Argasid ticks are vectors of viral and bacterial agents of humans and animals. Recent reports indicate that some ornithophilic argasids harbored rickettsial agents. A Nearctic tick, Argas monolakensis Schwan, Corwin, Brown is ornithophilic and has not previously been examined for rickettsial agents. Thirty adult A. monolakensis were tested by PCR for DNA from Rickettsia or Coxiella. Amplicons from a Coxiella sp. that were divergent from Coxiella burnetii were detected in 16/30 A. monolakensis. These molecular isolates were similar but not identical to C. burnetii, the Coxiella spp. of other ticks, and “Coxiella cheraxi” a pathogen of crayfish. Dark-field microscopy was used to determine the number of Borrelia spirochetes in 630 standard preparations obtained from adult ixodid ticks (344 Ixodes persulcatus and 286 I. ricinus) collected in 1989-92 in the Leningrad region of Russia. The average numbers of Borrelia in I. persulcatus and I. ricinus preparations were 34.7 and 23.3 per 100 microscopic fields, respectively. The maximal individual values registered each year for ticks of both species were several hundred times greater than the minimal values. Ticks carrying relatively small numbers of Borrelia generally predominated. Proportions of more heavily infected ticks varied considerably from year to year. These parameters were significantly higher in foci with predominance of I. persulcatus ticks. As a consequence, risk to acquire Lyme borreliosis in such foci is considered greater than in foci where I. ricinus predominates.  相似文献

An intertidal population of Iridaea capensis in the south-western Cape Province of South Africa has been studied in detail for a year. The species is generally restricted to sand-inundated rocky shores, which appears not to be the case for Iridaea species in the Pacific. In early winter (June) biomass, plant density and plant size were at a minimum. A small increase in these parameters occurred in August, but there was a massive increase in plant numbers and biomass in October. The largest plants were recorded in summer (February). The population was over 70% sporophytes in April, but switched to almost 80% gametophytes following heavy recruitment between August and October, and remained significantly gametophyte-dominated through April of the following year. Carrageenan levels were high (ca 42% of dry weight) in late winter-spring, and lowest (ca 30–35%) in summer-autumn. Strong offshore wind conditions in early summer caused a reduction in biomass, and it is hypothesized that the associated upwelling of nutrient rich water may be implicated in the lower carrageenan levels at this time.  相似文献

Ticks play an important role in human and veterinary medicine, in particular due to their ability to transmit a wide spectrum of pathogenic micro-organisms of protozoal, rickettsial, bacterial and viral origin. Pathogens in ticks can be identified by conventional methods such as indirect immuno-fluorescence, isolation in cell culture or by using histological staining techniques. However, the advent of the polymerase chain reaction (PCR) has resulted in tremendous improvements in the specific and sensitive detection of pathogen DNA in ticks. In this paper, literature on DNA extraction methods, PCR protocols, primers and probes, which are in use for the successful detection and identification of pathogens in ticks, are critically reviewed. Some recommendations are also given towards the end of tht review.  相似文献

Elasmobranch fossils recovered from the Fishburne Formation (lower Eocene/Ypresian) of Berkeley County, South Carolina, USA, include species from four genera of sharks and six genera of rays. Of particular interest was the recovery of multiple isolated teeth from a new genus and species of the cownosed ray family Rhinopteridae, which is the focus of this study. The unique crown morphology separates this genus and species from Rhinoptera. Eorhinoptera grabdai, gen. et sp. nov., is represented by small, bar-shaped teeth in the shape of greatly elongated hexagons. These teeth are the isolated elements of a dental plate. The holotype, with 12 wide root lobes, is the most elongated in the sample being 1 cm long and 1.5 mm wide, indicating an origin in the central region of the plate. Paratypes are less elongated, have 4–8 root lobes and are from more lateral rows. The crown is smooth and has a distinctly convex occlusal surface. Eorhinoptera is only the second genus of cow-nosed ray. Its distinctive crown morphology may have allowed it to exploit different kinds of prey than those favoured by rays that lacked convex tooth crowns.  相似文献

