The aim of this study was to determine the effects of the exposure of pregnant females of a viviparous teleost Cnesterodon decemmaculatus to sublethal Cd solutions on the survival of their offspring. Ovigerous females were acclimated and accomodated in aquaria containing the following solutions: artificial freshwater medium (AFW) and AFW with 25, 50 and 100 microg Cd(2+) /L (as chloride). Part of the offspring born from control females was transferred to media contain-ing 25, 50 and 100 microg/L of cadmium; the rest of the fry stayed in AFW solutions without cadmium. The offspring born from the females maintained in solutions of cadmium were transferred to aquaria with AFW, and to solutions containing metal at the same concentration as their mother. In all cases mortality was recorded daily for 168 h. The results demonstrated that the survival of newborn offspring was always related to the concentration of Cd(2+) to which they were exposed at birth, regardless of the concentration of the metal to which the mother was exposed during its pregnancy. When exposed to Cd(2+), alevins from non-exposed females showed a constant linear death rate while in those from Cd-exposed it resulted exponential.  相似文献

Newly hatched nymphs of the insect Oncopeltus fasciatus were exposed to various concentrations of CdCl2 administered in drinking water until the end of adult life. Significant nymphal mortalities were observed at concentrations above 30 mg Cd/l (corresponding to the LC50). The duration of the nymphal stages increased in proportion to the Cd concentration; at the lowest Cd concentration of 10 mg Cd/l, the median duration was significantly prolonged by one day, while at the highest concentration of 100 mg Cd/l it was increased by 10 days over the control group. The weight of newly emerged adults lineally decreased with Cd concentration, being reduced to half the weight of controls at 100 mg Cd/l. In addition, a proportionality between delay in development and weight reduction was found in Cd-treated adults. Survival of adult females was decreased at concentrations higher than 10 mg Cd/l, while males were only affected at 30 mg Cd/l or higher doses. Reproduction was the most affected parameter. Oviposition rate, fecundity and fertility of females exposed to 10 mg Cd/l were significantly lower than controls (73%, 58% and 55% relative to controls, respectively). Hatchability of the eggs laid by treated females was also reduced. These results show that development and reproduction of O. fasciatus are seriously impaired at sublethal Cd concentrations.  相似文献

Cadmium removal by living cells of the marine microalga Tetraselmis suecica was tested in cultures exposed to different cadmium concentrations (0.6, 3, 6, 15, 30 and 45 mg/l). The EC50 for growth was 7.9 mg Cd/l after six days of exposure. The cadmium removed was proportional to the concentration of this metal in the medium and it was dependent on the time of exposure; cultures with higher cadmium concentration removed a higher amount of this metal. In cultures exposed to 0.6 mg/l, T suecica cells removed 98.1% of added cadmium with 0.392 x 10(-6) microg Cd/cell, whereas in cultures with 45 mg/l only 7.7% was removed with 16.052 x 10(-6) microg Cd/cell. The highest amount of cadmium removed per liter of culture was observed in cultures exposed to 6 mg/l, with 3.577 mg/l of cadmium. After six days of incubation, the higher proportion of cadmium was bioaccumulated intracellularly in all cultures except in 45 mg/l cultures, the percentage of intracellular cadmium being always more than 50%. The highest percentage of bioadsorbed cadmium (60.1%) was found in cells of cultures with the highest cadmium concentration (45 mg/l). Furthermore, a relation between intracellular cadmium and the concentration of sulfhydryl groups was observed.  相似文献

