The present study was conducted to evaluate the infection rates of Entamoeba histolytica, Entamoeba dispar, and Entamoeba moshkovskii among asymptomatic individuals in Erbil City, northern Iraq. The research intent was to discover whether pathogenic or nonpathogenic species cause a high rate of symptomless Entamoeba infections. Stool samples were microscopically examined, and the 18S-rRNA gene was targeted utilizing the nested PCR technique in the positive specimens. Initial results based on morphological features showed that the Entamoeba prevalence rate was 7.4%. Significantly higher rates of infections were seen in females than in males and in low-income people than in moderate-income people. The incidence rates among the asymptomatic individuals, as determined by molecular analysis, were as follows: E. histolytica – 6%, E. dispar – 4.3%, and E. moshkovskii – 0.3%. Of all the Entamoeba positive samples, a single infection with E. histolytica was identified in 41.4% samples; the single infection with E. dispar in 18.6% samples, 35.7% samples had mixed infections with two Entamoeba species, and 4.3% had mixed infections with three species. The current study concluded that 7.4% of healthy people, who live in the endemic area under investigation, carry Entamoeba species asymptomatically. Additionally, the majority of asymptomatic Entamoeba infections were caused by the pathogenic E. histolytica (81.4%) compared to E. dispar (58.6%), and E. moshkovskii with the lowest rate of infection. Single and co-infections with E. histolytica and E. dispar were noted. DNA content was determined by means of diphenylamine reaction in trophozoites of exponentially growing, axenized Entamoeba histolytica (strains HK-9:NIH, HM-2:IMSS, and HM-3:IMSS), E. invadens (strain PZ), and E. moshkovskii (strain FIC). DNA content was variable in all strains. Variations generally, but not always, occurred within a range characteristic of each species. Average DNA content in strains analyzed was in decreasing order: E. histolytica > E. invadens > E. moshkovskii. Two types of variation were clearly seen in E. histolytica: (i) In one strain (HM-2) the initial content was higher, but, after subculturing it for 6 months (24 passages), the amount of DNA decreased almost four times and became similar to that of the other strains; (ii) a clonal derivative of HK-9 had a small but significant increase and less dispersion in DNA content than the parental strain. The proportion of trophozoites with more than one nucleus was variable; average DNA content per nucleus was slightly smaller than that per trophozoite. We believe that small variations in DNA content may be due to (i) slight changes in ploidy, (ii) genomic heterogeneity, or (iii) differences in the degree of synchrony of the cultures. The ribosomal RNA genes in Entamoeba histolytica are located on circular DNA molecules in about 200 copies per genome equivalent. Nucleotide sequence analysis of the 5.8S rRNA gene and the flanking internal transcribed spacers was carried out to determine the degree of sequence divergence in the multiple rRNA gene copies of a given strain; amongst three different E. histolytica strains (HM-1:IMSS, Rahman and HK-9); and amongst four species of Entamoeba (Entamoeba histolytica, Entamoeba dispar, Entamoeba moshkovskii and Entamoeba invadens). The results show that all rRNA gene copies of a given strain are identical. Few nucleotide positions varied between strains of a species but the differences were very pronounced amongst species. In general, the internal transcribed spacer 2 sequence was more variable and may be useful for strain- and species-identification. The 5.8S rRNA gene and the internal transcribed spacer 2 of E. invadens were unusually small in size.  相似文献

Aims:  To determine the impact of natural sunlight in disinfecting water contaminated with cysts of Giardia duodenalis and Entamoeba histolytica/dispar using plastic containers.
Methods and Results:  Known quantities of Giardia duodenalis and Entamoeba histolytica/dispar cysts in sterile water were exposed to the sun. Containers were made of polyethylene terephthalate, eight painted black on one side, one not painted and another cut open at the top and the last was a high density polypropylene container. Viability testing was performed using vital and fluorescent dyes. The same assays were conducted under cloudy conditions. Thermal control tests were also performed using heat without ultra violet light from the sun. Results show that 99·9% of parasites was inactivated when water temperatures reached 56°C after sunlight exposure.
Conclusion:  Both solar radiation and heat produced by the sun have a synergistic effect in killing cysts of Giardia duodenalis and Entamoeba histolytica/dispar when temperatures rise above 50°C, with complete death at 56°C, using painted 2-l PET containers.
Significance and Impact of the Study:  Solar disinfection system using PET containers painted black on one side can be used to disinfect water against Giardia duodenalis and Entamoeba histolytica/dispar using natural sunlight.  相似文献

