本文旨在探讨线粒体ATP敏感钾(mitochondrial ATP-sensitive K+,MitoKATP)通道对大鼠肺动脉平滑肌细胞低氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)表达和细胞增殖的影响。原代培养大鼠肺动脉平滑肌细胞,分为常氧对照组、常氧+diazoxide(MitoKATP通道的选择性开放剂)组、常氧+5-hydroxydecanoate(5-HD,MitoKATP通道的选择性阻断剂)组、低氧对照组、低氧+diazoxide组、低氧+5-HD组,共6组,分别应用罗丹明123荧光技术检测各组大鼠肺动脉平滑肌细胞的线粒体膜电位,免疫组化检测HIF-1α的表达及酶联免疫检测仪检测细胞增殖的变化。结果显示,常氧+ diazoxide组与常氧对照组比较,罗丹明123荧光、HIF-1α表达及细胞增殖明显增强(P〈0.05);低氧+diazoxide组与低氧对照组比较,罗丹明123荧光、HIF-1α表达及细胞增殖明显增强(P〈0.05):常氧+5-HD组与常氧对照组比较,罗丹明123荧光、HIF-1α表达、细胞增殖没有明显变化(P〉0.05);但低氧+5-HD组与低氧对照组比较,罗丹明123荧光明显减弱、HIF-1α表达及细胞增殖有所减弱(P〈0.05)。结果提示:MitoKATP通道的开放能引起大鼠肺动脉平滑肌细胞线粒体膜去极化,并可以促进HIF-1α的表达及细胞增殖。  相似文献

Since hypothermic conditions augment sensitivity to vasoactive amines like serotonin (5-HT) and 5-HT is associated with the etiology of Raynaud's phenomenon, this amine perhaps plays a role in cold-induced vasoconstriction. To determine if 5-HT participated in normal peripheral cooling and if ketanserin (KET), a 5-HT blocker, modulated such cooling, four groups of New Zealand white rabbits (N = 33) were studied. The femoral artery was cannulated to allow perfusion of a hindlimb. Thermistors were implanted in the footpad and rectum. The hindfoot was exposed to a 15 degrees C bath for 30 min, while footpad and rectal temperatures were recorded. During cold exposure, 5-HT (5 x 10(-2) M, group 1), KET (0.1 mg/kg) + 5-HT (group 2), KET (group 3), or saline (group 4) was perfused through the hindlimb. Groups 2 and 3 were also pretreated with KET (0.1 mg/kg perfused over 30 min). The rabbit footpad cooled rapidly when exposed to exogenous 5-HT (group 1). KET treatment in the presence of exogenous 5-HT (group 2) was associated with a significantly (P less than 0.05) reduced cooling rate. KET treatment in the absence of exogenous 5-HT (group 3) was also associated with a significantly (P less than 0.05) elevated limb temperature when compared to controls (group 4). This suggested that endogenous 5-HT participated in limb cooling. Therefore, as noted for Raynaud's disease, 5-HT may also influence peripheral cooling of tissues free of such pathologies.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

We investigated the effects of food availability on the seasonal testicular growth in the photoperiodic house sparrow (Passer domesticus). Two experiments were performed, each lasting 4 weeks. In experiment 1, sparrows were exposed to natural (NDL; group 1), short (8L:16D; group 2) and long (16L:8D; groups 3-5) day lengths with access to food ad libitum (groups 1-3) or for 10 h (zeitgeber time (zt) 0-10, group 4; zt 0 is the time of light onset) or for 8 h (zt 8-16, group 5). Testes recrudesced under long, but not short and natural, day lengths, and the recrudescence under long days was influenced by the duration of food availability. In experiment 2, the sparrows were exposed to short (8L:16D, group 1) and long (16L:8D, groups 2-5) day lengths with access to food ad libitum (for groups 1 and 2) or for 6 h (for groups 3-5) at different times during the 16 h light period (group 3- zt 0-6, group 4- zt 5-11, group 5- zt 10-16). As the expected, the testes recrudesced only under long lengths, but the photoinduction was variable among the 4 groups. The testes grew to full size in groups 2 and 3 that received food either ad libitum or for 6 h at zt 0-6, but to sub-maximal size in the groups that received 6 h food either at zt 5-11 (group 4) or at zt 10-16 (group 5). Altogether, these results support the idea that the photoperiodic regulation of reproduction in a seasonally breeding species is influenced both by the duration and the time of food availability.  相似文献

