Protein synthesis in dormant embryos of sugar maple ( Acer saccharum ) was investigated in seeds stratified at 4°C or incubated at 15°C. Seeds stratified at 4°C germinated after 27 days; seeds incubated at 15°C failed to germinate. Stratification increased the embryo's capacity for protein synthesis by day 11 as measured by in vivo incorporation of [35 S]-methionine into purified protein. At 4°C protein synthesis in the embryonic axis rose in a linear fashion prior to germination, whereas in cotyledons it increased until day 20 and then declined. Analysis of radiolabelled proteins by two-dimensional gel electrophoresis revealed that the levels of specific proteins were altered by temperature, primarily in the cotyledons. Several proteins were expressed in the cotyledons at 15°C but were absent in unstratified embryos and in embryos stratified at 4°C. That is, the expression of these proteins was repressed during stratification and release from dormancy. Levels of other proteins in the cotyledons declined at 4°C during stratification. We suggest that one or more of these proteins may be associated with the inhibition of growth of the embryonic axis imposed by the cotyledons. 相似文献

Simulated nitrogen deposition affects community structure of arbuscular mycorrhizal fungi in northern hardwood forests 总被引:1,自引:0,他引:1
Our previous investigation found elevated nitrogen deposition caused declines in abundance of arbuscular mycorrhizal fungi (AMF) associated with forest trees, but little is known about how nitrogen affects the AMF community composition and structure within forest ecosystems. We hypothesized that N deposition would lead to significant changes in the AMF community structure. We studied the diversity and community structure of AMF in northern hardwood forests after more than 12 years of simulated nitrogen deposition. We performed molecular analyses on maple (Acer spp.) roots targeting the 18S rDNA region using the fungal‐specific primers AM1 and NS31. PCR products were cloned and identified using restriction fragment length polymorphism (RFLP) and sequencing. N addition significantly altered the AMF community structure, and Glomus group A dominated the AMF community. Some Glomus operational taxonomic units (OTUs) responded negatively to N inputs, whereas other Glomus OTUs and an Acaulospora OTU responded positively to N inputs. The observed effect on community structure implies that AMF species associated with maples differ in their response to elevated nitrogen. Given that functional diversity exists among AMF species and that N deposition has been shown to select less beneficial fungi in some ecosystems, this change in community structure could have implications for the functioning of this type of ecosystem. 相似文献

Vesicular-arbuscular mycorrhizal spore populations in sugar maple (Acer saccharum marsh. L.) forests
The numbers and types of spores of vesicular-arbuscular mycorrhizal fungi occurring in the top 15 cm of the soil in three
maple forests in Eastern Canada were investigated using traditional wet-sieving/decanting methods. In the most acid site,
at St. Hippolyte, Québec, where the soil had been amended with base cations, after 1 year there was no effect on the numbers
of spores present. Vesicular-arbuscular mycorrhizal spores present at St. Hippolyte consisted of Glomus rubiforme, other Glomus spp. and Acaulospora spp. Although the sporocarpic species, G. aggregatum, G. macrocarpum and G. rubiforme occurred at St. Hippolyte, they were not found at the two less acid sites (Waterloo, Ontario and Lacolle, Québec) . of Acaulospora spp. were found at all three sites, but were most abundant at St. Hippolyte. At St. Hippolyte the total number of spores
was much higher than at the other two sites; at Waterloo numbers were an order of magnitude lower than at St. Hippolyte. It
is suggested that G. rubiforme and Acaulospora species may be adapted to acid conditions. Seasonal patterns of spore abundance suggested that Acaulospora spp. may sporulate during the spring, whereas G. rubiforme may sporulate during the fall.

Relatively high levels of dihydrozeatin and trans-zeatin were detected in unstratified seeds of Acer saccharum Marsh. Both cytokinins increased substantially over the first 20 days of stratification at 5°C and then fell rapidly to values well below original levels by first germination on day 55. In seeds held at 20°C, a non-afterripening temperature, cytokinin levels remained constant for the first 10 days and then declined to their lowest levels by day 20. Levels of putrescine, spermidine and spermine in the radicles and cotyledons did not change during the full course of the afterripening process, but large increments were noted during radicle emergence. A large increase in ethylene production at germination suggests that competition for S-adenosyl-methionine by the ethylene and polyamine biosynthetic pathways did not inhibit synthesis of ethylene or polyamines during seedling emergence and establishment. In seeds stratified at 20°C, ethylene showed an exceptionally large peak early in the stratification period, but polyamine contents remained low throughout the test. The present results are consistent with the hypothesis that cytokinins play a significant role in overcoming the metabolic block present in dormant seeds. This conclusion is supported by data showing that high levels of cytokinins develop concurrently with the start of tissue differentiation and at the time when abscisic acid and phenolic inhibitors decline in stratifying seeds. Changes in ethylene and polyamine contents did not correlate with any events in the afterripening process; however, large increases in levels of these substances were closely associated with the germinative process and, in the case of polyamines, specifically with the start of cell division. 相似文献

