Aerobic granules for sulphide and ammonium removal were cultivated in a sequencing batch reactor, and the microbial community of the aerobic granules was investigated by denaturing gradient gel electrophoresis. The loading rate increased from 0.15 to 0.9 kg S2? m?3 d?1, and the removal efficiencies of sulphide, chemical oxygen demand, and NH4 +-N were higher than 99, 80, and 98%, respectively. However, sludge settleability became poorer when the loading rate exceeded 0.3 kg S2? m?3 d?1. The denitrifying bacteria in the aerobic granules were Thauera sp., Pseudomonas alcaligenes, and uncultured planctomycetes, indicating that multiple N-removing processes occurred simultaneously in the aerobic granules. These processes could include nitrification and denitrification, aerobic denitrification, and anaerobic ammonia oxidation. Sludge settleability became poorer because of the overgrowth of uncultured Thiothrix sp.  相似文献

Members of the uncultured bacterial genus Candidatus Accumulibacter are capable of intracellular accumulation of inorganic phosphate in activated sludge wastewater treatment plants (WWTPs) performing enhanced biological phosphorus removal, but were also recently shown to inhabit freshwater and estuarine sediments. Additionally, metagenomic sequencing of two bioreactor cultures enriched in Candidatus Accumulibacter, but housed on separate continents, revealed the potential for global dispersal of particular Candidatus Accumulibacter strains, which we hypothesize is facilitated by the ability of Candidatus Accumulibacter to persist in environmental habitats. In the current study, we used sequencing of a phylogenetic marker, the ppk1 gene, to characterize Candidatus Accumulibacter populations in diverse environments, at varying distances from WWTPs. We discovered several new lineages of Candidatus Accumulibacter which had not previously been detected in WWTPs, and also uncovered new diversity and structure within previously detected lineages. Habitat characteristics were found to be a key determinant of Candidatus Accumulibacter lineage distribution while, as predicted, geographic distance played little role in limiting dispersal on a regional scale. However, on a local scale, enrichment of particular Candidatus Accumulibacter lineages in WWTP appeared to impact local environmental populations. These results provide evidence of ecological differences among Candidatus Accumulibacter lineages.  相似文献

Proteins synthesized by the mixed microbial community of two sequencing batch reactors run for enhanced biological phosphorus removal (EBPR) during aerobic and anaerobic reactor phases were compared, using mass spectrometry‐based proteomics and radiolabelling. Both sludges were dominated by polyphosphate‐accumulating organisms belonging to Candidatis Accumulibacter and the majority of proteins identified matched closest to these bacteria. Enzymes from the Embden–Meyerhof–Parnas pathway were identified, suggesting this is the major glycolytic pathway for these Accumulibacter populations. Enhanced aerobic synthesis of glyoxylate cycle enzymes suggests this cycle is important during the aerobic phase of EBPR. In one sludge, several TCA cycle enzymes showed enhanced aerobic synthesis, suggesting this cycle is unimportant anaerobically. The second sludge showed enhanced synthesis of TCA cycle enzymes under anaerobic conditions, suggesting full or partial TCA cycle operation anaerobically. A phylogenetic analysis of Accumulibacter polyphosphate kinase genes from each sludge demonstrated different Accumulibacter populations dominated the two sludges. Thus, TCA cycle activity differences may be due to Accumulibacter strain differences. The major fatty acids present in Accumulibacter‐dominated sludge include palmitic, hexadecenoic and cis‐vaccenic acid and fatty acid content increased by approximately 20% during the anaerobic phase. We hypothesize that this is associated with increased anaerobic phospholipid membrane biosynthesis, to accommodate intracellular polyhydroxyalkanoate granules.  相似文献

