The nonequilibrium electrical potentials across an artificial membrane bathed by solutions of a single salt have been measured and calculated using the Goldman-Hodgkin-Katz equation and the irreversible thermodynamic equation. The latter equation predicts the observed potential differences over a 2500-fold concentration range, while application of a modified Goldman-Hodgkin-Katz equation leads to difficulties.  相似文献

An analysis is made of the effect of solution-membrane partition of ions on the electrostatic potential and ion concentration profiles in fixed charge membranes. It is shown that the inclusion of partition effects gives rise to large solution-membrane “Donnan” potentials even when the concentration of fixed charges is of the same order as the concentration of the external solution. This effect renders the system and the simplified analysis of the double fixed charge membrane (FCM) previously given more applicable to biological membranes. An analysis is also given of the voltage dependence of the fluxes of individual ion species in the double FCM when it separates different ionic solutions and an expression is deduced for the membrane resting potential. Although the latter is similar in form to the Goldman-Hodgkin-Katz (GHK) equation the corresponding value of the permeability ratio PC1/PK is under certain specified conditions both concentration and potential dependent.  相似文献

The level of nerve membrane cholesterol was altered by in vitro incubation of rat brain synaptosomal plasma membrane with liposomes having varying cholesterol contents. The normal plasma membrane cholesterol/phospholipid ratio of 0.3-0.4 (mol/mol) could be decreased by about one-half or increased more than 100%. Fluorescence polarization measurements were made using the probe 1,6-diphenyl-1,3,5-hexatriene. At temperatures below 35 percent C, lowering membrane cholesterol led to increased apparent microviscosity, while raising cholesterol content produced little change. However, at 45 percent C a continuous direct relationship existed between experimental membrane cholesterol/phospholipid ratio (ranging from 0.18 to 0.73) and apparent microviscosity. Under standard liposome-synaptosomal plasma membrane exchange conditions, 80% of the initial specific [(3)H]saxitoxin binding activity to the voltage-dependent sodium channel and at least 95% of the (Mg2+,K+)-p-nitrophenylphosphatase activity were preserved. Our results indicate that neither the characteristics of toxin binding nor the kinetics of this enzyme activity is dependent upon membrane cholesterol content.  相似文献

(1) Fluctuations of the membrane potential states are essential for the brain functions from the response of individual neurons to the cognitive function of the brain. It has been reported in slice preparations that the action potential duration is dependent on the membrane potential states. (2) In order to examine whether dependence of action potential duration on the membrane potential could happen in isolated individual neurons that have no network connections, we studied the membrane potential dependence of the action potential duration by artificially setting the membrane potentials to different states in individual cultured rat hippocampal neurons using patch-clamp technique. (3) We showed that the action potential of individual neurons generated from depolarized membrane potentials had broader durations than those generated from hyperpolarized membrane potentials. (4) Furthermore, the membrane potential dependence of the action potential duration was significantly reduced in the presence of voltage-gated K+ channel blockers, TEA, and 4-AP, suggesting involvement of both delayed rectifier I K and transient I A current in the membrane potential dependence of the action potential duration. (5) These results indicated that the dependence of action potential duration on the membrane potential states could be an intrinsic property of individual neurons. 目的:考察山楂醇提取液对大鼠离体胃、肠平滑肌的影响.方法:本研究以大鼠的离体胃、肠平滑肌条为模型,考察在正常克氏液条件下加入山楂提取液大鼠胃、肠平滑肌条收缩状况.同时观察乙酰胆碱、阿托品作用下加入山楂提取液大鼠肠平滑肌条收缩状况.结果:山楂醇提取液在5-20mg生药/ml浓度范围内可显著抑制大鼠胃、肠平滑肌条的运动,且具有明显剂量依赖性.山楂醇提取液(20mg生药/ml)可拮抗乙酰胆碱引起的胃肠平滑肌的强烈收缩,山楂醇提取液(20mg生药/ml)可对抗阿托品引起的肠平滑肌的舒张作用.结论:山楂醇提取液对大鼠胃、肠平滑肌条的收缩具有明显的抑制作用.  相似文献

The objective of this study was to observe the effects of dietary supplementation with zinc oxide nanoparticles (ZnO-NPs) on performance, egg quality, tissue Zn content, bone parameters, superoxide dismutase activity (SOD), and egg malondialdehyde (MDA) content in laying hens. A total of 288 laying hens at 64 weeks of age were randomly assigned to 4 treatments, 6 replicates, with 12 birds each. Experimental diets included the based diet (without Zn supplementation), and basal diet supplemented with 40, 80, and 120 mg Zn/kg from ZnO-NPs. Feed intake and egg mass were significantly higher in the 40 and 80 ZnO-NPs groups than the other groups. The birds in the 80 ZnO-NPs group had significantly higher egg product than the 120 ZnO-NPs and control groups. Egg shell thickness and shell strength significantly increased in the 40 and 80 ZnO-NPs groups. Moreover, Haugh unit significantly improved in the groups supplemented with ZnO-NPs, compared to the control group. Bone-breaking strength was significantly greater in the 80 ZnO-NPs group than the 120 ZnO-NPs and control groups. Also, ash weight was significantly greater in the 40 and 80 ZnO-NPs groups than the control group. There were significant differences among the groups in the Zn content in plasma, tibia, liver, pancreas, and egg. Relative to the control group, ZnO-NPs supplementation significantly increased the activities of SOD in the liver, pancreas, and plasma. The MDA content in egg was significantly reduced in the groups supplemented with ZnO-NPs. In conclusion, this study demonstrates that ZnO-NPs as dietary supplementation can improve the performance of laying hens, and levels of 40 to 80 ZnO-NPs are the optimal concentrations.

