树鼩作为多种人类疾病研究模型的可能性已受到广泛关注,但尚缺乏研究其免疫功能的基本标志以及单克隆抗体。该实验首先以树鼩外周血总RNA为材料,通过RT-PCR扩增得到长度为1365bp的树鼩CD4全长编码序列,并确定了数据库中缺失的两个片段,进而通过ClustalW等软件对其序列和分子特征进行分析,发现树鼩CD4氨基酸序列胞外和胞内域保守性较好,且与人类和猴的亲缘关系较近。虽然树鼩和人CD4分子表面大部分区域均带正电荷,但与人CD4胞外域D1相比,树鼩CD4D1结构区域表面带负电荷较多,且多出两个N-糖基化位点。这些差异对抗体的结合可能存在影响。该研究为今后树鼩CD4单克隆抗体制备及功能研究奠定了基础。  相似文献

The major histocompatibility complex (MHC) class I genes play a pivotal role in the adaptive immune response among vertebrates. Accordingly, in numerous mammals the genomic structure and molecular characterization of MHC class I genes have been thoroughly investigated. To date, however, little is known about these genes in tree shrews, despite the increasingly popularity of its usage as an animal model. To address this deficiency, we analyzed the structure and characteristic of the tree shrew MHC class I genes (Tube-MHC I) and performed a comparative gene analysis of the tree shrew and other mammal species. We found that the full-length cDNA sequence of the tree shrew MHC class I is 1074 bp in length. The deduced peptide is composed of 357 amino acids containing a leader peptide, an α1 and α2 domain, an α3 domain, a transmembrane domain and a cytoplasmic domain. Among these peptides, the cysteines, CD8+ interaction and N-glycosylation sites are all well conserved. Furthermore, the genomic sequence of the tree shrew MHC class I gene was identified to be 3180 bp in length, containing 8 exons and 7 introns. In 21 MHC class I sequences, we conducted an extensive study of nucleotide substitutions. The results indicated that in the peptide binding region (PBR) the rate of non-synonymous substitutions (dN) to synonymous substitutions (dS) was greater than 1, suggesting balancing selection at the PBR. These findings provide valuable contributions in furthering our understanding of the structure, molecular polymorphism, and function of the MHC class I genes in tree shrews, further improving their utility as an animal model in biomedical research.  相似文献

The epiphyseal union, eruptive sequence of the permanent teeth, and sexual maturity are examined in 150 common tree shrews (Tupaia glis), and are compared with those in other mammals including primates. The sequence of epiphyseal union is: elbow-hip-ankle-wrist-knee-shoulder. The sequence of epiphyseal union in mammals is not so fixed as has been previously considered. The eruptive sequence of the permanent teeth in the common tree shrew is more similar to that of insectivores than primates. The gonads are examined histologically to determine the sexual maturation. The order of completion of these three developmental aspects is also investigated and is compared with those in other mammals. Although the tree shrews have many similarities with platyrrhines in the development of the eco-sensitive organs such as the epiphyses of the extremities, their inclusion in the order Primates is still doubtful.  相似文献

Copies of two repetitive elements of the genome of common tree shrew (Tupaia glis) were cloned and sequenced. The first element, Tu III, is a approximately 260 bp long short interspersed element (SINE) with the 5'-end derived from glycine RNA. Tu III carries long polypurine- and polypyrimidine-rich tracts, which may contribute to the specific secondary structure of Tu III RNA. This SINE was also found in the genome of smooth-tailed tree shrew of another genus (Dendrogale). Tu III seems to be confined to the order Scandentia (tree shrews) since it was not found in DNA of other tested mammals. The second element Tu-SAT1 is a tandem repeat with a monomer length of 365 bp. Some properties of its nucleotide sequence suggest that Tu-SAT1 is a centromeric satellite.  相似文献