The technical quality of 600 electrocardiograms (ECG''s) was assessed for missing leads and clipping, and graded from 1 to 5 for each of noise, lead drift and beat-to-beat drift. Three subgroups of 200 ECGs each were studied: group A, those obtained by emergency department staff (non-technicians); group B, records obtained by ECG technicians; and group C, telephone-transmitted records obtained by technicians performing all the laboratory work at a smaller, outlying hospital. Records with missing leads, clipping, grade 4 or 5 noise, grade 5 lead drift or grade 5 beat-to-beat drift were classified as unsatisfactory or rejected. With these stringent criteria the rejection rate was 71.0% for group A records, 58.5% for group B and 44.5% for group C. The proportions of records with peak quality (no missing leads or clipping, and grade 1 noise, lead drift or beat-to-beat drift) were 4.5% for group A, 5.5% for group B and 23.0% for group C. Suggested revisions in the grading of technical quality of ECGs are presented.  相似文献

为探讨合欢花对慢性应激大鼠生长和脑单胺类神经递质的影响,采用15只大鼠,设置了对照组、应激组和合欢花组3组实验。应激组和合欢花组均接受7天的应激刺激,之后合欢花组再灌胃合欢花10天。实验结束后,取3组大鼠的脑组织,用高效液相色谱法测定高香草酸(HVA)、去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)的含量。结果表明,应激组大鼠日增重显著低于对照组(P=0.011);而合欢花组大鼠的日增重极显著高于应激组(P=0.002)。应激组大鼠海马、纹状体和前额叶中的HVA含量与对照组相比,虽有升高的趋势,但无显著差异;两组间的NE、DA和5-HT也无显著差异。合欢花组大鼠海马中的HVA、DA含量明显高于应激组,而前额叶中的多巴胺和5-羟色胺,以及纹状体中的5-羟色胺均明显低于应激组。这表明合欢花对慢性应激引起的大鼠生长受抑有缓解作用,对其脑内单胺类神经递质有调节作用。  相似文献

