Summary Analytical microscopy was used to study the distribution and chemical composition of thorium deposits in bone marrow and liver after injection of thorium dioxide and thorium nitrate. Thorotrast (thorium dioxide) was identified as being localized in bone marrow macrophages of a patient who had undergone cerebral arteriography forty two years ago. Large thorotrast deposits were also present in liver cells. We show that non-colloidal thorium (thorium nitrate) injected in rats concentrates in a non soluble form in bone marrow macrophages, heptocytes and Kupffer cells. These deposits of thorium associated with phosphorus can be explained by the formation of thorium phosphate in lysosomes and we demonstrate that they remain in tissue for a long time. Microanalysis was performed with ion microscopy, and electron probe microanalysis by X ray spectrometry, which can identify and localize thorium and associated elements at cellular or intracellular level.  相似文献

A case of hepatoma induced by thorium dioxide (Thorotrast) is reported. The literature concerning neoplasia associated with this agent is reviewed, with emphasis on the long latent period before the development of these tumours and the equally long latent period which preceded the recognition of their iatrogenic nature.In particular, this report is intended to illustrate the diagnosis of such tumours on radiological and histological findings, provided that one is aware of their existence and familiar with the distribution and appearance of thorium dioxide deposits in the tissues. Such a diagnosis can be made without a history of thorium dioxide administration, which often may not be available.  相似文献

The attachment to the surface of the ameba (Chaos chaos L. (Pelomyxa carolinensis, Wilson)) of two proteins, ribonuclease and ferritin, and two colloidal suspensions, thorium dioxide and gold, was studied in the electron microscope. The initial step in the pinocytosis of ferritin and thorium dioxide particles by amebas is shown to be the attachment of these substances to the "hairlike" extensions of the plasmalemma. Ribonuclease caused alterations in the structure of the plasmalemma, but on account of its relative lack of density, it could not be definitely localized. Colloidal gold did not appear to be active with respect to pinocytosis in amebas. Since molecules in solution and particles in suspension are taken up by the same mechanism, the first step of which is their attachment to the cell surface, it is suggested that a single mechanism underlies phagocytosis, pinocytosis, ropheocytosis, cytopempsis, and potocytosis.  相似文献

Endothelium of large arteries from several species was studied in thin sections with the electron microscope. Before sacrifice, some animals received an intravenous injection of colloidal thorium dioxide which was visualized in the sections. Surface replicas were prepared by carbon evaporation on either frozen-dried endothelium or on endothelium dried by sublimation of naphthalene with which the tissue had been impregnated. Cell boundaries, stained with silver, were observed in sections and also from the surface by stripping off the inner part of the endothelium. In addition to the usual cytoplasmic organelles, the endothelial cells showed certain characteristic features, namely, large invaginated pockets communicating with the arterial lumen, numerous much smaller vesicular structures immediately under the plasma membrane and apparently also communicating with the lumen, and inclusions, into which injected thorium particles were incorporated. Intercellular boundaries appeared as regular double membranes in thin sections, and they were outlined by a double row of silver granules after silver staining. No evidence was obtained of permeation of intracellular spaces by colloidal thorium.  相似文献

Summary Smooth muscle cells were enzymatically isolated from the tunica media of the aorta of 5-day-old rats. Following in vitro exposure to colloidal thorium dioxide particles the cells displayed numerous lysosomes filled with these particles. Intramuscular injection of isolated cells into the tongues of young rats of the same strain, resulted in the formation by both freshly isolated and thorium dioxide-labeled cells of a characteristic tissue similar to that found in the intact aortic wall. Thus, the transplants consisted of typical smooth muscle cells surrounded by an extracellular matrix consisting of a microfibrillar network, patches and fibers of elastin, collagen fibrils and small polygonal granules believed to represent proteoglycans. Colloidal mercuric sulfide or thorium dioxide injected intraperitoneally passes into the cytoplasm of the mesothelium of the mesentery and of the diaphragm as early as 15 to 30 minutes after the injection. 2. Between 15 minutes and 12½ hours the number of particles within the mesothelial cells increases as the time between injection and termination of the experiment is lengthened. 3. The particulate matter is usually localized in the cytoplasm within clear vacuoles or bodies having a relatively dense matrix. 4. A greater quantity of the absorbed material is commonly observed within the cytoplasm of the diaphragmatic than of the mesenteric mesothelium.  相似文献

