Three kinds of Schiff bases of carboxymethyl chitosan (CMCTS) were prepared, and their antifungal activities were assessed according to Jasso de Rodríguez's method. The results indicated that 2-(2-hydroxybenzylideneamino)-6-carboxymethylchitosan (HNCMCTS) and 2-(5-chloro-2-hydroxybenzylideneamino)-6-carboxymethylchitosan (HCCMCTS) had better inhibitory effects than those of chitosan or CMCTS against Fusarium oxysporium f. sp. vasinfectum, Alternaria solani, and Valsa mali.  相似文献

Schiff bases of chitosan, N-substituted chitosan, and quaternized chitosan were synthesized and their antifungal properties were analyzed against Botrytis cinerea Pers. (B. cinerea pers.) and Colletotrichum lagenarium (Pass) Ell.et halst (C. lagenarium (Pass) Ell.et halst) based on the method of D. Jasso de Rodríguez and co-workers. The results showed that quaternized chitosan had better inhibitory properties than chitosan, Schiff bases of chitosan, and N-substituted chitosan.  相似文献

Superoxide anion scavenging activity of graft chitosan derivatives   总被引:9,自引:0,他引:9  
Two kinds of graft chitosan derivatives (CMCTS-g-MAS and HPCTS-g-MAS) were prepared by the graft copolymerization of maleic acid sodium onto etherified chitosans-carboxymethyl chitosan (CMCTS) and hydroxypropyl chitosan (HPCTS), respectively. Superoxide anion scavenging activity of the derivatives was evaluated in a luminal-enhanced autoxidaton of pyrogallol by chemiluminescence techniques. Compared with chitosan, the graft chitosan derivatives have much improved scavenging ability against superoxide anion. They have similar 50% inhibition concentrations (IC50s) as ascorbic acid and superoxide dismutase (SOD). Graft chitosan derivatives with hydroxypropyl groups have relatively higher superoxide anion scavenging ability owing to the incorporation of hydroxyl groups. The graft chitosan derivatives (HPCTS-g-MAS 1, 2, and 3) with different grafting percentages exhibit IC50s values ranging from 243 to 308 μg/mL, which could be related to the contents of active hydroxyl and amino groups in the polymer chains.  相似文献

Selective N-arylation of chitosan was performed via a Schiff bases formed by the reaction between the 2-amino group of glucosamine residue of chitosan with an aromatic aldehyde under acidic condition followed by reduction of the Schiff base intermediate with sodium cyanoborohydride (Borch reduction). Aromatic aldehydes bearing either an electron donating or electron withdrawing substituent were used. The chemical structures and thermal properties of the N-aryl chitosans were characterized by FT-IR, (1)H NMR, (13)C NMR, TGA, and DSC. The extent of N-substitution (ES) was influenced by the molar ratio of the aldehyde to the glucosamine residue of chitosan, the reaction time and the substituent on the aromatic ring. Lower ESs resulted from N-arylation using an aldehyde with an electron donating substituent. A linear relationship between the targeted ES and the ES obtained was observed when aldehydes bearing electron withdrawing substituents were employed.  相似文献

Chemical modification of chitosan by introducing quaternary ammonium moieties into the polymer backbone renders excellent antimicrobial activity to the adducts. In the present study, we have synthesized 17 derivatives of chitosan consisting of a variety of N-aryl substituents bearing either electron-donating or electron-withdrawing groups. Selective N-arylation of chitosan was performed via Schiff bases formed by the reaction between the 2-amino groups of the glucosamine residue of chitosan with aromatic aldehydes under acidic conditions, followed by reduction of the Schiff base intermediates with sodium cyanoborohydride. Each of the derivatives was further quaternized using N-(3-chloro-2-hydroxypropyl)trimethylammonium chloride (Quat-188) as the quaternizing agent that reacted with either the primary amino or hydroxyl groups of the glucosamine residue of chitosan. The resulting quaternized materials were water soluble at neutral pH. Minimum inhibitory concentration (MIC) antimicrobial studies of these materials were carried out on Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive) bacteria in order to explore the impact of the extent of N-substitution (ES) on their biological activities. At ES less than 10%, the presence of the hydrophobic substituent, such as benzyl and thiophenylmethyl, yielded derivatives with lower MIC values than chitosan Quat-188. Derivatives with higher ES exhibited reduced antibacterial activity due to low quaternary ammonium moiety content. At the same degree of quaternization, all quaternized N-aryl chitosan derivatives bearing either electron-donating or electron-withdrawing substituents did not contribute antibacterial activity relative to chitosan Quat-188. Neither the functional group nor its orientation impacted the MIC values significantly.  相似文献