The specificity of yeast NAD-specific isocitrate dehydrogenase for the structures of the allosteric effector 5'-AMP was examined with analogues modified in the purine ring, pentosyl group, and 5'-phosphate group. An unsubstituted 6-amino group was essential for activation as was the phosphoryl group at the 5'-position. Activity was retained when an oxygen function of the 5'-phosphoryl was replaced by sulfur (Murry & Atkinson, 1968) or by nitrogen (phosphoramidates). 2-NH2-AMP, 2-azido-AMP, and 8-NH2-AMP were active; 8-azido-AMP and 8-Br-AMP were inactive. The configuration or nature of substituents about carbons 2' and 3' of the pentosyl portion of AMP was not critical for allosteric activation since AMP analogues containing, e.g., 2',3'-dideoxyribose or the bulky 2',3'-O-(2,4,6-trinitrocyclo-hexadienylidene) substituent (TNP-AMP) were active. TNP-AMP was bound to the enzyme with fluorescence enhancement and had an S0.5 for activation similar to the S0.5 for AMP. Positive effector activity was decreased when the pentosyl moiety of 5'-AMP was replaced by the six-membered nitrogen-containing morpholine group, indicating that the pentosyl group may be critical as a spacer for the proper geometry of binding to enzyme at the 6-amino and 5'-phosphoryl groups of 5'-AMP. A comparison of molecular models of 5'-AMP with 8,5'-cycloAMP suggests that the species of 5'-AMP required for binding to the enzyme contains the purine and ribose moieties in an anti conformation and positioning of the 5'-phosphate trans with respect to carbon 4'.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Ammonia, amino acids (AA), and middle molecules (MM) have been implicated in the pathogenesis of experimentally induced acute hepatic coma in the pig. Hemodialysis (HD) using either a low- (Cuprophan = CU) or a high-permeability (polyacrylonitrile = AN 69) membrane has demonstrated the role of MM. Selective hemodialysis (SHD) of AA or NH3 and MM was performed by adding either NH3 (group I) or AA (group II) to the dialysate during AN 69 HD; for MM, SHD only was performed by adding NH3 and AA to the dialysate (group III). In group I the brain levels of tyrosine were similar to those in undialyzed animals with decreased striatal dopamine and decreased norepinephrine in the midbrain only. Brain tryptophan was higher than normal, but brain levels of 5-hydroxytryptamine and 5-hydroxyindoleacetic acid (5-HT, 5-HIAA) were within normal limits. In group II, despite an efficient NH3 clearance, brain NH3 levels were as high as in group I and did not correlate with plasma levels. Brain tyrosine (despite tyrosine overload of the dialysate) was lower than in group I; striatal dopamine decreased (but to a lesser extent than in group I), and norepinephrine was normal. Brain tryptophan was higher than normal, with an increase in brain 5-HT and 5-HIAA. In group III, results were similar to group I, except for a limited increase of 5-HT in the pons. Brain octopamine levels increased only in undialyzed and CU-HD animals, demonstrating a specific relation with MM. These experiments demonstrate the interrelationship between NH3 and neutral AA with regard to passage through the blood-brain barrier and to intracerebral metabolism.  相似文献

In this study, the developmental capacity and cytogenetic composition of different oocyte activation protocols was evaluated following intracytoplasmic sperm injection (ICSI) of in vitro matured bovine oocytes. Motile spermatozoa selected by Percoll density gradient were treated with 5 mM dithiothreitol (DTT) and analysed for ultrastructural changes of the head using transmission electron microscopy (TEM). The alterations in sperm morphology after DTT treatment for different times (15, 30 and 60 min) were 10%, 45-55% and 70-85%, respectively. Further, a partial decondensation of sperm heads was observed after DTT treatment for 30 min. Oocytes were injected with sperm treated with DTT for 30 min. In group 1, sperm injection was performed without any activation stimulus to the oocytes. In group 2, sham injection without sperm was performed without activating the oocytes. Oocytes injected with sperm exposed to 5 microM ionomycin for 5 min (group 3), 5 microM ionomycin + 1.9 mM dimethylaminopurine (DMAP) for 3 h (group 4) and 5 microM ionomycin + 3 h culture in M199 + 1.9 mM DMAP (group 5) were also evaluated for cleavage, development and chromosomal abnormality. Cleavage and development rates in groups 1, 2 and 3 were significantly (p < 0.05) lower than those in groups 4 and 5. The incidence of chromosomal abnormality in the embryos treated directly with DMAP after ionomycin (group 4) was higher than in group 5. We conclude that immediate DMAP treatment after ionomycin exposure of oocytes results in arrest of release of the second polar body, and thus leads to changes in chromosomal pattern. Therefore, the time interval between ionomycin and DMAP plays a crucial role in bovine ICSI.  相似文献

