This study was designed to study xanthine oxidase (XO) and xanthine dehydrogenase (XD) activity in the lung of rats exposed to prolonged restraining immobilization stress. Immobilization caused more than twofold increase of xanthine oxidase activity in the rat lung. The activity of xanthine oxidase decreased in lung homogenates incubated at -20 degrees C for 24 h. The same incubation of homogenates from control rats caused a non-significant increase of the activity. No measurable NAD(+)-dependent xanthine dehydrogenase activity could be established in the lungs of both control rats and rats subjected to immobilization. All rats revealed methylene blue-dependent xanthine dehydrogenase activity which was more than two-times higher in the immobilized animals. Incubation at -20 degrees C for 24 h increased the methylene blue-dependent xanthine dehydrogenase activity in homogenates from control rats and decreased the enzyme activity in homogenates from immobilized rats. A working hypothesis was proposed for the sequence of events explaining the results obtained: XO-catalyzed generation of activated oxygen species may take place in the initiation of lipid peroxidation in the lung of rats immobilized for prolonged periods of time.  相似文献

Significant hypoalgesia (tail-flick reflex at 60 degrees C) was observed in rats during the whole period of 24-hour immobilization in cramped cages, as compared to food-and-water deprived and control animals. This hypoalgesia was not antagonized by naloxone (1 mg/kg), however, the animals periodically receiving the drug during immobilization revealed aggressiveness and significant hypoalgesia after immobilization was discontinued (30-120 min observation).  相似文献

Experiments with rats have shown that thermoregulation under normal conditions and in response to stressful factors (immobilization, emotionally significant sound) is different in animals of different age. Pregnant primiparous rats were exposed to low immobilization stress or ACTH injections (one unit) throughout pregnancy. We measured the following parameters in the female offsprings: puberty (vaginal opening and first oestrous), oestrous cycle and sexual behaviour. Compared to the controls, female sexual receptivity measured by means of lordosis and the lordosis quotient (LQ) increased in both the experimental groups, but the puberty parameters of the offspring did not alter.  相似文献

The role of gluconeogenesis on the increase in plasma glucose and liver glycogen of rats exposed to hyper-G (radial acceleration) stress was determined. Overnight-fasted, male Sprague-Dawley rats (250-300 g) were injected i.p. with uniformly labeled 1 4C lactate, alanine, or glycerol (5 microCi/rat) and immediately exposed to 3.1G for 0.25, 0.50, and 1.0 hr. 1 4C incorporation of the labeled substrates into plasma glucose and liver glycogen was measured and compared to uncentrifuged control rats injected in a similar manner. Significant increases in 1 4C incorporation of all three labeled substrates into plasma glucose were observed in centrifuged rats at all exposure periods; 1 4C incorporation into liver glycogen was significantly increased only at 0.50 and 1.0 hr. The i.p. administration (5 mg/100-g body wt) of 5-methoxyindole-2-carboxylic acid, a potent gluconeogenesis inhibitor, prior to centrifugation blocked the increase in plasma glucose and liver glycogen during the first hour of centrifugation. The increase in plasma glucose and liver glycogen was also abolished in adreno-demedullated rats exposed to centrifugation for 1.0 hr. Propranolol, a beta-adrenergic blocker, suppressed the increase in plasma glucose of rats exposed to centrifugation for 0.25 hr. From the results of this study, it is concluded that the initial, rapid rise in plasma glucose as well as the increase in liver glycogen of rats exposed to hyper-G stress can be attributed to an increased rate of gluconeogenesis, and that epinephrine plays a dominant role during the early stages of exposure to centrifugation.  相似文献

The in vitro uptake of 131I by the thyroid gland was investigated in rats after immobilization stress with special respect to animals lesioned in the septal area. Lesions in a septal area performed 10 days before decreased the iodide accumulation in the thyroid, while stress by immobilization increased it to the control basal value. Repeated immobilization in control rats did not produce any changes in the iodide uptake in vitro. ACTH injected in vivo stimulated the iodide 131I uptake in vitro by thyroid glands of hypophysectomized rats. It is concluded that immobilization stress in rats with septal lesions increases 131I-iodide uptake in vitro and that the increase was probably influenced by both catecholamines and glucocorticoids.  相似文献

The function of the isolated perfused rat hearts was studied in four groups of experiments. Group 1--included the hearts of intact animals ("absolute control"), group 2--the hearts of rats subjected to 24 hour immobilization in supine position against the background of triple intramuscular injections of placebo (control), group 3 included the hearts of rats, which during 24 hour immobilization stress were thrice injected a synthetic analogue of endogenous opioids Dalargin in a dose of 3 g/kg, and group 4 included the hearts of animals, which during immobilization were administered Dalargin in a dose of 10 g/kg of body mass. Ulcer index as indicator of stress injury of gastric mucosa was also determined. In the control group of experiments (group 2) 24 hour immobilization stress resulted in complete depression of cardiac performance as compared with group 1, and ulcer index approximated 1. In group 3 the indices of cardiac performance even exceeded those in group 1 (intact animals). As compared with group 2, ulcer index in group 3 decreased by 9 times. In application of Dalargin in a dose of 10 g/kg complete preservation of heart function indices and complete prevention of stress injury of gastric mucosa were also observed. Thus, Dalargin possesses cardioprotective and anti-ulcerogenic effect in immobilization stress in rats. Most probably, this phenomenon can be attributed to its ability to inhibit the activity of sympathoadrenal system, which gets enhanced during stress.  相似文献

