Summary A total of 1475 individuals belonging to 43 natural populations of seven diploid (2x) blueberry species (Vaccinium section Cyanococcus) and two natural interspecific 2x hybrid populations were evaluated for unreduced pollen production. Significant differences were found in the frequency of unreduced pollen producers between species and within and between populations of the same species. Individuals with 1% or more unreduced pollen were considered unreduced pollen producers. The average frequency of unreduced pollen producers in these diploid species was 13.5%, ranging from 7.4% (V. corymbosum) to 18.4% (V. darrowii). The frequency of unreduced pollen grains in individual clones varied from <-1% to 28.6%. The production of unreduced pollen was not associated with male fertility. Dehydrins are plant proteins that may play a critical role in stabilizing cell functions during freezing and other dehydrative stresses. This study examines whether dehydrin expression in leaves is associated with varying levels of freezing-tolerance among F2 segregants, species, and cultivars of evergreen Rhododendron. Experiments were also conducted to determine whether physiological and chronological aging affects freezing-tolerance and dehydrin accumulation in Rhododendron leaf tissues. Our results indicate that in cold-acclimated F2 populations, levels of a 25-kDa dehydrin were closely associated with differences in leaf freezing-tolerance (LFT) among segregants. Studies of wild and cultivated plants indicated that LFT increased with both chronological age and developmental phase-change (juvenile to mature plants) and that this trend was accompanied by increased accumulation of the 25-kDa dehydrin. It is suggested that presence or absence of the 25-kDa dehydrin could serve as a genetic marker to distinguish between super cold-hardy and less cold-hardy rhododendron genotypes. Similarly, the relative level of this protein within a genotype can serve as a physiological indicator of freezing-tolerance status under a range of phenological (acclimation) or developmental (age) conditions. The level of three major polypeptides of 65, 60, and 14 kD increased in response to chilling unit accumulation in floral buds of a woody perennial, blueberry (Vaccinium, section Cynaococcus). The level of the polypeptides increased most dramatically within 300 h of chilling and decreased to the prechilling level with the initiation of budbreak. Cold-hardiness levels were assessed for dormant buds of Vaccinium corymbosum and Vaccinium ashei after different chilling treatments until the resumption of growth. These levels coincided with the level of the chilling-responsive polypeptides. Like some other previously described cold-induced proteins in annual plants, the level of the chilling-induced polypeptides also increased in leaves in response to cold treatment; the chilling-induced polypeptides were heat stable, resisting aggregation after incubation at 95 degrees C for 15 min. By fractionating bud proteins first by isoelectric point (pI) and then by molecular mass, the pI values of the 65- and 60-kD polypeptides were found to be 7.5 to 8.0 and the pI value of the 14-kD polypeptide was judged to be 8.5. Purification of the 65- and 60-kD polypeptides, followed by digestion with endoproteinase Lys-C and sequencing of selected fragments, revealed similarities in amino acid composition between the 65- and 60-kD polypeptides and dehydrins. Indeed, antiserum to the lysine-rich consensus sequence EKKGIMDKIKEKLPG of dehydrin proteins cross-reacted to all three of the major chilling-responsive polypeptides of blueberry, identifying these as dehydrins or dehydrin-like proteins.  相似文献

Blueberry is a widely grown and easily perishable fruit crop. An efficient post-harvest handling is critical, and for that purpose gene technology methods have been part of ongoing programmes to improve crops with high food values such as blueberry. Here we report the isolation, cloning, characterization and differential expression levels of two cDNAs encoding Polygalacturonase-Inhibitor Protein (PGIP) and Cinnamoyl-Coa Reductase (CCR) from blueberry fruits in relation to various storage conditions. The open reading frame of PGIP and CCR encodes a polypeptide of 329 and 347 amino acids, respectively. To assess changes in the expression of blueberry PGIP and CCR after harvest, a storage trial was initiated. The northern blots hybridization showed a clear differential expression level of PGIP and CCR between freshly harvested and stored fruits as well as between fruits stored under various storage conditions. Although the prospects of exploiting such a strategy for crop improvement are limited, the results provide further insight into the control of the quality over the storage period at the molecular level.  相似文献

