AIMS: A laboratory-scale autotrophic membrane-attached biofilm reactor was developed to remove nitrate from drinking water. METHODS AND RESULTS: Hydrogen and carbon dioxide flowed together into the lumem side of a gas-permeable silicone tube. The gases diffused through the membrane wall to feed Alcaligenes eutrophus that formed a biofilm on the surface of the silicone tube for autotrophic denitrification. Hydrogen provided the energy source, and carbon dioxide, besides serving as the carbon source, was employed to neutralize the alkalinity from denitrification. The optimal carbon dioxide concentration in the silicone tube was between 20% and 50%. CONCLUSION: This study has demonstrated that a gas-permeable silicone tube is a convenient and efficient method to feed A. eutrophus for autotrophic denitrification. Supplying a suitable amount of carbon dioxide together with hydrogen into the silicone tube solved the problem that alkalinity formation caused during denitrification. The pH of the bioreactor was maintained at about 7 to avoid nitrite accumulation, and then the nitrogen removal rate was increased. A high specific nitrogen removal rate (1.6-5.4 g Nm(2)d(-1-1) of surface area of silicone tube) was achieved. SIGNIFICANCE AND IMPACT OF THE STUDY: In addition to combining the advantages of the hydrogenotrophic denitrification process and a membrane feeding substrate bioreactor (MFSB), this bioreactor achieved a high nitrogen removal rate and is simple to operate. Halophilic nitrifiers belonging to the genus Nitrosomonas and Nitrospira were enriched from seawater and marine sediment samples of the North Sea. The maximal ammonia oxidation rate (AOR) in batch enrichments with seawater was 15.1 mg N L−1 day−1. An intermediate nitrite accumulation was observed. Two fixed-bed reactors for continuous nitrification with either polyethylene/clay sinter lamellas (FBR A) or porous ceramic rings (FBR B) were run at two different ammonia concentrations, three different ammonia loading rates (ALRs), ± pH adjustment, and at an increased upflow velocity. A better overall nitrification without nitrite accumulation was observed in FBR B. However, FBR A revealed a higher AOR and nitrite oxidation rate of 6 and 7 mg N L−1 h−1, compared to FBR B with 5 and 5.9 mg N L−1 h−1, respectively. AORs in the FBRs were at least ten times higher than in suspended enrichment cultures. Whereas a shift within the ammonia-oxidizing population in the genus Nitrosomonas at the subspecies level occurred in FBR B with synthetic seawater at an increasing ALR and a decreasing pH, the nitrite oxidizing Nitrospira population apparently did not change.  相似文献

The performance of acetic acid-supported pH-heterogenized heterotrophic denitrification (HD) facilitated with ferrous sulfide-based autotrophic denitrification (AD) was investigated in upflow activated carbon-packed column reactors for reliable removal of highly elevated nitrate (42 mg NO3-N l−1) in drinking water. The use of acetic acid as substrate provided sufficient internal carbon dioxide to completely eliminate the need of external pH adjustment for HD, but simultaneously created vertically heterogenized pH varying from 4.8 to 7.8 in the HD reactor. After 5-week acclimation, the HD reactor developed a moderate nitrate removal capacity with about one third of nitrate removal occurring in the acidic zone (pH 4.8–6.2). To increase the treatment reliability, acetic acid-supported HD was operated under 10% carbon limitation to remove >85% of nitrate, and ferrous sulfide-based AD was supplementally operated to remove residual nitrate and formed nitrite without excess of soluble organic carbon, nitrite or sulfate in the final effluent.  相似文献

The dispersion of nitrogenous compounds and heavy metals into the environment is frequent during mining activities. The effects of nickel (Ni) and cobalt (Co) on denitrification of simulated mine waters were investigated in batch bioassays and fluidized-bed reactors (FBRs). At pH 7, batch tests revealed that Co did not exhibit inhibition on denitrification even at 86.6 mg/L. Ni showed to be inhibitory at 50 and 100 mg/L by decreasing nitrate removal efficiencies of 18 and 65 %, respectively. In two FBRs, operated at 7–8 and 22 °C, 5.5 mg/L Ni did not affect nitrate and nitrite removals because of FBR potential of diluting soluble Ni feed concentration. On the contrary, the effluent pH clearly decreased in both FBR1 and FBR2 because of nickel sulfide precipitation and Ni inhibition of the last two steps of denitrification. When Ni injection was stopped, the process recovered more slowly at 22 than 7–8 °C. This is the first study reporting the effect of Ni on denitrification in biological FBRs.  相似文献

