that dopamine modulates aldosterone biosynthesis in the monkey probably by regulating glomerulosa 18-hydroxylase activity.  相似文献

Methods für the determination of urinary free corticosterone, free aldosterone and free 11-deoxycorticosterone (DOC) in rats are described. The free corticosteroids were measured in urine samples of 0.1–0.5 (2.0) ml by radioimmunoassay after purification by column chromatography. The validity of the methods is demonstrated by the data of the free urinary corticoids under basal conditions and after adrenal suppression and various forms of adrenal stimulation. The basal excretion of free corticosterone, free aldosterone and free DOC was 123.71 ± 15.31 (x? ± SD), 3.87 ± 1.29 and 10.61 ± 2.24 ng/day, respectively, exhibiting a decrease to 26.20 ± 5.21, 1.05 ± 0.47 and 1.35 ± 1.20 ng/day after adrenal suppression by dexamethasone. Irrespective of the mode of adrenal stimulation i.e., synthetic ACTH and systemic (cold, hunger) or neurotrophic (ether, reserpine) stress stimuli free corticosterone increased to about 450 ng/day, while free aldosterone excretion decreased during hunger and cold and was strongly enhanced after the application of reserpine. Furthermore, determination of urinary free DOC, which increased by a factor of 4, may be applied in the metyrapone test. There was a good correlation between the excretion of free corticosterone and that of free aldosterone and free DOC under basal conditions and after ACTH application, demonstrating that ACTH is responsible for the secretion of all the 3 corticoids measured. It is concluded, that the measurement of the urinary excretion of corticosterone, aldosterone and DOC is a valuable parameter of adrenal function in rats. Furthermore, in small laboratory animals like rats steroid measurements in urine are often more advantageous than Measurements in plasma.  相似文献

The relationship among impaired selenium status, lipid peroxidation, and liver function was examined in 19 hospitalized patients with severe alcoholic cirrhosis. Plasma selenium was found to be significantly lower (mean±SD: 54±13 μg/L) than in healthy controls (83±11 μg/L) and plasma malondialdehyde, assessed as thiobarbituric acid reactants, which reflects lipid peroxidation, was increased (2.0±1.2 μmol/L vs <1.2 μmol/L in controls). The mean14C aminopyrine breath test, an indicator of liver function, was lower than normal (2.7±1.9 vs 6.3±0.9% in controls) and found to be significantly correlated with plasma selenium (r=0.59,p<0.05). A prospective, randomized selenium supplementation trial was conducted in a group of 16 patients who received either daily 100 μg selenium as enriched yeast during 4 mo or a placebo. Among the 10 patients who completed the study, plasma selenium significantly increased in the supplemented group (n=4; before: 58±10 μg/L, and after 101±12 μg/L,p<0.01) contrary to the placebo group (n=6, before: 47±10 μg/L, after: 57±9 μg/L, n.s.),14C aminopyrine breath test improved in three out of four selenium-supplemented patients and in three out of six placebo patients, but the small number of patients did not allow statistical evaluation. These results demonstrate that low selenium status in alcoholic cirrhosis is correlated to liver function and could be improved by supplementation.  相似文献

In this study we compared the insemination efficiency of two alternative mating tactics (courtship and sneak mating) in the guppy Poecilia reticulata by quantifying the number of sperm delivered during a copulation. During a single copulation, guppies delivered between zero and 92% of the sperm available, as determined by mechanically stripping the males'' sperm reserve at rest. The absolute number of sperm delivered after courtship was three times larger than that delivered through sneak mating; nonetheless, the variance was large with both tactics and the two distributions largely overlapped. The number of sperm available at rest increased with male size. With both tactics, the number of sperm delivered was positively correlated with the sperm available. Contrary to courtship copulations, in sneak copulations there was no correlation between the number of sperm delivered and male size. However, once the data were standardized for sperm reserve, small males delivered a larger proportion of their available sperm during sneak copulation. The rate of sexual acts (sigmoid and thrust rate) before copulation was not correlated with the number of sperm available. After the occurrence of a copulation in both the courtship and sneak copulation groups, the sexual activity of the male decreased in proportion to the amount of sperm he previously inseminated.  相似文献

Oxytocin (OT) is a versatile neuropeptide that is involved in a variety of mammalian behaviors, and its role in reproductive function and behavior has been well established. The majority of pharmacological studies of the effects of OT on male sexual behavior have focused on the paraventricular nucleus (PVN), ventral tegmental area (VTA), hippocampus, and amygdala. Less attention has been given to the medial preoptic area (MPOA), a major integrative site for male sexual behavior. The present study investigated the effects of intra-MPOA administration of OT and (d(CH2)51, Tyr(Me)2, Thr4, Orn8, Tyr-NH29)-vasotocin, an OT antagonist (OTA), on copulation in the male rat. The relationship between OT receptor (OTR) binding levels in the MPOA and sexual efficiency was also explored. Microinjection of OT into the MPOA facilitated copulation in sexually experienced male rats, whereas similar injections of an OTA inhibited certain aspects of copulation but had no significant effect on locomotor activity in an open field. Contrary to expectation, sexually efficient males had lower levels of OTR binding in the rostral MPOA compared to inefficient animals. The present data suggest that OT activity in the MPOA is not necessary for the expression of male sexual behavior but is sufficient to facilitate copulatory behaviors and improve sexual efficiency in sexually experienced male rats. These data also suggest that OTR activity in the MPOA stimulates anogenital investigation, facilitates the initiation of copulation, and plays a role in the sensitization effect of the first ejaculation on subsequent ejaculations.  相似文献