A meta-analysis of 520 parents and 51,439 individuals was used to develop two equations for predicting age-age genetic correlations in Pinus taeda L. Genetic and phenotypic family mean correlations and heritabilities were estimated for ages ranging from 2 to 25 years on 31 sites in the southern U.S. and Zimbabwe. Equations for predicting age-age correlations based on P. taeda populations from west and east of the Mississippi River proved statistically different. Both predictive equations proved conservative for validation datasets consisting of younger tests in the U.S. and Zimbabwe. Age-dependent log-linear predictive equations were favored over growth-dependent equations. All P. taeda predictive equations based on genetic correlations favored earlier selection when compared to a generalized conifer predictive equation based on phenotypic correlations. Olive growth and productivity are limited by low temperatures mainly during winter, but sometimes also in spring and fall. The most effective way to avoid these damages in areas subjected to these climatic conditions is to select least susceptible varieties, but the choice of the right method to determine cold hardiness is extremely difficult. The aims of the work were (1) to assess LT50 (lethal temperature at which 50% of damage in plants subjected to low temperatures occurs) of some olive varieties in two seasons (summer and winter) and (2) to assess the reliability of different methods to evaluate cold hardiness. LT50 was determined on 21 different olive (Olea europaea L.) Italian varieties by leaf and shoot electrolyte leakage, shoot impedance spectroscopy and leaf color determination of fractal spectrum. All the experiments were conducted on non-acclimated and cold-acclimated plants. Our results showed that all the three methods were able to detect damages on olive plants after exposure to low temperatures, with leaves appearing more sensitive to cold stress than shoots. Among these methods, fractal analysis could be very useful in assessing cold hardiness of plants on the basis of visible injury, without sophisticated or expensive instruments and in a reliable and cost-effective way, using only a scanning device, a personal computer and dedicated freeware software.  相似文献

Genetic control of aluminium tolerance in rye (Secale cereale L.)   总被引:4,自引:0,他引:4  
 Aluminium (Al) tolerance in roots of two cultivars (“Ailés” and “JNK”) and two inbred lines (“Riodeva” and “Pool”) of rye was studied using intact roots immersed in a nutrient solution at a controlled pH and temperature. Both the cultivars and the inbred lines analysed showed high Al tolerance, this character being under multigenic control. The inbred line “Riodeva” was sensitive (non-telerant) at a concentration of 150 μM, whereas the “Ailes” cultivar showed the highest level of Al tolerance at this concentration. The segregation of aluminium-tolerance genes and several isozyme loci in different F1s, F2s and backcrosses between plants of “Ailés” and “Riodeva” were also studied. The segregation ratios obtained for aluminium tolerance in the F2s analysed were 3 : 1 and 15 : 1 (tolerant : non-tolerant) while in backcrosses they were 1 : 1 and 3 : 1. These results indicated that Al tolerance is controlled by, at least, two major dominant and independent loci in rye (Alt1 and Alt3). Linkage analyses carried out between Al-tolerance genes and several isozyme loci revealed that the Alt1 locus was linked to the aconitase-1 (Aco1), nicotinamide adenine dinucleotide dehydrogenase-2 (Ndh2), esterase-6 (Est6) and esterase-8 (Est8) loci, located on chromosome arm 6RL. The order obtained was Alt1-Aco1-Ndh2-Est6-Est8. The Alt3 locus was not linked to the Lap1, Aco1 and Ndh2 loci, located on chromosome arms, 6RS, 6RL and 6RL respectively. Understanding the genetic mechanisms for the phenotypic plasticity and developmental instability of a quantitative trait has important implications for breeding and evolution. Two clonally replicated plantations of two 3-generation inbred pedigrees derived from the highly divergent species Populus trichocarpa and P. deltoides were used to examine the genetic control of macro- and micro-environmental sensitivities and their genetic relationships with the trait mean across two contrasting environments. For all stem-growth traits studied, the trait mean had a higher broad-sense heritability (H2) level