乳腺癌是严重危害女性健康的常见恶性肿瘤,建立合适的乳腺癌动物模型对于研究人类乳腺癌的生物学机制及发展新的防治方法至关重要。相对于常用的啮齿类动物,树鼩(Tupaia belangeri chinensis,tree shrew)因在进化层次上更接近于人类而可用于建立更适合的乳腺癌模型。该文详细了介绍一例树鼩自发性乳头状良性乳腺肿瘤。免疫组化结果显示该例肿瘤孕激素受体阳性且Ki-67阳性细胞比例显著增加;而活化的Caspase3阳性细胞比例较低;且肿瘤的形态和病理与人导管内乳头状肿瘤非常接近。提示利用树鼩建立乳腺肿瘤模型的可行性。  相似文献

Tree shrew(Tupaia belangeri) is currently placed in Order Scandentia and has a wide distribution in Southeast Asia and Southwest China.Due to its unique characteristics,such as small body size,high brain-to-body mass ratio,short reproductive cycle and life span,and low-cost of maintenance,tree shrew has been proposed to be an alternative experimental animal to primates in biomedical research.However,there are some debates regarding the exact phylogenetic affinity of tree shrew to primates.In this study,we determined the mtDNA entire genomes of three Chinese tree shrews(T.belangeri chinensis) and one Malayan flying lemur(Galeopterus variegatus).Combined with the published data for species in Euarchonta,we intended to discern the phylogenetic relationship among representative species of Dermoptera,Scandentia and Primates.The mtDNA genomes of Chinese tree shrews and Malayan flying lemur shared similar gene organization and structure with those of other mammals.Phylogenetic analysis based on 12 concatenated mitochondrial proteinencoding genes revealed a closer relationship between species of Scandentia and Glires,whereas species of Dermoptera were clustered with Primates.This pattern was consistent with previously reported phylogeny based on mtDNA data,but differed from the one reconstructed on the basis of nuclear genes.Our result suggested that the matrilineal affinity of tree shrew to primates may not be as close as we had thought.The ongoing project for sequencing the entire genome of Chinese tree shrew will provide more information to clarify this important issue.  相似文献

Infection with hepadnaviruses and exposure to aflatoxin B1 (AFB1) are considered to be major risk factors in the development of hepatocellular carcinoma (HCC) in humans. A high rate of p53 mutations at codon 249 has been reported in these tumors. The tree shrew (Tupaia belangeri chinensis) is a useful animal model for the development of HCC after human hepatitis B virus (HBV) infection or AFB1 treatment. Therefore, it was of particular interest to determine whether the p53 gene in tree shrew HCCs associated with HBV infection and/or with exposure to AFB1 is affected in the same manner as in human HCCs. We determined the tree shrew p53 wild-type nucleotide sequences by RT-PCR and automatic DNA-sequencing. Tree shrew wild-type p53 sequence showed 91.7 and 93.4% homologies with human p53 nucleotide and amino acids sequences, respectively, while it showed 77.2 and 73.7% homologies in mice. One HCC and normal liver tissue from AFB1 treated and one HCC from AFB1- and HBV-treated tree shrew showed no change in p53 sequences, while three HCCs from AFB1- and HBV-treated tree shrews showed point mutations in p53 sequences. One HCC showed point mutations at codon 275, which is on the DNA-binding domain of p53 gene, which might be a cause of gain-of-function during the development of HCC. As a result, our finding indicates that tree shrews exposed to AFB1 and/or HBV had neither codon 249 mutations nor significant levels of other mutations in the p53 gene, as is the case with humans.  相似文献

