A biotype of Avena sterilis ssp. ludoviciana is highly resistantto a range of herbicides which inhibit a key enzyme in fattyacid synthesis, acetyl-CoA carboxylase (ACCase). Possible mechanismsof herbicide resistance were investigated in this biotype. Acetyl-CoAcarboxylase from the resistant biotype is less sensitive toinhibition by herbicides to which resistance is expressed. I50values for herbicide inhibition of ACCase were 52 to 6 timesgreater in the resistant biotype than in the susceptible biotype.This was the only major difference found between the resistantand susceptible biotypes. The amount of ACCase in the meristemsof the resistant and susceptible is similar during ontogenyand no difference was found in distribution of ACCase betweenthe two biotypes. Uptake, translocation and metabolism of [14C]diclofop-methylwere not different between the two biotypes. In vivo, ACCaseactivity in the meristems of the susceptible biotype was greatlyinhibited by herbicide application whereas only 25% inhibitionoccurred in the resistant biotype. Depolarisation of plasmamembrane potential by 50 µM diclofop acid was observedin both biotypes and neither biotype showed recovery of themembrane potential following removal of the herbicide. Avena sterilis (sterile oat) is one of the most extended and harmful weeds in Mediterranean cereal crops. A process‐based niche model for this species was developed using CLIMEX. The model was validated and used to assess the potential distribution of A. sterilis in Europe under the current climate and under two climate change scenarios. Both scenarios represent contrasting temporal patterns of economic development and CO2 emissions. The projections under current climate conditions indicated that A. sterilis does not occupy the full extent of the climatically suitable habitat available to it in Europe. Under future climate scenarios, the model projection showed a gradual advance of sterile oat towards Northeastern Europe and a contraction in Southern Europe. The infested potential area increases from the current 45.2% to 51.3% in the low‐emission CO2 scenario and to 59.5% under the most extreme scenario. These results provide the necessary knowledge for identifying and highlighting the potential invasion risk areas and for establishing the grounds on which to base the planning and management measures required. The main actions should be focused on controlling the large‐scale seed scattering, preventing seed dispersal into potentially suitable areas.  相似文献

Many herbicide-resistant weed species are polyploids, but far too little about theevolution of resistance mutations in polyploids is understood. Hexaploid wild oat(Avena fatua) is a global crop weed and many populations have evolved herbicideresistance. We studied plastidic acetyl-coenzyme A carboxylase (ACCase)-inhibitingherbicide resistance in hexaploid wild oat and revealed that resistant individuals canexpress one, two or three different plastidic ACCase gene resistance mutations(Ile-1781-Leu, Asp-2078-Gly and Cys-2088-Arg). Using ACCase resistance mutations asmolecular markers, combined with genetic, molecular and biochemical approaches, we foundin individual resistant wild-oat plants that (1) up to three unlinked ACCase gene lociassort independently following Mendelian laws for disomic inheritance, (2) all three ofthese homoeologous ACCase genes were transcribed, with each able to carry its own mutationand (3) in a hexaploid background, each individual ACCase resistance mutation confersrelatively low-level herbicide resistance, in contrast to high-level resistance conferredby the same mutations in unrelated diploid weed species of the Poaceae (grass) family. Lowresistance conferred by individual ACCase resistance mutations is likely due to a dilutioneffect by susceptible ACCase expressed by homoeologs in hexaploid wild oat and/ordifferential expression of homoeologous ACCase gene copies. Thus, polyploidy in hexaploidwild oat may slow resistance evolution. Evidence of coexisting non-target-site resistancemechanisms among wild-oat populations was also revealed. In all, these results demonstratethat herbicide resistance and its evolution can be more complex in hexaploid wild oat thanin unrelated diploid grass weeds. Our data provide a starting point for the daunting taskof understanding resistance evolution in polyploids.  相似文献

The growth and development of field-grown Avena fatua plants were studied for autumn and spring sowings in two consecutive years. The duration of various growth stages from sowing until anthesis was quantified in terms of thermal time (accumulated degree days) or photothermal time (degree days modified by photoperiod). Base temperatures and photoperiods for developmental phases were estimated as those which minimised the coefficient of variation among sowing dates. Relationships were derived between leaf emergence, canopy height, plant leaf area, and photothermal time. Stem extension and flowering occurred earlier in autumn-sown plants than spring-sown plants. Autumn-sown plants produced more leaves on the main stem, and had greater leaf area and above-ground biomass at anthesis than spring-sown plants.  相似文献

