Salt Tolerance in the Triticeae: Ion Discrimination in Rye and Triticale   总被引:5,自引:0,他引:5  
When rye and triticale accessions were grown in saline hydroponicculture they exhibited the low Na and high K concentrationsin their leaves which are characteristic of the enhanced K/Nadiscrimination trait originally found in the D genome of wheat.This trait was not consistently improved by the presence ofthe D genome in octaploid triticale or in D genome substitutionlines of hexaploid triticale. The presence of the rye genomedid not significantly affect anion concentrations within theleaves. At high salt concentrations (250 mol m–3 NaCl+12.5mol m–3 CaCl2) the triticales were more tolerant thanthe rye accessions or a DDRR-genome tetraploid, with two triticalelines being almost as salt-tolerant as barley. Concentrations of ions were measured in the youngest fully-expandedleaves of Triticum aestivum, T. durum, Hordeum vulgare, H. spontaneum,Secale cereale, and Aegilops squarrosa accessions grown in hydroponicculture in the presence of salt (NaCl+CaCl2). Triticum aestivum,Secale cereale, and Ae. squarrosa had the low leaf Na and highleaf K concentrations typical of plants which contain the enhancedK/Na discrimination character originally found in Ae. squarrosa.T. durum and the Hordeum species did not have this character.The better growth of H. vulgare than of T. durum with similarsalt concentrations in the youngest fully-expanded leaves maybe a result of better compartmentation of Na, Cl, and K betweendifferent tisssues or between different compartments withincells. The enhanced K/Na discrimination character was expressedin disomic addition lines of H. vulgare chromosomes in Triticumaestivum. The H. vulgare variety Herta and its slender mutantboth had similar leaf cation concentrations, although they differedin growth rate when grown at 60 mol m–3 NaCl. H. vulgareand T. durum seedlings grown in the absence of monovalent cationsaccumulated more 22Na in their shoots than seedlings of otherspecies when incubated in 1.0 mol m–3 NaCl labelled with22Na. Inorganic ion concentrations were measured in the leaves ofAegilops species growing in 50 or 75 mol m–3 NaCl (+2.5or 3.75 mol m–3 CaCl2). The low leaf Na and high leafK concentrations characteristic of the enhanced K/Na discriminationcharacter, originally found in Aegilops squarrosa and in hexaploidwheat, were also found in other Aegilops species containingthe D genome, but not in Ae. ventricosa. The S genome diploidAegilops species (section Sitopsis) all lacked the enhancedK/Na discrimination trait, as did the C genome species Ae. caudataand the N genome species Ae. uniaristata. Most of the U genomespecies (section Polyeides), except Ae. biuncialis, Ae. kotschyiand Ae. variabilis, also exhibited the trait. Examination ofamphiploid hybrids suggested that the trait was dominant incrosses involving Ae. squarrosa or Ae. umbellulata with otherspecies in which the trait was absent. Ion concentrations were measured in the leaves of synthetichexaploid wheats and their parents growing in saline hydroponicculture. The synthetic hexaploids contained genomes from a tetraploidwheat (Triticum diccocum, T. durum, T. araraticum or T. timopheevi)and from a diploid species (T. monococcum, T. urartu, T. boeoticumor Aegilops squarrosa). Leaf Na concentrations were low, andK concentrations high, in Ae. squarrosa, T. araraticum and allof the synthetic hexaploids, but high in T. dicoccum and T.durum. At low salinities leaf Na concentrations were particularlyhigh in T. durum in comparison with the other species. Theseresults suggest that the enhanced K/Na discrimination character,originally found in Ae. squarrosa and BBAADD genome hexaploidwheats, is also present in diploid wheat and in GGAA genometetraploid wheats. Inorganic cation concentrations were measured in shoots of hexaploidbread wheat (Triticum aestivum L.) and its presumed ancestorsgrown at 100 mol m–3 external NaCl. Aegilops squarrosaand T. aestivum had high K/Na ratios while T. dicoccoides andAe. speltoides had low K/Na ratios. T. monococcum although havinga high K/Na ratio, had the highest total salt load of the fivespecies tested. The effect of the D genome (from Ae. squarrosa)was further investigated in seedlings of synthetic hexaploidwheats, and was again found to improve cation selectivity. Differentresponses were obtained from root and shoot tissue in this experiment.One synthetic hexaploid and its constituent parents were grownto maturity at 100 mol m-3 NaCl and the yields recorded. Despitecomplications