山玉兰花粉离体萌发和花粉管生长特性的研究   总被引:1,自引:0,他引:1  
山玉兰(Magnolia delavayi)是木兰科木兰属的常绿乔木或大型灌木,是重要的园林造景、庭院绿化素材,也是重要的育种资源。山玉兰花粉的研究对其杂交育种的成败具有重要影响,但目前尚未见其花粉活力的相关报道。该研究以新鲜的山玉兰花粉为对象,采用悬滴培养法分析了温度、光照以及培养液的蔗糖和硼酸浓度对山玉兰花粉萌发的影响。结果表明:(1)山玉兰花粉萌发时,最适宜的温度为27℃。(2)光暗条件下,山玉兰花粉以浓度为5%的蔗糖培养效果最佳,其萌发率在16%以上;而硼酸浓度则以0.001%的培养效果最佳。(3)蔗糖与硼酸共同作用可有效促进花粉萌发和花粉管生长。其中,在光照条件下,以5.0%蔗糖+0.001%硼酸为最适宜的培养液,花粉萌发率达41.27%,花粉管长达281.49μm;而在黑暗条件下,则以5.0%蔗糖+0.01%硼酸为最适宜的培养液,花粉萌发率达45.71%,花粉管长达254.00μm。该研究结果为进一步开展人工辅助授粉、发掘山玉兰的种质资源工作奠定了基础。  相似文献

Pollen tube growth in vitro requires calcium for most species but the in vivo source or reservoir of this calcium is not known. Using methods to localize calcium in situ, we confirm that low levels of calcium are detected in the transmitting tract extracellular matrix (ECM) in unpollinated lily styles. Pollination in lily induces an increase in the detectable levels of calcium in the transmitting tract ECM binding to the stylar cell and pollen tube walls. Studies on the initial germination of pollen of Pinus mugo showed no significant influence of ions on O2 uptake and uptake of 32P-labelled phosphate. At the onset of tube growth O2 uptake decreased in the absence of calcium. In inorganic media tube growth and 32P uptake were reduced in the absence of calcium or boric acid. In the absence of calcium a requirement for magnesium was observed. When the medium was deprived of polyvalent ions with EDTA, growth and 32P uptake ceased. The presence of calcium in the medium was found to be essential for the maintenance of the structural and functional integrity of the cell membranne. — The ion requirement was more pronounced when tube growth was stimulated with sucrose. Calcium, magnesium, boric acid, and nitrate (as nitrogen source) were essential constitutents of the medium. The stimulation due to calcium required either magnesium or boric acid. — A density effect was observed which can be related to diffusible substances from the pollen into the medium. This was not observed when calcium and magnesium were present in the medium. The phenomenon is explained as an enrichment of the medium with diffusible substances from non-germinated dead pollen. — Germination and the tube growth were found to be greatly dependent on a short period of equilibration of pollen at room temperature before sowing.  相似文献

Well before pollen tube penetration, ethylene has begun to disseminate from pollinated styles of Petunia hybrida flowers. Previous stigmatic application of aminoethoxyvinylglycine (AVG) completely prevented this ethylene synthesis, indicating that the endogenous 1-aminocyclopropane-1-carboxylic acid (ACC) in pollen is not readily converted on the stigma. Compared to other flower parts, the capacity of the ethylene forming enzyme was largest in the stigma. When applied to the stigma, ACC caused ethylene synthesis, but did not accelerate wilting, unless high concentrations (20 nanomols) were used. Upon pollination or stigma wounding, the early ethylene evolved exclusively from the gynoecium, much later followed by the synthesis of corolla ethylene. Employing wideneck Erlenmeyer flasks, the competitive inhibitor of ethylene action, norbornadiene, was applied to entire flowers in situ, with delaying effects on wound-induced wilting. In contrast, norbornadiene treatment of styles alone, using capillaries, could not postpone wilting. Pollination with foreign pollen species did not lead to accelerated corolla wilting, notwithstanding considerable synthesis of ethylene during the first 5 hours. In situ treatment of the stigma with AVG considerably delayed wound- and pollination-induced wilting. Removal of the entire AVG-treated style 6 hours after stigma wounding still allowed for the postponement of the accelerated wilting, even at very low concentrations of AVG. It is concluded that early stylar ethylene does not play a role in the acceleration of wilting but that, much later, corolla ethylene does, induced by a mobile wilting factor from the stigma, which is ACC.  相似文献

