This study was carried out to evaluate the antifeedant and larvicidal activities of Hyptis suaveolens Poit. (Lamiaceae) leaves crude extracts and their fractions against four lepidopteran pests namely Helicoverpa armigera (Hbn.), Spodoptera litura (Fab.), Earias vittella (Fab.) and Leucinodes orbonalis (G.). Hexane, chloroform and ethyl acetate extracts were tested by leaf disc and fruit disc no-choice methods at 1% and 1,000?ppm concentrations for crude and fractions, respectively. Ethyl acetate extract of H. suaveolens exhibited the maximum antifeedant and insecticidal activity at 1% concentration against all the tested insects. It was subjected to fractionation using silica column chromatography with different combinations of hexane and ethyl acetate used as mobile phase. Among the 15 fractions obtained, fraction 2 showed the maximum antifeedant and insecticidal activity against all tested insects at 1000?ppm concentration. Preliminary phytochemical analysis of fraction 2 showed the presence of terpenoids and alkaloids. H. suaveolens could be considered as a safe and eco-friendly insecticide for lepidopteron pest management.  相似文献

Abstract

Millettia pachycarpa (Bentham), a perennial climbing tree, found endemic in north-east India used by tribes for treating various ailments. To ascertain the potential of M. pachycarpa for pest management, the present investigation was formulated against a polyphagous pest, Spodoptera litura (Fabricius). Growth inhibitory activity of various extracts and extract fractions was evaluated against third instar larvae of S. litura through topical as well as diet incorporation method. It was observed that dichloromethane extract showed very good growth inhibition in both topical (GI50 7.77?ppm) and diet incorporation methods (GI50< 5?ppm). The growth inhibitory potential of this plant is also demonstrated by the presence of various abnormalities and reduction in larval and pupal weight.  相似文献

Helicoverpa armigera (Hübner), Earias vittella (Fabricius), Spodoptera litura (Fabricius), Spodoptera exigua (Hübner) (all Lepidoptera: Noctuidae), Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae), and Chilo partellus (Swinhoe) (Lepidoptera: Crambidae) are the major pests of cotton and maize. Mass‐rearing of these insects under controlled conditions is necessary to obtain the numbers needed to conduct bioassays to screen insecticides, proteins, and other compounds, as tools for insect pest management. We present a diet suitable for rearing the six lepidopteran pests (five cotton and one maize pest). We further show that this diet is on par with or superior to the published diet recipes for each of the insect species, which were studied for three generations. We also discuss the advantages of antimicrobials other than formalin for keeping microbial growth under check. A combination of antimicrobial solution and benomyl provided effective control and suppressed the growth of microbes for a longer period than a formalin‐containing diet. A common diet for six pests provide opportunities for automation of diet preparation in addition to improved throughput and consistency in the process, while eliminating diet‐batch related errors.  相似文献

Bioefficacy of hexane, chloroform and ethyl acetate extracts of the seeds of Ceasalpinea bonduc (L.) Roxb. was studied against third instar larvae of Spodoptera litura at 0.25, 0.5, 1, 2.5 and 5.0% concentrations. Significant antifeedant, larvicidal and pupicidal activities and least LC50 values were observed in chloroform extract. The chloroform extract was subjected to fractionation using silica gel column chromatography. Six fractions were obtained; among these, the third fraction showed high antifeedant, larvicidal and pupicidal activities at 1000 ppm concentration. Abnormalities in adults were also observed. All the activities were concentration dependent. C. bonduc could be useful in integrated pest management programme.  相似文献

In order to find novel strains of Bacillus thuringiensis that are toxic to some of the major pests that impact economically important crops in Argentina, we initiated a search for B. thuringiensis isolates native to Argentina. We succeeded in assembling a collection of 41 isolates, some of which show a high potential to be used in biological control programs against lepidopteran and coleopteran pests. About 90% of the strains showed toxicity against Spodoptera frugiperda and Anticarsia gemmatalis, two important lepidopteran pests in Argentina. It is noteworthy that only one of these strains contained a cry1-type gene, while another isolate showed a dual toxicity against the lepidopteran and coleopteran insects assayed. Genetic characterization of the strains suggests that the collection likely harbors novel Cry proteins that may be of potential use in biological insect pest control.  相似文献

