Extracellular catalases produced by fungi of the genusPenicillium, i.e.,P. piceum, P. varians, andP. kapuscinskii, were purified by consecutive filtration of culture liquids. The maximum reaction rate of H2O2 decomposition, the Michaelis constants, and specific catalytic activities of isolated catalases were determined. The operational stability was characterized by the effective rate of catalase inactivation during enzymatic reaction (k in at 30°C). The thermal stability was determined by the rate of enzyme thermal inactivation at 45°C (k in * , s-1). Catalase fromP. piceum displayed the maximum activity, which was higher than the activity of catalase from bovine liver. The operational stability of catalase fromP. piceum was twofold to threefold higher than the stability of catalase from bovine liver. The physicochemical characteristics of catalases of fungi are better than the characteristics of catalase from bovine liver and intracellular catalase of yeastC. boidinii.  相似文献

对我国古老特有植物青檀叶片进行内生和附生真菌的研究,以了解青檀叶片内生和附生真菌的组成特点和探讨内生和附生真菌菌群之间的可能联系,为研究真菌资源多样性、植物附生和内生真菌的相互演化关系及真菌与宿主植物协同进化等提供有益参考资料。研究结果表明,从健康的青檀叶片获得可培养内生真菌839株,附生真菌1857株,共计2696株,鉴定其分属于4目,5科,43属。在目的分类水平上,内生和附生真菌均以丛梗孢目Moniliales为优势菌群,分别占90.23%和92.51%;在科的水平上,内生真菌以暗梗孢科Dematiaceae和丛梗孢科Moniliaceae为优势菌群,分别占47.56%和42.67%,附生真菌以丛梗孢科Moniliaceae和暗梗孢科Dematiaceae为优势菌群,分别占67.04%和25.47%;在属的水平上,内生真菌以黑团孢属Periconia和青霉属Penicillium为优势菌群,分别占31.47%和10.73%,附生真菌以小球霉属Glomerularia、膝葡孢属Gonatobotrys和青霉属Penicillium为优势菌群,分别占20.03%、13.95%和12.22%。青檀叶片内生真菌和附生真菌均存在的菌群数量达到23个属,占53.49%。内生真菌特有的属有6个,共分离19株,占0.70%,附生真菌特有的属有14个,共分离120株,占4.45%。内生真菌的Shannon-Wiener index(H')多样性指数(2.44)和Margalef index(R)丰富度指数(2.88)分别小于附生真菌ShannonWiener index(H')多样性指数(2.57)和Margalef index(R)丰富度指数(3.32),但两者的Evenness index(E)均匀度指数几乎相等。青檀叶片内生和附生真菌菌群组成具有较高的相似性,相似性系数达0.70。通过Fisher's exact test分析表明青檀叶片内生和附生真菌菌群组成无明显差异(P=0.072)。  相似文献

Fungi have been only rarely isolated from glacial ice in extremely cold polar regions and were in these cases considered as random, long-term preserved Aeolian deposits. Fungal presence has so far not been investigated in polar subglacial ice, a recently discovered extreme habitat reported to be inhabited exclusively by heterotrophic bacteria. In this study we report on the very high occurrence (up to 9000 CFU L−1) and diversity of filamentous Penicillium spp. in the sediment-rich subglacial ice of three different polythermal Arctic glaciers (Svalbard, Norway). The dominant species was P. crustosum, representing on the average half of all isolated strains from all three glaciers. The other most frequently isolated species were P. bialowiezense, P. chrysogenum, P. thomii, P. solitum, P. palitans, P. echinulatum, P. polonicum, P. commune, P. discolor, P. expansum, and new Penicillium species (sp. 1). Twelve more Penicillium species were occasionally isolated. The fungi isolated produced consistent profiles of secondary metabolites, not different from the same Penicillium species from other habitats. This is the first report on the presence of large populations of Penicillium spp. in subglacial sediment-rich ice.  相似文献

A total of 28 strains of 19 Penicillium species were isolated in a survey of extracellular enzyme-producing fungi from macroalgae along the coast of Jeju Island of Korea. Penicillium species were identified based on morphological and β-tubulin sequence analyses. In addition, the halo-tolerance and enzyme activity of all strains were evaluated. The diversity of Penicillium strains isolated from brown algae was higher than the diversity of strains isolated from green and red algae. The commonly isolated species were Penicillium antarcticum, P. bialowiezense, P. brevicompactum, P. crustosum, P. oxalicum, P. rubens, P. sumatrense, and P. terrigenum. While many strains showed endoglucanase, β-glucosidase, and protease activity, no alginase activity was detected. There was a positive correlation between halo-tolerance and endoglucanase activity within Penicillium species. Among 19 Penicillium species, three species–P. kongii, P. olsonii, and P. viticola–have not been previously recorded in Korea.  相似文献

