4-O-Glycosylation of 2-azidoethyl 2,3,6-tri-O-benzyl-4-O-(2,3-di-O-benzyl-6-O-benzoyl--D-galactopyranosyl)--D-glucopyranoside with a disaccharide donor, 4-trichloroacetamidophenyl 4,6-di-O-acetyl-2-deoxy-3-O-(2,3,4,6-tetra-O-acetyl--D-galactopyranosyl)-1-thio-2-trichloroacetamido--D-galactopyranoside, in dichloromethane in the presence of N-iodosuccinimide and trifluoromethanesulfonic acid resulted in a tetrasaccharide, 2-azidoethyl (2,3,4,6-tetra-O-acetyl--D-galactopyranosyl)-(1 3)-(4,6-di-O-acetyl-2-deoxy-2-trichloroacetamido--D-galactopyranosyl)-(1 4)-(2,3-di-O-benzyl-6-O-benzoyl--D-galactopyranosyl)-(1 4)-2,3,6-tri-O-benzyl--D-glucopyranoside, in 69% yield. The complete removal of O-protecting groups in the tetrasaccharide, the replacement of N-trichloroacetyl by N-acetyl group, and the reduction of the aglycone azide group to amine led to the target aminoethyl glycoside of -D-Gal-(1 3)--D-GalNAc-(1 4)--D-Gal-(1 4)--D-Glc-OCH2CH2NH2 containing the oligosaccharide chain of asialo-GM1 ganglioside in 72% overall yield. Selective 3-O-glycosylation of 2-azidoethyl 2,3,6-tri-O-benzyl-4-O-(2,6-di-O-benzyl--D-galactopyranosyl)--D-glucopyranoside with thioglycoside methyl (ethyl 5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-2-thio-D-glycero--D-galacto-2-nonulopyranosyl)oate in acetonitrile in the presence of N-iodosuccinimide and trifluoromethanesulfonic acid afforded 2-azidoethyl [methyl (5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero--D-galacto-2-nonulopyranosyl)oate]-(2 3)-(2,6-di-O-benzyl--D-galactopyranosyl)-(1 4)-2,3,6-tri-O-benzyl--D-glucopyranoside, the selectively protected derivative of the oligosaccharide chain of GM3 ganglioside, in 79% yield. Its 4-O-glycosylation with a disaccharide glycosyl donor, (4-trichloroacetophenyl-4,6-di-O-acetyl-2-deoxy-3-O-(2,3,4,6-tetra-O-acetyl--D-galactopyranosyl) 1-thio-2-trichloroacetamido--D-galactopyranoside in dichloromethane in the presence of N-iodosuccinimide and trifluoromethanesulfonic acid gave 2-azidoethyl (2,3,4,6-tetra-O-acetyl--D-galactopyranosyl)-(1 3)-(4,6-di-O-acetyl-2-deoxy-2-trichloroacetamido--D-galactopyranosyl)-(1 4)-{[methyl (5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero--D-galacto-2-nonulopyranosyl)onate]-(2 3)}-(2,6-di-O-benzyl--D-galactopyranosyl)-(1 4)-2,3,6-tri-O-benzyl--D-glucopyranoside in 85% yield. The resulting pentasaccharide was O-deprotected, its N-trichloroacetyl group was replaced by N-acetyl group, and the aglycone azide group was reduced to afford in 85% overall yield aminoethyl glycoside of -D-Gal-(1 3)--D-GalNAc-(1 4)-[-D-Neu5Ac-(2 3)]--D-Gal-(1 4)--D-Glc-OCH2CH2NH2 containing the oligosaccharide chain of GM1 ganglioside.  相似文献

Summary To determine whether sweet potato (Ipomoea batatas (L.) Poir.) takes up organic matter through the roots from the medium, the concentrations of natural14C (14C) in plant organic matter, atmospheric CO2 and compost applied to media were examined under soil and sand culture conditions. In these experiments, three kinds of composts of different 14C were used. CO2 derived from the mineralization of compost was continuously pumped out from the pots and its direct uptake by the leaves was prevented.14C of plant parts harvested after the 43 days experimental period were affected by the 14C of the compost in the treatments where the compost of rice straw was applied, and which suggested that a significant amount of plant carbon was derived from the compost.  相似文献

Our objectives were to quantify and compare soil CO2 efflux of two dominant soil types in an old-growth neotropical rain forest in the Atlantic zone of Costa Rica, and to evaluate the control of environmental factors on CO2 release. We measured soil CO2 efflux from eight permanent soil chambers on six Oxisol sites. Three sites were developed on old river terraces (old alluvium) and the other three were developed on old lava flows (residual). At the same time we measured soil CO2 concentrations, soil water content and soil temperature at various depths in 6 soil shafts (3 m deep). Between old alluvium sites, the two-year average CO2 flux rates ranged from 117.3 to 128.9 mg C m–2 h–1. Significantly higher soil CO2 flux occurred on the residual sites (141.1 to 184.2 mg C m–2 h–1). Spatial differences in CO2 efflux were related to fine root biomass, soil carbon and phosphorus concentration but also to soil water content. Spatial variability in CO2 storage was high and the amount of CO2 stored in the upper and lower soil profile was different between old alluvial and residual sites. The major factor identified for explaining temporal variations in soil CO2 efflux was soil water content. During periods of high soil water content CO2 emission decreased, probably due to lower diffusion and CO2 production rates. During the 2-year study period inter-annual variation in soil CO2 efflux was not detected.  相似文献