由于工业活动的影响,墨西哥水体环境中的重金属浓度在上升.浮游动物,尤其是轮虫类,由于对环境变化十分敏感而且是淡水中的常见组成部分,因此被广泛用于生态毒理试验以确定水质标准.在不同的胁迫途径下(如通过培养基或食料),重金属的毒性是不同的.在本研究中,通过在轮虫Brachionus rubens的培养基和食料中添加重金属这两种途径,我们评估了镉和铅的效应.对于这两种重金属,均采用将轮虫置于含0.5×106个/ml绿藻的培养基中或每天喂食经5倍于LC50值的金属处理(1,2和4h)的绿藻.对于在培养基中添加镉,使用了三个毒性水平(0.1,0.2和0.4 mg/L),铅的浓度分别为0.005,0.010 和0.015 mg/L.基于LC50的数据,B.rubens对铅的敏感性要比镉高24倍.镉浓度为0.4 mg/L时,培养基中加入镉造成B.rubens的生长趋缓.而喂食经不同时间处理的绿藻后,轮虫的密度随着食料在重金属中处理时间的延长而减小.培养基中或食料中添加铅时,轮虫种群生长的的趋势与在镉处理下的情况类似.随着培养基中重金属浓度的增加,每天种群增长率(r值)会减小.在培养基和食料处理两种不同途径下,r值会在0.33(对照)到0.02 d-1(经重金属处理)间变化[动物学报 51(1)46-52,2005].  相似文献

To assess the impact of enhanced root vacuole cadmium (Cd) sequestration on leaf Cd accumulation under a low Cd dose, as generally occurs in agriculture, leaf Cd accumulation was examined in field-grown tobacco plants expressing genes encoding the high-capacity-Cd, tonoplast-localized, divalent cation/H antiporters AtCAX4 and AtCAX2 (AtCAX, Arabidopsis cation exchanger). It has been shown previously that root tonoplast vesicles isolated from plants expressing these genes, directed by root-selective promoters, show enhanced Cd transport activity, and young plants show enhanced root Cd accumulation when grown in solution culture containing 0.02 µ m Cd, a moderate Cd dose. In this article, we present results which show that the lower leaves of mature plants expressing AtCAX2 or AtCAX4 , under the control of two different root-selective promoters, accumulate 15%–25% less lamina Cd than control plants when grown in the field (3 years, three different collection methods). Reciprocal grafting experiments of AtCAX2 shoots onto control roots (and vice versa), grown in solution culture with 0.005 µ m Cd, indicated that the root controls Cd translocation and accumulation in the shoot in control and AtCAX2 and AtCAX4 tobacco plants exposed to low Cd concentration. The results are consistent with a model in which supplementation of Cd/H antiporter activity in root cell tonoplasts enhances root Cd sequestration, resulting in decreased translocation of Cd to the shoot of field-grown plants. These results suggest that human Cd intake from food and tobacco use could be reduced via the enhancement of root vacuolar sequestration of this pollutant.  相似文献

The effect of cadmium (Cd) on the thyroid and parathyroid glands was examined following a 12-week exposure of rats to 5 or 50 mg Cd/dm3. Immunohistochemical methods were used to determine calcitonin (CT), calcitonin-gene related peptide (GGRP), parathormone (PTH), somatostatin (ST), synaptophysin (SPh) and neuron-specific enolase (NSE). Calcium and cadmium concentrations in the femoral bone were assayed. The animals exposed to cadmium showed attenuation of all reactions, which was most distinct after 50 mg Cd/dm3. Exposure-dependent cadmium accumulation and a decrease in calcium concentration in the femoral bone were noted.  相似文献

采用暴露实验方法,研究了海水中镉在牙鲆内脏、肌肉、鳃组织内蓄积及在清洁海水中排放规律,以及海水中总有机碳(TOC)浓度对镉吸收的影响.结果表明,镉暴露浓度为0.5 mg·L-1时,牙鲆各组织镉蓄积量随暴露时间增加而增大,第10天达吸收平衡,Cd蓄积量顺序为内脏(147.73 mg·kg-1)>鳃(15.55 mg·kg-1)>肌肉(4.578 mg·kg-1),表明内脏是重金属镉蓄积的主要部位.染毒13 d后,将牙鲆移入清洁海水中,结果表明,随排放时间增加,各组织镉蓄积量明显下降,排放至第15天时各组织镉排出率为内脏团(80.66%)>鳃(73.66%)>肌肉(56.84%),表明受镉污染牙鲆经清洁海水排放后,有望达到食用标准.海水中TOC浓度对镉蓄积量有明显影响.随着TOC浓度升高,各组织内镉蓄积量明显下降,表明海水TOC能降低镉的生物有效性.  相似文献

