Summary Endogenous SS14- as well as SS28-like immuno-reactive materials were detected in both male and female rats by radioimmunoassay and by immunocytochemistry on ultrathin frozen sections. The content of somatostatin-like immunoreactivity was 0.39±0.08 pg per mg adenohypophysis. Immunoreactivity was localized by immunocyto-chemistry in three pituitary cell types: somatotrophs, lactotrophs and thyrotrophs, but not in corticotrophs and gonadotrophs. In these three pituitary cell types the SS28- and the SS14-like immunoreactive materials were localized in the cytoplasm and in the nucleus. In the cytoplasm the immunoreactivity was seen in the cytoplasmic matrix and in the secretory granules. In the nucleus it was present mainly in the euchromatin close to the heterochromatin. In somatotrophs and lactotrophs, SS14- and SS28-like immunoreactive materials have been detected at the plasma membrane level. These results suggest that (1) endogenous SS14 and SS28 are present in adenohypophysis in somatotrophs, lactotrophs and thyrotrophs, and (2) the two peptides act on both the cytoplasmic components and the nucleus.  相似文献

Behavioral effects of somatostatin-14, and some of its fragments [somatostatin(3–8), somatostatin(9–14), somatostatin(7–10)] after intracerebroventricular (ICV) administration have been investigated in male rats. In a passive avoidance learning test, somatostatin-14 (0.6 nM) given immediately after the learning session increased the avoidance latency at 24 hr after the injection, when compared to a somatostatin(3–8) (0.6 nM)-treated group. However, compared to a saline-treated group, the peptides did not significantly influence the avoidance latency. Somatostatin-14 administered in higher dose (6.0 nM) decreased the avoidance latency compared to the saline-treated group, while its fragments did not influence it. In an open field behavioral test, immediately after the 24-hr passive avoidance test, 6 nM of somatostatin-14 decreased the rearing activity, while the fragments did not influence this behavior. Somatostatin-14 produced barrel rotation in a dose-related manner, but after the injection of a high dose of the peptide (12 nM) all of the animals died in cardiorespiratory failure (apnea, pulmonary oedema). Summary The physiological effects of the pancreatic peptides somatostatin-14 and somatostatin-25 on lipid metabolism in rainbow trout were evaluated by in vitro culture of liver and adipose tissue. The culture medium was subsequently analyzed for glycerol and fatty acid content and triacylglycerol lipase activity was measured within the tissues. Both somatostatin-14 and somatostatin-25 stimulated hepatic fatty acid and glycerol release within 3 h after treatment. Liver triacylglycerol lipase activity was elevated following treatment with somatostatin-14 (76% above control) or somatostatin-25 (94% above control). Somatostatin-14 and somatostatin-25 also significantly stimulated the release of fatty acid and glycerol from adipose tissue. Triacylglycerol lipase activity in adipose tissue also was enhanced by both somatostatins. 目的:Lin28是一种高度保守的RNA结合蛋白,对生物体的生命活动具有重要的调节作用.为了研究人Lin28A与Lin28B基因的功能,我们构建了分别过表达人Lin28A或Lin28B基因的两种转基因小鼠,表型分析发现小鼠体重变化,为研究Lin28A和Lin28B基因的生物学功能提供模型动物.方法:分别将人类Lin28A与Lin28B cDNA插入PCAG启动子下游,构建Lin28A与Lin28B转基因表达载体,通过显微注射方法,分别建立Lin28A转基因小鼠与Lin28B转基因小鼠.PCR鉴定转基因小鼠的基因型,筛选出人类Lin28A与Lin28B表达的小鼠,统计两种转基因小鼠体重变化.结果:①经PCR鉴定与公司测序证明Lin28A和Lin28B两种载体构建成功.②PCR鉴定小鼠基因型,筛选出可以特异性表达人Lin28A或Lin28B的转基因小鼠,并比较转基因小鼠与同窝野生型小鼠体重,发现两种转基因小鼠体重均大于同窝野生型小鼠,说明在小鼠体内过表达Lin28A或Lin28B均能引起小鼠体重增加.结论:人Lin28A与Lin28B在转基因小鼠体内能正常表达,并且能发挥生物学功能.又因为Lin28A和Lin28B在小鼠体内过表达会引起小鼠体重增加,我们推测其可能通过影响小鼠糖代谢过程引起小鼠体重改变.构建的Lin28A和Lin28B的转基因小鼠为进一步研究Lin28A和Lin28B在人新陈代谢、生长和发育过程中的作用提供了动物模型.  相似文献