Photosystem II chlorophyll fluorescence and leaf net gas exchanges (CO2 and H2O) were measured simultaneously on bean leaves (Phaseolus vulgaris L.) submitted either to different ambient CO2 concentrations or to a drought stress. When leaves are under photorespiratory conditions, a simple fluorescence parameter F/ Fm (B. Genty et al. 1989, Biochem. Biophys. Acta 990, 87–92; F = difference between maximum, Fm, and steady-state fluorescence emissions) allows the calculation of the total rate of photosynthetic electron-transport and the rate of electron transport to O2. These rates are in agreement with the measurements of leaf O2 absorption using 18O2 and the kinetic properties of ribulose-1,5bisphosphate carboxylase/oxygenase. The fluorescence parameter, F/Fm, showed that the allocation of photosynthetic electrons to O2 was increased during the desiccation of a leaf. Decreasing leaf net CO2 uptake, either by decreasing the ambient CO2 concentration or by dehydrating a leaf, had the same effect on the partitioning of photosynthetic electrons between CO2 and O2 reduction. It is concluded that the decline of net CO2 uptake of a leaf under drought stress is only due, at least for a mild reversible stress (causing at most a leaf water deficit of 35%), to stomatal closure which leads to a decrease in leaf internal CO2 concentration. Carbon-isotope ratios were examined as 13C values in several C3, C4, and C3–C4 Flaveria species, and compared to predicted 13C, values generated from theoretical models. The measured 13C values were within 4 of those predicted from the models. The models were used to identify factors that contribute to C3-like 13C values in C3–C4 species that exhibit considerable C4-cycle activity. Two of the factors contributing to C3-like 13C values are high CO2 leakiness from the C4 pathway and pi/pa values that were higher than C4 congeners. A marked break occurred in the relationship between the percentage of atmospheric CO2 assimilated through the C4 cycle and the 13C value. Below 50% C4-cycle assimialtion there was no significant relationship between the variables, but above 50% the 13C values became less negative. These results demonstrate that the level of C4-cycle expression can increase from, 0 to 50% with little integration of carbon transfer from the C4 to the C3 cycle. A simple approach to determine CO2/O2 specificity factor () of ribulose 1,5-bisphosphate carboxylase/oxygenase is described. The assay measures the amount of CO2 fixation at varying [CO2]/[O2] ratios after complete consumption of ribulose 1,5-bisphosphate (RuBP). Carbon dioxide fixation catalyzed by the carboxylase was monitored by directly measuring the moles of 14CO2 incorporated into 3-phosphoglycerate (PGA). This measurement at different [CO2]/[O2] ratios is used to determine graphically by several different linear plots the total RuBP consumed by the two activities and the CO2/O2 specificity factor. The assay can be used to measure the amounts of products of the carboxylase and oxygenase reactions and to determine the concentration of the substrate RuBP converted to an endpoint amount of PGA and phosphoglycolate. The assay was found to be suitable for all [CO2]/[O2] ratios examined, ranging from 14 to 215 micromolar CO2 (provided as 1–16 mM NaHCO3) and 614 micromolar O2 provided as 50% O2. The procedure described is extremely rapid and sensitive. The Maricopa cotton and wheat FACE (free-air CO2 enrichment) experiments offer propitious opportunity to quantify carbon turnover. Soil CO2 and 13C of soil organic carbon (SOC) in CO2-enriched and Control plots were measured between the final cotton FACE project (October 1991) and the end of the second wheat experiment (June 1994). The initial 13C-depletion in SOC of cotton FACE plots (measured by the difference in 13C between FACE and Control plots) persisted at the same level (1.9) 1.5 years after the experiment ended. A similar depletion was observed in soil CO2 evolved in the same plots, indicating ongoing decomposition of the new SOC. The SOC 13C of wheat plots before and after two growing seasons showed increasing 13C-depletion in FACE relative to Control. Isotopic mass balance was consistent with 5–6% new carbon input from the two wheat crops. This is lower than the 12–13% calculated for FACE cotton and perhaps a consequence of the larger root system of cotton or the 3-year duration of the cotton experiments versus 2 years for the wheat.  相似文献

