Summary In an attempt to discover A and B genome donor(s) to finger millet, Eleusine coracana, or its progenitor species, E. africana (both allotetraploid 2n=4x=36), five diploid species, E. Indica, E. Floccifolia, E. multiflora, E. tristachya and E. intermedia, were crossed to finger millet and its progenitor taxon. Crosses were successful only with E. coracana. Three combinations of triploid hybrids E. coracana x E. indica, E. coracana x E. floccifolia, and E. coracana x E. multiflora were obtained and analysed. Meiotic behaviour was perfectly normal in parental species. The regular number of 18 bivalents in E. coracana, 9 bivalents in E. indica, E. intermedia, E. tristachya and E. floccifolia and 8 bivalents in E. multiflora were invariably noticed. In E. coracana x E. indica hybrids a mean chromosome pairing of 8.84I+8.80II+0.03III+0.10IV per cell was found. About 86.5% of the cells showed the typical 9I+9II configuration, suggesting that E. indica (AA) is one of the diploid genome donors to cultivated species E. coracana. A mean chromosome pairing of 11.08I+7.63II+0.16III+0.04IV per cell was found in E. coracana x E. floccifolia hybrids. Two to ten bivalents and varying numbers of univalents were seen in 55% of the cells. About 45% of the cells showed the 9I+9II configuration. Various evidence suggests that perennial E. floccifolia is a primitive member of the A genome group of Eleusine species, and it may not be a genome donor to E. coracana. In E. coracana x E. multiflora hybrids (2n=26) mean chromosome pairing of 21.45I+1.97II+0.13III+0.04IV per cell was found. About 91% of the cells were observed to have 20–26 univalents. Only a small percentage of the cells contained bivalents or multivalents. This pairing behaviour indicates that E. multiflora lacks genomic homology with the A or B genome of E. coracana. Genomically E. multiflora is a distinct species and a genomic symbol of C is assigned to it. Identification of the B genome donor species to cultivated millet. E. coracana remains elusive.  相似文献

Summary We report here that similar to E6-chicken retinal cells, dissociated cells from 5.5-day-old (E5.5) quail retinae reaggregate in rotary culture, multiply about tenfold and reestablish histotypical areas. These cellular aggregates include all nuclear layers either with inversed or correct laminar polarity, depending on the local origin of the cells (called rosetted and laminar in-vitro-retinae (IVR), respectively; Layer and Willbold 1989). In combined cultures, chick and quail cells are evenly mixed only during the first two days of culture. Along with the assembly of single cells into rosettes and then into discrete laminae, sectors of chick and quail cells begin to segregate. They are delineated by borders running radially through all three nuclear layers. Thus, interspecies migration of cells at this advanced stage of differentiation is strongly inhibited. Concomitant with this segregation, coherent radial columns spanning all three layers but containing cells from either species only, can be traced histologically. We studied how the responses of four species of eucalypt to leaf litter related to their germination responses to light and water availability. Two of the species (Eucalyptus obliqua and E. baxteri) have a mesic distribution, while the other two (E. oleosa, and E. incrassata) are more xerophytic. We studied the effect of litter on emergence of the four species in a glasshouse experiment. Litter did not affect the emergence of E. incrassata and E. oleosa, but enhanced the emergence of E. obliqua and E. baxteri. Litter increased the seedling mortality of all four species. Germination responses to light and water availability were studied in growth cabinets under controlled conditions. The germination of E. obliqua and E. baxteri was substantially lower under fluorescent light than in darkness, but that of E. oleosa and E. incrassata was not affected by the light environment. The germination of E. obliqua and E. baxteri was significantly reduced by reduced water potential (a). Substantial germination of E. oleosa and E. incrassata occurred even at very low a (less than –1.05 MPa). We conclude that both the shade and the humid micro-environment provided by litter may have contributed to the emergence responses of the four species to litter, and these responses may correspond to ecological adaptations to the different environments in which they live.  相似文献

