Reconstituted proteoliposomes of tonoplast ATPase are formedon solubilization of tonoplast membranes from mung bean (Vignaradiata L.) with deoxycholate (DOC) in the presence of a mixtureof soybean phospholipids (asolectin), after removal of DOC bypassage through a PD-10 column (Pharmacia). This method is idealbecause of its simplicity and rapidity. Selective insertionof sets of tonoplast H+-ATPase polypeptides (68 kDa, 60 kDa,16 kDa and several minor polypeptides) into liposomes usingthis method was confirmed by SDS-PAGE and immuno-blotting withantibodies raised against 68-kDa and 60-kDa polypeptides. Pumping of protons across the membranes of the proteoliposomeswas demonstrated by quinacrine-fluorescence quenching in thepresence of ATP-Mg2+. ATP-Mg2+ was shown to be the preferredsubstrate in both reconstituted and native tonoplast vesicles,and its optimum concentration was 0.75 to 3.0 mM. Quenchingwas completely abolished by a channel-forming ionophore, gramicidinD, and an inhibitor of tonoplast H+-ATPase, KNO3. Antibodiesto 68-kDa and 60-kDa peptides partially inhibited the pumpingof protons. The rate of pumping of protons increased with thenumber of proteoliposomes, the maximal concentration of whichwas equivalent to 250 µg of protein per reaction mixture.The optimum pH for pumping was 6.5 when inside of proteoliposomeswere loaded pH at 7.2. The rate of pumping of protons was reducedwhen proteoliposomes were made using asolectin and cholesterolat 3 : 1 (w/w), as compared with those made with asolectin alone. The ATPase activity in reconstituted proteoliposomes was inhibitedby KNO3, with half-maximal inhibition at approximately 7 mM.The enzyme actively hydrolyzed ATP in preference to GTP, CTP,UTP, and ADP, but it did not hydrolyze pNPP or AMP. Antibodiesagainst the 60-kDa polypeptide strongly inhibited ATPase activityas compared to antibodies against the 68-kDa polypeptide. Highly purified oilseed rape (Brassica napus) root plasma membranefractions were prepared and their polypeptide patterns analysedby two-dimensional gel electrophoresis. Sulphur starvation enhancesthe sulphate uptake capacity of B. napus roots. The relativeabundance of several polypeptides increased significantly andspecifically after sulphur starvation. Several of them (37,38, 60, and 65 kDa), found in sulphur-starved plants, were moreabundant in a phase-partitioned membrane fraction treated withTriton X-100/KBr, indicating that they are intrinsic polypeptides.One polypeptide (47 kDa) was identified in the in vitro translationproducts of the roots mRNAs as specific for S-starved plants.It was also present among the intrinsic polypeptides specificfor —S plants. Plasma membranes were isolated using the aqueous polymer two-phasepartition method from the algae Chara corallina and Chara longifolia,algae which differ in their ability to grow in saline environments.Enrichment of plasma membrane and depletion of tonoplast relativeto the microsomal fraction was monitored using phosphohydrolaseassays and crossreactions to antibodies raised against higherplant transporters. Antibodies to the vacuolar ATPase and pyrophosphatasecross-reacted with epitopes in the microsomal fraction, butshowed little affinity for the plasma membrane fraction. Pyrophosphataseactivity also declined in the plasma membrane fraction relativeto the microsomal fraction. The V-type H+ -ATPase activity,sensitive to nitrate or bafilomycin, was low in both fractions,though the cross-reaction to the antibody was reduced in theplasma membrane fraction. By contrast, the antibody recognitionof a P-type H+-ATPase amino acid sequence from Arabidopsis didnot occur strongly in the anticipated 90–100 kDa range.While there was enhanced recognition of a polypeptide at around140 kDa in the plasma membrane fraction, salt treatment of Charalongifolia resulted in plasma membrane fractions with reducedamounts of this epitope, but no change in vanadate-sensitiveATPase activity, suggesting that it does not represent the onlyP-type ATPase. The in vitro conversion of [3H]tryptophan by a plasma membraneenriched fraction from Arabidopsis thaliana (L.) Heynh. seedlings,grown in liquid culture, revealed indole-3-acetaldoxime (IAOX)as the only detectable reaction product. The pH optimum of thereaction was at pH 8, the Km value for tryptophan 12 µM.The formation of IAOX was stimulated