Bacterial strains CVO and FWKO B were isolated from produced brine at the Coleville oil field in Saskatchewan, Canada. Both strains are obligate chemolithotrophs, with hydrogen, formate, and sulfide serving as the only known energy sources for FWKO B, whereas sulfide and elemental sulfur are the only known electron donors for CVO. Neither strain uses thiosulfate as an energy source. Both strains are microaerophiles (1% O2). In addition, CVO grows by denitrification of nitrate or nitrite whereas FWKO B reduces nitrate only to nitrite. Elemental sulfur is the sole product of sulfide oxidation by FWKO B, while CVO produces either elemental sulfur or sulfate, depending on the initial concentration of sulfide. Both strains are capable of growth under strictly autotrophic conditions, but CVO uses acetate as well as CO2 as its sole carbon source. Neither strain reduces sulfate; however, FWKO B reduces sulfur and displays chemolithoautotrophic growth in the presence of elemental sulfur, hydrogen, and CO2. Both strains grow at temperatures between 5 and 40°C. CVO is capable of growth at NaCl concentrations as high as 7%. The present 16s rRNA analysis suggests that both strains are members of the epsilon subdivision of the division Proteobacteria, with CVO most closely related to Thiomicrospira denitrifcans and FWKO B most closely related to members of the genus Arcobacter. The isolation of these two novel chemolithotrophic sulfur bacteria from oil field brine suggests the presence of a subterranean sulfur cycle driven entirely by hydrogen, carbon dioxide, and nitrate.  相似文献

Increasing concentrations of carbon dioxide (CO2) in the atmosphere or continuous nitrogen (N) deposition might alter the carbon (C) cycle in boreal mires and thus have significant impacts on the development of climate change. The atmospheric impact of the C cycle in mires is twofold: C accumulation attenuates and CH4 release strengthens the natural greenhouse effect. We studied the effects of an increased supply of CO2 or NH4NO3 on the vegetation and annual CO2 exchange in lawns of a boreal oligotrophic mire in eastern Finland over a 2-year period. Ten study plots were enclosed with mini-FACE (Free Air Carbon Dioxide Enrichment) rings. Five plots were vented with CO2-enriched air (target 560 ppmv), while their controls were vented with ambient air; five plots were sprayed with NH4NO3, corresponding to a cumulative addition of 3 g N m−2 a−1, while their controls were sprayed with distilled water only. A raised NH4NO3 supply seemed to affect the composition of the moss layer. Raised CO2 did not affect the vegetation, but gross photosynthesis increased significantly. The change in net CO2 exchange depended on the annual weather conditions. Our results suggest that C accumulation may increase in wet years and compensate for the warming effect caused by the increase in CH4 release from this mire. In contrast, a relatively dry and warm growing period favors decomposition and can even make the CO2 balance negative. Along with the increased CH4 release under raised CO2, the decreased C accumulation then increases the radiative forcing of boreal mires. Received 22 October 2001; accepted 13 May 2002.  相似文献