A cross-sectional study on the prevalence and morbidity of schistosomiasis was conducted in the main settlement of the municipality of Alhandra, in the southern coastal region of the state of Paraíba, in 2010. The results of this study were compared with the results of a previous study conducted in the same area in 1979. The systematic sampling per family conglomerate included approximately 10% of the resident population in the urban area of Alhandra. Faecal examinations were performed using the Kato-Katz method. The clinical forms of the disease were classified in accordance with FS Barbosa as Type I - intestinal form, Type II - hepatointestinal form and Type III - hepatosplenic form. The prevalence of the infection in 2010 was 10.05%, whereas in 1979 it was 46.6% among untreated patients. The percentages of the three clinical forms in 2010 were as follows: 95.3% Type I, 4.6% Type II and 0% Type III; in 1979, the percentages were 94.4%, 3% and 2.6% for Types I, II and III, respectively. In 1979, 6.07% of the Biomphalaria glabrata specimens (the intermediate host in this area) excreted cercariae, where in 2010 only 1.27% of the specimens caught excreted the parasite.  相似文献

The causative agent of amoebiasis is currently attributed to two distinct species (E. histolytica and E. dispar). The aim of this study was to differentiate these species by PCR in stool samples. Isolated genomic DNA was amplified by PCR and band products of 101 bp (E. dispar) were obtained. All seven stool samples were found to be E. dispar, not E. histolytica. Our results demonstrated the significance of E. histolytica/dispar differentiation in the diagnosis of amoebiasis. This study is preliminary to our current research project entitled "Investigation of the prevalence of amoebiasis and Entamoeba species in Izmir and its hinterland".  相似文献

Entamoeba histolytica Schaudinn, 1903 and Entamoeba dispar Brumpt. 1925 are two of eight species of Entamoeba that sometimes inhabit the human colon. The former is an invasive organism capable of causing life-threatening intestinal and extra-intestinal disease: the latter appears not to be invasive. Because the two species, when viewed by light microscopy appear morphologically similar, they were long regarded as a single species. However, recent biochemical. immunological, and genetic studies provided convincing evidence that they belong to separate species. Our ultrastructural studies revealed distinct differences in at least two features of the trophozoites. 1) The cell surfaces of the trophozoites of each species differ with regard to structures exposed on the surface, and the distribution and arrangement of intra-membranous proteins. 2) The phagocytosis of bacteria differs in respect to the formation of the phagocytic vacuoles. Loose vacuoles containing several bacteria were seen in E. histolytica whereas tight vacuoles containing a single bacterium were observed in E. dispar. Furthermore, bacteria were found only within vacuoles in E. histolytica; in E. dispar, bacteria were found within vacuoles and some were found free in the cytoplasm.  相似文献

In Brazil, malaria remains a disease of major epidemiological importance because ofthe high number of cases in the Amazonian Region. Plasmodium spp infections duringpregnancy are a significant public health problem with substantial risks for thepregnant woman, the foetus and the newborn child. In Brazil, the control of malariaduring pregnancy is primarily achieved by prompt and effective treatment of the acuteepisodes. Thus, to assure rapid diagnosis and treatment for pregnant women withmalaria, one of the recommended strategy for low transmission areas by World HealthOrganization and as part of a strategy by the Ministry of Health, the NationalMalaria Control Program has focused on integrative measures with woman andreproductive health. Here, we discuss the approach for the prevention and managementof malaria during pregnancy in Brazil over the last 10 years (2003-2012) usingmorbidity data from Malaria Health Information System. Improving the efficiency andquality of healthcare and education and the consolidation of prevention programmeswill be challenges in the control of malaria during pregnancy in the next decade.  相似文献

A new biozonation is proposed for the Late Carboniferous strata of the northeastern Paraná Basin (São Paulo and Paraná States), based on well-preserved palynological assemblages recovered from outcrops and core samples retrieved from the Itararé Subgroup. Assemblages include forty-nine species of trilete spores and twenty-five pollen species. The oldest biozone, the Ahrensisporites cristatus Interval Zone (AcZ), in the basal portion of the Itararé Subgroup, includes eleven exclusive spore species, and is assigned a Pennsylvanian (late Bashkirian to Kasimovian) age. The overlying proposed biozone, the Crucisaccites monoletus Interval Zone (CmZ), dated late Pennsylvanian (Kasimovian to Gzhelian), ranges approximately from the top of the lower portion to the middle portion of the Itararé Subgroup, bears only one exclusive species of pollen. Both zones are characterized by the dominance of trilete spores and monosaccate pollen grains, including also few taeniate pollen grains. Furthermore, the basal Protohaploxypinus goraiensis Subzone of the Vittatina costabilis Interval Zone (VcZ) is broadly recognized for the first time in the upper portion of the Itararé Subgroup in the northeastern Paraná Basin. This subzone is attributed to the Early Permian (Early Cisuralian), and is characterized by the dominance of monosaccate pollen grains, a significant increase of taeniate grains (mainly Protohaploxypinus spp.), apart from the appearance of polyplicate pollen (Vittatina spp.). Reference sections and the main characteristics of the two new zones (AcZ and CmZ) are presented, along with a preliminary palynological correlation and discussion on their ages.  相似文献