Interactions of APE1 (human apurinic/apyrimidinic endonuclease 1) and DNA polymerase beta with various DNA structures imitating intermediates of DNA repair and replication were investigated by gel retardation and photoaffinity labeling. Photoaffinity labeling of APE1 and DNA polymerase beta was accomplished by DNA containing photoreactive group at the 3 -end in mouse embryonic fibroblast (MEF) cell extract or for purified proteins. On the whole, modification efficiency was the same for MEF-extract proteins and for purified APE1 and DNA polymerase beta depending on the nature of the 5 -group of a nick/gap in the DNA substrate. Some of DNA duplexes used in this work can be considered as short-patch (DNA with the 5 -phosphate group in the nick/gap) or long-patch (DNA containing 5 -sugar phosphate or 5 -flap) base excision repair (BER) intermediates. Other DNA duplexes (3 -recessed DNA and DNA with the 5 -hydroxyl group in the nick/gap) have no relation to intermediates forming in the course of BER. As shown by both methods, APE1 binds with the highest efficiency to DNA substrate containing 5 -sugar phosphate group in the nick/gap, whereas DNA polymerase beta binds to DNA duplex with a mononucleotide gap flanked by the 5 -p group. When APE1 and DNA polymerase beta are both present, a ternary complex APE1-DNA polymerase beta-DNA is formed with the highest efficiency with DNA product of APE1 endonuclease activity and with DNA containing 5 -flap or mononucleotide-gapped DNA with 5 -p group. It was found that APE1 stimulates DNA synthesis catalyzed by DNA polymerase beta, and a human X-ray repair cross-complementing group 1 protein (XRCC1) stimulates APE1 3 -5 exonuclease activity on 3 -recessed DNA duplex.  相似文献

摘要 目的:研究异氟烷预处理对化疗性大鼠异食癖恶心呕吐模型神经功能及呕吐相关神经递质的影响。方法:选用SD大鼠作为研究对象,腹腔注射顺铂以建立化疗性大鼠异食癖恶心呕吐模型(Model组),腹腔注射等量生理研究作为对照组(Control),在腹腔注射顺铂前1 h和12 h吸入异氟烷预处理作为异氟烷治疗组(Isoflurane)。记录腹腔注射顺铂0~12 h和12~24 h内各组大鼠摄入高岭土量;在腹腔注射顺铂0 h、12 h和24 h后,通过神经功能缺损评分法对各组大鼠神经功能进行评分;并在腹腔注射顺铂24 h后处死大鼠,收集大鼠回肠和延髓组织以检测5-羟色胺(5-Hydroxytryptamine,5-HT)、5-羟基-吲哚乙酸(5-hydroxy-indole acetic acid,5-HIAA)、色氨酸羟化酶(Tryptophan hydroxylase,TPH)以及单胺氧化酶(Monoamine oxidase,MAOA)含量。结果:与Control组相比,Model组和Isoflurane组大鼠在腹腔注射顺铂0~12 h,12~24 h以及0~24 h内摄入的高岭土量均显著升高(P<0.05),并且Isoflurane组大鼠均显著Model组。三组大鼠在腹腔注射顺铂0 h、12 h和24 h后,神经功能评分均无显著差异(P>0.05)。与Control组大鼠相比,Model组和Isoflurane组大鼠在腹腔注射顺铂24 h后回肠/延髓组织内5-HT和TPH含量均显著升高,并且Isoflurane组大鼠显著低于Model组大鼠(P<0.05);与Control组相比,Model组大鼠回肠和延髓组织中5-HIAA/5-HIT比值和MAOA含量均显著降低(P<0.05);与Model组大鼠相比,Isoflurane组大鼠回肠5-HIAA含量、回肠/延髓5-HIAA/5-HT比值和MAOA含量均显著升高(P<0.05)。结论:异氟烷预处理可用于预防腹腔注射顺铂诱导的恶性呕吐,其机制可能与下降THP含量和提高MAOA含量,抑制5-HT合成以及促进5-HT代谢有关。  相似文献

