The effect of 2'-deoxycoformycin (DCF) on the PFC responses of AKR mice to SE, TNP-Ficoll, and TNP-B. abortus was examined. Subcutaneous injection of DCF 4 days before antigen caused suppression of all three responses by 70 to 78%. In contrast, injection of DCF 1 day after antigen caused enhancement of both the anti-SE and the anti-TNP-Ficoll responses. Although a single high dose of cortisone acetate injected 4 days before antigen caused a similar suppression, the effect of DCF was not mediated via a steroid release, inasmuch as DCF also suppressed the immune response in adrenalectomized mice. The response of BALB/c mice to TNP-Ficoll was also inhibited by DCF pretreatment and enhanced by injection of DCF after antigen. In contrast, in athymic mice DCF caused suppression of the anti-TNP-Ficoll PFC response, whether injected before or after antigen. These results are interpreted as suggesting that DCF causes suppression primarily via an effect on B cells. The enhancement seen in normal but not in athymic mice may possibly be ascribed to an effect on suppressor T cells. Apparently the enhancement of both TD and TI responses caused by DCF injected 1 day after antigen in normal mice is the net result of these two opposing effects. The results imply that helper T cells are resistant to DCF.  相似文献

Streptococcal pyrogenic exotoxin (SPE), a toxic protein, secreted by Group A streptococci modifies antibody responses in two ways. It suppresses the early peak plaque-forming cell (PFC) and serum antibody responses to sheep erythrocytes (SE) and it engenders a late burst of PFC detected at 12–14 days. We have termed the late phase a deregulated response. This effect has been observed in rabbits and NIH (+/+ and +/nu) mice. NIH athymic nude (nu/nu) mice exhibit the early suppressed response but do not show the late phase. In reconstruction experiments to delineate the responsible target site of SPE we have conferred upon the nude or nude spleen cells in vitro, +/nu PFC responsiveness to SE by transfer of +/nu spleen cells in vivo or by supplementation with +/nu spleen cells in Marbrook cultures. When this is done, complementation of nude PFC responses and their ability to exhibit a deregulated response after SPE treatment is conferred coordinately. Pretreatment of donor cells with a B-cell inhibitory dose of X-ray or with a B-cell inhibitory dose of anti-Ig serum + C′ does not inhibit complementation of nude cells to +/nu responsiveness. Moreover, such donor suspensions when treated with SPE retain the ability to complement and to confer upon nude cells the ability to exhibit the late burst of PFC (a deregulated response). A similar pretreatment of the donor cell suspension with an anti-T-cell serum and C′, however, markedly inhibits both the adoptive complementation and the deregulation of the nude mouse PFC response. Thus, it is demonstrated that the target cell affected in this way by SPE is a T-cell. We presume from this evidence that SPE inhibits a T-cell which is involved in the regulation of antibody formation.  相似文献

The in vivo immunosuppressive effects of Concanavalin A (Con A) on the thymus (T) helper dependent response to sheep erythrocytes (SRBC) and the T helper independent response to E. coli lipopolysaccharide 055: B5 have been investigated. Maximum suppression was observed in BALB/c mice treated with 3 successive ip injections of 100 μg each of Con A administered on Days ?1, 0, and +1 relative to the day of immunization (Day 0) with SRBC (splenic PFC on Day 4 reduced from 74,000 down to 1400). As little as 10 μg × 3 of Con A was capable of depressing both the PFC and serologic response while 2.5 μg × 3 was ineffective. A single ip injection of 300 μg of Con A administered simultaneously at the time of immunization with SRBC reduced splenic PFC from 74,000 down to 9990 and serum antibody titers by 3–4 log2 units. Significant depression was noted if mice were treated 1, 2, or 3 days prior to but not following immunization. Immunosuppression was noted in mice which had been treated and immunized ip or iv or treated iv and immunized ip. Heat inactivation reduced if not abolished the immunosuppressive properties of Con A.Mice immunized with varying doses of a bacterial vaccine of E. coli 055: B5 (15–1500 × 106 killed organisms) and treated with Con A on days ?1, 0, and +1 had no significant depression of splenic PFC when compared to nontreated controls. Mice treated with Con A and simultaneously immunized with both SRBC and E. coli had a 37-fold reduction in the PFC response to SRBC but only a 2-fold reduction in the response to E. coli. This differential immunosuppressive effect on T helper dependent and independent responses is consistent with the recently reported in vitro specificity which Con A has for theta antigen bearing lymphocytes.  相似文献