A steady-state kinetic analysis of the activation of bovine Factor X, by bovine Factor Xa, was undertaken. The activation was found to be dependent on the presence of divalent cations; Ca2+ showing the greatest stimulatory effect and Mn2+ exhibiting a lower degree of activity for this reaction. Although Sr2+ and Mg2+ were ineffective when present alone, each contributed synergistically to the activation rate at suboptimal levels of Ca2+. The effect of phospholipid (phosphatidylcholine:phosphatidylserine, 4:1, w:w) on the rate of activation and on the activation pathway was investigated. Phospholipid (PL) concentrations of up to 40 μm had no effect on the activation rate; whereas, concentrations of 40–180 μm were slightly inhibitory. In the absence of PL, the major product of the activation was Factor α-Xa, while in the presence of PL, lower-molecular-weight forms of Factor X (Factor β-X) and Factor Xa (Factor β-Xa were produced. At saturating levels of Ca2+, the Km app for the activation, at pH 7.4 and 37 °C, in the absence of PL, was found to be 0.6 ± 0.1 μm and the V was 1.7 ± 0.3 mol Factor X cleaved min?1 mol?1 Factor Xa. The corresponding values, in the presence of 90 μm PL, were 1.4 ± 0.2 μm and 2.2 ± 0.2 mol Factor X cleaved min?1 mol?1 Factor Xa.  相似文献

In order to obtain reference values from normal babies, Cr status of full-term newborns has been studied. Plasma and urine values were (mean±SEM) 0.7±0.1 μg/L and 0.9±0.3 μg/L, respectively, for the first month of life (n=19), and 0.6±0.1 μg/L and 0.8±0.2 μg/L for the second-to-third-month period (n=31). Premature newborns (gestational age 28–36 wk) were compared to these control values; concentrations were 0.9±0.1 μg/L and 1.1±0.2 μg/L for the first month (n=47), and 1.0±0.2 μg/L and 1.5±0.3 μg/L for the second to third months (n=27). For the whole group, there was a positive correlation between plasma and urine concentrations (p=0.0001); multiple regression analysis was performed between plasma levels and gestational age at birth (p=?0.002) and postnatal age (NS). Plasma levels of prematures and full terms were statistically different (p=0.03) only for the second- to third-month period. It is suggested that these high Cr levels result from high dietary intakes and/or high absorption rates.  相似文献

Trace elements (selenium, zinc, copper) \gb2 microglobulin levels, CD4, and CD8 cell counts have been determined in 80 HIV1 seropositive patients. The study group consisted of 19 females and 61 males with age mean of 35±10 yr, at stage IV of infection (CDC—Atlanta classification) and treated by AZT. No severe renal or liver diseases or hypoalbuminemia were observed in this group. Se values were significantly lower than in normal adults, 48.3±17 μg/L vs 71±12 μg/L; Zn was moderately diminished, 1±0.2 mg/L vs 1.2±0.2 mg/L, whereas copper values were in the normal range, 1.2±0.3 mg/L vs 1.1±0.5 mg/L. Se or Zn deficiency was found in 60 and 30 subjects, respectively. Blood Se and Zn decreases were associated in 23 patients. Moreover, all patients showed higher \gb2 microglobulin values than the upper normal limit of 2.4 mg/L. Negative correlations were found between Zn and \gb2 microglobulin (p<0.005) and between Se and \gb2 microglobulin (p<0.05). Moreover, there was a positive correlation between Se and Zn values (p<0.05). Nineteen subjects died 1 yr later (group I), and 61 remained alive (group II). With respect to the clinical evolution, a significant difference between both groups was found in Se and \gb2 microglobulin levels as well as in CD4 cell counts. The correlations previously observed persisted in group II, whereas no correlation was noted in group I. In addition, the patients of group I had significantly lower Se values, which were below 30 μg/L in 10 cases. These results confirm the prevalence of abnormalities in Se and Zn levels and their relationships with nonspecific markers of immune system activity in more advanced HIV disease. Impairment of trace element status and mainly Se status appeared, at least partially, to reflect the disease activity/progression and subsequently the immune dysregulation.  相似文献

We examined copulation patterns and associated sexual behaviour in the colonial Eurasian Griffon vulture Gyps fulvus during the pre-laying period. Eurasian Griffon vulture pairs conducted an average of 71.7 copulation attempts per clutch, with an average copulation frequency of 1.2 copulation attempts per day. Low copulation frequencies compared to other raptors and absence of mate-guarding suggest that this species does not possess adaptive behaviour aimed at increasing paternity assurance. However, the gradual increase in copulations during the fertile period is consistent with the sperm competition hypothesis.  相似文献