目的:建立一个合适的乳腺癌动物模型将在研究人类乳腺癌的发生、发展、转移等方面中发挥着越来越重要的作用。7,12-二甲基苯并蒽(7,12-dimethylbenz anthracene,DMBA)在实验中能诱导大鼠产生乳腺肿瘤。树鼩的基因的结构与人类的相似程度比啮齿类动物要高,而且树鼩的自发性乳腺癌已经有被报道,因而树鼩很有可能是研究乳腺肿瘤更合适的动物模型。因此我们想用致癌剂DMBA诱导树鼩产生乳腺肿瘤而建立树鼩的乳腺肿瘤模型。方法:在这个研究中,我们采用了十只在分娩之后失去幼崽的雌树鼩,其中一半的树鼩在腰部双侧乳房的脂肪垫注射100 mg/kg的DMBA,其余的树鼩作为对照组没有作DMBA处理。对生成的肿瘤组织进行病理切片HE染色的形态特点分析以及免疫组化化学法测定Ki-67、雌激素受体、孕酮受体、人表皮生长因子受体-2、E-钙粘蛋白、P120连环蛋白的表达。结果:通过诊断在DMBA处理的树鼩中,5分之1发展浸润性导管癌,其余发展成原位导管癌。结果还证明了诱导出来的乳腺肿瘤的形态学和病理学特征与人类的浸润性导管癌相似。结论:结果显示我们采用DMBA注射失去幼崽的雌树鼩的乳腺来诱导乳腺肿瘤是有效的,诱导出来的肿瘤组织学特征与人的乳腺癌相似,诱导的肿瘤组织表达目前人常用的乳腺癌相关分子生物学标记,并且表达情况与人的乳腺癌相似。这表明了DMBA诱导树鼩乳腺癌可以提供一个适合于研究人类乳腺癌发生、发展、转移和治疗的动物模型。  相似文献

干扰素(IFN)是在"危险信号"刺激下,由细胞分泌的具有抗病毒、抗肿瘤、抑制细胞增殖和免疫调节等多重作用的糖蛋白家族,在机体免疫系统中具有重要地位。树鼩作为多种人类疾病研究模型的前景已受到广泛关注,但对其IFN家族的研究尚属空白。该研究在现有的树鼩全基因组数据基础上,应用大片段核酸序列比对、基因预测等方法,对树鼩IFN家族的基本构成和分子特征进行预测和分析。结果显示,树鼩具有I型IFN:α(5个亚型)、β、ω、κ、ε、δ;II型IFN-γ;III型IFN:IFN-λ1、λ2/3,所编码的氨基酸序列及蛋白空间结构与其它哺乳动物具有较高的相似性,但在半胱氨酸位置和N糖基化个数上具有部分差异。该研究以全基因组数据对树鼩IFN家族信息进行系统挖掘和分析,为树鼩IFN的基因克隆以及其在感染免疫学中的作用和机理研究奠定了基础。  相似文献

This study was conducted to determine whether chronic psychosocial conflict alters the expression of glucocorticoid receptor (GR) mRNA in the hippocampus of male tree shrews (Tupaia belangeri).2. To generate probes for thein situ hybridization, the tree shrew GR gene was partly cloned. There was a 90% homology between the deduced amino acid sequence of the cloned tree shrew GR and that of the corresponding human GR sequence.35S-Labeled riboprobes which had been transcribed from the tree shrew GR clone hybridized to pyramidal neurons in all subregions of the tree shrew hippocampal formation and to granule neurons in the dentate gyrus.3. Afterin situ hybridization, the expression of GR mRNA was semiquantitatively determined by counting silver grains over single neurons of the hippocampal formation of psychosocially stressed tree shrews and control animals. After 12 days of social conflict, the number of silver grains in the CA1 and CA3 pyramidal neurons of stressed animals was significantly lower than in controls. No statistically significant differences in mRNA expression were observed in the pyramidal neurons of the subiculum and in the granule neurons of the dentate gyrus.4. The present results suggest that psychosocial stress leads to a site-specific down-regulation of hippocampal GR via modification of mRNA expression.  相似文献

