The monophyly of the litostomes was tested using nine newly sequenced and four previously unpublished small subunit ribosomal RNA (SSrRNA) gene sequences from free-living Haptoria as well as from endosymbiotic Trichostomatia: the vestibuliferids Balantidium coli and Isotricha prostoma, the cyclotrichiid Mesodinium pulex, and the haptorids Loxophyllum rostratum, Dileptus sp., Enchelyodon sp., Enchelys polynucleata, Epispathidium papilliferum (isolates A and B), Spathidium stammeri, Arcuospathidium muscorum, Arcuospathidium cultriforme, and the unusual Teuthophrys trisulca. Phylogenetic analyses depicted the litostomes as a monophyletic group consisting of the trichostomes (subclass Trichostomatia) and the free-living haptorians (subclass Haptoria). The cyclotrichiids Mesodinium and Myrionecta (order Cyclotrichiida) branched either basally within or outside the Litostomatea. In most analyses, the haptorians did not receive support as a monophyletic group. Instead, Dileptus branched basally to all litostome taxa, and Epispathidium papilliferum grouped with the Subclass Trichostomatia. Some subgroupings, however, of haptorian genera corresponded to suggested superfamilial taxa (e.g., orders Spathidiida and Pleurostomatida). Within the monophyletic trichostomes, we can distinguish three clades: (1) an Australian clade; (2) the order Entodiniomorphida; and (3) the order Vestibuliferida. However, Balantidium, currently classified in the Vestibuliferida, did not group with the other vestibuliferids, suggesting that this order may be paraphyletic.  相似文献

The phylogenetic relationships of members of the ciliate class Litostomatea were determined by a molecular phylogeny using the small subunit of the ribosomal RNA (ssu-rRNA) gene and a morphological phylogeny based on ultrastructural analyses of the group. Molecular analyses consistently supported the monophyly of Trichostomatia, Entodiniomorphida and the "Australian" trichostomes but provided limited support for a monophyletic Vestibuliferida and Haptoria. The results of the morphological analyses depended on the way in which the dataset was treated: "unordered" and "ordered" recovered a monophyletic Trichostomatia, Haptoria and the "Australian" trichostomes but challenged the monophyly of Entodinimorphida and Vestibuliferida; "dollo" recovered a monophyletic Trichostomatia and Entodiniomorphida but at the cost of a greatly longer tree than either "unordered" or "ordered" datasets. The monophyly of each "Australian" trichostome family was supported in all analyses and by both approaches. These results suggest that the trichostome ciliates may have become associated with mammals in Gondwana with the "Australian" trichostome ciliates entering Australia with primitive herbivorous marsupials. Subsequent diversification of the "Australian" families was probably a result of dietary specialization and oral and cortical synapomorphies define each family. We decline at this time to erect a formal taxon name for the "Australian" trichostomes due to the instability of other superfamilial taxa within the Litosomatea and concerns about the stability of tree topology until a better taxon sample of litostome ciliates is available.  相似文献

Bozasella gracilis n. sp. in the order Entodiniomorphida was found in fecal samples of an Asian elephant kept in a zoo. The ciliate has general and infraciliary similarities to the families Ophryoscolecidae and Cycloposthiidae. Phylogenetic trees were inferred from 18S rRNA gene sequences of B. gracilis, 45 entodiniomorphids, 10 vestibuliferids, 5 macropodiniids, and an outgroup, using maximum likelihood, Bayesian inference, and neighbor joining analyses. Of them, there were 32 new sequences; 26 entodiniomorphid species in the genera, Bozasella, Triplumaria, Gassovskiella, Ditoxum, Spirodinium, Triadinium, Tetratoxum, Pseudoentodinium, Ochoterenaia, Circodinium, Blepharocorys, Sulcoarcus, Didesmis, Alloiozona, Blepharoconus, Hemiprorodon, and Prorodonopsis, and 6 vestibuliferid species in the genera, Buxtonella, Balantidium, Helicozoster, Latteuria, and Paraisotricha. Thirty additional sequences were retrieved from the GenBank database. Phylogenetic trees revealed non-monophylies of the orders Entodiniomorphida and Vestibuliferida, the suborders Entodiniomorphina and Blepharocorythina, and the families Cycloposthiidae and Paraisotrichidae. Bozasella gracilis was sister to Triplumaria. In addition, to avoid homonymy, we propose Gilchristinidae nom. nov., Gilchristina nom. nov. and Gilchristina artemis (Ito, Van Hoven, Miyazaki & Imai, 2006) comb. nov.  相似文献