Arbuscular mycorrhizal (AM) fungi are important below-ground carbon (C) sinks that can be sensitive to increased nitrogen (N) availability. The abundance of AM fungi (AMF) was estimated in maple (Acer spp.) fine roots following more than a decade of experimental additions of N designed to simulate chronic atmospheric N deposition. Abundance of AMF was measured by staining and ocular estimation, as well as by analyzing for the AMF indicator fatty acid 16:1omega5c in phospholipid (biomass indicator) and neutral lipid (lipid storage indicator) fractions. Arbuscular mycorrhizal fungal biomass, storage structures and lipid storage declined in response to N addition measured by both methods. This pattern was found when AM response was characterized as colonization intensity, on an areal basis and in proportion to maple above-ground biomass. The phospholipid fraction of the fatty acid 16:1omega5c was positively correlated with total AMF colonization and the neutral lipid fraction with vesicle colonization. Decreased AMF abundance with simulated N deposition suggests reduced C allocation to these fungi or a direct soil N-mediated decline. The fatty acid (phospholipid and neutral lipid fractions) 16:1omega5c was found to be a good indicator for AMF active biomass and stored energy, respectively. 相似文献

Inorganic polyphosphate (poly P) has been considered to be a translocatable form of phosphate (Pi) in arbuscular mycorrhizal fungi (AMF). Here we examined time-course changes in poly P content during the AMF colonization process. Onion (Allium cepa) plants were cultured with or without inoculation with Gigaspora margarita for 2-8 wk with periodic sampling. Poly P in the extracts, purified through gel filtration, was quantified by the reverse reaction of polyphosphate kinase. The length of poly P in mycorrhizal roots appeared to be shorter than in extraradical hyphae or in spores of the AMF, indicating that AMF depolymerize poly P before providing Pi to the host. The poly P content increased as colonization proceeded, and was highly correlated with the weight of the colonized roots. These results support the model that AMF supply Pi to the host through the poly P pool, and that the poly P content of a mycorrhizal root can be a good indicator of the Pi-supplying activity of AMF. 相似文献

A study was conducted in the Lower Laurentians of southern Quebec to test the hypothesis that base cation fertilization would improve the nutrient status of declining sugar maples (Acer saccharum Marsh.) and alter the partitioning of leaf C and N. Six 40×40 m plots were delineated in an 80 year old stand of sugar maple. Three plots received a mixture of fertilizer and liming materials (500 kg ha–1 of K2SO4, 250 kg ha–1 of CaCO3 and 250 kg ha–1 of CaMg(CO2)2) in the spring of 1989. Leaves from mid crown of dominant or co-dominant maples were sampled monthly during the 1990 growing season. Trees were cored in 1992 to measure their response in diameter growth. Fertilization increased diameter growth and foliar K concentration of trees but reduced foliar Ca concentration. Fertilization resulted in lower starch concentrations and higher ratios of soluble sugars to starch in June and September, and in higher free amino acid concentrations but lower ratio of total non-structural carbohydrates to free amino acids in September. Leaf proline concentration was correlated with leaf starch concentration (r=0.39). The results suggest that amelioration of K deficiencies in sugar maple through fertilization with a mixture of base cations can increase tree growth and affect the seasonal dynamics of foliar C and N pools.Abbreviations FAA