最近基因组研究表明树鼩属于灵长类或是与灵长类亲缘关系最密切的姐妹种.因此,树鼩可能是应用于建立人类疾病动物模型的最佳动物之一.该文报道一种抑郁症的社会竞争失败病因学树鼩(Tupaia belangerichinensis)模型.一对雄性树鼩被饲养在一个双笼中,用网格把双笼隔开,网格上有一小门.适应1周后,把小门打开,这一对树鼩产生短暂的争斗,每天一次,连续21天.其中争斗失败者被称为服从者.这个过程可导致每天1 h的直接社交冲突和23 h的间接相互影响(比如通过气味、视觉等).与正常对照相比,失败者在第三周也就是社交冲突的最后一周显示了体重、自主活动、躲避行为、尿液皮质醇水平等的变化,并且这种改变可持续至少2周以上.此外,还报道全新的记忆模型,一种被捕获条件反射树鼩模型.在一个封闭的小房间中放置捕获笼,其中挂有一片苹果,小房间中有一只自由活动的树鼩.训练的前4次树鼩进入捕获笼吃苹果并不触发捕获笼关闭,但在第5次时触发捕获笼关闭,并持续一小时才释放树鼩.第1-5次树鼩进入捕获笼的延迟时时间作为适心性指标,其中第5次才是作为被捕获的一次学习训练.24 h后,测试树鼩进入捕获笼的延迟时间作为被捕获记忆能力指标.树鼩经过第5次被捕获训练,能形成很好的被捕获记忆,因为24 h后的延迟时间极人地增加.在训练前腹腔注射已知能阻断记忆形成的NMDA受体拮抗剂MK-801(0.2 mg/kg,腹腔注射),对适应指标没有显著影响,但足极大地缩短了24 h后测试的延迟时间,即阻断了被捕获记忆.这些结果表明了一种抑郁症的慢性社会竞争失败与学习和记忆的一次被捕获条件反射树鼩模型.这两种树趵模型对抑郁症与学习和记忆的机理研究、抗抑郁症新药的临床前药效学评价具有潜在的重要意义.
Abstract:
Recent genome studies indicate that tree shrew is in the order or a closest sister of primates, and thus may be one of the best animals to model human diseases. In this paper, we report on a social defeat model of depression in tree shrew (Tupaia belangeri chinensis). Two male tree shrews were housed in a pair-cage consisting of two independent cages separated by a wire mesh partition with a door connecting the two cages. After one week adaptation, the connecting door was opened and a brief fighting occurs between the two male tree shrews and this social conflict session consisted of 1 h direct conflict (fighting) and 23 h indirect influence (e.g. smell, visual cues) per day for 21 days. The defeated tree shrew was considered the subordinate. Compared with naive animals, subordinate tree shrews at the final week of social conflict session showed alterations in body weight, locomotion, avoidance behavior and urinary cortisol levels.Remarkably, these alterations persisted for over two weeks. We also report on a novel captive conditioning model of learning and memory in tree shrew. An automatic trapping cage was placed in a small closed room with a freely-moving tree shrew. For the first four trials, the tree shrew was not trapped when it entered the cage and ate the bait apple, but it was trapped and kept in the cage for 1 h on the fifth trial. Latency was defined as the time between release of the tree shrew and when it entered the captive cage. Latencies during the five trials indicated adaptation. A test trial 24 h later was used to measure whether the one-trial trapping during the fifth trial could form captive memory. Tree shrews showed much longer trapping latencies in the test trial than the adaptation trials. The N-methyl-d-aspartate (NMDA) receptor antagonist MK-801 (0.2 mg/kg, i.p.), known to prevent the formation of memory, did not affect latencies in the adaptation trails, but did block captive memory as it led to much shorter trapping latencies compared to saline treatment in the test trial. These results demonstrate a chronic social defeat model of depression and a novel one-trial captive conditioning model for learning and memory in tree shrews, which are important for mechanism studies of depression, learning,memory, and preclinical evaluation for new antidepressants.  相似文献