Typical human immunodeficiency virus-1 subtype B (HIV-1B) sequences present a GPGR signature at the tip of the variable region 3 (V3) loop; however, unusual motifs harbouring a GWGR signature have also been isolated. Although epidemiological studies have detected this variant in approximately 17-50% of the total infections in Brazil, the prevalence of B"-GWGR in the southernmost region of Brazil is not yet clear. This study aimed to investigate the C2-V3 molecular diversity of the HIV-1B epidemic in southernmost Brazil. HIV-1 seropositive patients were ana-lysed at two distinct time points in the state of Rio Grande do Sul (RS98 and RS08) and at one time point in the state of Santa Catarina (SC08). Phylogenetic analysis classified 46 individuals in the RS98 group as HIV-1B and their molecular signatures were as follows: 26% B"-GWGR, 54% B-GPGR and 20% other motifs. In the RS08 group, HIV-1B was present in 32 samples: 22% B"-GWGR, 59% B-GPGR and 19% other motifs. In the SC08 group, 32 HIV-1B samples were found: 28% B"-GWGR, 59% B-GPGR and 13% other motifs. No association could be established between the HIV-1B V3 signatures and exposure categories in the HIV-1B epidemic in RS. However, B-GPGR seemed to be related to heterosexual individuals in the SC08 group. Our results suggest that the established B"-GWGR epidemics in both cities have similar patterns, which is likely due to their geographical proximity and cultural relationship.  相似文献

AIMS: To evaluate the utility of random amplification of polymorphic DNA (RAPD) technique for routine practice in public health laboratories for epidemiological studies of Vibrio cholerae O1 isolates. MATERIALS AND RESULTS: Seventy-nine strains were examined by PCR for the toxin genes (ctx A, zot and ace), virulence-associated genes (tcp A and tox T) and RAPD sequences. Except for one strain (no. 1123) from the Amazonas State, all the strains analysed carried the genes ctx A, zot, ace, tcp A and tox T. RAPD fingerprinting revealed variability but no correlation with serotype, biotype or geographical origin of the isolates was found. CONCLUSION: A standardized RAPD method does not enable the establishment of a pattern data bank for the identification of V. cholerae O1 strains. SIGNIFICANCE AND IMPACT OF THE STUDY: The simplicity and discriminative capacity of this technique make it useful for detecting genetic diversity among micro-organisms from a defined group or for outbreak investigation.  相似文献

The study was carried out in the Private Nature Reservation Nossa Senhora do Outeiro de Maracaípe, located along the southern coast of Pernambuco state, in the Nossa Senhora do Ó District (08°31′48″S and 35°01′05″W), Ipojuca municipality, northeastern Brazil. The reserve has 76.20 ha of tropical coastal vegetation (Restinga forest). The aims were to determine the existence of seasonal phenological pattern of woody species as a function of rainfall. During the study period, the rainfall was 26.8 mm in the driest month (November 2003) and 538.9 in the wettest (June 2004). Observations on flowering, fruiting, leaf fall, and leaf flushing were recorded monthly, during 16 months (April 2003 to July 2004). All species with a ground level perimeter >10 cm and represented by at least three individuals, were selected and tagged. A total of 51 species represented by 156 individuals were recorded. Leaf flushing and leaf fall did not have seasonal variation and the species continued to gain and loose leaves during the dry (August to February) and rainy (March to July) seasons. Flowering, when evaluated by the Fournier's percentage, varied between 1.9% and 20.2%, with maximum intensity in December 2003 (16.4%) and January 2004 (20.2%). Likewise, fruiting varied between 3.8% and 21.4% with the highest percentages in February (17.2%) and March (21.4%) 2004. The analysis comparing reproductive phenological patterns with monthly rainfall did not show a significant correlation, which may suggest investigations concerning other factors, including biotic ones, as pollination and/or dispersal modes, which can be influencing reproductive events of plants in the studied Restinga forest.  相似文献

