Effects of water stress duration and intensity on gas exchange and leaf water potential were investigated in 7-month-old seedlings of a humid coastal provenance (Gympie) and a dry inland (Hungry Hills) provenance of E. cloeziana F. Muell. and in a dry inland (Chinchilla) provenance of E. argophloia Blakely supplied with 100% (T100), 70% (T70), 50% (T50) of their water requirements, or were watered only after they were wilted at dawn (T0). Seedlings of E. argophloia had the highest midday net photosynthetic rate (A), stomatal conductance (g s), stomatal density and predawn leaf water potential (pd) in all treatments. The E. cloeziana provenances did not differ in these attributes. The T70 and T50 treatments caused reductions in A of 30% in E. argophloia, and 55% in the E. cloeziana provenances. Under the T0 treatment, E. argophloia maintained higher rates of gas exchange at all levels of water stress than E. cloeziana provenances. The estimates of pd and midday water potential (md) at which plants remained wilted overnight were respectively: –2.7 and –4.1 MPa for E. cloeziana (humid), –2.8 and –4.0 MPa for E. cloeziana (dry) and, –3.7 and –4.9 MPa for E. argophloia. Following stress relief, both A and g s recovered more quickly in E. argophloia and in the dry provenance of E. cloeziana than in the humid provenance. We conclude that E. argophloia is more drought tolerant and has a potential for cultivation in the humid and semi humid climates, whilst E. cloeziana has greater potential in the humid subtropical climates.  相似文献

Host status of four leguminous cover crops [Canavalia ensiformis (L.) DC. (Jack bean), Crotalaria ochroleuca G. Don (Sunnhemp), Lablab purpureus L. (Hyacinth bean) and Mucuna pruriens (L.) DC. (velvet bean)] to Pratylenchus zeae Filipjev and effects of intercropping C. ensiformis and M. pruriens with Pan5195, H627 and Emap11 maize cultivars on P. zeae population and disease severity on maize were determined in greenhouse and field tests. Pratylenchus zeae significantly (P<0.05) reduced growth of C. ochroleuca by 36% but had no effect on C. ensiformis, M. pruriens and L. purpureus. While C. ensiformis, M. pruriens and L. purpureus reduced P. zeae population, C. ochroleuca increased it. In the greenhouse test, intercropping maize with C. ensiformis significantly (P<0.05) improved maize growth by up to 34%, Nematode populations in the roots of maize intercropped with either C. ensiformis or M. pruriens were significantly (P<0.05) reduced by up to 32% while nematode disease severity in these intercropping systems was reduced by up to 26%. In the field test, intercropping Emap11, Pan5195 and H627 with C. ensiformis significantly (P<0.05) increased maize grain yield by 190, 29 and 22%, respectively. Intercropping H627 with M. pruriens significantly (P<0.05) increased maize grain yield by 12%, but grain yields of Pan5195 and Emap11 declined by 79 and 40%, respectively. Root necrosis and soil nematode populations in the C. ensiformis–maize intercrop declined by up to 50 and 30%, respectively. Under pure maize stands, soil nematode populations increased by up to 35% in 90 days relative to the initial nematode population of three nematodes g−1 of fresh soil. Section Editor: P. A. M. Changes in body core temperature (T cor) and heat balance after an abrupt release of lower body negative pressure (LBNP) were investigated in 5 volunteers under the following conditions: (1) an ambient temperature (T a) of 20 °C or (2) 35 °C, and (3)T a of 25 °C with a leg skin temperature of 30°C or (4) 35°C. The leg skin temperature was controlled with water perfusion devices wound around the legs. Rectal (T re), tympanic (T ty) and esophageal (T es) temperatures, skin temperatures (7 sites) and oxygen consumption were measured. The intensity of LBNP was adjusted so that the amount of blood pooled in the legs was the same under all conditions. When a thermal balance was attained during LBNP, application of LBNP was suddenly halted. The skin temperatures increased significantly after the release of LBNP under all conditions, while oxygen consumption hardly changed. The release of LBNP caused significant falls inT cor s under conditions (1) and (3), but loweredT cor s very slightly under conditions (2) and (4). The changes inT es were always more rapid and greater than those ofT ty andT re. The falls inT ty andT re appeared to be explained by changes in heat balance, whereas the sharp drop ofT es could not be explained especially during the first 8 min after the release of LBNP. The results suggest that a fall inT cor after a release of LBNP is attributed to an increase in heat loss due to reflexive skin vasodilation and is dependent on the temperature of venous blood returning from the lower body. It is presumed thatT es may not be an appropriate indicator forT cor when venous return changes rapidly.  相似文献