The process of oviposition in A. walkerae was observed and found to be a sequence of exactly coordinated, interlocking events independent of the age of ticks. Egg-laying always commenced with numerous dorso-ventral movements of the capitulum. The pedipalps were spread during the last dorso-ventral movement and lowered to the ventral body wall embracing the genital aperture on both sides. Simultaneously, the cuticular sac of Gene's organ emerged and immediately everted maximally. Immediately afterwards the vestibulum vaginae prolapsed from the genital aperture touching the cuticular sac. The vestibulum vaginae handed over an egg to the cuticular sac after a brief contact. Then the vestibulum vaginae invaginated, the pedipalps closed and the cuticular sac was retracted. Finally, the capitulum very frequently moved up and down and the free egg was pushed in front of the ticks. The total time of laying an egg averaged 27min in both 4-week-old and 12-month-old ticks. In 4-week and 12-month-old ticks, egg-laying was preceded by a mean preoviposition period of 6.1 days and 7.5 days, respectively, while the mean oviposition was completed in 14 and 10 days and the average egg totals were 119 and 103.  相似文献

The effects of different temperatures and relative humidities (RHs) were tested on various reproductive parameters of Ornithodoros turicata, an argasid tick that inhabits gopher tortoise burrows in Florida, USA. The pre-oviposition, oviposition and incubation periods of the ticks decreased as temperature increased. These periods were also affected by the RH. The number of eggs oviposited was affected significantly by the combined effect of temperature and RH. Fewer eggs were laid by ticks in the 24°C regimes and the 27°C/95%RH regime compared to those in the other temperature/RH groups. There was an inverse relationship between the number of eggs oviposited and the percentage of hatched larvae that was correlated with the temperature and RH. Ticks reared at 27°C/90%RH and 30°C/90%RH laid more eggs than those reared in the other combinations of temperature and humidity but fewer larvae hatched from these eggs. The reproductive fitness index (RFI) values were highest in females held in the 24°C groups and the 30°C/95%RH group, although significantly more larvae hatched at the lower temperatures. The optimum reproductive conditions for O. turicata under laboratory conditions appear to be 24°C and 90–95%RH. While mating occurred at all temperatures, none of the females laid eggs at 22°C. The ticks may move preferentially to low temperatures when not feeding to remain above the critical equilibrium humidity and/or below the critical metabolic level necessary for prolonged survival. However, most female ticks oviposited after 45 days when moved to 27°C/95%RH. Ornithodoros turicata females may have a limited capability to delay oviposition until an optimal microenvironment for egg deposition can be located in the burrow.  相似文献

Paralysis caused by feeding female Ixodes rubicundus ticks is a major problem in large areas of South Africa. As the life cycle of the tick extends over a period of 2 years, it was hypothesized that strategic treatment of sheep with an acaricide over a 2 year period, timed to kill most engorging females, should markedly lower the biotic potential of the tick. Two flocks of sheep grazing in separate paddocks known to be infested with I. rubicundus were treated either strategically or on a threshold basis (i.e. only when tick challenge exceeded a predetermined critical level in terms of paralysis) for a 2 year period. The tick burdens of untreated control sheep running with the two flocks were monitored over a 4 year period and their seasonal dynamics determined. The times at which peak infestations occurred were similar for both flocks of sheep, but significant differences in mean tick burdens between the two flocks were recorded. Tick numbers on sheep in the strategically treated flock did not decrease during the third and fourth years of the trial as was expected. Possible reasons for this were low stocking densities, especially during times of peak abundance of adults and the presence of wild hosts which maintained tick populations.  相似文献

We collected and tested 616 tropical rat mites (Ornithonyssus bacoti (Hirst)) from rats (Rattus norvegicus (Berkenhout) and R. rattus (Linnaeus)) throughout 14 governorates in Egypt and tested DNA extracts from pools of these mites for Bartonella spp., Coxiella burnetii, and Rickettsia spp. by PCR amplification and sequencing. Three different mite-associated bacterial agents, including one Bartonella and two Rickettsia spp., were detected in eight pools of mites. Further research could demonstrate the vector potential of mites and pathogenicity of these agents to humans or animals. Doxomysis algoaensis sp.nov. is described from Algoa Bay, South Africa where it is common in nearshore marine waters just beyond the breaker line. D. algoaensis sp.nov. is morphologically similar to D. australiensis, but can readily be distinguished by the shape and armature of the telson and the length of the exopod of the fourth male pleopod. The apical cleft is one fifth the telson length in the former species and one third the length in D. australiensis. The telson apex on each side of the cleft is also armed with five and four stout spines in the two species, respectively. In D. australiensis, the exopod of the fourth male pleopod is almost three times the length of the endopod; in D. algoaensis sp.nov., the exopod is only slightly longer than the endopod. Other distinctive features of D. algoaensis sp.nov. include the maxillary palp, which is only slightly broader than long and the greater number of spines on the endopod of the uropod.  相似文献