The common cutworm, Spodoptera litura (Lepidoptera: Noctuidae) is one of the most destructive phytophagous pests of crops. Body coloration affects how animals interact with the environment, and pigmentation also influences behavior and immunity. To explore the mechanisms that drive pigmentation, the CRISPR/Cas9 genome editing system was applied to target the ebony gene in the non‐model insect S. litura. The mutants showed the typically deep coloration during the pupal and adult stage. This study clearly demonstrates the function of ebony in the body coloration and also provides a potentially useful marker gene for functional studies in S. litura as well as other lepidopteran pests (see pages 1011–1019). Six different crude extracts from bark powder of an important medicinal plant, Acacia nilotica (L.) were investigated for their toxicity against Spodoptera litura (Fab.). Total phenolic content in six crude extracts was measured. Ultra-High Performance Liquid Chromatography (UHPLC) was also performed for qualitative analysis of six crude extracts which revealed the presence of 11 phenolic compounds. Bioassays conducted with all crude extracts revealed maximum adverse effect with acetone extract. Duration of larval period and total development period was found to be prolonged. Various aberrations were observed in pupae and adults in the form of wrinkled pupae, blackened pupae, bulged out head part of pupae, partially emerged adults and adults with crumpled wings. All nutritional indices viz. RGR, RCR, ECI, ECD and AD decreased with treatment. The findings indicated that A. nilotica had considerable potential in the management of this serious insect pest.  相似文献

Although genetically modified (GM) plants expressing toxins from Bacillus thuringiensis (Bt) protect agricultural crops against lepidopteran and coleopteran pests, field-evolved resistance to Bt toxins has been reported for populations of several lepidopteran species. Moreover, some important agricultural pests, like phloem-feeding insects, are not susceptible to Bt crops. Complementary pest control strategies are therefore necessary to assure that the benefits provided by those insect-resistant transgenic plants are not compromised and to target those pests that are not susceptible. Experimental GM plants producing plant protease inhibitors have been shown to confer resistance against a wide range of agricultural pests. In this study we assessed the potential of AtSerpin1, a serpin from Arabidopsis thaliana (L). Heynh., for pest control. In vitro assays were conducted with a wide range of pests that rely mainly on either serine or cysteine proteases for digestion and also with three non-target organisms occurring in agricultural crops. AtSerpin1 inhibited proteases from all pest and non-target species assayed. Subsequently, the cotton leafworm Spodoptera littoralis Boisduval and the pea aphid Acyrthosiphon pisum (Harris) were fed on artificial diets containing AtSerpin1, and S. littoralis was also fed on transgenic Arabidopsis plants overproducing AtSerpin1. AtSerpin1 supplied in the artificial diet or by transgenic plants reduced the growth of S. littoralis larvae by 65% and 38%, respectively, relative to controls. Nymphs of A. pisum exposed to diets containing AtSerpin1 suffered high mortality levels (LC50 = 637 µg ml−1). The results indicate that AtSerpin1 is a good candidate for exploitation in pest control.  相似文献

Applications of three concentrations of oil-free neem seed extracts (Azadirachta indica A. Juss; Meliaceae) to cabbage plants in cages did not deter oviposition by individuals of three species of noctuid moths,Trichoplusia ni, Peridroma saucia, andSpodoptera litura. The concentrations used corresponded to 10, 50, and 100 ppm of the main active ingredient, azadirachtin. The total number of eggs laid per female, female longevity, and median day of oviposition were not affected. Sprays of the neem oil-based insecticide Margosan-O R , and a 1% aqueous emulsion of a refined neem seed oil similarly had no effect on any of the parameters studied. However, a 1% crude oil emulsion significantly reduced the proportion of eggs laid byS. litura on treated plants. Our results suggest that literature reports of significant neem-based oviposition deterrence toS. litura are the result of compounds that are removed by higher levels of processing and thus not likely to be found in most commercial neem seed formulations. Sprays consisting of highly processed neem seed extracts, used at concentrations that provide larval control, are unlikely to be generally effective as oviposition deterrents to noctuid pests.  相似文献