Microbial community of acetate utilizing denitrifiers in aerobic granules   总被引:2,自引:0,他引:2  
Nitrite accumulates during biological denitrification processes when carbon sources are insufficient. Acetate, methanol, and ethanol were investigated as supplementary carbon sources in the nitrite denitrification process using biogranules. Without supplementary external electron donors (control), the biogranules degraded 200 mg l−1 nitrite at a rate of 0.27 mg NO2–N g−1 VSS h−1. Notably, 1,500 mg l−1 acetate and 700 mg l−1 methanol or ethanol enhanced denitrification rates for 200 mg l−1 nitrite at 2.07, 1.20, and 1.60 mg NO2–N g−1 VSS h−1, respectively; these rates were significantly higher than that of the control. The sodium dodecyl sulfate polyacrylamide gel electrophoresis of the nitrite reductase (NiR) enzyme identified three prominent bands with molecular weights of 37–41 kDa. A linear correlation existed between incremental denitrification rates and incremental activity of the NiR enzyme. The NiR enzyme activity was enhanced by the supplementary carbon sources, thereby increasing the nitrite denitrification rate. The capacity of supplementary carbon source on enhancing NiR enzyme activity follows: methanol > acetate > ethanol on molar basis or acetate > ethanol > methanol on an added weight basis.  相似文献

To investigate the diversities of Accumulibacter phosphatis and its polyhydroxyalkanoate (PHA) synthase gene (phaC) in enhanced biological phosphorus removal (EBPR) sludge, an acetate-fed sequencing batch reactor was operated. Analysis of microbial communities using fluorescence in situ hybridization and 16S rRNA gene clone libraries showed that the population of Accumulibacter phosphatis in the EBPR sludge comprised more than 50% of total bacteria, and was clearly divided into two subgroups with about 97.5% sequence identity of the 16S rRNA genes. PAO phaC primers targeting the phaC genes of Accumulibacter phosphatis were designed and applied to retrieve fragments of putative phaC homologs of Accumulibacter phosphatis from EBPR sludge. PAO phaC primers targeting G1PAO, G2PAO, and G3PAO groups produced PCR amplicons successfully; the resulting sequences of the phaC gene homologs were diverse, and were distantly related to metagenomic phaC sequences of Accumulibacter phosphatis with 75-98% DNA sequence identities. Degenerate NPAO (non-PAO) phaC primers targeting phaC genes of non- Accumulibacter phosphatis bacteria were also designed and applied to the EBPR sludge. Twenty-four phaC homologs retrieved from NPAO phaC primers were different from the phaC gene homologs derived from Accumulibacter phosphatis, which suggests that the PAO phaC primers were specific for the amplification of phaC gene homologs of Accumulibacter phosphatis, and the putative phaC gene homologs by PAO phaC primers were derived from Accumulibacter phosphatis in the EBPR sludge. Among 24 phaC homologs, a phaC homolog (G1NPAO-2), which was dominant in the NPAO phaC clone library, showed the strongest signal in slot hybridization and shared approximately 60% nucleotide identity with the G4PAO group of Accumulibacter phosphatis, which suggests that G1NPAO-2 might be derived from Accumulibacter phosphatis. In conclusion, analyses of the 16S rRNA and phaC genes showed that Accumulibacter phosphatis might be phylogenetically and metabolically diverse.  相似文献

Aerobic granules are the potential tools to develop modern wastewater treatment technologies with improved nutrient removal efficiency. These granules have several promising advantages over conventional activated sludge-based wastewater treatment processes. This technology has the potential of reducing the infrastructure and operation costs of wastewater treatment by 25%, energy requirement by 30%, and space requirement by 75%. The nutrient removal mechanisms of aerobic granules are slightly different from that of the activated sludge. For instance, unlike activated sludge process, according to some reports, as high as 70% of the total phosphorus removed by aerobic granules were attributed to precipitation within the granules. Similarly, aerobic granule-based technology reduces the total amount of sludge produced during wastewater treatment. However, the reason behind this observation is unknown and it needs further explanations based on carbon and nitrogen removal mechanisms. Thus, as a part of the present review, a set of new hypotheses have been proposed to explain the peculiar nutrient removal mechanisms of the aerobic granules.  相似文献

Enhanced biological phosphorus removal (EBPR) is one of the most advanced and complicated wastewater treatment processes applied today, and it is becoming increasingly popular worldwide as a sustainable way to remove and potentially reuse P. It is carried out by complex microbial communities consisting primarily of uncultured microorganisms. The EBPR process is a well-studied system with clearly defined boundaries which makes it very suitable as a model ecosystem in microbial ecology. Of particular importance are the transformations of C, N, and P, the solid-liquid separation properties and the functional and structural stability. A range of modern molecular methods has been used to study these communities in great detail including single cell microbiology, various -omics methods, flux analyses, and modeling making this one of the best studied microbial ecosystems so far. Recently, an EBPR core microbiome has been described and we present in this article some highlights and show how this complex microbial community can be used as model ecosystem in environmental biotechnology.  相似文献