* It is commonly hypothesized that stand-level fine root biomass increases as soil fertility decreases both within and among tropical forests, but few data exist to test this prediction across broad geographic scales. This study investigated the relationships among fine roots, arbuscular mycorrhizal (AM) fungi and soil nutrients in four lowland, neotropical rainforests. * Within each forest, samples were collected from plots that differed in fertility and above-ground biomass, and fine roots, AM hyphae and total soil nutrients were measured. * Among sites, total fine root mass varied by a factor of three, from 237+/-19 g m-2 in Costa Rica to 800+/-116 g m-2 in Brazil (0-40 cm depth). Both root mass and length were negatively correlated to soil nitrogen and phosphorus, but AM hyphae were not related to nutrients, root properties or above-ground biomass. * These results suggest that understanding how soil fertility affects fine roots is an additional factor that may improve the representation of root functions in global biogeochemical models or biome-wide averages of root properties in tropical forests. 相似文献

Current ecosystem model predictions concerning the effects of global temperature increase on forest responses do not account for factors influencing long‐term evolutionary dynamics of natural populations. Population structure and genetic variability may represent important factors in a species' ability to adapt to global‐scale environmental change without experiencing major alterations in current range limits. Genetic variation and structure in sugar maple (Acer saccharum Marsh.) were examined across three regions, between two stands within regions, and among four to five open‐pollinated families within stands (total N = 547 genotypes) using 58 randomly amplified polymorphic DNA (RAPD) markers. Differences within open‐pollinated families account for the largest portion of the total variation (29%), while differences among regions represent less than 2% of the total variation. Genetic diversity, as indicated by estimates of percent polymorphic loci, expected heterozygosity, fixation coefficients, and genetic distance, is greatest in the southern region, which consists of populations with the maximum potential risk due to climate change effects. The high level of genetic similarity (greater than 90%) among some genotypes suggests that gene flow is occurring among regions, stands, and families. High levels of genetic variation among families indicate that vegetational models designed to predict species' response to global‐scale environmental change may need to consider the degree and hierarchical structure of genetic variation when making large‐scale inferences. 相似文献

The nutritional benefits that mycorrhizal associations provide to plants may be constrained by acidic soil conditions resulting in decreased photosynthetic function. Sugar maple (Acer saccharum) and red maple (Acer rubrum) seedlings were grown on a native acidic (pH 4.1) soil both unamended and amended with base cations (pH 6.2). In a second study a fungicide treatment was included. Foliar nutrition, mycorrhizal colonization, photosynthesis and their relationships were assessed. On the native soil, red maple maintained higher levels of mycorrhizal colonization and photosynthesis than sugar maple but showed little response to base cation amendments. Mycorrhizal colonization and photosynthesis of sugar maple increased significantly in response to base cation amendments. Correlations were observed among mycorrhizal colonization, foliar nutrition and photosynthesis. The fungicide treatment indicated that 50% of the base cation-induced increase in sugar maple photosynthesis was mycorrhiza related. The results suggest that base cation stimulation of mycorrhization and photosynthesis of sugar maple on acid soils are coupled by foliar nutrient dynamics. Red maple exhibits much less sensitivity to these same edaphic conditions. 相似文献

Hydraulic lift and its influence on the water content of the rhizosphere: an example from sugar maple,Acer saccharum 总被引:2,自引:0,他引:2
Hydraulic lift, the transport of water from deep in the soil through plant root systems into the drier upper soil layers, has been demonstrated in several woody plant species. Here the volume of water involved in hydraulic lift by a mature sugar maple tree is estimated. Twenty-four intact soil cores were collected from the vicinity of a sugar maple tree at the same positions at which thermocouple psychrometers had been placed. Desorption measurements were made on the soil cores and the data were fitted to the Campbell relation for soil matric potential versus soil water content . The psychrometer data were filtered to obtain the diurnal component contributed by hydraulic lift. The diurnal component in was combined with the Campbell relation for each soil core to obtain the increase in soil water content due to hydraulic lift. The additional water contents were numerically integrated to obtain a volume of 102±54 1 of water which was hydraulically lifted each night. The volume of hydraulically lifted water (HLW) is sufficiently great that in ecosystems where hydraulic lift occurs it should be included in models for calculating the water balance. However, a previous analysis of the stable hydrogen isotope composition (D) of water in understory plants around trees conducting hydraulic lift implies a much greater volume of HLW than that calculated from the analysis performed above. To reconcile these differences, it is hypothesized that some understory plants preferentially extract HLW due to its higher matric potential and that the proportion of this water source within the xylem sap of at least some understory plants that use HLW was so great that the roots of these plants must therefore be in close proximity to the tree roots from which the HLW comes. The results of this study have implications for studies of plant competition where positive associations may exist as well as for ion uptake, nutrient cycling and the design of agroforestry systems. 相似文献