The effect of the iron-binding glycoprotein lactoferrin (LF) on the in vitro primary antibody response of mouse splenic cells to a T-lymphocyte-dependent antigen, sheep erythrocytes (SE), and a T-lymphocyte-independent antigen, trinitrophenolated Brucella abortus, was examined. Both iron-saturated and native LF (8% saturated) at 10?10 to 10?6M concentrations but not transferrin (an iron-binding glycoprotein similar to LF) significantly (P < 0.05) decreased the number of direct (IgM) plaque-forming cells (PFC) to SE and trinitrophenol (TNP) as determined by the hemolytic plaque assay. LF was equally effective in decreasing the PFC response to TNP in T-lymphocyte-depleted splenic cell cultures. Concentrations of LF which decreased the PFC response were noncytotoxic. A 1-hr exposure of splenic cells to LF at the beginning of the 5-day culture period but not 1 hr prior to assaying for PFC, or exposure of isolated macrophage-rich but not lymphocyte-rich populations to LF prior to reconstitution resulted in a significant decrease in the anti-SE response. These data suggest that LF which is synthesized and released by polymorphonuclear leukocytes and present in inflammatory lesions may play a role in modulating the antibody response through an effect on the macrophage.  相似文献

Previous studies have shown that cellular and humoral antibody production to type III pneumococcal polysaccharide (SSS-III) is not appreciably altered in neonatally thymectomized mice and is enhanced in animals which have been treated with ALS. In order to determine what effect ALS has on the response to another antigen which does appear to require helper T cells, immunity to E. coli 055:B5 has been investigated. BALB/c mice were injected i.p. with 0.25 ml of ALS on days ?1, 0, and +1 relative to the day of immunization (d.0) with a killed E. coli bacterial vaccine. Splenic plaque forming cells (PFC) and serum hemolysin and hemagglutinin titers were determined 6 days later using sheep erythrocytes which had been coated with purified E. coli lipopolysaccharide (LPS). Mice treated with ALS or normal heterologous serum and immunized with an optimal immunogenic dose of bacteria (150 × 106) had similar numbers of splenic PFC and serum antibody titers. No significant immunosuppressive effect was noted over a wide range of antigen (0.015–1500 × 106) although dose related variations were seen. In contrast to its effect on the response to SSS-III, no enhancement was noted. ALS treated mice which had been simultaneously immunized with E. coli and sheep RBC had specific depression of the T helper dependent response to SRBC but not to LPS. The lack of immunosuppressive effect on antibody production to E. coli LPS provides strong evidence that ALS preferentially acts on T lymphocytes. It further indicates that enhancement occurs with some but not all T helper independent antigens.  相似文献