due to increased tillering in the stressed hexaploid,it was possible to show that the addition of the D genome enhancedyield characteristics in the hexaploid wheat. An experimentwith synthetic hexaploids derived from the tetraploid wheatvariety "Langdon" and several Ae. squarrosa accessions revealeddifferences in vegetative growth rates between the differentsynthetic hexaploids in the presence or absence of 150 or 200mol m–3 external NaCl. The possibility of transferringsalt tolerance genes from Ae. squarrosa to hexaploid wheat usingsynthetic hexaploids as bridging species is discussed. Amounts of the enzyme ribulose-1,5-bisphosphate carboxylasewere estimated in seedling leaves of barley (Hordewn vulgareL.) and flag leaves of wheat (Triticum aestitum L.) by radialimmuno diffusion. A fourfold variation among barley varietiesfor amount of RuBPCase at the seedling stage was observed (c.3.5–15mg g–1 fr. wt). Range in variation for amountof flag leaf RuBPCase among wheat varieties was 6-09-9.39 mgRuBPCase g–1 fr. wt. F1 hybrids from interspecific andintergeneric crosses of crested wheatgrasses (Agropyron andElymus spp.) and their amphidiploid analogues were comparedfor amount of RuBPCase in the most recent fully expanded leavesharvested before seed set. Amount of enzyme varied from 3.4to 77.6 mg g–1 fr. wt among the hybrids. No effect chromosomenumber on enzyme concentration was observed among 13 hybridsand their amphidiploid counterparts. Elymus dahuhcus, Leymus giganteus, L. angustus, L. sabulosusand, to a lesser extent, L. triticoides, were found to tolerate200 mol m–3 NaCl in solution culture. Elymus dahuricusdiffered from the Leymus species in its ion-uptake characteristics,showing a greater uptake of Cl and Na and a greater loss ofK from the shoots. In a more detailed experiment on Leymus sabulosusit was found that transpiration rates altered rapidly in responseto changes in external salinity whereas the accumulation ofNa and Cl in the leaves exhibited a lag of several days. Insalt stressed L. sabulosus Cl partially replaced the high levelsof nitrate found in the leaves of control plants. Glycinebetainelevels increased in the leaves from 8.0 mol m–3 plantsap in the controls to 28 mol m–3 plant sap at 250 molm–3 NaCl. Summary A number of accessions of the three species of diploid wheat, Triticum boeoticum, T. monococcum, and T. urartu, were grown in 50 mol m-3 NaCl+2.5 mol m-3 CaCl2. Sodium accumulation in the leaves was low and potassium concentrations remained high. This was not the case in T. durum grown under the same conditions, and indicates the presence in diploid wheats of the enhanced K/Na discrimination character which has previously been found in Aegilops squarrosa and hexaploid wheat. None of the accessions of diploid wheat showed poor K/Na discrimination, which suggests that if the A genome of modern tetraploid wheats was derived from a diploid Triticum species, then the enhanced K/Na discrimination character became altered after the formation of the original allopolyploid. Another possibility is that a diploid wheat that did not have the enhanced K/Na discrimination character was involved in the hybridization event which produced tetraploid wheat, and that this diploid is now extinct or has not yet been discovered.  相似文献

Using excised low-salt roots of barley and Atriplex hortenslsthe transport of endogenous potassium through the xylem vesselswas studied It was enhanced by nitrate and additionally by sodiumions which apparently replaced vacuolar potassium which wasthen available in the symplasm of root cells for transport tothe shoot Vacuolar Na/K exchange also has been investigatedby measurements of longitudinal ion profiles in single rootsof both species. In Atriplex roots a change in the externalsolution from K+ to Na+ induced an exchange of vacuolar K+ forNa+, in particular in the subapical root tissues and led toincreased K+ transport and loss of K+ from the cortex. In inverseexperiments a change from Na+ to K+ did not induce an exchangeof vacuolar Na+; merely in meristematic tissues Na+—apparentlyfrom the cytoplasm—was extruded in exchange for K+. Inroots of barley seedlings without caryopsis, as in excised roots,a massive exchange of K+ for Na+ was observed in the continuouspresence of external 1.0 mM Na and 0.2 mM K. This exchange alsowas attributed to the vacuole and was most pronounced in theyoung subapical tissues. It did not occur, however, in the correspondingtissues in roots of fully intact barley seedlings. In these,the young tissues retained a