In order to investigate the action of somatostatin-28 (SS-28) on the metabolic homeostasis of insulin-dependent diabetics, we compared its effects to those of somatostatin-14 (SS-14) in terms of insulin sparing, changes in dextrose demands, glucose fluctuations and behavior of growth hormone and glucagon secretion. Eight insulin-dependent subjects were connected to Artificial Endocrine Pancreas (Biostator) for 84 hours during which they received intravenous infusions of either SS-14, SS-28 or isotonic saline in a randomized order, after a steady state of metabolism had been achieved. Five of the patients received SS-28 100 micrograms/h and SS-14 250 micrograms/h for 10 hours and three of them SS-28, 50 micrograms/h and SS-14 250 micrograms/h for 12 hours. Identical doses of both peptides were administered as bolus infusions prior to the continuous ones. Under SS-28 100 micrograms/h and SS-14 250 micrograms/h patients required 13.5 +/- 2.3 and 14.5 +/- 1.9 U of insulin respectively vs 40 +/- 5.6 U under isotonic saline infusion (mean +/- SEM, P less than 0.005 and P less than 0.01). At the same period the apparatus delivered 15 times more dextrose under SS-28 and 20 times more under SS-14. The magnitude of glucose fluctuations diminished from 64.6 +/- 2.47 mg% without to 41.4 +/- 2 mg% under SS-14 (P less than 0.01) and 46 +/- 3.8 mg% under SS-28 (P less than 0.02). The conversion of synthetic somatostatin-28 (S-28) to somatostatin- 14 (S-14, SRIF) by subcellular fractions of rat hypothalamus has been investigated. The conversion products were identified by two techniques: (1) two separate RIAs using antibodies directed toward the central (RIA R149) or the N- terminal (RIA S39) region of the S-14 molecule, (2) gel chromatography of the reaction mixture followed by analysis of the column fractions by RIA R149. Maximal S-28 to S-14 converting activity was observed with the particulate fraction of the lysed synaptosomal pellet sedimenting at the density interface 8–16% Ficoll in 0.32 M sucrose in a discontinuous sucrose/ Ficoll gradient. Concomitant with conversion, degradation of total somatostatin-like immunoreactivity (SLI) was also observed with this fraction (t12 ~ 24 min). Relatively little converting activity was found in the remaining subcellular fractions. These data suggest that hypothalamic synaptosomes contain membrane bound enzymes which are able to catalyze the conversion of S-28 to S-14. Tissue specific differences in this converting activity may account for the reported variability in the S-28:S-14 ratios in different tissues.  相似文献

We have isolated form extracts of ovine hypothalami two molecules characterized as somatostatin-28 and somatostatin-4-28 (referred to as somatostatin-25). They were reproduced by solid hase synthesis. In equimolar ratio and depending upon the experimental conditions, synthetic somatostatin-28 ans somatostatin-25 are 3-14 times more potent than somatostatin-14 to inhibit the basal in vitro secretion of growth hormone or as stimulated by prostaglandin (PGE2). In early studies in vivo, somatostatin-28 and somatostatin-25 are also more potent than somatostatin-14 in inhibiting the secretion of growth hormone acutely stimulated in the rat by injection of morphine; somatostatin-28 is also longer-acting than somatostatin-14. These results suggest that somatostatin-14, as originally isolated, is a biologically active fragment of a larger molecule of greater specific activity; it should be considered as another form of somatostatin with high biological activity present in some tissues and likely secreted y the tissues along with somatostatin-14 and possibly other somatostatin-peptides of diverse sizes.  相似文献