The effects of latent iron deficiency combined with parenteral subchronic or acute cadmium exposure during pregnancy on maternal and fetal tissue distribution of cadmium, iron and zinc, and on fetal viability were evaluated. Timed-pregnant Sprague-Dawley rats were fed on semisynthetic test diets with either high iron (240 mg kg) or low iron (10 mg kg), and concomitantly exposed to 0, 3 or 5 mg cadmium (as anhydrous CdCl2) per kilogram body weight. Animals were exposed to cadmium from gestation day 1 through 19 by subcutaneously implanted mini pumps (Subchronic exposure) or on gestation day 15 by a single subcutaneous injection (Acute exposure). All rats were killed on gestation day 19. Blood samples, selected organs and fetuses were removed and prepared for element analyses by atomic absorption spectrometry. Low iron diet caused decreases in maternal body weight, maternal and fetal liver weights, placental weights and tissue iron concentrations. By cadmium exposure, both subchronic and acute, tissue cadmium concentrations were increased and the increase was dose-related, maternal liver and kidney zinc concentrations were increased, and fetal zinc concentration was decreased. Cadmium concentration in maternal liver was additionally increased by low iron diet. Acute cadmium exposure caused lower maternal body and organ weights, high fetal mortality, and decreased fetal weights of survivors. In conclusion, parenteral cadmium exposure during pregnancy causes perturbations in essential elements in maternal and fetal compartments. Acute cadmium exposure in the last trimester of gestation poses a risk for fetal viability especially when combined with low iron in maternal diet.  相似文献

Accumulation and effects of cadmium were investigated in Chironomus thummi larvae exposed to 10, 100 and 250 mug radiolabeled Cd/1 for up to 4 days. (1) After 4 days, average cadmium accumulation was 6.6 ng Cd/mg dry weight (10 mug Cd/1 exposure) and 177 ng Cd/mg dry weight (250 mug Cd/1 exposure). (2) Dissection studies showed that by 32 hr of exposure to both cadmium concentrations, 63.5-81.4% of accumulated cadmium was confined to the posterior midgut epithelium. Light microscope autoradiography similarly showed accumulations of cadmium in posterior midgut epithelium and smaller amounts in fat body and muscle. Little cadmium was associated with Malphigian tubules, haemocoel, anterior midgut or exoskeleton. (3) After exposure to 10 or 250 mug Cd/1, 60-75% of cadmium in ultracentrifuged homogenates of whole animals or dissected guts was associated with the resulting supernatant. When supernatants were further analyzed by gel chromatography, cadmium eluted with both a high and low molecular weight peak. The relative proportions of cadmium in the two peaks varied with concentration and length of exposure. (4) Transmission electron microscopy of posterior midgut cells from animals exposed to cadmium demonstrated frequent mitochondrial lesions. Exposure to high cadmium concentrations caused some posterior midgut cells to undergo generalized structural degeneration.  相似文献

The resultant effects of cadmium exposure are seen in almost all the systems of the body, however, this study is designed to quantify its accumulation in tissues of animals exposed to cadmium. The rats were divided into two distinct groups of males and females, which were then divided into three groups, each for the monitoring of exposure. Group 1 served as control male and female and received normal rat chow and tap water. Group 2 males and females were treated with 5 mg/kg body weight of cadmium chloride (Cd) intraperitoneally for eight days while Group 3 males and females rats received 100 ppm of Cd in drinking water for 18 days. The accumulation of cadmium in rat liver cadmium binding protein induced by single and repeated intraperitoneal injections of CdCl2 and the de novo biosynthesis of CdBP were studied by using 109Cd to measure cadmium binding in the CdBP and 35S incorporation as indicator of protein synthesis. The biosynthesis of CdBP is controlled by the cadmium concentrations. For single doses up to 1 mg Cd2+/Kg b.w. about 50% of the cadmium is present in the soluble fraction of liver bound to CdBP and the incorporation of 35S-cysteine is linear with the cadmium concentration. When single doses ranging from 1 to 3 mg Cd2+/Kg b.w. are administered the fractions of both 35S-cysteine and cadmium incorporated into de novo synthesized CdBP gradually decrease. For single doses higher than 3 mg Cd2+/Kg b.w. the biosynthesis capability is maximum and 20 mug Cd/g liver can be incorporated into the de novo biosynthesized CdBP. When rats are treated every day with amounts of cadmium of about 0.8 mg Cd2+/Kg b.w. for up to 8 days a dose-proportional increase in both Cd incorporation and CdBP biosynthesis are observed. This shows a cumulative incorporation of cadmium in the de novo biosynthesized CdBP. Experiments carried out by injecting 65ZnCl2 and 203HgCl2 every day showed that they are not accumulated like cadmium and do not induce the biosynthesis of rat liver CdBP after repeated administration over 7 days.  相似文献