The evolution of sophisticated differentiations of the gastro-intestinal tract enabled herbivorous mammals to digest dietary cellulose and hemicellulose with the aid of a complex anaerobic microbiota. Distinctive symbiotic ciliates, which are unique to this habitat, are the largest representatives of this microbial community. Analyses of a total of 484 different 18S rRNA genes show that extremely complex, but related ciliate communities can occur in the rumen of cattle, sheep, goats and red deer (301 sequences). The communities in the hindgut of equids (Equus caballus, Equus quagga), and elephants (Elephas maximus, Loxodonta africanus; 162 sequences), which are clearly distinct from the ruminant ciliate biota, exhibit a much higher diversity than anticipated on the basis of their morphology. All these ciliates from the gastro-intestinal tract constitute a monophyletic group, which consists of two major taxa, i.e. Vestibuliferida and Entodiniomorphida. The ciliates from the evolutionarily older hindgut fermenters exhibit a clustering that is specific for higher taxa of their hosts, as extant species of horse and zebra on the one hand, and Africa and Indian elephant on the other hand, share related ciliates. The evolutionary younger ruminants altogether share the various entodiniomorphs and the vestibuliferids from ruminants.  相似文献

Various echinoderms are colonized by species from several classes of the Phylum Ciliophora, indicating that the echinoderm "habitat" has been invaded independently on numerous occasions throughout evolutionary history. Two "echinoderm" ciliates whose phylogenetic positions have been problematic are Licnophora macfarlandi Stevens, 1901 and Schizocaryum dogieli Poljansky and Golikova, 1957. Licnophora macfarlandi is an endosymbiont of the respiratory trees of holothuroids, and S. dogieli is found in the esophagus of echinoids. A third species, Lechriopyla mystax Lynch, 1930, is a plagiopylid ciliate found in the intestine of echinoids. Host echinoderms were collected near the Friday Harbor Laboratories, San Juan Island, WA. Specimens of S. dogieli and L. mystax were obtained from the esophagus and intestine, respectively, of the sea urchin Strongylocentrotus pallidus. Specimens of L. macfarlandi were collected from the fluid obtained from the respiratory trees of Parastichopus californicus. Using small subunit ribosomal RNA (SSrRNA) sequences of these three ciliates and a global alignment of SSrRNA sequences of other ciliates, we established the following. 1) Licnophora is a spirotrich ciliate, clearly related to the hypotrichs and stichotrichs; this is corroborated by its possession of macronuclear replication bands. 2) Lechriopyla is the sister genus to Plagiopyla and is a member of the Class Plagiopylea, which was predicted based on its cytology. 3) Schizocaryum clusters in the Class Oligohymenophorea and is most closely related to the scuticociliates; there are currently no morphological features known to relate Schizocaryum to the scuticociliates.  相似文献