An effective malaria vaccine is a public health priority. Proteins expressed during the blood-stage of the parasite life cycle have been proposed as good vaccine candidates. No such blood-stage vaccine, however, is available against Plasmodium falciparum, the deadliest Plasmodium species. We show here that P. falciparum serine repeat antigen 5 (SERA5) is a potential vaccine immunogen. We have constructed a new recombinant molecule of SERA5, namely SE36, based on previously reported SE47′ molecule by removing the serine repeats. Epidemiological study in the holo-endemic population of Solomon Islands shows highly significant correlation of sero-conversion and malaria protective immunity against this antigen. Animal experiments using non-human primates, and a human phase 1a clinical trial assessed SE36 vaccine immunogenicity. Vaccination of squirrel monkeys with SE36 protein and aluminum hydroxyl gel (SE36/AHG) conferred protection against high parasitemia and boosted serum anti-SE36 IgG after P. falciparum parasite challenge. SE36/AHG was highly immunogenic in chimpanzees, where serum anti-SE36 IgG titers last more than one year. Phase 1a clinical trial (current controlled trials, ISRCTN78679862) demonstrated the safety and immunogenicity of SE36/AHG with 30 healthy adults and 10 placebo controls. Three subcutaneous administrations of 50 and 100 μg dose of SE36/AHG were well-tolerated, with no severe adverse events; and resulted in 100% sero-conversion in both dose arms. The current research results for SE36/AHG provide initial clinical validation for future trials and suggest clues/strategies for further vaccine development.  相似文献

Young-adult and old (CS7BL × C3H)F1 mice were injected with either of two tolerogens followed by challenge with dinitrophenylated human γ globulin (DNP-HGG). The primary IgM and IgG responses of spleen cells to the DNP determinant were evaluated by a modified hemolytic plaque assay. Carrier-specific thymus-derived cell (T-cell) tolerance was induced in mice with deaggregated HGG. Hapten-specific bursal-derived cell (B-cell) tolerance was induced with DNP coupled to isogeneic mouse IgG. The dose of these two tolerogens was successively decreased by 10-fold decrements until a response similar to that of control mice was achieved. The minimum tolerizing dose (MTD) was then determined for young-adult and aged B-and T-cells. We found that the MTD for old B-cells was 10 times greater than that obtained with young B-cells for both the direct and indirect PFC responses. No difference in MTD was observed between young and old T-cells when assessed by the indirect response; the MTD for old T-cells was 10-fold greater than that obtained for young-adult T-cells when the direct response was evaluated. A double threshold of tolerance was found for T-cells of young-adult mice.  相似文献

Mice of the NZB and NZW strains and their F1 hybrid produce antihapten plaque-forming cell (PFC) responses to T-dependent antigens (trinitrophenylated bovine gamma globulin and dansylated keyhole limpet hemocyanin) which are of unusually restricted heterogeneity of affinity, are relatively lacking in low-affinity PFC, and are of relatively high average affinity. Since some low-affinity PFC are present in NZB mice early after immunization, the results suggest a particularly marked down-regulation of low-affinity antibody production by these strains. The non-autoimmune-prone F1 hybrid (NZB × CBA) produces a typical heterogeneous response containing a high proportion of low-affinity PFC. Thus, the tendency to down-regulate low-affinity PFC is not inherited as a simple Mendelian dominant trait. The response of NZB mice to T-independent antigens does not show the same restricted heterogeneity of affinity. In fact, late after injections of trinitrophenylated Ficoll, NZB mice tend to have more heterogeneous responses than nonautoimmune-prone BALB/c mice in which a marked down-regulation of high-affinity antibody-producing PFC is seen. The possible relationship between these unusual features of the immune response of NZB and some related strains and their tendency to develop autoimmune disease is discussed.  相似文献

To further define requirements for T cell help in the stimulation of an in vivo IgE response, we studied a system in which the injection of mice with a goat antibody to mouse IgD (GaMD) stimulates large polyclonal IgG1 and IgE responses. In this system, both responses are blocked by anti-CD4 antibody, but only the IgE response is blocked by (anti-IL-4) antibody. Anti-CD4 antibody, if injected 5 days after GaMD, was found to inhibit the GaMD-induced IgE response to a much greater extent than the IgG1 response, even though both responses occur simultaneously and are inhibited to an equal extent by optimal or suboptimal doses of anti-CD4 antibody administered 2 days after GaMD. Even a suboptimal, 50-micrograms dose of anti-CD4 antibody, when injected 5 days after GaMD, inhibited the IgE response to a much greater extent than did an optimal 10-mg dose of anti-IL-4 antibody injected at the same time, even though 10 mg of anti-IL-4 antibody more completely inhibited GaMD-induced IgE production than did 50 micrograms of anti-CD4 antibody when injected 2 days after GaMD. These observations provide evidence that a late acting form of T cell help other than IL-4 is important for the generation of an IgE response but not an IgG1 response in GaMD-immunized mice.  相似文献