The osteological development of the vertebral column and fins in shi drum Umbrina cirrosa was studied in order to improve knowledge for its introduction in Mediterranean aquaculture. The osteological development was studied in 171 individuals, of total length (LT) from 2·7 to 30·2 mm that were reared under the mesocosm technique. Vertebral ontogeny starts at 3·4 and 4·0 mm LT, with the formation of the first cartilaginous neural and haemal arches, and spines, respectively, and is completed with the full attainment of epicentrals (12·5 mm LT). The formation of vertebral centra occurs between 4·1 and 7·4 mm LT. Pectoral supports are the first fin elements to develop (3·0 mm LT), followed by those of the caudal fin (3·8 mm LT), pelvic fin (3·9 mm LT) and finally by those of the dorsal and anal fins (4·5 mm LT). The caudal fin is the first to develop fin rays and attain the full count of principal fin rays (4·5–6·8 mm LT), but the last to be fully completed with the formation of procurrent fin rays (6·9–17·5 mm LT). The next fins starting to present rays are the dorsal (5·3 mm LT) and the pectoral fins (5·6 mm LT), while the anal and pelvic fins are the last (5·7 mm LT). Following the caudal principal fin rays (6·8 mm LT), the dorsal, anal (6·9 mm LT), pelvic (7·4 mm LT) and pectoral fins (9·8 mm LT) are the next with fully completed ray counts. Aggregation of qualitative changes, such as the appearance of cartilages, the beginning and the complement of the ossification process and the full complement of elements in U. cirrosa were measured as cumulative frequency counts. These measurements reveal three ontogenetic intervals: one very developmentally active period during early life stages (from 3 to 5·9 mm LT), a second slower developmental period (from 6·0 to 8·9 mm LT) and finally a period of ontogeny more focused on structure refinement up to metamorphosis and settlement (>9·0 mm LT).  相似文献

Sublethal effects of Thymus vulgaris were investigated on life-table parameters of Tetranychus urticae Koch (Acari: Tetranychidae) in the laboratory conditions at 25?±?1?°C, 70?±?10% RH and a 14:10?h (L:D) photoperiod. Bioassay was conducted to assess the effect of different concentrations of T. vulgaris essential oil on two-spotted spider mite. LC50 and LC20 of T. vulgaris on T. urticae were calculated as 18.86 and 6.24?μl/litre air for 24?h after treatment. In the sublethal effects, after 24?h exposure to LC50 concentration, mortality, fecundity and longevity of the females that survived from treatment were determined. Results showed that T. vulgaris caused significant reductions in fecundity and longevity of adults. Life-table assay indicated that the intrinsic rate of increase (r m), net reproductive rate (R 0), finite rate of increase (λ), mean generation time (T), significantly reduced in treated females compared to control. These results suggest that T. vulgaris could be incorporated in integrated pest management programmes of T. urticae.  相似文献

Summary Lectins of different carbohydrate specificities (GNA (Galanthus nivalis), con A (Canavalia ensiformis), VFL (Vicia faba), PSL (Pisum sativum), LCA (Lens culinaris), PNA (Arachis hypogaea; with or without prior neuraminidase treatment), WGA (Triticum vulgare), SBA (Glycine max), UEA-I (Ulex europaeus), LPA (Limulus polyphemus), BS-I B4 (Bandeiraea simplicifolia, isolectin B4)) were explored for use as differentiation markers of rumen epithelial cellsin vivo andin vitro. Lectins specific for mannose (GNA), mannose/glucose (con A, VFL, PSL and LCA),N-acetylglucosamine (WGA) or forN-acetylneuraminic acid (LPA) reacted generally with all types of rumen epithelial cell from both rumen tissue and cell culture. They were, therefore, not suitable markers of epithelial differentiation. SBA was unsuitable because, although it reacted with both tissue and cultured rumen epithelial cells, it was also bound to non-stratified areas of primary rumen epithelial cell cultures. Both BS-I B4 and PNA (after neuraminidase treatment) had to be ruled out because they did not react with differentiated rumen tissue epithelial cells, although they did bind to both stratified and non-stratified cultured cells. In contrast, UEA-I reacted strongly with differentiated rumen epithelial cells both from rumen tissue and cell cultures and therefore appears to be a good general marker for rumen epithelial cell differentiation.  相似文献