Adhesion of lily pollen tubes on an artificial matrix   总被引:2,自引:0,他引:2  
 We proposed that pollination in lily is a case of cell adhesion and cell movement, but experimental evidence for the adhesion event is lacking. In this study, we developed an artificial extracellular matrix that mimics the in vivo lily stylar transmitting tract. This artificial matrix was created by applying the transmitting tract exudate extracted from lily styles onto a nitrocellulose membrane. When in vitro-grown pollen tubes were applied to the matrix, they adhered by their tips to the area of the stylar exudate which is rich in arabinogalactan proteins. Once they adhered, they grew on the in vitro artificial matrix at rates faster than normal. This is the first experimental evidence demonstrating the adhesion of in vitro-grown pollen tubes, an event that has been described as common in vivo. The adhesion event is stylar exudate specific, concentration dependent, and is affected by the developmental age of the pollen tube. Upon germination on the stigma, pollen tubes elongate in the stylar transmitting tract, aided by female factors, with speed and directionality not mimicked in in vitro pollen tube growth cultures. We have shown that a stylar transmitting tissue arabinogalactan protein (AGP) from Nicotiana tabacum (tobacco), TTS protein, stimulates pollen tube growth in vivo and in vitro and attracts pollen tubes grown in a semi-in vivo culture system. It has been reported that the self-incompatible Nicotiana alata produced a stylar glycoprotein, GaRSGP, which had a backbone polypeptide that shared 97% identity with those of TTS proteins but some of its properties were different from those described for TTS proteins. We report here the characterization of a family of stylar transmitting tissue glycoproteins from N. alata that is virtually identical to tobacco TTS proteins and which we refer to as NaTTS proteins. Like their tobacco counterparts, NaTTS proteins are recognized by the traditional AGP-diagnostic reagent beta-glucosyl Yariv reagent, and they are also recognized by JIM13, a monoclonal antibody against AGP. NaTTS proteins also stimulate pollen tube elongation in vitro and attract pollen tubes in a semi-in vivo pollen tube culture system. Biochemical and immunological characterization of NaTTS proteins revealed that they have extraordinary variability in the extent of sugar modifications of their polypeptide backbones. The extent of sugar modifications on NaTTS proteins significantly affects their biochemical properties, influences how they interact with the transmitting tissue extracellular matrix, and affects their solubility from this matrix. Our results suggest that the strategy used to purify GaRSGP only recovered a less glycosylated, more tightly extracellular matrix-bound sub-population of the entire spectrum of N. alata TTS proteins.  相似文献