about 10-fold by H2O2 Incubationexperiments with solubilized proteins and membrane vesiclesshowed that the investigated enzyme(s) were bound covalent tothe plasma membrane. Tryptophan oxidizing enzyme (TrpOxE) andperoxidase activity were not only found in the plasma membrane,but also in the culture medium. Specific IAOX forming activitywas 74-fold and 6-fold higher compared to the crude extractand the plasma membrane fraction, respectively. After isoelectricfocusing of solubilized plasma membrane and precipitated mediumproteins, TrpOxE activity co-migrated with two prominent highpI peroxidase bands stained with benzidine-guaiacol. The zonesof the IEF gel with peroxidase and TrpOxE activity were analyzedby SDS PAGE and revealed in all fractions a main protein bandof ca. 55 kDa. TrpOxE activity and peroxidase activity wereboth inhibited by antisera directed against tobacco and horseradishperoxidase. TrpOxE activity and peroxidase activity were determinedduring plant development. TrpOxE activity peaked after 8 and42 days, whereas peroxidase activity was consistently presentduring the whole life cycle. The inhibitory effects of indolederivatives, especially indole-3-glyoxylic acid, on (i) seedlingdevelopment and (ii) on TrpOxE and peroxidase activity werealso compared. Cell fractions enriched in endoplasmic reticulum, tonoplast, plasma membrane, and cell walls were isolated from roots of barley (Hordeum vulgare L. cv CM 72) and the effect of NaCl on polypeptide levels was examined by two-dimensional (2D) polyacrylamide gel electrophoresis. The distribution of membranes on continuous sucrose gradients was not significantly affected by growing seedlings in the presence of NaCl; step gradients were used to isolate comparable membrane fractions from roots of control and salt-grown plants. The membrane and cell wall fractions each had distinctive polypeptide patterns on 2D gels. Silver-stained gels showed that salt stress caused increases or decreases in a number of polypeptides, but no unique polypeptides were induced by salt. The most striking change was an increase in protease resistant polypeptides with isoelectric points of 6.3 and 6.5 and molecular mass of 26 and 27 kilodaltons in the endoplasmic reticulum and tonoplast fractions. Fluorographs of 2D gels of the tonoplast, plasma membrane, and cell wall fractions isolated from roots of intact plants labeled with [35S]methionine in vivo also showed that salt induced changes in the synthesis of a number of polypeptides. There was no obvious candidate for an integral membrane polypeptide that might correspond to a salt-induced sodium-proton anti-porter in the tonoplast membrane.  相似文献

The Permeability of the Guard Cell Plasma Membrane and Tonoplast   总被引:4,自引:0,他引:4  
Uptake experiments and efflux compartmental analysis of planthormones, osmotica and toxins using ‘isolated’ guardcells of Valerianella locusta and guard cell protoplasts (GCP)of Vicia faba were performed in order to study the permeabilityproperties of guard cell plasma membrane and tonoplast. Theplasma membrane of guard cells exhibits a higher permeabilitythan plasma membranes of mesophyll cells for most solutes investigated.The permeability coefficients (Ps calculated for the guard cellplasma membranes are also significantly higher than the Ps valuesfor the guard cell tonoplast. This applies also for protonatedABA. We suppose that the high permeability for ABAH could bepart of the target cell properties. A Collander analysis demonstratesa linear correlation between Ps, values and the ratio Kr/Mr1,5for both plasma membrane (r = 0.87) and for the tonoplast (r=0.93). Because of deviations from the observed correlations,the permeation of some solutes (ABA, GA, IAA through the tonoplast;methylamine through the plasma membrane) seems to be facilitatedby an additional transport mechanism. The Collander analysisof the plasma membrane of GCP shows very similar results tothe analysis of the plasma membrane of ‘isolated’guard cells, indicating that isolation of protoplasts does notalter the permeability of the guard cell plasma membrane. The polypeptide composition of nuclear envelopes prepared fromhypocotyls of mung bean (Vigna radiata) was investigated. Thetissue was homogenized in the presence of Triton X-100 and nucleiwere isolated by differential and discontinuous Percoll