An eco-physiological simulation model of the growth and development of Avena fatua was parameterised and tested. The model simulates growth ofA. fatua, in kg dry matter ha-1 day-1 from sowing to maturity as a function of irradiance, temperature and various species characteristics. Parameter values were derived from the literature and from field experiments, including both autumn and spring sowings of A. fatua over three years at two sites in southern England. With two exceptions, a single set of parameter values was sufficient to accurately simulate the emergence, growth and development of both autumn and spring cohorts over all years and sites. The two exceptions were the result of differences between autumn and spring cohorts of A. fatua in the rate of early leaf area growth and in the relationship between specific leaf area and developmental stage.  相似文献

Experiments were done to examine the effects of controlling wild-oats and autumn-germinating broad-leaved weeds in winter wheat, early in winter or late in spring. The herbicides used were barban (winter), chlortoluron or isoproturon (winter), and benzoylprop-ethyl, at the recommended doses and at half doses. Sequential treatments of two herbicides at half doses were also examined. All treatments were given a routine broad-leaved herbicide treatment in spring. Yields of wheat were influenced more by the time of weed removal than by the degree of control achieved. Grain yields at three sites with dense autumn broad-leaved weed populations were greatest following the use of chlortoluron or isoproturon. At three other sites with moderate to dense wild-oat populations (60 to 240 plants/m1), the use of barban at the crop three-leaf stage gave larger yields than benzoylprop-ethyl in late spring at the early stem elongation stage of the crop. Seed formation from surviving A. fatua was similar with both wild-oat herbicides. The treatment which reduced seed production of A. fatua and maintained crop yield most consistently was barban followed by benzoylprop-ethyl, each at half the normal recommended dose.  相似文献

Allohexaploid wild oat, Avena fatua L. (Poaceae; 2n = 6x = 42), is one of the world's worst weeds, yet unlike some of the other Avena hexaploids, its genomic structure has been relatively little researched. Consequently, in situ hybridisation was carried out on one accession of A. fatua using an 18S-25S ribosomal DNA (rDNA) sequence and genomic DNA from A. strigosa (AA-genome diploid) and A. clauda (CC-genome diploid) as probes. Comparing these results with those for other hexaploids studied previously: (i) confirmed that the genomic composition of A. fatua was similar to the other hexaploid Avena taxa (i.e., AACCDD), (ii) identified major sites of rDNA on three pairs of A/D-genome chromosomes, in common with other Avena hexaploids, and (iii) revealed eight chromosome pairs carrying intergenomic translocations between the A/D- and C-genomes in the accession studied. Based on karyotype structure, the identity of some of these recombinant chromosomes was proposed, and this showed that some of these could be divided into two types, (i) those common to all hexaploid Avena species analysed (3 translocations) and (ii) one translocation in this A. fatua accession not previously observed in reports on other hexaploid Avena species. If this translocation is found to be unique to A. fatua, then this information, combined with more traditional morphological data, will add support to the view that A. fatua is genetically distinct from other hexaploid Avena species and thus should retain its full specific status.  相似文献

The response of wild oat to imazamethabenz varies with the growth stage, but the role of tillers in this regard is unclear. Removal of tillers at the three-leaf stage before spraying with imazamethabenz did not significantly affect the total shoot fresh weight measured 3 weeks later. The leaf area and dry weight of intact plants at the three-leaf stage were 17–21% greater than for plants with coleoptilar and first leaf main shoot tillers (T0 and T1) removed. The greater leaf area may have increased herbicide interception per plant. Similar fresh weight reductions in main shoot, total tillers, and total shoots were found whether imazamethabenz was applied to the plant at the two-leaf without tillers or the three-leaf with two tillers stage. Imazamethabenz applied only to the main shoot reduced total shoot dry weight more than an equivalent amount of imazamethabenz applied only to tiller T1 or applied over the whole shoot. Imazamethabenz had the least inhibitory effect on whole plant growth when applied only to T1. When 14C-herbicide was applied to the first main shoot leaf of plants at the three-leaf stage with two tillers, the 14C translocated 38% to roots, 33% to the main shoot, and nearly 30% to all tillers. When 14C-herbicide was applied to the first leaf of T1 then the 14C translocated 50% to T1, 25% to the main shoot, 20% to roots, and 5% to all other tillers. The translocation pattern and fresh weight values suggested that the presence of early tillers during herbicide application neither increased nor decreased imazamethabenz efficacy in wild oat. Background and Aims