The increasing world demand for fuels makes it necessary to exploit the largest reserve of extra-heavy crude oil (EHCO) of the Orinoco Oil Belt from Venezuela. We propose the use of extracellular oxidative enzymes, in particular, lignin-degrading enzyme systems (LDS) of fungi, for enzymatic improvement of EHCO. Autochthonous non-white rot fungal strains able to use EHCO, and several polycyclic aromatic hydrocarbons (PAHs) as sole carbon source and energy, were isolated from EHCO-polluted soils and identified as belonging to the genera Fusarium, Penicillium, Trichoderma, Aspergillus, Neosartorya, Pseudallescheria, Cladosporium, Pestalotiopsis, Phoma and Paecillomyces. Phenotypic and biochemical assays revealed the ability of these filamentous fungi to synthesize extracellular oxidative enzymes, and suggested a relationship between the LDS and EHCO bioconversion. This work reports, for the first time, the use of o-phenylenediamine dihydrochloride (OPD) as substrate to measure extracellular ligninolytic peroxidases (ELP) in culture broths of filamentous fungi (Fusarium solani HP-1), and constitutes the first formal study of the fungal community associated with the EHCO of the Orinoco Oil Belt.  相似文献

The predominant fungi present in samples of reject and retail red kidney beans were Aspergillus glaucus, Penicillium spp. and Alternaria spp. Together with A. ochraceus, A. flavus, Fusarium spp., and Trichoderma, these isolates from the reject beans were screened for numerous mycotoxins by TLC. The most consistently produced mycotoxins were penicillic acid (from A. ochraceus and Penicillium spp.) and Alternaria toxins (tenuazonic acid and alternariol). A. glaucus strains were tested for cytotoxicity in three tissue culture cell lines with positive results.  相似文献

The distribution of microorganisms in four soil samples taken near Casey Station, Wilkes Land, and on Dewart Island, Frazier Islands (East Antarctica), was studied using isolation cultures at different temperatures. The fungal assemblages comprised Penicillium, Alternaria, Cladosporium, Phoma, Verticillium, Phialophora, Candida and Rhodotorula. Microalgal Chlorophyta were also common. Among the bacteria representatives of cyanobacteria, Staphylococcus, Bacillus, Pantoea, Streptomyces, Micromonospora, and coryne- and nocardioform species were found. Some of the isolated actinomycete strains, determined as Arthrobacter, Rhodococcus, Streptomyces and Micromonospora, produced extracellular substances with antibacterial and antifungal activities. Their potential as biological agents against phytopathogenic bacteria and fungi is discussed.  相似文献

Fourteen species of keratinophilic fungi belonging to ten genera (Chrysoporium, Malbranchea, Chaetomium,Sepedonium, Microascus, Scopulariopsis, Curvularia, Fusarium, Aspergillus, Penicillium) were isolated from feathers of about one hundred living poultry birds. The isolated fungi were compared for their keratinase activity after growing them on two different media: (1) basal salts solution containing natural keratin (human hair) as the only source of carbon and nitrogen; (2) the medium was supplemented with a minor amount of readily assimilable source of carbon along with natural keratin. All the test fungi could grow on keratinous material, degrading it and releasing sulphydryl containing compounds detected as cysteine, total proteins and extracellular keratinase. Maximum enzyme release by these fungi occurred in the broth supplemented with glucose and vitamins, thereby indicating a correlation between the mycelial biomass and production of proteolytic keratinases. Summary Six strains of fungi: two Aspergilli of the A. glaucus group, Penicillium sp. Trichoderma viride, and two unidentified dark fungi, were found able to use thiourea as a source of nitrogen. One of the Aspergilli and the Penicillium showed the best utilization, but the growth was always slow, and no more than 50% (usually much less) of the thiourea-nitrogen was recovered in the mycelium.Assimilation of thiourea-nitrogen by Aspergillus and Penicillium was accompanied by oxidation of the sulphur to sulphate in roughly approximate stoichiometric proportion. The same fungi were able to produce sulphate from cystine as combined source of nitrogen and sulphur.  相似文献