Summary Djungarian dwarf hamsters,Phodopus s. sungorus, were kept in natural photoperiodic conditions throughout the year, either inside at a constantT a of 23°C or outside subjected to seasonally varyingT a. Comparisons were made between hamsters from both conditions to evaluate the significance of seasonal changes in photoperiod and/orT a as environmental cues for seasonal acclimatization inPhodopus. Basal metabolic rate was lowest in July (1.68 ml/g·h) and highest in January (2.06 ml/g·h inPhodopus living outside), combined with a decrease inT 1c from 26°C in July to 20°C in January. This was parallelled by seasonal changes in body weight (summer 42 g, winter 25g), fur colouration, fur depth and the occurrence of short daily torpor.AtT a below thermoneutrality total energy requirements for thermoregulation in winter acclimatizedPhodopus were found 36% lower than summer values (e.g. at O°CT a in summer 1,160 mW, in winter 760 mW), which were effected by a combined strategy of reducing body weight (19%) together with improvements of thermal insulation of the body surface (17%). Desulfovibrio vulgaris lipopolysaccharide and outer membrane proteins (OMPs) were incorporated into vesicles ofD. vulgaris phospholipid and studied for [55Fe]binding activity. Both lipopolysaccharide and an extract of two major OMPs caused large increases in55Fe uptake over control (phospholipid only) vesicles. CommercialSalmonella typhimurium lipopolysaccharide gave a similar result, but the effect was inhibited by calcium ions; this was not the case forDesulfovibrio. The lipid A portion ofS. typhimurium lipopolysaccharide had a high iron-binding ability, whereasDesulfovibrio lipid A iron binding was little different from control values;D. vulgaris lipopolysaccharide thus has a specific iron-binding site within its polysaccharide side chain.  相似文献

Summary Minute ventilation (V E), tidal volume (V T), respiratory frequency (f) and clavicular air sac gas composition were measured in conscious domestic fowl breathing air and hypoxic gas mixtures at neutral (18±1°C) and raised (33±1°C) air temperatures. Increases inV E caused by inhalation of 10%, 8% or 6.5% O2 in N2, respectively, were independent of temperature although at each level the absoluteV E was ca. 21·min−1 greater in the panting birds. Changes in respiratory pattern during hypoxia were markedly dependent on temperature. At 18°C almost all of the increasedV E resulted from increasedf. At 33°C hypoxia led to a strong suppression off and increase inV T. It is concluded that hyperthermia and hypoxia are additive and non-interactive in their effects on ventilatory drive, in agreement with previously reported effects of hypercapnia and physical exercise on breathing in panting fowl.  相似文献

The higher proportion of repeated DNA sequences in the garden pea (Pisum sativum) than in the mung bean (Vigna radiata), as well as other differences between these legume genomes, are consistent with a higher rate of sequence amplification in the former. This hypothesis leads to a prediction that repeated sequence families inPisum are mostly heterogeneous, as defined by Bendich and Anderson (1977), whileVigna families are homogeneous. An assay developed by these authors to distinguish between the two types of families, by comparison of reassociation rates at different temperatures, was utilized. The results forVigna defied the predictions of the assay for either homogeneous or hetereogeneous model. Evaluation of the kinetic data in light of the great diversity of repeated family copy numbers in both genomes enabled an interpretation of the results as consistent with hetereogenous families inPisum and homogeneous families inVigna. The reproductive biology of Centropomus parallelus was described from 589 individuals captured in estuarine and coastal waters in Southern Brazil. Length-frequency distribution showed the dominance of males in smaller length-classes (132–290 mm LT), whereas females were dominant in larger length-classes (>290 mm LT). Total length at maturity (L50) was 180 mm LT and corresponded to 29% of the maximum length recorded. Histological sections revealed one hermaphrodite (205 mm LT) and few immature females. Life history traits provided herein can contribute to sustainable fisheries management practices.  相似文献

Edible basidiomycetesFlammulia velutipes andPleurotus ostreatus were cultivated in the usual manner on media based on sawdust and rice bran, and the cultures were exposed to slowly flowing CO2-enriched air (550 (control), 3,000, 6,000, and 9,000µl/l) for seven days at different stages of cultivation. When the cultures were exposed at the primordium stage (less than 10 mm in length), length and yield of fruit-bodies increased and pileus expansion was slightly inhibited inF. velutipes, while inP. ostreatus length increased, yield decreased, and pileus expansion was greatly inhibited. When the cultures with fruit-bodies larger than 10 mm were exposed, length and yield were insensitive and pileus expansion was greatly inhibited inF. velutipes, while inP. ostreatus length was insensitive, but pileus expansion was heavily damaged by trumpet-like deformation and yield decreased. The different action of CO2 on the two species appeared to be due to the different anatomical structures of their fruit-bodies.  相似文献

