Mycelium ofBeauveria bassiana can be grown in liquid culture, filtered, and the mycelium dried. After rehydration the mycelium sporulates. Two carbohydrate sources (sucrose and maltose), and one nitrogen/vitamin source (yeast extract) were tested for mycelium growth and subsequent conidial production. Maximum mycelium growth (12.31 mg/ml), in liquid culture, was in the sucrose (3.5%)/yeast extract (3.5%) medium, but mycelium from a maltose (2%)/yeast extract (0.75%) medium produced the maximum of 4.62×106 conidia/mg dry mycelium after incubation in moist Petri dishes. Using the data on mycelium yield (in liquid culture) and conidial production (by dry mycelium) it is calculated that the sucrose (3.5%)/yeast extract (3.5%) and the maltose (2%)/yeast extract (0.75%) media produce most conidia per media volume (an equivalent of 3.52–3.72×107 conidia/ml).   相似文献

Growth and differentiation of mycelial strands in Rigidoporus lignosus have been shown to depend on suitable combinations of the pH of the media and the nature of the nitrogen and carbon sources. Amino acids as sole nitrogen sources gave rise to vegetative mycelium. At pH 4.5, growth and mycelial strand differentiation required asparagine, as the fungus failed to grow in the absence of this amino acid. However, at pH 6, differentiation of strands occurred appreciably in asparagine-deficient media, suggesting a close balance between pH and amino acid requirements. Ammonium was required for strand differentiation, while nitrate, as a sole nitrogen source, maintained the fungus undifferentiated. Of the carbohydrates tested, only glucose, fructose and mannose supported strand differentiation. Starch was found to be particularly effective in promoting growth of vegetative mycelium. Strand differentiation required more specific conditions than growth of the vegetative mycelium.  相似文献

A chitinolytic actinomycete complex in chernozem soil has a specific taxonomic composition, which differs from that of the actinomycete complex typically isolated on standard nutrient media containing sugars and organic acids as carbon sources. The actinomycete complex that was isolated by using nutrient media with chitin as the source of carbon and nitrogen was dominated by representatives of the genus Streptosporangium, and the actinomycete complex that was isolated by using nutrient media with sugars and organic acids as the carbon sources was dominated by representatives of the genus Streptomyces. The confirmation of the ability of actinomycetes to utilize chitin as a sole source of carbon and nitrogen came from the augmented length and biomass of the mycelium, the increased number and biomass of the actinomycete spores, the production of carbon dioxide, and the accumulation of NH4 + ions in the culture liquid of the actinomycetes grown in the nutrient media with chitin.  相似文献

It was shown that change of medium growth composition of photopathogenic fungus Rhizoctonia solani Kühn, especially accessible sources of nutrition, leads to change of both quantity of produced proteinases and their action specificity. The mineral source of nitrogen suppressed the fungus proteinase secretion on cultivatiin medium containing potato thermostable proteins but an organic source of nitrogen accelerated mycelium growth and increased proteinase secretion. On the basis of an analysis of a fungus extracellular proteinase substrate-specificity, it is established that the presence of thermostable proteins of a potato in the cultural liquid induces the secretion of trypsin-like proteinases mainly, and the addition of yeast extract to this growth medium induces the secretion of subtilisin-like ones, thus suppressing the trypsin-like enzymes production. This fact can indicate that mycelium of fungus R. solani loses pathogenic properties and becomes saprophytes when the growth medium was enriched by an organic source of nitrogen.  相似文献

Microalgae cultures are receiving attention because of increasing biotechnological and biomedical production of active biomolecules. We evaluated various fertilizer-based culture media to scale up production of the marine microalga Phaeodactylum tricornutum for production of exocellular polysaccharides (EPS), soluble proteins, and cellular superoxide dismutase (SOD). The standard source of sodium nitrate was the same as that used in the synthetic f/2 culture medium and ammonium nitrate, urea, ammonium sulfate, and calcium nitrate as alternative sources of nitrogen. The maximum production of EPS was achieved in microalgae cells grown in the culture media containing 63 and 23% nitrogen from ammonium sulfate, and also in microalgae cells grown in the culture media containing 3% nitrogen from ammonium nitrate. The maximum production of cellular SOD was achieved in microalgae cells grown in the culture media containing 35 and 26% nitrogen from ammonium sulfate, and in the culture media containing 17% nitrogen from urea. The results suggest that it is possible to use a source of nitrogen, other than sodium nitrate, to scale up growth of P. tricornutum for production of EPS and SOD at reduced costs.  相似文献

