The objectives of the present work were in vitro propagation of Araucaria excelsa R. Br. var. glauca Carrière (Norfolk Island pine) with focus on the evaluation of the mean number of shoots per explant (MNS/E) and mean length of shoots per explants (MLS/E) produced by different parts of the orthotropic stem of A. excelsa R. Br. var. glauca in response to plant growth regulators. Norfolk Island pine axillary meristems responded very well to the 2-iso-pentenyl adenine (2iP) and thidiazuron (TDZ) levels. Explants taken from stem upper segments in the media containing 2iP had a higher MNS/E (3.47) and MLS/E (6.27 mm) in comparison to those taken from stem lower segments, which were 0.71 and 0.51 mm, respectively. Using 0.045 μM TDZ in the MS medium not only resulted in 4.60 MNS/E with 7.08 mm MLS/E but proliferated shoots showed a good performance as well. Investigating the best position of stem explant on mother plant as well as the best concentrations of growth regulators were performed which were useful for efficient micropropagation of this plant. Thirty three percent of explants were rooted in the MS medium containing 3 % sucrose, supplemented with 7.5 μM of both NAA and IBA for 2 weeks before transferring to a half strength MS medium without any growth regulator. Plantlets obtained were acclimatized and transferred to the greenhouse with less than 20 % mortality. This procedure considered the first successful report for regeneration and acclimatization of A. excelsa R. Br. var. glauca plantlet through main stem explants.  相似文献

Root treatments of barley (Hordeum distichum L.) plants with 10-7 to 10-4 M abscisic acid (ABA) caused an increase in proline content, especially at higher concentrations, within 2–3 h. Even 3 h after the removal of ABA from the medium the plants continued to accumulate proline. The higher the concentration of the ABA, the higher was the proline level at 6 h. When the highest ABA concentration, 10-4 M, was tested with polyethylene glycol (PEG) (-5.0 bars) in the medium, the ABA treatment resulted in a higher proline content than in control plants. The treatments PEG alone and PEG + ABA resulted in heavy accumulation of proline, especially, 3 h after releasing the plants from the stress. The proline content in PEG+ABA-treated plants was always higher than plants treated with PGE or ABA alone. In peas (Pisum sativum L. cv Alaska) the same trend occurred although to a lesser degree. From a quantitative inventory of sulphur forms and sulphur budget, the relation between the distribution of the various sulphur forms and the sulphate fluxes in three soil profiles has been addressed. These profiles are located in two forested watersheds at Mont Lozère. One has been sampled in a beech forest and the other two in a spruce forest and in a harvested plot of this spruce forest, respectively. The mean annual input-output budgets showed a sulphur immobilization in the soil cover of the three plots. In the preserved spruce forest plot, because of larger dry depositions, the sulphur immobilization is much greater than in other plots and occurs essentially in the B horizons. In the other two profiles, the dominant immobilization occurs in the parent material.The total sulphur content is very high in the forest floor reaching 2065 g S g-1 in the litter of one of the soils under spruce. In the organo-mineral horizons of soils under spruce, the total sulphur content decreases with depth and ranges from 310 to 520 g S g-1 in the A horizons to 100–200 g S g-1 in the parent material. In the profile under beech, the total sulphur content is lower except in the parent material. In all cases, the organic sulphur is the major part of sulphur often representing more than 90% of total sulphur. In organo-mineral soil horizons of the spruce forest, the part of the sulphateesters is more important than in the soil of the beech forest, probably related to the different nature of the microbial activity in the spruce forest. In contrast, the humification processes are more efficient in the soil under beech, which can be due to the greater input of organic sulphur by litterfall. It appears that the dominant organic sulphur form varies as a function of microbial ecology and sulphate flux. The maximum of the inorganic sulphate is located at the base of the B horizons in the soil of the spruce forest and in the parent material of the soil under beech. In these horizons, the high content of inorganic sulphate can be related to the higher amounts of amorphous Fe and Al phases.  相似文献

