Phototaxis in the flagellateEuglena gracilis was studied by means of a microvideographic analysis, and the light-induced directional movement was determined by computer-based statistical treatments. Lateral white light with an illuminance of 25 lx (0.105 Wm–2) caused the cells to preferentially swim toward the light source (positive phototaxis), while an illuminance of 1,000 lx (4.2 Wm–2) induced negative phototaxis. The lipophilic membranepenetrating cation triphenylmethyl phosphonium ion (TPMP+) specifically inhibited positive phototaxis, while it hardly affected negative phototaxis. The uncoupler sodium azide, on the other hand, impaired negative phototaxis substantially.  相似文献

In this paper we report studies on photosynthetic formation of inorganic pyrophosphate (PPi) in three phototrophic bacteria. Formation of PPi was found in chromatophores from Rhodopseudomonas viridis but not in chromatophores from Rhodopseudomonas blastica and Rhodobacter capsulatus. The maximal rate of PPi synthesis in Rps. viridis was 0.15 mol PPi formed/(min*mol Bacteriochlorophyll) at 23°C. The synthesis of PPi was inhibited by electron transport inhibitors, uncouplers and fluoride, but was insensitive to oligomycin and venturicidin. The steady state rate of PPi synthesis under continuous illumination was about 15% of the steady-state rate of ATP synthesis. The synthesis of PPi after short light flashes was also studied. The yield of PPi after a single 1 ms flash was equivalent to approximately 1 mol PPi/500 mol Bacteriochlorophyll. Aggregation of Dictyostelium amoebae is inhibited by light. White light intensities 102 W · cm-2 cause an inhibition which reaches a saturation at 2 · 103 W · cm-2. The action spectrum, based on photon fluence-response curves, shows a major peak around 405 nm and extends through most of the visible spectrum with a secondary maximum at about 530 nm. The action spectrum of the inhibition of aggregation resembles the action spectrum of accumulations of amoebae in light traps and the action spectrum of photodispersal from light traps; it does not resemble the action spectrum of phototaxis in pseudoplasmodia.  相似文献

Photokinesis of the red alga Porphyridium cruentum was studied with the aid of a population method. Because of the slow spreading velocity (0.35 m/min) the duration of the experiments was 7 days in general. According to the white light illuminance-response curve the zero threshold of photokinesis lies below 10 lx and the optimum around 10,000 lx. With further increasing illuminance the photokinetic effect decreases, reaching zero at about 100,000 lx. The action spectrum indicates that the photokinetically active radiation is absorbed by photosynthetic pigments, namely the biliproteins B-phycoerythrin, R-phycocyanin and allo-phycocyanin, as well as by chlorophyl a, although the photokinetic effect of blue light is relatively low. To analyze early effects of auxin application, an apparatus was developed which continuously and simultaneously registered the curvature of 10 individual maize (Zea mays L.) coleoptiles. Resolution was less than 5 m over a range of ±0.5 mm. The data were evaluated and plotted via paper tape and Hewlett-Packard-computer. Unilateral application of 3×10-5 M indoleacetic acid (IAA) resulted in a transient inhibition of growth on the side of application for ca. 10 min (Phase I), followed by a strong stimulation (Phase II). The phytotoxin fusicoccin (FC) caused an immediate stimulation of elongation. The initial negative reaction of Phase I is auxin-specific. Only active auxins such as IAA and 1-naphtaleneacetic acid produced this initial inhibition; chemical analogs-inhibitory or neutral in long-term growth tests, e.g. phenylacetic acid-did not show any significant effects on Phase I. When the coleoptiles were symmetrically preloaded with different levels of auxin, only a large step-up of subsequent unilateral auxin application resulted in a negative phase I; a small step-up led to an immediate positive reaction. When rotated horizontally in a cuvette in a strong lateral light beam, the flagellateEuglena gracilis effectively corrects its course and shows negative phototaxis, provided the angular velocity does not exceed 200s–1. Faster rotations cannot be corrected effeciently. In two strong light beams of equal illuminance perpendicular to each other, the cells move along the resultant away from the light beams. Decreasing the illuminance of one beam causes increasing numbers of the organisms to orient with respect to the stronger light source. In two perpendicular low illuminance beams (>200lx), the population splits into two components moving towards either light source. The percentage of cells in each component depends on the relative illuminances. The results can be explained by the shading hypothesis combined with a dichroic orientation of the photoreceptor molecules perpendicular to the long axis of the cells. Externally applied electric dc fields have no effect on positive or negative phototaxis; this supports the hypothesis that electrical potential changes are not involved in the sensory transduction chain of photoorientation inEuglena.  相似文献