为研究不同性别桑树在镉(Cd)与酸雨(AR)复合处理条件下的生长与镉积累差异,本试验以‘强桑1号’(雌株)和‘农桑14号’(雄株)两种不同性别桑树为材料,研究重金属镉(100 mg·kg-1干土)和模拟酸雨(pH 3.0)处理下,两者生物量积累和分配、最大净光合速率(Amax)和叶绿素荧光参数,以及Cd积累与分配的差异.试验共设置无Cd和AR(对照)、单独Cd、单独AR、Cd+AR复合4个处理.结果表明:单独Cd处理虽然显著增加了雌株和雄株根茎叶中Cd含量,而且雌株各部分Cd含量显著高于雄株,但是只有雄株根、茎和总生物量、最大净光合速率显著下降,而雌株除了Amax显著下降外,各部分生物量积累下降均不显著,且两者的最大光化学效率(F_v/F_m)、光化学淬灭系数(q_P)和非光化学淬灭系数(q_N)均未发生显著变化.复合处理下,雄株和雌株的总生物量、Amax均显著降低,与单独镉处理相比,雌株根和叶中的Cd含量显著升高,但雄株没有明显变化,两者的F_v/F_m、q_P没有显著变化,但q_N均显著降低.短时间处理下桑树雌株对单独镉胁迫的耐受性高于雄株,但是酸雨的加入会降低雌株对镉的耐受性,可能是因为酸雨促进雌株根系对镉的吸收,并导致叶片积累了更多的镉.  相似文献

Dexamethasone was administered orally for 7.5 or 10 d to each of 20 pregnant bitches beginning at an estimated 28 to 51 d of gestation, using 1 of 2 dose regimens. Five bitches were given dexamethasone 3 times a day for 10 d, with the highest dose of 0.2 mg/kg for 5 d and then at progressively decreasing doses of 0.16-0.02 mg/kg for 5 d. The 15 remaining bitches were given dexamethasone 2 times a day for 7.5 d, increasing from 0.1 to 0.2 mg/kg over the first 3 administrations, then remaining at 0.2 mg/kg on Days 2 to 5, and decreasing from 0.16 to 0.02 mg/kg over the last 5 administrations. The side effects, including mild polydipsia and polyuria, disappeared when treatment was discontinued. Depending on the stage of pregnancy, uterine contents were either resorbed or aborted, or both. Pregnancy was terminated within 2 to 16 d after the start of treatment in all treated bitches, at 2 to 5 d of treatment in 2 of 3 bitches treated at 40 to 51 d of pregnancy, and at 0 to 4 d after the end of treatment in most of the 17 bitches treated at 28 to 35 days of pregnancy. Oral administration of dexamethasone appears to be a potentially useful pharmacologic treatment for the termination of unwanted pregnancy in the bitch.  相似文献

The aim of this study was to evaluate the effect of ethylenediaminetetraacetic acid (EDTA) on cadmium (Cd) induced renal dysfunction. Seventeen workers (14 males, 3 females) were diagnosed with occupational Cd poisoning in 1986. These individuals had between 7 to 39 years of Cd exposure. From 1986 to 1999, patients received periodic EDTA therapy as part of their follow-up, all at the same hospital. Levels of urinary cadmium (UCd) and urinary beta2-microglobulin (B2M) were measured before and after each annual EDTA treatment period. Renal dysfunction was defined as urinary B2M > 0.8 mg/g Cr (creatinine). In these workers, patients with UCd level higher than 10 microg/g Cr in 1986 had abnormal B2M excretions (> or = 0.8 mg/g Cr) or trended to have abnormal B2M levels during the treatment period. However, in subjects with UCd concentration lower than 10 microg/g Cr in 1986, their urinary B2M excretions either remained normal (< 0.8 mg/g Cr) or returned to normal during the treatment period. The prevalence of renal dysfunction increased during the follow up period regardless of whether UCd levels increased or not, indicating a progressive renal dysfunction despite removal from Cd exposure. Our results suggest that reversibility of renal dysfunction caused by Cd related to the level of Cd exposure at the time of removal from exposure: renal dysfunction could be reversed if initial UCd < 10 microg/g Cr, but was irreversible when UCd > 10 microg/g Cr. Repeated examinations on these 17 Cd exposed workers from 1986 to 1999 also revealed that periodic administration of EDTA had no beneficial effects on chronic Cd-induced renal dysfunction.  相似文献