The effect of CO2 enrichment (700 and 1050 ppm) on phytomass, soluble sugars, leaf nitrogen and secondary chemicals of three Salix myrsinifolia clones was studied in plants cultivated at very poor (sand seedlings) and moderate (peat seedlings) nutrient availability and under low illumination. The total shoot phytomass production of sand scedlings was less than 10% of that of the peat seedlings. Carbon dioxide increased the total shoot phytomass of peat seedlings. When the ambient carbon supply was doubled (to 700 ppm) the growth of sand seedlings was slightly enhanced but 1050 ppm CO2 gave growth figures similar to those at the control CO2 level. Leaf nitrogen content and total soluble sugar contents were significantly higher in peat seedlings than in sand seedlings. Leaf nitrogen showed a decreasing trend in relation to CO2 increase. On the other hand, CO2 did not have any clear-cut effect on total sugars. At the control CO2 level the content of salicortin, which is a dynamic phenolic, was higher in the peat seedlings than in the sand seedlings, but salicin showed the opposite trend. CO2 enrichment considerably decreased these phenolics in the peat seedlings. At the control CO2 level, the content of more static phenolics, such as proanthocyanidins, was higher in sand seedlings. An increased carbon supply considerably increased static phenolics in the peat seedlings. Willow defence against generalist herbivores is moderately decreased by enhancement of atmospheric carbon dioxide.  相似文献

The influence of pH on the denitrification activity of a continuous culture of Paracoccus denitrificans was studied in relation to the presence of nitrite. After a transition from aerobic to anaerobic conditions at the suboptimal pH of 6.8, P. denitrificans was not able to build up a functional denitrification pathway. Nitrite accumulated in the medium as the predominant denitrification product. Although the nitrite reductase gene was induced properly, the enzyme could not be detected at sufficient amounts in the culture. These observations indicate that either translation was somehow inhibited, or once synthesized nitrite reductase was inactivated, possibly by the high concentrations of nitrous acid (HNO2. Interestingly, when a P. denitrificans culture which was grown to steady-state under anaerobic conditions was then exposed to suboptimal pHs, cells exhibited a reduced overall denitrification activity, but neither nitrite nor any other denitrification intermediate accumulated.  相似文献

Photosynthetic capacities of five species of brown algae in red light were found to be strongly limited by the inorganic carbon supply of natural sea water. Under these conditions, pH 8·2 and dissolved inorganic carbon concentration (DIG) of 2·1 mol m?3, a short pulse of blue light was found to increase the subsequent rate of photosynthesis in saturating red light. The degree of blue light stimulation varied between species, ranging from an increase of over 200% of the original rate in Colpomenia peregrins to only 10% in Dictyota dichotoma. Increasing the DIG concentration of sea water by bicarbonate addition resulted in carbon saturation of photosynthesis in all five species. Blue light stimulation was greatly reduced at these higher DIG concentrations. The response in Laminaria digitata was examined in more detail by manipulation of pH and DIG to produce solutions with different concentrations of dissolved CO2. At a CO2 concentration typical of normal sea water (12·4 mmol m?3), blue light treatment increased photosynthetic rate by approximately 50%. Blue light stimulation was increased to over 150% at CO2 concentrations below that of sea water, whereas at concentrations above that of sea water, the effect was diminished. Therefore, the effect of blue light on photosynthetic capacity appears to involve an increase in the rate of supply of carbon dioxide to the plant.  相似文献

The effects of carbon dioxide supply within the range of 1–5 % (along with purified air), on cell culture of Panax ginseng were investigated in a balloon type bubble bioreactor containing 4 dm3 of Murashige and Skoog (MS) medium supplemented with 7.0 mg dm−3 indolebutyric acid, 0.5 mg dm−3 kinetin and 30 g dm−3 sucrose. A 1 % CO2 supply was found beneficial for the production of cell mass; however, increasing CO2 concentration to 2.5 and 5 % decreased the biomass accumulation. CO2 enrichment was not beneficial for saponin production and 1, 2.5, and 5 % CO2 supply resulted in decrease in saponin accumulation up to 11.6, 19.5, and 50.6 %, respectively.  相似文献

脱氮假单胞菌发酵生产维生素B12过程中,副产物粪卟啉Ⅲ的积累对产物的代谢合成和分离提取有很大的影响。建立了发酵液中粪卟啉Ⅲ的HPLC快速测定方法,发酵液处理后直接进样测定,检测线性范围为12~275 μg/ml,重复性好。研究了不同供氧水平、二氧化碳浓度和pH值对发酵过程中维生素B12和粪卟啉Ⅲ代谢合成的影响,并在120吨发酵罐中进行了发酵过程优化控制。结果表明:在发酵过程产物的合成期控制适当的供氧水平、控制二氧化碳浓度在8.6±0.8%、维持pH值在7.0±0.12能明显抑制卟啉Ⅲ的生物合成,同时使维生素B12产量提高15%。  相似文献