In order to evaluate the potential application of microsporidia as a microbial control agent against lepidopteran insect pests, microsporidian infection in a field population of the common cutworm, Spodoptera litura (Fabricius), was surveyed in vegetable crop fields in Can Tho City, Vietnam, in March 2007. The infection rate of microsporidia was 46.7% (99/212 individuals) in adult S. litura, and 16 samples of infected adults were used to characterize the microsporidia at the molecular level. Analysis of the small subunit ribosomal RNA (SSU rRNA) sequences indicated that microsporidian strains isolated from S. litura were closely related to Nosema bombycis from the silkworm, Bombyx mori (Linnaeus); however, phylogenetic analysis based on genome profiling produced a different result from the SSU rRNA sequences. Temperature gradient gel electrophoresis profiles of 12 microsporidian strains from S. litura were closely related to N. bombycis strains, while the profiles of three microsporidian strains formed a different cluster. The Vietnamese strains did not form a single group, but were classified into at least three groups. These results suggested that the microsporidia isolated from S. litura in the Mekong Delta, Vietnam, are genetically diverse.  相似文献

Antifeedant and larvicidal activities of rhein (1,8-dihydroxyanthraquinone-3-carboxylic acid) isolated from the ethyl acetate extract of Cassia fistula flower were studied against lepidopteron pests Spodoptera litura and Helicoverpa armigera. Significant antifeedant activity was observed against H. armigera (76.13%) at 1000 ppm concentration. Rhein exhibited larvicidal activity against H. armigera (67.5), S. litura (36.25%) and the LC50 values was 606.50 ppm for H. armigera and 1192.55 ppm for S.litura. Under laboratory conditions, the biocontrol potential of Steinernema thermophilum was tested against eggs and larval stages of two important lepidopteran insect pests, Helicoverpa armigera and Spodoptera litura (polyphagous pests), as well as Galleria mellonella (used as a model host). In terms of host susceptibility of lepidopteran larvae to S. thermophilum, based on the LC50 36 hr after treatment, G. mellonella (LC50 = 16.28 IJ/larva) was found to be more susceptible than S. litura (LC50 = 85 IJ/larva), whereas neither host was found to be significantly different from H. armigera (LC50 = 54.68 IJ/larva). In addition to virulence to the larval stages, ovicidal activity up to 84% was observed at 200 IJ/50 and 100 eggs of H. armigera and S. litura, respectively. To our knowledge this is the first report of entomopathogenic nematode pathogenicity to lepidopteran eggs. Production of infective juvenile (IJ) nematodes/insect larva was also measured and found to be positively correlated with rate of IJ for H. armigera (r = 0.990), S. litura (r = 0.892), as well as G. mellonella (r = 0.834). Both Phase I and Phase II of symbiotic bacteria Xenorhabdus indica were tested separately against neonates of H. armigera and S. litura by feeding assays and found to be virulent to the target pests; phase variation did not affect the level of virulence. Thus S. thermophilum as well as the nematode’s symbiotic bacteria applied separately have the potential to be developed as biocontrol agents for key lepidopteran pests.  相似文献

Lepidopterans such as Helicoverpa armigera are emerging pests of corn in Korea, causing huge yield losses and deteriorating the quality of the corn crop. We monitored four major lepidopteran pests in major corn growing areas of Korea by employing sex pheromone traps from 2012 to 2015 to understand population dynamics. H. armigera, Ostrinia furnacalis, Spodoptera litura, Spodoptera exigua and Mythimna separata were collected from April to October. Among these species, H. armigera was found to be the main pest based on crop damage intensity (80–90% of the total damage). H. armigera was generally observed during the earing stage of corn and would migrate to other fields during September to October for overwintering. We also tested select insecticides against larvae of H. armigera in the laboratory. The most effective insecticide was indoxacarb, which was applied at specific times to manage H. armigera in cornfields. Optimal timing of spraying was estimated at about 70 days after planting (earing season) to control for H. armigera. Additionally, two species of parasitoid (Therion circumflexum and Ophioninae sp.) were identified from H. armigera. However, more extensive surveys are needed to organize a control program based on natural enemies.  相似文献

