The new model proposed to account for the energy requirement for growth includes both a constant maintenance energy term (m) independent of the specific growth rate and a term (m′) which decreases linearly with increase in specific growth rate and becomes zero at the maximum specific growth rate. The available data for testing the model do not deviate significantly from the relations predicted. Consistent values of the maximum growth yield (Y G) can be derived, irrespective of whether the cultures are energy limited or energy sufficient. Attention is drawn to the possibility that the constant maintenance energy term may be estimated from the maximum specific growth rate.  相似文献

An improved bioassay is described for a factor that promotes tumor growth which was first obtained from extracts of pinto bean leaves with crown gall tumors. Sixteen primary pinto bean leaves per sample are inoculated with sufficient Agrobacterium tumefaciens to initiate about 5 to 10 tumors per leaf and treated with tumor growth factor at day 3 after inoculation. The diameters of 30 to 48 round tumors (no more than 3 randomly selected per leaf) are measured per test sample at day 6. Mean tumor diameter increased linearly with the logarithm of the concentration of tumor growth factor applied. The tumor growth factor was separated by column chromatography from an ultraviolet light-absorbing compound previously reported to be associated with fractions having maximal tumor growth factor activity. Partly purified tumor growth factor showed no activity in a cytokinin bioassay or an auxin bioassay, and negligible activity in gibberellin bioassays. Representatives of these three classes of growth factors did not promote tumor growth. Extracts from crown gall tumors on primary pinto bean leaves, primary castor bean leaves, Bryophyllum leaves, carrot root slices, and tobacco stems showed tumor growth factor activity, whereas extracts from healthy control tissues did not. Extracts from actively growing parts of healthy pinto beans, Bryophyllum, and tobacco, however, showed tumor growth factor activity. Tumor growth factor is proposed to be a normal plant growth factor associated with rapidly growing tissues. Its synthesis may be activated in nongrowing tissues by infection with Agrobacterium sp.  相似文献

A methanogenic coccoid organism, Methanosarcina mazei LYC, was isolated from alkaline sediment obtained from an oil exploration drilling site. The isolate resembled M. mazei S-6 by exhibiting different morphophases during its normal growth cycle. It differed from M. mazei S-6 by undergoint a spontaneous shift from large, irregular aggregates of cells to small, individual, irregular, coccoid units. In batch cultures at pH 7.0, M. mazei LYC grew as aggregates during the early growth stage. As the batch culture began exponential growth, the cell aggregates spontaneously dispersed: the culture liquid became turbid, and myriads of tiny (diameter, 1 to 3 μm) coccoid units were observed under phase-contrast microscopy. Disaggregation apparently was accomplished by the production of an enzyme which hydrolyzed the heteropolysaccharide component of the cell wall; the enzyme was active on other Methanosarcina strains as well. Although the enzyme was active when tested at pH 6.0, it apparently was not produced at that pH: when strain LYC was grown at pH 6.0, only cell aggregates were present throughout batch growth. Individual coccoid cells of M. mazei LYC were sensitive to sodium dodecyl sulfate, but the large aggregates of cells were not. Strain LYC rapidly used H2-CO2, in addition to methanol, and mono-, di-, and trimethylamine as methanogenic substrates; acetate was used very slowly. Its optimum growth temperature was 40°C, and its optimum pH was 7.2.  相似文献

Aeromonas hydrophila is frequently reported from arsenic affected areas. Present study was aimed to determine the effect of arsenic and temperature on growth of A. hydrophila. The bacteria were isolated from naturally infected fish from a water body in Birbhum, West-Bengal, India, which is reported to be an arsenic-free area. Arsenic concentration in natural aquatic reservoirs (e.g., pond, lake or river) varies from 0–6 mg/L. No significant change in bacterial growth was observed within this range of arsenic exposure. However, variation in temperature impacted the growth of A. hydrophila. A single dimension model was constructed using simple logistic equation. Rate parameters of the model were derived from the experimental observations. Comparison of model results and laboratory observations gives a good conformity regarding the effect of variation of arsenic concentration and temperature change on growth of this bacterium. From the analysis of this model we further get the idea that the maximum growth of A. hydrophila is supposed to be at 31.4°C in absence of arsenic, whereas at 477 mg/L arsenic concentration, the growth of the bacteria totally stops at 30°C.  相似文献