A wide range of fungi, amounting to fifty-six species belonging to twenty-two genera, have been recovered from animal feeds and foodstuffs.The most frequent fungi were Aspergillus niger, and A. flavus, followed by Mucor racemosus, Alternaria alternata, Rhizopus stolonifer, Penicillium corylophilum and P. notatum. Three genera were found to be of moderate occurrence, namely, Mucor, Rhizopus and Alternaria. The three following genera were of low occurrence: Cladosporium, Fusarium, and Neurospora.The fluorescence method of detecting aflatoxin-producing strains demonstrated that only one isolate of A. flavus possesses this property. Certain species of Penicillium and Aspergillus produced fluorescent substances (metabolites) similar in color to B and G aflatoxin. These substances were subsequently proved not to be aflatoxin by (TLC) chromatography.The animal and public health significance from such toxins was also discussed.  相似文献

Out of 36 strains of fungi of the genus Penicillium, some of which were isolated from ancient permafrost, 14 strains synthesized mycophenolic acid (MPA). Maximal (over 500 mg/l) accumulation of MPA in culture liquid was observed in P. brevicompactum strains (VKM F-457, VKM F-477, and VKM F-1150). This was the first study to detect MPA in representatives of the species P. rugulosum; in three strains of this species (VKM FW-665, VKM FW-717, and VKM FW-733), the level of MPA accumulation exceeded 300 mg/l. The time course of the synthesis of MPA by P. rugulosum strain VKM FW-733 was studied. Production of extracellular catalase by microscopic mycelial fungi (255 strains) belonging to different taxonomic groups was studied. Producers of extracellular catalase were found among fungi of the genera Penicillium, Talaromyces and Aspergillus. Strains of the genus Penicillium were the most active producers. The formation of catalase depended on the initial pH, carbon and nitrogen sources and their ratio, and the content of microelements in the medium. The yield of extracellular catalase produced by the strains selected (P. chrysogenum, P. funiculosum, P. pinophilum, and P. minioluteum) was not less than 400-1400 U per ml culture liquid.  相似文献

The genus Penicillium is one of the largest and widely distributed fungal genera described to date. As a result, its taxonomic classification and species discrimination within this genus has become complicated. In this study, 52 isolates that belonged to the Penicillum genus and other related genera were characterized using two DNA-based methods: (i) analysis of the nucleotide sequences of internal transcribed spacers in ribosomal DNA and (ii) analysis of DNA fingerprints that were generated by polymerase chain reactions with specific primers for enterobacterial repetitive intergenic consensus (ERIC) and repetitive extragenic palindromic (REP) sequences, and BOX elements. Using both methods, Penicillium species were discriminated from other fungal genera. Furthermore, Penicillium species that include strains which are used as biocontrol agents, such as P. glabrum, P. purpurogenum, and P. oxalicum, could be distinguished from other Penicillium species using these techniques. Based on our findings, we propose that a polyphasic approach that includes analysis of the nucleotide sequences of ribosomal DNA and detecting the presence of highly conserved, repeated nucleotide sequences can be used to determine the genetic relationships between different Penicillium species. Furthermore, we propose that our results can be used as a start point to develop a strategy to monitor the environmental presence of particular strains of Penicillium species when they are used as biocontrol agents.  相似文献

Toxigenic saprophytic fungi were isolated from samples of shellfish, sediment and seawater obtained from marine shellfish farming areas. The 456 strains identified included 12 different genera, with a clear predominance (68%) of Penicillium, Aspergillus, Trichoderma and Cladosporium. To assess the risk of poisoning due to the presence of these fungi in shellfish farming areas, the strains were cultured in liquid medium, filtered, and tested on larvae of Artemia salina, a small crustacean highly sensitive to mycotoxins. Thirty-five point five percent of the strains proved active with this test. This study confirms the existence of fungi in shellfish farming areas, as suggested by our earlier work showing that filter-feeding shellfish accumulate toxic metabolites of fungal origin. The presence of fungi in the marine environment represents a real risk of poisoning through the consumption of contaminated shellfish.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

