Aims: Aim of this study is to determine the genetic variation of rhizobia associated with horse gram [Macrotyloma uniflorum (Lam.) Verdc.] plants grown in different regions of Andhra Pradesh, India. Methods and Results: Four representative isolates having most representative characters from the previous characterization were selected for 16S rRNA sequence. The sequences were submitted to the NCBI GenBank and Ribosomal Database Project (RDP). The isolates HGR‐4, 6 and 13 showed more than 99% homology between them and they were grouped with Rhizobium reference strains where as the isolate HGR‐25 showed 87·1, 87·4 and 87·2% homology with the isolates HGR‐4, 6 and 13, respectively, and were grouped with reference strains for Caulobacter. The nodulation ability of these isolates on horse gram was confirmed by inoculation tests. Conclusions: The isolate HGR‐25 was identified as Caulobacter isolated from the plants growing in soil samples collected from Khareemnagar district, Andhra Pradesh, India. Inoculation tests revealed that Caulobacter formed nodules on horse gram. It was also confirmed by RDP. Significance and Impact of the Study: This is the first report that a legume was nodulated by a member of the genus Caulobacter, which belongs to the family Caulobacteriaceae in the order Caulobacterales of Alphaproteobacteria. 相似文献

The contemporary data of the participation of phenylalanine in the biosynthesis of fluorescent pigment pyoverdine PM of Pseudomonas putida strain M are presented. Using aro1phu1 mutant of this strain, it has been shown that one of the precursors of the dihydroxyquinoline moiety of the pyoverdine PM is phenylalanine in the D- or L-form. These results were confirmed in experiments with 14C-phenylalanine incorporation. Pyoverdine PM that was synthesized by aro1phu1 mutant from exogenous phenylalanine is identical with the pigment from wild type cells. 相似文献

A group of 85 isolates of haloalkaliphilic obligately chemolithoautotrophic sulphur-oxidizing bacteria belonging to the genus Thioalkalivibrio were recently obtained from soda lakes in Mongolia, Kenya, California, Egypt and Siberia. They have been analyzed by repetitive extragenic palindromic (rep)-PCR genomic fingerprinting technique with BOX- and (GTG)5-primer set. Cluster analysis was performed using combined fingerprint profiles and a dendrogram similarity value (r) of 0.8 was used to define the same genotype. Fifty-six genotypes were found among the isolates, revealing a high genetic diversity. The strains can be divided into two major clusters, including isolates from the Asiatic (Siberia and Mongolia) and the African (Kenya and Egypt) continents, respectively. The majority (85.9%) of the genotypes were found in only one area, suggesting an endemic character of the Thioalkalivibrio strains. Furthermore, a correlation between fingerprint clustering, geographic origin and the characteristics of the lake of origin was found. 相似文献

Several extreme thermophilic Gram negative bacteria found in a thermally polluted river in Belgium have been compared with Thermus strains isolated from widely distant geographical areas. This analysis has become possible after the design of a new culture medium (162).All strains examined (including the isolate successively denominated Flavobacterium thermophilum and Thermus thermophilus) were found to be morphologically identical with strain YT-1 of Thermus aquaticus. The cells are immotile, rod-like, strictly aerobic, catalase and oxidase positive. They produce amylase, hydrolyze gelatin and are confirmed to be highly sensitive towards penicillin.The nutritional pattern of all strains has been analysed extensively, by testing a broad spectrum of possible substrates.The strains display a uniform response to the microbiological tests applied and most probably belong to the same species: Thermus aquaticus.Abbreviations GC

Bacteria belonging to the Pseudomonas genus and isolated from zonal soils in different geographical zones of the USSR as well as from the rhizosphere of cultivated and wild plants were tested for their esterase activity. The studied collection of cultures included 205 strains of different pigmented Pseudomonas species which, according to the conventional taxonomy, were assigned to the so-called "Pseudomonas fluorescens complex". As was shown in this study, many Pseudomonas species are potential producers of nonspecific esterases. P. maltophilia and P. geniculata synthesizing pyomelanin have the highest activity of esterase. The activity of esterase correlates with the formation of a melanin-like pigment in Pseudomonas cultures. It also correlates with the species to which a culture belongs, which makes it possible to use this property as an additional criterion for the identification of Pseudomonas species. 相似文献