than macroenvironmental sensitivity, both with much higher values than microenvironmental sensitivity. Genetic correlation analyses indicated that the trait mean was more or less independent of macro- or micro-environmental sensitivity in stem height. Thus, for this trait, the genetic difference in response to the two environments might be mainly due to epistasis between some regulatory loci for plasticity and loci for trait mean. However, for basal area and volume index, pleiotropic loci might be more important for their genetic differences between the two environments. No evidence was found to support Lerner’s (1954) homeostasis theory in which macro- or micro-environmental sensitivity is the inverse function of heterozygosity.  The freezing and freezing survival of the Antarctic nematode Panagrolaimus davidi after exposure to solutions of different osmotic concentrations has been examined using a thermoelectric cooling stage and multi-specimen cooling block to see if there is any evidence that freeze-induced desiccation prevents inoculative freezing. The nematodes froze in all the test solutions used (up to 1138 mosmol ⋅ l-1) and at all cooling rates and nucleation temperatures tested. Freezing survival was at its maximum in 0.1 mol ⋅ l-1 NaCl in artificial tap water after 1 h exposure to the test solution and in artificial tap water after 24 h exposure. Hyperosmotic and hyposmotic stress adversely affected the nematodes’ ability to survive freezing. In non-frozen controls survival declined with increasing osmolality of the test solution. Measurements of the osmolality of water extracted from a variety of moss samples indicate that the nematodes are exposed to an osmotic concentration of about 9 mosmol ⋅ l-1 in their natural habitat. This is close to that of artificial tap water. Our experiments, and measurements of freeze concentration effects in the literature, indicate that freeze-induced desiccation is unlikely to prevent inoculative freezing and the survival of nematodes over the winter. Diplocarpon rosae is the causal agent of rose blackspot, one of the most severe diseases of field-grown roses. The genetics of resistance to this pathogen was investigated in crosses between tetraploid rose genotypes. The hybrid breeding line 91/100-5, which exhibits a broad resistance to all isolates tested so far, was selfed to produce an F2 population, backcrossed to the susceptible tetraploid variety ‘Caramba’ and crossed to the susceptible varieties ‘Heckenzauber’, ‘Pariser Charme’ and ‘Elina’. Infection experiments resulted in segregation ratios consistent with the presence of a single dominant resistance locus in the duplex configuration in the hybrid 91/100-5. This suggests, together with previous data on the race structure of the fungus, a “gene-for-gene” type of interaction in the pathosystem Diplocarpon/Rosa. We propose to designate this gene Rdr1, which is the first resistance gene described in the genus Rosa. The advantages and limitations of such an interaction type for future rose breeding programmes and for marker-assisted selection strategies are discussed. Colletotrichum gloeosporioides causes anthracnose, the most severe foliar disease of field-grown water yam (Dioscorea alata). The inheritance of resistance to a moderately virulent (FGS) strain of the pathogen was investigated in crosses between tetraploid D. alata genotypes: TDa 95/00328 (resistant)×TDa 95–310 (susceptible) (cross A), and TDa 85/00257 (resistant)×TDa 92–2 (susceptible) (cross B). Segregation of F1 progeny fitted genetic ratios of 3:1, 5:1 (crosses A and B) and 7:1 (cross A) resistant:susceptible when inoculated with the FGS strain, indicating that resistance is dominantly inherited and suggesting that more than one gene controls the inheritance of resistance to this strain in the accessions studied. When parental and progeny lines of cross A were inoculated with an aggressive (SGG) strain of the pathogen, all plants expressed a susceptible phenotype, indicating strain-specific resistance in TDa 95/00328. Screening of 20 cultivars/landraces confirmed the high susceptibility of D. alata accessions to the SGG strain and revealed the presence of apparent strain non-specific resistance in TDa 85/00257. TDa 85/00257 and TDa 87/01091 which were resistant to the SGG strain, will be useful both as sources of resistance and in the development of a host differential series for D. alata. Greenhouse pollinations were performed to determine whether early-acting inbreeding depression is contributing to low levels of self-fertility in three Vaccinium species: V. myrtilloides