When an aconidial strain (STL6A) of Neurospora crassa is grown on carbon sources such as glucose, maltose, sucrose, etc., trehalase activity per unit weight of mycelium is very low. By contrast, media containing arabinose, glutamic acid, glycine, etc., which support growth only poorly, produce mycelium with very high trehalase activity. Retarding growth limiting the supply of a necessary nutrient, altering the pH and temperature, or adding toxic substances, however, does not derepress trehalase activity. Repression and derepression of trehalase was found to be reversible through the transfer of cultures to appropriate media. It is likely that the increase in trehalase activity results from de novo synthesis because labeled enzyme can be isolated from acrylamide gels after isolation from medium containing C14-labeled leucine and after purification by other means. These experiments are interpreted in terms of catabolite repression which may be correlated with events during growth and conidiation.  相似文献

In this paper, we described the development of a potassium-chloride-based-salt formulation containing low sodium concentrations (5.0 to 11 mM) to support the growth of Salinispora tropica strain NPS21184 and its production of salinosporamide A (NPI-0052). The sodium present in the media was essentially derived from the complex nitrogen sources Hy Soy, yeast extract, and peptone used in the media. We demonstrated that good growth rate and yield of S. tropica strain NPS21184 were detected in both agar and liquid media containing the potassium-chloride-based-salt formulation with sodium concentration as low as 5.0 mM, significantly less than the critical seawater-growth requirement concentration of 50 mM sodium for a marine microorganism. We also observed good production of NPI-0052 (176 to 243 mg/l) by S. tropica strain NPS21184 grown in production media containing the potassium chloride-based-salt formulation. The production of deschloro analog, salinosporamide B (NPI-0047), was significantly lower in the low-sodium-salt-formulation medium than in the high-sodium-salt-formulation media. We demonstrated that while S. tropica strain NPS21184 is a novel marine actinomycete that requires high salt content for growth, it does not require sodium-chloride-based seawater-type media for growth and production of NPI-0052.  相似文献

Six non-amino acid nitrogen compounds were examined as nitrogen source for growth of Streptomyces hygroscopicus and biosynthesis of rapamycin. Of the nitrogen sources studied, ammonium sulfate was the best with respect to formation of rapamycin, and supported cell growth comparable to the organic nitrogen sources used in the control chemically defined medium, ie, aspartate, arginine plus histidine. In the new chemically defined medium, which is buffered with 200 mM 2-(N-morpholino)ethanesulfonic acid to prevent decline of pH during fermentation, an ammonium sulfate concentration of 40 mM was optimal for biosynthesis of rapamycin. Rapamycin production increased by more than 30% on both volumetric and specific bases as compared to the previous medium containing the three amino acids as nitrogen source. The effect of inorganic nitrogen source on population growth of Euglena mutabilis, an acidophillic benthic protozoa colonizing on the sediment of acid mine drainage, was investigated. Sodium nitrate, ammonium chloride, and ammonium sulfate were tested as nitrogen sources. The population density of E. mutabilis at equilibrium density cultivated in ammonium chloride‐ and ammonium sulfate‐containing media was 9–11 times higher than that in sodium nitrate‐containing medium at the optimal salt medium concentration. The population growth of E. mutabilis in ammonium sulfate‐containing medium was rapid and reached half of the equilibrium density after ca. 228 h, which was ca. 77 h earlier than that in ammonium chloride‐containing medium. Culture medium with ammonium sulfate as the nitrogen source achieved the highest maximum population density and the fastest growth rate among the three nitrogen salts used as nitrogen sources.  相似文献