The aim of the present study was to define the stress-induced pattern of cytosolic glucocorticoid receptor (GR) and Hsp70 protein in the liver of male Wistar rats exposed to different stress models: acute (2 h/day) immobilization or cold (4 degrees C); chronic (21 days) isolation, crowding, swimming or isolation plus swimming and combined (chronic plus acute stress). Changes in plasma levels of corticosterone were studied by radioimmunoassay (RIA). The results obtained by Western immunoblotting showed that both acute stressors led to a significant decrease in cytosolic GR and Hsp70 levels. Compared to acute stress effects, only a weak decrease in the levels of GR and Hsp70 was demonstrated in chronic stress models. Chronically stressed rats, which were subsequently exposed to novel acute stressors (immobilization or cold), showed a lower extent of GR down-regulation when compared to acute stress. The exception was swimming, which partially restores this down-regulation. The observed changes in the levels of these major stress-related cellular proteins in liver cytosol lead to the conclusion that chronic stressors compromise intracellular GR down-regulation in the liver.  相似文献

Chronic stress has been related to multiple diseases. Inflammation is proposed strongly to link stress to stress-related diseases in different organs, such as small intestine, colon, and brain. However, stress cellular effect on the pancreatic tissue, especially the exocrine one, had received relatively little attention. This work aimed to evaluate the cellular effect of chronic immobilization stress on the pancreatic tissue function and structure along with evaluating the sex role in this type of pancreatic injury. Thirty rats were equally divided into 5 groups: control male, control female, stressed male, stressed female, and stressed female with bilateral ovariectomy. Stressed rats were exposed to immobilization for 1 h/day, 6 days/week, for 3 weeks. Rats were then decapitated for further biochemical, histological, histo-morphometric, and immunohistochemical study. The results showed that, in male and female rats, chronic immobilization stress produced hypoinsulinemia and hyperglycemia, with increasing exocrine pancreatic injury markers by increasing oxidative and inflammatory status of the pancreatic tissue, and exhibited a degenerative effect on the pancreatic tissue. However, the stress-induced pancreatic effects were more obvious in male rats and female rats with bilateral ovariectomy than that in female rats. It could be concluded that male animals were more susceptible to stress-induced pancreatic damage than females. The ovarian hormones are responsible, at least partly, for pancreatic tissue protection since the stress-induced pancreatic injury in females was exacerbated by ovariectomy. In this study, inflammatory and oxidative stress differences in both sexes could provide a plausible explanation for sex differences.  相似文献

In case of a life-threatening, stressful event, the body prepares for an emergency. Indeed, the lung is unique in which alveolar cells are constantly exposed to physical and chemical stresses. This study aimed to study the impact of immobilization stress on the blood–air barrier and how it initiate and maintain an inflammatory response, plus determining the resolution of lung inflammation and repair. There was a significant increase in the plasma levels of stress markers “corticosterone and catecholamines” with a decrease in surfactant protein A (a lung-injury marker). Chronic stress produced a significant increase in the pulmonary oxidative and inflammatory markers malondialdehyde, tumor necrosis factor α, and induced nitric oxide synthase when compared with that of acute stress. Both stresses provoked marked pulmonary morphological and ultrastructural changes with a significant increase in caspase-3 immunoexpression. There was increasing evidence of lung’s capacity for repair. This process involved edema resolution, cell proliferation, and tissue remodeling in improving the lung-injury, oxidative, and inflammatory markers.  相似文献

An investigation was made into the effects of cold/restraint stress in rats on renal function, plasma constituents, blood count and endocrine function. Cold/restraint caused an increased volume of urine output, with increased K+ and urea excretion, and decreased Na+ and Cl? excretion. There was a fall in plasma glucose, and a rise in plasma urea. A marked leucopenia was found, and the blood pH was significantly lowered. Cold/restraint was also found to cause marked increases in corticosterone and thyroxine levels, and a fall in the insulin level.  相似文献

The changes in DOPA and catecholamine (adrenaline, noradrenaline, dopamine) levels were investigated in noradrenaline- and dopamine-synthesizing brain nuclei of Wistar rats after prolonged immobilization stress on catecholamine analyzer (BAS, USA) using HPLC technique. Distinct DOPA and catecholamine changes were observed in locus ceruleus + nucleus subceruleus (1. c + n. sc) and substantia nigra at any stage after immobilization (right after immobilization and 15 and 30 days later). The most prominent alterations in noradrenaline content were detected in 1. c + n. sc. 30 days after immobilization NA level in these nuclei was 1.5 times higher, as compared to the control one. It is suggested that the increasing noradrenaline level in 1. c + n. sc. plays a defensive role in survival of rats after immobilization stress.  相似文献