The prevalence of Borrelia burgdorferi sensu lato in several tick species was studied over a 2 year period in two ecologically different areas in Spain. One area was an endemic area for Lyme disease, with a number of autochthonous human cases and supported large populations of Ixodes ricinus on cattle and birds; the second area was characterized by the absence of I. ricinus together with the presence of foxes and their associated tick species. While I. ricinus was the main vector of B. burgdoreri in the endemic area (with a mean prevalence of 14% in adults and 51% in nymphs), adults of both Ixodes canisuga and Ixodes hexagonus had high rates of B. burgdorferi prevalence (30 and 28%, respectively) in the zone where I. ricinus was absent. Immatures of Ixodes frontalis were found to be carriers of the spirochete only in those zones where I. ricinus is present, suggesting evidence for reservoir competence in a tick-bird cycle.  相似文献

An inhibitor of activated coagulation factor X (fXa) was isolated from salivary gland extracts prepared from Ornithodoros savignyi using a two-step procedure, involving reversed-phase high-performance liquid chromatography (RP-HPLC) and diethylaminoethyl (DEAE) ion-exchange chromatography. From its behaviour during DEAE chromatography it could be deduced that it possesses an acidic pI (4.6). Capillary zone electrophoresis (CZE) of the purified inhibitor showed it to be homogeneous. The molecular mass was determined as 12 kDa using capillary gel electrophoresis (CGE) and as 7183.4 using laser desorption mass spectrometry (LDMS). The N-terminal amino acid sequence (residues 1–12) was determined and found to share a 66% identity with tick anticoagulant peptide (TAP). The O. savignyi peptide is a slow, tight-binding inhibitor of fXa (K i=0.83±0.10 nM). The interaction of the fXa-inhibitor was found to be competitive and dependent on ionic strength. From January 2002 to December 2004, 152 ticks were collected from 40 wild birds recovered in Santo André Natural Reserve and Monsanto Forestal Park, Portugal mainland. Five ticks species were identified from 22 species of birds, and new host record were provided for some species. In addition, 32 (21%) ticks were screened by PCR to detect infections with agents belonging to order Rickettsiales: Anaplasma phagocytophilum, Ehrlichia chaffeensis, and Rickettsia spp. PCR amplicons were obtained in 5 (15.6%) tick samples. Rickettsia DNA exhibiting gltA sequences similar to those of Rickettsia aeschilimannii, R. helvetica and R. massiliae were identified in Hyalomma marginatum, Ixodes ventalloi and in Rhipicephalus turanicus, respectively. This is the first report of rickettsiae infections in ticks collected from wild birds in Portugal. Giving the results presented above wild birds play an important role in the maintenance and dissemination of several tick species and associated rickettsiae.  相似文献

Ticks are ectoparasites that cause considerable damage to their hosts while feeding. The feeding process is facilitated by anti-haemostatic factors present in the tick saliva. Apyrase (ATP diphosphohydrolase, EC 3.6.1.5) is a platelet aggregation inhibitor found in most haematophagous organisms studied. The present study describes the identification and characterization of such an activity in the tick Ornithodoros savignyi. The enzyme conformed to many properties common to apyrases. These included a low substrate specificity, dependence on bivalent metal ions for activity and insensitivity to the classical ATPase inhibitors. Heat denaturation studies, pH optima and similar effects of inhibitors on the enzyme's ATP and ADP hydrolysing activities supported its classification as an apyrase. Salivary gland extracts inhibited the platelet aggregation induced by ADP, collagen and thrombin and disaggregated aggregated platelets. The results suggest the presence of two or more anti-platelet factors present in the salivary glands of this tick species.  相似文献

The present study evaluated the rickettsial infection in Amblyomma parvum ticks collected in Northwestern Córdoba Province, Argentina. Each tick was subjected to DNA extraction and tested by polymerase chain reaction (PCR) targeting fragments of the rickettsial genes gltA and ompB. Nine (69.2%) out of 13 adult ticks yielded expected PCR products for the two rickettsial genes. Products from the ompB PCR were sequenced, generating DNA sequences 100% identical for the nine PCR-positive ticks. Three of these ticks were tested in another battery of PCR targeting fragments of the rickettsial genes gltA, htrA, and ompA. Products from the gltA, htrA, and ompA PCRs were sequenced generating DNA sequences 100% identical for the three PCR-positive ticks. The rickettsia detected in the A. parvum ticks was designated as Rickettsia sp. strain Argentina. Phylogenetic analyses performed with partial sequences of the rickettsial genes gltA, htrA, ompB, and ompA showed that Rickettsia sp. strain Argentina belonged to the spotted fever group, being distinct from all known Rickettsia species and genotypes available in GenBank, representing possibly a new Rickettsia species. This was the first evidence of rickettsial infection in the tick A. parvum, and the third report of rickettsial infection among the Argentinean tick fauna. The role of Rickettsia sp. strain Argentina as a human pathogen is unknown. Further studies are needed to obtain tissue-cultured isolates of Rickettsia sp. strain Argentina, in order to better characterize it and to determine its taxonomic status as a new species.  相似文献