relatively high K/Na ratio alsoin their vacuoles. Similarly, contrasting results were obtainedwith intact and excised roots of Zea mays L. Based on theseresults a scheme of the events that lead to selective cationuptake in intact barley roots is proposed. In this scheme acrucial factor of selectivity is sufficient phloem recirculationof K+ by the aid of which K+ rich cortical cells are formednear the root tip. When matured these cells are suggested tomaintain a high cytoplasmic K/Na ratio due to K+ dependent sodiumextrusion at the plasmalemma and due to recovery of vacuolarK+ by Na/K exchange across the tonoplast. Ricinus communis L. (castor bean) plants were grown in the absence(control) and in the presence of 100molm–3NaCl with areciprocal split-root system, in which K+ was supplied to oneand NO3 to the other part of the root system. In theseplants shoot and, to a lesser extent, total root growth wereinhibited compared to plants with non-split roots. Without andwith NaCl, growth of roots receiving NO3 but noK+ (‘minusK/plus N-roots’) was substantially more vigorous thanunder the reverse conditions (‘plus K/minus N-roots1).100mol m–3 NaCl inhibited growth of minus K/plus N-roots1to the same extent as that of non-split roots, indicating thatexternally supplied K+ was not required for root growth undersaline conditions. In growth media without added K+ the rootdepleted the external low K + levels resulting from chemicalsdown to a minimum value Cmln (1.0 to 1.4 mmol m–3); inthe presence of 100 mol m–3 NaCl, Cmin, was higher (10–18mmol m–3) and resulted from an initial net loss of K +.Cmin, was pH-dependent The distribution of K+, Na+ and Mg2+along the root was measured. In meristematic root tissues, K+ concentrations were scarcely affected by external K+ or byNaCl, where Na + concentrations were low, but somewhat elevatedat low external K+ and/or high NaCl. In differentiated, vacuolatedtissues K + concentrations were low and Na+ concentrations high,if K + was not supplied externally and/or NaCl was present.The longitudinal distribution of ions within the root was usedto estimate cytoplasmic and vacuolar ion concentrations. Thesedata showed a narrow homoeostasis of cytoplasmic K+ concentrations(100–140 mol m–3) independent of external K + supplyeven in the presence of 100 mol m –3 NaCl. CytoplasmicNa + concentrations were maintained at remarkably low levels.Hence, external K+ concentrations above Cmin, were not requiredfor maintaining K/Na selectivity, i.e. for controlling Na+ entry.The results are discussed with regard to mechanisms of K/Naselectivity and to the importance of phloem import of K+ forsalt tolerance of roots and for cytoplasmic K+ homoeostasis. The diploid wheatgrass Thinopyrum bessarabicum was found towithstand prolonged exposure to 350 mol m–3 NaCl in hydroponicculture. During the gradual addition of salt to the externalmedium, osmotic adjustment was rapidly achieved by the accumulationof Na and Cl. Following osmotic adjustment constant leaf Naand Cl concentrations were maintained, and K was retained ata high level. Thinopyrum bessarabicum may be described as anosmoconformer, adjusting its internal osmotic pressure to 400–500mOsmol kg–1 above that of the external medium in hydroponicculture. Both slower shoot initiation and reduced leaf lengthcontributed to the reduced growth rates at higher salinities.Leaf width was not affected. Increasing salinity resulted inincreases in leaf concentrations of phosphate, glycinebetaine,sucrose and proline, and in decreases in the concentrationsof nitrate, sulphate, magnesium, calcium, total amino acidsand organic acids. The growth of garden orache, A triplex hortensis was studiedunder conditions of mild NaCl or Na2SO4 salinity. Growth, drymatter production and leaf size were substantially stimulatedat 10 mM and 50 mM Na+ salts. Increased growth, however, appearedto be due to a K+-sparing effect of Na+ rather than to salinityper se. The distribution of K+ and Na+ in the plant revealeda remarkable preference for K+ in the roots and the hypocotyl.In the shoot the K/Na ratio decreased strongly with leaf age.However, the inverse changes in K+ and Na+ content with leafage were dependent on the presence of bladder hairs, which removedalmost all of the Na+ from the young leaf lamina. Measurementsof net fluxes of K+ and Na+ into roots and shoots of growingAtriplex plants showed a higher K/Na selectivity of the netion flux to the root compared to the shoot. With increasingsalinity the selectivity ratio SK, Na* of net ion fluxes tothe roots