The use of tree shrews as experimental animals for biomedical research is a new practice. Several recent studies suggest that tree shrews are suitable for studying cancers, including breast cancer, glioblastoma,lung cancer, and hepatocellular carcinoma. However, the telomeres and the telomerase of tree shrews have not been studied to date. Here, we characterize telomeres and telomerase in tree shrews. The telomere length of tree shrews is approximately 23 kb, which is longer than that of primates and shorter than that of mice, and it is extended in breast tumor tissues according to Southern blot and flow-fluorescence in situ hybridization(FISH) analyses. Tree shrew spleen, bone marrow, testis, ovary, and uterus show high telomerase activities, which are increased in breast tumor tissues by telomeric repeat amplification protocol assays. The telomere length becomes shorter, and telomerase activity decreases with age. The tree shrew TERT and TERC are more highly similar to primates than to rodents. These findings lay a solid foundation for using tree shrews to study aging and cancers.  相似文献

目的调查滇西亚种树鼩体外寄生虫的自然感染状况,为建立树鼩质量控制标准提供依据。方法对野生的滇西亚种树鼩和自繁F1代树鼩各60只分别用尸检法和活体法检测,随即采用拔毛或用透明胶带粘取腋下、耳根、腹股沟、肛门附近等处的毛发制片,置于体视镜和显微镜下观察虫体和虫卵。结果野生滇西亚种树鼩和自繁F1代树鼩体外寄生虫自然感染率分别为:虱子25%/6%、蚤5%/0、蜱1.6%/0、水蛭1.6%/0、虱子和蚤混合感染1.6%/0。结论野生树鼩体外寄生虫的自然感染多为虱子和蚤,其次还有蝉、水蛭,总感染率明显高于自繁F1代。使用灭虱灵对于驱除树鼩体外寄生虫具有较好的疗效。  相似文献

We have sequenced four new mitochondrial genomes to improve the stability of the tree for placental mammals; they are two insectivores (a gymnure, Echinosorex gymnurus and Formosan shrew Soriculus fumidus); a Formosan lesser horseshoe bat (Rhinolophus monoceros); and the New Zealand fur seal (Arctocephalus forsteri). A revision to the hedgehog sequence (Erinaceus europaeus) is also reported. All five are from the Laurasiatheria grouping of eutherian mammals. On this new data set there is a strong tendency for the hedgehog and its relative, the gymnure, to join with the other Laurasiatherian insectivores (mole and shrews). To quantify the stability of trees from this data we define, based on nuclear sequences, a major four-way split in Laurasiatherians. This ([Xenarthra, Afrotheria], [Laurasiatheria, Supraprimates]) split is also found from mitochondrial genomes using either protein-coding or RNA (rRNA and tRNA) data sets. The high similarity of the mitochondrial and nuclear-derived trees allows a quantitative estimate of the stability of trees from independent data sets, as detected from a triplet Markov analysis. There are significant changes in the mutational processes within placental mammals that are ignored by current tree programs. On the basis of our quantitative results, we expect the evolutionary tree for mammals to be resolved quickly, and this will allow other problems to be solved.  相似文献

目的:探讨树鼩乳汁的基本成份,并和其它乳制品成份进行了比较分析。方法选取10只分娩后(1-21)d期间的哺乳期母树鼩,用人工被动母乳喂养方法让仔树鼩自由吮吸母乳,立即采用无菌操作的方法用注射器直接采取仔树鼩胃内的乳汁,1次/2 d,连续3-5次,每只母树鼩采取18 mL乳汁,按照国家标准的方法进行成分检测。结果树鼩乳汁总固形物为43.63%、脂肪为26.01%、蛋白质为10.41%、乳糖为0.45%、灰分为0.99%。树鼩乳的总固形物、灰分、蛋白质、脂肪、乳糖含量分别为牛乳的3.36、1.24、2.74、6.67、0.09倍;树鼩乳的总固形物、灰分、蛋白质、脂肪、乳糖含量分别为婴幼儿配方乳的1.44、0.20、0.58、1.53、0.06倍;与牛乳及婴幼儿配方乳矿物质成分含量相比,树鼩乳的矿物质成分钙、磷、钾、钠、镁、铁、锌含量均大于牛乳,分别为1.83、2.73、1.25、1.93、1.28、1.48倍,而均小于婴幼儿配方乳,分别为0.66、0.85、0.34、0.26、0.85、0.24、0.49倍。结论树鼩乳的主要营养成分呈现高脂高蛋白低糖的类型,可为树鼩人工育幼和饲养工作提供依据。  相似文献