Agonist activation of the hP2Y1 receptor expressed in Xenopus oocytes stimulated an endogenous voltage-gated ion channel, previously identified as the transient inward (Tin) channel. When human P2Y1 (hP2Y1) and skate P2Y (sP2Y) receptors were expressed in Xenopus oocytes, time-to-peak values (a measure of the response to membrane hyperpolarization) of the Tin channel were significantly reduced compared to oocytes expressing the hB1-bradykinin receptor or the rat M1-muscarinic (rM1) receptor. Differences in activation were also observed in the Tin currents elicited by various P2Y receptor subtypes. The time-to-peak values of the Tin channel in oocytes expressing the hP2Y4, hP2Y11, or hB1-bradykinin receptors were similar, whereas the channel had significantly shorter time-to-peak values in oocytes expressing either the hP2Y1 or sP2Y receptor. Amino acid substitutions at His-132, located in the third transmembrane domain (TM3) of the hP2Y1 receptor, delayed the onset of channel opening, but not the kinetics of the activation process. In addition, Zn2+ sensitivity was also dependent on the subtype of P2Y receptor expressed. Replacement of His-132 in the hP2Y1 receptor with either Ala or Phe increased Zn2+ sensitivity of the Tin current. In contrast, truncation of the C-terminal region of the hP2Y1 receptor had no affect on activation or Zn2+ sensitivity of the Tin channel. These results suggested that TM3 in the hP2Y1 receptor was involved in modulating ion channel function and blocker pharmacology of the Tin channel.  相似文献

Summary D. maculata, the white-faced hornet, stabilized (regulated) thoracic temperature (T Th) over wide ranges of ambient temperature (T a), whileV. vulgaris, the common yellowjacket, regulatedT Th poorly. The hornets also maintained a higherT Th than the wasps, sometimes heating 38°C aboveT a. Attacking individuals of both species had higherT Th than those either leaving or returning to the nest from foraging. The hornets, who are primarily hunters of live prey, showed peak activity near dawn, and they were as active atT a=2°C as at 20°C. Being able to regulate theirT Th and fly at the lowT a should enhance their ability to capture small insects that are usually torpid at theseT a. The yellowjacket wasps, on the other hand, who are scavengers as well as hunters, did not leave the nest at 2°C; their activity decreased greatly with decreasingT a. Chromosome numbers for 26 different species of the generaPiper, Peperomia andPothomorphe (Piperaceae) are reported. The basic chromosome numbers are 2n = 26, x = 13 (Piper, Pothomorphe) and 2n = 22, x = 11 (Peperomia), polyploid series are characteristic forPiper andPeperomia. Piper has the smallest chromosomes and prochromosomal interphase nuclei,Peperomia the largest ones and mostly reticulate to euchromatic nuclei.Pothomorphe is intermediate in both characters. The karyomorphological differences betweenPothomorphe andPiper underline their generic separation. Interspecific size variation of chromosomes occurs inPiper andPeperomia. Infraspecific polyploidy was observed inPiper betle. C-banding reveals different patterns of heterochromatin (hc) distribution between the genera investigated. Kinetics of Catalase Inactivation Induced by Ultrasonic Cavitation   总被引:1,自引:0,他引:1  
Kinetic patterns of sonication-induced inactivation of bovine liver catalase (CAT) were studied in buffer solutions (pH 4.0–11.0) within the temperature range from 36 to 55o. Solutions of CAT were exposed to LF (20.8 kHz) ultrasound (specific power, 48–62 W/cm2). The kinetics of CAT inactivation was characterized by effective first-order rate constants (s–1) of total inactivation (k in), thermal inactivation (*k in), and ultrasonic inactivation (k in(us)). In all cases, the following inequality was valid: k in > *k in. The value of k in(us) increased with the ultrasound power (range, 48–62 W/cm2) and exhibited a strong dependence on the pH of the medium. On increasing initial concentration of CAT (0.4–4.0 nM), k in(us) decreased. The three rate constants were minimum within the range pH 6.5–8.0; their values increased considerably at pH < 6.0 and pH > 9.0. At 36–55o, the temperature dependence of k in(us) was characterized by an activation energy (E act) of 19.7 kcal/mol, whereas the value of E act for CAT thermoinactivation was equal to 44.2 kcal/mol. Bovine and human serum albumins (BSA and HSA, respectively) inhibited sonication-induced CAT inactivation; complete prevention was observed at concentrations above 2.5 g/ml. Dimethyl formamide (DMFA), a scavenger of hydroxyl radicals (O ), prevented sonication-induced CAT inactivation at 10% (k in and *k in increased with the content of DMFA at concentrations in excess of 3%). The results obtained indicate that free radicals generated in the field of ultrasonic cavitation play a decisive role in the inactivation of CAT, which takes place when its solutions are exposed to low-frequency ultrasound. However, the efficiency of CAT inactivation by the radicals is determined by (1) the degree of association between the enzyme molecules in the reaction medium and (2) the composition thereof.  相似文献