Wild carrot is the ancestor of cultivated carrot and is the most important gene pool for carrot breeding. Transgenic carrot may be released into the environment in the future. The aim of the present study was to determine how far a gene can disperse in wild carrot populations, facilitating risk assessment and management of transgene introgression from cultivated to wild carrots and helping to design sampling strategies for germplasm collections.

Methods

Wild carrots were sampled from Meijendel and Alkmaar in The Netherlands and genotyped with 12 microsatellite markers. Spatial autocorrelation analyses were used to detect spatial genetic structures (SGSs). Historical gene dispersal estimates were based on an isolation by distance model. Mating system and contemporary pollen dispersal were estimated using 437 offspring of 20 mothers with different spatial distances and a correlated paternity analysis in the Meijendel population.

Key Results

Significant SGSs are found in both populations and they are not significantly different from each other. Combined SGS analysis indicated significant positive genetic correlations up to 27 m. Historical gene dispersal σg and neighbourhood size Nb were estimated to be 4–12 m [95 % confidence interval (CI): 3–25] and 42–73 plants (95 % CI: 28–322) in Meijendel and 10–31 m (95 % CI: 7–∞) and 57–198 plants (95 % CI: 28–∞) in Alkmaar with longer gene dispersal in lower density populations. Contemporary pollen dispersal follows a fat-tailed exponential-power distribution, implying pollen of wild carrots could be dispersed by insects over long distance. The estimated outcrossing rate was 96 %.

Conclusions

SGSs in wild carrots may be the result of high outcrossing, restricted seed dispersal and long-distance pollen dispersal. High outcrossing and long-distance pollen dispersal suggest high frequency of transgene flow might occur from cultivated to wild carrots and that they could easily spread within and between populations.  相似文献

The effects of a short exposure to red, far-red or alternate red/far-red light on the germination of seeds after-ripened for different periods of time were studied in dormant lines of wild oat ( Avena fatua L.). Three stages were distinguishable in the after-ripening period in the response of germination to light. Seeds stayed dormant and showed no response to light during stage I. Phytochrome-mediated germination was observed in seeds during stage II. The phytochrome action disappeared during stage III, i.e. seeds fully germinated following treatments of all light qualities. When the seeds were imbibed in polyethylene glycol solutions, dark germination was reduced and phytochrome again had an effect, which suggested the involvement of phytochrome in water uptake of the seed.  相似文献

A multivariate analysis to differentiate morphologically the populations of wild potatoes Solanum cardiophyllum ssp. cardiophyllum and S. cardiophyllum ssp. ehrenbergii was carried out. An analysis of the morphology and the viability of pollen of these potato populations was also made. The results of the morphometric analysis indicate that both subspecies are phenetically different. The pollen grain shape and size in ssp. cardiophyllum are different in northern and southern populations. Pollen diameter is significantly different between subspecies. The shape of the fitness function relating the decline in fitness with coefficient of inbreeding (f) can provide evidence concerning the genetic basis of inbreeding depression, but few studies have examined inbreeding depression across a range of f using noncultivated species. Futhermore, studies have rarely examined the effects of inbreeding depression in the maternal parent on offspring fitness. To estimate the shape of the fitness function, we examined the relationship between f and fitness across a range off from 0.000 to 0.875 for components of both male and female fitness in Cucurbita pepo ssp. texana. Each measure of female fitness declined with f, including pistillate flower number, fruit number, seed number per fruit, seed mass per fruit, and percentage seed germination. Several aspects of male fitness also declined with f, including staminate flower number, pollen number per flower, and the number of days of flowering, although cumulative inbreeding depression was less severe for male (0.34) than for female function (0.39). Fitness tended to decline linearly with f between f = 0.00 and f = 0.75 for most traits and across cumulative lifetime fitness (mean = 0.66), suggesting that individual genes causing inbreeding depression are additive and the result of many alleles of small effect. However, most traits also showed a small reduction in inbreeding depression between f = 0.75 and f = 0.875, and evidence of purging or diminishing epistasis was found for in vitro pollen-tube growth rate. To examine inbreeding depression as a maternal effect, we performed outcross pollinations on f = 0.0 and f = 0.5 mothers and found that depression due to maternal inbreeding was 0.07, compared to 0.10 for offspring produced through one generation of selfing. In at least some families, maternal inbreeding reduced fruit number, seed number and mass, staminate flower number, pollen diameter, and pollen-tube growth rate. Collectively these results suggest that, while the fitness function appears to be largely linear for most traits, maternal effects may compound the effects of inbreeding depression in multigenerational studies, though this may be partially offset by purging or diminishing epistasis.  相似文献