目的:探讨脂肪因子分泌型卷曲相关蛋白(secreted frizzled-related protein 5,SFRP5)对骨形态发生蛋白9(bone morphogenetic protein 9,BMP9)诱导人脐带间充质干细胞(human umbilical cord-derived mesenchymal stem cells,hUC-MSCs)成骨分化的影响。方法:将人脐带间充质干细胞根据不同的处理因素分为4组:对照组、BMP9组、BMP9+SFRP5组和SFRP5组;分别在3天、5天和7天进行碱性磷酸酶(alkaline phosphatase,ALP)活性读数,7天进行ALP染色,21天进行茜素红染色检测钙盐沉积及油红O染色;收集不同组的细胞用于裸鼠皮下注射成骨模型的建立,4周后取出离体骨进行Micro-CT扫描和分析,获取的标本进行HE、Masson染色,Alcian blue染色及油红O染色检测。Western blot检测成骨分化相关蛋白Runx2和OPN的表达。结果:BMP9组的ALP活性读数和染色结果和茜素红染色结果均较对照组增加,而BMP9+SFRP5组则较BMP9组降低;BMP9处理后出现少量脂滴,而BMP9+SFRP5组脂滴明显增加,SFRP5组脂滴最多;裸鼠皮下注射成骨模型的观察结果显示,对照组和SFRP5组没有形成肉眼可见的皮下包块,BMP9组和BMP9+SFRP5组能生成异位骨;4周后观测大体标本以及进行MicroCT检测,发现BMP9+SFRP5组的骨密度值小于BMP9组(P0.05)。HE、Masson染色,Alcian blue染色结果显示,BMP9组的骨分化程度大于BMP9+SFRP5,油红O染色结果示BMP9+SFRP5组有较多的成脂分化;SFRP5能抑制BMP9诱导的Runx2、OPN的蛋白质表达。结论:SFRP5抑制BMP9诱导的人脐带间充质干细胞成骨分化。  相似文献

为探索宫腔注入无水乙醇建立薄型子宫内膜模型的可行性,将25只大鼠分3组:对照组(宫腔注入生理盐水),5分钟组(宫腔注入无水乙醇并贮留5 min),10分钟组(宫腔注入无水乙醇并贮留10 min),经测量子宫内膜变薄者为造模组,5分钟组有14个子宫内膜变薄,10分钟组无一例纳入造模组.免疫组织化学检测角蛋白、波形蛋白、血管内皮生长因子(vascular endothelial growth factor,VEGF)、雌激素受体α(estrogen related recep-tor alpha,ERα)的表达.结果发现5分钟组造模组与对照组单位内膜面积中角蛋白面积、单位间质面积中波形蛋白面积和子宫内膜中VEGF平均光密度差异有统计学意义(P<0.05),造模组与对照组子宫内膜ERα组织学积分无差异(P>0.05).表明宫腔注入无水乙醇并贮留5 min可以成功建立薄型子宫内膜模型,成功率为70%.  相似文献

5-氨基水杨酸纳米粒对TNBS诱导小鼠结肠炎的治疗作用   总被引:1,自引:0,他引:1  
目的:评价超临界分散法制备的5-氨基水杨酸(5-ASA)结肠定位纳米粒对小鼠结肠炎的治疗作用。方法:将24只6周龄BABL/c小鼠随机分为空白对照组,结肠炎模型组,5-ASA治疗组,5-ASA纳米粒治疗组。三硝基苯磺酸(TNBS)造模后的第2-7 d分别给予0.9%氯化钠,5-ASA和5-ASA纳米粒灌胃,于第8 d处死所有小鼠,评估小鼠的疾病活动指数(DAI),组织病理形态以及结肠组织学损伤情况;检测结肠组织髓过氧化物酶(MPO)活性。结果:5-ASA和5-ASA纳米粒均能明显减轻结肠重量,缩小溃疡面积,降低MPO活性。超临界分散法制备的5-ASA纳米粒在DAI,减轻局部的炎症程度(MPO水平)方面,其疗效与5-ASA组(50 mg/Kg)相当。结论:超临界分散法制备的5-ASA纳米粒对TNBS诱导小鼠的结肠炎有明显的治疗作用。  相似文献