This study examined sexual and other social behavior in relation to menstrual cycle phase in four mixed-sex social groups of Cebus apella. Groups consisted of two adult males and either six or nine adult females. Menstrual cycles of high (rank 1–2) and low (rank 3–7) dominance-rank females from each group were monitored via vaginal swabs, and correlated with data collected from ongoing behavioral observations. Only cycles bounded by positive detection of menstrual blood were included in this analysis (n = 15 females, 182 cycles; mean ± s.d., cycle length = 20.8 ± 1.2 days). Rates of copulation and female solicitation of males varied significantly with cycle phase, with highest rates at midcycle. While total rates of solicitation and copulation did not vary with female dominance rank, copulation rates with the dominant male were significantly greater for high ranking females than for low ranking ones. Variance observed in affiliative and agonistic behaviors, including those with males, was attributable to female rank rather than cycle phase. Females and males were also observed attempting to interfere with copulations of lower ranking same-sex individuals. Although further study, particularly of wild populations, is needed, these results indicate that female-female reproductive competition should be included as a component of the capuchin breeding system. Precopulatory courtship plays an essential role in the insemination process and influences postcopulatory behavior between males and females. Male precopulatory oral stimulation of female genitals is rare for invertebrates. Here, we describe an intriguing oral sexual courtship in a cryptic desert beetle Platyope mongolica Faldermann. The males repeatedly contact the female''s genitals using their mouths to gain consent to mate. Furthermore, the rate at which males contact the female''s genitals relates to the copulation success in a series of observations. However, interference in oral sexual contacts decreased the proportion of successful copulation. Further no‐choice tests found homosexual behavior between males with antenna removed. We report the precopulatory oral sexual behavior and its important role for copulation success in P. mongolica for the first time. These findings highlight the significance of oral sexual courtship in sexual selection.  相似文献

The mating behavior of Mahanarva spectabilis (Distant) was studied under laboratory conditions. Virgin adults were individually maintained in transparent cages for observation of the mating sequence. Mating behavior of 23 couples was monitored during the photophase portion of the day. Copulation was observed in 17 of the 23 pairs. The mating sequence includes male approaching the female, mounting of the female, and copulation. Of the 17 mating pairs, 70.6 % mated on the second day after emergence. Most of the copulations began between 8 and 9 h after the onset of photophase. The average duration of copulation was 268?±?24.9 min, and most pairs mated once or twice during their lifetimes. The mating behavior of M. spectabilis is important to understand because it determines the age and ideal time for further behavioral testing, which is essential for determining the cues involved in the communication system of the species.  相似文献

We have studied the hepatic microsomal 17β-hydroxysteroid dehydrogenase (17β-HSD) capacity of koala (Phascolarctos cinereus) and tammar wallaby (Macropus eugenii). A detailed comparison of the activity in hepatic fractions from koala and rat was made. Hepatic microsomal NADP-supported 17β-HSD activity was significantly higher in koala (11.64±3.35 nmoles/mg protein/min), (mean±S.D.) than in tammar wallaby liver (1.52±0.79 nmoles/mg protein/min). However, when NAD was utilised as cofactor the activity was similar in both marsupial species (2.83±2.03 nmoles/mg protein/min, koala; 0.70±0.71 nmoles/mg protein/min, tammar wallaby). Data for rat indicated a cofactor preference for NAD rather than NADP (17.94±6.40 nmoles/mg protein/min, NAD; 2.18±1.04 nmoles/mg protein/min, NADP). Michaelis–Menten parameters for the kinetics of 17β-HSD testosterone oxidation by NADP and NAD were determined in the koala. The Km for testosterone was of the order of 10.0–24.0 μM (n=6) irrespective of the cofactor used, whilst the Km for NADP was 0.28–0.43 μM (n=2) and for NAD was 13.9–18.5 μM (n=2). 17β-estradiol was found to be an inhibitor of both NAD- and NADP- supported 17β-HSD activity. These findings indicate that NADP-mediated, but not NAD-mediated testosterone dehydrogenation is a major pathway of steroid biotransformation in koala liver; the reaction is less extensive in fractions from wallaby, human and rat. Such species-related differences in cofactor preference may contribute along with species differences in gene expression to observed rates of 17β-HSD activity in mammals.  相似文献

Hippocampal CA1 neurons express both mineralocorticoid and glucocorticoid receptors. Due to the difference in affinity of the two receptor types for corticosterone and variations in endogenous steroid levels, occupation of the receptors will range between a situation of predominant mineralocorticoid receptor activation and conditions where both receptor types are occupied. It was observed that local signal transduction is regulated by activation of the corticosteroid receptors. Particularly, transmission mediated by biogenic amines appears to be sensitive to steroid control. The data indicate that cholinergic and serotonergic responses are small with predominant mineralocorticoid receptor activation, while additional glucocorticoid receptor activation results in large responses; the reverse has been found for noradrenalin. The steroid-dependent control over transmission by biogenic amines will influence local excitability and therefore functional processes in which the hippocampal system is involved.  相似文献