Controlled environmental experiments were carried out to determine the phytotoxicity of several graminicides on wild oat (Avena futua L.) as influenced by combination of drought and temperature stress or drought and low relative humidity. Compared with unstressed conditions (20/15°C plus adequate soil moisture), imazamethabenz phytotoxicity to wild oat was reduced significantly when plants were exposed to a combination of drought and high temperature (30/20°C) stress. Imazamethabenz phytotoxicity was reduced almost as much by high temperature stress alone as by a combined temperature and drought stress. When herbicides were applied to wild oat plants subjected to drought alone or to drought plus high temperature, the observed reduction in phytotoxicity from greatest to least was: fenoxaprop = diclofop > flamprop > imazamethabenz. Fenoxaprop performance was most inhibited by the combination of drought plus high temperature, although drought alone and to a lesser degree, high temperature alone, inhibited fenoxaprop action. High temperature had an adverse effect on the efficacy of fenoxaprop at lower application rates. Raising fenoxaprop application rates to 400 g ha−1 overcame the inhibition caused by high temperature alone but only partially alleviated the effect of drought combined with high temperature. When plants were grown under a low temperature regimen the imposition of drought stress had little effect on imazamethabenz phytotoxicity but did reduce fenoxaprop phytotoxicity. At 25/15°C drought reduced the phytotoxicity of fenoxaprop and diclofop greatly but had no significant impact on the performance of any of the herbicides examined, regardless of soil moisture regimen. Bitou bush (Chrysanthemoides monilifera ssp. rotundata; Asteraceae) is a major woody weed that competes with the native legume Acacia sophorae in coastal ecosystems of eastern Australia. Three glasshouse experiments examined whether litter or soil from beneath bitou bush or Acacia plants could influence seed germination and seedling growth of A. sophorae. The presence of litter decreased seed germinability and this effect was greater for bitou bush litter than for Acacia litter. Shoot growth was increased by the addition of Rhizobium after 40 days, irrespective of soil type. After 78 days, shoot and root biomass were significantly lower for seedlings grown in bitou bush soil than for those grown in Acacia soil. There was a non‐significant trend towards a lower median population of Rhizobium in the soil beneath bitou bush than in that beneath Acacia. The results demonstrated a slight effect of bitou bush on the growth of A. sophorae, which could, however, be overshadowed by the judicious use of herbicides or fire for weed control and revegetation.  相似文献