Summary Generally, the CRF-like activity of vasopressin is studied in experiments involving adrenalectomy and corticosteroid replacement. In order to avoid this complex type of stress, male and female (diestrus, estrus) rats were exposed for 5min to immobilization stress and sacrificed 5, 15, and 30 min thereafter. After a survival period of 5 min the vasopressin-synthesizing part of the paraventricular nucleus exhibited an increased activity. Vasopressin-reactive axons in the pericapillary layer of the median eminence and among the solid cell clusters of the pars tuberalis became more conspicuous and increased in number. In this group of experimentally treated animals the prechiasmatic division of the supraoptic nucleus did not show any changes in immunoreactivity. The same holds true for the neurohypophyses in all experimental groups. In animals with increased survival times the supraoptic nucleus exhibited a slightly increased activity, whereas the staining intensity of the paraventricular nucleus decreases gradually. From these results it can be concluded that the paraventricular nucleus is involved in the first phase of the stress response. The problem of vasopressin or a very similar peptide synthesized in this nucleus and exerting a CRF-releasing function is discussed.The author wishes to express her sincere gratitude to Dr. A. The integration of innate andadaptive immune responses is required for efficient control ofCandida albicans. The present work aimed to assess, at thelocal site of the infection, the immunocompetence of macrophages inrats infected intraperitoneally with C. albicans and exposedsimultaneously to stress during 3 days (CaS group). We studied the1) ability to remove and kill C. albicans,2) tumor necrosis factor- (TNF-) release,3) balance of the inducible enzymes NO synthase (iNOS) andarginase, and 4) expression of interleukin (IL)-1 andIL-1 receptor antagonist (ra) mRNA. Compared with only infected animals(Ca group), the number of colony-forming units was significantly higherin CaS rats (P < 0.01), and the macrophagecandidicidal activity was ~2.5-fold lower (P < 0.01). Release of TNF- was diminished in both unstimulated andheat-killed C. albicans restimulated macrophages of the CaSgroup (Ca vs. CaS, P < 0.03 and P < 0.05, respectively). In Ca- and CaS-group rats, the rates for both thearginase activity and the NO synthesis were significantly enhanced.However, the stress exposure downregulated the activity of both enzymes(CaS vs. Ca, P < 0.05). After in vitro restimulation,the IL-1ra/IL-1 ratio was significantly diminished in CaS-group rats(P < 0.05). Our results indicate that a correlationexists between early impairment of macrophage function and stress exposure.

Oxidative stress and protein carbonylation is implicated in aging and various diseases such as neurodegenerative disorders, diabetes, and cancer. Therefore, the accurate identification and quantification of protein carbonylation may lead to the discovery of new biomarkers. We have developed a new method that combines avidin affinity selection of carbonylated proteins with iTRAQ labeling and LC fractionation of intact proteins. This simple LC-based workflow is an effective technique to reduce sample complexity, minimize technical variation, and enable simultaneous quantification of four samples. This method was used to determine protein oxidation in an iron accumulating mutant of Saccharomyces cerevisiae exposed to oxidative stress. Overall, 31 proteins were identified with 99% peptide confidence, and of those, 27 proteins were quantified. Most of the identified proteins were associated with energy metabolism (32.3%), and cellular defense, transport, and folding (38.7%), suggesting a drop in energy production and reducing power of the cells due to the damage of glycolytic enzymes and decrease in activity of enzymes involved in protein protection and regeneration. In addition, the oxidation sites of seven proteins were identified and their estimated position also indicated a potential impact on the enzymatic activities. Predicted 3D structures of peroxiredoxin (TSA1) and thioredoxin II (TRX2) revealed close proximity of all oxidized amino acid residues to the protein active sites.  相似文献

Exposure to an open field is psychologically stressful and leads to an elevation in core temperature (T(c)). Methyl scopolamine (MS), a muscarinic antagonist, and pyridostigmine (PYR), a carbamate that inhibits acetylcholinesterase, do not cross the blood-brain barrier and have little effect on T(c) in resting, nonstressed animals. However, we have found that MS has an antipyretic effect on T(c) that is caused by handling and cage-switch stress. PYR should act pharmacologically to reverse the effects of MS. To this end, we assessed the effects of MS and PYR on stress-induced hyperthermia. Male Sprague-Dawley rats at 90 days of age were housed individually at an ambient temperature of 22 degrees C. T(c) and motor activity were monitored by radiotelemetry in an open-field chamber. Rats were dosed intraperitoneally at 1200 with 1.0 mg/kg MS, 0.1 mg/kg PYR, a combination of MS and PYR, or saline and placed immediately inside the open-field chamber for 60 min. Stress-induced hyperthermia was suppressed immediately by MS and enhanced by PYR. T(c) only increased by 0.3 degrees C in the MS-treated animals. The hyperthermic response in the PYR group was nearly 0.6 degrees C above that of rats dosed with saline. Coadministration of PYR and MS led to a stress-induced hyperthermia response nearly identical to that of rats injected with saline. Overall, open-field stress exacerbated the effects of MS and PYR on body T(c) and provides support for a peripheral cholinergic mechanism that mediates stress-induced hyperthermia.  相似文献