and to the shoots was increased. The data suggestthat recirculation of K+ from leaves to roots is an importantlink in establishing the K/Na selectivity in A. hortensis plants.The importance of K+ recirculation and phloem transport forsalt tolerance is discussed. Summary Triticum aestivum cv. Chinese Spring wheat,Elytrigia elongatum (tall wheatgrass), and theTriticum-Elytrigia amphiploid were grown in complete nutrient culture containing, in addition, 0, 40, 80 and 120 mM NaCl. The 3 genotypes responded quite differently to increasing salinity; the Na concentration of wheat shoots increased in direct proportion to the increase in salinity of the external medium whereas the Elytrigia response was interpreted as showing high affinity for Na at low external Na (40 mM) but comparative exclusion of Na at high salinities (120 mM). In contrast, Na levels of the amphiploid were less than those of either wheat or Elytrigia under both low and high salinities. Thus the amphiploid behaved like wheat at 40 mM NaCl but more like Elytrigia at 120 mM NaCl because Na transport to the amphiploid shoot was restricted over the whole salinity range. The K concentration of the amphiploid shoot at high salinities was significantly greater than the K concentrations of either wheat or Elytrigia.  相似文献

Water loss was studied in regenerated plantlets of Brassicaoleracea var. botrytis cv. Currawong derived through apicalmeristem culture. Hardening of plantlets was eliminated by asingle application of a polyvinyl resin (S600) sprayed immediatelyafter transplanting. Plantlets sprayed with S600 had highercuticular resistances than unsprayed plantlets; this treatmenthad no effect on stomatal resistance. Leaves formed during theculture period showed very little wax formation and using markedleaves it was found that only reduced levels of wax formed onthese leaves even after transplanting. New leaves formed aftertransplanting, showed typical wax formation compared to seedgrown plants. Abscisic acid (ABA) at 10–4 M applied as a leaf sprayto transplants did not cause a substantial increase in stomatalresistance in leaves which had been initiated during the cultureperiod. Leaves of seed-grown plants as well as leaves of plantletsformed after transplanting did respond to a leaf spray of ABAat 10–4 M by a large increase in stomatal resistance. Relative concentrations of K, Na, Ca, P, S and Mg in guard cellswere calculated for each leaf type by X-ray micro-probe analysis.K/Na values decreased in the order: seedling > leaves formedafter transplanting > leaves intiated during culture. A highpositive correlation was also found between K/Na and K/P forthe three leaf types. K:Mg and K:Ca ratios for leaves formedduring culture were low in comparison to the values obtainedfor leaves formed after transplanting and seedlings for whichthe values were similar. Halosarcia pergranulata(P. G. Wilson) subsp.pergranulatais amember of the Salicornioideae and is native to Australia. Salttolerance inH. pergranulatasubsp.pergranulatawas assessed bygrowing plants for 83 d at seven NaCl concentrations from 10to 800 mol m-3. Shoot biomass was greatest for plants grownat 10 to 200 mol m-3NaCl, while at salinities of 300 mol m-3orhigher it was inhibited. Salt tolerance of plants depends on HKT transporters (High-affinityK+ Transporter), which mediate Na+-specific transport or Na+-K+co-transport. Gene sequences closely related to rice HKT geneswere isolated from hexaploid bread wheat (Triticum aestivum)or barley (Hordeum vulgare) for genomic DNA southern hybridizationanalysis. HKT gene sequences were mapped on chromosomal armsof wheat and barley using wheat chromosome substitution linesand barley–wheat chromosome addition lines. In addition,HKT gene members in the wild diploid wheat ancestors, T. monococcum(Am genome), T. urartu (Au genome), and Ae. tauschii (Dt genome)were investigated. Variation in copy number for individual HKTgene members was observed between the barley, wheat, and ricegenomes, and between the different wheat genomes. HKT2;1/2-like,HKT2;3/4-like, HKT1;1/2-like, HKT1;3-like, HKT1;4-like, andHKT1;5-like genes were mapped to the wheat–barley chromosomegroups 7, 7, 2, 6, 2, and 4, respectively. Chromosomal regionscontaining HKT genes were syntenic between wheat and rice exceptfor the chromosome regions containing the HKT1;5-like gene.Potential roles of HKT genes in Na+ transport in rice, wheat,and barley are discussed. Determination of the chromosome locationsof HKT genes provides a framework for future physiological andgenetic