The proteasome activation properties of recombinant REG gamma molecules depend on purification procedures. Prior to ammonium sulfate precipitation recombinant REG gamma activates the trypsin-like catalytic subunit of the proteasome; afterwards it activates all three catalytic subunits. The expanded activation specificity is accompanied by reduced stability of the REG gamma heptamer providing support for the idea that a "tight" REG gamma heptamer suppresses the proteasome's chymotrypsin-like and postglutamyl-preferring active sites. In an attempt to determine whether REG gamma synthesized in mammalian cells also exhibits restricted activation properties, extracts were prepared from several mammalian organs and cell lines. Surprisingly, endogenous REG gamma was found to be largely monomeric. In an alternate approach, COS7 cells were cotransfected with plasmids expressing FLAG-REG gamma and REG gamma. The expressed FLAG-REG gamma molecules were shown to form oligomers with untagged REG gamma subunits, and the mixed oligomers preferentially activated the proteasome's trypsin-like subunit. Thus, REG gamma molecules synthesized in mammalian cells also exhibit restricted activation properties.  相似文献

Somatostatin-14 (S14) and its precursor, somatostatin-28 (S28), are widely distributed throughout the rat brain, suggesting that they could act as neurotransmitter or neuromodulator in the central nervous system. The present study was undertaken to study the localization of S14 and S28 receptors in the rat brain determined by "in vitro" radioautography. The study performed on slide mounted frozen brain section with iodinated S14 and S28 analogs revealed an identical distribution of binding sites for the two forms of somatostatin. A good correlation could be observed between receptor distribution and immunohistologically localized neuropeptides except for striatum and hypothalamus. However, receptors were not detectable in the hypothalamus and were found in low concentration in the caudate-putamen nucleus, two regions containing high amounts of S28 and S14, suggesting a high occupancy of receptors in these areas by endogenous peptides or an inverse correlation between receptor and peptide concentrations.  相似文献

The molecular forms of somatostatin contained in the rat striatum were separated by size-exclusion HPLC. Three major peaks of somatostatin-like immunoreactivity (SLI) were resolved. Two peaks cochromatographed with synthetic somatostatin-14 (SS-14) and somatostatin-28 (SS-28), respectively. One peak exhibited a higher molecular weight (about 10,000) and may contain a proform of somatostatin. Local injection of the neurotoxin kainic acid (1 microgram) into the left striatum resulted in a persistent decrease (65-85%) of all three forms of somatostatin. In the contralateral--not injected--striatum a decrease of SLI was also observed which was maximal (45%) after 2 days and was largely abolished after 7 days. This decrease of SLI in the contralateral striatum, however, was due mainly to a decrease of SS-14 and SS-28 but not of the putative proform. Our data suggest that kainic acid causes a destruction of somatostatin-containing perikarya in the injected striatum, whereas in the contralateral striatum increased release with subsequent inactivation of SS-14 and SS-28 takes place. The putative somatostatin proform may serve as neurochemical marker for somatostatin-containing perikarya in the striatum.  相似文献

Simultaneous and direct recording of temperature from the body, hypothalamus, and cortex in animals exposed to acute thermal challenges lack evidence. This study was conducted to assess the usual concept, that brain temperature is rather stable when animals are exposed to different ambient temperatures. In this study, we report the characteristic changes in the body, hypothalamic, and cortical temperature, when the rats were acutely exposed to cold (6 °C) and hot (36 °C) ambient temperature. The results of our study show that the body temperature is robustly regulated while hypothalamic and cortical temperatures vary on challenges to ambient cold (6 °C) and warm (36 °C) exposure in awake rats. The onset of response was observed quickest in the cortex, indicating that the cortical thermal sensing may relay intracranial thermal input to the hypothalamus for the regulation of body temperature within narrow limits. The present findings contradict earlier evidence, which stated that the brain does not participate in thermal sensing.  相似文献

Summary Immunoreactivity of regulatory peptides has been demonstrated in the fetal lung of Macaca mulatta by the peroxidase anti-peroxidase method. Serotonin-immunoreactive neuroepithelial bodies are distributed in the airways from the bronchi to the alveolar ducts. Many neuroepithelial bodies also show bombesin-like immunoreactivity; a very few are immunoreactive to somatostatin antiserum. Four populations of neuroepithelial bodies were identified which contain immunoreactivity for 1) serotonin alone, 2) serotonin and bombesin, 3) serotonin and somatostatin, and 4) serotonin, bombesin, and somatostatin. Since bombesin and somatostatin have been demonstrated to have opposite effects on the release of other peptide hormones, it seems likely that the presence of these same peptides in neuroepithelial bodies may have a similar regulatory role in the lung.  相似文献