In order to verify the occurrence of diapause, preference for pupation sites and hymenopteran parasitism, the pupae of the horn fly, Haematobia irritans (Diptera: Muscidae), were collected from undisturbed cattle dung pats in pastures, and adults of the fly were sampled from cattle in São Paulo State, south-eastern Brazil, from April 1993 to July 1994. Diapause was verified in 7.7% of pupae sampled from pastures in June and July of 1993 and in 9.9% of those sampled in May, June and July of 1994 (overall rate of 9.1%). Approximately 8.3% of the pupae were parasitized by microhymenopterans, mostly Spalangia nigroaenea and S.cameroni (Hymenoptera: Pteromalidae). Horn fly pupae were found almost exclusively inside the pat or in the soil immediately beneath and adjacent to it, and very few were collected elsewhere. Pupa mortality was 54.4% and did not change significantly during the year, but mortality was greater among pupae collected in pastures when compared to those obtained from experimental pats, lacking natural enemies.  相似文献

A biostratigraphic study was carried out in the Lower Cretaceous Araripe basin, northeastern Brazil, allowing the recognition of several chronostratigraphic units: the Dom João (Jurassic?-Lower Cretaceous?), the Rio da Serra (Neocomian) and the Alagoas (Aptian/Albian) local stages. For the first time a large hiatus between the Rio da Serra and Alagoas local stages is carefully documented. The palynomorphs and the ostracode associations throughout the Jurassic?-Aptian/Albian sequence allow the interpretation of the paleoenvironmental evolution of the Araripe basin which otherwise confirms that a polycyclical sedimentation occurred in the basin, being one of the controlling factors on the distribution of ostracodes and palynomorphes.  相似文献

A case-control study was conducted to determine the presence of Mycobacterium leprae DNA in nasal secretions of leprosy cases and nonleprosy individuals in Fortaleza, Brazil. It included 185 cases identified by physicians at the Dona Libânia National Reference Centre for Sanitary Dermatology (CDERM). A control group (Co) (n = 136) was identified among individuals from CDERM not diagnosed as leprosy cases. To augment the spatial analysis of M. leprae specific repetitive element (RLEP) positive prevalence, an external group (EG) (n = 121), a convenience sample of healthy students, were included. Polymerase chain reaction for the RLEP sequence was conducted for all participants. Prevalence of RLEP positivity for cases and Co were 69.2% and 66.9%, respectively, significantly higher than for EG (28.1%), and reported elsewhere. Male sex, belonging to a lower socioeconomic status (D/E), history of a previous contact with a case and being older, were associated with being a leprosy case. Our geographical analysis demonstrated that the bacillus is widespread among the healthy population, with clusters of RLEP positive multibacillary cases concentrated in distinct areas of the city. Our results suggest that in endemic areas, as in Fortaleza, surveillance for both nonhousehold leprosy contacts and members of the general population living in cluster areas should be implemented.  相似文献