To create plots with differential levels ofEuseius, malathion sprays, highly toxic to phytoseiids but of low toxicity to citrus red mite,Panonychus citri (CRM), and citrus thrips,Scirtothrips citri, were applied to blocks of Valencia oranges for three seasons (1984, 1985 and 1986) and Eureka lemons for one season (1985). Releases ofEuseius spp. were made in plots within the treated blocks. In 1984, CRM populations were highest in the treated/no release plot, lowest in the untreated and intermediate in the treated/Euseius stipulatus-release plot. In 1985, CRM populations were high in all plots in the oranges (including theE. tularensis release) except theE. stipulatus-release plot, and in all plots in the lemons except the untreated.Euseius releases in the lemons had no effect, apparently because they were followed by very hot weather. In 1986, CRM was very low in all plots.Euseius stipulatus became the dominant phytoseiid species even in theE. tularensis-release plots after a few weeks, indicating that this species colonized more readily and spread to other trees more rapidly than didE. tularensis. The results support the hypothesis thatE. stipulatus is an important mortality factor contributing to CRM control at low levels.Euseius stipulatus also may have a negative impact on the citrus rust mite,Phyllocoptruta oleivora. Citrus thrips populations were low during the entire study period and did not differ significantly between plots.  相似文献

Competition in a natural system may be interspecific or intraspecific. In semiarid ecosystems, competition for resources between established neighboring grass species and newly recruited seedlings is very high. To examine the effects of grass species density, growing space and time of establishment on Eucalyptus victrix seedlings (interspecific competition), and the effect of density and growing space within E.victrix (intraspecific competition) we conducted an experiment under controlled conditions. We tested four hypotheses (i) E.victrix seedling growth is not affected by grass density; (ii) there is no difference in E.victrix survival and growth between early and later grass establishment; (iii) interspecific competition is not more intense than intraspecific competition in E.victrix; and (iv) growth of E.victrix seedlings is not dependent on available growing space. In a monoculture of E.victrix, seedling mortality was higher (10%) in large pots. In mixed culture pots, where E.victrix seedlings and grass seedlings were planted on the same day, E.victrix seedlings survived for up to 4weeks, but started to die after week five in the smallest pots. However, mortalities occurred in pots of all sizes when grass was established before E.victrix seedlings. Results also indicated that the resources necessary for the growth of individual E.victrix seedlings were more limiting under conditions of increased density of neighboring grass species rather than intraspecific competition. In particular, photosynthetic area of E.victrix seedlings was drastically reduced in mixed cultures. Although density, pot size and time of planting had impacts on E.victrix seedlings, the patterns of these impacts were variable.  相似文献

A near-shore coastal mangrove island receiving untreated sewage and a coastal cove receiving rum distillery effluent in Puerto Rico were examined for their ability to support survival and activity ofKlebsiella pneumoniae andEscherichia coli. Pure cultures of both bacteria were monitored for 96 hours in situ at both locations using membrane diffusion chambers.K. pneumoniae survived at all sites as measured by AODC and Coulter Counter direct counts. However, at the mangrove island less than 20% of theK. pneumoniae population was active (AODC) after the first 3 hours and less than 10% of this population was respiring (INT). In contrast, the coastal area which was receiving rum distillery effluent was able to maintain 40% of theK. pneumoniae population in an active state with 90% respiring. TheE. coli population declined by two orders of magnitude at the mangrove island, but remained unchanged at the rum distillery outfall. TheE. coli population had a higher proportion of active cells and respiring cells thanK. pneumoniae at all sites. At the rum distillery site, theE. coli population was remarkable in that 95% remained active and 99% were respiring. This study suggests that, when sufficient organic loading exists,E. coli, a nonsurvivor, can overcome the bactericidal effects of tropical marine waters.K. pneumoniae, a survivor, could survive under all conditions but could not maintain the activity or respiration that theE. coli population could, even when high organic loads were present. Morphological changes related to nutrient stress in the tropical marine environment were apparent inE. coli, but not inK. pneumoniae. Based on physiological activityE. coli is just as much a survivor asK. pneumoniae in tropical marine waters.  相似文献