Long-, mid-, and short-styled clonal accessions of oca (Oxalis tuberosa) were intercrossed in a complete diallelic design. Pollen tube growth in styles was monitored in all diallelic crosses. Pollen fertility was estimated by two tests: staining of pollen grains with aceto-carmine and detection of β-galactosidase activity by the substrate X-Gal. The two methods of pollen fertility estimation were equally useful to detect fertility levels. Pollen originating from short stamens had the highest fertility (85%) and pollen from long stamens had the lowest fertility (70%). Pollen fertility was high throughout, but its degree varied with the stylar morph on which the pollen was formed. Long-styled accessions had the highest rates of fertile pollen. Differences in pollen fertility at different anther levels in the same style morph were also apparent. Pollen grain diameter of the six morph-anther level combinations was inversely correlated with pollen fertility. Pollen grains from long stamens were the largest and pollen grains from short stamens were the smallest. Neither pollen fertility nor pollen grain size had an influence on pollen tube growth in the style or on fruit and seed set. Pollen tubes growing within the styles were inhibited at a different level for each of the 18 cross combinations in the diallel. Although legitimate crosses had greatest pollen tube growth, some of the illegitimate inter- and intramorph crosses had equally high scores. Of all illegitimate crosses, mid-styled seed parents had the lowest level of stylar incompatibility. Fruit and seed set were highly correlated with the extent of pollen tube growth in the style. The number of pollen tubes entering ovules in a flower was in good agreement with the number of seeds produced per fruit. Therefore, it is concluded that stylar incompatibility is the major determinant of limited seed formation in oca even in the most successful legitimate cross combinations. Summary A factorial cross design was used to evaluate the influence of differences among pollen donors and recipients on variation in stylar attrition of pollen tubes in self-fertile plants of Petunia hybrida. Pollinations to flower buds were used to reduce the inhibitory ability of the style and these crosses were compared with flower pollinations to assess the degree of stylar influence on pollen fertilization ability. There was less pollen tube attrition after bud pollinations than after flower pollinations, indicating that styles of buds were less able to inhibit pollen tubes. The variance component for plants acting as pollen donors tended to be greater after flower pollinations than after bud pollinations. The lower variance in male success after bud pollinations indicates that differences among pollen donors after flower pollinations were due to stylar inhibition of pollen rather than differences in pollen vigor. Since the level of variation in pollen growth after pollination to flowers was greater among clones than among ramets within clones, the differences in pollen fertilization ability are probably genetically based.  相似文献

Summary The excised pistil technique was used to study effects of pollen treatments on self-incompatibility in red clover. Pollen was treated directly with infrared or ultraviolet radiation, and indirectly with chemicals that were applied to the stigmas before pollination. The chemicals (boric acid, calcium nitrate, colchicine, cyclohexamide, gibberellic acid, glycine, and indole butyric acid) were each used at five concentrations. High dosage rates of most treatments inhibited pollen growth in styles that were genetically compatible, but none of the treatments had a specific effect on the self-incompatibility mechanism.The effects of growth of compatible or incompatible pollen tubes in a style on subsequent growth of both types of pollen tubes in the same style were studied in an experiment with 6 hr between pollinations. The passage of some compatible pollen tubes through the style did not influence either self- or cross-compatibility in the style after the second pollination. The passage of some self-incompatible tubes into the styles caused a slightly higher number of incompatible tubes to penetrate the style after the second pollination. Various factors (pollen diameter, in vitro germination and tube length, in vivo growth rate in selfed and nonselfed styles) which could possibly contribute to the competitive ability of pollen were investigated on 30 Zea mays L. inbred lines. The only factor with which pollen diameter was positively correlated was in vitro pollen-tube growth. Traits related to the early stages of growth (in vitro germination, in vitro tube length, early in vivo pollen growth rate) were all positively correlated with each other, and these early characteristics were negatively correlated with late in vivo tube growth rate, which is largely influenced by the stylar genotype.  相似文献

In the absence of cations, the release of proteins from pollen tubes ofNicotiana tabacum in culture is greatly dependent on boron concentration and inversely related to growth stimulation. The minimum of proteins in the medium occurs at 100 mg 1? of boric acid, which is the optimum concentration for growth. The shift of boron level to this optimum further increases the proportion of proteins bound to the insoluble pollen tube fraction; on the other hand the amount of soluble proteins is not affected inside pollen tubes, but greatly decreased in the medium. The loss of proteins into the medium is considerably reduced by oalcium and also at the optimal boron concentration and in the presence of K+ and Mg2+ ions. The rate of tube elongation, however, is slightly decreased and the duration of pollen growth activity is not extended. The release of proteins is not affected by potassium and is slightly reduced by magnesium. The possible mechanism of calcium and boron action on protein release is discussed in relation to the exocytotic function of secretory vesicles and it is suggested that boron supports the incorporation of proteins transported to the growing tip into the pollen tube wall structures.  相似文献