Streptococcus intermedius is well known to produce severe infections in various areas of the body. In this study, we evaluated the ability of S. intermedius to utilise human transferrin as a source of iron and investigated the mechanism by which iron can be obtained from this plasma protein. Adding either ferrous sulfate or holotransferrin to an iron-deficient culture medium allowed growth of S. intermedius. Cultivation of S. intermedius under an iron-poor condition was associated with the over expression of a 58 kDa cell surface protein. Neither siderophore activity nor reductase activity could be detected. Moreover, cells of S. intermedius did not show transferrin-binding activity or proteolytic activity toward transferrin. It was found that S. intermedius could rapidly decrease the pH of the medium during cell growth, resulting in a release of iron from holotransferrin. When the buffering capacity of the culture medium was significantly increased, the holotransferrin could not support growth of S. intermedius. It is suggested that under certain circumstances, S. intermedius may migrate from its normal niche (oral cavity), reach a particular site and create a localised environment where the pH can be lowered with the subsequent release of iron from transferrin. This would allow bacterial growth and initiation of the infectious process.  相似文献

The chlorophyll a:b ratio was shifted in Chlorella vannielii by varying the illuminance under which the cells were cultured—the ratio increased from 2.9, 3.0, 4.0, and 4.8 to 6.2, respectively, at 100, 300, 900, 2,700 and 6,000 foot candles. The 6,000-foot candle cells retained an optimal growth rate at the chlorophyll a:b ratio of 6.2 which was the upper limit of normal growth. Comparisons were made between the 300-and 6,000-foot candle cultures to determine the significance to the photosynthetic mechanism of a shift in the chlorophyll a:b ratio.  相似文献

Experimental studies on the secondary metabolism characteristic of lichens have been impeded by the slow growth of the fungi and by the inconsistent results of many attempts to induce the pathways in the fungi isolated from their photosynthetic partners. In the present study, a lichen-specific secondary pathway was consistently induced in a lichen fungus (Cladonia grayi) grown in the absence of the alga. The depside (4-O-demethylsphaerophorin) and two depsidones (grayanic and 4-O-demethylgrayanic acids) found in the natural lichen began to accumulate a few days after the transfer of lightly fragmented mycelia from liquid to solid medium. Induction was enhanced on drier substrates and was correlated with the proliferation of aerial hyphae, where the major product (grayanic acid) accumulated in extracellular patches visible by fluorescence microscopy. The time course was analyzed by quantitative high-performance liquid chromatography of extracts from small cultures grown on nylon filters. Induction was rapid in view of the slow growth of the fungus, and secondary productivity was comparable to that of some nonlichen fungi. These results confirm that the alga is not needed for catalysis in lichen depside and depsidone biosynthesis and suggest instead that the characteristic secondary metabolism of natural lichen is linked to their aerial habit of growth.  相似文献

Invaders generally show better individual performance than non-invaders and, therefore, vital rates (survival, growth, fecundity) could potentially be used to predict species invasiveness outside their native range. Comparative studies have usually correlated vital rates with the invasiveness status of species, while few studies have investigated them in relation to population growth rate. Here, I examined the influence of five vital rates (plant establishment, survival, growth, flowering probability, seed production) and their variability (across geographic regions, habitat types, population sizes and population densities) on population growth rate (λ) using data from 37 populations of an invasive, iteroparous herb (Lupinus polyphyllus) in a part of its invaded range in Finland. Variation in vital rates was often related to habitat type and population density. The performance of the populations varied from declining to rapidly increasing independently of habitat type, population size or population density, but differed between regions. The population growth rate increased linearly with plant establishment, and with the survival and growth of vegetative individuals, while the survival of flowering individuals and annual seed production were not related to λ. The vital rates responsible for rapid population growth varied among populations. These findings highlight the importance of both regional and local conditions to plant population dynamics, demonstrating that individual vital rates do not necessarily correlate with λ. Therefore, to understand the role of individual vital rates in a species ability to invade, it is necessary to quantify their effect on population growth rate.  相似文献

An important emerging question in the area of signal transduction is how information from different pathways becomes integrated into a highly coordinated response. In budding yeast, multiple pathways regulate filamentous growth, a complex differentiation response that occurs under specific environmental conditions. To identify new aspects of filamentous growth regulation, we used a novel screening approach (called secretion profiling) that measures release of the extracellular domain of Msb2p, the signaling mucin which functions at the head of the filamentous growth (FG) MAPK pathway. Secretion profiling of complementary genomic collections showed that many of the pathways that regulate filamentous growth (RAS, RIM101, OPI1, and RTG) were also required for FG pathway activation. This regulation sensitized the FG pathway to multiple stimuli and synchronized it to the global signaling network. Several of the regulators were required for MSB2 expression, which identifies the MSB2 promoter as a target “hub” where multiple signals converge. Accessibility to the MSB2 promoter was further regulated by the histone deacetylase (HDAC) Rpd3p(L), which positively regulated FG pathway activity and filamentous growth. Our findings provide the first glimpse of a global regulatory hierarchy among the pathways that control filamentous growth. Systems-level integration of signaling circuitry is likely to coordinate other regulatory networks that control complex behaviors.  相似文献

