New micropaleontological data coming from three cores collected on the meso-Adriatic continental shelf (Vasto area) are studied. Comparisons to foraminiferal assemblages and radiocarbon dates previously collected from cores in the San Benedetto del Tronto and Tremiti areas allow the correlation of patterns observed in shallow water areas with those in deeper parts of the basin. We focused on the response of benthic foraminifera during the Holocene high-stand, corresponding to the installation of the recent sedimentary and trophic system. An influence of anthropogenic impacts cannot be ruled out; its effects consist of a depletion of oxygen level with a consequent modification of the structure of benthic foraminiferal assemblages. During the glacial/post glacial cycle, three phases, characterized by a total of six foraminiferal biofacies including different species assemblages were recognized. The first phase, from 14 kyr BP to 11 kyr BP, corresponds to the Bölling/Alleröd and Younger Dryas cold event, before the Holocene sea-level rise. During this phase, the continental shelf was characterized by an infralittoral environment with productive waters owing to the proximity of the Po river delta at the edge of the Mid-Adriatic Deep. The second phase, from 11 kyr BP to 4 kyr BP, represents the Holocene sea-level rise and is characterized by a condensed sedimentation spread over the entire basin. The third phase corresponds to the Holocene high-stand, during which time the modern current system became established. During this phase, the eastern portion of the shelf underwent to the central part of the mud-belt, corresponding to the sub-recent conditions. Recent eutrophication resulting from human activities over the last few centuries is evidenced by frequency fluctuations of typically opportunistic taxa such as Nonionella turgida and Epistominella vitrea.  相似文献

The capacity of eight wetland seeds to be internally dispersed by common teal (Anas crecca L.) after a dry period typical of natural Mediterranean wetlands was experimentally quantified. These plant taxa (Chara spp., Echinochloacrus-galli (L.) P. Beauv., Eleocharis palustris (L.) Roem. & Schult, Polygonum lapathifolium L., Potamogeton nodosus Poir., Potamogeton pusillus L., Schoenoplectus mucronatus (L.) Palla, Scirpus maritimus L.) were frequently ingested by teal in the field. Captive teal were fed with known quantities of dry seeds and faecal samples were collected at intervals of 1-2 h for 48 h. The proportion of seeds retrieved after gut passage that germinated varied between taxa from 3% to 83%. From 2% to 63% of the seeds ingested were recovered intact, and 18-95% of these were still viable. Therefore, an overall 32% of the seeds ingested were evacuated in a viable condition, ranging from 0.2% for Chara spp. to 54% for S. mucronatus. Mean seed retention time was 3-5 h. Overall, 94% of seeds were evacuated within 12 h, but 2% were recovered after more than 24 h. Seed viability was reduced at longer retention times, although viable seeds of all taxa except Chara spp. were recovered 44-48 h after the experiment began. Germinability was increased by gut passage, although for some species this effect was only significant after stratification, indicating that they exhibit both mechanical and physical dormancy. Seed germination was accelerated by gut passage, especially at longer retention times. Gut passage increased the proportion of viable seeds, suggesting selective digestion of non-viable ones. Endozoochorous transport by teal appears to be a widespread dispersal mechanism among wetland plants.  相似文献