The regulation of carbonic anhydrase by environmental conditions was determined forChlamydomonas reinhardtii. The depression of carbonic anhydrase in air-grown cells was pH-dependent. Growth of cells on air at acid pH, corresponding to 10 m CO2 in solution, resulted in complete repression of carbonic-anhydrase activity. At pH 6.9, increasing the CO2 concentration to 0.15% (v/v) in the gas phase, corresponding to 11 M in solution, was sufficient to completely repress carbonic-anhydrase activity. Photosynthesis and intracellular inorganic carbon were measured in air-grown and high-CO2-grown cells using a silicone-oil centrifugation technique. With carbonic anhydrase repressed cells limited inorganic-carbon accumulation resulted from non-specific binding of CO2. With air-grown cells, inorganic-carbon uptake at acid pH, i.e. 5.5, was linear up to 0.5 mM external inorganic-carbon concentration whereas at alkaline pH, i.e. 7.5, the accumulation ratio decreased with increase in external inorganic-carbon concentration. It is suggested that in air-grown cells at acid pH, CO2 is the inorganic carbon species that crosses the plasmalemma. The conversion of CO2 to HCO 3 - by carbonic anhydrase in the cytosol results in inorganic-carbon accumulation and maintains the diffusion gradient for carbon dioxide across the cell boundary. However, this mechanism will not account for energy-dependent accumulation of inorganic carbon when there is little difference in pH between the exterior and cytosol.  相似文献

This study investigated inorganic carbon accumulation in relation to photosynthesis in the marine dinoflagellate Prorocentrum micans. Measurement of the internal inorganic carbon pool showed a 10-fold accumulation in relation to external dissolved inorganic carbon (DIC). Dextran-bound sulfonamide (DBS), which inhibited extracellular carbonic anhydrase, caused more than 95% inhibition of DIC accumulation and photosynthesis. We used real-time imaging of living cells with confocal laser scanning microscopy and a fluorescent pH indicator dye to measure transient pH changes in relation to inorganic carbon availability. When steady-state photosynthesizing cells were DIC limited, the chloroplast pH decreased from 8.3 to 6.9 and cytosolic pH decreased from 7.7 to 7.1. Re-addition of HCO3 led to a rapid re-establishment of the steady-state pH values abolished by DBS. The addition of DBS to photosynthesizing cells under steady-state conditions resulted in a transient increase in intracellular pH, with photosynthesis maintained for 6 s, the amount of time needed for depletion of the intracellular inorganic carbon pool. These results demonstrate the key role of extracellular carbonic anhydrase in facilitating the availability of CO2 at the exofacial surface of the plasma membrane necessary to maintain the photosynthetic rate. The need for a CO2-concentrating mechanism at ambient CO2 concentrations may reflect the difference in the specificity factor of ribulose-1,5 bisphosphate carboxylase/oxygenase in dinoflagellates compared with other algal phyla.  相似文献

The biological utilization of CO2 and H2 for the formation of short-chain fatty acids was studied by using a mixed culture of bacteria. Optimization of a medium was carried out in continuous culture to identify limiting factors which controlled growth and production of organic acids. The optimal pH for growth and acid production was 7.0 at 37°C; the maximal cell concentration obtained was 5.9 g of cells per liter (dry weight), and the maximal amount of volatile acids formed was 4.7 g/liter, with acetic acid as the predominant acid. With the optimized medium, it was found that the rate of transfer of hydrogen or carbon dioxide, or both, from gas to liquid was the limiting factor which controlled growth and production of acids.  相似文献