Erythrina lectins possess similar structural and carbohydrate binding properties. Recently, tri- and tetra-antennary complex type carbohydrates with non-reducing terminal galactose residues have been shown to be precipitated as tri- and tetravalent ligands, respectively, with certainErythrina lectins [Bhattacharyya L, Haraldsson M, Brewer CF (1988) Biochemistry 271034-41]. The present work describes a comparative study of the binding and precipitating activities of fourErythrina lectins,viz. E. corallodendron, E. cristagalli, E. flabelliformis, andE. indica, with multi-antennary complex type carbohydrates and synthetic cluster glycosides. The results show that though their binding affinities are very similar, theErythrina lectins show large differences in their precipitating activities with the carbohydrates. The results also indicate significant dependence of the precipitating activities of the lectins on the core structure of the carbohydrates. Zusammenfassung Das orangerote Lipochrom im Gefieder des Roten Farbenkanarienvogels und des Kapuzenzeisigs (Spinus cucullatus) ist eine Mischfarbe, die aus Canthaxanthin und Kanarienxanthophyll besteht. Auch im Eidotter ist Canthaxanthin als Begleitpigment gelber Carotinoide vorhanden. Der Entzug der Nahrungscarotinoide hat in kürzester Zeit die fast völlige Depigmentierung der Federn zur Folge.Von fünf am Rotvogel getesteten definierten Carotinoidfarbstoffen sind vier von Einfluß auf die Gefiederfarbe: während Rhodoxanthin und Carotin nur schwache Rosafärbung der Federn hervorrufen und Lutein die normale orangerote Gefiederfarbe bedingt, ist die Wirkung von Canthaxanthin am intensivsten, indem es starke Rotfärbung zur Folge hat. Die Ergebnisse am Gelbvogel sind vergleichsweise erwähnt.Es werden die Möglichkeiten der Enstehung roter Lipochrome aus gelben Nahrungscarotinoiden besprochen.  相似文献

Summary Dihaploids (n=2x=14, SY) of two Elymus species, i.e., E. dolichatherus (Keng) Löve (2n=4x=28, SSYY) and E. brevipes (Keng) Löve (2n=4x=28, SSYY), were obtained from the interspecific hybrid combinations E. dolichatherus () x E. tibeticus (Meld.) G. Singh () and E. brevipes () x E. panormitanus (Parl.) Tzvelev (). The dihaploids were probably formed through selective elimination of male parental chromosomes in early embryo development. Meiotic chromosome behavior was studied in E. dolichatherus, E. brevipes, and their dihaploids. The two parental Elymus species had regular meioses with predominantly ring bivalent formation. A low frequency of homoeologous chromosome pairing was observed, with an average of 0.81 bivalents and 0.03 trivalents in the dihaploid of E. dolichatherus, and 0.26 bivalents in the dihaploid of E. brevipes. Up to two chromatid bridges accompanied by small fragments were present at anaphase I of the E. dolichatherus dihaploid. It is concluded from this study that: (i) both E. dolichatherus and E. brevipes are allotetraploid species; (ii) a low affinity exists between the S and Y genomes of the two Elymus species.  相似文献

Specimens of Euplotes were collected from Ross Sea (Antarctica), allowed to multiply in the laboratory, and taxonomically studied on the basis of classical diagnostic traits as seen under the optical and scanning electron microscope. They were assigned to three different morphospecies, all characterized by a dorsal silver-line system of the double type and a set of 10 fronto-ventral cirri. Two morphospecies were identified as new and named E. nobilii and E. euryhalinus; one was judged reconcilable with E. rariseta.  相似文献