The influence of ethylene and CO2 on pollen germination and tube growth was investigated employing ventilated culture systems. Ethylene had no effect on pollen germinability or tube growth. Germinating pollen did not produce a detectable amount of ethylene (less than 0.1 nl/g·hr). Supplementing the cultures with CO2 caused a marked increase in germination and tube growth. The half-maximal response for germination was less than 0.5%. CO2 levels ranging from 1.08 to 2.22% were found in the internal cavity of lily styles. CO2 derived from stylar metabolism may, therefore, modulate pollen tube growth thus integrating the events leading to fertilization.  相似文献

Most species of Dalechampia have expanded stigmatic surfaces that extend from the stylar tip part way down the sides of the elongated styles. Pollen grains landing at the stylar tip grow tubes directly down the style to the ovary. Pollen grains landing on the lateral stigmatic surfaces, however, exhibit indirect pollen-tube growth: the tubes grow first to the stylar tip, bend 180°, and then grow to the ovary. Indirect pollen-tube growth appears to be reproductively important: more than half the arriving pollen lands on the lateral stigmatic surfaces, and virtually all of these grains produce tubes that grow indirectly to the ovary; pollen on the lateral surface fertilizes seeds about as effectively as pollen at the tip (with direct tube growth). Indirect pollen-tube growth may be the result of a correlated response to selection for expanded stigmatic surfaces in Dalechampia, or it may be an adaptation to increase the intensity of pollen competition.  相似文献

以‘丰水’和‘幸水’梨花柱及花粉为试材,用激光共聚焦显微技术,研究了离体条件下G蛋白活性调节剂和花柱S-RNA酶对花粉管生长及其游离Ca~(2 )浓度的影响。结果表明:G蛋白激活剂CTX可促进花粉管生长,且可解除花柱S-RNA酶对自身花粉管生长的抑制作用;G蛋白抑制荆PTX和花柱S-RNA酶共同处理使异体的花粉管生长受到抑制。CTX处理使花粉管尖端区的[Ca~(2 )]_i明显升高,花柱S-RNA酶处理引起自身花粉管尖端区的[Ca~(2 )]_i梯度消失;CTX和花柱S-RNA酶共同处理则使自身花粉管内的[Ca~(2 )J_i表现出两者单独处理时的综合特征;而花柱S-RNA酶和PTX共同处理后,异体的花粉管内[Ca~(2 )]_i表现出先升高后下降的趋势。  相似文献

Effect of two different seasons and hormones on pre- and postfertilization barriers in crossability were studied inVigna mungo, V. unguiculata and their crosses. Significant differences between the two seasons for pollen fall, pollen germination, tube growth and pod set were observed. In kharif season with an average temperature of 26 to 30 °C and 75 to 85 % humidity all the characters showed higher values than in the spring season with a higher temperature of 32 to 36 °C and low humidity of 40 to 50 %. Application of gibberellic acid (GA) alone and a combination of gibberellic acid, naphthaleneacetic acid (NAA) and kinetin (KIN) significantly increased the pollen fall by 7–13 %, pollen germination by 10–12 % and pollen tube growth by 27–30 %. More pollen tube abnormalities in interspecific crosses only indicates partly incompatible reaction of pollen tube in the stylar tisue. Hormonal treatments, especially GA, significantly increased the pod set and pod harvest by 20 % and 34 %, respectively. Higher pod harvest was seen in crosses withV. unguiculata as female parent. GA treatment enhanced pollen germination and pollen tube growth, and by partially overcoming embryo abortion for 10 to 12 d, immature embryos were successfully rescued forin vitro production of hybrid plantlets.  相似文献