Algal biomass refineries for sustainable transportation fuels, in particular biodiesel, will benefit from algal strain enhancements to improve biomass and lipid productivity. Specifically, the supply of inorganic carbon to microalgal cultures represents an area of great interest due to the potential for improved growth of microalgae and the possibility for incorporation with CO2 mitigation processes. Combinations of bicarbonate (HCO3?) salt addition and application of CO2 to control pH have shown compelling increases in growth rate and lipid productivity of fresh water algae. Here, focus was placed on the marine organism, Nannochloropsis gaditana, to investigate growth and lipid accumulation under various strategies of enhanced inorganic carbon supply. Three gas application strategies were investigated: continuous sparging of atmospheric air, continuous sparging of 5% CO2 during light hours until nitrogen depletion, and continuous sparging of atmospheric air supplemented with 5% CO2 for pH control between 8.0 and 8.3. These gas sparging schemes were combined with addition of low concentrations (5 mM) of sodium bicarbonate at inoculation and high concentration (50 mM) of sodium bicarbonate amendments just prior to nitrogen depletion. The optimum scenario observed for growth of N. gaditana under these inorganic carbon conditions was controlling pH with 5% CO2 on demand, which increased both growth rate and lipid accumulation. Fatty acid methyl esters were primarily comprised of C16:0 (palmitic) and C16:1 (palmitoleic) aliphatic chains. Additionally, the use of high concentration (50 mM) of bicarbonate amendments further improved lipid content (up to 48.6%) under nitrogen deplete conditions when paired with pH-controlled strategies.  相似文献

Biomass production by the green algae Scenedesmus obliquus and Chlorella vulgaris in intensive laboratory continuous cultures was considerably affected by the pH at which the cultures were maintained. Carbon photoassimilation experiments revealed that pH values in the range of 8 to 9 were important for determining the free CO2 concentrations in the medium. With higher pH values, additional pH effects were observed involving a decrease in the relative high affinity of low CO2-adapted algae to free CO2. The carbon uptake rate by high CO2-adapted algae after transfer to low free CO2 medium was characterized by a lag period of about 30 min, after which the affinity of the algae to CO2 increased considerably. Both continuous growth and carbon uptake experiments indicated that artificially maintained high free CO2 concentrations are recommended for maximal production in intensive outdoor algal cultures.  相似文献

Forests in the south-eastern United States experienced a prolonged dry spell and above-normal temperatures during the 1995 growing season. During this episode, nearly continuous, eddy covariance measurements of carbon dioxide and water vapour fluxes were acquired over a temperate, hardwood forest. These data are used to examine how environmental factors and accumulating soil moisture deficits affected the diurnal pattern and magnitude of canopy-scale carbon dioxide and water vapour fluxes. The field data are also used to test an integrative leaf-to-canopy scaling model (CANOAK), which uses micrometeorological and physiological theory, to calculate mass and energy fluxes. When soil moisture was ample in the spring, peak rates of net ecosystem CO2 exchange (NF) occurred around midday and exceeded 20 μmol m?2 s?1. Rates of NK were near optimal when air temperature ranged between 22 and 25°C. The accumulation of soil moisture deficits and a co-occurrence of high temperatures caused peak rates of daytime carbon dioxide uptake to occur earlier in the morning. High air temperatures and soil moisture deficits were also correlated with a dramatic reduction in the magnitude of NE. On average, the magnitude of NE decreased from 20 to 7 μmol m?2 s?1 as air temperature increased from 24 to 30°C and the soil dried. The CANAOK model yielded accurate estimates of canopy-scale carbon dioxide and water vapour fluxes when the forest had an ample supply of soil moisture. During the drought and heat spell, a cumulative drought index was needed to adjust the proportionality constant of the stomatal conductance model to yield accurate estimates of canopy CO2 exchange. The adoption of the drought index also enabled the CANOAK model to give improved estimates of evaporation until midday. On the other hand, the scheme failed to yield accurate estimates of evaporation during the afternoon.  相似文献