We investigated a range of microbiological community assays performed on scrapes of biofilms formed on artificial diffusing substrates deployed in 8 streams in eastern Scotland, with a view to using them to characterize ecological response to stream water quality. The assays considered were: Multiplex Terminal Restriction Fragment Length Polymorphism or M-TRFLP (a molecular method), Phospholipid Fatty Acid or PLFA analysis (a biochemical method) and MICRORESP (a physiological method) alongside TDI, diatom species, and chlorophyll a content. Four of the streams were classified as of excellent status (3-6 μg/L Soluble Reactive Phosphorus (SRP)) with respect to soluble P content under the EU Water Framework Directive and four were of borderline good/moderate or moderate status (43-577 μg/L SRP). At each site, 3 replicates of 3 solute diffusion treatments were deployed in a Latin square design. Solute diffusion treatments were: KCl (as a control solute), N and P (to investigate the effect of nutrient enrichment), or the herbicide isoproturon (as a “high impact” control, which aimed to affect biofilm growth in a way detectable by all assays). Biofilms were sampled after 4 weeks deployment in a low flow period of early summer 2006.The chlorophyll a content of biofilms after 4 weeks was 2.0 ± 0.29 mg/m2 (mean ± se). Dry matter content was 16.0 ± 13.1 g/m2. The M-TRFLP was successfully used for generating community profiles of cyanobacteria, algae and bacteria and was much faster than diatom identification. The PFLA and TDI were successful after an increase in the sample size, due to low counts. The MICRORESP assays were often below or near detection limit. We estimated the per-sample times for the successful assays as follows: M-TRFLP: 20 min, PLFA 40 min, TDI 90 min. Using MANOVA on the first 5 principal co-ordinates, all the assays except MICRORESP showed significant differences between sites, but none of the assays showed a significant effect of either initial stream trophic status (as classified by the EU Water Framework Directive using chemical standards for soluble P), or of the diffusing solute treatment. Multiple Procrustes analysis on the ordination results showed that the diatom and M-TRFLP data sets hold distinct, though as yet unexplored, information about the ecological factors affecting stream biofilms. The diatom data were subjected to principal components analysis, to identify which taxa were more strongly influenced by site variables, trophic status or treatment effects. These were Acnanthes lanceolata, A. minutissimma, Nitzchia spp., Coccineis spp. and Navicula spp. Further experimentation and data analysis on a larger number of sites, to identify specific M-TRFLP bands that could be used as indicators linked to specific taxa, are desirable. Results highlight the need for a multifactorial approach to understanding controls on stream ecology.  相似文献

Increases in sediment and drift algae accumulations have caused degradation of coastal lagoons worldwide. It is well known that these factors are stressors of seagrass beds, sediment fauna and coral reefs. However, little is known about the impacts on temperate hard-bottom assemblages within soft-bottom lagoons. To test if accumulations of sediment and drift algae (stress) affected recruitment of sessile oyster reef organisms, we constructed cages in Hog Island Bay, Virginia that trapped drifting macroalgae (≈ 2.7 kg WW m2) and facilitated sedimentation (≈ 7 mm per 2-3 month). The stress treatments and unstressed controls were placed in front, between, and behind reefs (position) to represent wave exposed (≈ 0.3 m, windy conditions), current exposed (≈ 0.2 m s 1, peak tide) and protected (≈ 0 m, 0.0 m s 1) habitats. The percentage cover of recruited taxa onto bricks was mapped 4 times during a 1-year period. There were strong significant effects of stress on the total assemblage, plant (but not animal) richness, total plant and animal cover, and cover of the most common taxa. Unstressed bricks had high plant richness, high animal and plant cover, and high cover of the oyster Crassostrea virginica, the alien algae Gracilaria vermiculophylla and Codium fragile, the alga Agardhiella subulata, and high to medium cover of the opportunistic algae Ulva curvata and Enteromorpha spp. In comparison, sediment-stressed bricks had low plant richness, low animal and plant cover, and low cover of C. virginica, G. vermiculophylla, C. fragile, A. subulata, U. curvata and Enteromorpha spp. Similarly, algae-stressed bricks also had low cover of animals, C. virginica, G. vermiculophylla, C. fragile, and A. subulata, but intermediate plant richness and plant cover and high cover of U. curvata and Enteromorpha spp. Although reef position caused significant multivariate results, this factor was clearly less important than the stress factor. Our study shows that accumulations of sediments and drift algae have an adverse impact on sessile temperate reef organisms, reducing richness and abundance, but favoring a few small opportunistic taxa. As the reef-generating oysters themselves performed poorly under these stressors, the long-term impact of the causes of these stressors, eutrophication and urbanization, is likely to be diminished reefs with cascading adverse effects on sessile reef organisms.  相似文献