The purpose of this work was to determine optimum sequence retention times for nutrient removal with low-cost using very short aeration time in an SBR treating domestic wastewater. During the study, four different CYCLEs were evaluated, with the highest removal efficiencies recorded for the CYCLE with fill, anaerobic, aerobic1, anoxic, aerobic2, settle, and decant sequences operated at retention times of 0.5, 2, 2, 1, 0.75, 1, and 0.5 h, respectively. For this CYCLE, the removal efficiencies of chemical oxygen demand (COD), total Kjeldahl nitrogen (TKN), ammonia nitrogen (NH3–N), total phosphorus (TP), and ortho-phosphate (PO4–P) were found, on average, to be 91, 78, 85, 87, and 83%, respectively. The optimum sequence retention time was determined via the analysis of variance (ANOVA) using the Matlab software (Mathworks Inc.). The data indicated that the total time of the aerobic sequences was shorter than those of previous studies for similar level of removal efficiencies in all parameters including N and P.  相似文献

Aerobic granules are dense microbial aggregates with the potential to replace floccular sludge for the treatment of wastewaters. In bubble-column sequencing batch reactors, distinct microbial populations dominated propionate- and acetate-cultivated aerobic granules after 50 days of reactor operation when only carbon removal was detected. Propionate granules were dominated by Zoogloea (40%), Acidovorax, and Thiothrix, whereas acetate granules were mainly dominated by Thiothrix (60%). Thereafter, an exponential increase in enhanced biological phosphorus removal (EBPR) activity was observed in the propionate granules, but a linear and erratic increase was detected in the acetate ones. Besides Accumulibacter and Competibacter, other bacterial populations found in both granules were associated with Chloroflexus and Acidovorax. The EBPR activity in the propionate granules was high and stable, whereas EBPR in the acetate granules was erratic throughout the study and suffered from a deterioration period that could be readily reversed by inducing hydrolysis of polyphosphate in presumably saturated Accumulibacter cells. Using a new ppk1 gene-based dual terminal-restriction fragment length polymorphism (T-RFLP) approach revealed that Accumulibacter diversity was highest in the floccular sludge inoculum but that when granules were formed, propionate readily favored the dominance of Accumulibacter type IIA. In contrast, acetate granules exhibited transient shifts between type I and type II before the granules were dominated by Accumulibacter type IIA. However, ppk1 gene sequences from acetate granules clustered separately from those of propionate granules. Our data indicate that the mere presence of Accumulibacter is not enough to have consistently high EBPR but that the type of Accumulibacter determines the robustness of the phosphate removal process.  相似文献

Atmospheric concentrations of carbon tetrachloride (CCl(4)) were removed by bulk aerobic soils from tropical, subtropical, and boreal environments. Removal was observed in all tested soil types, indicating that the process was widespread. The flux measured in field chamber experiments was 0.24 ± 0.10 nmol CCl(4) (m(2) day)(-1) (average ± standard deviation [SD]; n = 282). Removal of CCl(4) and removal of methane (CH(4)) were compared to explore whether the two processes were linked. Removal of both gases was halted in laboratory samples that were autoclaved, dry heated, or incubated in the presence of mercuric chloride (HgCl(2)). In marl soils, treatment with antibiotics such as tetracycline and streptomycin caused partial inhibition of CCl(4) (50%) and CH(4) (76%) removal, but removal was not affected in soils treated with nystatin or myxothiazol. These data indicated that bacteria contributed to the soil removal of CCl(4) and that microeukaryotes may not have played a significant role. Amendments of methanol, acetate, and succinate to soil samples enhanced CCl(4) removal by 59%, 293%, and 72%, respectively. Additions of a variety of inhibitors and substrates indicated that nitrification, methanogenesis, or biological reduction of nitrate, nitrous oxide, or sulfate (e.g., occurring in possible anoxic microzones) did not play a significant role in the removal of CCl(4). Methyl fluoride inhibited removal of CH(4) but not CCl(4), indicating that CH(4) and CCl(4) removals were not directly linked. Furthermore, CCl(4) removal was not affected in soils amended with copper sulfate or methane, supporting the results with MeF and suggesting that the observed CCl(4) removal was not significantly mediated by methanotrophs.  相似文献