Certain phototropism mutants ofPhycomyces blakesleeanus show defective bending responses (tropisms) to stimuli besides light, such as gravity, wind, and barriers. These so-called stiff mutants are affected in four genes (madD tomadG). Using two-dimensional gel electrophoresis, we have analyzed polypeptides from microsomal and soluble fractions obtained from the wild type, four single mutants, and six double mutants affected in all pairwise combinations of the four genes. Consistent differences in spot patterns formadE andmadF mutants were found in microsomal fractions but not in soluble fractions. InmadE mutants, two spots designated E1 (52 kDa,pI 6.65) and E2 (50 kDa,pI 6.65) were altered. E1 appeared denser in the wild type than in themadE mutants, while the reverse was true for E2. The spots E1 and E2 are probably under regulatory control bymadE, perhaps involving posttranslational modification. A protein spot, F1 (53 kDa,pI 6.1), was present on the wild-type gels but absent from all gels formadF mutants. Summary The ribosomal proteins of seven different Enterobacteriaceae were compared with those of E. coli by two-dimensional gel electrophoresis and by immunological methods. The ribosomal proteins of all Enterobacteriaceae were found to be very similar in molecular weight and in their electrophoretic properties. However, more dissimilarities could be detected by immunological methods thus indicating that few, if any, of the ribosomal proteins among the tested Enterobacteriaceae are identical. Ribosomes from all Enterobacteriaceae possess a protein which is electrophoretically identical with protein S7 from strain B (and not strain K) of E. coli.Paper No. 67 on Ribosomal Proteins. Effects of water stress duration and intensity on gas exchange and leaf water potential were investigated in 7-month-old seedlings of a humid coastal provenance (Gympie) and a dry inland (Hungry Hills) provenance of E. cloeziana F. Muell. and in a dry inland (Chinchilla) provenance of E. argophloia Blakely supplied with 100% (T100), 70% (T70), 50% (T50) of their water requirements, or were watered only after they were wilted at dawn (T0). Seedlings of E. argophloia had the highest midday net photosynthetic rate (A), stomatal conductance (g s), stomatal density and predawn leaf water potential (pd) in all treatments. The E. cloeziana provenances did not differ in these attributes. The T70 and T50 treatments caused reductions in A of 30% in E. argophloia, and 55% in the E. cloeziana provenances. Under the T0 treatment, E. argophloia maintained higher rates of gas exchange at all levels of water stress than E. cloeziana provenances. The estimates of pd and midday water potential (md) at which plants remained wilted overnight were respectively: –2.7 and –4.1 MPa for E. cloeziana (humid), –2.8 and –4.0 MPa for E. cloeziana (dry) and, –3.7 and –4.9 MPa for E. argophloia. Following stress relief, both A and g s recovered more quickly in E. argophloia and in the dry provenance of E. cloeziana than in the humid provenance. We conclude that E. argophloia is more drought tolerant and has a potential for cultivation in the humid and semi humid climates, whilst E. cloeziana has greater potential in the humid subtropical climates.  相似文献

Autoparasitoids are species of parasitic wasps in the family Aphelinidae which produce females as solitary primary endoparasitoids of homopterans such as whitefly and scale insects (primary hosts), and males as solitary hyperparasitoids. Males generally develop on immature conspecific females or on individuals of other primary parasitoid species (secondary hosts). Encarsia pergandiella is an autoparasitoid that has been introduced to Italy for control of greenhouse whitefly Trialeurodes vaporariorum, in greenhouses and field crops. In this study we examined the secondary host selection behaviour of this species with regard to conspecific females and females of two thelytokous species, E. formosa and E. meritoria. Encarsia formosa has been used successfully for greenhouse whitefly control in Northern Europe, but has not been effective in Southern Italy in winter crops in unheated greenhouses. E. meritoria has recently spread in Italy, and may have potential for biological control of whitefly in the greenhouse environment. In the first experiment, female E. pergandiella were exposed to one of three pair-wise combinations of the three species in petri dish arenas. Parasitism was determined by dissection of the hosts. The number of hosts parasitized by E. pergandiella females did not differ with host species. However, significantly greater numbers of eggs were laid in E. meritoria in both treatments in which it was present; these hosts were more likely to be superparasitized. In a second experiment, observations of females in arenas with equal numbers of all three host species indicated that females encountered and parasitized all host species with approximately equal frequency, although the length of time females spent in the oviposition posture differed with host species.  相似文献