Abstract

Streptozotocin-induced diabetic rat liver was analyzed for glucocorticoid receptor (GR) content by saturation and Scatchard analysis. The hepatic GR content of streptozotocin-induced diabetic rats was significantly decreased from a control level of 0.17 ± .01 pmol/mg protein to 0.11 ± .01 pmol/mg protein. Insulin replacement therapy to the diabetic rat dramatically increased the hepatic GR content to 0.26 ± 0.02 pmol/mg protein as compared to the diabetic value of 0.11 ± 0.01 pmol/mg protein.

A time course study of GR content in the diabetic rat liver demonstrated that after an initial decrease in hepatic GR content at 14 days, the 25-day diabetic receptor level elevated back to control levels. A significant increase in GR content over controls was observed in the 110-day diabetic rats. These results suggest that insulin has a role in the regulation of hepatic GR content.  相似文献

In cardiac myocytes a complex network of membrane tubules - the transverse-axial tubule system (TATS) - controls deep intracellular signaling functions. While the outer surface membrane and associated TATS membrane components appear to be continuous, there are substantial differences in lipid and protein content. In ventricular myocytes (VMs), certain TATS components are highly abundant contributing to rectilinear tubule networks and regular branching 3D architectures. It is thought that peripheral TATS components propagate action potentials from the cell surface to thousands of remote intracellular sarcoendoplasmic reticulum (SER) membrane contact domains, thereby activating intracellular Ca2+ release units (CRUs). In contrast to VMs, the organization and functional role of TATS membranes in atrial myocytes (AMs) is significantly different and much less understood. Taken together, quantitative structural characterization of TATS membrane networks in healthy and diseased myocytes is an essential prerequisite towards better understanding of functional plasticity and pathophysiological reorganization. Here, we present a strategic combination of protocols for direct quantitative analysis of TATS membrane networks in living VMs and AMs. For this, we accompany primary cell isolations of mouse VMs and/or AMs with critical quality control steps and direct membrane staining protocols for fluorescence imaging of TATS membranes. Using an optimized workflow for confocal or superresolution TATS image processing, binarized and skeletonized data are generated for quantitative analysis of the TATS network and its components. Unlike previously published indirect regional aggregate image analysis strategies, our protocols enable direct characterization of specific components and derive complex physiological properties of TATS membrane networks in living myocytes with high throughput and open access software tools. In summary, the combined protocol strategy can be readily applied for quantitative TATS network studies during physiological myocyte adaptation or disease changes, comparison of different cardiac or skeletal muscle cell types, phenotyping of transgenic models, and pharmacological or therapeutic interventions.  相似文献

The present study was conducted to investigate whether the combined treatment with Se and Zn offers more beneficial effects than that provided by either of them alone in reversing Cd-induced oxidative stress in the kidney of rat. For this purpose, 30 adult male Wistar albino rats, equally divided into control and four treated groups, received either 200 ppm Cd (as CdCl2), 200 ppm Cd + 500 ppm Zn (as ZnCl2), 200 ppm Cd + 0.1 ppm Se (as Na2SeO3), or 200 ppm Cd + 500 ppm Zn + 0.1 ppm Se in their drinking water for 35 days. The results showed that Cd treatment decreased significantly the catalase (CAT) and glutathione peroxidase (GSH-Px) activities, whereas the superoxide dismutase (SOD) activity and the renal levels of lipid peroxidation (as malondialdehyde, MDA) were increased compared to control rats. The treatment of Cd-exposed rats with Se alone had no significant effect on the Cd-induced increase in the MDA concentrations but increased significantly the CAT activities and reversed Cd-induced increase in SOD activity. It also partially prevented Cd-induced decrease in GSH-Px activity. The treatment of Cd-exposed animals with Zn alone increased significantly the CAT activity and partially protected against Cd-induced increase in the MDA concentrations, whereas it had no significant effect on the Cd-induced increase in SOD activity and decrease in GSH-Px activity. The combined treatment of Cd-exposed animals with Se and Zn was more effective than that with either of them alone in reversing Cd-induced decrease in CAT and GSH-Px activities and Cd-induced increase in MDA concentrations. Results demonstrated beneficial effects of combined Se and Zn treatment in Cd-induced oxidative stress in kidney and suggest that Se and Zn can have a synergistic role against Cd toxicity. I. Zinc deficiency impairs the hepatic lipid metabolism. Previous studies were focused on the negative effects of zinc deficiency on the hepatic lipid metabolism. A few studies investigated the effects of high zinc levels on the lipid metabolism in hepatocytes. In this study, rat hepatocytes were cultured and treated with different and high concentrations of zinc to investigate the effects of high zinc levels on the lipid synthesis in hepatocytes in vitro. The levels of hepatocytes functional markers, including alkaline phosphatase, lactate dehydrogenase, and albumin, were significantly higher in the zinc treatment groups than in the control group (p?<?0.05, p?<?0.01). The mRNA and protein levels of sterol regulatory element-binding protein 1c (SREBP-1c) were significantly higher in the zinc treatment groups than in the control group (p?<?0.05, p?<?0.01). Furthermore, the mRNA expression levels of acetyl-CoA carboxylase 1 (ACC1) and fatty acid synthase (FAS) were significantly higher in the medium- and high-dose zinc treatment groups than in the control group (p?<?0.01). The mRNA levels of stearoyl-CoA desaturase-1 (SCD-1) were significantly higher in the high-dose group (p?<?0.01). These results indicate that high levels of zinc increase hepatocytes activity and SREBP-1c expression, which upregulate the expression of ACC1, FAS, and SCD-1, thereby improving the lipid metabolism in the hepatocytes.  相似文献