The in vivo effects of histamine injection in LAF1 male mice on the immune reactivity to trinitrophenylated bovine γ-globulin was studied using plaque-forming cell (PFC) responses and their avidity distributions. Splenic anti-trinitrophenyl (anti-TNP) PFC responses of mice treated with histamine (5 × 10?6 mol or 1 mg, intravenously) were significantly reduced in number and restricted in heterogeneity and characterized by a preferential loss of high-avidity IgG PFCs. The reduced PFC response in histamine-treated mice was dose and time dependent. No evidence of suppressor cell activity in the spleens from histamine-treated mice was demonstrable. Only histamine-treated mice produced a significantly high percentage of anti-idiotype-blocked, hapten-augmentable IgG PFCs, suggesting the presence of auto-anti-idiotypic activity. Immune sera taken from histamine-treated mice caused an inhibition of anti-TNP PFC in vitro. This PFC-inhibiting factor in immune sera of histamine-treated mice was an antibody of the IgG1 and IgG2a class, lacked anti-TNP antibody activity, but reacted with anti-TNP antibody of LAF1 origin. Passive hemagglutination study of this sera showed anti-(anti-TNP F(ab′)2-IgG) titer. Thus, the results of this study suggest that histamine in combination with antigen induces auto-anti-idiotypic antibody which, in turn, is involved in the normal regulation of the immune response to trinitrophenylated bovine γ-globulin in vivo.  相似文献

The effect of injection of the synthetic corticosteroid dexamethasone sodium phosphate upon the primary response to Escherichia coli lipopolysaccharide (LPS) was studied in mouse spleen and bone marrow. Daily corticosteroid injections, starting 1 day before immunization with LPS, could suppress the anti-LPS plaque-forming cell (PFC) response in the spleen. The higher the dose of corticosteroids, the more the splenic PFC response was suppressed. On the other hand, the bone marrow PFC response showed a dose-dependent enhancement after corticosteroid injections. This effect was maximal when tested 7 days after antigen injection, and constituted a 3- to 15-fold increase after daily injection of 16 mg dexamethasone/kg body wt. The same effect was found in genetically athymic nude mice, showing that the corticosteroid-mediated enhancement of the anti-LPS PFC response in the bone marrow is not due to elimination of T suppressor cells. Probably the differential effect of corticosteroids upon antibody formation in spleen and bone marrow is due to a redistribution of B-lineage cells, with a resulting accumulation in the bone marrow.  相似文献

Mice homozygous for the nu gene fail to develop a thymus. In comparison to spleen cells from +/nu mice spleen cells from nu/nu mice have a deficient 19S PFC response to SRBC when tested in culture or in vivo. This deficiency is due to a lack of “helper” T cells in nu/nu spleen; A cells and B cells appear to be normal. The capacity of nu/nu spleen cells to produce a PFC response in culture can be corrected by the addition of T cells obtained from either the thymuses or the spleens of +/nu mice. In contrast to “helper” T cells obtained from the spleen, “helper” T cells obtained from the thymus appear to require the capacity for proliferation during the response to SRBC.  相似文献