Measurements of root-length density (RLD) in a range of 31 apple, kiwifruit, peach, Asian pear and grape orchards were used to derive indices to describe the exploration and exploitation of rooting volumes. Orchards were of various ages and located on a range of soil types, geographic regions, management systems etc. Data were obtained from core samples of volume 1.66×10-4 m3 randomly taken within a standard volume, determined by average planting grids, of 2 m radius centred on tree stems, and 1 m depth. Root systems were described using an exploitation index, E(), and an exploration index, E(0). E() is defined as the proportion of the soil volume which contains roots at RLD greater than or equal to some specified value, . E(0) is defined as the proportion of the soil volume which contains roots at any RLD greater than zero. These indices are dependent on sample size, as are all volumetric or soil-coring data.Estimates of E(0) for each orchard were obtained as the proportions of cores containing any RLD>O and assessed for dependence on species. Peach trees had a significantly higher value of E(0), equal to almost 1.0, compared to the other four species where E(0) was approximately 0.8 (p0.01) or less. There was also some variation with age. E(0) was lower for very young plants which had not fully occupied the sampled soil volume. Exploration indices for woody roots increased with rootstock age but otherwise did not explain large differences in E() between species for given values.For example at =0.05×104 m.m-3, E() was approximately 0.45 for peach and kiwifruit, and 0.05 for apple, Asian pear and grape, whereas at =0.5×104 m.m-3 the corresponding values were 0.1 and almost zero. Negative exponential curves relating E(), scaled by dividing by E(0), to were fitted for each of the 31 orchards. Exponents for these curves, k, were significantly smaller for kiwifruit and peaches than apples, grapes and Asian pears (p0.05), and smaller for apples than grapes and Asian pears (p0.05). A larger k implies a rapid fall-off in E() as increases. Although all five species contained zero and low RLD samples, only kiwifruit and peaches contained higher RLD values and consequently have higher mean RLD. This trend was consistent across all soils, regions, sampling dates, and plant ages.The analyses demonstrate that core sampling can give useful insights into macro-scale root-system distribution, such as the proportion of a soil volume explored and how it is exploited. If positions of core samples are noted during sampling using angular direction, depth and radial distance as spatial coordinates the method can be used to describe root-system structures.  相似文献

Summary The morphology of starch grains isolated from the latex of 2 species of Euphorbia was compared by scanning electron microscopy. In E. terracina they are elongated and greater, in diameter at the midregion than at the tips while in E. tirucalli they are osteoid. Starch grains varied in size in both, species although in E. tirucalli the largest grains, which measured 49 , were approximately twice the length of those in E. terracina (27 ).  相似文献

Summary Voltage-clamp currents and resting membrane potential of squid giant axons have been studied at extracellular pH varying between 4 and 10. The membrane currents, analyzed according to the Hodgkin-Huxley equations, showed that sodium permeability,P Na (E), and potassium conductance,g K (E), curves were shifted toward positive voltages by different amounts and slightly depressed as the external pH was lowered. Under the same conditions, m(E) and n(E) were found to be enhanced and shifted to a larger extent in the same direction. The rate constants m and n were shifted substantially toward positive voltages, but m and n changed hardly at all. The shift of the m(E) curve was analyzed in terms of a fixed surface charge model; it indicates that unspecific negative groups with an approximate pKa of 4.5 are located in the vicinity of sodium active sites with an average charge separation of 8 Å. A similar figure is obtained for the potassium system from the shift of the n(E) curve.  相似文献