Chlorella sp. strain 3.83, a symbiotic Chlorella isolated from the heliozoan Acanthocystis turfacea, excreted between 8% and 16% of assimilated 14CO2 as maltose in the light (15000 lx), with a pH optimum around 4.8. This percentage increased when the illuminance was lowered (36% at 1700 lx). Release of [14C]maltose continued in darkness and could be inhibited by the uncoupler carbonyl cyanide p-trifluoro-methoxyphenylhydrazone and by diethylstilbestrol. Net efflux of maltose was observed even at a concentration ratio of extracellular/intracellular maltose of 7.8. Exogenous [14C]maltose (5 mM) was taken up by the cells with a rate <2% of that of simultaneous maltose release, indicating a practically unidirectional transport. Summary Synchronous cultures of the green algaScenedesmus quadricauda (Turp.) Bréb. grown at mean irradiances 25Wm–2, 75Wm–2, and 130Wm–2 PhAR were exposed to different illumination regimes (ratio of light to dark interval varied from 2:22 to 24:0 hours). The populations of daughter cells released under these conditions differed markedly in their progress in the cell cycle. The cells from these populations were stained with DAPI and the shape, localization and number of chloroplast nucleoids were examined. The nucleoids were of spherical shape, divided asynchronously having dumbbell shape during fission. In the chloroplast, nucleoids were located symmetrically about the transverse axis of the cells. The mean number of nucleoids varied from two in the least developed daughter cells to 16 in the daughter cells of the highest developmental stage. Summary The active sites of actin of oneCharaceae species were found to interact with the endoplasmic factor from a different species. Protoplasm was suqueezed out of cells ofChara australis with vacuoles that had been perfused beforehand with a medium containing EGTA and Mg · ATP. Centrifugation of this protoplasmic mixture divided it into the supernatant composed of endoplasmic granules and the precipitate composed of chloroplasts and nuclei. When the endoplasmic granular aggregates were introduced into a tonoplast-freeNitella axilliformis cell treated with NEM to inactivate the endoplasmic factor, they became attached to theNitella gel and streamed longitudinally with the polarity. The elongation of the fourth internode of fully green Chenopodium polyspermum L. is strongly stimulated by far-red light (FR) given at the end of the day. The end-of-day effect is more important when the plants had been cultivated for several days with a main light period of 140 Wm-2 than with a main light period of 85 Wm-2. Selection-induced mutations, sometimes called directed, adaptive, or Cairnsian mutations, are spontaneous mutations that occur as specific responses to environmental challenges, usually during periods of prolonged stress, and that occur more often when they are selectively advantageous than when they are selectively neutral. In this study I show that lesions in uvrA, uvrB, uvrC, or uvrD increase the mutation rate from trpA46 to trpA + by 102– to 104–fold during tryptophan starvation, but those same lesions do not affect random mutation rates in growing cells when tryptophan is present. The increased selection-induced mutation rates remain specific to the gene that is under selection in that no increase in the mutation rate from trpA46 to trpA + is detected during proline starvation.Evidence is presented showing that proline starvation produces a state of cellular stress which results in a burst of mutations from trpA46 to trpA + when proline-starved cells are plated onto medium lacking tryptophan but containing proline.These results are consistent with the hypermutable state model for selection-induced mutagenesis.  相似文献