14CO2 assimilation, 14C incorporation into glycolate and glycolate accumulation in -HPMS treated bean leaves at various O2 and CO2 concentrations were studied. In 1% CO2 oxygen concentration had no significant effect on glycolate accumulation and 14C incorporation into glycolate. In the CO2 concentration range of 0.03% to 0.01%, increased oxygen concentration decreased not only 14CO2 assimilation but also glycolate accumulation and 14C incorporation into glycolate. In 1% and 0.1% CO2, no matter what O2 concentration was supplied, and in 0.03% CO2 with 2% and 21% O2, all of the glycolate accumulated was formed from newly assimilated carbon. In 0.01% CO2 and 2%, 21% and 100% O2, and in 0.03% CO2 with 100% O2, a substantial portion of the glycolic acid that accumulated in leaves originated from endogenous unlabelled substrates. These findings are discussed in terms of possible changes in the ratio of RuBP carboxylation to RuBP oxygenation and of changes of RuBP pool size, induced by changing O2 and CO2 concentrations.This work was supported by the Polish Academy of Sciences, Contract No. 10.2.10.  相似文献

Onion (Allium cepa L.) plants were examined to determine the photosynthetic role of CO2 that accumulates within their leaf cavities. Leaf cavity CO2 concentrations ranged from 2250 L L–1 near the leaf base to below atmospheric (<350 L L–1) near the leaf tip at midday. There was a daily fluctuation in the leaf cavity CO2 concentrations with minimum values near midday and maximum values at night. Conductance to CO2 from the leaf cavity ranged from 24 to 202 mol m–2 s–1 and was even lower for membranes of bulb scales. The capacity for onion leaves to recycle leaf cavity CO2 was poor, only 0.2 to 2.2% of leaf photosynthesis based either on measured CO2 concentrations and conductance values or as measured directly by 14CO2 labeling experiments. The photosynthetic responses to CO2 and O2 were measured to determine whether onion leaves exhibited a typical C3-type response. A linear increase in CO2 uptake was observed in intact leaves up to 315 L L–1 of external CO2 and, at this external CO2 concentration, uptake was inhibited 35.4±0.9% by 210 mL L–1 O2 compared to 20 mL L–1 O2. Scanning electron micrographs of the leaf cavity wall revealed degenerated tissue covered by a membrane. The influence of different foliar applications of the triacontanol (Tria.) based plant grow regulator-Miraculan, on growth, CO2 exchange and essential oil accumulation in Lemongrass (Cymbopogon flexuosus L.) Steud. Watts. was studied in glass-house conditions. The yield components, viz., plant height, tillers per plant, biomass yield, accumulation of essential oil, net CO2 exchange and transpiration rates increased with Miraculan treatment of 0.4 g/ml compared to the untreated control, but the number of leaves per tiller remained unaffected. Application of Miraculan at 0.4 g/ml increased micronutrient uptake, total chlorophyll and citral content but decreased chlorophyll a/b ratio and stomatal resistance. Increase in shoot biomass, photosynthesis and chlorophyll were significantly correlated with essential oil content.  相似文献

Responses of the rate of net CO2 assimilation (A) to the intercellular partial pressure of CO2 (p i ) were measured on intact spinach (Spinacia oleracea L.) leaves at different irradiances. These responses were analysed to find the value of p i at which the rate of photosynthetic CO2 uptake equalled that of photorespiratory CO2 evolution. At this CO2 partial pressure (denoted ), net rate of CO2 assimilation was negative, indicating that there was non-photorespiratory CO2 evolution in the light. Hence was lower than the CO2 compensation point, . Estimates of were obtained at leaf temperatures from 15 to 30°C, and the CO2/O2 specificity of ribulose 1,5-bisphosphate (RuBP) carboxylase/oxygenase (E.C. 4.1.1.39) was calculated from these data, taking