We present an overview of long-term changes in the floristic composition and growth areas in L. Peipsi (3555 km2, unregulated water level) that have occurred since the 1960s and a list of plant taxa containing 140 species of higher plants and 4 charophytes. A significant correlation was found between the relative abundance and frequency per stations (Fs) (Rs = 0.93). Data on five inhabitants of the eulittoral revealed significant (p < 0.05) inter-annual differences in Fs. Comparison of the data of Fs for 67 taxa for 1970-1980 (87 stations) and 1997-2007 (139 stations) showed a significant change in the Fs distribution (p < 0.03) and a decline (p < 0.05) for 20 taxa; for 15 species Fs had decreased two times or more. However, 14 of the markedly declined taxa, e.g. the long-term dominating submergent Potamogeton perfoliatus, belong still among the top 33 in the list. A significant (χ2 = 11.8; p < 0.028) change was observed in the species number of different frequency classes. The number of taxa in the Fs class 46-100 (92)% was 17 in 1970-1980 but only 3 in 1997-2007. The top of the list of macrophytes is dominated by circumpolar species and vicariants. Impoverishment of the flora in the course of eutrophication is expressed by the decrease in Fs; at the same time, the total number of species had not changed. Among the 20 declined taxa 14 are characteristic of the temporarily flooded and/or shallow-water zone of eutrophic water bodies (amphibious and emergent plants); the remaining taxa are shallow-water submergents. The simpliest explanation for their decrease is the expansion of thick reeds occupying suitable eulittoral habitats.  相似文献

In the genus Carthamus (2n = 20, 22, 24, 44, 64; x = 10, 11, 12), most of the homologues within and between the chromosome complements are difficult to be identified. In the present work, we used fluorescent in situ hybridisation (FISH) to determine the chromosome distribution of the two rRNA gene families, and the two isolated repeated DNA sequences in the 14 Carthamus taxa. The distinctive variability in the distribution, number and signal intensity of hybridisation sites for 18S–26S and 5S rDNA loci could generally distinguish the 14 Carthamus taxa. Active 18S–26S rDNA sites were generally associated with NOR loci on the nucleolar chromosomes. The two A genome taxa, C. glaucus ssp. anatolicus and C. boissieri with 2n = 20, and the two botanical varieties of B genome C. tinctorius (2n = 24) had diagnostic FISH patterns. The present results support the origin of C. tinctorius from C. palaestinus. FISH patterns of C. arborescens vis-à-vis the other taxa indicate a clear division of Carthamus taxa into two distinct lineages. Comparative distribution and intensity pattern of 18S–26S rDNA sites could distinguish each of the tetraploid and hexaploid taxa. The present results indicate that C. boissieri (2n = 20) is one of the genome donors for C. lanatus and C. lanatus ssp. lanatus (2n = 44), and C. lanatus is one of the progenitors for the hexaploid (2n = 64) taxa. The association of pCtKpnI-2 repeated sequence with rRNA gene cluster (orphon) in 2–10 nucleolar and non-nucleolar chromosomes and the consistent occurrence of pCtKpnI-1 repeated sequence at the subtelomeric region in all the taxa analysed indicate some functional role of these sequences.  相似文献

This study investigated the possibility of utilizing detoxified sugarcane bagasse hydrolysate (DSCBH) as an alternative carbon source to culture Yarrowia lipolytica Po1g for microbial oil and biodiesel production. Sugarcane bagasse hydrolysis with 2.5% HCl resulted in maximum total sugar concentration (21.38 g/L) in which 13.59 g/L is xylose, 3.98 g/L is glucose, and 2.78 g/L is arabinose. Detoxification of SCBH by Ca(OH)2 neutralization reduced the concentration of 5-hydroxymethylfurfural and furfural by 21.31% and 24.84%, respectively. Growth of Y. lipolytica Po1g in DSCBH with peptone as the nitrogen source gave maximum biomass concentration (11.42 g/L) compared to NH4NO3 (6.49 g/L). With peptone as the nitrogen source, DSCBH resulted in better biomass concentration than d-glucose (10.19 g/L), d-xylose (9.89 g/L) and NDSCBH (5.88 g/L). The maximum lipid content, lipid yield and lipid productivity of Y. lipolytica Po1g grown in DSCBH and peptone was 58.5%, 6.68 g/L and 1.76 g/L-day, respectively.  相似文献