A substantial amount of sediment phosphorus can be bound in bacterial biomass. In this study the fractional composition of phosphorus in the bacteria Pseudomonas was determined by sequential extraction with ammonium chloride, sodium hydroxide and hydrochloric acid according to the scheme of Hieltjes & Lijklema (1980). Both non-labelled and 32P-labelled bacteria were used for fractionation. Up to 80% of the bacterial phosphorus was found in the NaOH-nRP fraction, which is in agreement with the results of Hupfer & Uhlman (1992) for Acinetobacter and activated sludge obtained with the sequential extraction scheme of Psenner et al. (1985). A significant correlation was found between bacterial biomass and the amount of phosphorus retained in the NaOH-nRP fraction when sediments were fractionated. Additional experiments with 32P-labelled Pseudomonas in sediment-water systems were performed in order to follow bacterial release of phosphorus under aerobic and anaerobic conditions. These studies did not sustain the hypothesis that anaerobic conditions lead to rapid release of phosphorus from bacterial cells. 相似文献

In 290 strains of bacteria belonging to the genus Pseudomonas, 120 morphological and physiologo-biochemical characters were studied and the results obtained thereby were analyzed by the methods of numerical taxonomy using computers. The majority of strains were subdivided into 11 clusters: Ps. aeruginosa (1), Ps. putida (2), Ps. rathonis (5), Ps. syringae (8), Ps. pseudoalcaligenes (9), Ps. maltophilia (10), Ps. acidovorans (11), Ps. testosteroni (12), Ps. mendocina (13), Ps. cepacia (14), Ps. fluorescens (3). The latter cluster included also the strains identified earlier as Ps. aurantiaca, Ps. lemonnieri, Ps. fluoro-violaceus, and Ps. aureofaciens. Three clusters contained strains which could not be identified and probably should be regarded as distinct species. The characteristics have been selected useful for diagnostics of the above Pseudomonas bacteria and the subgroups of Ps. fluorescens. 相似文献

The nature of phenylalanine auxotrophy in Pseudomonas sp. strain M9 was studied. A metabolic defect was found after the aromatic amino acid pathway branches from chorismate, so that the latter could not be converted to phenylpiruvate. It was shown that enzymatic activity of the strain could convert phenyllactate to phenylalanine. The reaction mechanisms were studied, and approaches to increasing the rate of the process and phenylalanine yield were proposed. 相似文献

Fatty acid composition during naphthalene utilization was investigated in three strains of bacteria Pseudomonas vesicularis, Pseudomonas stutzeri and Pseudomonas sp. JS150 that expressed different naphthalene degradation abilities. All strains significantly changed their cellular fatty acid profiles as a response to naphthalene exposure. Since naphthalene was present in the medium P. stutzeri increased ratio of saturated/unsaturated fatty acids from 1.1 to 2.1 and Pseudomonas sp. JS150 from 7.5 to 12.0, respectively. In contrast, this ratio decreased from 2.1 to 1.1 in P. vesicularis under the same growth conditions. The changes comprised also alterations in the percentage of selected groups of fatty acids: iso and anteiso, hydroxy and cyclopropane fatty acids. Our results showed that naphthalene induced in tested strains different changes in fatty acids composition. It may suggest that in the presence of naphthalene microorganisms used different adaptive mechanisms to maintain the cells in appropriate physiological state. 相似文献

A strain of the genera Pseudomonas genera was found to possess hemolytic, fibrinolytic and thrombolytic activities. The fibrinolytic activity of the lyophilized unpurified preparation was 900 conventional units/mg. After incubation in the blood plasma, the activity completely remained. The preparation (1 microgram/ml, 750 micrograms of protein) obtained by precipitation with ammonium sulfate (80% saturation) completely lysed in vitro human blood thrombi for 50 min. Active sulphate-reducing microorganism which belongs to the genus Pseudomonas has been distinguished and described. The culture is a facultative aerobe, optimum Eh is -170-180 mV. Pseudomonas sp. being cultivated under strictly anaerobic conditions sulphate-reduction proceeds more intensively than under aerobic conditions.