The cortical development during binary fission of the relatively poorly known stichotrich ciliate, Trachelostyla pediculiformis (Cohn, 1866) Borror, 1972, found in coastal waters near Qingdao, China, was investigated using the protargol impregnation method. The morphogenetic process reveals some pretty unusual characteristics, which do not follow the Oxytricha-pattern: (1) the parental oral apparatus is entirely renewed from an oral primordium formed de novo in the proter; (2) in the proter, the parental undulating membranes are not involved in the formation of the newly formed oral primordium; both undulating membrane-anlagen (UM-anlage) and frontoventral-transverse cirral anlagen (FVT-anlagen) develop from the oral primordium in the proter; (3) the dorsal kineties (DK) are generated in a unique way, that is, in both dividers, two separate groups of DK-anlagen develop in the right- and left-most DK, generate all the DK and evolve to replace the old structures; (4) three caudal cirri are formed at the posterior ends of three right-most dorsal kinety anlagen; (5) eight frontal, five ventral and five transverse cirri are derived from six streaks, namely, the UM-anlage and 5 FVT-anlagen; the cirri are segregated from these anlagen in the pattern 1:3:3:3:4:4 (from left to right) in the Oxytricha mode. Based on both SSrRNA gene sequencing and morphogenetic data, the systematic positions of the genus Trachelostyla Borror, 1972 as well as the family Trachelostylidae Small and Lynn, 1985 are briefly analyzed. The results indicate that this genus/family could be a highly isolated lineage and might be ancestral to other well-known oxytrichids.  相似文献

Laboea strobila Lohmann, 1908 is a conspicuous oligotrich ciliate in the marine plankton. In order to compare different populations, the morphology of specimens from the Mediterranean Sea, North Sea, and Irish Sea was investigated using live observation, protargol impregnation, and scanning electron microscopy. Furthermore, the PCR-amplified products of the SSrRNA gene from a monoclonal culture of L. strobila from the Mediterranean Sea were sequenced and aligned with sequences of other oligotrichs, including a population of L. strobila from the Atlantic coast of the USA. Finally, the data from the ecological literature were summarized and the cultivation methods were described. The SSrRNA gene sequences of the two distantly located L. strobila populations from the North Atlantic are identical. Likewise, the morphometrics of most populations so far investigated after protargol impregnation (i.e. from the North Atlantic) do not show obvious differences. In all computed phylogenetic trees, L. strobila groups with Strombidium species, forming a monophyletic taxon corresponding to the subclass Oligotrichia. These results are corroborated by the ontogenetic comparison. Since no type species was fixed for Laboea Lohmann, 1908, L. strobila was designated in the present paper.  相似文献

从12种限制性内切酶中筛选出6种可获得种间多样性的内切酶:Alu Ⅰ,Taq Ⅰ,Hae Ⅲ,Hinf Ⅰ,Msp Ⅰ,Xba Ⅰ,对广义双眉虫Diophrys-complex5个种(伪寡毛双眉虫Diophrys apoligothrix、悬游双眉虫D.appendiculata、盾圆双眉虫D.scutum、秀丽拟双眉虫Paradiophrys irmgard、针毛类双眉虫Diophryopsis hystrix)共7个种群的核糖体基因(18S小亚基、部分23S大亚基及其内转录间隔区域)进行多位点酶切。结果显示,种间差异明显大于种内差异。利用RAPDistance1.04软件构建的邻接树表明,盾圆双眉虫的3个种群表现出高度的同源性;3个近缘属(双眉虫属、拟双眉虫属、类双眉虫属)可以被明确区分,并支持类双眉虫属与拟双眉虫属的独立性。从GenBank/EMBL数据库中获得广义双眉虫及相近种的小亚基单位核糖体RNA(SSrRNA)基因序列,利用邻接法(NJ),贝叶斯法(Bayesian)和最大简约法(MP)构建的系统发生树具有基本一致的拓扑结构,结果显示:狭义双眉虫属Diophrys为单源发生系,并与类双眉虫属Diophryopsis组成姐妹群;尽管拟双眉虫属Paradiophrys具有广义双眉虫典型的形态学特征,但与尾刺虫属Uronychia有着较近的亲缘关系。本工作同时表明,核糖体DNA限制性酶切(ARDRA)技术可靠地区分纤毛虫的形态相似种,并在双眉虫属间水平的系统关系推定中存在一定的适用性。  相似文献