The novel N-heterocyclic chitosan aerogel derivatives were prepared by reacting 79% deacetylated chitosan separately with 4-pyridinecarboxaldehyde and 2,6-pyridinedicarboxaldehyde followed by subsequent solvent exchange into acetone, filteration and lyophilization. The identity of the Schiff bases was confirmed by UV–vis absorption spectroscopy and Fourier transform infrared (FTIR) spectroscopy. The N-heterocyclic chitosan derivatives were evaluated by X-ray diffraction (XRD), thermo-gravimetric analysis (TGA), differential scanning calorimetry (DSC), scanning electron microscopy (SEM), rheological studies and biological activity. Overall, the N-heterocyclic chitosan derivative based gels open new perspectives in biomedical applications.  相似文献

Some reactional parameters as mol ratio (salicylaldehyde:free amino groups), reaction time and temperature were investigated in order to improve the substitution degree (DS) in the preparation of biopolymeric Schiff bases from chitosan. In this case, the reaction of chitosan and salicylaldehyde was used as a probe system in order to produce the Schiff base. The use of 50% (mol/mol) salicylaldehyde excess, reaction time of 18 h and temperature of 55 °C permitted to obtain a DS of 60% without evidences of hydrolysis of the biopolymeric matrix or changes in its acetylation degree.  相似文献

N-substituted chitosan and quaternized chitosan were synthesized and their antioxidant activity against hydroxyl radicals was assessed, respectively. Compared with the antioxidant activity of chitosan, the results indicated that the two kinds of chitosan derivatives had different scavenging ability on hydroxyl radicals, which should be related to the form of amido in the two kinds of chitosan derivatives.  相似文献

Chitosan, a biocompatible, biodegradable, non-toxic polymer, is prepared from chitin, which is the second most naturally occurring biopolymer after cellulose. The Schiff base of chitosan was synthesized by the reaction of chitosan with citral working under high-intensity ultrasound. The effect of the molar ratio of chitosan to citral, reaction time, and temperature on the yield has been investigated. The optimal conditions were a temperature of 50 °C, a molar ratio of chitosan to citral of 1:6, and a reaction time of 10 h. The maximum yield achieved was 86.4% under optimum conditions. The structure of the Schiff base was characterized by FTIR spectroscopy, elemental analysis, and X-ray diffraction studies. The strong peaks at 1648.3 and 1610.6 cm−1 are due to CN and CC stretching vibrations. The results confirmed that amino groups on chitosan reacted with citral to form the Schiff base. The antimicrobial activities of chitosan and Schiff base of chitosan were investigated against Escherichia coil, Staphylococcus aureus, and Aspergillus niger. The results indicate that the antimicrobial activity of the Schiff base increases with an increase in the concentration. It was also found that the antimicrobial activity of the Schiff base was stronger than that of chitosan.  相似文献

N-Arylated chitosans were synthesized via Schiff bases formed by the reaction between the primary amino group of chitosan with aromatic aldehydes followed by reduction of the Schiff base intermediates with sodium cyanoborohydride. Treatment of chitosan containing N,N-dimethylaminobenzyl and N-pyridylmethyl substituents with iodomethane under basic conditions led to quaternized N-(4-N,N-dimethylaminobenzyl) chitosan and quaternized N-(4-pyridylmethyl) chitosan. Methylation occurred at either N,N-dimethylaminobenzyl and N-pyridylmethyl groups before the residual primary amino groups of chitosan GlcN units were substituted. The total degree of quaternization of each chitosan varied depending on the extent of N-substitution (ES) and the sodium hydroxide concentration used in methylation. Increasing ES increased the total degree of quaternization but reduced attack at the GlcN units. N,N-dimethylation and N-methylation at the primary amino group of chitosan decreased at higher ES’s. Higher total degrees of quaternization and degrees of O-methylation resulted when higher concentrations of sodium hydroxide were used. The molecular weight of chitosan before and after methylation was determined by gel permeation chromatography under mild acidic condition. The methylation of the N,N-dimethylaminobenzyl derivative with iodomethane was accompanied by numerous backbone cleavages and a concomitant reduction in the molecular weight of the methylated product was observed. The antibacterial activity of water-soluble methylated chitosan derivatives was determined using Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive) bacteria; minimum inhibitory concentrations (MIC) of these derivatives ranged from 32 to 128 μg/mL. The presence of the N,N-dimethylaminobenzyl and N-pyridylmethyl substituents on chitosan backbone after methylation did not enhance the antibacterial activity against S. aureus. However, N-(4-N,N-dimethylaminobenzyl) chitosan with degree of quaternization at the aromatic substituent and the primary amino group of chitosan of 17% and 16–30%, respectively, exhibited a slightly increased antibacterial activity against E. coli.  相似文献