Seagrass meadows harbour diverse faunal assemblages, but the relative importance of landscapes attributes, settlement processes and biological traits of individual species for recruitment patterns is poorly understood. To quantify the influence of habitat heterogeneity on larval, juvenile and adult post-larval dispersal, invertebrates (> 125 µm) were collected in benthic settlement traps at five occasions (June-August) in three habitats; continuous seagrass, seagrass patches and bare sand. The study was carried out by SCUBA diving in a subtidal (2.5 m depth) seagrass landscape dominated by Zostera marina L. in the Baltic Sea. Traps collected totally > 30 taxa, with non-significant effects of habitat on species richness and total abundance. Total number of invertebrates exhibited strong temporal variability, probably driven by wind-induced bedload and water column transport. Surprisingly, traps located in small (< 12 m2) patches contained on average almost twice as many individuals as traps located in the continuous vegetation. Dominating taxa such as nematodes, copepods, and oligochaetes were found in similar densities across the landscape. In contrast, location within the landscape had strong effects on bivalve settlement and redistribution patterns, resulting in significantly lower densities in continuous vegetation compared to patches and bare sand. A biological trait analysis indicated that semi-mobile taxa with annual protracted direct development, and short-distance dispersal are probably of higher importance for the community assembly process in this environment than long-distance larval dispersal. Results suggest that seagrass landscapes are highly dynamic environments, characterized by time and species-specific effects of landscape attributes on animal dispersal and recruitment. A conceptual model illustrating interactions between settling larvae and landscape heterogeneity is presented.  相似文献

Despite high taxonomic diversity elsewhere in North America during the Pleistocene, vertebrate faunas are exceedingly rare in the region of northern Mexico. Térapa, a unique fossil site located in the present-day desert of Northcentral Sonora, Mexico (29°41′N, 109°39′W, 605 m elevation), contributes to our understanding of the paleoecology and paleoclimate of the region during the Late Pleistocene, ca. 43,000-40,000 cal. yr BP. At least 60 vertebrate taxa, including amphibians, turtles, a crocodilian, snakes, birds and many mammals, have been recovered from an 11-m thick sequence of fossiliferous sediments. The diversity and tropical affinity of these taxa suggest a more-forested environment than the thornscrub desert habitat present in this region today.Isotopic analyses of tooth enamel carbonate from ancient mammalian herbivores suggest that the Sonoran desert has undergone considerable climate change since the Late Pleistocene. Bulk carbon (δ13C) and oxygen (δ18O) isotopes from nine mammalian fossils indicate a habitat mosaic with variations in diet that include browsers, mixed feeders and C4 hyper-grazers (δ13C range of − 10‰ to 2‰). Unique to this site are δ13C tooth enamel values of − 6.1 and − 5.6 ‰ for the deer Odocoileus, which suggest a more variable diet than strict browsing, including possibly feeding on CAM and/or C4 plants. Serial sampling of carbon (δ13C) and oxygen (δ18O) isotopes for ancient mammal teeth with hypsodont dentitions (fossil Equus and Bison,) as well as δ18O meteroric water estimates from well-supported climatic models suggest a cooler and more equable environment at Térapa during the Late Pleistocene. These results also support previous habitat reconstructions inferred from the macrobotanical and packrat midden records of northern Sonora (Mexico). High-resolution stable isotope geochemistry indicates that: 1) ancient Térapa was covered with forest and grassland habitats that extended northward into Mexico by about 350 km relative to their present-day northern limits during the Late Pleistocene; and 2) an Amount Effect (AE) is demonstrated in the fossil record at Térapa even though the climate was less seasonal compared to the modern desert habitat.  相似文献