A bacteriocinogenic factor of Enterobacter cloacae   总被引:24,自引:0,他引:24  
Summary Enterobacter cloacae strain DF13 produces a bacteriocin which is able to kill other strains of Enterobacter and Klebsiella. This property can be transmitted to Enterobacter cloacae strain O 2 (up to 90% of the acceptor population became bacteriocinogenic), to E. coli K12F- and E. coli K 12 Hfr. Transfer of chromosome material was never observed, suggesting that the production of the bacteriocin is determined by a plasmid. However all attempts to eliminate this plasmid failed. The plasmid F trp cys Col B Col V could be transferred from E. coli into Ent. cloacae DF13 and subsequently it could be eliminated by acridine orange treatment. Ent. cloacae DF13 harbours in addition two independently transferable R-factors, one determining resistance against streptomycin and sulfanilamide and the other resistance against penicillin.Most but not all Ent. cloacae O2 recombinants which have received only the bacteriocinogenic factor upon conjugation with Ent. cloacae DF 13, can transfer this property to Ent. cloacae O2 but not to E. coli. E. coli F- recombinants, which have received only the bacteriocinogenic factor cannot transfer this property. The results suggest that the bacteriocinogenic factor cannot mediate its own transfer, but can be transferred when another transmissible plasmid is present. This may be the R(str sul) factor, the F-factor in E. coli Hfr or a transfer factor () in Ent. cloacae O2.Closed circular DNA molecules were selectively isolated from these strains and investigated by electron microscopy and velocity sedimentation. Ent. cloacae DF13 harbours small closed circular DNA molecules ranging from 0.5 to 3.2 in contour length, 98% of which corresponds to a size class of 0.7±0.1 . Ent. cloacae O2 also harbours closed circular DNA ranging from 0.8 to 3.0 in contour length, with major size classes of 0.9 and 1.4 respectively. Circular DNA of a contour length of 3.0±0.2 (S20,w=26 S) corresponding to a molecular weight of 6.0×106 daltons was transferred to E. coli and Ent. cloacae O 2 concomitantly with the ability to produce the bacteriocin. A significant difference was observed in the number of copies of the plasmid between Ent. cloacae and E. coli.  相似文献

Summary Four hybrids were obtained between three Australian Elymus taxa and three cereal grains: wheat, rye, and barley. Mean meiotic metaphase-I configurations were 41.14 I, 0.42 rod II, 0.003 ring II, and 0.01 III for E. scabrus var plurinervis x Triticum aestivum (1 hybrid plant), 22.27 I, 2.63 rod II, 0.06 ring II, and 0.12 III for E. scabrus var scabrus x Secale cereale (4 hybrid plants), and 26.65 I, 0.66 rod II, 0.00 ring II, and 0.01 III for E. scabrus var plurinervis x Hordeum vulgare (13 hybrid plants). The I genome of barley also paired very little in a BIII hybrid of apomictic E. rectisetus x H. vulgare (2 hybrid plants). Megasporogenesis in this BIII hybrid was at least facultatively apomeiotic, with the same sort of nuclear elongation, apomeiotic division, and dyad formation seen previously in E. rectisetus itself. All four hybrid combinations were sterile. While spike morphology in the E. scabrus x T. aestivum and E. scabrus x H. vulgare hybrids were intermediate to their parents, E. scabrus x S. cereale and E. rectisetus x H. vulgare looked like their maternal parents.  相似文献

Organophosphate insecticide (parathion/diazinon) resistance in housefly (Musca domestica L.) is associated with the change in carboxylesterase activity. The product of MdE7 gene is probably playing a role in detoxification of xenobiotic esters. In our research, we have isolated, cloned and sequenced the MdE7 gene from five different Turkish housefly strains. High doses of malathion (600 g/fly) were applied in a laboratory environment for one year to Ceyhan1, Ceyhan2, Adana, and Ankara strains while no insecticide treatment was performed in the laboratory to Kirazli strain. Trp251 Ser substitution was found in the product of MdE7 gene in all malathion-resistant and Kirazli stocks. In addition, we checked the malathion carboxylesterase (MCE), percent remaining activities in acetylcholinesterase (AChE), glutathion-S-transferase (GST), and general esterase activities in all five strains used in this study. In comparing with universal standard sensitive control WHO, a high level of MCE and GST activities were observed while lower level of general esterase activities was detected in the tested strains. In addition, a higher percent remaining activities in AChE than WHO susceptible strain were observed in all malathion-resistant strains.  相似文献