ABSTRACT. Three complete 18S ribosomal RNA gene sequences from the rumen ciliates, Entodinium coudatum (1,639 bp), Epidinium caudarum (1,638 bp), and Polyplastron multivesiculatum (1,640 bp) were determined and confrimed in the opposite direction. Trees produced using maximum parsimony and distance-matrix methods (lest squares and neighbour-joining). with strong bootstrap support, depict the rumen ciliates as a monophyletic group, Entodinium caudatum is the earliest branching rumen ciliate. However, Entodiniwn simplex does not pair with En. caudatum , but rather with Polyplastron multivesiculatum. Signature sequences for these rumen ciliates reveal that the published SSrRNA gene sequence from En. simplex is in fact a Polyoplastron species. The free-living haptorian ciliates, Loxophyllum, Homalozoon and Spathidium (Subclass Hoptoria), are monophyletic and are the sister group to the rumen cilates. The litostomes (class Litostomatea), consisting of the haptorians and the rumen ciliates, are also a monophyletic group.  相似文献

ABSTRACT. The cortical development of Diophryopsis hystrix , a small marine euplotid ciliate, was observed during binary division. The pattern of morphogenesis is generally similar to that of other Diophrys- like species, with the exception of the marginal cirrus, which develops in a unique manner in D. hystrix being formed from the posterior end of the dorsal kinety anlage, in a similar manner to the formation of the caudal cirri. In the small subunit (SS) rRNA gene sequence tree, Diophryopsis represents a distinct clade separate from Diophrys . Furthermore, our ontogenetic and SSrRNA gene sequence data both support the suggestion that the Diophrys complex is sister to Uronychia , and should be considered as a distinct subfamily within the family Uronychiidae, i.e. Diophryinae Jankowski, 1979, comprising Diophrys , Diophryopsis , and Paradiophrys . Diagnoses of the subfamilies Diophryinae and Uronychiinae are provided.  相似文献

Endobiotic ciliates of native Yakut horse are investigated. 57 ciliate species have been found. From 17 up to 43 species of ciliates have been revealed in each host specimen. The specimens of Alloiozona trizona Hsiung, 1930 and Triadinium caudatum Fiorentini, 1890 were present in 100 % of the horses. The endemic genus and species of allantosomides, Strelkowella urunbasiensis Kornilova, 2004, has been found in 11 horses. Allantoxena japonensis (Imai, 1979) and Cycloposthium ishikawai Gassovsky, 1919 being recorded previously in Japan only were found for the first time in Russia. The species Cycloposthium ponomarevi Kornilova, 2001 peculiar to the Turkmenistan koulan has been found in the Yakut horses.  相似文献

Morphological research on over 50 species of ciliates recorded as endosymbionts of echinoids suggests that invasion of the echinoid microhabitat occurred on at least 4 occasions. Gene sequence data confirm the phylogenetic distinctness of spirotrichean, armophorean, plagiopylean, and oligohymenophorean endosymbionts. It is also likely that oligohymenophoreans have repeatedly invaded the gut habitat. To test this hypothesis, we sequenced small subunit rRNA (SSrRNA) genes of 6 species representing the larger scuticociliate species found in the intestine of Strongylocentrotus pallidus from the northeast Pacific Ocean: Entodiscus borealis (Entodiscidae); Plagiopyliella pacifica and Thyrophylax vorax (Thyrophylacidae); and Entorhipidium pilatum, Entorhipidium tenue, and Entorhipidium sp. (Entorhipidiidae). SsrRNA genes were amplified by PCR, and sequences obtained in both directions. In all phylogenetic analyses, the scuticociliates are well supported as a clade. Entodiscus is distinct from these other echinoid taxa and is the sister taxon to the facultatively parasitic Uronema marinum. The other 5 echinoid species always form a clade whose basal species is the free-living Parauronema longum. The greatest genetic distance among these latter 5 species is less than 1.5%. This probably explains why the Thyrophylacidae and Entorhipidiidae are paraphyletic based on the SSrRNA gene sequences.  相似文献