Distinctive expressions and incidences of discrete dental traits at the outer enamel surface (OES) contribute to the diagnoses of many early hominin taxa. Examination of the enamel-dentine junction (EDJ), imaged non-destructively using micro-computed tomography, has elucidated the morphological development of dental traits and improved interpretations of their variability within and among taxa. The OES expressions of one of these dental traits, the protostylid, have been found to differ among African Plio-Pleistocene fossil hominin taxa. In this study protostylid expression is examined at the OES and at the EDJ of Paranthropus robustus (n = 23) and Australopithecus africanus (n = 28) mandibular molars, with the goals of incorporating EDJ morphology into the definition of the protostylid and assessing the relative contribution of the EDJ and enamel cap to its expression in these taxa. The results provide evidence a) of statistically significant taxon-specific patterns of protostylid morphology at the EDJ that are not evident at the OES; b) that in P. robustus, thick enamel reduces the morphological correspondence between the form of the protostylid seen at the EDJ and the OES, and c) that if EDJ images can be obtained, then the protostylid retains its taxonomic value even in worn teeth.  相似文献

Biotic factors such as interactions among co-occurring taxa are frequently cited as important in determining how nekton use habitat, yet their effects have rarely been tested experimentally. We investigated the general hypothesis that depth distributions of five taxa common in salt marsh intertidal creeks are affected by the presence of other taxa. Two experiments were conducted in a pair of large (15 m3), multi-level (20, 40, and 60 cm water depth) flow-through tanks under fairly stable environmental conditions. During the first experiment, when grass shrimp (Palaemonetes spp.), mummichog (Fundulus heteroclitus), white mullet (Mugil curema), spot (Leiostomus xanthurus), and pinfish (Lagodon rhomboides) were each tested alone, the highest proportions of individuals occupied the intermediate and deepest depths. When all taxa were tested together, the distributions of resident creek taxa, mummichog and grass shrimp, shifted to shallower depths, but the distributions of the seasonal transient taxa, white mullet, spot, and pinfish, did not change. A second experiment was conducted to test the effect of predator presence on grass shrimp depth selection. Neither predatory pinfish nor non-predatory white mullet had a strong influence on grass shrimp depth distribution when tested separately, but grass shrimp shifted to shallower depths in the combined presence of both fish. This suggests that a multiple-taxa effect such as increased disturbance or higher encounter frequency influenced the depth selection of grass shrimp. Our results demonstrate that biotic factors are important in shaping distributions of grass shrimp and mummichog in shallow water habitat.  相似文献

During the Messinian, the Mediterranean area experienced fast and prominent paleoenvironmental changes, culminating in the so-called Messinian Salinity Crisis, with the deposition of the evaporitic series. This work investigates the micropaleontological assemblages in the pre-evaporitic sediments of the Sant’Agata Fossili Marls (SAF) of the Pollenzo section (Cuneo area, North Western Italy). A semiquantitative analysis is carried out on the upper part of the marly and pelitic sediments of the SAF underlying the first gypsum bed, ascribed to the Vena del Gesso Fm. (VDF). The studied interval belongs to the planktonic foraminifer Globorotalia conomiozea Zone and “non distinctive Zone” of Iaccarino and to the calcareous nannofossil MNN11b/c Zone of Raffi et al. (1998, 2003) ( [Raffi et al., 1998] and [Raffi et al., 2003]). Decrease of diversity and abundance of the foraminifer and calcareous nannofossil assemblages is recorded 12 m below the VDG and clearly reflects environmental stress. From bottom to top, six paleoecological events are recorded: (1) the first peak abundance of “small” Reticulofenestra and the last recovery (LR) of planktonic foraminifers; (2) the peak abundance of Pontosphaera japonica and the last recovery of warm water taxa Discoaster spp.; (3) the last recovery of benthic foraminifers; (4) the co-occurring peak abundances of Helicosphaera carteri and Sphenolithus abies, and the last recovery of warm water taxa Amaurolithus spp.; (5) the second peak of “small” Reticulofenestra; (6) the definitive disappearance of calcareous nannofossils. These paleoecological events describe a progressive isolation of the basin from the world ocean and increasingly stressed environment (LR planktonic foraminifers; LR Discoaster spp.), increasing dysoxic to anoxic conditions at the sea floor (LR benthic foraminifers), shallowing of the water column (peak of H. carteri), increasing salinity in surface waters (peak of S. abies), and enhanced nutrient concentration in surface waters (peak of “small” Reticulofenestra); these are related to paleoenvironmental changes predating gypsum deposition at Pollenzo and affecting the whole Mediterranean basin.  相似文献