Alkaloid extractables of Tylophora asthmatica (Asclepiadaceae) have been tested for the antifeedant activity against Spodoptera litura by incorporating them in semisynthetic and cellulose diets. The alkaloid extractables at 0.01% concentration inhibited feeding of the larvae. Three compounds, tylophorine, tylophorinine and pergularinine, were isolated; and their EC50 values were found to be 2.9, 8.6 and 12.00 ppm, respectively.  相似文献

A member of the potato proteinase inhibitor II (PPI II) gene family that encodes for a chymotrypsin iso-inhibitor has been introduced into tobacco (Nicotiana tabacum) usingAgrobacterium tumefaciens-mediated T-DNA transfer. Analysis of the primary transgenic plants (designated R0) confirmed that the introduced gene is being expressed and the inhibitor accumulates as an intact and fully functional protein. For insect feeding trials, progeny from the self-fertilization of R0 plants (designated R1) were used. Leaf tissue, either from transgenic or from control (non-transgenic) plants, was fed to larvae ofChrysodeixis eriosoma (Lepidoptera: Noctuidae, green looper),Spodoptera litura (F.) (Lepidoptera: Noctuidae) andThysanoplusia orichalcea (F.) (Lepidoptera: Noctuidae) and insect weight gain (increase in fresh weight) measured. Consistently,C. eriosoma larvae fed leaf tissue from transgenic plants expressing thePPI II gene grew slower than insects fed leaf tissue from non-transgenic plants or transgenic plants with no detectablePPI II protein accumulation. However, larvae of bothS. litura andT. orichalcea consistently demonstrated similar or faster growth when fed leaf tissue from transgenic plants compared with those fed non-transgenic plants. In agreement with the feeding trials, the chymotrypsin iso-inhibitor extracted from transgenic tobacco effectively retarded chymotrypsin-like activity measured inC. eriosoma digestive tract extracts, but not in extracts fromS. litura. We conclude, therefore, that for certain insects the use of chymotrypsin inhibitors should now be evaluated as an effective strategy to provide field resistance against insect pests in transgenic plants, but further, that a single proteinase inhibitor gene may not be universally effective against a range of insect pests. The significance of these observations is discussed with respect to the inclusion of chymotrypsin inhibitors in the composite of insect pest resistance factors that have been proposed for introduction into crop plants.  相似文献

An indigenously isolated strain of Bacillus thuringiensis subsp. kenyae exhibited toxicity against lepidopteran as well as dipteran insects. The lepidopteran active cry1Ac protoxin gene coding sequence of 3.5 kb from this strain was cloned into vector pET28a(+). However, it could not be expressed in commonly used Escherichia coli expression hosts, BL21(DE3) and BL21(DE3)pLysS. This gene is classified as cry1Ac17 in the B. thuringiensis toxic nomenclature database. The coding sequence of this gene revealed that it contains about 3% codons, which are not efficiently translated by these expression hosts. Hence, this gene was expressed in a modified expression host, Epicurian coli BL21-Codonplus (DE3)-RIL. The expression of gene yielded a 130-kDa Cry1Ac17 protein. The protein was purified and its toxicity was tested against economically important insect pests, viz., Helicoverpa armigera and Spodoptera litura. LC50 values obtained against these insects were 0.1 ng/cm3 and 1231 ng/cm2, respectively. The higher toxicity of Cry1Ac17 protein, compared to other Cry1Ac proteins, toward these pests demonstrates the potential of this isolate as an important candidate in the integrated resistance management program in India.  相似文献

