Changes in soluble sugar content in individual parts (embryo, endosperm and pericarp) of maize caryopses exposed to the influence of 24°, 10° and 6° were studied. At the same time the eluates from sand in which the seeds were planted and exposed for various periods were analysed by paper chromatography. It was found that the lower was the temperature of exposure the greater were the amounts of sucrose, glucose, raffinose and other sugars diffused from the caryopses into the medium before the start of germination. The exosmose course and the changes in sugar content in kernel tissues proved that the hydrolysis of storage polysaccharides is not inhibited by low temperatures but that under such conditions the use of products of hydrolysis for the growth of the embryo is inhibited. It is concluded that not only surface parts of endosperm but also the embryo participates in exosmose. The relatively high sugar exosmose found from seeds germinated at low temperatures explains the well-known interaction of pathogenic and parasitic microorganism in cold resistance during emergence.  相似文献

Floral gradient in flowering tobacco in relation to free amino acids   总被引:1,自引:0,他引:1  
By employing TCLs (thin cell layers) culture, the floral gradient in flowering tobacco of different developmental stages was confirmed. The TCLs from early flowering tobacco regenerated more floral buds than those from the tobacco plants in full blooming or fruiting stages. Analysis of free amino acid levels revealed the acropetal gradient of Pro in flowering tobacco stem. L-Pro. L-Trp. D,L-Met and L-Arg were respectively added into the culture medium for testing their influence on floral bud formation from tobacco pedicel segments. Only L-Trp evidently enhanced the floral bud neoformation.  相似文献

A comparative study of free amino acids and protein fractions of normal with a double mutant (su1 o2) was made, during endosperm development in segregating ears of a maize synthetic. Zein content showed striking differences in the two genotypes, being 7.7 and 6 times greater in the normal endosperm at 24 and 47 days after pollination respectively. This observed decrease in zein synthesis, coded by sugary-1/opaque-2 genes, causes an accumulation of alanine, glutamic and aspartic acids, glutamine and asparagine in the high lysine endosperm mutant.  相似文献

The changes in the concentration of free radicals and bound amino acids in developing maize inflorescences and fruits were ascertained. The concentration rises markedly when tissues disintegrate and desiccate. 赤眼蜂的工厂化繁育常使用米蛾卵作为中间寄主卵,为了满足赤眼蜂生产的需要,需对米蛾卵进行冷藏,但冷藏米蛾卵影响赤眼蜂的生长发育。利用核磁共振技术(NMR)测定了新鲜米蛾卵(U)、新鲜杀胚米蛾卵(CK)、杀胚米蛾卵在4℃条件下冷藏15 d(N15)、30 d(N30)、45 d(N45)和60 d(N60)后,米蛾卵卵液游离氨基酸种类和含量的变化。结果共鉴定到24种游离氨基酸及其衍生物,包括昆虫发育必需的10种氨基酸。U和CK之间的游离氨基酸组分没有显著差异,N15、N30和N45游离氨基酸总量随冷藏时间的延长而显著升高,N60中的氨基酸总量与N45比较没有显著增加。对24种游离氨基酸及其衍生物的含量进行主成分分析,结果表明丙氨酸、谷氨酰胺、鸟氨酸、天冬氨酸、组氨酸、赖氨酸、丝氨酸、焦谷氨酸等8种氨基酸的含量随冷藏时间的增加而有着显著的变化,其中,丙氨酸变化幅度最为明显,随着冷藏时间的增加,含量从0.1624 mmol/L增加到8.6192 mmol/L;组氨酸在N30、N45和N60处理之间显著下降,从0.7553 mmol/L降低到0.2495 mmol/L。因此,冷藏会导致米蛾卵内游离氨基酸含量发生一定变化,这可能是冷藏米蛾卵影响赤眼蜂生长发育的重要原因之一。  相似文献

When 5-mm maize root tips were excised and placed in an inorganic salts solution for 6 hours, there was a loss of alcohol-insoluble nitrogen. The levels of threonine, proline, valine, isoleucine, leucine, tyrosine, phenylalanine, and lysine in the alcohol soluble fraction were severely reduced, whereas those of glutamate, aspartate, ornithine, and alanine were scarcely affected. There was a 4-fold increase in the level of γ-aminobutyrate. Those amino acids whose synthesis appeared to be deficient in excised root tips also showed poor incorporation of acetate carbon. In addition, the results show that asparagine and the amino acids of the neutral and basic fraction were preferentially transported to the root tip region. The results therefore suggest that the synthesis of certain amino acids in the root tip region is restricted, and that this requirement for amino acids in the growing region could regulate the flow of amino acids to the root tip.  相似文献

In the years 1963–1965 the content of proline, asparagine, glutamic and γ-aminobutyric acids in young apple (Malus cultivars) shoots was studied. Two groups of apple varieties differing in frost resistance were included into experiments: the Yellow Transparent and Oberländer Himbeerapfel varieties as frost-resistant ones and-the Ontario and Reinette du Canada varieties as sensitive to frost. In the course of experiment γ-aminobutyric acid showed a relatively balanced level but glutamic acid, asparagine and especially proline were found in greater amount in the frost-resistant varieties. According to the results of this experiment the amount of proline—along with many other factors—can be considered as one of the indicators of the length of winter dormancy and also of the frost resistance of apple-trees at the beginning of the winter period.  相似文献