在实验条件下,将黑斑蛙暴露于12.5mg/L和25.0mg/L浓度的镉溶液中30d,分别测定了黑斑蛙在暴露10、20和30d时肝、肾组织中镉(Cd)含量、过氧化产物丙二醛(MDA)的含量、还原型谷胱甘肽(GSH)含量和超氧化物歧化酶(SOD)活性,以探讨镉对机体的脂质过氧化作用及机体的抗氧化损伤机制。实验结果表明,不同剂量组黑斑蛙肝、肾镉含量、MDA含量均随着镉暴露时间的延长而升高,且肝MDA含量与镉在肝中的蓄积量呈显著正相关(R^2=0.8643,n=9)。肝脏GSH含量随镉暴露时间的延长而被显著诱导,且与MDA含量呈显著正相关(R^2=0.5933,n=9);肾GSH含量则随暴露时间的延长而显著下降,与MDA含量呈显著负相关(R^2=0.8609,n=9)。不同剂量组肝SOD活性随镉暴露时间的延长而升高,肾SOD活性在高剂量组随镉暴露时间的延长表现为先升高后回落下降的趋势。可见,在镉的长期暴露下,细胞膜过氧化增强是镉伤害机体的主要原因,而GSH含量、SOD活性的升高则可能是机体抗过氧化的机理之一。  相似文献

The effects of acute exposure to cadmium (Cd) on the blood antioxidant defense system, lipid peroxide concentration and hematological parameters, as well as the possible protective role of vitamin E were studied. Male Wistar albino rats (3 months old) were treated with cadmium (0.4 mg Cd/kg b.m., i.p., 24 h before the experiment) or with vitamin E + Cd (20 IU Vit E/kg b.m., i.m., 48 h + 0.4 mg Cd/kg b.m., i.p., 24 h before the experiment). The hematological parameters were assessed: red blood cell counts, hematocrit value and hemoglobin concentration were significantly decreased in the blood of Cd-treated rats. Intoxication with cadmium was also followed by significantly increased lipid peroxide concentrations. We also observed increased activity of antioxidant defense enzymes: copper zinc containing superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase and glutathione-S-transferase as well as concentrations of non-enzymatic components of antioxidant defense system: reduced glutathione, vitamin C and vitamin E. Pretreatment with vitamin E exhibited a protective role on the toxic effects of cadmium on the hematological values, lipid peroxide concentration as well as on enzymatic and non-enzymatic components of antioxidant defense system.  相似文献

The objective of this study was to examine the influence of oral supplementation with Zn or Mg on Cd content in the blood and organs of rabbits exposed to prolonged Cd intoxication. Rabbits were divided into the following groups: Cd group-received orally every day for 4 weeks 10 mg Cd/kg body weight (b.w.), Cd+Zn group and Cd+Mg group-exposed to Cd and supplemented with 20 mg Zn/kg b.w. or 40 mg Mg/kg b.w. 1 h after Cd treatment. Cd content in biological material was determined by atomic absorption spectrophotometry. Blood Cd concentration was determined in all investigated groups at time 0 and after 10, 14, 18, 22, 25, and 28 days, whereas Cd content in the brain, heart, lungs, liver, kidney, spleen, pancreas, skeletal muscle, and bone was determined after 28 days. Blood Cd concentration was significantly increased in all groups from the 14th day of Cd intoxication and lasted till the end of the experiment. Zn or Mg supplementation significantly reduced blood Cd content on the 18th and 25th days. Supplementation with Zn or Mg significantly decreased Cd concentration in the kidney, spleen, and bone and, in addition, Zn reduced Cd content in the brain. Supplementation with Zn or Mg in Cd-intoxicated rabbits caused similar reduction of blood Cd concentration; however, reduction of tissue Cd content was more pronounced in Zn- than in Mg-supplemented group.  相似文献