Michaux, V. angustifolium Aiton, and V. corymbosum L. All three species showed a significant reduction in self fruit set and in the proportion of fertilized ovules that developed into mature seed in self compared to outcross fruit. Reductions were more severe in V. myrtilloides and V. angustifolium than in V. corymbosum; however, early-acting inbreeding depression appeared to be the primary factor limiting self-fertility in all three species. Evidence for early- acting inbreeding depression included the presence and higher proportion of aborted ovules in self fruit than in outcross fruit, a correlation between levels of self and outcross seed set, and pollen chase experiments demonstrating that self-pollen does fertilize the ovules. Grain yield is a complex trait, strongly influenced by the environment: severe losses can be caused by drought, a stress common in most maize-growing areas, including temperate climatic zones. Accordingly, drought tolerance is one of the main components of yield stability, and its improvement is a major challenge to breeders. The aim of the present work was the identification, in maize genotypes adapted to temperate areas, of genomic segments responsible for the expression of drought tolerance of yield components: ear length, ear weight, kernel weight, kernel number and 50-kernel weight. A linkage analysis between the expression of these traits and molecular markers was performed on a recombinant inbred population of 142 families, obtained by repeated selfing of the F1 between lines B73 and H99. The population, genotyped at 173 loci (RFLPs, microsatellites and AFLPs), was evaluated in well-watered and water-stressed conditions. A drought tolerance index was calculated as the ratio between the mean value of the trait in the two environments. For the traits measured, a highly positive correlation was found over the two water regimes, and more than 50% of the quantitative trait loci (QTLs) detected were the same in both; moreover, the direction of the allelic contribution was always consistent, the allele increasing the trait value being mostly from line B73. Several QTLs were common to two or more traits. For the tolerance index, however, most of the QTLs were specific for a single component and different from those controlling the basic traits; in addition, a large proportion of the alleles increasing tolerance were provided by line H99. The data suggest that drought tolerance for yield components is largely associated with genetic and physiological factors independent from those determining the traits per se. The implications of these results for developing an efficient strategy of marker-assisted selection for drought tolerance are discussed. Inheritance and linkage of isozyme loci in pear (Pyrus communis L.)   总被引:2,自引:0,他引:2  
 The polymorphism of 11 enzymes was analysed in 11 progenies from controlled crosses between pear varieties, using acrylamide and starch electrophoresis gels. Twenty-two loci were identified and segregation was scored for 20 of them. Three pairs of duplicated loci forming intergenic hybrid bands were detected, these correspond to equivalent duplicated genes in apple. A total of 49 active alleles and 1 null allele were identified. Joint segregation analysis revealed three linkage groups, which could all be related to existing groups on the apple map. The conservation of isozyme patterns, duplicated genes and linkage groups indicates a high degree of synteny between apple and pear. Received: 8 July 1996 / Accepted: 19 July 1996  相似文献

Summary The salt-tolerant cultivated tomato (Lycopersicon esculentum) accession, PI174263, and a sensitive cv, UCT5, were crossed to develop reciprocal F1, F2 and BC1 populations for genetic analysis of salt tolerance in tomatoes during seed germination. Variation was partitioned into embryo, endosperm and maternal (testa and cytoplasmic) components. Generation means analysis indicated that there were no significant embryo (additive, dominance or epistatic) effects on germination performance under salt stress. Highly significant endosperm additive and testa dominance effects were detected. The proportion of the total variance explained by the model containing these two components was R2=98.2%. Variance component analysis indicated a large genetic variance with additive gene action as the predominant component. Furhter inspection indicated that this variance was attributable to endosperm additive effects on germinability under salt stress. Narrow-sense heritability was estimated as moderately high. Implications for breeding procedures are discussed.  相似文献