Insect body pigmentation and coloration are critical to adaption to the environment. To explore the mechanisms that drive pigmentation, we used the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) genome editing system to target the ebony gene in the non-model insect Spodoptera litura. Ebony is crucial to melanin synthesis in insects. By directly injecting Cas9 messenger RNA and ebony-specific guide RNAs into S. litura embryos, we successfully induced a typical ebony-deficient phenotype of deep coloration of the puparium and induction of melanin formation during the pupal stage. Polymerase chain reaction-based genotype analysis demonstrated that various mutations had occurred at the sites targeted in ebony. Our study clearly demonstrates the function of ebony in the puparium coloration and also provides a potentially useful marker gene for functional studies in S. litura as well as other lepidopteran pests.  相似文献

1 The fall armyworm Spodoptera frugiperda is a voracious pest of numerous crops of economic importance throughout the New World. In its native Mexico, larvae can be attacked by several species of parasitic wasps, which are candidate biological control agents against this and other lepidopteran pests. 2 We attempted to survey the parasitoid fauna on S. frugiperda in maize and sorghum fields throughout Mexico. However, our efforts have been hampered by the incomplete development of parasitoid larvae emerging from collected Spodoptera caterpillars. 3 This problem was solved by developing a method to identify seven species of parasitic wasps using polymerase chain reaction amplification and restriction enzyme digestion. This enables the precise determination of the species of those parasitoid larvae that are usually not morphologically identifiable.  相似文献

Host–parasites interaction is a common phenomenon in nature. Diffusive coevolution might maintain stable cooperation in a fig–fig wasps system, in which the exploiter might diversify their genotype, phenotype, or behavior as a result of competition with pollinator, whereas the figs change flower syconia, fruits thickness, and syconia structure. In functionally dioecious Ficus auriculata, male figs and female figs contain two types of florets on separate plant, and share high similarities in outside morphology. Apocryptophagus (Sycophaginae, Chalcidoidea, Hymenoptera) is one of few groups of nonpollinating fig wasps that can reproduce within both male and female figs. On the basis of the morphology and DNA barcoding, evidence from partial sequences of mitochondrial cytochrome c oxidase I and nuclear internal transcribed spacer 2, we found that there are two nonsibling Apocryptophagus species living on male and female F. auriculata figs, respectively. We estimated that these two species diverged about 19.2 million years ago. Our study suggests that the host shift from Ficus variegate or Ficus prostrata fig species to male figs is a preference way for Apocryptophagus wasps to adapt to the separation of sexual function in diecious figs. Furthermore, to escape the disadvantage or sanction impact of the host, the exploiter Apocryptophagus wasps can preferably adapt to exploiting each sex of the figs, by changing their oviposition, niche shift, and habitat.  相似文献

Petroleum ether, acetone, water and aqueous extract of Pedalium murex L. (Pedalaceae) leaves, root and fruit were tested against fourth instar larvae of the tobacco cutworm, Spodoptera litura (Fabr.) under laboratory conditions using leaf dip method. Larval mortality; larval, pupal and adult periods; pupal weight, pupation and adult emergence; larval and pupal deformities of S. litura were recorded. All the tested solvent extracts of P. murex were effective against S. litura life stages by causing mortality in a dose dependent manner. However, the efficacy was more significant with respect to acetone (leaf and root) and petroleum ether (root) at higher concentrations (0.8%) which leads to 100% larval mortality. The water and aqueous extracts of root caused 86.6 and 88.0% larval mortality respectively at 4% concentration. Among the plant parts tested, root had more importance followed by leaf and fruit. Plant extracts extended the larval, pupal and adult periods; reduced the pupal weight, pupal and adult emergence and caused larval–pupal deformities. Total life time was highly prolonged in acetone extracts of P. murex root. Though all the extracts were found to have insecticide activity, acetone and petroleum ether extracts of P. murex root can be used as an effective alternative to modern synthetic insecticides. Bioactive principles from these extracts can be isolated, identified and integrated in S. litura management.  相似文献