Four groups of goldfish were exposed to cadmium in a concentration of 20 mg Cd/l water under aquarium conditions. The duration of exposure was 1, 4, 7 and 15 days. It was shown that the activity of superoxide dismutase (SOD) in the red blood cells (RBC) significantly decreased after the first day of cadmium exposure. However, the SOD activity increased after 7 and 15 days of cadmium treatment. Elevated activity of catalase (CAT) was found in erythrocytes of cadmium-treated fishes after 15 days, whereas plasma GOT levels was increased after 7 and 15 days and GPT levels after 1, 4, 7 and 15 days of cadmium treatment. This was accompanied by a significant decrease of blood hemoglobin concentrations (after 15 days) and hematocrit values (after 7 and 15 days). However, the concentration of blood glucose significantly increased after 1, 4, 7 and 15 days of cadmium exposure. These results indicate that cadmium causes oxidative stress and tissue damage in the exposed fishes.  相似文献

Cadmium is well known for its toxicity to the animal body. However, its effect on pregnancy and the development of young animals is still not well understood. This study examined such effects, using bank voles captured from the wild to make the results closer to those which could be expected in the natural environment. One group of animals was fed 7 microg g(-1) cadmium in the food, a second 35 microg g(-1), and a third no cadmium, as a control. The concentrations of cadmium in the whole bodies of young bank voles were determined on the 3rd, 5th, or 10th day of life. The cadmium level in the bodies of animals exposed to 35 microg g(-1) of cadmium was significantly higher than in those from either the control group or the group receiving 7 microg g(-1) of cadmium, which did not differ from each other. The cadmium level did not change with animal age in any of the study groups. Concentrations of Zn, Cu, and Fe were also determined in the whole body of young animals, as cadmium is known to disturb the metabolism of these essential metals through antagonistic activity. Both Cu and Fe levels were negatively correlated with cadmium concentrations, while a positive correlation was found between zinc and cadmium in the young animal bodies. Also found was higher offspring mortality in the group receiving 35 microg g(-1) of cadmium in food. There was no difference in young animal body weight between the study groups.  相似文献

The aim of this study was to assess the effects of subchronic exposure to cadmium (Cd) on the antioxidant defense system of red blood cells (RBCs) and lipid peroxide concentration in the plasma, as well as the possible protective role of zinc (Zn). For this purpose, 60 male Wistar rats (8 weeks old) were divided into three groups: the first group was exposed to Cd in the form of CdCl2, administered in five doses (each of 0.4 mg Cd/kg BW) on days 5, 10, 15, 20 and 25, giving a total dose of 2 mg Cd/kg BW, i.p.; the second group was simultaneously exposed to Zn and Cd with the same timeline and the same doses of Cd as the first group but with, in addition, injections of Zn in the form of ZnCl2, administered in doses of 0.8 mg Zn/kg BW, giving a total dose of 4 mg Zn/kg BW, i.p.; a control group received 0.5 mL of physiological saline in an identical manner.

It was shown that exposure to Cd induced a significant decrease (p<0.05) in superoxide dismutase (Zn/Cu SOD) and catalase (CAT) activities in RBCs. Increased lipid peroxide concentration, measured by thiobarbituric acid reactive substances (TBARS), was also observed in the plasma of cadmium-exposed rats. Cd had no effect on glutathione peroxidase (GSH-Px) activity. Zn administration had a beneficial effect on the Cd-induced decrease in Zn/Cu SOD activity (p<0.05) but not on CAT activity. Animals receiving Cd and Zn simultaneously had significantly (p<0.05) lower concentrations of lipid peroxides than rats exposed to Cd alone. Our results indicate that Cd causes oxidative stress and that Zn supply in conditions of exposure to Cd can partially protect against Cd-induced oxidative stress.  相似文献