Polyclonal plaque-forming cell (PFC) responses in murine spleen cells induced by Staphylococcus aureus and S. epidermidis were studied. Injection of Balb/c mice with S. aureus strain 248βH resulted in the generation of anti-trinitrophenyl (TNP) and anti-sheep red blood cell PFC in their spleens. Cultures of Balb/c spleen cells in the presence of S. aureus 248βH, Cowan I, or a protein A-deficient mutant yielded many anti-TNP PFC. The larger the number of organisms that were added to the cultures, the better was the PFC response. Both living and killed organisms, were capable of inducing the response, but an excess of living 248βH organisms in the cultures abrogated the response. All of the organisms (12 strains of S. aureus and 11 strains of S. epidermidis) freshly isolated from patients had the ability to induce the polyclonal PFC response in cell cultures. These organisms stimulated cultured C3H/HeJ mouse spleen cells, which were unresponsive to bacterial lipopolysaccharide (LPS). Cultured cells from the spleens of athymic nu/nu mice also responded to these organisms, and the number of PFC in nu/nu cell cultures was always greater than that in nu/+ cells prepared from a haired litter mate. Moreover, the responses of nu/nu spleen cell cultures to which nylon wool column-filtered splenic nu/+ T cells were added were lower than expected. These findings suggest that the polyclonal PFC response to staphylococci is thymus independent, but that the magnitude of the response is regulated by mature T cells. Cultures of macrophage-depleted spleen cells responded to the organisms to an extent similar to that of the control. The 248βH organisms were less capable of stimulating spleen cells of 2-week-old mice (i.e., early maturing B cells) than LPS. However, spleen cells from adult (7-week-old) and aged (9-month-old) mice responded well to both the organisms and LPS. Previous sensitization with the organisms in vivo did not affect any polyclonal responses of spleen cells in vitro to either the organisms or LPS. The role of staphylococcal protein A in the polyclonal PFC response to staphylococci is discussed.  相似文献

Spleen cultures prepared from mice injected 24 hr earlier with 2 × 106?2 × 108 sRBC and challenged in vitro with sRBC produced 10 times more anti-sRBC IgM PFC than cultures prepared from uninjected mice. The effect was specific for the particular species of foreign RBC injected in vivo. In vitro responses to TNP were also increased in spleen cultures prepared from animals injected 24 or 12 hr earlier with carrier RBC alone, directly implicating carrier-specific T cells in this process. Similar enhancements of PFC formation occurred in cultures prepared from mice which had been injected with sRBC 24 and 48 hr earlier, but which were exposed to lethal irradiation at 1 hr after injection of antigen, if their spleens were shielded extracorporeally during irradiation. This finding indicated that in vivo recruitment of antigen-reactive extrasplenic X-ray-sensitive cells from the circulating lymphocyte pool by the spleen could not account for the observed enhancement.Proliferation in the spleen of antigen-reactive T cells, commencing 12–20 hr after the administration of antigen, was demonstrated by the tritiated thymidine pulse technique. An 8-hr hot-pulse given to spleen cell cultures from normal animals at 20 hr after in vitro challenge with antigen did not affect the rate of generation of IgM-producing cells; however, administration of a similar pulse to cultures which were initiated at 12 or at 20 hr after the in vivo injection of sRBC eliminated the enhanced generation of PFC and delayed the in vitro response to sRBC by 24 hr.Spleen cell cultures were prepared from mice which had been injected in vivo with sRBC at 12, 20, and 70 hr earlier, and 8- to 10-hr hot pulses were given immediately after initiation of the cultures. The cultures were then challenged with sRBC-TNP; antibody responses to TNP were greatly reduced in hot-pulsed cultures prepared from mice injected in vivo with carrier RBC at 12 or 20 hr prior to initiation of the cultures. In contrast, antibody responses to TNP observed in hot-pulsed cultures prepared from mice which had been injected with carrier RBC at 70 hr prior to initiation of the cultures were generally similar to those of nonpulsed 70 hr control cultures. This result suggests that the onset of T helper cell proliferation begins within 12–20 hr after injection of antigen, but subsides in vivo within 70 hr. By that time, the antigen-reactive T cells have already differentiated to perform their helper function.In spite of the triggering of T-cell proliferation during the first 24 hr after injection of antigen, spleen cell cultures prepared from mice which had been injected 24 hr earlier in vivo with 2 × 108 sRBC produced only minimal numbers of anti-sRBC PFC if no antigen was added to the cultures. The presence of unprocessed antigen thus appears to be a requirement for B-cell proliferation in vitro, even after T-cell division has been triggered. This finding is consistent with earlier suggestions that the function of “helper” T cells may not be limited to passive transport of antigenic determinants to B cells. Evidence is also presented to support the contention that the antigen-reactive T cell involved in this process may have to undergo cell division in order to develop “helper” capacity.  相似文献