This study was designed to examine the protective potential of zinc on the histoarchitecture distortion induced by nickel in rats. Male Sprauge Dawley (S.D) rats received either nickel alone in the form NiSO4·6H2O at a dose of 800 mg/l in drinking water, zinc alone in the form of ZnSO4·7H2O at a dose of 227 mg/l in drinking water, or nickel plus zinc or drinking water alone for a total duration of eight weeks. The effects of different treatments were studied on rat liver histoarchitecture by using both light and transmission electron microscopes. Normal control and zinc treated animals revealed normal histology of liver, however, nickel treated animals resulted in drastic alterations of normal hepatic histoarchitecture, after 8 weeks of treatment. Administration of zinc to nickel treated rats resulted in marked improvement in the structure of hepatocytes, thus emphasizing the protective potential of zinc in restoring the altered hepatic histoarchitecture close to the histoarchitecture of normal animals.  相似文献

One hundred male rats were randomly divided into four groups (n = 25) and fed a Zn-adequate diet (ZA, 46.39 mg/kg), Zn-deficient diet (ZD, 3.20 mg/kg), Zn-overdose diet (ZO, 234.39 mg/kg), or were pair-fed a Zn-adequate diet (PF) for 5 weeks, respectively. The body weight, femur weight, and activity of alkaline phosphatase (ALP) were reduced in the ZD group but were increased in the ZO group. Zn concentrations in both liver and femur were elevated in the ZO group, whereas femur Zn was decreased in the ZD group. The concentrations of calcium and phosphorus were lower in the ZD than those in other groups. Serum calcium concentration was decreased in the ZD. The relative expression level of ALP was decreased in both ZD and PF, and no significant differences were observed between ZO and ZA. Insulin-like growth factor-I (IGF-I) mRNA level was reduced in the ZD but unchanged in the ZO and PF group. Zn deficiency also decreased ALP mRNA level as compared with that of PF group. Carbonic anhydrase II mRNA level was not affected by Zn. Nevertheless, dietary Zn influenced the growth, bone metabolism, and expression of IGF-I and ALP in male growing rats.  相似文献

Abstract: The normal stability of the cell membrane bilayer depends on its lipid composition being appropriate to the ambient (physiological) temperature, Tp. Membrane lipid composition may be altered by disease such that the bilayer is only stable at a new critical temperature, T, which may differ from Tp. In Alzheimer's disease (AD) temporal cortex, a defect of lipid composition has previously been identified, namely, a decrease in the ratio of plasmalogen to nonplasmalogen ethanolamine glycerophospholipids. Furthermore, for AD temporal cortex neural membranes, T≪ Tp, a finding confirmed in the present study in a larger series than previously, using a new method for obtaining T. This inequality between T and Tp has been proposed as a putative contributory pathogenetic mechanism leading to membrane destabilisation in AD brain. The plasmalogen deficiency could account for the change in T in AD, as shown by experiments where T was measured for artificial lipid mixtures simulating brain membranes with varying plasmalogen/nonplasmalogen ratios. The critical temperature was found to be very sensitive to small alterations in plasmalogen content.  相似文献

To assess the nutritional defects of some trace elements caused by an excess supplementation of calcium, an in vitro study was undertaken on brush border membrane vesicles (BBMV) isolated from the small intestinal mucosa of normal rats. The uptake of 65Zn2 + tended to be saturable with increasing concentration of Zn2 +, which was decreased by adding an excess concentration of IIa cations or of Mn2 +. The degree of inhibition was inversely proportional to the ionic radius of these divalent cations, except for manganese. All of these inhibition processes proceeded without change in the maximum velocity of Zn2 + uptake, indicating the intervention by a common carrier for these cations during the course of mucosal uptake.  相似文献