The interaction of (−)-reboxetine, a non-tricyclic norepinephrine selective reuptake inhibitor, with muscle-type nicotinic acetylcholine receptors (AChRs) in different conformational states was studied by functional and structural approaches. The results established that (−)-reboxetine: (a) inhibits (±)-epibatidine-induced Ca2+ influx in human (h) muscle embryonic (hα1β1γδ) and adult (hα1β1εδ) AChRs in a non-competitive manner and with potencies IC50 = 3.86 ± 0.49 and 1.92 ± 0.48 μM, respectively, (b) binds to the [3H]TCP site with ∼13-fold higher affinity when the Torpedo AChR is in the desensitized state compared to the resting state, (c) enhances [3H]cytisine binding to the resting but activatableTorpedo AChR but not to the desensitized AChR, suggesting desensitizing properties, (d) overlaps the PCP luminal site located between rings 6′ and 13′ in the Torpedo but not human muscle AChRs. In silico mutation results indicate that ring 9′ is the minimum structural component for (−)-reboxetine binding, and (e) interacts to non-luminal sites located within the transmembrane segments from the Torpedo AChR γ subunit, and at the α1/ε transmembrane interface from the adult muscle AChR. In conclusion, (−)-reboxetine non-competitively inhibits muscle AChRs by binding to the TCP luminal site and by inducing receptor desensitization (maybe by interacting with non-luminal sites), a mechanism that is shared by tricyclic antidepressants.  相似文献

Expression in Escherichia coli of his-tagged human mevalonate diphosphate decarboxylase (hMDD) has expedited enzyme isolation, characterization, functional investigation of the mevalonate diphosphate binding site, and crystal structure determination (2.4 Å resolution). hMDD exhibits Vmax = 6.1 ± 0.5 U/mg; Km for ATP is 0.69 ± 0.07 mM and Km for (R,S) mevalonate diphosphate is 28.9 ± 3.3 μM. Conserved polar residues predicted to be in the hMDD active site were mutated to test functional importance. R161Q exhibits a ∼1000-fold diminution in specific activity, while binding the fluorescent substrate analog, TNP-ATP, comparably to wild-type enzyme. Diphosphoglycolyl proline (Ki = 2.3 ± 0.3 uM) and 6-fluoromevalonate 5-diphosphate (Ki = 62 ± 5 nM) are competitive inhibitors with respect to mevalonate diphosphate. N17A exhibits a Vmax = 0.25 ± 0.02 U/mg and a 15-fold inflation in Km for mevalonate diphosphate. N17A’s Ki values for diphosphoglycolyl proline and fluoromevalonate diphosphate are inflated (>70-fold and 40-fold, respectively) in comparison with wild-type enzyme. hMDD structure indicates the proximity (2.8 Å) between R161 and N17, which are located in an interior pocket of the active site cleft. The data suggest the functional importance of R161 and N17 in the binding and orientation of mevalonate diphosphate.  相似文献