The green micro-algae Chlamydomonas reinhardtiiand Dunaliella tertiolecta were cultivated undermedium-duration square-wave light/dark cycles with acycle time of 15 s. These cycles were used to simulatethe light regime experienced by micro-algae inexternally-illuminated (sunlight) air-lift loopbioreactors with internal draft tube. Biomass yieldin relation to light energy was determined as gprotein per mol of photons (400–700 nm). Between 600and 1200 mol m-2 s-1 the yield at a10/5 s light/dark cycle was equal to the yield atcontinuous illumination. Consequently, provided thatthe liquid circulation time is 15 s, a considerabledark zone seems to be allowed in the interior ofair-lift loop photobioreactors (33% v/v) without lossof light utilization efficiency. However, at a 5/10 slight/dark cycle, corresponding to a 67% v/v darkzone, biomass yield decreased. Furthermore, bothalgae, C. reinhardtii and D. tertiolecta,responded similarly to these cycles with respect tobiomass yield. This was interesting because they werereported to exhibit a different photoacclimationstrategy. Finally, it was demonstrated that D.tertiolecta was much more efficient at low (average)photon flux densities (57–370 mol m-2s-1) than at high PFDs (> 600 mol m-2s-1) and it was shown that D. tertiolectawas cultivated at a sub-optimal temperature (20 °C).  相似文献

At fluence rates below 45 W· m-2 cells of the flagellate stage of Haematococcus lacustris react only positively phototactically with a rather high degree of orientation (indicated by r values up to 0.66 with the Rayleigh test). The directedness of orientation decreases with decreasing irradiance. The degree of directedness of the phototactic response depends on the intensity of preirradiation: Low light intensity applied after strong light application results in a dark reaction (low r values), low light given after darkness stimulates a rather high degree of directedness of positive phototaxis. Weak blue light (=483 nm; 0.4 W · m-2) stimulates positive phototactic response, whereas comparable red light (=658 nm; 0.5 W · m-2) does not.Cells which were grown in a medium containing 10-4 M Norflurazon (effective in inhibition of carotenoid biosynthesis) although maintaining motility completely lose the ability to react positively phototactically. The possible role of carotenoids in the phototactic orientation is discussed.  相似文献