Summary Chondrocytes isolated from fetal, guinea-pig epiphyses were grown in monolayer culture, exposed to thorium dioxide particles, and studied ultrastructurally after varying intervals. The exogenous marker was ingested by endocytosis and subsequently accumulated in lysosomes. After intramuscular injection into young guinea pigs, the thorium dioxidelabeled chondrocytes formed a typical hyaline cartilage. This consisted mainly of rounded or polygonal cells with large, eccentrically located nuclei. The cytoplasm showed an extensive granular endoplasmic reticulum and a well-developed Golgi complex, suggesting active synthesis and secretion of matrix components. Among the other cytoplasmic organelles, lysosomes containing variable amounts of marker particles were observed. After 2–3 weeks the transplants showed signs of cellular degeneration and disintegration. During these processes, lysosomes remained structurally intact and, furthermore, retained the incorporated marker. Thus, thorium dioxide-labeled bodies were found in former chondrocyte lacunae and in the intercellular substance proper. In the latter location labeled bodies could be observed in close proximity to early mineral deposits. The ability of the mouse mast cell to phagocytize colloidal thorium dioxide, Thorotrast, was investigated employing the mouse connective tissue air pouch. The connective tissue mast cell of mouse was found to have limited capability to ingest the particulate Thorotrast in comparison to the rat peritoneal mast cell which ingested this material readily. Fibroblasts and macrophages in the connective tissue removed injected Thorotrast very rapidly in contrast to mast cells that demonstrated limited phagocytic capabilities. The tissue mast cell of the mouse, therefore, should not be considered a part of the reticuloendothelial system.  相似文献

Several approaches have been used to study the determinants of phagosome-lysosome fusion in intact mouse macrophages. Lysosomes were labeled with the fluorescent vital dye acridine orange and the rate and extent of their fusion with yeast-containing phagosomes was monitored by fluorescence microscopy. Fusion was also assayed by electron microscopy, using horseradish peroxidase or thorium dioxide as a marker for secondary lysosomes. Good agreemen samples with an enzymatic marker, and thorium dioxide-labeled samples evaluated by stereology. The rate of usion as assayed by fluorescence was not affected by the number of particles ingested, serum concentration, or prior uptake of digestible or nondigestible substances. With this assay it was possible to observe the rate of fusion separate from and uninfluenced by the phagocytic rate. Both the rate and extent of fusion were dramatically increased after several days in culture and similar changes were found by use of the EM assays. Fusion was strongly affected by incubation temperature, having a Q10 of 2.5 No detectable fusion occurred below 15 degrees C, and this inhibition was rapidly reversed when cells were returned to 37 degrees C.  相似文献

Physiological studies have demonstrated that ions, as well as large molecules such as hemoglobin or fluorescein, can diffuse across and within the cornea. Most of the substrates for corneal metabolism are obtained from aqueous humor filling the anterior chamber. In order to receive its nutrients and in order to maintain its normal conditions of hydration, the avascular cornea must transport relatively large amounts of solute and solvent across the cellular layers which cover this structure. It has been suggested in the past that there may be a morphological basis for the transport of large amounts of solvents and solutes by cells by the mechanism of pinocytosis. The use of electron-opaque markers to study fluid movements at the electron microscope magnification level was described by Wissig (29). The present study describes the fine structure of the normal rabbit cornea and the pathways of transport of colloidal particles by the cornea in vivo. Rabbit corneas were exposed in vivo to suspensions of saccharated iron oxide, thorium dioxide, or ferritin by injection of the material into the anterior chamber. In other experiments thorium dioxide or saccharated iron oxide was injected into the corneal stroma, producing a small bleb. Particles presented at the aqueous humor surface of the rabbit corneal endothelium are first attached to the cell surface and then pinocytosed. It appears that the particles are carried around the terminal bar by an intracellular pathway involving the pinocytosis of the particles and their subsequent transport in vesicles to the lateral cell margin basal to the terminal bar. Particles introduced at the basal surface of the endothelium (via blebs in the corneal stroma) are apparently carried through the endothelial cells in membrane-bounded vesicles without appearing in the intercellular space. There appears to be free diffusion of these particles through Descemet's membrane and the corneal stroma. The stromal cells take up large quantities of the particles when blebs are injected into the stroma.  相似文献