参与人自体花结(A花结)形成的分子(如CD2/LFA-3),与免疫细胞的粘附和激活有关。我们曾发现,人和猴淋巴细胞表面的树鼩红细胞(TRBC)受体不同于绵羊红细胞(SRBC)受体(CD2),可能是一种新的白细胞分化抗原。花结试验表明,树鼩的外周血淋巴细胞(TPBL)和胸腺细胞都能形成A花结,结花率分别为20.9%和11.1%;而绵羊红细胞花结(E花结)形成率分别是20.9%和1.1%。以四种单克隆抗体(McAb)(Leu 5,0-275,AICD2.1和E2 McAb)进行树鼩A花结和E花结的抑制与抗原调变试验,结果表明,这些抗体对树鼩的A花结都没有明显的抑制或调变作用,但对E花结的抑制及调变作用明显。说明TPBL表面的TRBC受体不同于SRBC受体,与CD2/LFA-3及E2分子无关。因此,TPBL的A花结与E花结形成机制不同。  相似文献

Shrew species of the subfamily Soricinae have unusually high metabolic rates when compared to Crocidurinae shrews and other similar-sized mammals. The aim of this study was to clarify whether the high basal metabolic rate of Soricinae shrews is reflected in a high capillary density in their muscles. To this end, the capillary supply of four limb muscles and diaphragm of the common shrew (Sorex araneus) was quantified from cross-sectioned muscles. The capillary densities of the limb muscles were 2575 ± 329, 3111 ± 299, 2812 ± 197 and 2752 ± 173 capillaries mm−2 fibre area in gastrocnemius lateralis, g. medialis, plantaris and soleus, respectively. Capillary density of the shrew diaphragm (6691 ± 1057) was double that of the limb muscles. This value is among the highest ever measured in mammals. In general, the capillary supply in the hind limb of the common shrew is about 3–4 times higher than commonly found in the leg muscles of the laboratory rat or other bigger mammals, but similar to those in Crocidurinae shrews and some small rodents. Thus the high resting metabolism of the common shrew is not associated with an extraordinarily high capillary density. The apparent disparity between basal metabolic rate and muscle capillary supply in S. araneus is probably due to the small aerobic scope of shrews in the subfamily Soricinae. The tree shrew, a new experimental animal model, has been used to study a variety of diseases, especially diseases of the nervous system. 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is the gold standard for toxin-based animal models of Parkinson’s disease (PD) because MPTP treatment replicates almost all of the pathological hallmarks of PD. Therefore, in this study, the effects of MPTP on the motor function of the tree shrew were examined. After five daily injections of a 3 mg/kg dose of MPTP, the motor function of MPTP-injected tree shrews decreased significantly, and the classic Parkinsonian symptoms of action and resting tremor, bradykinesia, posture abnormalities, and gait instability were observed in most MPTP-injected tree shrews. HPLC results also showed significantly reduced striatal dopamine and 3,4-dihydroxyphenylacetic acid levels in tree shrews after MPTP injection. Increased oxidative stress levels are usually considered to be the cause of dopaminergic neuron depletion in the presence of MPTP and were observed in the substantia nigra of MPTP-treated tree shrews, as indicated by a significant reduction in superoxide dismutase and glutathione peroxidase activity and increased levels of malondialdehyde. In addition, elevated α-synuclein mRNA levels in the midbrain of MPTP-treated tree shrews were observed. Furthermore, MPTP-treated tree shrews showed the classic Parkinsonian symptoms at a lower MPTP dosage compared with other animal models. Thus, the MPTP-treated tree shrew may be a potential animal model for studying the pathogenesis of PD.  相似文献