studies investigating the relationships between HKTgenes and salt tolerance in wheat and barley. There are conflicting reports about whether guard cells of Paphiopedilumspp. accumulate K during stomatal opening. In this study X-raymicroprobe analysis and histochemical staining for K indicatedthat K accumulated in guard cells in leaves of Paphiopedilumspp. when stomata opened. Additionally, stomatal opening inepidermal strips of P. harrisseanum could only be induced inan MES buffered KC1 medium. Element analysis ofP. harrisseanumepidermis also indicated substantial levels of K, Na and Cawithin the tissue. We conclude that K is involved in the stomatal movements ofPaphiopedilum. These experiments were performed to determine the effects ofreducing Ca2+ influx(Cain) onK+ currents(IK) inmyocytes from rat small mesenteric arteries by1) adding externalCd2+ or2) lowering externalCa2+ to 0.2 mM. When measured froma holding potential (HP) of 20 mV(IK20),decreasing Cain decreasedIK at voltageswhere it was active (>0 mV). When measured from a HP of 60 mV(IK60),decreasing Cain increasedIK at voltagesbetween 30 and +20 mV but decreased IK at voltagesabove +40 mV. Difference currents(IK) weredetermined by digital subtraction of currents recorded under controlconditions from those obtained whenCain was decreased. At testvoltages up to 0 mV,IK60 exhibitedkinetics similar to controlIK60, with rapidactivation to a peak followed by slow inactivation. At 0 mV, peakIK60 averaged75 ± 13 pA (n = 8) withCd2+ and 120 ± 20 pA(n = 9) with lowCa2+ concentration. At testvoltages from 0 to +60 mV,IK60 always had an early positive peak phase, but its apparent "inactivation" increased with voltage and its steady value became negative above +20mV. At +60 mV, the initial peakIK60 averaged115 ± 18 pA with Cd2+ and 187 ± 34 pA with low Ca2+. With 10 mM pipette BAPTA, Cd2+ produced asmall inhibition ofIK20 but stillincreased IK60 between 30 and +10 mV. InCa2+-free external solution,Cd2+ only decreased bothIK20 andIK60. In thepresence of iberiotoxin (100 nM) to inhibitCa2+-activatedK+ channels(KCa),Cd2+ increasedIK60 at allvoltages positive to 30 mV while BAY K 8644 (1 µM) decreasedIK60. Theseresults suggest that Cain, through L-type Ca2+ channels and perhapsother pathways, increases KCa(i.e., IK20) and decreases voltage-dependent K+currents in this tissue. This effect could contribute to membrane depolarization and force maintenance.

Effluxes of K+ and Ca2+ from root segments of both wheat, Triticunaestivum L. cv. Capelle and mung bean, Vigna radiata (L.) Wilczek,were measured in the presence or absence of 20 mol m–3para-fluorophenylalanine (p-FPA). The results were used to estimatethe compartment contents and transmembrane K+ and Ca2+ fluxesin root cortex cells. Using the Ussing-Teorell flux equationas the criterion, it was concluded that entry of K+ from theoutside solution to the cytoplasm, and from the cytoplasm tothe vacuole were active in both wheat and mung bean. Also, inboth species, Ca2+ entered the cytoplasm passively across theplasmalemma and was actively pumped back to the external solution.However, interpretation of the direction of active transportacross the tonoplast depends upon an assumption about Ca2+ activityin the cytoplasm. The only qualitative effect of p-FPA was to alter the drivingforce for K+ influx, across the plasmalemma in wheat, from anactive to a passive one. Quantitative effects of the analoguewere seen for K+ fluxes in both wheat and mung bean and forCa2+ fluxes in wheat. The p-FPA reduced transport of K+ in bothspecies, while transport of Ca2+ was unaffected. The implicationsof these results for the ‘two pump hypothesis’ arediscussed. Studies on the germination of Cassia acutifolia (senna) seedsindicated, that they were highly tolerant of salinities up to16.0 mmhos cm–1, and 50 per cent reduction in germinationoccurred, at about 20.0 mmhos cm.1. Air temperature had a significanteffect on senna germination at substrate salinities between10.0 and 25.0 mmhos cm–1. Seedling growth was more sensitive to salinity and alkalinitythan the germination stage. Young plant survival and total podyield were significantly reduced in soil salinities higher than11.0 minhoa cm–1, and this was more pronounced when plantswere irrigated at short intervals with saline waters. The sensitivityof senna to higher levels of salinities was correlated withthe higher rate of chloride accumulation in the tissue resultingin specific chloride injury. However, the possibility of sodiuminjury cannot be excluded.  相似文献