The small subunit rRNA (SSrRNA) gene was sequenced for Cardiostomatella vermiforme, a large marine ciliate the systematic position of which is uncertain but which has been regarded as a scuticociliate for about forty years. The present work indicates that this organism, together with a closely related species, Dexiotrichides pangi, always form a separate assemblage as a sister group to the scuticociliates sensu stricto. The fact that the clade comprising Cardiostomatella and Dexiotrichides branches between the typical scuticociliates and Hymenostomatia, and shares a series of morphological and morphogenetical characters with both, supports the conclusion that it belongs to an intermediate group between the two. We suggest that this group represents a new order, Loxocephalida n. ord. within the subclass Scuticociliatia, which possibly contains all taxa within the families Loxocephalidae and Cinetochilidae and with Loxocephalidae as the type family.  相似文献

The morphologically unique ciliate Psammomitra has long been considered as a systematically uncertain stichotrich. This is mainly because of its highly specialized morphology and a lack of either detailed information concerning its ontogenesis, or molecular data. Based on the small subunit rRNA (SSrRNA) gene and alpha‐tubulin gene sequences, we re‐evaluated the phylogenetic position of Psammomitra retractilis using multiple algorithms. Phylogenetic trees inferred from the SSrRNA gene sequences representing a total of 53 spirotrichs demonstrated the closest relationship of Psammomitra was with Holosticha‐like taxa, with strong support, which clearly suggested that Psammomitra should be placed into the order Urostylida although it branched at a rather deep level, and is likely to be closely related to Holostichidae. With consideration to molecular evidence and morphological characters, Psammomitra should be a clearly outlined taxon at about the rank of family, i.e. Psammomitridae stat. nov. , within the order Urostylida. The improved diagnosis for this family is as follows: Urostylida possessing extremely contractile, elongated body which consists of three parts: head, trunk, and slender tail; midventral complex composed of midventral pairs only and restricted to about anterior 1/3 of ventral surface; frontal, frontoterminal, and transverse cirri present; one left and one right marginal rows which commence near proximal end of adoral zone and extend to near rear body end.  相似文献

Three new ciliate species presumably belonging to the family Paraisotrichidae were recognized in the fecal samples from zoo-kept Asian and African elephants. All the ciliates possess a unique but similar arrangement of somatic ciliature, thus a new genus Latteuria n.g. has been created for them. The genus is characterized by the presence of a tapered frontal ‘spout’ at the anterior end of the body, posterior ciliary rows in narrow grooves encircling the posterior half of the body and an anterior arch of cilia. Latteuria polyfaria n.sp. is the largest species in the genus with 9–11 posterior ciliary rows. In L. media n.sp., of medium body size, the number of rows varies from four to six, and the smallest species, L. trifaria n.sp., has only three-four posterior ciliary rows.  相似文献

Observations are reported on seasonal changes in the age structure of populations of nematodes of the family Trichonematidae recovered at regular intervals from the lumens of the large intestines of horses slaughtered in S.W. England. The results show that changes in the size of parasitic populations of Trichonema nassatum follow seasonal variations in the rate of infection, more individuals maturing during summer/autumn than during winter/spring because of proportional differences in the numbers of infective larvae ingested from the pasture. In contrast, larvae of T. longibursatum, T. catinatum and T. goldi ingested by grazing horses during summer accumulate in the gut wall rather than promptly returning to the lumen and developing to maturity. Their development is apparently inhibited until the following spring when 4th-stage larvae emerge en masse and quickly reach the adult stage. The relationship between fluctuations in the size of the adult populations of the 4 species studied and the characteristic seasonal variations in horse faecal egg counts is discussed.  相似文献