When pea plants (Pisum sativum L.) were subjected to flooding,abscisic acid (ABA) content in shoots and roots increased upto 8-fold in the following days and stomatal conductance significantlydecreased. Although young leaves of flooded plants had a slightlyhigher water potential than those of the unflooded plants, oldleaves had lower water potential and lost turgor at the timewhen a substantial ABA increase was detected. In plants wherethe old leaves were clipped off, flooding did not cause anyABA increase during 7 d of the experimental period, except underconditions of higher transpiration demand, when the increasein ABA content was both delayed and small in scale (only I-fold).When intact plants were flooded and ABA was assayed separatelyin both old and young leaves, the ABA increase in old leavespreceded that in young leaves. Evidence here suggests that theflooding-induced ABA increase mainly results from the wiltingof old leaves. This suggests that young leaves may be protectedfrom wilting by ABA originating in old leaves under unfavourableenvironmental conditions. Metabolite profiles and activities of key enzymes in the metabolism of organic acids, nitrogen and amino acids were compared between chlorotic leaves and normal leaves of ‘Honeycrisp’ apple to understand how accumulation of non-structural carbohydrates affects the metabolism of organic acids, nitrogen and amino acids. Excessive accumulation of non-structural carbohydrates and much lower CO2 assimilation were found in chlorotic leaves than in normal leaves, confirming feedback inhibition of photosynthesis in chlorotic leaves. Dark respiration and activities of several key enzymes in glycolysis and tricarboxylic acid (TCA) cycle, ATP-phosphofructokinase, pyruvate kinase, citrate synthase, aconitase and isocitrate dehydrogenase were significantly higher in chlorotic leaves than in normal leaves. However, concentrations of most organic acids including phosphoenolpyruvate (PEP), pyruvate, oxaloacetate, 2-oxoglutarate, malate and fumarate, and activities of key enzymes involved in the anapleurotic pathway including PEP carboxylase, NAD-malate dehydrogenase and NAD-malic enzyme were significantly lower in chlorotic leaves than in normal leaves. Concentrations of soluble proteins and most free amino acids were significantly lower in chlorotic leaves than in normal leaves. Activities of key enzymes in nitrogen assimilation and amino acid synthesis, including nitrate reductase, glutamine synthetase, ferredoxin and NADH-dependent glutamate synthase, and glutamate pyruvate transaminase were significantly lower in chlorotic leaves than in normal leaves. It was concluded that, in response to excessive accumulation of non-structural carbohydrates, glycolysis and TCA cycle were up-regulated to “consume” the excess carbon available, whereas the anapleurotic pathway, nitrogen assimilation and amino acid synthesis were down-regulated to reduce the overall rate of amino acid and protein synthesis.  相似文献

Proline is synthesized either from glutamate or from ornithine in plants. Relatively little is known about the contribution of the pathway from ornithine to proline biosynthesis. In this paper we investigated the contribution of ornithine--aminotransterase (OAT), an enzyme responsible for ornithine pathway, to proline accumulation in water-stressed detached rice leaves. Although OAT activity increased with the increase of water stress duration, a pattern similar to that obtained for proline accumulation, the ornithine pathway in rice leaves seems to contribute little, if any, to proline accumulation under water stress condition. This conclusion was based on the observations that (a) gabaculine (50 M), an inhibitor of OAT, inhibited about 75% OAT activity caused by water stress but reduced only 20% of proline content and (b) cycloheximide, a protein synthesis inhibitor, had no effect on OAT activity induced by water stress but significantly reduced proline accumulation.  相似文献