We have examined the effects of centrally applied neuropeptides on ventilation (respiratory rate, tidal volume, and minute ventilation) in urethane-anesthetized rats. TRH caused an increase in respiratory rate, a decrease in tidal volume, but an increase in net minute ventilation. One TRH metabolite, acid TRH, caused similar changes, but no effect was observed from the other TRH metabolite, cHis-Pro. Both bombesin and calcitonin caused increases in minute ventilation due to increases in respiratory rate and tidal volume. Additionally, bombesin induced periodic sighing respirations at rates up to 15/minute which was observed with no other neuropeptide. Substance P, somatostatin, and neurotensin had no effect upon ventilation variables.  相似文献

Gel-filtration chromatography of an acid-extract of a phaeochromocytoma, under dissociating conditions, revealed 4 peaks of immunoreactive somatostatin (IRS) of approx. 8-10 kilodaltons (K), 6K, 3.5K and 1.6K as detected by an antiserum (R9) directed against the central region of tetradecapeptide somatostatin (S14). The 3.5K and 1.6K forms of IRS co-eluted with synthetic cyclic S28 and S14 respectively on reversed phase HPLC. Using another radioimmunoassay for the 1-14 sequence of S28 (N-peptide) a peak of immunoreactive N-peptide (IRN) with a molecular weight of approx. 4500 was observed. The antiserum (N3) used in the N-peptide assay was raised against N-Tyr N-peptide and cross-reacts less than 5% with synthetic S28. Two peaks were further characterised by partial tryptic digestion and gel-filtration chromatography. The 3.5K IRS peak was partially converted to a 1.6K IRS form together with an approximately equimolar amount of IRN with apparent molecular weight of 2500. This 2.5K IRN co-eluted both with N-Tyr N-peptide and with the IRN generated by tryptic digestion of synthetic cyclic S28. No IRN peak of this size was observed in the original extract. Tryptic digestion of the 6K IRS peak generated 3.5K and 1.6K IRS and 2.5K IRN. These results suggested that (1) this human phaeochromocytoma contains IRS very similar to the known structure of ovine and porcine S28 and S14. (2) The 6K IRS is composed of an unknown peptide sequence attached via trypsin-susceptible bond to the N-terminus of S28. (3) In this tumour S14 is being generated directly from 6K IRS and not via S28.  相似文献

In order to investigate changes in levels of monoamines and their related substances together with those of other neurotransmitters (acetylcholine and GABA), choline and substances related to energy metabolism (ATP, lactate and glucose) accompanying incomplete cerebral ischemia, a bilateral common carotid artery occlusion model of spontaneously hypertensive rats (SHR) was utilized. Animals were subjected to 1 or 2 h ischemia. Then the concentrations of substances were measured in the cerebral cortex, hippocampus and striatum and compared with control values. Due to the incomplete ischemia, ATP showed a moderate decrease, while lactate and choline increased remarkably, and GABA underwent a moderate increase. With regard to monoamines, both noradrenaline and serotonin levels were reduced in the cerebral cortex and hippocampus, whereas dopamine levels increased in the hippocampus. All monoamine metabolites, i.e. metabolites by monoamine oxidase (MAO), metabolites by catechol-O-methyltransferase (COMT), and metabolites by both MAO and COMT, underwent increases. The 3-methoxytyramine level in particular showed marked increases. Furthermore levels of precursor amino acids as well as 5-hydroxytryptophan rose. Acetylcholine decreased moderately only in the cerebral cortex. Among these changes, sustained increases in all the monoamine metabolites were characteristic in the incompletely ischemic brain, suggesting that both COMT and MAO retain their activities in the incompletely ischemic brain.  相似文献