A method for the measurement of oxygen uptake and carbon dioxide production rates in mammalian cell cultures using membrane mass spectrometry is described. The small stirred reactor with a volume of 15 ml and integrated pH-control permits the economical application of isotopically labelled substrates and 13C-labelled bicarbonate buffer. Repetitive experiments showed the reproducibility of the method. In one case bicarbonate-free HEPES buffer was used and carbon dioxide production was measured using the intensity of the peak at m/z = 44(12CO2). In all other cases H13CO3 -buffer was applied and also12CO2 was measured. The minimum cell density required was only 2 × 104 cells ml−1. In the hybridoma T-flask cultivation studied here the measured specific oxygen uptake and carbon dioxide production rates were reasonably constant during the exponential growth phase and decreased significantly afterwards. Estimated respiratory quotients were always between0.90 and 0.92 except in HEPES-buffer, where a value of 0.67 was found. In the latter case specific oxygen uptake rate was higher than in bicarbonate buffered culture, however, carbon dioxide production rate was lower, and viable cell density was lowest. The addition of phenazine methosulfate, an artificial electron acceptor, increased both rates resulting in highest viable cell density but also highest lactate production rate. Glucose and glutamine pulse-feeding increased final cell density. The method described is directly applicable for samples from batch, fed-batch and continuous cultivations. The efforts to explain the ‘missing sink’ for anthropogenic carbon dioxide (CO2) have included in recent years the role of nitrogen as an important constraint for biospheric carbon fluxes. We used the Nitrogen Carbon Interaction Model (NCIM) to investigate patterns of carbon and nitrogen storage in different compartments of the terrestrial biosphere as a consequence of a rising atmospheric CO2 concentration, in combination with varying levels of nitrogen availability. This model has separate but closely coupled carbon and nitrogen cycles with a focus on soil processes and soil–plant interactions, including an active compartment of soil microorganisms decomposing litter residues and competing with plants for available nitrogen. Biological nitrogen fixation is represented as a function of vegetation nitrogen demand. The model was validated against several global datasets of soil and vegetation carbon and nitrogen pools. Five model experiments were carried out for the modeling periods 1860–2002 and 2002–2100. In these experiments we varied the nitrogen availability using different combinations of biological nitrogen fixation, denitrification, leaching of soluble nitrogen compounds with constant or rising atmospheric CO2 concentrations. Oversupply with nitrogen, in an experiment with nitrogen fixation, but no nitrogen losses, together with constant atmospheric CO2, led to some carbon sequestration in organismic pools, which was nearly compensated by losses of C from soil organic carbon pools. Rising atmospheric CO2 always led to carbon sequestration in the biosphere. Considering an open nitrogen cycle including dynamic nitrogen fixation, and nitrogen losses from denitrification and leaching, the carbon sequestration in the biosphere is of a magnitude comparable to current observation based estimates of the ‘missing sink.’ A fertilization feedback between the carbon and nitrogen cycles occurred in this experiment, which was much stronger than the sum of separate influences of high nitrogen supply and rising atmospheric CO2. The demand‐driven biological nitrogen fixation was mainly responsible for this result. For the modeling period 2002–2100, NCIM predicts continued carbon sequestration in the low range of previously published estimates, combined with a plausible rate of CO2‐driven biological nitrogen fixation and substantial redistribution of nitrogen from soil to plant pools.  相似文献

This review reports the physiological and metabolic changes in plants during development under elevated atmospheric carbon dioxide concentration and/or limited-nitrogen supply in order to establish their effects on leaf senescence induction. Elevated CO2 concentration and nitrogen supply modify gene expression, protein content and composition, various aspects of photosynthesis, sugar metabolism, nitrogen metabolism, and redox state in plants. Elevated CO2 usually causes sugar accumulation and decreased nitrogen content in plant leaves, leading to imbalanced C/N ratio in mature leaves, which is one of the main factors behind premature senescence in leaves. Elevated CO2 and low nitrogen decrease activities of some antioxidant enzymes and thus increase H2O2 production. These changes lead to oxidative stress that results in the degradation of photosynthetic pigments and eventually induce senescence. However, this accelerated leaf senescence under conditions of elevated CO2 and limited nitrogen can mobilize nutrients to growing organs and thus ensure their functionality.  相似文献