Ménière's disease (MD) is a disorder of the inner ear characterized by an insidious onset and aspecific symptoms, such as dizziness, vertigo, tinnitus, and hearing loss, that may become very debilitating. The presence of endolymphatic hydrops is a common feature in MD patients, but the pathophysiology is still largely unknown. In this study, we have used a proteomics‐driven approach to identify potential biomarkers of MD. To this end, plasma was obtained from whole blood of 16 individuals previously diagnosed as suffering from MD and compared to plasma from healthy donors. A depletion of the highly abundant proteins (i.e., albumin, IgG, transferrin, etc.) was performed in order to enhance the chance of detection of the less represented ones, therefore reducing the noise‐background. Two‐dimensional gel electrophoresis, followed by in‐gel tryptic digestion of the selected spots and LC‐MS/MS analysis, allowed us to identify a set of proteins whose expression appears to be differentially modulated in patients versus controls. In particular: complement factor H and B, fibrinogen alpha and gamma chains, beta‐actin and pigment epithelium derived factor are over expressed; on the other hand, the levels of beta‐2 glycoprotein‐1, vitamin D binding protein and apolipoprotein‐1 are significantly decreased in the plasma of MD‐affected individuals. Even though preliminary and not necessarily linked directly to the molecular pathogenesis of the disease, our original findings suggest that a molecular signature, represented by the plasma protein profile previously described, might represent a potentially powerful, innovative and not invasive tool for early diagnosis and clinical management of MD patients. Crude homogenates or acetone powder preparations from oat leaves efficiently catalyse the glucosylation of a steroidal sapogenin, nuatigenin [22,25-epoxy-(20S)(22S)(25S)-furost-5-en-3β,26-diol], using UDP-glucose as the sugar donor. The reaction product was identified as nuatigenin 3β-D-monoglucoside. In contrast to the glucosylation of phytosterols, which is also catalysed by enzyme preparations from oat leaves, the formation of nuatigenin glucoside is not stimulated by Triton X-100. This result suggests that glucosyltransferases with different specifirity patterns are involved in sterol and nuatigenin glucosylation in oat leaves. Enzymatic acylation of nuatigenin glucoside to its monoacyl derivative with the use of an endogenous acyl source was also observed with a crude homogenate or a crude membranous fraction as the enzyme preparation.  相似文献

The aim of this study was to record and compare the occurrence of adults and immature stages of honeydew moth, Cryptoblabes gnidiella (Millière), related with grape phenology in two Pinot Noir (Vitis vinifera) vineyards in Bento Gon?alves (29 degrees 10'S 51 degrees 32'O), RS, during 2004-2005. Fortnightly, immature stages were monitored in the clusters and grapevine branches, while the adults, in delta traps baited with synthetic sex pheromone. There were significant differences in the number of individuals in different phenologic stages of vineyards. The highest catches of honeydew moth were registered in the period of dry clusters. The permanence of dry cluster after harvest, could serve as refuge for C. gnidiella immatures between seasons.  相似文献

Summary

Light and electron microscopy were used to analyze the morphology and ultrastructure of the spermatozoon of Fissurella nigra and Fissurella picta, two sympatric species of keyhole limpets from the southern Chilean coast. The spermatozoa of both species are of the aquasperm type, typical of species with external fertilization. Each species had its own distinctive spermatic morphology, particularly in relation to size. Proceeding from anterior to posterior, the spermatozoa of both species each consists of an elongated head with a conical acrosome having a deep subacrosomal space and truncated conical nucleus, followed by a midpiece containing five mitochondria associated in a compact ring around the proximal and distal centrioles, and, at the end, a flagellum. The spermatozoon head of F. nigra is almost twice as long as that of F. picta. This difference constitutes the first morphological evidence that the size of the spermatozoon could represent a candidate for the maintenance of reproductive isolation between these two sympatric species.  相似文献