gradientcentrifugation. The nuclei were subjected to sonication in 2M KC1 or 50 mM lithium diiodosalicylate and then the nuclearenvelopes were collected by centrifugation. Proteins in theenvelope fraction were analyzed by sodium dodecylsulfate-polyacrylamidegel electrophoresis and blotting techniques. When the envelopefraction was incubated with [-32P]ATP, 10 to 15 polypeptideswere labeled and the intensity of labeling of some of thesepolypeptides was enhanced by the addition of calcium ions. Theresults suggest the presence of a protein-phosphorylation systemin nuclear envelopes. Three polypeptides of 100, 42, and 40kDa stained blue with the cationic carbocyanine dye "Stains-all",and they were labeled with 45Ca2+ on a transfer membrane. Thelectin concanavalin A recognized glycoproteins that migratedas polypeptides of 50, 49, 47, 43, 35 and 32 kDa, respectively.Of these polypeptides the two larger ones were prominent andwere solubilized by treatment of the envelope fraction withKCl at 2 M but not at less than 100 mM. These results suggestthat the mung bean nuclear envelope contains some calcium-bindingproteins and glycoproteins. A cytochrome b6f complex was isolated and purified from Spirulinasp. The complex was solubilized with n-heptyl ß-D-thioglucosideand chromatographed on a DEAE-Toyopearl 650M column. The purifiedcomplex contained a small amount of chlorophyll and carotenoid.At least four polypeptides were present in the complex: cytochromef (29 kDa), cytochrome b6(23 kDa), iron-sulfur protein (ISP,23 kDa), and a 17 kDa polypeptide. Each polypeptide was separatedfrom the complex treated with 2-mercaptoethanol or urea. Theabsorption spectra of cytochrome b6 and cytochrome f were similarto those of Anabaena and spinach as expected. The complex wasactive in supporting ubiquinol-cytochrome c oxidoreductase activity.Fifty percent inhibition of the activity was accomplished by1 µM dibromothymoquinone (DBMIB). The vacuolar membrane, the tonoplast, is a proteinrich membranehitherto only few proteins in it have been identified. As anapproach for the identification of tonoplast proteins by monoclonalantibodies (MABs), purified tonoplast from cress roots (Lepidiumsativum L.) were used for immunization and plasma membranesas a control membrane to test the absence of antigen. The MABTOP 35 identified a glycoprotein of about 35 kDa in purifiedtonoplast of cress roots. Triton X-114 phase separation showedthat it was a hydrophobic integral membrane protein. In immunocytochemistrythe MAB TOP 35 strongly labelled the vacuolar membrane. Theabsence of cell wall or plasma membrane labelling by TOP 35indicates a distinct biosynthetic pathway of this protein tothe vacuolar membrane in plants. Greening of etiolated seedlings of wild and Chl b-less barley(Hordeum vulgare L.) genotypes in the presence of D-threochloramphenicol(CAP) led to macrogranal arrangements accompanying the inhibitionof Chl synthesis and an enhancement of the total protein contentin differentiated etiochloroplasts. In treated mutant plastids,protein/Chl ratio reached up to 100. No light-dependent O2 evolution was detected in CAP-treatedplastids which had deficiency in polypeptides belonging to thephotosystem II (PSII) centres. On the other hand, plastids displayeda high photosystem I (PSI) activity despite the absence of the92 kDa polypeptide linked to the PSI centre. The accumulationof polypeptides ranging from 16 to 20 kDa suggest that theycould originate from primary complexes consisting of few Chlmolecules, but they were sufficient to allow the activity ofthe reaction centres. No accumulation of the 25–27 kDapolypeptides linked to the PSII antenna was detected. The increase in the proportion of trans-3hexadecenoic acid (16:1tr) in phosphatidylglycerol (PG) of etiochloroplasts from bothtypes after CAP treatment could indicate an alteration of theregulation process of 16:1 tr biosynthesis occurring in plastids.The formation of macrograna could optimize the energy transferin altered thylakoid membranes. The accumulation of PG-16:1tr molecules could be related to the formation of active primarycomplexes in thylakoid when Chl synthesis is altered. Tonoplast and plasma membrane vesicles were prepared from chilling-sensitive(CS) and chilling-insensitive (CI) cultured cells of rice (Oryzasativa L.) to examine how they would respond to low