In the present series of experiments we have studied the effects of anti-lymphocyte serum (ALS) and concanavlin A (Con A) on the immune response to technetium-99m-labeled sheep erythrocytes (SRBC) and have related this to the localization and persistence of antigen at the site of induction and antibody synthesis. The number of 99mTc-labeled SRBC in the spleen and liver was quantified by gamma scintillation counting and the cellular kinetics of the splenic antibody response was determined by means of the hemolytic plaque technique. After injection of normal rabbit serum (NRS)-treated control mice with 4 × 10899mTc-labeled SRBC, the number of cells localizing in the spleen ranged from a high of 4.2 × 106 on Day 1 to a low of 1.7 × 106 on Day 4, while the number in the liver ranged from a high of 68.8 × 108 on Day 1 to 18.6 × 106 on Day 4. The number of splenic plaque-forming cells (PFC) increased from 321–429 on Day 1 to 93,000–101,000 PFC on Day 4 and this was paralled by a rise in serum hemagglutinin and hemolysin titers. In mice treated with ALS on the other hand, splenic localization initially was increased 10-fold, hepatic localization was unchanged, and the antibody response was markedly suppressed. Splenic PFC ranged from approximately 100 between days 1 and 3 and increased to only 500 on Day 4. Mice which received Con A on Day — 1 had a reduction in splenic PFC which ranged from 150 on Day 1 to 1900 on Day 4. Splenic localization of 99mTc-labeled RBC initially was three- to fourfold greater than that in NRS-treated mice and then decreased to control levels. The increased numbers of SRBC detected in the spleens of immunosuppressed mice at the time of peak response can be attributed to decreased in vivo lysis by reduced numbers of splenic antibody-producing cells.  相似文献

An adoptive transfer system is described to measure serum helper activity in the primary antibody response to sheep red blood cells (SRBC). Mice injected with a high dose of cyclophosphamide and reconstituted with rabbit anti-thymocyte serum-treated spleen cells were used as recipients. Serum obtained 9 hr after ip injection of normal mice with 2 × 108 SRBC (S(SRBC)) injected i.v. in the recipients caused a significant enhancement of the antibody response to 2 × 107 SRBC. The serum helper activity was not generated in thymectomized animals and could be absorbed from S(SRBC) by normal and formalinized SRBC. The SRBC-specific serum helper activity (SSHA) is heat labile (30 min 56 °C) and shows allogeneic restriction. Another test system described in literature for measuring T-cell help in vivo was less suited to measure SSHA in the response to 2 × 107 SRBC. A system using normal mice injected with 105 SRBC for determining specific immune response-enhancing factor (SIREF), demonstrated SIREF activity in S(SRBC). It did, however, not measure SSHA, as absorption of S(SRBC) with formalinized SRBC did not abolish the activity in that system.  相似文献