The coffee leaf miner Leucoptera coffeella (Guérin-Mèneville), one of the major pests of coffee crops in Brazil, is mainly controlled with insecticides. The objective of this study was to evaluate the residues and the translocation of the insecticide thiamethoxam in coffee leaves, as well as to study its effect on the coffee leaf miner control, comparing it with aldicarb, used as standard. One experiment was set up in the county of Gar?a, SP from December/2001 to August/2002. The treatments used were: aldicarb 150 G at the rates of 2.25 and 4.50 g a.i./pit, thiamethoxam 10 GR, at the rates of 0.15 and 0.30 g a.i./pit and check. Twig samples were collected prior to and 30 , 60, 90, 120, 150, 180, 210 and 240 days after the application, at three coffee plant heights (lower, middle and upper third), and the percentage of mined leaves was evaluated. The determination of aldicarb residues, including their sulphoxide and sulfone metabolites and of thiamethoxam were performed by gas chromatography with a nitrogen-phosphorus and mass spectrometer detectors, respectively. The results indicated a uniform translocation of both insecticides in all three thirds of the coffee plants when applied to the soil. A higher persistence of thiamethoxam was verified with its residues being found for as far long as eight months following the application, while aldicarb residues, including the sulphoxide and sulfone metabolites, were found only until four to six months after the application. Control of the coffee leaf miner was observed with both insecticides.  相似文献

The production of both ABA and H?O? is induced by drought and can act as signals under stress conditions. We investigated the relationships between ABA, H?O? and catalase (CAT) in rice leaves when rice seedlings were treated with polyethylene glycol as water stress treatment. As a key gene in ABA biosynthesis, OsNCED3 was significantly induced in rice by water stress treatment and such induction preceded the rapid increase in ABA. Water stress inhibited the expression of CATA and CATC but substantially enhanced the expression of CATB. Exogenously applied ABA promoted the expression of CATB also and inhibited the expression of CATC in a concentration-dependent manner. When ABA production was inhibited by using ABA biosynthesis inhibitors nordihydroguaiaretic acid and tungstate, expression of CATB was also subdued while CATC was enhanced under the water stress. Accumulation of H?O? was also reduced when endogenous ABA production was inhibited and showed a correlation with the total activity of catalases. Our results suggest that water stress-induced ABA prevents the excessive accumulation of H?O?, through the induction of the expression of CATB gene during water stress.  相似文献

An interspecies deer hybrid resource population developed from a cross of Père David's and red deer was used to detect QTL that account for species differences. A genome scan, coupled with composite interval mapping, was conducted to search for QTL controlling body measurements at pre-pubescent age (6 months of age) and puberty (15 months of age) in this interspecies hybrid. Five linkage groups that harbour QTL affecting morphology were identified. A joint-traits analysis was used to search for putative pleiotropic QTL on four of these linkage groups, and three were significantly associated with pleiotropic QTL for nose width and foot length (metacarpal and phalanges), which collectively accounted for 29-58% of the phenotypic difference between the two deer species. This study suggests that a few loci with large pleiotropic effects may be responsible for species-specific differences in growth and structure-related traits.  相似文献

During the 20th century, the first dinosaur tracks of the Causses Basin were identified at Saint-Laurent-de-Trèves, in the Parc National des Cévennes (southern France). A recent excavation reveals a new theropod tracksite in the Hettangian deposits from Le Mazel, 2 km from the historical tracksite at Saint-Laurent-de-Trèves. The tracks are here described combining a biometric approach and 3D imaging photogrammetry. The main track-bearing surface bears 64 in situ tridactyl footprints preserved as concave epireliefs. Two morphotypes were identified, a “Grallatorid” morphotype and a “Kayentapus” morphotype. Footprints belonging to the first morphotype are closely similar to Grallator lescurei, Grallator minusculus, and Grallator sauclierensis. This study shows the difficulty to distinguish quite similar tridactyl tracks from an ichnotaxonomic point of view and highlights the importance of detailed biometric comparisons. Tracks are preserved in a brown to yellowish dolomudstone showing abundant cryptalgal laminites and mud cracks. These deposits were accrued in shallow environments such as intertidal and supratidal zones of a tidal flat.  相似文献