Forested ecosystems diversified more than 350 Ma to become major engines of continental silicate weathering, regulating the Earth''s atmospheric carbon dioxide concentration by driving calcium export into ocean carbonates. Our field experiments with mature trees demonstrate intensification of this weathering engine as tree lineages diversified in concert with their symbiotic mycorrhizal fungi. Preferential hyphal colonization of the calcium silicate-bearing rock, basalt, progressively increased with advancement from arbuscular mycorrhizal (AM) to later, independently evolved ectomycorrhizal (EM) fungi, and from gymnosperm to angiosperm hosts with both fungal groups. This led to ‘trenching’ of silicate mineral surfaces by AM and EM fungi, with EM gymnosperms and angiosperms releasing calcium from basalt at twice the rate of AM gymnosperms. Our findings indicate mycorrhiza-driven weathering may have originated hundreds of millions of years earlier than previously recognized and subsequently intensified with the evolution of trees and mycorrhizas to affect the Earth''s long-term CO2 and climate history.  相似文献

The soil bacterium Bradyrhizobium (Chamaecytisus) strain BGA-1 produces an extracellular polymeric substance (EPS) that, in the presence of Fe3+, Al3+ or Th4+ solutions, forms a gel-like precipitate composed of polysaccharide, protein, lipopolysaccharide and the metal. Precipitation of the main component of the EPS, the extracellular polysaccharide, and thorium was studied. The precipitate was stable, but redissolved at pH values below 3.0 or in the presence of 10 mM EDTA. In the precipitate, the ratio thorium/basic repeating unit of the polysaccharide ranged from 0.4 to 0.8 mol/mol. Soluble polysaccharide–thorium complexes were not found, and larger polysaccharide molecules were precipitated in preference to smaller ones. Kinetic studies showed a non-linear dependence of the precipitate on the concentrations of both thorium and polysaccharide. The behaviors of the purified polysaccharide and of whole EPS with the thorium-containing precipitate were compared. The results suggested that EPS components other than polysaccharide are able to modify the precipitating ability of the polysaccharide. Thus, whole EPS is a better substrate than the purified polysaccharide for the removal of thorium from its solutions.  相似文献

Inactive cells of Rhizopus arrhizus have been documented to exhibit a high thorium biosorptive uptake (170 mg/g) from aqueous solutions. The mechanism of thorium sequestering by this biomass type was investigated following the same method as for the uranium biosorption mechanism. The thorium sequestering mechanism appeared somewhat different from that of uranium. Experimental evidence is presented which indicates that, at optimum biosorption pH (4), thorium coordinates with the nitrogen of the chitin cell wall network and, in addition, more thorium is absorbed by the external section of the fungal cell wall. At pH 2 the overall thorium uptake is reduced. The kinetic study of thorium biosorption revealed a very rapid rate of uptake. Unlike uranium at optimum solution pH, Fe(2+) and Zn(2+) did not interfere significantly with the thorium biosorptive uptake capacity of R. arrhizus.  相似文献