The in vitro mitogen responses of spleen cells from mice injected ip with the nonantigenic adjuvant, Al(OH)3, are markedly depressed. This depressed reactivity was found to be mediated by a population of nylon wool adherent, Fc-receptor-bearing suppressor cells. Suppressor cells were detected only in the spleens of the adjuvant-treated mice, as the response of lymph node cells to mitogenic stimulation in vitro was found comparable to that of normal controls. Moreover, elevated levels of suppressive activity could be detected in sera of Al(OH)3-treated mice during the first week after adjuvant administration, which, however, did not correlate with either the long-lasting presence of suppressor cells or the in vivo normal immune response of the adjuvant-treated animals. Studies designed to test the effect of suppressor cells on the generation of splenic PFC in vivo revealed that both the direct and indirect PFC responses against SRBC inoculated iv were enhanced rather than suppressed, as compared to those of the normal controls. Furthermore, the level of cytotoxic lymphocytes generated in spleens of Al(OH)3-treated mice immunized with allogeneic tumor cells was equal to or higher than that of the normal controls. In view of the present results, we feel that the concept that splenic, nonspecific suppressor cells (macrophages) are immunosuppressive in vivo as well as the in vivo relevance of in vitro findings should be carefully reevaluated.  相似文献

Mice exposed to 35 cercariae of the human helminth Schistosoma mansoni develop chronic (greater than 16wk) infections characterized by immunoregulation of their cell-mediated granulomatous responses to schistosome eggs. Evidence was sought regarding the possible development of anti-idiotypic responses against the responses to soluble egg antigens (SEA). Sera were collected from CBA/J mice with chronic S. mansoni infections. Multiclonal idiotypic, anti-SEA antibody (id) was prepared from these pooled sera by affinity chromatography on an SEA immunoadsorbent column. Analysis of the id preparations by polyacrylamide gel electrophoresis demonstrated that this material contained only immunoglobulin heavy and light chains. A modified reverse plaque-forming cell (PFC) assay was developed to quantify anti-idiotypic (anti-id) PFC in spleen cell preparations from infected and age-matched control CBA/J mice. Expression of anti-id PFC began 2 to 3 wk after onset of egg production and continued throughout the course of infection. Positive selection of anti-id-reactive spleen cells by panning cell preparations from chronic mice on id-coated plates resulted in an enrichment of anti-id PFC in the id-adherent population. Conversely, the number of PFC reactive with SEA (id-producing PFC) was lowered by panning on id-coated plates. These data demonstrate the occurrence of anti-id responses during schistosomiasis mansoni. It is possible that such an immunoregulatory mechanism could play an important role in how an animal modulates the granulomatous response that leads to the formation of pathologic lesions and in the maintenance of this chronic infection.  相似文献

Synergistic effects of two synthetic adjuvants, dimethyldioctadecylammonium bromide (DDA) and dextran sulfate (DXS) on the humoral response to sheep red blood cells (SRBC) were investigated. Mice received intraperitoneal (ip) injections of adjuvant and antigen simultaneously. The number of plaque-forming cells (PFC) in the spleen were determined 5 days later and circulating anti-SRBC antibodies were measured till 16 weeks after immunization. Although combinations of DDA and DXS were very effective in enhancing the PFC response to both moderate (2 X 10(7] and low (2 X 10(6] doses of SRBC, synergy between the adjuvants was only observed at the low dose of SRBC. Optimal augmentation of the primary response to the low antigen dose was evoked by the combination of the highest dose tested of either adjuvant (1 mumol DDA and 1 nmol DXS) resulting in a 560-fold increase of the number of PFC in the spleen as compared to controls. Even combinations of relatively small amounts of both adjuvants were very effective in augmenting the response to SRBC. Mice receiving half the amounts of both adjuvants with 2 X 10(6) SRBC displayed increased numbers of PFC in the spleen at Day 5 as well as increased titers of total anti-SRBC antibodies at Week 1 and Week 2 and 2-mercaptoethanol-resistant antibodies from Week 4 till Week 16 as compared to the calculated sum of responses in mice which received either DDA (0.05 mumol per mouse) or DXS (0.05 nmol per mouse). The mechanism behind the synergy between these adjuvants is discussed and the possibility of discerning adjuvants on their modes of action is suggested.  相似文献