into account changes in CO2 and O2 solubilities with temperature. The CO2/O2 specificity decreased with increasing temperature. Therefore we concluded that temperature effects on the ratio of photorespiration to photosynthesis were not solely the consequence of differential effects of temperature on the solubilities of CO2 and O2. Our estimates of the CO2/O2 specificity of RuBP carboxylase/oxygenase are compared with in-vitro measurements by other authors. The rate of nonphotorespiratory CO2 evolution in the light (R d ) was obtained from the value of A at . At this low CO2 partial pressure, R d was always less than the rate of CO2 evolution in darkness and appeared to decrease with increasing irradiance. The decline was most marked up to about 100 mol quanta m-2 s-1 and less marked at higher irradiances. At one particular irradiance, however, R d as a proportion of the rate of CO2 evolution in darkness was similar in different leaves and this proportion was unaffected by leaf temperature or by [O2] (ambient and greater). The reaction of methanol dehydrogenase with cytochrome c L from Methylophaga marina and the reactions of the non-physiological substrates, Wurster's blue and ascorbic acid, with both proteins were studied as a function of temperature (4–32 °C), pressure (1–2000 bar) and ionic strength using the optical high pressure stopped-flow method. The thermodynamic parameters H, S and V were determined for all reactions where electron transfers are involved. These data allowed the determination of the Maxwell relationships which proved the internal thermodynamic consistency of the system under study. Two-year-old sweet chestnut trees were grown outside in normal or double CO2 atmospheric concentration. In spring and in autumn of two growing seasons, a six day labelling pulse of14C labelled CO2 was used to follow the carbon assimilation and distribution in the plant-soil system. Doubling atmospheric CO2 had a significant effect on the tree net carbon uptake. A large proportion of the additional C uptake was lost through the root system. This suggests that increased C uptake under elevated CO2 conditions increases C cycling without necessarily increasing C storage in the plant. Total root derived material represented a significant amount of the extra-assimilated carbon due to the CO2 treatment and was strongly correlated with the phenological stage of the tree. Increasing root rhizodeposition led to a stimulation of microbial activity, particularly near the end of the growing season. When plant rhizodeposition was expressed as a function of the root dry weight, the effect of increasing CO2 resulted in a higher root activity. The C to N ratios were significantly higher for trees grown under elevated CO2 except for the fine root compartment. An evaluation of the plant-soil system nitrogen dynamics showed, during the second season of CO2 treatment, a decrease of soil N mineralization rate and total N uptake for trees grown at elevated CO2 levels.  相似文献

The rate of CO2- and p-benzoquione-dependent photosynthetic O2 evolution by Anabaena variabilis cells remained unaltered and the rate of O2 uptake observed after switching off the light (endogenous respiration) was enhanced by a factor of 6–8 when the O2 concentration was increased from 200 to 400 M. Photosystem-I-linked O2 uptake and respiration of the cells incubated with ascorbate and N,N,NN-tetramethyl-p-phenylenediamine was not appreciable influenced by the O2 concentration. 2-Iodo-6-isopropyl-3-methyl-2,4,4-trinitrodiphenyl ether, blocking electron transfer at the plastoquinone level, suppressed O2 evolution and had no influence on endogenous respiration. 2-n-Heptyl-4-hydroxyquinoline-N-oxide, an inhibitor of electron transfer between photosystems II and I, as well as the cytochrome-oxidase inhibitors N 3 - , CN- and NH2OH, caused a 35–50% retardation of endogenous respiration and blocked photosynthetic O2 evolution. The molar ratio of cytochromes b6, f, c-553, aa3 and photosystem-I reaction centers in the isolated membranes equalled approx. 