 The expression of freezing tolerance was examined in interspecific F1 and somatic hybrids of potatoes using 20 species and 34 different combinations between hardy and sensitive species. In the field, the frost tolerance of hybrids resembled either that of the hardy parent, the sensitive parent, or the parental mean, depending on the species combination and the genomic ratio (ratio of the number of sets of chromosomes contributed from each parent). Similar phenomena were observed when the non-acclimated freezing tolerance (NA) and the acclimation capacity (ACC) (two independent genetic components of freezing tolerance) were evaluated separately under controlled environments. In general, the expression level of freezing tolerance was higher in hybrids with more genomes contributed from the hardy parent than from the sensitive parent. In addition, the effectiveness or combining ability of genes conferring freezing tolerance from the hardy species also showed some influence on the expression of freezing tolerance. All three parameters, namely NA, ACC and acclimated freezing tolerance (AA) (NA plus ACC), were significantly correlated to the frost tolerance exhibited in the field. This indicates that the controlled freezing test used in this study could provide a good estimate of field performance. In the temperate climate of the northern hemisphere, winter survival of woody plants is determined by the ability to acclimate to freezing temperatures and to undergo a period of dormancy. Cold acclimation in many woody plants is initially induced by short photoperiod and low, non-freezing temperatures. These two factors (5°C and short photoperiod) were used to study changes in the proteome of bark tissues of 1-year-old peach trees. Difference in-gel electrophoresis technology, a gel-based approach involving the labeling of proteins with different fluorescent dyes, was used to conduct a quantitative assessment of changes in the peach bark proteome during cold acclimation. Using this approach, we were able to identify differentially expressed proteins and to assign them to a class of either ‘temperature-responsive’ or ‘photoperiod-responsive’ proteins. The most significant factor affecting the proteome appeared to be low temperature, while the combination of low temperature and short photoperiod was shown to act either synergistically or additively on the expression of some proteins. Fifty-seven protein spots on gels were identified by mass spectrometry. They included proteins involved in carbohydrate metabolism (e.g., enolase, malate dehydrogenase, etc), defense or protective mechanisms (e.g., dehydrin, HSPs, and PR-proteins), energy production and electron transport (e.g., adenosine triphosphate synthases and lyases), and cytoskeleton organization (e.g., tubulins and actins). The information derived from the analysis of the proteome is discussed as a function of the two treatment factors: low temperature and short photoperiod. The freeze tolerance and accumulation of cryoprotectants was investigated in three geographically different populations of the enchytraeid Enchytraeus albidus (Oligochaeta). E. albidus is widely distributed from the high Arctic to temperate Western Europe. Our results show that E. albidus is freeze tolerant, with freeze tolerance varying extensively between Greenlandic and European populations. Two populations from sub Arctic (Nuuk) and high Arctic Greenland (Zackenberg) survived freezing at −15 °C, whereas only 30% of a German population survived this temperature. When frozen, E. albidus responded by catabolising glycogen to glucose, which likely acted as a cryoprotectant. The average glucose concentrations were similar in the three populations when worms were frozen at −2 °C, approximately 50 μg glucose mg−1 tissue dry weight (DW). At −14 °C the glucose concentrations increased to between 110 and 170 μg mg−1 DW in worms from Greenland. The average glycogen content of worms from Zackenberg and Nuuk were about 300 μg mg−1 DW, but only 230 μg mg−1 DW in worms from Germany showing that not all glycogen was catabolised during the experiment. Nuclear magnetic resonance spectrometry (NMR) was used to screen for other putative cryoprotectants. Proline, glutamine and alanine were up regulated in frozen worms at −2 °C but only in relatively small concentrations suggesting that they were of little significance for freeze survival. The present study confirms earlier reports that freeze tolerant enchytraeids, like other freeze tolerant oligochaete earthworms, accumulate high concentrations of glucose as a primary cryoprotectant.  相似文献