temperature.With CS cells, the specific activity of ATPase in tonoplastvesicles was relatively higher than that of plasma membraneATPase. Tonoplast ATPase activity was decreased by low temperaturetreatment, and a slight decrease in plasma membrane ATPase activitywas also observed. The decrease in the specific activity ofthe tonoplast ATPase by low temperature may reflect a decreasein Vmax. However, no change was noted in Km. The break pointof the Arrhenius plots of the tonoplast ATPase was ca. 32?C,this value being ca. 9?C higher than that of the plasma membraneATPase. With CI cells, the specific activity of tonoplast ATPasewas somewhat less than that of the plasma membrane ATPase. TonoplastATPase activity was decreased by low temperature at 5?C, whereasan increase in plasma membrane ATPase activity was observed.The break point of the tonoplast ATPase activity was ca. 22?C,which was 3?C higher than that of the plasma membrane ATPase.Using ATPase solubilized from the plasma membrane or tonoplast,the Arrhenius plots of log ATPase activity against the reciprocalof absolute temperature gave a straight line fit from 5?C to45?C with no obvious break point. The break point appeared onadding a phospholipid mixture (asolectin) to a reaction mixturecontaining solubilized enzyme. The slope of the curve of theArrhenius plot was very different between the CS and CI cells.The plasma membrane and tonoplast ATPases from the CS cellshad a higher Ea above 20?C, whereas that from the CI cells hada lower one. These findings indicate that the tonoplast ATPase in a riceplant is more sensitive to low temperature than the plasma membraneATPase, with this response possibly being due to interactionsbetween the proteins and phospholipids. Peripheral and integral subunits of the tonoplast H+-ATPase from oat roots   总被引:10,自引:0,他引:10  
The subunit organization of the tonoplast H+-pumping ATPase from oat roots (Avena sativa L. var. Lang) was investigated. Tonoplast vesicles were treated with low ionic strength solutions (0.1 mM 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid buffer or 0.1 mM Na EDTA), carbonate, or a chaotropic reagent (KI), and then centrifuged to give a soluble fraction and a pellet. Treatments with low ionic strength solutions or KI resulted in 70-80% reduction in the membrane-associated ATPase activity, but did not affect the K+-stimulated pyrophosphatase activity. Polypeptides of 72, 60, and 41 kDa were solubilized from tonoplast vesicles by these wash treatments. These polypeptides reacted with polyclonal antibodies against the holoenzyme of tonoplast ATPase (anti-ATPase) and copurified with the tonoplast ATPase activity during gel filtration chromatography (Sepharose CL-6B). Mono-specific antibody against the 72- or 60-kDa polypeptide reacted with the solubilized 72- or 60-kDa polypeptide, respectively. However, the N,N-[14C]dicyclohexylcarbodiimide-binding 16-kDa polypeptide and a 13-kDa polypeptide that also reacted with anti-ATPase and copurified with the tonoplast ATPase activity during gel filtration remained in the pellets after the wash treatments. We conclude that the 72- and 60-kDa polypeptides appear to be peripheral subunits of the tonoplast ATPase and that the 16-kDa polypeptide is probably embedded in the membrane bilayer. Additional subunits of the ATPase complex may include a 41-kDa (peripheral) and a 13-kDa (integral) polypeptide. Based on these results, a working model of the tonoplast ATPase analogous to the F1F0-ATPase is proposed.  相似文献

Vacuoles were isolated from primary leaves of barley (Hordeum vulgare L.) by mechanical breakage of protoplasts, and their polypeptide composition analyzed by two-dimensional gel electrophoresis. Vacuoplasts which consist of the vacuole, a portion of the plasmalemma and of the cytoplasma were prepared from protoplasts by ultracentrifugation. By comparing the vacuolar polypeptide pattern with polypeptide patterns of isolated chloroplasts and of vacuoplasts, vacuolar polypeptides could clearly be distinguished from polypeptides derived from cross-contaminating cell compartments. At least 14 polypeptides of apparent molecular mass between 12 and 76 kilodaltons and an isoelectric point between 4.5 and 7.6 could be attributed to the tonoplast fraction of the vacuole, and 35 polypeptides to the soluble fraction of the vacuole. Several lectins