2:1:2:0.7:2. The regulation of Crassulacean acid metabolism (CAM) in the fern Pyrrosia piloselloides (L.) Price was investigated in Singapore on two epiphytic populations acclimated to sun and shade conditions. The shade fronds were less succulent and had a higher chlorophyll content although the chlorophyll a:b ratio was lower and light compensation points and dark-respiration rates were reduced. Dawn-dusk variations in titratable acidity and carbohydrate pools were two to three times greater in fronds acclimated to high photosynthetically active radiation (PAR), although water deficits were also higher than in shade fronds. External and internal CO2 supply to attached fronds of the fern was varied so as to regulate the magnitude of CAM activity. A significant proportion of titratable acidity was derived from the refixation of respiratory CO2 (27% and 35% recycling for sun and shade populations, respectively), as measured directly under CO2-free conditions. Starch was shown to be the storage carbodydrate for CAM in Pyrrosia, with a stoichiometric reduction of C3-skeleton units in proportion to malic-acid accumulation. Measurements of photosynthetic O2 evolution under saturating CO2 were used to compare the light responses of sun and shade fronds for each CO2 supply regime, and also following the imposition of a photoinhibitory PAR treatment (1600 mol·m-2·s-1 for 3 h). Apparent quantum yield declined following the high-PAR treatment for sun- and shade-adapted plants, although for sun fronds CAM activity derived from respiratory CO2 prevented any further reduction in photosynthetic efficiency. Recycling of respiratory CO2 by shade plants could only partly prevent photoinhibitory damage. Photosynthetic carbon fixation of freshly isolated cells of Prochloron, the symbiont of Lissoclinum patella, proceeded at high rates (80–180 mol O2·mgChl-1·h-1) in buffered seawater and showed a typical light response, saturating at about 300 E·m-2·s-1. However, in NaCl solutions osmotically equivalent to seawater CO2-dependent O2 evolution ceased or was severely inhibited. Hypotonic or hypertonic conditions induce degrees of swelling or shrinkage, respectively, apparently causing similar increases in the plasmamembrane's permeability to ferricyanide. Initially high, but rapidly declining, rates of electron transport were observed when the cells were suspended in distilled water. This inhibition was not caused by rupture of the cells, indicating instead diffusive loss of some essential factor(s) which normally exchange easily and rapidly between the cells and/or the host environment. The light dependence of quantum yields of Photosystem II (II) and of CO2 fixation were determined in C3 and C4 plants under atmospheric conditions where photorespiration was minimal. Calculations were made of the apparent quantum yield for CO2 fixation by dividing the measured rate of photosynthesis by the absorbed light [A/I=CO2 and of the true quantum yield by dividing the estimated true rate of photosynthesis by absorbed light [(A+Rl)/Ia=CO2·], where RL is the rate of respiration in the light. The dependence of the II/CO2 and II/CO2 * ratios on light intensity was then evaluated. In both C3 and C4 plants there was little change in the ratio of II/CO2 at light intensities equivalent to 10–100% of full sunlight, whereas there was a dramatic increase in the ratio at lower light intensities. Changes in the ratio of II/CO2 can occur because respiratory losses are not accounted for, due to changes in the partitioning of energy between photosystems or changes in the relationship between PS II activity and CO2 fixation. The apparent decrease in efficiency of utilization of energy derived from PS II for CO2 fixation under low light intensity may be due to respiratory loss of CO2. Using dark respiration as an estimate of RL, the calculated II/CO2 * ratio was nearly constant from full sunlight down to approx 5% of full sunlight, which suggests a strong linkage between the true rate of CO2 fixation and PS II activity under varying light intensity. Measurements of photosynthesis rates and II were made by illuminating upper versus lower leaf surfaces of representative C3 and C4 monocots and dicots. With the monocots, the rate of photosynthesis and the ratio of II/CO2 exhibited a very similar patterns with leaves illuminated from the adaxial versus the abaxial surface, which may be due to uniformity in anatomy and lack of differences in light acclimation between the two surfaces. With dicots, the abaxial surface had both lower rates of photosynthesis and lower II values than the adaxial surface which may be due to differences in anatomy (spongy versus palisade mesophyll cells) and/or light acclimation between the two surfaces. We