An animal (BALB/c mice) model of catalpol associated with bronchial asthma in vivo was established, and the effects of catalpol and its relationship with cytokines were investigated. A total of 30 adult BALB/c mice were randomly divided into a positive control group, a model group, and a catalpol group, with 10 mice in each group. The lung function of mice, the cell count, and the cytokine concentrations in bronchoalveolar lavage fluid (BALF) were detected. The levels of cytokines [interleukin 4 (IL-4), interleukin 5 (IL5), and interferon gamma (IFN-γ)] in BALF were measured with enzyme-linked immunosorbent assay methods. The total number of cells in the BALF of the group treated with catalpol was significantly lower than the model group. After treatment with catalpol, the eosinophils and neutrophils of the mice were remarkably reduced compared with the model group. The malondialdehyde content in the lung tissue homogenate of the mice was also decreased in the catalpol group. The cytokines IL-5 and IL-4 exhibited a similar tendency: the concentrations of IL-4 and IL-5 for the catalpol group were dramatically decreased compared with the model group. However, the IFN-γ concentration for the catalpol group was higher than the model group. The results indicated that IL-5 may involve in the pathologic process of asthma-like IL-4, and an inflammatory reaction may still exist in the airway during the remission stage of asthma. The imbalances of the cytokine network might be an important molecular basis in the asthma pathogenesis. It is suggested that catalpol may be a potential drug for the clinical treatment of asthma.  相似文献

Acute virus necrobiotic virus (AVNV) is one of the main pathogens for large scale mortality of Chinese scallop Chlamys farreri. In this paper, C. farreri were infected by different dilutions of AVNV supernatant (5(0), 5(-1), 5(-2), 5(-3), 5(-4), 5(-5), 5(-6), 5(-7), respectively), and dead individuals were counted every day for 15 days. Samples from groups of 5(-3) and 5(-5) were taken every day till 15 days and the activities of acid phosphatase (ACP), alkaline phosphatase (ALP), superoxide dismutase (SOD), myeloperoxidase (MPO), phenoloxidase (PO), peroxidase (POD) and catalase (CAT) in haemocytes were measured. The results of virus challenge showed that survival rates of scallops in groups of 5(0) and 5(-1) decreased sharply after the first day and died out completely on the 5th and 4th day, respectively. In other groups (5(-2), 5(-3), 5(-4), 5(-5), 5(-6) and 5(-7)), survival rates decreased gradually till 6 or 7 days, then kept steady till 15 days, and they were dose-dependent, increasing from 12% to 80% as the dose decreased from 5(-2) to 5(-7) viral supernatant. In the control group, survival rate was 88%. Enzyme activities for groups of 5(-3) and 5(-5) illustrated that activities of ACP, SOD, MPO, PO in groups of 5(-3) and 5(-5) were significantly higher than the control group in the first 9 or 10 days, and went back to the control group levels gradually after 10 days. Moreover, their activities in group of 5(-3) varied more than that in the group of 5(-5), especially activities of MPO, PO. Differently, the activities of POD and CAT were reduced or induced by virus infection and showed no regular trends in the experiments. The activity of ALP was not detected.  相似文献

We investigated the protective and therapeutic effects of molsidomine (MOL) in a rat model of whole brain radiotherapy (RT). Forty female rats were divided into five groups of eight: group 1, control; group 2, 15 Gy single dose RT (RT); group 3, 4 mg/kg MOL treated for 5 days (MOL); group 4, 4 mg/kg MOL for 5 days, 10 days after RT treatment (RT + MOL); group 5, 4 mg/kg MOL treatment for 5 days before RT treatment and for 5 days after RT treatment (MOL + RT). All rats were sacrificed on day 16. Neurodegenerative changes in the brain and tissue levels of oxidants and antioxidants were evaluated. The oxidative parameters were increased and antioxidant status was decreased in group RT compared to groups MOL + RT and RT + MOL. Histopathological examination showed that treatment with MOL after RT application and treatment with MOL before RT treatment decreased neuronal degeneration. No difference in neuronal appearance was found between groups RT + MOL and MOL + RT. MOL treatment protected the nervous system of rats and may be a treatment option for preventing RT induced neural injury.  相似文献