【背景】松树的内生真菌会影响红脂大小蠹及其伴生真菌的生长及扩散,从而影响红脂大小蠹的入侵。【目的】掌握赤峰地区红脂大小蠹寄主树种内生真菌的物种多样性,筛选对其伴生菌有拮抗作用的菌株,为红脂大小蠹的生物防治提供资源。【方法】采用组织分离法、形态学鉴定和internal transcribed spacer (ITS)序列分析相结合的方法对红脂大小蠹寄主树种油松、樟子松和潜在寄主落叶松进行内生真菌多样性研究,并用两点对峙法进行拮抗菌株筛选。【结果】松树内生真菌鉴定为2门6纲10目18科19属,其中落叶松韧皮部分离到的内生真菌数量最多,为39株,隶属于9属12种,真菌检出率为43.33%;油松次之,为30株,隶属于7属8种,真菌检出率为33.33%;樟子松最少,为29株,隶属于10属13种,真菌检出率为32.22%。3个树种的内生真菌相似性较低,无共有的菌种,青霉属(Penicillium)和篮状菌属(Talaromyces)是唯二的共有菌属,且青霉属在3个树种中均为优势菌属。平板对峙结果表明90%以上的树木内生真菌均能够与伴生真菌形成稳定的对峙,抑制率在50%-86%之间,且Phialocephala sp.和Pochonia bulbillosa对伴生菌的抑制率能高达93.7%。【结论】松树韧皮部的内生真菌具有较高的生防潜力,Phialocephala sp.和P. bulbillosa对红脂大小蠹伴生菌有较好的抑制效果,可作为红脂大小蠹的生防资源。  相似文献

A number of Penicillium isolates were recovered in association to Rhizoctonia solani strains pathogenic on tobacco and from soil on plates pre-colonized by the pathogen itself. Their antagonism toward R. solaniAG-2-1 was evaluated in dual cultures in vitro. Inhibition of growth was evident to some extent in most pairings, while hyphal interactions referable to mycoparasitic relationships were not observed. However, the occurrence of plasmolysis and/or vacuolisation and the induction of monilioid cells were indicative of the release of bioactive compounds. Therefore, production of fungitoxic metabolites was tested by adding concentrated culture filtrates of each Penicillium isolate to the growth medium of R. solani. Complete and lasting inhibition was incited by culture filtrates of some isolates belonging to P. brevicompactum, P. expansum, and P. pinophilum. Three purified compounds, respectively mycophenolic acid, patulin and 3-O-methylfunicone, which were extracted from culture filtrates, were able to inhibit R. solani in vitro. Their production was also detected in dual cultures of the same Penicilliumstrains with R. solani prepared in sterilized soil and when the Penicilliumstrains were cultured directly on R. solani mycelium harvested from liquid cultures. The possible role of such metabolites in antagonism of the above-mentioned Penicilliumspecies against R. solani is discussed.  相似文献

A correlation was established between species specificity and synthesis of specific secondary metabolites by the Penicillium fungi. Strains of the subgenus Aspergilloides usually synthesize metabolites of polyketide nature. Most strains of the subgenus Furcatum produce clavine ergot alkaloids and metabolites of diketopiperazine nature. The only clavine ergot alkaloids and diketopiperazine alkaloids produced by strains of the subgenus Biverticillium are rugulovasines and rugulosuvines, respectively. Species designations of the strains of the subgenus Penicillium isolated from permafrost soil, the Mir orbital complex, and sites undergoing anthropogenic load were refined based on the marker secondary metabolites. Changes in the taxonomic position of some strains in the genus Penicillium are suggested.  相似文献

Species of Penicillium and Trichoderma were found to dominate the rhizosphere of established tea bushes in a detailed study conducted from various tea growing locations in India. Penicillium erythromellis, P. janthinellum, P. raistrickii, Trichoderma pseudokoningii and T. koningii were found to be closely associated with tea roots. While seasonal fluctuation was observed in the case of Penicillium spp., the population of Trichoderma spp. showed less variation during the year. Both species were sensitive to low temperatures. In general, fungi associated with the tea rhizosphere were found to prefer a mesophillic temperature range (15 °C to 35 °C). The dominant species of Penicillium and Trichoderma also exhibited tolerance to lower temperatures, i.e., 5 to 10 °C on agar plates. Most fungi were able to grow in a wide range of pH (4 to 12). Lowering of soil pH in the rhizosphere of tea bushes was positively correlated with the age of the bush and may have affected the development of a specific microbial community in the rhizosphere.The populations of Penicillium and Trichoderma species were inversely correlated with the populations of two most dominant rhizosphere bacteria, Bacillus subtilis and B. mycoides. Both Bacillus species have been shown to have antagonistic activity against these two fungi under in vitro conditions. The present study demonstrates the existence of a similar antagonism under in situ conditions in the rhizosphere of established tea bushes.  相似文献