This review presents the chemistry of secondary metabolites isolated from the Cystoseiraceae and its contribution to the identification of species belonging to the major genera of this family. It completes Piattelli's review on the chemistry and taxonomy of Sicilian Cystoseira species which was presented at the Sixth International Symposium on Marine Natural Products, held in Dakar, 1989 [Piatelli, M. (1990) New J. Chem. 14, 777–782]. In particular, the chemical studies of Cystoseira species from Atlantic and some other Mediterranean Coasts are reported with those of other genera from the Cystoseiraceae family, such as Bifurcara, Halidrys and Cystophora. This review is limited to secondary metabolite—diterpenoids essentially—isolated from the lipid extract of studied species (literature up to January 1995).  相似文献

An autoinoculative device was used to test the ability of sap beetles (Coleoptera: Nitidulidae) to carry a specific strain of Beauveria bassiana (Balsamo) Vuillemin to overwintering sites in a multiyear field study. The device was baited with the pheromone and coattractants for the dusky sap beetle (Carpophilus lugubris Murray) and placed in the field in the fall of each year. The introduced strain occurred at high frequency among the B. bassiana isolated in the fall of all four years tested (100% of all isolates from 21 of 22 collection dates). The introduced strain of B. bassiana was isolated at high frequency from all the B. bassiana-contaminated sap beetles recovered from the overwintering traps (100% of all isolates from 13 of 23 trap/beetle species combinations) and was highest after the longest fall exposure. The introduced strain was primarily isolated from C. lugubris and C. antiquus, but species distribution was also dependent on the overwintering trap design used. Few non-sap beetles species of insects were recovered from the artificial overwintering sites. Although B. bassiana was isolated from free flying sap beetles caught in traps in the spring of each year, none were infected with the introduced strain. The autoinoculating device provides selective contamination of sap beetles in overwintering sites when used in the fall. It may be useful in providing some control of sap beetles or other insects where limited numbers of mass overwintering sites (such as tree holes) occur.  相似文献

Two endemic subterranean freshwater amphipod species have been discovered in groundwater of the volcanic active zone in Iceland, Crangonyx islandicus and Crymostygius thingvallensis. At least five different ciliate sequences were isolated from C. islandicus and the analysis of variation at the 18S ribosomal DNA gene suggests that they present previously unsequenced species, from the orders Apostomatida and Philasterida. Apostome ciliates are well known to be exuviotrophic epibionts on crustaceans. Analyses of ciliate epibionts from different groundwater amphipod species from North America and mainland Europe revealed distinct groups of ciliates and higher diversity on the epigean amphipod Crangonyx pseudogracilis. Analysis of geographic patterns of the most common ciliate within Iceland revealed population differentiation supporting limited current connectivity between the different groundwater systems. Our study reports an occurrence of previously undescribed ciliate species in a groundwater ecosystem characterized by low species diversity.  相似文献

Within carnivorous plants, the bladderworts (Utricularia) possess the most complicated traps whose mechanisms are not yet completely understood. For the first time, a representative survey of different traps from both subgenera (Utricularia and Polypompholyx) is presented.

Based on scanning- and transmission electron microscopy, traps of 14 species of Utricularia (out of 215 species) representing 11 sections (out of 35 sections) and including all life forms (aquatic, epiphytic, and terrestrial) were investigated. Additionally, it was tested whether life forms correlate with trapping mechanisms. Most morphological and anatomical features of the traps vary considerably between the different life forms, e.g. position of trap and trap entrance as well as form and position of trap appendages. Morphological data support the basal position of subgenus Polypompholyx within the genus. Some characteristics of the traps of terrestrial Utricularia multifida (subgenus Polypompholyx) differ remarkably from traps of the other species, e.g. trap-door anatomy and trap walls. This might be an indication for a primordial (non-suction) trapping mechanism in the former species, similar to that of the eel-traps of the closely related genus Genlisea.  相似文献