determined for Vicia faba L the influence of nitrogen uptake and accumulation on the values of photon saturated net photosynthetic rate (P Nmax), quantum yield efficiency (), intercellular CO2 concentration (C i), and carboxylation efficiency (C e). As leaf nitrogen content (NL) increased, the converged onto a maximum asymptotic value of 0.0664±0.0049 mol(CO2) mol(quantum)–1. Also, as NL increased the C i value fell to an asymptotic minimum of 115.80±1.59 mol mol–1, and C e converged onto a maximum asymptotic value of 1.645±0.054 mol(CO2) m–2 s–1 Pa–1 and declined to zero at a NL-intercept equal to 0.596±0.096 g(N) m–2. fell to zero for an NL-intercept of 0.660±0.052 g(N) m–2. As NL increased, the value of P Nmax converged onto a maximum asymptotic value of 33.400±2.563 mol(CO2) m–2 s–1. P N fell to zero for an NL-intercept of 0.710±0.035 g(N) m–2. Under variable daily meteorological conditions the values for NL, specific leaf area (L), root mass fraction (Rf), P Nmax, and remained constant for a given N supply. A monotonic decline in the steady-state value of Rf occurred with increasing N supply. L increased with increasing N supply or with increasing NL.  相似文献

Micropropagated plantlets are fragile and often lack sufficient vigour to survive the acclimatization shock during transplantation to the soil. Effects of photosynthetic photon flux densities (PPFDs) on growth, photosynthesis and anatomy of micropropagated Doritaenopsis were studied after 4 months of acclimatization in a greenhouse at 25 °C. The plantlets were transferred to three different PPFDs for four months, i.e. low light (175), intermediate light (270) and high light (450 mol m–2 s–1). For most of the growth parameters measured i.e. leaf length, leaf area, leaf width, fresh weight, dry weight, chlorophyll (Chl) a/b ratio, were greater for the intermediate light levels after 4 months of acclimatization. The only exception was leaf thickness, which was increased more under high light levels. Results showed that the survival of Doritaenopsis plantlets was greatest (90%) in low light and intermediate light (89%) compared with only (73%) at high light. However, at low light levels, pigment concentrations (chlorophyll a, b and total chlorophyll) were higher. Net CO2 assimilation (A), stomatal conductance (g) and transpiration (E) were higher in plantlets grown at high level PPFD than at low after 4 months of acclimatization. Photosynthetic efficiency (Fv/Fm) decreased insignificantly; only at mid day for the high light treatment whereas leaf temperature and stomatal closure increased compared to low light. Scanning electron microscopic (SEM) images of leaves from acclimatized plantlets showed an increase in wax formation for the high light grown plantlets compared to those at low light. Microscopic analysis of acclimatized root sections showed highly developed multiseriate-velamen layers and higher root cell activity; while shoots had larger leaf air spaces than those of in vitro grown plantlets. These results suggest that physiological acclimation occurs at the intermediate PPFD (270 mol m–2 s–1) in Doritaenopsis compared to treatment at the high light level.  相似文献

Our investigations of diurnal variations of the 13C/12C ratio and CO2 content in soil air were carried out in three environments during periods of high biosphere activity. It has been observed that diurnal variation of CO2 concentration is negatively correlated 13. Particularly great variations occurred at shallow soil depths (10–30 cm) when the plant cover activity was high while the soil temperature was rather low. Under such conditions the 13 variations had the magnitude of 4, while the CO2 concentration varied more than doubly. The maximum of the 13C/12C ratlo and the minimum of the CO2 concentration in a cultivated field with winter wheat took place in the afternoon, whereas in deciduous forest similar patterns were observed at dawn. In these cases soil temperatures at 10 cm depths varied less than 2°C. Hence, under wheat the variation in root respiration rate seem to be the main reason of the recorded varations. In an uncultivated grass-field during the hottest period in summer we did not measure any distinct variations of CO2 properties in spite of the fact that soil temperature varied up to 5°C. This might be due to dominant microbial respiration at the high soil temperature, which exceeded 20°C.  相似文献