Sixteen peptides were injected intracerebroventricularly to test their effects on rectal temperature of rabbits in a thermoneutral environment. In initial tests 5 μg α-MSH, ACTH(1–24), oxytocin, vasopressin and glucagon altered body temperature while ACTH(1–10), cholecystokinin, contraceptive tetrapeptide, gastrin, insulin, interferon, leupeptin, LHRH, panhibin (somatostatin), and proctolin did not. Bombesin also altered body temperature but in no consistent direction. In further tests on the effective peptides 1.25–5.0 μg α-MSH and ACTH(1–24) produced dose-related decreases in rectal temperature as great as 1.0°C. The same doses of oxytocin and glucagon produced small, prolonged hyperthermias which did not exceed 0.4°C. Vasopressin caused rapid development of small increases in rectal temperature; temperature returned to normal in 2–3 hr. The results suggest that five of the peptides tested may have roles in central mediation of normal body temperature, hypothermia, hyperthermia and fever.  相似文献

The biosynthesis of pro-opiomelanocortin (POMC) and related peptides by the intermediate lobe of the pituitary gland was studied in the frog Rana ridibunda using the pulse-chase technique. Analysis of radioactive proteins by dodecyl sulfate polyacrylamide gel electrophoresis showed that during pulse incubations a 36,000 dalton (36K) glycosylated prohormone was synthesized. It disappeared slowly during chase incubations, giving rise to another glycosylated protein (Mr 18K), identified as the N-terminal fragment of POMC. This latter protein was secreted to the incubation medium. High performance liquid chromatography analysis of peptides synthesized during chase incubations revealed the biosynthesis of two peptides related to gamma-MSH, three peptides related to alpha-MSH, one endorphin-related and one CLIP-related peptides. These newly synthesized peptides were slowly secreted to the incubation medium. Among the alpha-MSH related peptides, only the des-N alpha-acetyl alpha-MSH form of the peptide was found to be present within the cells, in contrast to the incubation medium where the presence of des-N alpha-acetyl alpha-MSH and a modified alpha-MSH was demonstrated.  相似文献

A peptide with somatostatin-like immuno- and bioactivity has been isolated from an aqueous extract of 96 800 rat hypothalamic by means of immunoaffinity chromatography, gel filtration and reverse-phase high-performance liquid chromatography (HPLC). Chemical characterization by amino acid analysis, tryptic peptide mapping and retention behavior in two HPLC system showed that the peptide was indistinguishable from somatostatin-28 previously characterized from several species. This evidence suggests that rat hypothalamic somatostatin-28 is identical in structure to porcine and ovine somatostatin-28.  相似文献

The gastric exocrine inhibitory activities of somatostatin-28 (SS-28) and somatostatin-14 (SS-14) were determined in conscious cats prepared with gastric fistulae. Gastric acid and pepsin secretions were stimulated with pentagastrin. Expressed in terms of exogenous doses, SS-14 (ID50: 1.49 nmol . kg-1 . h-1) was 3.4 times more potent than SS-28 (ID50: 5.12 nmol . kg-1 . h-1) as an inhibitor of gastric acid secretion. Similarly SS-14 (ID50: 0.25 nmol . kg-1 . h-1) was 3.8 times more potent than SS-28 (ID50: 0.96 nmol . kg-1 . h-1) as an inhibitor of pepsin secretion. Expressed in terms of circulating plasma concentration measured by radioimmunoassay, SS-14 (ID50: H+, 232 and pepsin 73 pM) was 8-9 times more potent than SS-28 (ID50: H+, 2112 and pepsin, 611 pM) as an inhibitor of gastric exocrine secretions. The plasma immunoreactive half-life of SS-28 (6.1 min) was double that for SS-14 (2.4 min) possibly due to a slower theoretical metabolic clearance rate of the larger peptide (30 and 87 ml . kg-1 . min-1, respectively). Both peptides had similar apparent distribution volumes (SS-14, 306 and SS-28, 263 ml . kg-1). As judged by gel chromatography of plasma samples, there was no evidence for the conversion of SS-28 to SS-14 in vivo. The reduced activity of SS-28, compared with SS-14, against gastric exocrine secretions contrasts with its more potent effects in the pituitary and pancreas.  相似文献

Two b-type cytochrome components, differing in their mid-point redox potentials as well as their reactivities toward CO, are demonstrated in the membrane particle of Azotobactervinelandii by potentiometric titration. The low redox component is revealed to be CO-reactive while the higher redox component is not. Accordingly, the later cytochrome is believed not to be directly involved in the reduction of oxygen while the former component is the cytochrome o of Azotobacter.  相似文献