The process kinetics of a lab-scale upflow aerobic immobilized biomass (UAIB) reactor using simulated sugar-manufacturing wastewater as feed was investigated. The experimental unit consisted of a 22l reactor filled with high porosity pumice stone. The UAIB reactor was tested under different organic loads and different hydraulic retention times (HRT) and the substrate loading removal rate was compared with prediction of Stover-Kincannon model, second-order model and the first order substrate removal model. After obtaining steady-state conditions, organic loading rate was increased from 750 to 4500 g COD/m(3) day to resemble wastewater from sugar production lines, and hydraulic retention time was decreased from 1 to 0.5 days, stepwise. Nine different operational conditions were applied changing these two parameters in a certain program. As a result of the calculations, Stover-Kincannon model and second-order model known as "Grau" model were found to be the most appropriate models for this reactor. Stover-Kincannon model and Grau second-order model gave high correlation coefficients, which were 99.7% and 99.4%, respectively. Therefore, these models could be used in predicting the behavior or design of the UAIB reactors.  相似文献

The spatial distribution, quantity and diversity of different microorganisms within anaerobic granular sludge from a lab-scale expanded granular sludge bed (EGSB) reactor operated at different organic loading rates were studied using florescent in situ hybridization (FISH), real time quantitative — polymerase chain reaction (RTQ-PCR) and denaturing gradient gel electrophoresis (DGGE) techniques. The results indicated that most Eubacteria were located in the outer layer of granule, while the Archaea which mainly were methanogens and more sensible to the environmental conditions were located in the inner layer of the granule. The quantity of Archaea was obviously less than that of Eubacteria in the granules, but increased with the increasing of organic loading rates of the reactor. As the organic loading rate of the reactor increased and the operating time elapsed, the Archaea community in the granules changed significantly. Seven typical DGGE bands were collected and sequenced, and found that the dominant species of Archaea in the granules operated in the last period were mainly Methanocorpusculum, Methanobacterium, Methanosaeta.  相似文献

The spatial distribution and activities of nitrifying and denitrifying bacteria in sponge media were investigated using diverse tools, because understanding of in situ microbial condition of sponge phase is critical for the successful design and operation of sponge media process. The bacterial consortia within the media was composed of diverse groups including a 14.5% Nitrosomonas spp.-like ammonia oxidizing bacteria (AOB), 12.5% Nitrobacter spp.-like nitrite oxidizing bacteria (NOB), 2.0% anaerobic ammonium-oxidizing (ANAMMOX) bacteria and 71.0% other bacteria. The biofilm appeared to be most dense in the relatively outer region of the media and gradually decreased with depth, but bacterial viabilities showed space-independent feature. The fluorescent in situ hybridization results revealed that AOB and NOB co-existed in similar quantities on the side fragments of the media, which was reasonably supported by the microelectrode measurements showing the concomitant oxidation of NH(4) (+) and production of NO(3) (-) in this zone. However, a significantly higher fraction of AOB was observed in the center than side fragment. As with the overall biofilm density profile, the denitrifying bacteria were also more abundant on the side than in the center fragments. ANAMMOX bacteria detected throughout the entire depth offer another advantage for the removal of nitrogen by simultaneously converting NH(4) (+) and NO(2) (-) to nitrogen gas.  相似文献

This study studied the cultivation of granules from an expanded granular sludge bed reactor that simultaneously transforms sulfates, nitrates, and oxygen to elementary sulfur, nitrogen gas, and carbon dioxides, respectively. The living cells accumulate at the granule outer layers, as revealed by the multicolor staining and confocal laser scanning microscope technique. The microbial community comprises sulfate-reducing bacteria (SRB, Desulfomicrobium sp.), heterotrophic (Pseudomonas aeruginosa and Sulfurospirillum sp.), and autotrophic denitrifiers (Sulfurovum sp. and Paracoccus denitrificans) whose population dynamics at different sulfate and nitrate loading rates are monitored with the single-strand conformation polymorphism and denaturing gradient gel electrophoresis technique. The Desulfomicrobium sp. presents one of the dominating strains following reactor startup. At high sulfate and nitrate loading rates, the heterotrophic denitrifiers overcompete autotrophic denitrifiers to reduce SRB activities. Conversely, suddenly reducing nitrate loading rates completely removes the heterotrophic denitrifier Sulfurospirillum sp. from the granules and activates the autotrophic denitrifiers. The physical fixation of different groups of functional strains in granules fine-tunes the strains' activities, and hence the reactor performance.  相似文献