1. A method of isolation of individual afferent fibers from muscle has yielded a representative sample of the fibers which comprise groups 1 (12 to 20 µ) and II (4 to 12 µ) of the afferent fiber diameter distribution of muscle nerves in cat. 2. Afferent fibers from muscle stretch receptors account for groups I and II of the afferent diameter spectrum of muscle nerves to soleus and medial gastrocnemius. Fibers from tendon organs are largely confined to the diameter range above 12 µ. This fiber group, which has a simple one-peak diameter distribution, is termed group IB. Fibers from muscle spindles show a bimodal diameter distribution and account for the remainder of fibers in the 12 to 20 µ group (termed IA) and substantially all of group II (4 to 12 µ). 3. No significant difference has been found in the receptor characteristics of the large (group IA) and intermediate sized (group II) spindle afferent fibers other than a slightly higher threshold of the latter to steady external stretch.  相似文献

目的:探讨自杀未遂老年抑郁症患者血清5-羟色胺(5-HT)水平、总胆固醇(TC)、C反应蛋白(CRP)及白介素-6(IL-6)检测的临床意义。方法:选取2015年5月至2018年5月我院收治的首次住院的老年抑郁症患者155例,依据入院前2周内是否曾有过自杀行为将其分为自杀未遂组(n=75)和无自杀行为组(n=80)。比较两组患者的临床指标及血清5-HT、TC、CRP及IL-6水平;分析不同病情严重程度自杀未遂老年抑郁症患者血清5-HT、TC、CRP及IL-6水平;采用Pearson相关分析血清5-HT、TC、CRP及IL-6水平与汉密尔顿抑郁量表(HAMD)评分的相关性。结果:自杀未遂组HAMD评分高于无自杀行为组(P<0.05)。自杀未遂组血清5-HT及TC水平均低于无自杀行为组(P<0.05),自杀未遂组血清CRP及IL-6水平均高于无自杀行为组(P<0.05)。与轻度组比较,中度组、重度组患者血清5-HT及TC水平均明显降低,且重度组低于中度组(P<0.05);与轻度组比较,中度组和重度组患者血清CRP及IL-6水平均明显升高,且重度组高于中度组(P<0.05)。5-HT、TC与HAMD评分呈负相关(P<0.05);CRP、IL-6与HAMD评分呈正相关(P<0.05)。结论:自杀未遂老年抑郁症患者血清5-HT、TC水平降低,CRP、IL-6水平升高,检测血清5-HT、TC、CRP及IL-6有助于评价其病情严重程度及出现自杀行为的风险。  相似文献

目的研究不同浓度抗坏血酸(ascorbic acid)溶液对幽门螺杆菌(Helicobacter pylori,H.pylori)相关萎缩性胃炎小鼠模型的作用。方法将C57BL/6小鼠随机分为5组,每组10只。组1为空白对照组,余4组小鼠用幽门螺杆菌悉尼菌株(Helicobacter pylori-Sydney strain 1,H.pylori-SS1)感染2周,快速尿素酶法和免疫组织化学法检测H.pylori-SS1的定植;组3、组4和组5小鼠分别给予抗坏血酸低倍剂量(0.1 g/L)、中倍剂量(0.5 g/L)和高倍剂量(1.0 g/L)灌胃22周。灌胃结束后,处死所有小鼠取胃组织和血清,H&E染色观察组织学变化,ELISA检测血清胃泌素(gastrin)和白介素8(IL-8)水平。结果与H.pylori组相比,抗坏血酸干预组胃组织萎缩程度减轻、血清胃泌素水平升高、炎症因子水平降低(组2 vs组3:F=21.730,P=0.029;组2 vs组3:F=42.781,P=0.015;组2 vs组3:F=35.720,P=0.021),中倍剂量组较低倍剂量组有更显著的作用(组3 vs组4:F=21.730,P=0.041;组3 vs组4:F=42.781,P=0.034;组3 vs组4:F=35.720,P=0.043),但高倍剂量组与中倍剂量组差异无统计学意义(组4 vs组5:F=21.730,P=0.125;组4 vs组5:F=42.781,P=0.391;组4 vs组5:F=35.720,P=0.450)。结论抗坏血酸干预可减轻小鼠H.pylori相关萎缩性胃炎症状,高剂量抗坏血酸与中低剂量比无显著差异。  相似文献