This study examines the effects of ricinoleic acid esters from Ricinus communis castor oil on the vitellogenesis of Rhipicephalus sanguineus ticks attached to hosts that were fed with commercial rabbit food containing these esters. The oocytes of ticks from the treatment group (TG) showed cytoplasmic changes that inhibited the development of oocytes I and II to the advanced stages (IV and V) in addition to preventing the maturation of oocytes V, resulting in small ones. In addition, sperm was not observed in ampoules. Our findings confirm the acaricide potential of ricinoleic acid esters.  相似文献

This study showed the interference of esters extracted from Ricinus communis in the secretory cycle of salivary glands of Rhipicephalus sanguineus ticks, which consequently caused collateral effects on their feeding process. Ticks attached on hosts which were fed with commercial feed containing different concentrations of R. communis oil esters suffered damages such as cytoplasmic changes in their salivary glands, notably in the acinar cells, impairing the functioning of the acini and accelerating the organs degeneration as a whole. It was found that esters interfered with the activity of cellular secretion by changing the glycoprotein of salivar composition especially in acini II cells. It was also shown that the damages caused by esters in the salivary glands cells of these ectoparasites increased in higher concentrations of the product and degenerative glandular changes were more pronounced.  相似文献

In some studies the prevalence of tick infection (infection rate) and the intensity of infection are negatively correlated with unfed tick age (in the broad sense of this term). However, no special research has been carried out to consider the phenomenon thoroughly. The infection indices of the female taiga ticks, Ixodes persulcatus, infected with Borrelia burgdorferi s.l. were related to tick physiological age, an index that more precisely reflects tick physiological state than the time of tick collection in the field or the duration of tick survival under laboratory conditions. A novel quantitative technique of physiological age determination based on the evaluation of the ratios between sizes of the stable (scutum) and the changing (alloscutum) structures of the tick body was used. The age was estimated in accordance with the classical age-grade scale introduced by Balashov and a more fractional scale determined by the new technique. In total, 131 female ticks were examined for their infection and physiological age, 46 of which were infected with B. burgdorferi s.l. (mean infection rate 35.1%). The minimal intensity of infection was 0.4 bacterial cells per 100 fields of view whereas the maximal infection was 172 cells. There was no difference between the prevalence of infection in ticks of different physiological age. The intensity of infection obviously differed between ticks of different age groups in the scale introduced by Balashov but did not significantly differ between ticks of different age groups according to the fractional age-grade scale. The data concerning the relationships between Borrelia burgdorferi and unfed Ixodes ticks are considered.  相似文献

The prevalence of infection with Lyme borreliosis spirochetes in nymphal Ixodes ricinus and Haemaphysalis punctata was examined on three Swedish islands in the Baltic Sea. The proportion of H. punctata nymphs infected with Borrelia spirochetes (2%) was lower than that of I. ricinus nymphs (11–16%), even on the island of Stora Karlsö where both tick species feed on the single mammalian host present, the varying hare, Lepus timidus. Ten of the 12 infected questing H. punctata nymphs harboured very few spirochetes, whereas the remaining two harboured as many as 30–40 spirochetes. However, it remains to be seen whether H. punctata nymphs are capable of transmitting spirochetes during their blood meal and, thus, serving as vectors for Lyme borreliosis spirochetes. On Gotland and Fårö, I. ricinus was more abundant than H. punctata in deciduous and coniferous forest, whereas H. punctata was equally or more abundant than I. ricinus in juniper brush areas on open land. Host preference and biotope usage of H. punctata seems, in general, to prevent this tick from feeding on highly infective Borrelia reservoirs, such as Apodemus mice.  相似文献

Borrelia burgdorferi sensu lato was detected in one out of five cement plugs of female Ixodes persulcatus ticks. The spirochetes were found by dark field microscopy as early as 18 h after attachment of the ticks to the skin of a white mouse. The relevance of this finding is discussed in relation to the epidemiology of Lyme Borreliosis.  相似文献