Entamoeba histolytica is a deep-branching eukaryotic pathogen. Rhomboid proteases are intramembrane serine proteases, which cleave transmembrane proteins in, or in close proximity to, their transmembrane domain. We have previously shown that E. histolytica contains a single functional rhomboid protease (EhROM1) and has unique substrate specificity. EhROM1 is present on the trophozoite surface and relocalizes to internal vesicles during erythrophagocytosis and to the base of the cap during surface receptor capping. In order to further examine the biological function of EhROM1 we downregulated EhROM1 expression by >95% by utilizing the epigenetic silencing mechanism of the G3 parasite strain. Despite the observation that EhROM1 relocalized to the cap during surface receptor capping, EhROM1 knockdown [ROM(KD)] parasites had no gross changes in cap formation or complement resistance. However, ROM(KD) parasites demonstrated decreased host cell adhesion, a result recapitulated by treatment of wild-type parasites with DCI, a serine protease inhibitor with activity against rhomboid proteases. The reduced adhesion phenotype of ROM(KD) parasites was noted exclusively with healthy cells, and not with apoptotic cells. Additionally, ROM(KD) parasites had decreased phagocytic ability with reduced ingestion of healthy cells, apoptotic cells, and rice starch. Decreased phagocytic ability is thus independent of the reduced adhesion phenotype, since phagocytosis of apoptotic cells was reduced despite normal adhesion levels. The defect in host cell adhesion was not explained by altered expression or localization of the heavy subunit of the Gal/GalNAc surface lectin. These results suggest no significant role of EhROM1 in complement resistance but unexpected roles in parasite adhesion and phagocytosis.Entamoeba histolytica is an extracellular protozoan parasite and is a leading parasitic cause of death worldwide (48). The factors, which determine the outcome of amebic infection, are currently unknown, although it is likely that a combination of host and parasite determinants influence clinical outcome. A number of parasite factors required for amebic pathogenesis have been identified, including the Gal/GalNAc surface lectin, pore-forming proteins, and cysteine proteases (36,38, 41).Recently, we identified several members of a class of intramembrane rhomboid proteases in the E. histolytica genome (4). Rhomboid proteases are seven-pass transmembrane proteases first identified in Drosophila melanogaster whose active site lies within the lipid bilayer, allowing them to cleave transmembrane proteins (6, 32). Substrates of rhomboid proteases are largely single-pass transmembrane proteins whose transmembrane domain contains helix-breaking residues (52). Recent work has revealed that there are multiple classes of rhomboid proteases that recognize different types of sequences within the transmembrane domains of their substrates (3). Despite low sequence similarity between individual rhomboid proteases of each class, these enzymes share a remarkable ability to functionally replace one another (16, 28, 52).Rhomboid proteases have been studied in flies, bacteria, mammals, and parasites, and roles ranging from quorum sensing to host cell entry have been identified (3, 11, 25, 33, 35, 46, 47, 49, 54, 59). In apicomplexan parasites, such as Plasmodium falciparum and Toxoplasma gondii, it has been suggested that rhomboid proteases mediate cleavage of surface adhesin proteins to facilitate host cell entry (3, 11, 46, 47). The E. histolytica genome encodes four rhomboid-like genes, with only a single gene containing the necessary catalytic residues for proteolytic activity (4). This gene, EhROM1, is a functional protease with substrate specificity similar to the P. falciparum ROM4 (PfROM4) (3, 4). In trophozoites EhROM1 is localized to the parasite surface and relocalizes to internal vesicles during erythrophagocytosis and to the base of the cap during surface receptor capping. We have shown that the heavy subunit of the amebic surface Gal/GalNAc lectin (Hgl) is a substrate of EhROM1 in vitro. Mutational analyses using a COS cell cleavage assay demonstrated that the cleavage of Hgl requires the catalytic serine in EhROM1 as well as a helix-breaking glycine residue in the transmembrane domain of Hgl (4). These data indicate that EhROM1 is a functional rhomboid protease whose physiological substrate may be Hgl.In order to further elucidate the biological function of EhROM1 we have utilized the epigenetic silencing mechanism of the E. histolytica G3 strain (8, 9). The mechanism of gene silencing in G3 ameba is not well understood. However, it is known that the silencing mechanism is epigenetically maintained, and epigenetic changes in the chromatin state of the silenced genes have been noted (22). G3 parasites transfected with a plasmid containing an upstream region of the 5′ end of EhROM showed almost complete downregulation of expression; we have named these parasites ROM(KD) for ROM knockdown. Phenotypes examined in ROM(KD) parasites included cap formation, complement resistance, adhesion, phagocytosis, hemolysis, and motility. We observed defects in both adhesion and phagocytosis in the ROM(KD) parasites compared to the parent G3 strain but no changes in cap formation or complement resistance. Importantly, the reduced phagocytosis phenotype appears independent of the reduced adhesion phenotype, implying that EhROM1 has distinct roles in both pathways.  相似文献

Protein–protein interactions (PPI) play a key role in predicting the function of a target protein and drug ability to affect an entire biological system. Prediction of PPI networks greatly contributes to determine a target protein and signal pathways related to its function. Polyadenylation of mRNA 3′-end is essential for gene expression regulation and several polyadenylation factors have been shown as valuable targets for controlling protozoan parasites that affect human health. Here, by using a computational strategy based on sequence-based prediction approaches, phylogenetic analyses, and computational prediction of PPI networks, we compared interactomes of polyadenylation factors in relevant protozoan parasites and the human host, to identify key proteins and define potential targets for pathogen control. Then, we used Entamoeba histolytica as a working model to validate our computational results. RT-qPCR assays confirmed the coordinated modulation of connected proteins in the PPI network and evidenced that silencing of the bottleneck protein EhCFIm25 affects the expression of interacting proteins. In addition, molecular dynamics simulations and docking approaches allowed to characterize the relationships between EhCFIm25 and Ehnopp34, two connected bottleneck proteins. Interestingly, the experimental identification of EhCFIm25 interactome confirmed the close relationships among proteins involved in gene expression regulation and evidenced new links with moonlight proteins in E. histolytica, suggesting a connection between RNA biology and metabolism as described in other organisms. Altogether, our results strengthened the relevance of comparative genomics and interactomics of polyadenylation factors for the prediction of new targets for the control of these human pathogens.  相似文献