The Ranunculaceae are known to accumulate a wide range of unusual fatty acids in their seed lipids, and this variability has been advocated as a taxonomic marker. The Anemone species, Anemone leveillei L. and Anemone rivularis Buch.-Ham., have previously been reported to accumulate 5-desaturated fatty acids in their seed tissue [K. Aitzetmüller (1995) Plant Syst Evol 9:229–240]. Two cDNAs, AL1 and AL2, with similarity to plant cytochrome b5-fusion "front-end" desaturases were isolated from developing seeds of A. leveillei and their function identified by expression in Saccharomyces cerevisiae. AL2 was characterised as a sphingolipid long-chain-base 8-desaturase, while AL1 acted as a fatty acid desaturase. However, AL1 did not produce 5-desaturated fatty acids as expected; instead, when expressed in transgenic S. cerevisiae or Arabidopsis thaliana this enzyme was functionally characterised as a 6-desaturase. Northern analysis confirmed the expression of this gene in seed tissue and leaf tissue of A. leveillei, though 6-desaturated fatty acids were found to accumulate only in the leaf tissue. The unexpected characterisation of a 6-desaturase in A. leveillei has implications for the use of fatty acids in chemotaxonomic studies. Summary Synchronous cultures of the green algaScenedesmus quadricauda were grown at different mean irradiances (ranging from 15 Wm–2 to 130Wm–2). At each irradiance, the algae were exposed to illumination regimes which differed in light duration and dark intervals (222 to 240 hours). The cells from these cultures were sampled during their cycles, stained with DAPI and the number of nuclei and chloroplast nucleoids estimated.The nucleoids divided semisynchronously in steps which represented doublings in their number. For each doubling a constant amount of light energy (defined as the product of irradiance and light duration) had to be converted by the cells to become committed to this division. The times to the start of the nucleoid divisions were therefore inversely proportional to the irradiances applied and the final number of nucleoids was proportional to the light duration.Temporal relationships between nuclear and nucleoid divisions were also light dependent. Shortage of light energy caused delay in nucleoid division. The cell division rate was higher than the rate of nucleoid division and consequently, the cells tended to decrease their nucleoid number with decreasing irradiance. With increasing irradiance the start of nucleoid division was gradually shifted toward the beginning of the cell cycle. The growth inhibitory activities of 6 endogenous growth inhibitors isolated from light-grown dwarf peas (Pisum sativum cv. Progress No. 9) were examined in the epicotyl of dark-grown seedlings of the same cultivar in the dark in order to examine the possible contribution of these compounds to the growth inhibition brought about by red light. The activities of these natural inhibitors, including two A-2 and A-2 of as yet undetermined structure, were compared with those of synthetic growth retardants and benzyladenine. Samples were applied directly into the epicotyls via a glass capillary tube. In 24-h tests doses for a 25% inhibition (I25) were: A-2, 4.3 × 10-2: cis-xanthoxin, 1.2 × 10-1 ; A-2, 1.6 × 10-1; trans-xanthoxin, 1.2; R,S-dihydromaleimide, 3.5 × 102 and pisatin, 4.0 × 102 nmol plant-1 . In 72-h tests, I25's were: benzyladenine, 1.5; AMO-1618 (ammonium-(5-hydroxycarvacryl)-trimethylchloride piperidine carboxylate), 2.4; R,S-dihydromaleimide, 4.0 × 102 and CCC (chlorocholine chloride), 1.1 × 103 nmol plant-1. -D-Glucosyl-R-dihydromaleimide had no activity at all. Benzyladenine caused the thickening as well as elongation inhibition of the epicotyls of intact plants. TheN-linked carbohydrate chains of the-subunit of highly purified urinary human chorionic gonadotropin have been re-investigated. The oligosaccharides were released enzymatically by peptide-N 4-(N-acetyl--glucosaminyl)asparagine amidase-F, and fractionated by a combination of FPLC and HPLC. As a result of the application of improved fractionation methods, apart from the earlier reported carbohydrate chains, also small amounts of trisialo tri- and tri-antennary oligosaccharides were found. UV-B irradiation has a detrimental effect on the survival of populations of the filamentous cyanobacterium, Phormidium uncinatum, at levels slightly higher than those currently measured at the surface of the earth. The organisms are not damaged or killed by UV-B radiation at 300 nm of 200 Wm-2 for up to 20 h; but slightly increased levels of UV-B irradiation (2 h of 200 Wm-2 at 300 nm) drastically impair motility, phototactic orientation and photophobic responses. These photosynthetic organisms require a narrow light intensity range for growth so that any decrease in their ability to actively search for and move into areas of favorable light conditions is bound to affect the survival of a population. The fluorescence yield of both phycobilins and chlorophyll is not altered even after 20 h of UV-B irradiation (200 Wm-2 at 270 nm) indicating that UV-B at that dose does not affect the photosynthetic apparatus. The organisms are killed either by too bright intensities which bleach the photosynthetic pigments or by the lack of energy when they are unable to avoid moving into dark areas.  相似文献

Phototaxis of the unicellular red alga Porphyridium cruentum was studied by staining the slime tracks of individual cells as well as with the aid of a population method. Because of the increased straightness of the movement the mean linear velocity of a unilaterally illuminated population exceeds considerably that of an only photokinetically stimulated one. In white light the phototactic reaction is saturated already at 100 lx. The zero threshold lies at about 1 lx. Spectral sensitivity curves of phototaxis obtained at high photon fluence rates (>=10–11 mol cm–2 s–1) display two main peaks which shift against each other at intermediate irradiances and, finally, form a single maximum in the blue range (443 nm) at low photon fluence rates (10–12 and 10–13 mol cm–2 s–1). Photon fluence rate-response curves reveal that supraoptimal irradiances decrease the phototactic reaction, especially in the range of the highest sensitivity of the cells. The action spectrum of phototaxis was calculated on the basis of the photon fluence rate-response curves. It shows a maximum at 443 nm and shoulder at 416 nm and between 467 and 477 nm. Wavelengths longer than 540 nm are phototactically inactive even at very high irradiances (25 W m–2). Thus, this is the first phototactic action spectrum of a biliprotein-containing organism which does not indicate the participation of biliproteins in the absorption of phototactically active light.