Water-soluble chitosan derivatives were prepared by graft copolymerization of maleic acid sodium onto hydroxypropyl chitosan and carboxymethyl chitosan sodium. Their scavenging activities against hydroxyl radical *OH were investigated by chemiluminescence technique. They exhibit IC(50) values ranging from 246 to 498 microg/mL, which should be attributed to their different contents of hydroxyl and amino groups and different substituting groups.  相似文献

A chitosan Schiff base with an aromatic aldehyde was synthesized and characterized by FTIR and NMR spectroscopies. Furthermore, the degree of substitution was calculated based on the ratios of the area of the proton of the imine (Aimine) and the area of the peak of the proton of the pyranose ring (AH-2). The antimicrobial activities were determined against bacterial and fungal strains, as well as multiple drug-resistant (MDR) bacteria. The chitosan Schiff base was also tagged with medicinal plants, for example, Curcuma longa, Peganum harmala, Lepidium sativam, and cruciferous vegetables, and the biological activities determined against pathogenic bacterial and fungal strains. The chitosan Schiff base showed maximum zone of inhibition of 22 mm against Staphylococcus aureus with a minimum zone of inhibition of 15 mm against Bacillus cereus. The chitosan Schiff base was fused with C longa, isothiocyanates and a combined mixture of P harmala and L sativam that has shown activities against Escherichia coli with a zone of inhibition of 28, 24, and 30 mm, respectively. The Schiff base of chitosan fused with medicinal plants also showed significant inhibitory activities against MDR bacteria.  相似文献

Chitosan derivatives such as N-vanillyl chitosan and 4-hydroxybenzyl chitosan were prepared by reacting chitosan with 4-hydroxy-3-methoxybenzaldehyde (vanillin) and 4-hydroxybenzaldehyde. Amino groups on chitosan reacts with these aldehydes to form a Schiff base intermediate, which is later on converted into N-alkyl chitosans by reduction with sodium cyanoborohydride. The chemical reaction was monitored by 1H NMR spectroscopy and the absence of aldehydic proton at 9.83 ppm in NMR spectra was observed for both the modified chitosan derivatives confirming the reaction. Modified chitosan films were later prepared by solution casting method and their physico-mechanical, barrier, optical and thermal properties were studied. The results clearly indicated significant change in tensile strength, water vapour transmission rate, and haze properties of modified chitosans. Modified chitosan films were also studied for their antimicrobial activity against Aspergillus flavus. The results showed a marked reduction of aflatoxins produced by the fungus in the presence of the N-vanillyl chitosan and 4-hydroxybenzyl chitosan film discs to 98.9% and non-detectable levels, respectively.  相似文献

Chitosan (CS) and chitosan sulfates (CSS) with different molecular weight (Mw) were reacted with 4-acetamidobenzene sulfonyl chloride to obtain sulfanilamide derivatives of chitosan and chitosan sulfates (LSACS, HSACS, LSACSS, HSACSS). The preparation conditions such as different reaction time, temperature, solvent, and the molar ratio of reaction materials are discussed in this paper. Their structures were characterized by FTIR spectroscopy and elemental analyses. The antioxidant activities of the derivatives were investigated employing various established in vitro systems, such as hydroxyl-radical ((*)OH) superoxide anion (O2(*-)) scavenging and reducing power. All kinds of the compounds (HCS, LCS, HCSS, LCSS, HSACS, LSACS, HSACSS, LSACSS) showed stronger scavenging activity on hydroxyl radical than ascorbic acid (Vc). The inhibitory activities of the derivatives toward superoxide radical by the PMS-NADH system were obvious. The experiment showed that the superoxide radical scavenging effect of sulfanilamide derivatives of chitosan and chitosan sulfates was stronger than that of original CS and CSS. All of the derivatives were efficient in the reducing power. The results indicated that the sulfanilamide group were grafted on CS and CSS increased the reducing power of them obviously.  相似文献

Ascorbic acid immediately dissolves Euphausia superba chitosan upon mixing and forms chitosan ascorbate; during the 6-h period after dissolution in water at pH 5–7, ascorbate is oxidized to dehydroascorbate which undergoes Schiff reaction with the amino groups of chitosan, thus yielding a viscous solution of a polymeric ketimine. The latter is characterized by infrared spectrometry, circular dichroism spectropolarimetry, viscometry and alkalimetry. When brought into contact with transition metal ions, the chitosan ascorbate ketimine yields insoluble metal chelates. Upon reduction with sodium cyanoborohydride, the water-insoluble N-[2-(1,2-dihydroxyethyl)tetrahydrofuryl] chitosan (NDTC) is obtained, which shows enhanced capacity for uranium, up to 800 mg U/g from solutions at pH 4·5.  相似文献