Apical growth rates of Furcellaria lumbricalis (Huds.) Lamour. and Ahnfeltia plicata (Huds.) Fries (Gigartinales: Rhodophyceae), and Ascophyllum nodosum (L.) Le Jol. (Fucales: Phaeophyceae), were studied in spray culture, applying independent microcomputer systems for control, data-logging, and apical growth measurements. The system functioned with a minimum of maintenance. Sea-water temperature and light seemed to be more important for growth than nutrient supply under the prevailing conditions. Basal shoots (young, undivided parts of the plants close to the holdfast) of Furcellaria lumbricalis had growth rates three times higher than the apices (apical meristem on the distal end of mature plants) from the older parts of the thallus. For Ascophyllum nodosum apical growth rates of basal shoots, divided and undivided apices were similar. Whereas apical growth rate of A. nodosum was normal, the biomass yield was low. A. nodosum cultivated simultaneously in tanks and in continuous spray had similar growth rates. The spray cultivation technique facilitated control of the experimental conditions, and handling of the plants during the experimental work while excluding epiphytes.  相似文献

Microalgae are a promising feedstock for renewable biodiesel production. High productivity of biodiesel production from microalgae is directly related to growth rate as well as lipid content of cells. In the present study, an enrichment process in a continuous cultivation system was developed to screen a high-growth-rate microalga from a mixed culture of microalgal species; Chlorella vulgaris, Chlorella protothecoides, and Chlamydomonas reinhardtii were used as test organisms for our experiments. The time-dependent washout of mixed microalgal pool was executed to successfully enrich the C. reinhardtii, which exhibits the higher growth rate than C. vulgaris and C. protothecoides under turbidostat conditions within 75 h. The domination of C. reinhardtii in the mixed culture was validated by on-line monitoring of growth rate and flowcytometric analysis. For the time-efficient production of microalgal biomass, this screening process has a high potential to segregate the fast-growing microalgal strains from the pool of various uncharacterized microalgal species and random mutants.  相似文献

Nutrient limitation commonly constrains organisms in natural ecosystems. Typically, ecologists focus on limitation by N and P. However, other nutrients can limit growth or reproduction. Here we focus on K limitation of invertebrate consumers (Daphnia dentifera) and phytoplankton in freshwater lakes. All organisms require K for several metabolic processes. In freshwater, K could limit growth because low external concentrations can increase the energetic costs of accumulating K. Furthermore, in a study linking K to disease, we previously found that K enrichment of water from one low-K lake stimulated the growth and reproduction of Daphnia. Here we test whether K could limit the production of Daphnia and phytoplankton across lakes and years. We repeated a life table experiment using water collected from a low-K lake during a different year. K again stimulated Daphnia reproduction. We also enriched water from 12 lakes with K or P and measured short-term growth of Daphnia and the resident algal community. Both nutrients increased Daphnia growth in five lakes. However, only P enhanced algal production. P stimulation of Daphnia positively correlated with algal quantity and the ratio of C to P in seston. However, K stimulation of Daphnia was not correlated with these factors or the background concentration of K. Thus, this study shows repeatable K-limited animal physiology in nature. Further, we can exclude the hypothesis that K stimulates Daphnia indirectly by enhancing algal production. These patterns call for future physiological studies to uncover the mechanistic basis of K limitation in natural systems.  相似文献

Nonobligate bacterial predator strain N-1 was highly resistant to copper. In fact, it required more than minimal amounts of copper to initiate growth, but not for growth that followed growth initiation. Strain N-1 made a peptide growth initiation factor (GIF) to marshal copper from its environment for growth initiation. Production of this GIF occurred before the onset of growth initiation, but production was shut down if excess copper was present. At high copper levels, the time required for onset of growth initiation was directly related to the amount of copper that was present. At low copper levels, a similar graded response occurred for increments of added GIF. Agromyces ramosus is a predator in its own right, but it also is a prey species for strain N-1. A. ramosus was found to be very sensitive to copper and to the copper GIF produced by N-1. It is possible that the copper GIF is the means used by N-1 to kill A. ramosus.  相似文献