with different specificity were employed to characterize the degree and nature of glycosylation of vacuolar polypeptides. Concanavalin A bound to a large number of polypeptides. Cellular membrane fractions, including endoplasmic reticum (ER),Golgi-enriched membrane, plasma membrane and tonoplasts, wereisolated from Vigna radiata seedlings. Each of these membranefractions was associated with specific ATPases which were highlydependent on Mg2+. ATPases of ER, Golgi-enriched membrane andplasma membrane were sensitive to vanadate but the tonoplastATPase was not. ATPases were mostly dependent on Cl1, but aslight stimulation by K+ was observed in the case of ATPasesof Golgi-enriched membrane and plasma membrane. KNO3 inhibitedtonoplast ATPase but stimulated the other ATPases. ER ATPasecan be distinguished from other ATPases by the following characteristics:specific inhibition by KNO2 and Triton X-100, stimulation bylow concentrations of diethylstilbestrol and 4,4'-diisothiocyanostilbene-2,2'-disulfonicacid, and high sensitivity to heat. The ATPases showed typicalMichaelis-Menten kinetics and had Km values of 0.5 to 0.6 ITIMMg2+-ATP for ER, Golgienriched-membrane and tonoplast ATPases,and 2.27 msi Mg2+-ATP for plasma membrane ATPase. ATPases ofGolgi-enriched membranes and plasma membranes had similar properties,but they were still distinguishable by the differences in theirKm values and their responses to Triton X-100. Topoisomerase II was partially purified from Daucus carota cellsby a procedure including ammonium sulphate fractionation, ion-exchange,and affinity chromatography steps. The type II enzyme, identifiedfor its ability to unknot knotted P4 DNA and decatenate Trypanosomacruzi kDNA, requires ATP and Mg2+ for activity. The unknottingactivity was sensitive to an inhibitor of the mammalian typeII enzyme, the drug VP16 (IC50 32 mmol m–3), whereas inhibitorsof DNA gyrase showed a limited effect on activity. The SDS-PAGEanalysis of the dsDNA cellulose fraction revealed the presenceof four polypeptides of apparent molecular masses of 72, 71,34, and 33 kDa among which only a polypeptide of about 70 kDacrossreacted with antibodies against yeast topoisomerase II.Immunoprecipitation experiments with monoclonal antibodies tothe and ß isoforms of the human enzyme confirmedthe recognition of a polypeptide of 70 kDa. The sedimentationcoefficient (S) of the topoisomerase II in the phosphocellulosefraction, calculated by analytical glycerol gradient, was 6.1corresponding to a molecular mass of about 123 kDa. Resultssuggest the presence in carrot of a protein of molecular massof 70 kDa having the typical properties of an eukaryotic topoisomeraseII and carrying epitopes recognized by MoAbs to both human and ß enzymes. Nitrate regulation of protein synthesis and RNA translation in maize (Zea mays L. var B73) roots was examined, using in vivo labeling with [35S]methionine and in vitro translation. Nitrate enhanced the synthesis of a 31 kilodalton membrane polypeptide which was localized in a fraction enriched in tonoplast and/or endoplasmic reticulum membrane vesicles. The nitrate-enhanced synthesis was correlated with an acceleration of net nitrate uptake by seedlings during initial exposure to nitrate. Nitrate did not consistently enhance protein synthesis in other membrane fractions. Synthesis of up to four soluble polypeptides (21, 40, 90, and 168 kilodaltons) was also enhanced by nitrate. The most consistent enhancement was that of the 40 kilodalton polypeptide. No consistent nitrate-induced changes were noted in the organellar fraction (14,000g pellet of root homogenates). When roots were treated with nitrate, the amount of [35S]methionine increased in six in vitro translation products (21, 24, 41, 56, 66, and 90 kilodaltons). Nitrate treatment did not enhance accumulation of label in translation products with a molecular weight of 31,000 (corresponding to the identified nitrate-inducible membrane polypeptide). Incubation of in vitro translation products with root membranes caused changes in the SDS-PAGE profiles in the vicinity of 31 kilodaltons. The results suggest that the nitrate-inducible, 31 kilodalton polypeptide from a fraction enriched in tonoplast and/or endoplasmic reticulum may be involved in regulating nitrate accumulation by maize roots.  相似文献