In the present study, we have electrospun various grades of chitosan and cross-linked them using a novel method involving glutaraldehyde (GA) vapor, utilizing a Schiff base imine functionality. Chemical, structural, and mechanical analyses have been conducted by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and Kawabata microtensile testing, respectively. Additionally, the solubilities of the as-spun and cross-linked chitosan mats have been evaluated;solubility was greatly improved after cross-linking. SEM images displayed evidence that unfiltered low, medium, and high molecular weight chitosans, as well as practical-grade chitosan, can be electrospun into nanofibrous mats. The as-spun medium molecular weight chitosan nanofibers have a Young's modulus of 154.9 +/- 40.0 MPa and display a pseudo-yield point that arose due to the transition from the pulling of a fibrous mat with high cohesive strength to the sliding and elongation of fibers. As-spun mats were highly soluble in acidic and aqueous solutions. After cross-linking, the medium molecular weight fibers increased in diameter by an average of 161 nm, have a decreased Young's modulus of 150.8 +/- 43.6 MPa, and were insoluble in basic, acidic, and aqueous solutions. Though the extent to which GA penetrates into the chitosan fibers is currently unknown, it is evident that the cross-linking resulted in increased brittleness, a color change, and the restriction of fiber sliding that resulted in the loss of a pseudo-yield point.  相似文献

Chitosans with different degree of deacetylation were prepared from crab shell chitin in the presence of alkali. Aminoderivatized chitosan derivatives were prepared in addition of amino functional groups at a hydroxyl site in the chitosan backbone. Six kinds of aminoderivatized chitosan such as aminoethyl-chitosan (AEC90), dimethylaminoethyl-chitosan (DMAEC90), and diethylaminoethyl-chitosan (DEAEC90), which were prepared from 90% deacetylated chitosan, and AEC50, DMAEC50 and DEAEC50, which were prepared from 50% deacetylated chitosan, were prepared and their reactive oxygen species (ROS) scavenging activities were investigated against hydroxyl radical, superoxide anion radical and hydrogen peroxide. The electron spin resonance (ESR) spectrum revealed that AEC90 showed the highest scavenging effects against hydroxyl and superoxide anion radical, the effects were 91.67% and 65.34% at 0.25 and 5 mg/mL, respectively. For hydrogen peroxide scavenging effect, DEAEC90 exhibited the strongest activity. These results suggest that the scavenging effect depends on their degree of deacetylation and substituted group.  相似文献

研究低聚壳聚糖与木糖的美拉德反应,考察了两种体系(低聚壳聚糖与木糖的质量比分别为1∶1和1∶3)反应过程中pH、吸光度及荧光值的变化,醇沉法提取4 h和8 h的低聚壳聚糖美拉德反应衍生物,分别为CX11-4、CX13-4、CX11-8和CX13-8。对衍生物进行红外表征和分子量测定,并研究其对羟基自由基.OH和DPPH的清除能力以及还原能力。结果显示:壳聚糖衍生物的抗氧化能力都明显优于低聚壳聚糖,抗氧化活性顺序为CX13-4>CX11-4,CX11-8>CX13-8。可见,壳聚糖美拉德衍生物的抗氧化活性不仅与反应物的比例有关,还与反应的时间有关。  相似文献

Dihydroxyethyltetrahydrofuryl chitosan (which is obtained from Euphausia superba chitosan upon Schiff reaction with dehydroascorbic acid and subsequent reduction of the ketimine obtained) can accumulate uranium from aqueous solutions, including 3% saline waters. By this chemical modification, the characteristic property of the chitosan-glucan complexes of fungal origin which collect uranium by chelation and hydrolysis is conferred to krill and shrimp chitosans. The capacity of dihydroxyethyltetrahydrofuryl chitosan is 1·0 g U/g and is about one order of magnitude higher than that of the cells of Rhizopus arrhizus and about two orders of magnitude higher than the mycelia of Penicillium digitatum.  相似文献

pH- and thermo-sensitive (1→4)-2-amino-2-deoxy-β-d-glucan (i.e. chitosan) graft copolymer was prepared by reversible addition fragmentation chain transfer polymerizations of N-isopropylacrylamide with 4-methylbenzenesulfonic acid (i.e. tosylic acid)-chitosan complex. The polymerization was controlled well, and the amino group of chitosan could be deprotected easily and mildly with 15% Tris solution. The model aldehyde vanillin was conjugated with amino group of chitosan-g-PNIPAM via Schiff base bond (Loading efficiency, LE=77.6 mg/g), and the drug release could be controlled with temperature and pH. This property may promote the chitosan graft copolymer to be used in the field of "smart" drug delivery.  相似文献