区域旅游交通碳排放测度是分解旅游业减碳任务的一个难题。在剖析替代式自下而上法机理的基础上,以长三角为例,依托归纳法和变异系数法,尝试从人均GDP、人均消费水平和人均运输线路长度三方面测算出游距离,采取以实地调研数据为主、辅之以Mus TT模型法拟定各旅游交通方式的距离比例,立足区情确定碳排放系数,并以区域输入和输出的双向旅游流的人次比值法则对替代结果进行还原调整。研究表明:(1)2011年,长三角旅游交通碳排放总量为8.32 Mt,其中江苏省、浙江省和上海市分别为3.23、2.98 Mt和2.11 Mt;(2)飞机和自驾车共占排放量的71.64%,较明显低于世界平均比例,二者是降低旅游交通碳排放的关键;旅游公共交通的碳排放比例具有远高于发达国家甚至全球平均水平的"中国式"烙印;由高碳排放系数的旅游交通方式向低碳排放系数甚至零碳排放系数的转变,是旅游交通减排的基本方向。  相似文献

Mast cells from the peritoneal cavity of the rat were obtained at various times following in situ injection of a colloidal thorium dioxide preparation (Thorotrast). They were prepared for electron microscopy by aldehyde fixation, osmium tetroxide postfixation, and embedding in Epon. Thorotrast was rapidly taken up by mast cells through enhanced or newly elicited surface specializations. It was confined at first to large vesicles which moved to the Golgi area. Subsequently, in a matter of a few hours only, it became associated with progressively more mature granules, including "fully" mature ones. In addition to demonstrating a further phagocytic or pinocytotic activity of mast cells, the findings suggest that mast cell granules share a common membranous investment, and that substances from the tissue environment may theoretically percolate over and interact with the granules. Mast cell function could thus be served primarily by absorptive rather than secretory processes.  相似文献

OBJECTIVES--To seek associations between meteorological factors, concentrations of air pollutants or pollen, and an asthma epidemic which occurred in London on 24 and 25 June 1994 after a thunderstorm. DESIGN--Retrospective study of patients'' accident and emergency department records, with bivariate and multivariate analysis of environmental factors and data collection for the two months surrounding the epidemic. SETTING--The accident and emergency department of St Mary''s Hospital in west central London. SUBJECTS--148 patients presenting with asthma between 1 June and 31 July 1994, of whom 40 presented in the 24 hours after the storm. RESULTS--The asthma epidemic was significantly associated with a drop in air temperature six hours previously and a high grass pollen concentration nine hours previously. Non-epidemic asthma was significantly associated with lightning strikes, increase in humidity or sulphur dioxide concentration, a drop in temperature or high rainfall the previous day, and a decrease in maximum air pressure or changes in grass pollen counts over the previous two days. CONCLUSIONS--New episodes of asthma during the epidemic on 24 and 25 June 1994 were associated with a fall in air temperature and a rise in grass pollen concentration. Non-epidemic asthma was significantly associated with a greater number of environmental changes. This may indicate that the patients with thunderstorm associated asthma were a separate population, sensitive to different environmental stimuli.  相似文献

Selected samples of waste microbial biomass originating from various industrial fermentation processes and biological treatment plants have been screened for biosorbent properties in conjunction with uranium and thorium in aqueous solutions. Biosorption isotherms have been used for the evaluation of biosorptive uptake capacity of the biomass which was also compared to an activated carbon and the ion exchange resin currently used in uranium production processes. Determined uranium and thorium biosorption isotherms were independent of the initial U or Th solution concentration. Solution pH affected the exhibited uptake. In general, lower biosorptive uptake was exhibited at pH 2 than at pH 4. No discernible difference in uptake was observed between pH 4 and pH 5 where the optimum pH for biosorption lies. The biomass of Rhizopus arrhizus at pH 4 exhibited the highest uranium and thorium biosorptive uptake capacity (g) in excess of 180 mg/g. At an equilibrium uranium concentration of 30 mg/liter, R. arrhizus removed approximately 2.5 and 3.3 times more uranium than the ion exchange resin and activated carbon, respectively. Under the same conditions, R. arrhizus removed 20 times more thorium than the ion exchange resin and 2.3 times more than the activated carbon. R. arrhizus also exhibited higher uptake and a generally more favorable isotherm for both uranium and thorium than all other biomass types examined.  相似文献