The major storage protein of jackbean (Canavalia ensiformis) has been purified by a protocol involving ammonium-sulphate precipitation, gel filtration and ion-exchange chromatography. The protein was shown by partial amino-acid-sequence data to be homologous to vicilin, a major storage protein of pea (Pisum sativum), and is thus a member of the family of legume 7S proteins exemplified by pea vicilin. This protein is thus referred to as jack-bean vicilin rather than canavalin or precanavalin as previously used. Other properties of the jack-bean vicilin (e.g. subunit relative molecular mass (Mr) and structure, resistance to proteolysis) show similarity to phaseolin, the major 7S storage protein ofPhaseolus vulgaris. Jack-bean vicilin contained no detectable -mannosidase activity, either as isolated from mature or germinating seeds, or after proteolytic treatment. -Mannosidase was also purified from jack beans, and was shown to have a subunit Mr of approx. 120,000; it was separated completely from jack-bean vicilin by a similar protocol to that used for purifying the latter. The -mannosidase was proteolytically cleaved after seed germination, but did not give polypeptides of the same Mr as jackbean vicilin. Cell walls of two strains ofChlorella vulgaris (from fresh water, F, and saline water, S) and ofKirchneriella lunaris contained various proportion of saccharides and proteins (highest percentage in dry matter was found inK. lunaris). They differed also in presence of individual monosaccharides (5 inC. vulgaris F, 8 inC. vulgaris S and 6 inK. lunaris) and amino acids (11 in C.vulgaris F, 6 in C.vulgaris S and 7 inK. lunaris). The lipophilic yeastsMalassezia furfur andM. pachydermatis show an initial rapid uptake ofl-leucine followed by slower steady-state rates. At least two independent transport systems forl-leucine were present in both species. The high-affinity system forM. furfur had a KT of 0.047 µM with a Jmax of 222 fM/min/106 cells (65 pM/min/mg dry weight), whereas forM. pachydermatis the KT was 0.067 µM with a Jmax of 709 fM/min/106 cells (89 pM/min/dry weight). The low-affinity system forM. furfur had a KT of 646 µM with a Jmax of 1.62 pM/min/106 cells (0.5 nM/min/mg dry weight) and that ofM. pachydermatis had a KT of 3.3 µM with a Jmax of 9.97 pM/min/106 cells (1.3 nM/min/mg dry weight). Both transport systems were energy-dependent. Cells incubated with Tween 80 showedl-leucine uptake via both transport systems. Cells incubated with a combination of glucose (1%) and Tween 80 (0.01%) showed decreased transport rates for the high-affinity system for both species as compared with cells incubated only with glucose. The low-affinity transport system of both species in the presence of glucose plus Tween 80 showed an initial rapid uptake followed by greater efflux than influx ofl-leucine.l-Leucine demonstrated binding to Tween 80, but the major effect of Tween 80 on membrane transport inMalassezia appears to be on the efflux of transported molecules.  相似文献

doi: 10.1111/j.1741‐2358.2012.00676.x Effect of thermal cycling and disinfection on colour stability of denture base acrylic resin Objectives: The purpose of this study was to investigate the effect of thermal cycling and disinfection on the colour change of denture base acrylic resin. Materials and Methods: Four different brands of acrylic resins were evaluated (Onda Cryl, QC 20, Classico and Lucitone). All brands were divided into four groups (n = 7) determined according to the disinfection procedure (microwave, Efferdent, 4% chlorhexidine or 1% hypochlorite). The treatments were conducted three times a week for 60 days. All specimens were thermal cycled between 5 and 55°C with 30‐s dwell times for 1000 cycles before and after disinfection. The specimens’ colour was measured with a spectrophotometer using the CIE L*a*b* system. The evaluations were conducted at baseline (B), after first thermal cycling (T1), after disinfection (D) and after second thermal cycling (T2). Colour differences (ΔE) were calculated between T1 and B (T1B), D and B (DB), and T2 and B (T2B) time‐points. Results: The samples submitted to disinfection by microwave and Efferdent exhibited the highest values of colour change. There were significant differences on colour change between the time‐points, except for the Lucitone acrylic resin. Conclusions: The thermal cycling and disinfection procedures significantly affected the colour stability of the samples. However, all values obtained for the acrylic resins are within acceptable clinical parameters.  相似文献