Plasma membranes can be isolated from a variety of plant tissues by first preparing a post-mitochondrial membrane fraction enriched in plasma membranes, by differential centrifugation, and partitioning this on a dextran-polyethylene glycol two-phase system. With wild oat aleurone, however, we observed that differential centrifugation could not be used to produce a microsomal fraction enriched in plasma membrane. Approximately 70% of the plasma membrane in aleurone homogenates was pelleted by sequential centrifugation at 100 g× 10 min and 1000 g× 10 min. The remainder sedimented at 112 000 g× 1 h. All the material that was pelletable by centrifugation was, therefore, subjected to dextran-polyethylene glycol two-phase partitioning. The plasma membrane marker enzymes glucan synthase II (GSII, EC 2. 4. 1. 34) and UDP-glucose:sterol glucosyltransferase (SGT, EC 2. 4. 1.) were enriched in the upper phase, whereas cytochrome c oxidase activity (EC 1. 9. 3. 1), a mitochondrial marker enzyme, was depleted. The presence of endoplasmic reticulum (ER) and protein body membranes in the phase system was assessed by probing western blots, of SDS-PAGE separated proteins, with polyclonal antiserum either to binding protein (BiP, an ER marker) or to tonoplast intrinsic protein (TIP, a protein body membrane marker). BiP and TIP were present in the lower phase, but were not detected in the upper phase. In addition, the polypeptide patterns of material in the upper and lower phases were very different. These observations suggested that high purity aleurone plasma membrane had been isolated. Although the procedure for isolating plasma membranes was applicable to both aleurone protoplasts and layers, the polypeptide patterns of plasma membranes prepared from these sources were very different. The major protein components of wild oat aleurone were 7 S and 12 S storage globulins. These proteins were present in the lower phase, but not in the plasma membrane enriched upper phase, after aqueous two-phase partitioning. Differential centrifugation studies showed that it was necessary to homogenise aleurone in a buffer of pH 6. 0 or less if a soluble protein fraction, essentially devoid of storage globulins, was to be obtained. The use of these fractionation techniques is discussed in relation to photoaffinity labelling of gibberellin (GA)-binding proteins in aleurone.  相似文献

A comparative study of protein synthesis has been carried out with embryos excised from dormant (D) and non-dormant (ND) caryopses of the wild oat. Although D embryos imbibed in water or ND embryos imbibed in abscisic acid do not germinate, they incorporate [14C]leucine into TCA-insoluble material for the first 48 h as readily as embryos that do germinate (ND embryos imbibed in water, or D embryos imbibed in gibberellic acid). Pulsechase experiments with [14]leucine show that in both D and ND embryos the proteins associated with the membranes undergo turnover. The rates of decay of incorporated radioactivity are similar in both dormant and germinating embryos up to 98 h following embryo excision. Fractionation of the membrane proteins in SDS-polyacrylamide gels indicates that the different polypeptides have different rates of turnover. It is concluded that membrane proteins in imbibed D embryos are in a state of constant turnover, and that this is a part of the replacement processes necessary to maintain the integrity of hydrated cells. In the presence of gibberellic acid (GA3) aleurone layers and isolated aleurone protoplasts of Avena fatua accumulate specific isozymes of acid phosphatase (EC 3.1.3.2). Some of these may be involved in mobilizing aleurone-grain phosphate reserves during germination. The hormone also controls secretion of other specific molecular forms of the enzyme that probably assist in endosperm hydrolysis. The accumulation and secretion of putative cell-wall-associated isozymes are stimulated by the action of GA3 in isolated protoplasts. Sodium nitrate and nitrite induced germination in a partly after-ripened dormant line of Avena fatua L. The dose-response curves for their stimulation of germination and oxygen uptake were similar, indicating that these compounds may stimulate germination by promoting oxygen uptake. Time-sequence studies showed that nitrate and nitrite stimulated oxygen uptake as much as 70% prior to the first signs of germination. A similar ammonium chloride treatment failed to induce germination or elevate the rate of oxygen uptake, indicating that nitrate and nitrite may promote these events by acting as electron acceptors. The stimulation of germination and oxygen uptake by nitrate and nitrite were not significantly inhibited by salicylhydroxamic acid, an inhibitor of alternative respiration. Thus, stimulation of germination and oxygen uptake by nitrate and nitrite do not require the operation of the alternative pathway of respiration. The stimulation of germination and oxygen uptake by nitrate and nitrite were not inhibited by sodium azide, an inhibitor of cytochrome-mediated respiration; however both germination and oxygen uptake were prevented when salicylhydroxamic acid and sodium azide were applied together. Thus, stimulation of germination and oxygen uptake by nitrate and nitrite require the operation of only one or other of the pathways of respiration; however a specific requirement for the operation of the alternative pathway of respiration does not exist. The function of these compounds as promotors of respiration is discussed with reference to dormancy and the involvement of the Krebs cycle.  相似文献