In order to assess the effects of polyploidisation on distribution patterns of species in a polyploid complex, we compared potential and actual distribution ranges of representatives from the genus Leucanthemum Mill. (Compositae, Anthemideae), which occur on the Iberian Peninsula with 20 taxa spanning ploidy levels between 2n = 2x = 18 and 2n = 22x = 198. Actual distribution ranges of taxa were derived from geo-referenced herbarium specimens by defining buffer circles around the collection localities to account for geographical uncertainties and haphazardness of specimen collecting. Eco-climatologically potential ranges were modelled with the maximum entropy method (Maxent) based on the actual distributions. By comparison of sizes of realised and eco-climatologically potential areas for each taxon we computed taxon-specific overlap indices that express the filling of eco-climatological niches. The correlation between overlap indices and ploidy levels was tested by Spearman's rank correlation coefficient. We observed a significant negative correlation between ploidy level and sizes of realised distribution ranges and no significant correlation of ploidy level and sizes of potential ranges. Finally, we found that diploid and low-ploidy taxa are filling their potential eco-climatological distribution ranges more exhaustively than highly polyploid taxa. Assuming that ascending polyploidisation is the prevailing mode compared to descending polyploidisation, our results demonstrate that the distribution patterns of members of the Leucanthemum polyploid complex on the Iberian Peninsula are mainly influenced by the age of taxa and not by ecological advantages or disadvantages connected with polyploidy.  相似文献

A 2700-year high resolution pollen record from annually-varved Sugan Lake in the Qaidam Basin at 2793 m a.s.l was obtained to examine vegetation and climatic change on the NE Tibetan Plateau. Pollen data shows that Sugan Basin was constantly covered by open desert-steppe vegetation dominated by Chenopodiaceae, Artemisia, Poaceae and Ephedra. However, large variations in Artemisia/Chenopodiaceae (A/C) ratios suggest regional moisture fluctuations over the last 2700 years, including a dry and relatively stable climate prior to 300 AD, relatively wet climate from 300 to 1200 AD with variability during 1100-1200 AD, and unstable climate since 1200 AD with relatively moister climate during 1250-1400 AD and 1700-1800 AD. However, other proxies (varve thickness, Chironomid taxa, isotopes of oxygen in precipitated carbonate) show fresher water when regional moisture was lower inferred from A/C ratio. This inconsistency suggests the possible difference of in-lake lithology/environment and regional moisture change. Fresh water into the lake from ice melting on the surrounding mountains might have contributed to the in-lake lithology and environment variation. The effective moisture changes in the Basin are in opposite phases to snow accumulation records from Dunde ice core (5325 m a.s.l) and to the monsoon intensity inferred from Dongge Cave, suggesting that the regional topography might have played an important role in mediating moisture changes at regional scale. Pollen data from Sugan Lake shows the shift of moisture at 1200 AD, from stable to variable conditions. This event is well correlated with other paleoclimate proxies in China and other parts of the world; however, the mechanisms behind these patterns require further investigation.  相似文献

The c-type nitric oxide reductase (cNOR) from Paracoccus (P.) denitrificans is an integral membrane protein that catalyzes NO reduction; 2NO + 2e + 2H+ → N2O + H2O. It is also capable of catalyzing the reduction of oxygen to water, albeit more slowly than NO reduction. cNORs are divergent members of the heme-copper oxidase superfamily (HCuOs) which reduce NO, do not pump protons, and the reaction they catalyse is non-electrogenic. All known cNORs have been shown to have five conserved glutamates (E) in the catalytic subunit, by P. denitrificans numbering, the E122, E125, E198, E202 and E267. The E122 and E125 are presumed to face the periplasm and the E198, E202 and E267 are located in the interior of the membrane, close to the catalytic site. We recently showed that the E122 and E125 define the entry point of the proton pathway leading from the periplasm into the active site [U. Flock, F.H. Thorndycroft, A.D. Matorin, D.J. Richardson, N.J. Watmough, P. Ädelroth, J. Biol. Chem. 283 (2008) 3839-3845]. Here we present results from the reaction between fully reduced NOR and oxygen on the alanine variants of the E198, E202 and E267. The initial binding of O2 to the active site was unaffected by these mutations. In contrast, proton uptake to the bound O2 was significantly inhibited in both the E198A and E267A variants, whilst the E202A NOR behaved essentially as wildtype. We propose that the E198 and E267 are involved in terminating the proton pathway in the region close to the active site in NOR.  相似文献