Fish powders and fish protein hydrolysates (FPH) from sardinella (Sardinella aurita) were prepared and tested as growth media for alkaline protease production by Pseudomonas aeruginosa MN7. Cultivated in fish substrate as carbon source, the strain exhibited a slightly greater protease production (about 7800 U ml–1) than that obtained with commercial peptones (about 7222 U ml–1). Furthermore, P. aeruginosa MN7 produced the same amount of protease when cultivated in medium containing only fish substrate or that containing all ingredients, indicating that the strain can obtain its carbon and nitrogen requirements directly from whole fish proteins. Moreover, it was found that extensive hydrolysis of fish proteins did not increase protease formation. Protease production in media containing only FPH prepared by Alcalase was about 70% of those obtained with MN7 protease digest of fish protein or with meat-fish powder. These results indicate that sardinella substrates are an excellent carbon and nitrogen source for the growth of P. aeruginosa MN7 and the production of protease.  相似文献

Summary The production of protease and mycelium byNeurospora sitophila cultured on solid and liquid potato dextrose media was studied. Maximal activity of protease extracted from 4-day-old cultures occurred at pH 6.5 when an unfractionated peanut (groundnut) protein substrate was used. The greatest protease activity and mycelium production occurred during the first day of the 4-day test period. Potato dextrose media containing more than 0.2 M NaCl resulted in decreased protease and mycelium production, while tapioca starch was without affect at concentrations up to 1.4%. Addition of up to 0.3 M sucrose to growth media greatly stimulated protease production and mycelial growth. Maximal proteolytic activity was observed in extracts from mycelium cultured in potato dextrose media adjusted from pH 6.0 to 7.5. Activity was greater when soluble peanut protein was used as a substrate, compared to unfractionated or globulin protein substrates.  相似文献

Growth requirements of two predaceous hyphomycetes in the absence of eelworm prey were determined in specified culture media, in an investigation of the interrelationship of eelworm and fungi. Both fungi were able to utilize a wide range of energy sources and gave the highest yields of mycelium when the carbon sources supplied had the same configuration as glucose around carbon atoms, 3, 4, 5 and 6, or were oligosaccharides yielding glucose on hydrolysis. The species differed in their ability to utilize nitrogen sources, A. robusta producing very little growth when nitrate was the sole source of nitrogen, and comparatively little on aspartate nitrogen. Both responded to thiamin, biotin and p-aminobenzoic acid, but neither species showed absolute dependence on an external source of vitamin.  相似文献

The influence of carbohydrate utilisation on the growth of three strains of Vittad. mycelium (1BO, 17BO and 10RA) in culture was assessed using culture media containing glucose (control), mannose or mannitol. Mannose was the best substrate for growth of the strains and this was particularly evident for strain 17BO. Mannitol instead was metabolized only by 10RA and 1BO. In order to explain the different growth trends, analyses of enzyme levels, kinetic parameters, protein patterns and the morphology of the three strains were carried out. Our results show that these strains of mycelium were affected by the substrates used in the media. The aim of the present work was to optimise the in vitro production of T. borchii mycelium for use in experiments which require the fungus in precise and reproducible conditions, such as mycorrhizal synthesis or protein and nucleic acid extractions.  相似文献

Summary Pichia stipitis CBS 6054 ferments D-Xylose to ethanol in a medium containing corn steep liquor as the only source of nitrogen, amino acids, vitamins and other nutrients. The ethanol yield and fermentation rate compare favorably to those obtained with media containing more expensive sources of nitrogen, vitamins and amino acids. Corn steep liquor is a good source of nutrients that can support growth and fermentation activity of this xylose fermenting yeast.  相似文献

Investigations simulating solid-state fermentation, SSF, were carried out to study the influence of a varying N supply on the growth of a mycelium. Experiments with two fungi and two N sources demonstrated the high adaptability of the mycelium to a deficit of nitrogen over a wide range of N supplies. The mycelium of the fungi Gliogladium sp. and Aspergillus sp. can grow up to the very low crude protein content of 11–15%. But protein-poor biomass could not make up the deficit of protein even under optimal cultivation conditions, that means the state of the mycelium remains irreversible. This showed the limit of the adaptability of filamentous fungi, which has to be taken into account for mycelium growth in SSF.  相似文献