The evolution of a microbial community was investigated during sludge granulation using a wide range of micro-scale and molecular biology techniques. Experimental results demonstrate that polyphosphate-accumulating granules were successfully cultured during the anaerobic/aerobic cycle. Improvement in sludge sedimentation performance occurred prior to the formation of granular sludge and was not affected by change in granule size. Rod-shaped and filamentous bacteria appeared to initiate granule formation and generate the structures that supported further granule growth. It was observed that mature granules supported microbial populations that differed from nascent granules and were predominantly packed with coccoid bacteria. It was further observed that the diversity of the granular microbial community increased as the granules grew. Accumulibacter, Nitrosospira and Thauera were mainly responsible for nutrient removal while microorganisms such as Rhodocyclus and Hyphomicrobiaceae appeared to be primarily responsible for forming and maintaining the granule structure.  相似文献

Nutrient removal from synthetic wastewater was investigated using a four-step sequencing batch reactor (SBR) at different phenol (C6H5OH) concentrations in order to determine the inhibition effects of phenol on biological nutrient removal. The nutrient removal process consisted of anaerobic, oxic, anoxic, and oxic phases with hydraulic residence times (HRT) of 1 h/3 h/1 h/1 h and a settling phase of 3/4 h. Solids retention time (SRT) was kept constant at 10 days in all experiments. Initial phenol concentrations were varied between 0 and 600 mg l−1 at seven different levels. The effects of phenol on COD, NH4-N, and PO4-P removals and effluent nutrient levels were investigated. Phenol was almost completely degraded up to 400 mg l−1 phenol concentration resulting in almost negligible inhibition effects on COD, NH4-N, and PO4-P removals. Nutrient removals were adversely affected by phenol at concentrations above 400 mg l−1. Above 95% COD, 90% NH4-N and 65% PO4-P removal was obtained for phenol concentrations below 400 mg l−1. The sludge volume index (SVI) was almost constant around 45 ml g−1 for phenol concentrations below 400 mg l−1 but increased to 90 ml g−1 at a phenol level of 600 mg l−1.  相似文献

Sequencing batch operation was used for nutrient (COD, NH4-N, NO3-N, PO4-P) removal from synthetic wastewater by using different carbon sources. Operation consisted of anaerobic, anoxic, oxic, anoxic and oxic (An/Ax/Ox/Ax/Ox) phases with durations of 2/1/4.5/1.5/1.5 h. Glucose, acetate and a mixture of glucose/acetate were used as carbon source to yield a COD/N/P ratio of 100/5/1.5 in the feed. Sludge age was kept constant at 10 days. COD, NH4-N, NO3-N and PO4-P removal efficiencies were maximum at the levels of 96%, 87%, 81% and 90% respectively, when a mixture (50/50) of glucose and acetate was used.  相似文献

The use of phosphate rocks as low-solubility phosphorus fertilizers has been promoted to reduce the environmental impacts of agriculture, but adequate nutrient uptake by plants depends on solubilization of the rock, driven by soil microorganisms. Here, investigation was made of the microbial solubilization of low-solubility phosphate rocks, together with simultaneous bioprotective action involving the biocontrol of microorganisms. The aim was to enhance function and value by delivering two effects using a single bio-based product, in accordance with the concept of a “bioreactor-in-a-granule” system. A composite structure was developed, based on a starch matrix, comprising a combination of Trichoderma asperelloides, as a biocontrol agent, and Aspergillus niger, as an acidulant. A significant increase of up to 150% in P solubilization was achieved, indicating the positive effect of the microorganism-composite interaction. In vitro assays showed that the ability of T. asperelloides to inhibit Fusarium oxysporum mycelial growth was maintained in the presence of A. niger. Moreover, the estimated cost of the composite granule (0.35 US$/kg of product on a dry basis) revealed competitive. The results indicated that the association of T. asperelloides and A. niger is an effective way to increase nutrient availability and to inhibit plant pathogens, opening up possibilities for the design of multifunctional bio-based fertilizer composites.  相似文献