Previous studies suggested that certain protein(s) other thancarbonic anhydrase might play an important role in the facilitatedtransport of dissolved inorganic carbon (DIC) from the mediumto the site of CO2 fixation by ribulose-1,5-bisphosphate carboxylase/oxygenasein the unicellular green alga Chlorella regularis adapted tolow-CO2 (ordinary air) conditions [Shiraiwa et al. (1991) Jpn.J. Phycol. 39: 355; Satoh and Shiraiwa (1992) Research in Photosynthesis,Vol. III, p. 779]. The proteins that might be involved in thisfacilitated transport of DIC were investigated by pulse-labelingof induced proteins with 35S-sulfate during adaptation of cellsgrown under high-CO2 conditions to low CO2. Analysis by SDS-PAGErevealed that synthesis of two polypeptides, with molecularmasses of 98 and 24 kDa, respectively, was induced under low-CO2conditions. The 24-kDa polypeptide was induced at pH 5.5 butnot at pH 8.0, whereas the 98-kDa polypeptide was induced atboth pH 5.5 and pH 8.0. Acid Invertase of Melon Fruits: Immunochemical Detection of Acid Invertases   总被引:2,自引:0,他引:2  
Soluble (S) and cell wall-bound (CW) forms of acid invertases(AIs) were partially purified from mesocarp tissue of melon(Cucumis melo L. cv. AMS) fruits and some properties of theenzymes were examined. The optimum pH values for activity were5.5 and 4.5 for S- and CW-AI, respectively. Km values of S-and CW-AIs for sucrose were 4.2 and 1.7 mM, and those for raffinosewere 20.0 and 10.5 mM, respectively. The band of a polypeptideof about 70 kDa was detected with antibodies against AI of tomatofruits (CW form) and of mung bean seedlings (S form) by immunoblotanalysis. The 70-kDa polypeptide was isolated from each fractionand further analyzed by limited proteolytic digestion with Staphylococcusaureus V8 protease. The digested polypeptides generated almostidentical profiles. The localization of CW-AI was also studiedand it was found to be an extracellular enzyme. The activitiesof S- and CW-AIs per gram fresh weight increased and reacheda maximum on day 10 after anthesis, and then they decreasedrapidly during maturation of fruits. The relative levels ofS- and CW-AI polypeptides were correlated with the levels ofactivities of S- and CW-AIs. These results show that the decreasein activity of AI during maturation of melon fruits is due toa decline in levels of AI polypeptides in the fruits. The potential differences across the tonoplast and plasmalemmamembranes have been measured in the single cells of Nitellatranslucens, the cells being immersed in an artificial pondwater (composition: NaCl 1.0 mM., KC1 0.1 mM., CaCl2, 0.1 mM.).The potential of the cytoplasm is –138 m V with respectto the bathing medium and –18 mV with respect to the vacuole.The concentrations of Na, K, and Cl have been measured in thetwo cell fractions. The concentrations in the flowing cytoplasmare: Na 14 mM., K 119 mM., and Cl 65 mM.; the vacuolar concentrationsare: Na 65 mM., K 75 mM.,and Cl 160 mM. The observed potential differences across the two membranesare compared with the Nernst potentials for all three ions.This analysis shows that all three ions are actively transportedat the plasmalemma: Na is pumped outwards while K and Cl arepumped inwards. At the tonoplast Na is pumped into the vacuolewhile K and Cl are close to electrochemical equilibrium. The inhibitor, ouabain, has no effect on the cell resting potential.  相似文献

Tonoplast and plasma membrane vesicles were prepared from rice(Oryza sativa L. var. Yuukara) culture cells with step sucrosegradient (30% and 42.9%, w/v) and/or step dextran T-70 gradient(1% and 8%, w/w) to determine the inhibition of tonoplast andplasma membrane AT-Pases by local anesthetics. The degree towhich the anesthetics inhibited these ATPases was of the followingorder: dibucaine>lidocainetetracaine>procaineGABA. Dibucaineranging in concentration from 0.2 nui to 2 mM inhibited tonoplastATPase activity more than plasma membrane ATPase, the half inhibitionsbeing 0.8 and 1.1 mM, respectively. The Km values of tonoplastand plasma membrane ATPases were not affected by dibucaine,but various values were noted for Vmax. Dibucaine inhibitedtonoplast and plasma membrane ATPases solubilized from 0.1%DOC pellet by n-octylglucoside and zwittergent 3–14, respectively.The addition of a phospholipid mixture (asolectin) to solubilizedboth ATPases had no effect on the inhibition by dibucaine.