Both growth and sporulation increase linearly according to the initial water content of the solid substrate when Penicillium roquefortii is cultivated on buckwheat seeds. This indicates that water is the limiting factor for fungal development since neither carbon nor nitrogen sources were exhausted during these experiments. This feature validates the concept of available water for fungal growth, defined as initial water content of substrate minus its residual water content when vegetative growth stops. An efficient methodology, based on drawing a regression line of mycelium dry weight production as a function of the initial water content of substrate is presented; it allows estimation of both available water and water content of mycelium. Results show that growth stops when the residual water in the substrate is close to 0.52 g H2O/g initial dry matter, corresponding to a water activity (aw) close to 0.96, and that the initial water content in mycelium is near 76%. The presence of different carbon and nitrogen sources and bivalent metal compounds in the substrate medium influenced the mycelial growth of Macrophomina phaseolina and in vitro susceptibility to fungicides. The inoculum from such substrate media showed differences in pathogenicity on mung bean (Vigna radiata). Sucrose and asparagine significantly increased the mycelial growth as well as pathogenicity of the fungus. Absence of bivalent metal ions, viz., Fe++, Zn++ and Mg++ in the medium produced inoculum which caused maximum seedling mortality and foliage blight. Carbendazim and thiophanate-M as seed treatments were significantly less effective when the inoculum was from a medium containing glucose than when the inoculum was from a medium containing sucrose. Captafol and thiram gave significantly better disease control on mung bean when the inoculum used for soil inoculations was from media containing asparagine and ammonium nitrate compared to the inoculum grown on a medium containing sodium nitrate. Carbendazim, thiophanate-M, PMA, captafol and thiram gave good disease control when the inoculum used was raised on a medium devoid of bivalent metal ions. Carbendazim and thiophanate-M were the best fungicides as foliar treatments and controlled the disease irrespective of carbon, nitrogen and bivalent metal ion status of the substrate medium used for the production of inoculum.  相似文献

The capability of utilizing 20 amino acids and 2 amides as the sole nitrogen source for growth was studied in two green algae (Chlorophyceae). A comparison was made of the growth rate of algae in a mineral nutrient solution containing nitrate as the nitrogen source, with that in the same solution in which nitrogen in the form of nitrate was substituted by an equivalent nitrogen amount in the form of various amino acids. In addition to this, another series of experiments was carried out in whioh both culture media were supplied with glucose. The results show that both algae utilize a series of amino acids in dependence of their structure (mostly 3-carbon amino acids). The growth rate ofChlorella in the presence of these sources is the same as in nitrate, that ofScenedesmus even much higher. In the cultures containing glucose both algal species exhibit a higher growth rate in the media with the nitrate nitrogen source than in those with amino acids (with the exception of glycine inScenedesmus).  相似文献

The cultural characteristics of the fungusCladosporium phlei were assessed in order to develop an improved method for the production of the fungal pigment, phleichrome, which is an intermediate in the production of a photodynamic therapeutic agent. The growth ofC. phlei, as measured by the hyphal growth rate and increase in biomass, varies significantly depending on the culture media utilized (V8 juice-based medium proved optimal for both growth rate and biomass increase). How-ever, even on a V8 juice plate, the growth ofC. phlei occurred slowly and in a limited fashion, in that the colony covered only 75% of the agar surface after more than 4 weeks of cultivation at 20°C. Supplementations of glucose, fructose, galactose, and sucrose increased both hyphal expansion and mass production, whereas supplementations of other carbon sources, including glycerol and sorbitol, exerted no detectable effects. The effect of inorganic nitrogen supplementation was negligible, whereas organic nitrogen evidenced significant effects, with enhanced growth with malt extract and growth inhibition with yeast extract and tryptone. Sporulation was enhanced under conditions of continuous light, and a minimum of 103 spores per mL of liquid media was found to be necessary for the optimal mass increase. A simple extraction procedure was established in order to isolate the deep red pigment which was subsequently identified as phleichrome via NMR analysis. WhenC. phlei was cultured on V8 medium containing 5% glucose and 2% malt extract, the quantity of mycelial mass was estimated as 20.6 g (dry weight) per liter of culture. The expected phleichrome yields from the mycelia and culture filtrates were estimated to be 43 and 2 mg/L, repectively.