DszB desulfinase activity reached a sharp maximum at the early exponential phase, but it rapidly decreased at later growth phases. The characterization of a novel Pseudomonas strain exhibiting antagonism towards many important corn fungal pathogens is presented. This strain was isolated from the caryopses of the grass Tripsacum dactyloides and was identified as Pseudomonas cepacia. The antagonistic activity is due to the production of an antifungal compound. The chromatographic properties of this partially purified compound isolated from growth medium differ from those reported previously for other pseudomonads. The suppression of the growth of economically important phytopathogens by this strain and by the partially purified compound indicates a potential biocontrol agent.  相似文献

Plant-plant interactions influence how forests cope with climate and contribute to modulate species response to future climate scenarios. We analysed the functional relationships between growth, climate and competition for Pinus sylvestris, Quercus pyrenaica and Quercus faginea to investigate how stand competition modifies forest sensitivity to climate and simulated how annual growth rates of these species with different drought tolerance would change throughout the 21st century. Dendroecological data from stands subjected to thinning were modelled using a novel multiplicative nonlinear approach to overcome biases related to the general assumption of a linear relationship between covariates and to better mimic the biological relationships involved. Growth always decreased exponentially with increasing competition, which explained more growth variability than climate in Q. faginea and P. sylvestris. The effect of precipitation was asymptotic in all cases, while the relationship between growth and temperature reached an optimum after which growth declined with warmer temperatures. Our growth projections indicate that the less drought-tolerant P. sylvestris would be more negatively affected by climate change than the studied sub-Mediterranean oaks. Q. faginea and P. sylvestris mean growth would decrease under all the climate change scenarios assessed. However, P. sylvestris growth would decline regardless of the competition level, whereas this decrease would be offset by reduced competition in Q. faginea. Conversely, Q. pyrenaica growth would remain similar to current rates, except for the warmest scenario. Our models shed light on the nature of the species-specific interaction between climate and competition and yield important implications for management. Assuming that individual growth is directly related to tree performance, trees under low competition would better withstand the warmer conditions predicted under climate change scenarios but in a variable manner depending on the species. Thinning following an exponential rule may be desirable to ensure long-term conservation of high-density Mediterranean woodlands, particularly in drought-limited sites.  相似文献

The global increase in surface air temperature has produced an overall lengthening of the growing season by 3–5 days/decade in the Northern Hemisphere during the last 30 years. The direct impact of a longer growing season has not been well documented for Sphagnum moss communities in the Arctic. We hypothesized that an increase in the growing season length may be detrimental to Sphagnum growth as a result of photoinhibition caused by the plants emerging from snow near the seasonal peak of solar irradiance. We conducted an experiment from 1999 to 2002, lengthening the growing season in arctic Alaska, to determine the effects that this simulation of climate change had on the growth of hollows dominated by Sphagnum angustifolium. The lengthened growing season was associated with a decrease in annual moss height increment of 78 and 69 % for 1999 and 2000, respectively. These growth reductions may be related to freeze/thaw episodes and prolonged periods of cold in those years. The growth of individuals exposed to snow removal was also reduced by high global radiation. Overall, snow removal did not significantly affect the seasonal dynamics of growth, but seasonal patterns of growth strongly differed among years. These differences in seasonal dynamics suggest that Sphagnum growth is driven by opportunistic responses to favorable conditions rather than ontogenetic drivers. In addition, we compared environmental variation and growth between control and snow removal plots. Growth of Sphagnum in both treatments was stimulated by warmer soil temperatures and drier conditions. With earlier snowmelt as a result of warmer temperatures, it is likely that S. angustifolium will be subjected to higher levels of radiation and possibly greater photoinhibition which may lead to lower growth rates and significant implications for moss production in tussock tundra.  相似文献

A comparative study of growth cone morphology in cultured embryonic neurons derived from wild type PS1(+/+) and knockout PS1(?/?) mice has been performed. Growth cones from wild type PS1(+/+) mice were well spread and usually formed radially continuous and regular lamellar extensions, short filopodia. In contrast, most growth cones from knockout PS1(?/?) mice collapsed after 3–4 days in culture. Summarizing these data, we suggest that PS1 plays an important role in growth cone structure by stabilizing the integrity of the cytoskeleton. The growth cone collapse may be the main reason of abnormal neuronal migration and impaired synaptic function in PS1(?/?) mice.  相似文献