Summary A technique was developed for extracting and analyzing the free amino acid fraction of soil. Ethanol was used as an extracting agent. Ethanolextraction curves showed 20 per cent ethanol was the optimum percentage for extraction. Extraction-time curves indicated 18 to 20 hours of extraction with 20 per cent ethanol produced satisfactory results.The free amino acid fraction of soil was characterized and the limitations of the technique were determined. The naturally occurring amino acids extracted with 20 per cent ethanol were limited to acidic and neutral amino acids; basic amino acids were not extracted in sufficient quantities to permit detection. Based on the percent recovery of amino acids incorporated into soil and extracted with 20 per cent ethanol 90 to 95 per cent of the acidic, 80 to 85 per cent of the neutral and 1 to 5 per cent of the basic amino acids used were recovered with the technique.  相似文献

Conditions for extraction of free amino acids from tomato leaves were examined. Two methods of sample preservation were also tested. Best results were obtained when samples were preserved by lyophilization and extracted by Soxhlet for 6 h at 40°C at a ratio of 1 g plant material/80 mL 80% (v/v) ethanol.  相似文献

A new form of fructokinase has been identified from developing maize (Zea mays L.) kernels that utilizes CTP, UTP, and GTP from four to eight times more effectively than ATP at nonlimiting concentrations. Ten millimolar dithiothreitol was necessary to stabilize activity. A second form of fructokinase was nonspecific for nucleoside triphosphate whereas a third form was fairly specific for ATP.  相似文献

Heterosis is the superior performance of hybrids over their inbred parents. Despite its importance, little is known about the genetic and molecular basis of this phenomenon. Heterosis has been extensively exploited in plant breeding, particularly in maize (Zea mays, L.), and is well documented in the B73 and Mo17 maize inbred lines and their F1 hybrids. In this study, we determined the dry matter, the levels of starch and protein components and a total of 24 low-molecular weight metabolites including sugars, sugar-phosphates, and free amino acids, in developing maize kernels between 8 and 30 days post-pollination (DPP) of the hybrid B73 × Mo17 and its parental lines. The tissue specificity of amino acid and protein content was investigated between 16 and 30 DPP. Key observations include: (1) most of the significant differences in the investigated tissue types occurred between Mo17 and the other two genotypes; (2) heterosis of dry matter and metabolite content was detectable from the early phase of kernel development onwards; (3) the majority of metabolites exhibited an additive pattern. Nearly 10% of the metabolites exhibited nonadditive effects such as overdominance, underdominance, and high-parent and low-parent dominance; (4) The metabolite composition was remarkably dependent on kernel age, and this large developmental effect could possibly mask genotypic differences; (5) the metabolite profiles and the heterotic patterns are specific for endosperm and embryo. Our findings illustrate the power of metabolomics to characterize heterotic maize lines and suggest that the metabolite composition is a potential marker in the context of heterosis research.  相似文献

A method was developed to analyze quantitatively free amino acids and amino acids attached to transfer RNA (tRNA) in tissue samples by gas chromatography. Free amino acids were purified by ion-exchange chromatography after deproteinization. Total cellular aminoacyl-tRNA was extracted from rabbit reticulocytes and liver by a modified phenol extraction method under conditions which were designed to prevent deacylation of the attached amino acids. After deacylation and separation from tRNA by pressure ultrafiltration, eighteen amino acids were determined by gas chromatography as their N-heptafluorobutyryl isobutyl derivatives.  相似文献

Nové pfirustky plodonos? na stromech s plody neměly nikdy pr?kazně vyssí obsah rozpustného dusiku nez nové prir?stky plodonos? a nové prir?stky plodového obrostu na stromech bez plodu. U vsech sledovanych odrud jabloně byl zjistěn stejny kvalitativni obrazee volnych aminokyselin a amid? a v odpovídajících osních cástech take obdobny pomér volnych aminokyselin a amid?. Největsi mnozství volnych amino-kyselin a amidu - obzvláste asparaginu a argininu-bylo zjistěno v plodonosích. V plodonosíeh byl arginin akumulován ve velké mífe mnohem dííve nez v novych prir?stcich plodonos? a nez v novych pfir?stcich plodového obrostu. K akumulaci argininu a asparaginu v plodonosích docházelo i pri nedostateeném zásobení dusíkem, v novych pfir?stcich plodonos? byl vsak jejich obsah pii nedostatku dusiku velmi nizky. V plodonosích s plody byl také dosti zvyseny obsah glutaminu a v nekterych pfípadech i threoninu. Glutamová kyselina, glutamin a někdy také threonin byly zastoupeny о песо vice v mladsích osách nez v osách starsích, jiz zdievnatělych. Z vysledkú uvedenych v teto práci lze uzavrít, ze mezi obsahem a poměrem rozpustného a nerozpustného dusiku v plodovém obrostu jabloní na jedné strane a zakládáním květnieh pupenu na druhé strane není zádny zrejmy vztah. Mezi obsahem volnych aminokyselin a amid? v sledovanych osních cástech jabloně a zakládáním květnieh pupenu nebyl rovněz zjistěn zádny zrejmy vztah.  相似文献
